ABSTRACT

Photoperiodic treatments have been of practical interest in controlling seasonal reproduction in sheep, goats and horses. Melatonin is the principal mediator of the environmental photoperiodic message. To investigate the intra- and inter-subject variability of melatonin 24 h rhythm, ten female Italian Saddle horses (8–10 yrs old, mean body weight 525 ± 30 kg), ten female Sarda breed sheep (2–3 yrs old, mean body weight 40.5 ± 2.8 kg) and ten female Sarda breed goats (3–4 yrs old, mean body weight 38.9 ± 4.1 kg), housed individually in a 4 × 4 m soundproof box equipped with 50 × 100 cm opening windows, were subjected to a natural photoperiod of the vernal equinox (sunrise 06:00 h; sunset 18:00 h). Blood samples were collected from each animal, every 3 h over a 48 h period starting at 00:00 h of day 1 and ending at 00:00 h of day 3. Plasma melatonin concentrations were determined by direct radioimmunoassay (MelatoninDirect RIA, Labor Diagnostika Nord GmbH, Nordhorn, Germany). The application of single cosinor method substantiated a circadian rhythm of melatonin with a nocturnal peak in all studied species. The application of two-way ANOVA on the rhythmic parameters indicated statistically significant differences between the three species in all of the cosinor analysis-derived parameters of MESOR, amplitude, acrophase and robustness of rhythm. Analyses of intra- and inter-subject variability indicate that organization of the melatonin 24 h rhythm is characterized by great accuracy of control within and between the individuals of a breed. In conclusion, features of the 24 h rhythm of melatonin among species; however, the 24 h rhythmicity of melatonin each species showed high stability within the various subjects and within the same subject. These findings must be taken into consideration when applying photoperiod and melatonin treatments for breeding purposes.  相似文献

This study was designed to investigate any seasonal (spring, summer, autumn and winter) changes in haematological parameters in the blood of Tinca tinca measuring the number of red blood cells (RBC), haematocrit, white blood cells, and total plasma proteins.The results show significant changes in RBC and haematocrit in males comparing spring and summer with autumn and winter, whereas in females the RBC remained constant for all 4 seasons but the haematocrit decreased in autumn and winter compared to spring and summer. The white blood cells of male and female animals were significantly lower in spring and winter compared to summer and autumn. In male fish total protein contents significantly decreased in autumn and winter compared to spring and summer, whereas in females protein output significantly decreased in winter compared to the other seasons. The results indicate marked seasonal variation in the blood of male and female Tinca tinca. This variation may play a important protective role for the survival of the animals.  相似文献

Abstract. . Locomotor activity rhythms were studied in eight species of nemobiine crickets with different habitat preferences. They showed similar patterns with a major peak of activity around dusk and a minor one around dawn. These patterns did not change within the natural range of daylengm (12–16 h). The acrophase time of the rhythm varied between -3.2 and +3.5 h as measured from dusk, and the free-running period (T) varied between 22.8 and 24.5 h. In the Dianemobius fascipes species complex these circadian parameters varied among nine local strains (8oS to 43oN) but did not show any latitudinal trend. In a temperate strain of this species complex (Dianemobius nigrofasciatus from Teshikaga, 43oN), two lines which differed from each other in mean T by 1.4h were selected, and a high heritability of T, 0.78 + 0.20 (mean±SE) was obtained. On the other hand, in a subtropical strain (Dianemobius fascipes from Ishigaki Island, 24oN), low heritabilities, 0.35 ± 0.15 for acrophase and 0.17 ± 0.21 for T, were obtained from the full-sib correlations. We studied daily rhythmicity of body temperature (T b) before and during hibernation in Anatolian ground squirrels (Spermophilus xanthoprymnus) under natural and laboratory conditions using surgically implanted temperature loggers. Under both conditions, robust daily T b rhythmicity with parameters comparable to those of other ground squirrel species was observed before but not during hibernation. Euthermic animals had robust daily T b rhythms with a mean of 37.0°C and a range of excursion of approximately 4°C. No T b rhythm was detected during torpor bouts, either because T b rhythmicity was absent or because the daily range of excursion was smaller than 0.2°C. The general patterns of hibernation that we observed in Anatolian ground squirrels were similar to those previously observed by other investigators in other species of ground squirrels.  相似文献

