Abstract

The present study was conducted to assess the magnitude and health impacts of As in drinking water. Drinking water samples (n?=?60) were collected from twenty different sites of Shiekhupura District (Pakistan). Health risk assessment through average daily dose (ADD), hazard indices (HI), hazard quotient (HQ), carcinogenic risk (CR), and cancer indices (CI) for dermal and oral exposure were determined. Results revealed that As concentration ranged from 2 to 900?µg?L?1 in water samples, which was significantly greater than the safe limit of As (10?µg?L?1) in water. Health risk assessment of As showed that ADD (1.07E?02–9.85E?04), HQ (1.06E+01–9.85E+00), and CR (1.60E?02–9.85E?04) for oral exposure and ADD (1.03E?05–9.69E?06), HQ (1.19E?02–7.96E?03), and CR (1.11E?05–8.98E?05) for dermal exposure which were exceeded the toxic risk index value. Comparison of the two exposure pathways indicated that the oral exposure is much higher risk than the dermal contact. Both values of HI and CI were greater than WHO limit. It is concluded that residents of study area are at higher risk of As induced diseases and carcinogenicity.  相似文献

Europe has the world's most extensive network of conservation areas. Conservation areas are selected without taking into account the effects of climate change. How effectively would such areas conserve biodiversity under climate change? We assess the effectiveness of protected areas and the Natura 2000 network in conserving a large proportion of European plant and terrestrial vertebrate species under climate change. We found that by 2080, 58?±?2.6% of the species would lose suitable climate in protected areas, whereas losses affected 63?±?2.1% of the species of European concern occurring in Natura 2000 areas. Managers of New York and New Jersey Harbor dredging projects are developing strategies to dispose and manage the large volumes of sediment that must be dredged to maintain passable waterways. The various management alternatives include aquatic containment facilities, upland containment, and treatment with beneficial reuse. An important consideration in the selection of an appropriate alternative is the evaluation of potential risks to ecological and human receptors. This study presents a framework for a screening-level ecological and human health risk assessment that compares risks associated with management alternatives for contaminated dredged materials. The major objectives of the work were to identify exposure routes that show the potential for risk and develop a framework that can be used to compare relative potential risks among eight management alternatives. Managers can use this framework to: ? identify characteristics of the placement/treatment alternatives that contribute to potential risk, ? choose one alternative over another for sediments with high concentrations of contaminants, ? implement controls that mitigate risk, or ? identify the need for a more comprehensive site-specific risk assessment.  相似文献

For policy-making to address public risk perceptions effectively, policy-makers must have a clear understanding of the nature of public risks. Public opinion polls regularly solicit perceptions of risk toward a variety of topics. These assessments, though, tend to be general with no specificity offered for a nuanced interpretation. Yet, there is good reason to assume that risk perceptions are not based on the same criteria. If true, policy-makers may be unable to address risks adequately without a better understanding of the drivers of risk perceptions. This project focuses on two primary research questions: (1) Does the public weigh the risk associated with global climate change differently in specific sub-domains? (2) If so, which climate change sub-domains are various members of the public most concerned about when offering a general assessment of global climate change risk? We assess public risk perceptions of climate change in three sub-domains—public health, economic development, and environment—and find that two of the three sub-domains are predictors of a general assessment of risk.  相似文献

We revisit the assumptions associated with the derivation and application of species sensitivity distributions (SSDs). Our questions are (1) Do SSDs clarify or obscure the setting of ecological effects thresholds for risk assessment? and (2) Do SSDs reduce or introduce uncertainty into risk assessment? Our conclusions are that if we could determine a community sensitivity distribution, this would provide a better estimate of an ecologically relevant effects threshold and therefore be an improvement for risk assessment. However, the distributions generated are typically based on haphazard collections of species and endpoints and by adjusting these to reflect more realistic trophic structures we show that effects thresholds can be shifted but in a direction and to an extent that is not predictable. Despite claims that the SSD approach uses all available data to assess effects, we demonstrate that in certain frequently used applications only a small fraction of the species going into the SSD determine the effects threshold. If the SSD approach is to lead to better risk assessments, improvements are needed in how the theory is put into practice. This requires careful definition of the risk assessment targets and of the species and endpoints selected for use in generating SSDs.  相似文献

