Background The critical review is an important, though not always well understood and correctly applied element of Life Cycle Assessment studies. It is the intention of this paper to analyse the relevant standards and to present personal experiences in conducting critical reviews.Results and Discussion A peer review for LCA-studies was first proposed in the SETAC guidelines ‘A Code of Practice’ (1993). The ISO standard 14040 (1997) describes three types of ‘Critical review’ which are optional in general, but mandatory ‘for LCA studies used to make a comparative assertion that is disclosed to the public’. For this purpose, the most comprehensive form according to section 7.3.3, the panel method, has to be (‘shall’ in ISO terminology) be used. From personal experience, this method is found to be the most frequently performed in practice (60%), the average panel size being three experts. Large panels with more than 4 experts are rare.Recommendation Personal experience leads to supporting the recommendation by SETAC of the accompanying or ‘interactive’ critical review, which should be preferred, over the review ‘a posteriori’, which offers considerable risks in regards to the duration and costs of an LCA study. ISO 14040, on the other hand, does not recommend one or the other way of performing the critical review.  相似文献

Low-temperature absorption and fluorescence spectra of fully active cores and membrane-bound PS II preparations are compared. Detailed temperature dependence of fluorescence spectra between 5 and 70 K are presented as well as 1.7-K fluorescence line-narrowed (FLN) spectra of cores, confirming that PS II emission is composite. Spectra are compared to those reported for LHCII, CP43, CP47 and D1/D2/cytit b559 subunits of PS II. A combination of subunit spectra cannot account for emission of active PS II. The complex temperature dependence of PS II fluorescence is interpretable by noting that excitation transfer from CP43 and CP47 to the reaction centre is slow, and strongly dependent on the precise energy at which a ‘slow-transfer’ pigment in CP43 or CP47 is located within its inhomogeneous distribution. PS II fluorescence arises from CP43 and CP47 ‘slow-transfer’ states, convolved by this dependence. At higher temperatures, thermally activated excitation transfer to the PS II charge-separating system bypasses such bottlenecks. As the charge-separating state of active PS II absorbs at >700 nm, PS II emission in the 680–700 nm region is unlikely to arise from reaction centre pigments. PS II emission at physiological temperatures is discussed in terms of these results.  相似文献

The white-throated Dipper (Cinclus cinclus) is unique among passerine birds by its reliance on diving to achieve energy gain in fast-flowing waters. Consequently, it should have evolved behavioural adaptations allowing responding directly to runoff patterns (one of the assumptions of the Natural Flow Regime Paradigm—NRFP). In this study (October 1998–August 2001), we investigated how behavioural and energy use strategies in Dippers might vary under the natural flow regime of snowmelt-dominated streams in The Pyrénées (France) where natural flow regime is highly seasonal and predictable. We recorded time spent in each of 5 behavioural activities of ringed birds to estimate time–activity budgets and derive time–energy budgets enabling the modelling of daily energy expenditure (DEE). Annual pattern in ‘foraging’ and ‘resting’ matched perfectly the annual pattern of the natural regime flow and there was a subtle relationship between water stage and time spent ‘diving’ the later increasing with rising discharge up to a point where it fell back. Thus, time–activity budgets meet the main prediction of the NRFP. For males and females Dippers, estimates of feeding rates (ratio Eobs/Ereq = observed rate of energy gain / required foraging rate) and energy stress (M = DEE / Basal Metabolic Rate) also partly matched the NFRP. Maximum value for the ratio Eobs/Ereq was registered in May whilst M peaked in spring. These ratios indicated that Pyrenean Dippers could face high energy stress during winter but paradoxically none during high snowmelt spates when food is expected to be difficult to obtain in the channel and when individual birds were observed spending ca 75% of the day ‘resting’. Annual pattern in DEE did not match the NFRP ; two phases were clearly identified, the first between January to June (with oscillating values 240–280 kJ d− 1 ind− 1) and the second between July and December (200–220 kJ d− 1 ind− 1). As total energy expenditure was higher during the most constraining season or life cycle, we suggest that energy management by Dippers in Pyrenean mountain streams may fit the ‘peak total demand’ hypothesis. At this step of the study, it is not possible to tell whether Dippers use an ‘energy-minimisation’ or an ‘energy-maximisation' strategy.  相似文献