The timing and pattern of mammalian behavioral activities are regulated by an evolutionary optimized interplay of the genetically based biological (circadian) clock located in the brain’s suprachiasmatic nuclei and direct responses to environmental factors that superimpose and thus mask the clock-mediated effects, the most important of which is the photically induced phase-setting (synchronization) of the circadian rhythmicity to the 24-hour solar day. In wild and captive animals living under the natural conditions prevailing in their habitat, to date, only a few attempts have been made to analyze the role of these two regulatory mechanisms in the species’ adaptation to the time structure prevailing in their habitat. We studied the impact of housing conditions and season on the daily timing and pattern of activity in Mexican spider monkeys (Ateles geoffroyi). To this end, we carried out long-term activity recordings with Actiwatch® AW4 accelerometer/data-logger devices in 11 adult Ateles living under identical natural lighting and climatic conditions in either a large wire netting cage or a 0.25?ha forest enclosure in the primatological field station of Veracruz State University near Catemaco, Mexico. In a gravid female in the forest enclosure, we obtained first-hand information on the effect of late pregnancy and parturition on the monkey’s activity rhythm. The Ateles behaved strictly diurnal and undertook about 90% of daily total activity during this activity time. Due to a higher second activity peak in late afternoon, the bimodal activity pattern was more pronounced in monkeys living in the forest enclosure. Although the spider monkeys kept there had an earlier activity onset and morning activity peak than their conspecifics in the cage, no consistent differences were found in the parameters characterizing the phase-setting of the circadian system to the environmental 24-h periodicity, either by comparison or correlation with the external time markers of sunrise (SR) and sunset (SS). The most obvious effect of late pregnancy, parturition and lactation was a distinct reduction of the activity level during the week of parturition and the next. Seasonal variations in the form of significant differences between the long-day summer half year and the short-day winter half year were established in the phase-angle differences of the morning activity peak to SR, in the evening activity peak and activity offset to SS, as well as in the activity time and the peak-to-peak interval, but not in the phase position of activity onset to SR or in the height of the morning and evening activity peak. These findings in combination with a high variability of the phase angle differences indicate that in A. geoffroyi, a relatively weak circadian component and strong masking direct effects of environmental factors are involved in the regulation of the daily activity rhythm.  相似文献

The aim of the present study was to assess the effect of storage time at +4 °C on red blood cell count (RBC), haematocrit (Hct), haemoglobin (Hb), white blood cell count (WBC), thrombocyte count (TC), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH) and mean corpuscular hemoglobin concentration (MCHC) in mullet (Mugil cephalus) using an automatic method. After blood collection (T0), all samples were analyzed using both the manual and automatic method. To test the validation of the automatic method, a paired t‐test was applied, and no statistical difference was observed. The samples were successively divided into four different aliquots and stored at +4 °C to assess the haematological parameters using the automatic method. The first aliquot was refrigerated for 6 h, the second one for 24 h, the third one for 48 h and the last one at for 72 h. One‐way repeated‐measures ANOVA showed a significant effect of storage time (P < 0.05) on Hb, WBC, TC, MCH and MCHC. These results suggest that haematological parameters can be assessed within 6 h from blood collection when samples are stored at +4 °C because long‐term storage modifies the results of the analyses. Further studies on these parameters could be still needed in various fish species to validate an appropriate method for haematological analysis useful not only for the evaluation of the health status of animal living in captivity and in aquaculture but also to have reliability environmental haematological biomarkers. The aim of this study was to assess the accuracy of auricular temperature (AT) recording in the determination of body temperature as well as rectal temperature (RT). For this purpose we compared RT and AT in five clinically healthy horses. Data collections were performed every 3 h over 2 different 24 h photoperiods, 13/11 light/dark cycle and constant darkness. Repeated measures multifactor analysis of variance (MANOVA) was applied to determine a statistical significant effect of time of day, side of temperature collection and photoperiods on AT and RT. Our finding showed a significant effect of time of day on the temperature values recorded and an influence of side of temperature collection. RT values were higher of about 4 °C than the AT values, and the pattern of the two temperatures was not comparable. A daily rhythmicity of rectal temperature was observed. Auricular temperature no showed daily rhythmicity in both periods of monitoring. On the basis of these findings AT does not reflect body temperature as well as RT in horse.  相似文献