About 30?years ago, the first Dutch unifactorial guidelines on hypertension and hypercholesterolaemia were developed. These guidelines have been revised several times, often after publication of landmark studies on new generations of drugs. In 1978, cut-off points for pharmacological treatment of hypertension were based on diastolic blood pressure values ≥115?mmHg, and in 2000 they were lowered to >100?mmHg. From 1997 onwards, cut-off points for systolic blood pressure values >180?mmHg were introduced, which became leading. In 1987, cut-offs for hypercholesterolaemia of ≥8?mmol/l were set and from 2006 pharmacological treatment was based on a total/HDL cholesterol ratio >8. Around 2000, treatment decisions for hypertension and/or hypercholesterolaemia were no longer based on high levels of individual risk factors, but on a multifactorial approach based on total risk of cardiovascular diseases (CVD), determined by a risk function. In the 2006 multidisciplinary guideline on cardiovascular risk management, the Framingham risk tables were replaced by European SCORE risk charts. A cut-off point of 10% CVD mortality was set in the Netherlands. In 2011, this cut-off point changed to 20% fatal plus nonfatal CVD risk. Nowadays, 'the lower the risk factors, the lower the absolute risk' is the leading paradigm in CVD prevention.  相似文献

One often observes a familial resemblance of risk factors of disease. The following question then arises: How much familial aggregation of cases of disease would be expected because of this resemblance? This problem is attacked through a particular model where the risk is supposed to depend exponentially on the risk factors. Only pairs of relatives (father/son) are considered. The calculations are performed both with normally distributed risk factors and with particular skewed distributions. An application to coronary heart disease is given.  相似文献

Lead is a toxic element. It can damage multiple human organs and systems. In present study, we detected the blood lead levels (BLLs) during the whole pregnancy period and 6-12th weeks after delivery and analyzed their influencing factors by healthy pregnant women. We recruited 128 healthy pregnant women absent of pregnancy or obstetric complications or abnormal pregnancy outcomes as the gravida group. The control group consisted of 120 healthy non-pregnant women. The lead concentrations of all the three pregnancy trimesters and postpartum were: 5.95?±?2.27, 5.51?±?1.93, 5.57?±?1.85, and 6.88?±?1.90?μg/dl; and the mean lead concentration of control group was 6.87?±?2.29?μg/dl. We found that the BLLs of the gravida group were lower than that of control group during all three trimesters and occupations, supplement nutrition elements, and time of house painted could affect blood lead levels of pregnant women. Lead-related occupations, using cosmetics, and living in a house painted <1?year are risk factors of high BLLs among pregnant women, while calcium, iron, zinc, and milk supplements are protective factors. These may help people especially pregnant women to reduce lead exposure via supplement of calcium, iron, zinc, and milk or avoiding contacting above risk factors.  相似文献

Esophageal cancer was the fifth most commonly diagnosed cancer and the fourth leading cause of cancer-related death in China in 2009. Esophageal squamous cell carcinoma (ESCC) accounts for more than 90% of esophageal cancers. Besides environmental risk factors, genetic factors such as single-nucleotide polymorphisms (SNPs) play an important role in ESCC carcinogenesis. We performed a hospital-based case–control study to evaluate the Forkhead-box protein A1 (FOXA1) rs12894364 C?>?T, rs2145146 C?>?A and rs7144658 T?>?C tag SNPs in the risk of developing ESCC. We recruited 629 ESCC cases and 686 controls. Genotypes were determined using ligation detection reaction. Logistic regression analyses revealed that the three FOXA1 SNPs were not associated with ESCC risk. However, there was significantly decreased ESCC risk associated with the FOXA1 rs12894364 C?>?T and rs2145146 C?>?A polymorphisms among older patients. There was significantly increased ESCC risk associated with the FOXA1 rs7144658 T?>?C polymorphism among male patients. This study demonstrates an association between FOXA1 polymorphisms and ESCC susceptibility. Additional larger studies are required to confirm our findings.  相似文献

