When corpora allata from adult female Schistocercagregaria are incubated invitro with either 3H-trans, trans farnesenic acid or 3H-trans, trans, cis bishomo-farnesenic acid and [methyl-14C] methionine, they fabricate large quantities of the corresponding double labelled methyl 10, 11-epoxy esters. Radio GLC of these products indicates retention of geometric configuration at the C-2 and C-6 double bonds. Separate analyses of the contents of the glands and medium after incubation show that the epoxy esters are rapidly released from the glands into the medium and that only the glands contain the corresponding unepoxidized esters. We suggest that unepoxidized esters are the intracellular intermediates in the formation of juvenile hormones from the unsaturated acids. Gel filtration shows that the epoxy esters are not released as stable protein complexes but as simple solutes into the medium. Using this method of promoting the synthesis of juvenile-hormone-active compounds, rates of biosynthesis of epoxy esters of up to 33 ng. per pair of glands per hour have been achieved.  相似文献

The effect of P[N,N-bis(2-chloroethyl)amino]phenylacetate esters of 3β-hydroxy-methyl-17α-aza-d-homo-5α-androstan-17-one (compound 3) and 3β-hydroxy-17α-aza-d-homo-5α-androstane (compound 2) on sister-chromatid exchange (SCE) frequencies and on human lymphocytes proliferation kinetics was studied. The results are compared with those of the P[N,N-bis(2-chloroethyl)phenylacetate esters of 3β-hydroxy-17α-aza-d-homo-5α-androstan-17-one (compound 1). All compounds were found to be active in inducing markedly increased SCE rates and cell division delays. A correlation between potency for SCE induction, effectiveness in cell division delay and previously established antitumour activity of these compounds was observed.  相似文献

We wish to report here the syntheses of (5S, 6R)-5-hydroxy-, (5R, 6R)-5-hydroxy-, (5R, 6S)-5-hydroxy-, and (5S, 6S)-5-hydroxy-PGI1 and their methyl ester derivatives. Treatment of (5R, 6S)-5-epoxy- and (5S, 6R)-5-epoxy-PFG methyl esters with acid washed silica gel afforded (5R, 6R)-5-hydroxy- and (5S, 6S)-5-hydroxy-PGI1 methyl esters; correspondingly, silica promoted cyclization of (5S, 6S)-epoxy- and (5S, 6R)-5-epoxy- PGF1 methyl esters yielded (5S, 6R)-5-hydroxy- and (5R, 6S)-5-hydroxy- PGI1 methyl esters. Alternatively, the 5-hydroxyl group was introduced into the PGI1 skeleton via reaction of the 5-mercuric halides with sodium borohydride in the presence of oxygen. Stereochemical assignments were based on their mode of synthesis and 1H nmr shift differences.  相似文献

Gas chromatography-chemical ionization mass spectrometry has been applied successfully in the analysis of choline and its esters. This approach serves to extend further the potential of existing gas chromatographic procedures which are capable of the microestimation of choline esters following their N-demethylation by either chemical or physical means. Typical fragmentation patterns with ions at me = 72 and me = (M + 1) were obtained for each choline ester derivative. When methane was used as the reactant gas, the above fragments were approximately of equal abundance for each ester. Use of isobutane as reactant gas yielded almost 80% of the (M + 1) fragment, and only approximately 5% of the fragment ion at me = 72. Recovery of all fragments was linear for nondeuterated as well as deuterated analogs of choline ester derivatives. Recovery, as evident from the analysis of records of relative ratios of injected isotopic variants of these esters, indicated that this analysis of choline esters using chemical ionization mass spectrometry coupled with gas chromatography is quantitative and highly reproducible.  相似文献

Creatine phosphate, nucleotides and glycolytic phosphate esters were estimated in extract of beating, in situ freeze clamped, 1312 to 1912 day fetal rat hearts by automated phosphate ester chromatography. Creatine phosphate increased more than 4-fold to almost 9 n moles per mg. protein at 1912 days, while ATP remained relatively constant at about 19 to 21 n moles per mg. protein. Most other nucleotides decreased as gestation advanced. ATP rather than creatine phosphate appears to be the major energy source of fetal rat heart. Except for glucose-6-phosphate, which increased, the glycolytic phosphate esters decreased only very slightly with advancing gestational age, suggesting a relatively stable basal glycolytic activity. Methodology includes correction for phosphate esters of whole blood trapped in extracts of in situ freeze clamped tissues.  相似文献

