The routine metabolic rate R R and standard metabolic rate R S were measured in horse mackerel Trachurus trachurus at 13°C over weight range of 1·4–390 g. A data extraction method rather than the more commonly used method of extrapolating the swimming speed-metabolic rate curves back to zero swimming speed was developed to measure the R S . The relation between R R and R S and weight was expressed as a linear regression with the log transformed data. The mean slope of the regression was 0·752 for R S and 0·725 for R S . 相似文献

Mixed function oxidase-catalyzed dearylation of parathion was determined in microsomal fractions of gonadal and liver tissues of fetal and adult horse. When parathion metabolism was calculated on a basis of milligram of protein, the activity was found to be similar between fetal and adult gonads but not liver. However, on a basis of nmoles of cytochrome P-450, parathion metabolism was similar between fetal and adult liver but not gonadal tissues; the metabolism being 20 times higher in the adult than the fetal gonad. 相似文献

A microassay requiring as few as 2 X 10(5) cells per assay was developed for systematic analysis of 9 purine enzymes in lymphocytes from equine peripheral blood, spleen, lymph node, thymus and bone marrow. The activities of adenosine deaminase (ADA), purine nucleoside phosphorylase (PNP), adenosine kinase (AK), deoxyadenosine kinase (dAK), deoxycytidine kinase (dCK), 5'-nucleotidase (5'-N), AMP deaminase, hypoxanthine-guanine phosphoribosyl transferase (HGPRT or HPRT), and adenine phosphoribosyl transferase (APRT) were measured by this microassay in lymphocytes from peripheral blood from four different breeds of horses (Arabian, Quarter Horse, Thoroughbred and Shetland Pony). There were no significant differences in the enzyme activities among the various breeds. Peripheral blood lymphocytes (PBL) from foals exhibited enzyme activities similar to those observed for adult animals. All lymphoid tissue contained similar levels of activity for each kinase (AK, dAK and dCK). Spleen had the highest activity for ADA, PNP, 5'-N, and HGPRT. The lowest activity for ADA, APRT, PNP and AMP deaminase was found in thymus. Enzymatic activities that varied the most among the tissue were 5'-N, ADA, APRT, HGPRT and AMP deaminase. 相似文献

1. In intact horses, heat-induced sweating occurred initially as pulses, then as a continuous, synchronously fluctuating discharge. 2. I.V. adrenaline (Adr) induced sweating immediately; isoprenaline (Isop) elicited sweating after a delay; and phenylephrine (PhE) had no sudorific effect. 3. In isolated perfused skin, PhE induced an immediate small sweat discharge, Isop a slower sustained output and Adr a biphasic discharge. alpha- and beta-adrenergic antagonists blocked the first and second phases, respectively, of Adr-induced sweating. 4. The observed sweating patterns are consistent with independent activation of alpha-adrenergic myoepithelium and beta-adrenergic secretory cells in the sweat glands. 5. Changes in the inbreeding coefficient, F, in the Thoroughbred horse over the past 45 years have been investigated by genotyping 467 Thoroughbred horses (born between 1961 and 2006) using the Illumina Equine SNP50 bead chip, which comprises 54,602 SNPs uniformly distributed across the equine genome. The Spearman rank correlation coefficient, r, between the year of birth and F was estimated. The results indicate that inbreeding in Thoroughbreds has increased over the past 40 years, with r = 0.24, P < 0.001 demonstrating that there is a highly significant, though relatively weak correlation between the year of birth and inbreeding coefficients. Interestingly, the majority of the increase in inbreeding is post-1996 and coincides with the introduction of stallions covering larger numbers of mares. 相似文献

Recent studies demonstrated that zwitterionic buffers could be used for satisfactory storage of equine embryos at 5 degrees C. The success of freezing embryos is dependent upon size and stage of development. Morulae and blastocysts <300 microm can be slowly cooled or vitrified with acceptable pregnancy rates after transfer. The majority of equine embryos are collected from single ovulating mares, as there is no commercially available product for superovulation in equine. However, pituitary extract, rich in FSH, can be used to increase embryo recovery three- to four-fold. Similar to human medicine, assisted reproductive techniques have been developed for the older, subfertile mare. Transfer of in vivo-matured oocytes from young, healthy mares into a recipient's oviduct results in a 70-80% pregnancy rate compared with a 30-40% pregnancy rate when the oocytes are from older, subfertile mares. This procedure can also be used to evaluate in vitro maturation systems. In vitro production of embryos is still quite difficult in the horse. However, intracytoplasmic sperm injection (ICSI) has been used to produce several foals. Cleavage rates of 60% and blastocyst rates of 30% have been reported after ICSI of in vitro-matured oocytes. Gamete intrafallopian tube transfer (GIFT) is a possible treatment for subfertile stallions. Transfer of in vivo-matured oocytes with 200,000 sperm into the oviduct of normal mares resulted in a pregnancy rate of 55-82%. Oocyte freezing is a technique that has proven difficult in most species. However, equine oocytes vitrified in a solution of ethylene glycol, DMSO, and Ficoll and loaded onto a cryoloop resulted in three pregnancies of 26 transfers and two live foals produced. Production of a cloned horse appears to be likely, as several cloned pregnancies have recently been produced. 相似文献