Monte Carlo environmental risk assessment requires estimates of the exposure distributions. An exposure of principal concern is often soil ingestion among children. We estimate the long-term (annual) average soil ingestion exposure distribution using daily soil ingestion estimates from children who participated in a mass-balance study at Anaconda, MT. The estimated distribution is accompanied by uncertainty estimates. The estimates take advantage of developing knowledge about bias in soil ingestion estimates and are robust. The estimates account for small particle size soil, use the median trace element estimate for subject days, account for the small sample variance of the median estimates, and use best linear unbiased predictors to estimate the cumulative long term soil ingestion distribution. Bootstrapping is used to estimate the uncertainty of the distribution estimates. The median soil ingestion is estimated as 24?mg/d (sd = 4?mg/d), with the 95 percentile soil ingestion estimated as 91?mg/d (sd = 16.6?mg/d). Strategies are discussed for use of these estimates in Monte Carlo risk assessment.  相似文献

Prey usually adjust anti-predator behavior to subtle variations in perceived risk. However, it is not clear whether adult large carnivores that are virtually free of natural predation adjust their behavior to subtle variations in human-derived risk, even when living in human-dominated landscapes. As a model, we studied resting-site selection by a large carnivore, the brown bear (Ursus arctos), under different spatial and temporal levels of human activity. We quantified horizontal and canopy cover at 440 bear beds and 439 random sites at different distances from human settlements, seasons, and times of the day. We hypothesized that beds would be more concealed than random sites and that beds would be more concealed in relation to human-derived risk. Although human densities in Scandinavia are the lowest within bear ranges in Western Europe, we found an effect of human activity; bears chose beds with higher horizontal and canopy cover during the day (0700?C1900?hours), especially when resting closer to human settlements, than at night (2200?C0600?hours). In summer/fall (the berry season), with more intensive and dispersed human activity, including hunting, bears rested further from human settlements during the day than in spring (pre-berry season). Additionally, day beds in the summer/fall were the most concealed. Large carnivores often avoid humans at a landscape scale, but total avoidance in human-dominated areas is not possible. Apparently, bears adjust their behavior to avoid human encounters, which resembles the way prey avoid their predators. Bears responded to fine-scale variations in human-derived risk, both on a seasonal and a daily basis.  相似文献