The alignment of cholesteryl esters in multilayer phosphatidylcholine membranes was investigated using two spin-labelled cholesteryl esters: 10 : 3 ester (I) and 1 : 14 ester (II). The nitroxide label of I is aligned in the membrane with a very large angle of tilt (47° ± 1.5°) with respect to the normal to the membrane surface; II does not show such a tilt. I gives spectra corresponding to immobilized label while II gives nearly isotropic spectra. Ascorbate treatment of the multilayers shows that the labels in I and II are not present at the phosphatidylcholine-water interphase.The data supports a ‘horseshoe’ configuration for the cholesteryl ester in the bilayer, with both the fatty acid chain and the cholesteryl moiety extending deep into the hydrophobic region of the membrane and with the ester linkage near the surface.  相似文献

The enantiomer selection in the nucleophilic addition reaction of optically active amines such as α-amino acid esters to phenylalanine and N-methylphenylalanine N-carboxyanhydride in m-dimethoxybenzene as a solvent has been investigated. Stereoselectivity between the amines and the N-carboxyanhydrides was found to change markedly according to the reaction conditions. This experimental finding is in contrast to the idea hitherto accepted that in the nucleophilic addition-type polymerization of α-amino acid N-carboxyanhydride the growing chain end reacts preferentially with one of the enantiomorphic N-carboxyanhydrides having the same configuration, and indicates the importance of the investigation of stereoselectivity in the N-carboxyanhydride polymerization using suitable model reactions. Most (S)-α-amino acid esters reacted preferentially with (R)-phenylalanine N-carboxyanhydride, and this type of stereoselectivity increased with the N-methylation of N-carboxyanhydride and with increasing bulkiness of the Cα substituent of α-amino acid esters (alanine < norleucine < leucine < valine). The relationship observed between the stereoselectivity and the structures of amines and N-carboxyanhydrides was explained satisfactorily in terms of the transition state model in which the interaction of N-carboxyanhydride nitrogen and α-amino acid ester carbonyl as well as the interaction of N-carboxyanhydride carbonyl and α-amino acid ester nitrogen was taken into account. (S)-Proline ethyl ester did not show enantiomer selectivity toward phenylalanine N-carboxyanhydride, but reacted preferentially with (S)-(N)-methylphenylalanine N-carboxyanhydride. for the reaction of proline ester with N-carboxyanhydride a transition-state model was proposed, which was different from the transition state model proposed for other α-amino acid esters. Some experiments were carried out to examine the transition-state models proposed. The implications of the present investigation in stereoselectivity in the nucleophilic addition-type polymerization of N-carboxyanhydride hitherto reported are discussed.  相似文献

Haemolymph of Manduca sexta fifth instar larvae contains a high esterase activity capable of hydrolyzing Hyalophora cecropia C18 juvenile hormone (JH). In an attempt to characterize the substrate specificity of the enzyme (s) involved, dilute haemolymph was incubated in vitro with a series of JH-analogs. Ethyl esters with or without the 10,11-epoxide group were hydrolyzed readily but isopropyl esters and the (2Z)-isomer of JH were not affected. Indirect evidence was obtained for the hydrolysis of aziridine analogs of JH. Correlations of these results to biological activities are discussed.  相似文献

The current studies present evidence that bilirubin conjugates derived from rat bile undergo rapid transesterification, invitro, in solutions containing methanol. The conjugates of bilirubin and the methyl esters formed from them by exposure to methanol were isolated by thin layer chromatography. The isolates were chemically quantitated for their bilirubin and glucuronic acid composition. Characterization of the bilirubin methyl esters was performed by mass spectrometric analysis of the trimethylsilyl and phenylazo derivatives.  相似文献