In fluctuating environmental temperature, homeotherms are able to maintain stable their body temperature, which however reveals a rhythmic daily pattern as described in literature. Because of the importance of body temperature rhythmicity in the knowledge of thermal homeostasis and as a means to facilitate the study of biological rhythmicity in general, the aim of our study was to assess the influence of two different ambient temperature on the daily rhythmicity of body and auricle temperature in goat (Capra hircus). For our study 6 female adult Maltese goats (18 month old), not pregnant and clinically healthy were used. On each subject body and auricle temperature was recorded during two different periods, of 7 days each, every 3 hours for 24 hours from 07.00 to 04.00, at different environmental temperature (1st period 18.25 ± 1.48°C, 2nd period 28.25 ± 2.05°C). The analysis of the obtained result under two ambient temperature indicates the existence of a daily periodicity, with small differences in amplitude and acrophase between the two studied period, both in body and auricle temperature in Maltese breed goat so we can assume that environmental temperature can influence in a significant way the daily pattern of body and auricle temperature.  相似文献

Circadian rhythms of plasma insulin, Cortisol, and glucose concentrations were examined in scotosensitive (reproductively sensitive to inhibitory effects of short daylengths) and scotorefractory male and female Syrian hamsters (Mesocricetus auratus) maintained on short (LD 10:14) and long (LD 14:10) daylengths. The baseline concentration (mean of all values obtained every 4 hr six times of day) of insulin was much greater in female than in male scotosensitive hamsters kept on short daylengths. These differences in insulin concentration may account for the observed heavy fat stores in female and low fat stores in male scotosensitive hamsters kept on short daylengths. The baseline concentrations of Cortisol were approximately equal in both scotosensitive and scotorefractory males held on short and long daylengths, but were relatively low in females held on short daylengths and especially high in scotorefractory females held on long daylengths.

The plasma concentrations of both cortisol and insulin varied throughout the day in many of the groups tested. However, the variations were not equivalent. The circadian variations of cortisol were similar irrespective of sex, seasonal condition and daylength. Peak concentrations generally occurred about 12 hr after light onset. In contrast, the circadian variations of insulin differed markedly. For example in male hamsters, robust daily variations were found in scotosensitive hamsters held on short daylengths but not on long daylengths and in scotorefractory hamsters held on long daylengths but not on short daylengths. Furthermore, the daily peak occurred during the light in the scotosensitive hamsters and during the dark in the scotorefractory animals. Neither the daily feeding pattern (about 60% consumed during dark) nor the daily variations of glucose concentration varied appreciably with seasonal condition or daylength. They do not appear to determine nor directly reflect the variations in cortisol and glucose concentrations. It is postulated that the daily rhythms of cortisol and insulin are regulated by different neural pacemaker systems and that changes in the phase relations of circadian systems account in part for seasonal changes in body fat stores.  相似文献

Extra-cellular (e) heat shock protein (Hsp)72 has been shown to be elevated in a number of clinical conditions and has been proposed as a potential diagnostic marker. From a methodological and diagnostic perspective, it is important to investigate if concentrations of eHsp72 fluctuate throughout the day; hence, the purpose of the study was to measure resting concentrations of plasma eHsp72 throughout a 24-h period. Blood samples were taken every hour from 1200-2100 hours and from 0700-1200 hours the following day from seven healthy recreationally active males. Participants remained in the laboratory throughout the trial, performed light sedentary activities and were provided with standardised meals and fluids. Physical activity was quantified throughout by the use of an accelerometer. Ethylenediaminetetraacetic acid blood samples were analysed for eHsp72 concentration using a commercially available high-sensitivity enzyme-linked immunosorbent assay (intra-assay coefficient of variation = 1.4%). One-way repeated measures analysis of variance revealed that measures of physiological stress such as heart rate, systolic and diastolic blood pressure remained stable throughout the trial and subjects remained sedentary throughout (mean activity energy expenditure above resting metabolic rate-35.7 +/- 10.0 kcalh(-1)). Plasma Hsp72 concentration did not fluctuate significantly throughout the day and showed no apparent endogenous circadian rhythm in absolute (P = 0.367) or plasma volume change corrected data (P = 0.380). Individual coefficients of variation ranged from 3.8-7.7% (mean 5.4%). Mean Hsp72 concentration across all subjects and time points was 1.49 +/- 0.08 ngml(-1). These data show that in a rested state, plasma eHsp72 concentration shows no apparent endogenous circadian rhythm.  相似文献