The majority of Middle American cichlids are placed in the informal assemblage ‘Cichlasoma.’ The group is divided into eight sections which appear to be based primarily on trophic morphology. Although several members of ‘Cichlasoma’ have been used in ecomorphological, behavioral, and biogeographic studies, no phylogenetic hypotheses for the group exist. In an attempt to develop a better understanding of the phylogenetic relationships of ‘cichlasomine’ cichlids, we examined the evolution of the trophic specialization, substratum-sifting, in two sections, ‘Cichlasoma(Thorichthys)’ and ‘C.(Amphilophus),’ to determine whether the trait reflects common ancestry. We sequenced the complete mitochondrial cytochromebgene for 19 cichlids representing six sections of ‘Cichlasoma,’ and representatives of other Neotropical Cichlidae. Additional cichlid, and noncichlid outgroup sequences were included for a total of 22 taxa. The molecular phylogeny supports the recognition of the section ‘C.(Thoricthys)’ as a natural group, and we place those cichlids in the genusThorichthys.The phylogeny also depicts ‘C.(Amphilophus)’ as paraphyletic, with substratum-sifters and generalized predators forming separate nonsister clades. We recommend that the substratum-sifting clade of the section ‘C.(Amphilophus)’ be placed in the resurrected genusAstatheros.The generalized predator clade of ‘C.(Amphilophus)’ contains only two species, ‘C. (A.) citrinellum’ and ‘C. (A.) labiatum,’ which we place in the genusAmphilophus.The phylogenetic hypotheses generated indicate that the substratum-sifting generaThorichthysandAmphilophusdo not share a common ancestor. Reconstruction of the evolution of substratum-sifting is equivocal, requiring either the independent evolution of the trait on two separate occasions or its presence in a more inclusive clade and subsequent loss in nonsubstratum sifting species.  相似文献

The concentration (c) and shear rate (γ) dependence of viscosity (η) has been studied for a wide range of random coil polysaccharide solutions, and the following striking generalities are observed:
1. 1. The transition from dilute to concentrated solution behaviour occurs at a critical concentration , when ‘zero shear’ specific viscosity (ηsp) ≈ 10. ηsp varies as c1.4 for dilute solutions, and as c3.3 for concentrated solutions.
2. 2. The shear rate dependence of viscosity, and frequency dependence of dynamic (oscillatory) viscosity are closely superimposable.
3. 3. Double logarithmic plots of against (where η0 is ‘zero shear’ viscosity, and is the shear rate at which ) are essentially identical for all concentrated solutions studied, and thus the two parameters η0 and completely define the viscosity at all shear rates of practical importance.
Departures from points 1 and 2, but not 3, are observed for concentrated solutions of locust bean gum, guar gum, and hyaluronate at low pH and high ionic strength and are attributed to specific intermolecular associations (‘hyperentanglements’) of longer timescale than non-specific physical entanglements.  相似文献

The aim of this work was to investigate whether Fe reduction and antioxidant mechanisms were expressed differently in five Prunus rootstocks (‘Peach seedling,’ ‘Barrier,’ ‘Cadaman,’ ‘Saint Julien 655/2’ and ‘GF-677’). These rootstocks differ in their tolerance to Fe deficiency when grown in the absence of Fe (−Fe) or in presence of bicarbonate (supplied as 5 or 10 mM NaHCO3). Fe deficiency conditions, especially bicarbonate, were shown to decrease Fe and total chlorophyll (CHL) concentration. In the (−Fe)-treated roots of all rootstocks and in the 5 mM NaHCO3-treated ones of the tolerant ‘GF-677’ the Fe(III)-chelate reductase (FCR) activity was stimulated. On the contrary, apart from the ‘GF-677,’ FCR activity was greatly inhibited by the 10 mM NaHCO3. From the results obtained with decapitated rootstocks, it is not entirely clear whether or not the presence of shoot apex was a prerequisite to induce FCR function in all rootstocks tested. In the leaves of rootstocks exposed to the (−Fe) treatment, superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) activities were enhanced whereas the levels of the non-enzymatic antioxidants (FRAP values) were increased in the Fe-deprived leaves, irrespective of the rootstock. Except for ‘Peach seedling,’ foliar SOD activity was stimulated by the presence of NaHCO3. Furthermore, POD activity was increased in the ‘Saint Julien 655/2’ and ‘GF-677,’ but was depressed in the ‘Barrier’ rootstocks exposed to 10 mM NaHCO3. As a result of 10 mM NaHCO3, the expression of a Cu/Zn-SOD and a POD isoform was diminished in the leaves of ‘Peach seedling’ and ‘Barrier,’ respectively. By contrast, an additional isoform of both POD and Mn–SOD were expressed in the leaves of ‘GF-677’ exposed to 10 mM NaHCO3 suggesting that the tolerance of rootstocks to Fe deficiency is associated with induction of an antioxidant defense mechanism. Although CAT activity was increased in the 5 mM NaHCO3-treated leaves of ‘GF-677,’ specifically the 10 mM NaHCO3 treatment resulted in a decrease of CAT activity and an accumulation of H2O2, indicating that bicarbonate-induced Fe deficiency may cause more severe oxidative stress in the rootstocks, than the absence of Fe. A general link between Fe deficiency-induced oxidative stress and Fe reduction-sensing mechanism is also discussed.  相似文献