A novel synthesis of 16α-hydroxy-4-androstene-3,17-dione (3), 16α-hydroxy-4-androstene-3, 6,17-trione (4), 17β-amino-5-androsten-3β-ol (10) and 17β-amino-4-androsten-3-one (14) is described. 16α-Bromoacetoxy-4-androstene-3, 17-dione (5), 16α-bromoacetoxy-4-androstene-3, 6,17-trione (6) and 17β-bromoacetylamino-4-androsten-3-one (15) were synthesized as potentially selective irreversible inhibitors of androgen aromatases. 16α-Bromo-4-androstene-3,17-dione (1) and 16α-bromo-4-androstene-3, 6,17-trione (2) were converted to compounds 3 and 4 in 80–90% yield by controlled stereospecific hydrolysis using sodium hydroxide in aqueous pyridine. Reductive amination of 3β-hydroxy-5-androsten-17-one and 3-methoxy-3,5-androstadien-17-one (11) using ammonium acetate and sodium cyanohydridoborate (NaBH3CN) and a subsequent treatment with acid gave the amines 10 and 14 respectively, as a salt. The corresponding 17-imino compounds 9 and 13 were also isolated from the reaction mixtures when methanol was used as a solvent for the reaction. The 16α-hydroxyl compounds 3 and 4 and the 17β-amino compound 14 were con- verted to the corresponding bromoacetyl derivatives, 5, 6, and 15, with bromoacetic acid and N,N'-dicyclohexylcarbodiimide.  相似文献

The adenine nucleotide translocase, the transport protein for ADP and ATP, located in the inner mitochondrial membrane is an important site for the regulation of cell metabolism. Inhibition of the adenine nucleotide translocase by long chain fatty acyl CoA esters demonstrated invitro may also occur invivo when the complete oxidation of fatty acids by the myocardium has been compromised during ischemia. Reversal of this biochemical lesion may be of benefit in the preservation of the ischemic myocardium.  相似文献

The epididymis of adult rats metabolize 3H-testosterone by experiments in vivo. Thirty minutes after the injection of 100 μCi 3H-testosterone, some 10 per cent of the total radioactivity of the epididymis was found in the water-soluble fraction, whereas 90 per cent was found in the ether soluble fraction (free steroids). The free steroids were examined further and the following androgenic metabolites identified: testosterone (17β-hydroxy-4-androsten-3-one) 8, 9%, androstendipne (4-androstene-3, 17-dione, 2,7%,5α-A-dione (5α-androstane-3, 17-dione) 6,5%, DHT (17β-hydroxy-5α-androstan-3-one) 47, 2%, 3β-diol (5α-androstane-3β, 17β-diol) 4, 4%, 3α-diol (5α-androstane-3α,17β-diol) 20, 8% and androsterone (3α-hydroxy-5α-androstan-3-one) 3,4%. The relative amount of each metabolite is given in per cent of total radioactivity in the ether soluble fraction.  相似文献

A series of depsipeptides have been synthesized and used to demonstrate the esterase activity of rabbit pulmonary angiotensin converting enzyme. Among the esters studied, Bz-Phe-OPhe-Ala was found to have the highest kcatKm which is about 15 that of its exact peptide analog, Bz-Phe-Phe-Ala. Esters such as Bz-Gly-OGly-Phe, Bz-Gly-OPhe-Phe and Bz-Gly-OLeu-Ala were also hydrolyzed but at much lower rates. Normal Michaelis-Menten behavior is observed and the kinetic parameters obtained indicate that the esters and their peptide analogs bind to the enzyme equally well, but that peptides are hydrolyzed at much higher rates. Studies on the pH-rate profiles, chloride ion effect, inhibition and chemical modifications detect no mechanistic differences between ester and peptide hydrolysis.  相似文献

High mobility group (HMG) proteins 14 and 17 of rat C6 glioma cells are phosphorylated invivo on both serine and threonine. In HMG 14 about 60% of the total [32P]phosphate was identified as phosphoserine and 40% as phosphothreonine. In HMG 17, there was 88% phosphoserine and 12% phosphothreonine. Glioma cell nuclear protein kinase NII phosphorylates HMG 14 and 17 invitro on serine as well as threonine and the relative percentages of [32P]phosphoamino acid are similar to those seen invivo. Nuclear protein kinase NI and the type I and II cAMP-dependent protein kinases exhibit only minor phosphorylating activity towards HMG 14 and 17. We conclude that nuclear protein kinase NII is responsible for the phosphorylation of HMG 14 and 17 invivo.  相似文献

Liver alcohol dehydrogenase is found to possess, in addition to its dehydrogenase and dismutase activities, the ability to hydrolyze octanoate esters at a rate approximately 150011000 of that of the dehydrogenase reaction. The esterase and dehydrogenase activities exhibit an identical isozyme pattern indicating that the same protein catalyzes both reactions. Inhibition studies suggest that the esterase activity presumably shares the catalytic domain with the dehydrogenase activity.  相似文献