The control by light of the flowering response rhythm in the short-day plant Pharbitis nil Choisy cv. Violet was examined by giving a single pulse of light at various times between 1 and 6 h after a 24-h light period. When the first circadian cycle of the rhythm was monitored, it was found that a pulse of red light given at 1, 2 or 3 h into a 72-dark period caused a 1-h delay of the phase of the response rhythm, while a pulse at 6 h caused a 2-h delay. These results support the hypothesis that, when red-light pulses are given at hourly intervals, they are as effective as continuous light in preventing the onset of dark timing because they repeatedly return the rhythm to the circadian time at which it is apparently suspended in continuous light. The perception of and response to continuous light and red-light pulses are also briefly discussed.  相似文献

This study was carried out to examine the seasonal difference in the magnitude of the suppression of melatonin secretion induced by exposure to light in the late evening. The study was carried out in Akita (39° North, 140° East), in the northern part of Japan, where the duration of sunshine in winter is the shortest. Ten healthy male university students (mean age: 21.9±1.2 yrs) volunteered to participate twice in the study in winter (from January to February) and summer (from June to July) 2004. According to Japanese meteorological data, the duration of sunshine in Akita in the winter (50.5 h/month) is approximately one‐third of that in summer (159.7 h/month). Beginning one week prior to the start of the experiment, the level of daily ambient light to which each subject was exposed was recorded every minute using a small light sensor that was attached to the subject's wrist. In the first experiment, saliva samples were collected every hour over a period of 24 h in a dark experimental room (<15 lux) to determine peak salivary melatonin concentration. The second experiment was conducted after the first experiment to determine the percentage of melatonin suppression induced by exposure to light. The starting time of exposure to light was set 2 h before the time of peak salivary melatonin concentration detected in the first experiment. The subjects were exposed to light (1000 lux) for 2 h using white fluorescent lamps (4200 K). The percentage of suppression of melatonin by light was calculated on the basis of the melatonin concentration determined before the start of exposure to light. The percentage of suppression of melatonin 2 h after the start of exposure to light was significantly greater in winter (66.6±18.4%) than summer (37.2±33.2%), p<0.01). The integrated level of daily ambient light from rising time to bedtime in summer was approximately twice that in winter. The results suggest that the increase in suppression of melatonin by light in winter is caused by less exposure to daily ambient light.  相似文献

The aim of this work was to determine the role of intraperitoneally‐administered vitamin E and selenium on the biochemical and haematological parameters in the blood of rats. Thirty‐two adult male Wistar rats were used in this study. All rats were randomly divided into four groups. The first group was used as the control. The second group was intraperitoneally administered with vitamin E (±‐α‐tocopheroryl acetate, 10 mg day−1), the third group with Se (Na2SeO3 0·2 mg over a day), and the fourth group with vitamin E and Se (vitamin E 10 mg + Na2SeO3 0·2 mg over a day). This administration was done for 5 weeks. Blood samples were taken from animals at the end of the dosage period and biochemical parameters in serum samples and haematological parameters in total blood were determined. The levels of total cholesterol (p<0·01) and number of white blood cells (p<0·001) in blood were significantly higher in the vitamin E group than in the control group. The levels of ALP, total cholesterol (p<0·01) and number of white blood cells (p<0·01) in blood were significantly higher in the selenium group than in the controls. The levels of glucose (p<0·05), ALP (p<0·01), total cholesterol (p<0·001) and number of white blood cells (p<0·01) were higher in the vitamin E and selenium combined group than in the controls. Other parameters considered within this trial (ALT, LDH, creatinine, albumin, total protein, amylase, creatine kinase, HDL, triglycerides, total lipid, sodium, chloride, uric acids, red blood cell, haemoglobin, packed cell volume, MCV, MCH, MCHC) did not show statistically significant differences between the control and injected groups. The results indicated that blood glucose and total cholesterol levels, ALP activity and white blood cell counts were significantly increased by intraperitoneal administration of vitamin E and selenium in rats. Studies of the daily rectal temperature rhythms during May and October–November in three female subjects leading normal lives under natural conditions disclosed that although a rhythm of rectal temperature existed in all three subjects in both seasons, the temperature level seems to be lower in October–November than in May, especially during the nighttime. The phasing of the rectal temperature rhythm did not seem to differ between the two seasons.  相似文献