We have measured the proton longitudinal relaxation times of tissue water of healthy and tumor-bearing mice as a function of the Larmor frequency in the range 6.7 to 90 MHz. These data can be rationalized according to , where A and B are constants specific to the tissue species. We present an interpretation of this frequency dependence within the Fast Exchange Two States model. It is shown that involving a distribution of correlation times for water proton-proton interaction does not yield consistent results, whereas a physically meaningful translational diffusion model pertinent to the dipolar interaction between water protons and macromolecules protons leads to the required frequency dependence. Essentially tissues would differ by the ‘bound’ versus ‘free’ proportion, or by structural properties of cells, rather than by the time-scales governing water motion.  相似文献

Centaurea ragusina L. (Asteraceae) is an endemic Croatian plant species, which developed xeromorphic characteristics as a consequence of its natural environment – vertical limestone cliffs above the Adriatic sea. Cytogenetic status of C. ragusina long-term culture (94th subculture) and C. ragusina seedlings was analysed and compared after 4 weeks of growth. Cytogenetic stability was investigated in root meristem cells of C. ragusina cultured plants originated from Pen đa (cliffs near Dubrovnik) and seedlings originated from three different localities in south Adriatic (Penđa, Pasjača – cliffs near Dubrovnik and island of Komiža) using mitotic index and mitotic and chromosomal abnormalities as parameters. Mitotic indices of cultured plants and ‘Penđa’ seedlings were similar and showed significant increase compared to mitotic indices of ‘Komiža’ and ‘Pasjača’ seedlings. Although the highest number of mitotic abnormalities was recorded in root meristem cells of cultured plants, it was only a bit higher than in root tips of ‘Pasjača’ and ‘Penđa’ seedlings, while that of ‘Komiža’ was two times lower compared to cultured plants. Pattern of analysed mitotic abnormalities was very similar in root tips of cultured plants and ‘Pasjača’ and ‘Penđa’ seedlings, with exception of ‘Komiža’ seedlings. Presented results suggest that long-term cultivation of C. ragusina has almost no effect on culture ageing considering similar distribution of scored mitotic abnormalities as in ‘Penđa’ and ‘Pasjača’ seedlings.  相似文献

The existence of two types of binding sites for ouabain in human erythrocyte membranes is described. Receptor sites designated as ‘type I’, which may be identical to the K+-insensitive sites of intact cells, were detected at concentrations of ouabain as low as 10−7 M. The ‘type II’ receptor sites require the inclusion of Mg2+ + Pi to form complexes with ouabain; they may be identical to the K+-sensitive sites of intact cells. These sites were saturated at approx. 5 · 10−7 M ouabain but could not be detected at higher concentrations. The range of ouabain concentrations at which ‘type I’ receptors start to predominate (i.e. 5 · 10−8–5 · 10−7 M) was termed ‘critical digitalis concentrations’. The process of binding reached equilibrium within 1 and 4 h for ‘type I’ and ‘type II’ sites, respectively. The dissociation constant for ‘type II’ receptor-ouabain complexes was 7.6 · 10−9 M.Under similar experimental conditions, rat erythrocyte membranes exhibited only non-saturable sites.Alterations in the proportions of the two types of receptors were demonstrated by preincubation of the membranes, in the presence or absence of Mg2+ + Pi, prior to the addition of ouabain. In the first case, ‘type II receptor-ouabain’ complexes were stabilized at about 50% of the untreated membranes and ‘type I-ouabain’ complexes slowly approached equilibrium over a period of 24 h. In the latter instance, ‘type I’ receptors were not detected, and only ‘type II-ouabain’ complexes prevailed.  相似文献