The menisci of the knee are two crescent shaped cartilage shock absorbers sitting between the femur and the tibia, which act as load sharers and shock absorbers. Loss of a meniscus leads to a significant increase in the risk of developing arthritis in the knee. Replacement of a missing meniscus with allograft tissue can reduce symptoms and may potentially reduce the risk of future arthritis. Meniscal allograft transplantation is a complex surgical procedure with many outstanding issues, including ‘what techniques should be used for processing and storing grafts?’, ‘how should the allografts be sized?’ and ‘what surgical implantation techniques might be most appropriate?’ Further clinical research is needed and close collaboration between the users (surgeons) and the suppliers (tissue banks) is essential. X-ray repair cross-complementing group 1 gene (XRCC1) has been implicated in risk for lung cancer. However, the results from different studies remain controversial. In this meta-analysis, we have assessed 44 published case-control studies regarding associations of lung cancer risk with three common polymorphisms, codon 194, codon 280 and codon 399, and -77 T?>?C in the promoter region of XRCC1. The results in total population showed that the risk for lung cancer was increased among the variant homozygote Trp/Trp of codon 194 polymorphism, compared with the wild type Arg/Arg (OR: 1.19; 95?% CI 1.01-1.39), and the variant genotype CC of -77 T?>?C polymorphism showed a significantly increased risk of developing lung cancer, compared to wild-type genotype TT (OR: 1.91; 95?% CI 1.24-2.94). However, no associations were found between lung cancer risk and codon 280, codon 399. In the subgroup analyses by ethnicity, the OR for the variant homozygote Trp/Trp of codon 194 was 1.21(95?% CI 1.02-1.43) for Asian. When stratified by source of control, we found a protective effect of codon 194 Arg/Trp genotype (OR: 0.87; 95?% CI 0.77-0.98) and risk effect of codon 399 combined Arg/Gln?+?Gln/Gln variant genotype (OR: 1.09; 95?% CI 1.01-1.18) for lung cancer on the basis of hospital control. Subgroup analyses by histological types of lung cancer indicated that the heterozygote Arg/Trp in codon 194 could decrease and the combined variant genotype Arg/Gln?+?Gln/Gln in codon 399 could increase the risk of non-small cell lung cancer (OR: 0.69; 95?% CI 0.57-0.85 and OR: 1.14; 95?% CI 1.04-1.24). In conclusion, this meta-analysis has demonstrated that codon 194, codon 399 and -77 T?>?C polymorphisms of XRCC1 gene might have contributed to individual susceptibility to lung cancer. To further evaluate effect of XRCC1 polymorphisms, gene-gene interaction and gene-environment interaction on lung cancer risk, a single large sample size study with thousands of subjects is required to get conclusive results.  相似文献

Abstract

The p22phox is a critical component of vascular NADPH oxidases and is encoded by the CYBA gene. It was shown that functionally relevant polymorphisms of the CYBA gene ?930A?>?G, ?852C?>?G, ?675A?>?T, ?536C?>?T, 214C?>?T (previously described as 242C?>?T), *24A?>?G (previously described as 640A?>?G), and *49A?>?G modulate generation of reactive oxygen species (ROS). To analyse whether the CYBA gene polymorphisms ?852C?>?G, ?675A?>?T, and ?536C?>?T were associated with coronary artery disease (CAD), and to designate haplotype blocks. Four hundred and ninety subjects: 245 patients with CAD and 245 age and sex-matched controls. The polymorphisms were genotyped using the PCR-RFLP method and the TagMan® Pre-designed SNP Genotyping Assay. The analysed polymorphisms do not form haplotype blocks. Case–control study revealed that the ?930?G/-675T and ?930G/*49G diplotypes were a CAD risk factor. The 675T/*49G diplotype can modulate CAD risk in women. The protective effect reducing CAD risk in women was related to the ?930A/?675T and ?930A/*49A diplotypes. Carrier state of the ?852C allele (?852C?>?G) was associated with multivessel stenosis while the CC genotype of the ?536C?>?T polymorphism was more frequent in patients with peripheral artery disease. Hypercholesterolemic, cigarette smokers had an increased risk of CAD, especially C???852 allele (?852C?>?G) carriers (SIM?=?3.54; odds ratios (OR)?=?10.01, p?<?0.000). The CYBA gene polymorphisms modulate the risk of CAD but do not form a haplotype blocks.  相似文献

Abstract

The purpose of the study was to acquire the source and evaluate the risk posed by heavy metals in road dust of steel industrial city (Anshan), Liaoning, Northeast China. Potential ecological risk index (RI), pollution index (PI) and geo-accumulation index (Igeo) were applied to evaluate the heavy metal pollution level, and the carcinogenic risk (RI) and hazard index (HI) were calculated to estimate the human health risk. The geographic information system maps clearly reveal the hot spots of heavy metal spatial distribution. Principle component analysis (PCA) and cluster analysis (CA) classified heavy metals into three groups. The metal Zn and Pb originate from the traffic emission, while Cd, Cr, Fe, Mn, Ni and Sb primarily come from industrial activities. These two pathways were the major source of heavy metals pollution by positive matrix factorization (PMF). The Igeo and PI values of heavy metals were decreased in the following order: Cd?>?Sb?>?Zn?>?Fe?>?Pb?>?Cu?>?Cr?>?Sn?>?Mn?>?Ni. The RI index showed the heavy metals were moderate to very high potential ecological risk. The HI values for children and adults presented a decreasing order of Cr?>?Pb?>?Ni?>?Cu?>?Cd?>?Zn. The HI also predicted a possibility of non-carcinogenic risk for children living in urban areas in comparison with adults.  相似文献