This review first examines reliable and convenient ways of measuring core temperature for studying the circadian rhythm, concluding that measurements of rectal and gut temperature fulfil these requirements, but that insulated axilla temperature does not. The origin of the circadian rhythm of core temperature is mainly due to circadian changes in the rate of loss of heat through the extremities, mediated by vasodilatation of the cutaneous vasculature. Difficulties arise when the rhythm of core temperature is used as a marker of the body clock, since it is also affected by the sleep-wake cycle. This masking effect can be overcome directly by constant routines and indirectly by “purification” methods, several of which are described. Evidence supports the value of purification methods to act as a substitute when constant routines cannot be performed. Since many of the mechanisms that rise to the circadian rhythm of core temperature are the same as those that occur during thermoregulation in exercise, there is an interaction between the two. This interaction is manifest in the initial response to spontaneous activity and to mild exercise, body temperature rising more quickly and thermoregulatory reflexes being recruited less quickly around the trough and rising phase of the resting temperature rhythm, in comparison with the peak and falling phase. There are also implications for athletes, who need to exercise maximally and with minimal risk of muscle injury or heat exhaustion in a variety of ambient temperatures and at different times of the day. Understanding the circadian rhythm of core temperature may reduce potential hazards due to the time of day when exercise is performed.  相似文献

Various studies have been performed using the Social Rhythm Metric (SRM), though none has been developed with stroke patients. Stroke is a pathology that provokes a strong physical and social impact caused by an abnormality in cerebral circulation. Consequently, we performed two studies to validate the SRM and translate it into Portuguese, and to evaluate the regularity of the daily activities of stroke patients. Both healthy individuals and patients with unilateral cerebral lesions were evaluated. Subjects were of both sexes and between 45 and 65 yrs of age. Participants underwent clinical evaluation and recorded the time of 17 daily activities on the SRM for two weeks. Data were analyzed by the Pearson correlation and Fisher tests. After conceptual translation into Portuguese, corrections were made to arrive at the final version. Normative SRM scores varied from 3.2 to 7.0, suggesting that the activities presented in SRM adequately represented the daily routines of the patients. A correlation was found in SRM between the weeks (r=0.84; p=0.0001), indicating instrument reliability. The mean (±SD) score of the stoke patients was 4.8 (±1.0), and the correlation between the SRM and level of neurological damage showed that patients with lower SRM values were more physically compromised (r=?0.29; p=0.04), suggesting that SRM may be a clinical predictor. Activities related to eating and the sleep‐wake cycle were rated by most patients. In all, 71% of the patients did not work, while 84% of healthy individuals did (p=0.001). Only 64% of patients left home compared to 90% of the healthy subjects (p=0.001), and 59% of patients recorded the activity of going home compared to 82% of healthy individuals (p=0.001). According to the results, there is evidence of the validity and reliability of the SRM, enabling it to be reliably used in chronobiological studies of stroke patients. Given that a less regular lifestyle may be associated with neurological compromise and a decrease in social activities, we suggest new studies with the repeated application of this instrument over the clinical evolution of the disease to better define improvement or worsening of the patient's condition in terms of their social and health aspects.  相似文献

Many ungulates are reported to use natural or artificial licks with seasonal patterns around the world. From December 2016 to August 2017, we used infrared camera to record the use of artificial licks in wild sika deer (Cervus nippon) in Zhejiang Qingliangfeng National Nature Reserve, China. We explored the daily rhythm, seasonal pattern and sex difference in lick utilization. In total, 12,043 videos and 22,901 pictures were collected. Our results showed that: (1) the lick visiting frequency was higher at night than that during daytime; (2) the difference in lick visiting frequency between females and males disappeared after taking into account of sex ratio; (3) the lick duration peaked in April during a year. These findings suggested that there were clear daily rhythm and seasonal pattern of lick use in sika deer. Seasonal change in lick use intensity was consistent with our prediction. These variations in lick use might be driven by both the physiological needs of the mineral elements in different life stages and the seasonal changes in climate and food. The reserve management authority should pay more attention to the supplement of licks in spring and summer to fulfill animal’s physiological needs.  相似文献