The synthesis of 4-ethenylidene-5α-androstane-3β, 17β-diol (5) and of 4-ethenylidene-5α-androstane-3,17-dione (4) is described. Compound 5 is a competitive inhibitor of solubilized bovine microsomal adrenal Δ5-3β-hydroxysteroid dehydrogenase, with Ki =2.7μM, and is converted by the enzyme to the corresponding 3-ketone. Compound 4 shown to irreversibly inactivate the enzyme in a time-dependent manner (t12 =31 min; 55μM; pH =7.0). The substrate, dehydroepiandrosterone, protects against inactivation by compound 4. In contrast, compound 5 is not oxidized at the 3-position by the 3β-(and 17β)-hydroxysteroid dehydrogenase from P. testosteroni, but is oxidized at the 17-position. Nevertheless, the 4-ethenylidene-3,17-diketone (4) causes irreversible time-dependent inactivation (t12 =28min; 64μM; pH =7.0) when incubated directly with this bacterial enzyme, acting as an affinity label.  相似文献

A-ring enollactones 1a, 1b or 9 derived from 4-cholesten-3-one, testosterone benzoate or 3-oxo-4-estren-17β-yl benzoate were condensed with [1,2-13C2]acetyl chloride to give intermediates 2a, 2b or 10. 2a and 2b were cyclized by acid or base to give 3,4-13C2-labeled 4-cholesten-3-one and testosterone, respectively. [3,4-13C2]4-Cholesten-3-one was converted via reduction of its trimethylsilyl enol ether to [3,4-13C2]cholesterol. Acetyl enollactone 10 was cyclized in acetic acid to [3,4-13C2]3-oxo-4-estren-17β-yl benzoate followed by aromatization and hydrolysis to produce [3,4-13C2]estradiol-17β. Alternatively, cyclization of 10 with base afforded [3,4-13C2]3-oxo-4-estren-17β-ol directly, which was then oxidized and aromatized to yield [3,4-13C2]estrone. Ozonolysis of progesterone, conversion to the diketal ester 16 and acylation followed by acid hydrolysis furnished [3,4-13C2]progesterone.  相似文献

The epididymis of adult rats metabolizes 3H-testosterone by experiments invitro. After incubation of slices from epididymal tissue for 2 hrs at 37°C, 8% of the total radioactivity was found in the water-soluble fraction, whereas 92% in the ether soluble fraction (free steroids). The free steroids were examined further and the following metabolites identified: testosterone (17β-hydroxy-4-androsten-3-one) 10,4%, androstendione (4-androstene-3,17-dione) 6,2%, 5α-A-dione (5α-androstane-3,17-dione) 7,3%, DHT (17β-hydroxy-5α-androstane-3-one) 39,3%, 3α-diol (5α-androstane-3α,17β-diol) 22,7%, 3β-diol (5α-androstane-3β,17β-diol) 4,6% and androsterone(3α-hydroxy-5α-androstan-17-one) 8,9%. The relative amount of each metabolite is given in per cent of the total radioactivity in the ether soluble fraction. When segments (caput, corpus, cauda) of epididymis were incubated in the same way, differences in steroid metabolism were demonstrated. Characteristic for caput epididymidis was high formation of DHT (58,4%) and 3α-diol (23,5%). Corpus epididymidis showed lower formation of DHT (50,6%) and 3α-diol (12,7%), but an approximately 3 times higher formation of 5α-A-dione (12,0%) than caput (3,4%) and cauda (3,5%). Cauda epididymis showed the lowest formation of DHT (38,3%), whereas 3α-diol (29,1%) and androsterone (11,4%) formation were relatively high. The ratio between 17β-hydroxy metabolites (DHT and androstanediols) and 17-keto metabolites were much higher in the caput (8,8) than in the corpus (3,2) and cauda (3,6), indicating a higher 5α-reductase activity in this segment.  相似文献

The 9β isomers of estradiol-17β, estradiol-17α, estrone and 17-ethinylestradiol-17β were synthesized and compared with their 9α-counterparts in the rat uterine cytosol estrogen receptor, utero-tropic, and gonadotropin release inhibition assays. Except for 17-ethinyl-9β-estradiol-17β which was as active as its 9α isomer in the uterotropic assay, none of the 9β estrogens exhibited any biological activity which was equal to or greater than their 9α counterparts. For examples, 9β-estradiol-17β was 110 as active as estradiol-17β, and 9β-estrone was 14 as active as estrone in the uterotropic assay.  相似文献