Microevolution may determine both the safety and efficacy of classical biological control. Despite a growing body of literature, there are several key unanswered questions regarding the role of evolution in biological control: (1) How common is local adaptation of natural enemies to their hosts or the environment in the native range? How critical is it for success of biological control to find locally adapted agents for importation? (2) Does adaptive evolution following introductions play an important role in biological control? (3) Do introductions of biological control agents impose bottlenecks in population size that reduce genetic variation, and is reduced genetic variation associated with low fitness and poor performance? (4) How great is the risk of evolution of host range of biological control agents? (5) What is the risk of target pests evolving resistance to biological control agents? If pests evolve increased resistance, will biological control agents evolve mechanisms to overcome that resistance? Here, we review the four fundamental processes of microevolution, and discuss how they interact in the context of biological control. We discuss our current state of knowledge regarding the outstanding questions, highlight the types of experiments that can address them, and suggest ways to use microevolution to define risks, and enhance efficacy and safety of biological control.  相似文献

Diabet. Med. 29, e365-e368 (2012) ABSTRACT: Aim Endothelial dysfunction is defined by reduced bioavailability of nitric oxide and has been shown to be associated with cardiovascular risk. The global arginine bioavailability ratio and the arginine to ornithine ratio have recently been shown to be associated with cardiovascular outcome in patients with coronary artery disease. The aim of our study was to investigate the impact of a multifactorial risk factor intervention in subjects with Type?2 diabetes on these two potential new cardiovascular surrogate parameters. Methods In a single-centre and prospective study, we investigated 41 patients with Type?2 diabetes not reaching treatment targets according to current local diabetes guidelines in two out of three of the following measurements: HbA(1c) LDL cholesterol 2.6 or blood pressure. Within 3?months, therapy was intensified according to current guidelines aiming to reach the treatment targets. At baseline and 3?months, arginine, ornithine and citrulline were chromatographically determined after pre-column-derivatization followed by fluorescent detection, and arginine bioavailability ratios were calculated. Results Intensified risk factor management significantly improved the global arginine bioavailability ratio (0.33?±?0.12 at baseline vs. 0.38?±?0.14 after 3?months; P?=?0.018). Posing the question of whether the precautionary principle has a role in risk assessment effectively constrains any debate of the issue within a framework predicated on the assumption that application of risk assessment is inevitable in the formulation of regulatory decisions. The question can equally validly be expressed in terms of whether there is a role for risk assessment in the formulation of precautionary legislation. This allows the debate then to turn on consideration of two questions: Firstly, does the precautionary principle have a role in policy development? and secondly, is this role consistent and compatible with a risk based approach to regulation? When recast in these terms, a more holistic comparison of the aims and objectives of both approaches and of their relative power in the formulation of regulation becomes possible. This leads to the conclusion that the precautionary principle is, when defined and applied correctly, scientifically more robust than risk assessment. Precautionary approaches utilize scientific information and conform robustly to a scientific process but also explicitly incorporate indeterminacies into the decision making framework. Moreover, the precautionary principle when applied to environmental regulation, is more likely to lead to regulation consistent with global sustainability. On this premise, this paper argues that risk based approaches are essentially incompatible with approaches based on the precautionary paradigm, and that of the two, risk assessment is more likely to lead to unsustainable underprotection of the environment.  相似文献
