高活性大豆异黄酮发酵豆粕体外抗氧化能力研究   总被引:1,自引:0,他引:1  
[目的]研究高活性大豆异黄酮发酵豆粕(HSMF)的体外抗氧化能力。[方法]选取HSMF、普通发酵豆粕(SMF)和普通豆粕(SM)三种材料,分别制备不同浓度发酵豆粕的大豆异黄酮(SIF)提取液,测定其体外抗氧化能力及苷元转化效率。[结果]发现1,1-二苯基-2-苦肼基(DPPH)清除力和还原力与SIF浓度呈正相关,羟基自由基清除力和超氧阴离子清除力随着浓度的升高而升高,达到一定浓度后保持稳定。当在浓度为100%时,HSMF、SMF、SM的DPPH清除力分别为58.56%、45.32%和31.01%,还原力分别为1.04、0.80和0.44,羟基自由基清除力分别为78.56%、70.54%和65.42%,超氧阴离子清除率分别为61.59%、49.21%和17.53%,苷元型异黄酮所占比例分别为18.84%、50.90%和85.62%。[结论]利用复合益生菌发酵后的豆粕抗氧化能力显著高于SMF及SM(P0.05)。  相似文献

本文通过超临界CO2提取芝麻油的均匀设计实验和微波和超声波诱导提取芝麻油的正交实验比较,考察影响提取的主要因素,寻求最佳萃取工艺。超临界CO2萃取最佳工艺条件为:压力32MPa,温度60℃,CO2流量31kg/h,萃取时问80min,得率46.39%;微波萃取最佳工艺条件为:溶剂为丙酮,物料与溶剂比例1:7,辐射时间7min,辐射功率810W,得率23.01%。超声波萃取最佳工艺条件为:物料与溶剂比例1:7,溶剂为石油醚,浸泡时间30h,得率23.99%。结果表明超临界CO2萃取芝麻油品质最好,而且萃取也最高,质量最稳定。  相似文献

采用均匀设计,对影响建兰ISSR-PCR体系的引物、Mg^2+、dNTP和Taq DNA聚合酶浓度等进行4因素5水平和4因素3水平两轮优化,建立了适合于建兰ISSR-PCR的反应体系:在20μL反应体系中,舍引物0.25μmol/L、Mg^2+ 2.5mmol/L,dNTP0.2mmol/L、Taq DNA聚合酶1.5U和模板DNA40ng.在此基础上对扩增程序中的循环次数和退火温度,以及ISSR引物进行筛选.筛选获得的扩增程序为:94℃预变性5min;接着进行32个循环:94℃变性35S,52~56℃退火45S,72℃延伸90s;循环结束后,72℃延伸10min.同时筛选得到14个扩增稳定、多态性丰富的ISSR引物.  相似文献

本研究以毛竹叶片DNA为模板,利用均匀设计U10(54)表,对毛竹EST-SSRPCR反应体系的TaqDNA聚合酶、Mg2+、dNTPs及引物浓度4因素5个水平进行优化筛选。优化实验结果表明,毛竹EST-SSRPCR的25μL优化反应体系为:10×PCRBuffer(含Mg2+)2.5μL、TaqDNA聚合酶1.5U,Mg2+、dNTPs及引物的最适浓度分别是1.0mmol/L、0.1mmol/L、0.48μmol/L;通过梯度PCR试验筛选得到相应引物的最佳退火温度为52.4℃。对优化出的EST-SSRPCR反应体系进行稳定性检测,结果均能获得丰富、稳定、清晰可辨的DNA谱带。该优化体系为利用EST-SSR分子标记对毛竹进行遗传多样性等相关研究工作提供了基础条件。  相似文献

通过测定溶液粘度,判断超声波降解紫球藻(Porphyridium cruentumi)胞外多糖的效果。运用均匀设计对超声波降解紫球藻胞外多糖的影响因素(处理振幅、时间、脉冲)进行优化,获得超声波处理的最佳条件:振幅39%、处理时间245s和脉冲9.5s。在最佳条件下,胞外多糖的粘度为2.98mm2/s,与预测值一致。采用DPS软件对实验结果进行二次多项式分析与拟合,并对模型和回归系数进行显著性检验,建立了以胞外多糖粘度为目标的回归方程式。  相似文献

利用均匀设计法优化了高丽参多糖的提取过程.主要考察料液质量比、浸泡时间、提取温度和提取时间四个因素对提取率的影响.通过数学模型得到最优提取状况和3D曲面图.结果表明,提取温度对高丽参多糖得率影响最大,在料液质量比1:35、浸泡时间135 min、水提取温度81℃条件下提取156 min,高丽参多糖最大理论提取率为49.51%.最优化模型下实际多糖得率49.29%与理论得率相符.  相似文献