This article explores the connections between eugenics, politics and the state, taking the Swiss case as a particular focus. It is argued that Switzerland provides a historical example of what Bauman [Bauman, Z. (1989). Modernity and the Holocaust. Cambridge: Polity Press.] describes as ‘gardening states’: states that are concerned with eliminating the ‘bad weeds’ from the national garden and thereby constructing sharply exclusionary national identities. The Swiss experiments with eugenics (1920s–1960s) can be seen as an example of an ongoing struggle against ‘difference’. Against this backdrop I will examine, first, the ways in which state regulation of reproductive sexuality, and other eugenic measures, became central mechanisms for dealing with cultural and other ‘differences’ in the Swiss nation. Second, I will analyse the gendered nature of such mechanisms, as well as the preoccupation with racial ‘difference’ exemplified by eugenic policies towards ‘Gypsies’. To conclude, I will examine the impact of political institutions and political ideology, in particular, social democracy, on these eugenic gardening efforts.  相似文献

Two cyclic ether (CE) analogues of the prostaglandin endoperoxide PGH2, CE I and CE II, have been found to exert powerful stimulant effects on lung ‘irritant’ receptors and bronchial C-fiber endings after intravascular or aerosol administration in open-chest dogs under Dial-pentobarbital anesthesia. ‘Irritant’ receptors responded to a dose as small as 0.1 μg/kg CE II, injected into the right atrium. CE II was twice as effective as CE I and 10–20 times more potent than PGF. As an aerosol, it exceeded histamine in potency by more than 800 times. ‘Irritant’ receptor stimulation was always associated with decrease in lung compliance and increase in lung resistance. Isoproterenol which reduced the latter effects also diminished the response of ‘irritant’ receptors. Left atrial injection of GES had only weak and delayed effects. CE-induced ‘irritant’ receptor firing declined or ceased during ventilatory arrest in expiration and following hyperinflation of the lungs. In contrast to ‘irritant’ receptors, C-fibers responded more effectively and more rapidly, and in the absence of mechanical changes, when the drugs were injected into the left atrium as compared to right atrial injection. These findings suggest that CE-induced ‘irritant’ receptor stimulation is secondary to changes in lung mechanics, whereas C-fiber stimulation is a direct effect upon the nerve ending.  相似文献

The distribution of metals within the hepatopancreas of Oniscus asellus (Crustacea, Isopoda) from two uncontaminated sites, and two sites contaminated with zinc, cadmium and lead, has been studied by atomic absorption spectrophotometry, light microscopy, transmission and scanning electron microscopy and X-ray microanalysis. The hepatopancreas contains two types of intracellular granule. The first type, in the S cells, are spherical granules which contain copper, sulphur and calcium. In woodlice from contaminated sites, these ‘copper’ granules, also contain zinc, cadmium and lead. The second type, in the B cells, are flocculent deposits which contain iron. In woodlice from contaminated sites, these ‘iron’ granules also contain zinc and lead. Moribund woodlice from contaminated sites have large numbers of ‘copper’ and ‘iron’ granules in the hepatopancreas and a fine deposit of zinc and lead on the membranes of the cells. There are numerous microorganisms in close association with the microvillous border of the hepatopancreas of woodlice from all four sites. Within the microorganisms of Oniscus asellus from contaminated sites, there are deposits of material which contain zinc, lead, calcium and phosphorus ‘Copper’ and ‘iron’ granules could have evolved as storage sites for essential metals to be utilized when demand from the body exceeds uptake from the food. Woodlice in contaminated sites may be able to ‘detoxify’ potentially harmful amounts of essential and non-essential metals by storing them in a relatively insoluble form within these granules.  相似文献