The effects of exercise intensity and duration on blood gases in thoroughbred horses were studied to characterize the apparent exercise-induced failure in pulmonary gas exchange that occurs in these animals. In response to 2 min of exercise, arterial CO2 tension (PaCO2) decreased in mild and moderate exercise, returned to normocapnic levels in moderate to heavy exercise, and rose 5-10 Torr above resting values during very heavy exercise when CO2 production (VCO2) exceeded 20 times the resting value, and mixed venous CO2 tension approximated 140 Torr. Exercise-induced hypoxemia occurred at the onset of heavy exercise and was associated with the absence of a hyperventilatory response and an alveolar-arterial PO2 difference that increased four to six times above rest with very heavy exercise. PaCO2 was related to VCO2 but not fb, as changes in breathing frequency (fb) of 8-20 breaths/min at comparable VCO2 did not affect PaCO2. Prolonging very heavy exercise from 2 to 4 min caused a severe metabolic acidosis (arterial pH less than 7.15) and hypoxemia was maintained; however, CO2 was no longer retained, as PaCO2 gradually fell to below resting levels, due to an increased tidal volume at constant fb. We conclude that a truly compensatory hyperventilation to very heavy exercise in the horse is not achieved because of the excessive volumes and flow rates required by their extraordinarily high VCO2 and VO2. On the other hand, the frank CO2 retention during short-term high-intensity exercise occurs even though the horse is not apparently mechanically obligated to tolerate it. 相似文献

The effect of dexamethasone on the synthesis and degradation of type IV collagen was studied in human fibrosarcoma cells, HT-1080. A dexamethasone concentration as low as 0.1 microM markedly increased collagen synthesis in HT-1080 cells labelled with [14C]proline. The increase in type IV collagen synthesis was not specific, since total protein synthesis was also increased. Further studies indicated that part of the increase was due to an increase in the specific radioactivity of the intracellular proline pool, after dexamethasone treatment. In fact, with dexamethasone concentrations of 0.1-10 microM the relative collagen synthesis was decreased, indicating that synthesis of other protein was increased more than that of type IV collagen. This was also confirmed by measuring the relative amount of type IV collagen RNA by using recombinant plasmid cDNA specific for the human procollagen pro alpha l (IV) RNA. The results indicated that relative collagen synthesis and the relative amount of type IV collagen messenger RNA was decreased similarly, indicating that dexamethasone affected type IV collagen synthesis at the pre-translational level. The dexamethasone-induced effect on total protein and collagen synthesis was maximal after 12-24 h. Dexamethasone induced a marked accumulation of collagen into the cell layer, leading to diminished deposition of soluble collagen into the medium. Since bacterial-collagenase treatment of the cell layer drastically decreased the collagen content of the dexamethasone-treated cells, this indicates that dexamethasone caused an accumulation of collagen into the extracellular matrix of the cell layer. In contrast, the amount of fibronectin was markedly increased in the medium. Dexamethasone decreased the type IV collagen-degrading activity in HT-1080 cells. The HT-1080 cells contained glucocorticoid receptors, as demonstrated by two different methods: by a whole-cell binding assay and by using a cytosol-gel-filtration method. The number of specific binding sites was similar to that in human skin fibroblasts. In conclusion, glucocorticoids affect the metabolism of type IV collagen and fibronectin in HT-1080 cells, and, since these cells contain specific glucocorticoid receptors, the effects are apparently receptor-mediated. 相似文献

Oxygen consumption rates were measured in a school of 56 horse mackerel Trachurus trachurus while at rest and while swimming at steady sustained speeds. Resting values of 38.76 and 42.10mg O2 kg?1 h?1 were measured in a sealed cylindrical tank (535 l) while observing that the fish school remained neutrally buoyant and inactive with only gentle pectoral fin movements and no swimming motion. The same school was trained to swim with projected light patterns within a 10-m diameter annular doughnut respirometer. The oxygen consumption increased from the resting level through 51 mg O2 kg?1 h?1 at the slowest swimming speeds of 0.29 m s?1 (0.95 L s?1) to around 259 mg O2 kg?1 h?1 at the higher measured swimming speed of 0.87 m s?1 (2.82 L s?1). The data fitted a curve where oxygen consumption rose in proportion to velocity to the power of 2.56 with the intercept at the resting level. The maximum sustained speed (80 min) of 1.12 m s?1 (3.63 Ls?1) was not achieved within the respirometer but corresponded to an estimated oxygen consumption of 458.33 mg O2 kg?1 h?1 giving a scope for aerobic activity of 419.02 mg O2 kg?1 h?1. At a speed of 0.87 m s?1, there was a lower bound on the aerobic efficiency of at least 38% and at 1.12 m s?1, the highest aerobic speed, of 40%. Sustained speeds swum in a curved path as here should be increased by 5% for a straight path giving a maximum sustained 80 min speed of 1.18 m s?1. 相似文献

The metabolism and urinary excretion of 1,2(n)-3H-1-dehydrotestosterone were studied in cross-bred gelded horses. Approximately 40% of the dose was excreted in 24 h. The steroid metabolites were extracted by Amberlite XAD-2 resin and fractionated into glucuronides and sulphoconjugates. Unchanged 1-dehydrotestosterone was the only component identified by gas chromatography mass spectrometry after solvolysis of the sulphoconjugates. Positive and negative ion fast atom bombardment mass spectra were obtained on the purified 1-dehydrotestosterone sulphoconjugate isolated from horse urine and on the alkali metal salts of three standard steroid conjugates. Spectra obtained in the different modes were of comparable intensity. Positive ion spectra were generally more complex due to the formation of alkali metal adduct ions containing several sodium cations. The most abundant ion in the negative ion spectra corresponded to the loss of the alkali metal cation to give [M]-. Thus, the structure of a conjugate can be defined from the combination of mass spectrometric techniques. 相似文献

The Maremmano is an Italian warmblood horse breed from central Italy. We characterized the genetic diversity and the degree of admixture in Maremmano in comparison to 14 other European horse breeds using 30 microsatellites. Between-breed diversity explained about 9 per cent of the total genetic diversity. Cluster analysis, genetic distances and genetic differentiation coefficients showed a close relationship of Maremmano with Hanoverian and Lusitano in accordance with breed history. 相似文献