Soy isoflavones are thought to have a cardioprotective effect that is partly mediated by an inhibitory influence on the oxidation of low density lipoprotein (LDL). However, the aglycone forms investigated in many previous studies do not circulate in appreciable quantities because they are metabolised in the gut and liver. We investigated effects of various isoflavone metabolites, including for the first time the sulphated conjugates formed in the liver and the mucosa of the small intestine, on copper-induced LDL oxidation. The parent aglycones inhibited oxidation, although only 5% as well as quercetin. Metabolism increased or decreased their effectiveness. Equol inhibited 2.65-fold better than its parent compound daidzein and 8-hydroxydaidzein, not previously assessed, was 12.5-fold better than daidzein. However, monosulphated conjugates of genistein, daidzein and equol were much less effective and disulphates completely ineffective. Since almost all isoflavones circulate as conjugates, these data suggest that despite the increased potency produced by some metabolic changes, isoflavones may not be effective antioxidants in vivo unless they are deconjugated again.  相似文献

Three new isoflavones, 4′,8-dihydroxy-6,7-dimethoxyisoflavone (1), 4′,6-dihydroxy-8-methoxycarbonyl-7-methoxyisoflavone (2), 4′,7-dimethoxy-8-hydroxymethyl-6-hydroxyisoflavone (3), together with three known flavones (46), were isolated from the leaves of Nicotiana tabacum. Their structures were elucidated by spectroscopic methods, including extensive 1D- and 2D NMR techniques. Compounds 13 were evaluated for their anti-tobacco mosaic virus (anti-TMV) activities. The results showed that compounds 13 (at the concentration of 20 μM) exhibited anti-TMV activities with inhibition rates of 25.2, 22.6, and 27.4%, respectively.  相似文献

Black chickpea is a good source of bioactive compounds, particularly isoflavones. Sprouting improves nutraceutical value in chickpea seeds. This study aimed to explore the role of sprouting of black chickpea seeds on the synthesis of isoflavones and evaluate the impact of the soluble isoflavone on cellular antioxidant activity (CAA) and antiproliferative activity in breast cancer cells. Isoflavones were identified and quantified by HPLC-UV-MS. The CAA and antiproliferative activity were determined in HepG2 cells and MDA-MB-231 cancer cells, correspondingly. In sprouted black chickpea, six isoflavones (formononetin, biochanin-A, and its glycosides) were identified and the total isoflavones content increased (0.31 to 35.72 µgBA/mg of extract). The CAA was increased five times from 137.2 to 788.2 µMEQ/100 g of sample. The bioactive compounds in sprouted chickpea decreased the proliferation of MDA-MB-231 cell line. Also caused morphological changes such as cell shrinkage, rounding and nuclear fragmentation. The results herein suggest that bioactive compounds, as isoflavones, in sprouted black chickpea showed a potential antioxidant and antiproliferative activity. Therefore, it may be considered as a value-added product or ingredient for produce functional foods.  相似文献

将不同浓度的吐温-80添加到野葛毛状根悬浮培养液中,研究在一定的作用时间内其对毛状根生长及次生代谢物合成与分泌的影响。结果表明,采用2%浓度处理较为适宜,不仅可以提高毛状根内葛根素的含量,而且有利于培养液中葛根素、大豆甙元及总异黄酮的积累,与对照相比,其含量可分别提高24.2%、50%和46.7%。在该浓度下连续处理毛状根72h后,发现毛状根仍生长旺盛,其生长量已是对照的1.5倍。但不同时间的连续处理对毛状根及培养液中几种异黄酮物质的积累与释放作用不同,其中以处理48h最有利于培养液中总异黄酮的累积,其含量是毛状根中的38倍。  相似文献

The contents of isoflavones in the developing soybean seedling tissues under different light conditions were examined by high-performance liquid chromatography (HPLC). The results indicated that the total contents of isoflavones in soybean seedling tissues were remarkably different, being the highest in the cotyledons and lower in the leaves and roots. In the cotyledons, the isoflavones like daidzin, genistin and their malonyl conjugates were the major metabolites and were increased markedly with the time of light exposure. In contrast, there was a decrease in the content of isoflavones in the dark. When the dark grown seedlings were tranferred to the light, an increase of the isoflavone content was observed. In the leaves and roots, there was also a marked difference in the contents and types of isoflavones due to various light conditions. Leaves contained mainly the isoflavones such as genistin, malonyl gensistin and flavones rutin, which were increased gradually under light condition, but in the dark, daidzin and malonyl daidzin were the major metabolites and did not change markedly with time. In the roots, no isoflavone was detected in the dark, while five isoflavones were found in the light-grown seedlings, and all root sections were predominated by daidzein and its conjugates. These results provided the evidence that light strongly stimulates the accumulation of isoflavones in soybean seedling tissues.  相似文献