Pigments, chlorophyll fluorescence, dark respiration, and the antioxidant system in the shaded peel of green ‘Anjou’ pear (Pyrus communis L.) and its bud mutation, red ‘Anjou’, were compared in response to high peel temperature, high light alone or in combination to determine the protective role of anthocyanins under high temperature with or without light. Under high temperature treatment alone, no difference in the maximum quantum yield of PSII (FV/FM) was detected between red ‘Anjou’ and green ‘Anjou’; the superoxide dismutase activity and the glutathione pool were up-regulated in green ‘Anjou’ peel but remained unchanged in red ‘Anjou’ peel. Under high temperature coupled with high light, the FV/FM of green ‘Anjou’ peel was decreased to a lower value than that of red ‘Anjou’, and significant interaction was detected between temperature and light for both cultivars. Furthermore, the difference in FV/FM between red ‘Anjou’ and green ‘Anjou’ under high temperature coupled with high light was significantly larger than that under high light alone, indicating that this larger difference was caused by the interaction between high temperature and high light as no significant difference was detected in FV/FM between the two cultivars under high temperature treatment alone at any sampling point. It is concluded that the elevated anthocyanin level in the shaded peel of red ‘Anjou’ does not alter its thermotolerance in the dark, but makes it more tolerant of high temperature under high light.  相似文献

The kinetics and concentration dependence of the binding of dichlorophenyldimethylurea (DCMU) to Photosystem II (PS II) were monitored through fluorescence measurements. According to whether the acceptor system is in the ‘odd’ state (QB ag QB) or ‘even’ state (QB), very different results are obtained. The binding to centers in the even state is rapid ( at [DCMU] = 10−5 M and [chlorophyll] = 10 μg/ml), with a pH-independent rate. The concentration curve of the bound inhibitor (at equilibrium) corresponds to an association constant of about 3.3·107 M−1·1. The binding of the inhibitor to centers in the odd state is slow ( at pH 7, same DCMU and chlorophyll concentrations as above), and depends on pH. In the pH range 6–8, the lower the pH, the slower the kinetics. The association constant is also diminished by a factor of approx. 20 (at pH 7) compared to the even state centers. It is shown that these effects are in good agreement with predictions from Velthuys' hypothesis (Velthuys, B.R. (1981) FEBS Lett. 126, 277–281) that the mode of action of DCMU is a competition with plastoquinone for the binding to the secondary acceptor site. A large part of PS II photochemical quenching corresponds to acceptors which seem to possess a secondary acceptor distinct from B. They were called ‘non-B-type acceptors’ (Lavergne, J. (1982) Photobiochem. Photobiophys. 3, 257–285) and may be identified with Joliot's ‘Q2’ (Joliot P. and Joliot, A. (1977) Biochim. Biophys. Acta 462, 559–574). However, the rate at which the inhibition affects these non-B-type acceptors is similar to the rate of DCMU binding on the B site (i.e., slow in the odd state, fast in the even state).  相似文献

The analysis of benzene in urine of the general population or of exposed workers can be performed with different methods using the ‘purge and trap’ or ‘solid-phase microextraction’ techniques in combination with gas chromatographic analysis and photoionisation or mass spectrometric detection. The published results, however, are deeply conflicting. Differences in sample preparation by different research groups and our own preliminary observations prompted us to investigate pre-analytical and analytical factors potentially capable of modifying the urinary benzene quantification results. Benzene concentrations were measured in 20 urine samples in relation to different conditioning conditions (at 24, 40 and 80°C) and at basic or acid pH. Urinary protein concentrations were measured in the same samples. Urine heating at 80°C yields benzene concentrations on average five times higher than at 24°C. On acidification of urine, the benzene released increases up to 28-fold in comparison to that obtained at uncorrected ‘physiological’ pH. Despite a widely scattered data distribution, a statistically significant linear correlation was found between ‘heat-released’ and ‘acid-labile’ benzene values. There was no correlation between total urinary proteins present in ‘physiological’ concentrations (between 12 and 110 mg/l) and the different kinds of benzene in urine. Our results could perhaps be explained if it is supposed that part of the benzene in urine is absorbed onto sediment, or bound to specific proteins, or derived from parent molecules and is released with pH modification or heat administration. Our observations may also help to explain why the urinary benzene concentrations reported by different investigators vary considerably even when environmental levels are comparable.  相似文献

The mathematical model of Hung (Tsai and Hung, 1984) is empolyed to determine the wall shear rate acting on canine carotid arteries perfused in vitro. Model equations for pulsatile flow in a deformable vessel are coupled with experimental data of dynamic pressure drop, flow rate, vessel radius and radial wall motion. Derived quantities, e.g. velocity profiles and wall shear, are obtained for vessels exposed to ‘normotensive’ hemodynamics, ‘hypertension’ simulations and perfusions in which the compliance of the vessel wall is deliberately altered. Our results indicate that wall shear varies markedly as a function of the hemodynamic environment. The effects of vessel radius vs flow rate on the development of wall shear are also demonstrated. It is found that convective processes correlate with the magnitude of wall shear in the ‘hypertension’ simulations.The present findings and complementary published data may explain, at least in part, the variations in vessel wall transport and endothelial cell biology we observe as a function of the hemodynamic environment. For example we have documented that the exposure of canine carotids to ‘hypertensive’ (vs ‘normotensive’) hemodynamics is associated with an increased flux of lipoproteins (LDL) into the intima and luminal media. Alternations in wall compliance, on the other hand, profoundly influence endothelial shape, orientation and cytoskeletal array.  相似文献

Reduced feed intake and hence lower growth rates commonly occur when the environment of the pig changes, e.g. at weaning and when pigs are moved from growing accommodation to finishing accommodation. It is hypothesised that if environmental factors, such as feeder type, remain the same in the weaning and finishing accommodation this ‘growth check’ may be reduced. A total of 640 pigs in 32 pens of 20 pigs per pen were used to investigate the effects of two feeder types and changing or not changing the feeder type at 10 weeks of age on growth performance and behaviour in the periods from 4 to 10, 10 to finish (22) and from 4 to finish (22) weeks of age, respectively. The two feeder types tested were a ‘wet and dry’ single-space (S) feeder and a ‘dry’ multi-space feeder (M).In the period 4–10 weeks of age feeder type had no significant effect on growth performance though pigs on the ‘dry’ multi-space feeder tended to exhibit significantly better feed efficiency from 4 to 7 weeks of age. In the first week after changing accommodation the growth rate of pigs on all treatments was depressed (compared to the last 3 weeks of the grower period) and was reduced a further 60 g/day when the feeder type was changed. However, the “feeder” effect was transitory and had no effect on overall finisher performance except for pigs changed from a ‘dry’ multi-space feeder to ‘wet and dry’ single-spaced feeder. These animals had significantly faster growth rate over the finisher phase (P<0.05, 886 g/day) and between 4 weeks of age and finish (P<0.05, 730 g/day) than pigs on all other treatments. Feeder treatment had no significant effect on feed intake or feed efficiency in the finisher period or overall from wean to finish. Similarly, the treatments had no effect on the variation in growth rate. Behavioural observations showed that the average number of pigs at the feeder during the finishing period was significantly higher when feed was offered from treatment M–S (1.19 (freq/30 s), P<0.001). In conclusion, the present results suggest that, a change in feeder type, from a ‘dry’ multi-space feeder in growing accommodation to a ‘wet and dry’ single-space feeder in finishing accommodation appears to stimulate improved growth performance in the early finishing period and overall from weaning to finish.  相似文献

Crystalline polysaccharides with 1 → 3−β and 1 → 4−β glycosidic linkages are the most prevalent ones in nature. An interpretation of recent X-ray data on 1 → 3−α glucan shows that it has a ribbon-like crystalline conformation similar to cellulose. Comparison of the crystalline conformation of the four principal homoglucans shows that they fall either in the ‘ribbon-like’ or ‘large amplitude’ helix class. Heteroglucans with a regular sequence of glucosidic linkages show characteristics of the ‘extended conformation’ rather than the ‘coiled conformation’ even when there is 50% of a linkage which in a homoglucan leads to a large amplitude helix. It is concluded that X-ray diffraction analysis fully establishes the hypothesis that the glycosidic linkage type is the determinant of polysaccharide conformation. In this respect, polysaccharides are more like synthetic polymers than proteins or nucleotides; in the latter, it is variation in the substituents which are responsible for the conformational diversity.  相似文献
