Spermatogenesis in Blasia pusilla L. was reexamined with electron optics to help evaluate conflicting results of the two earlier light microscope studies of this species. The present micrographic resumé of spermatid transformation confirms Sharp's observation of centrosomes in very young spermatids and agrees with Woodburn's general account of spermatozoid maturation. Further, male gamete development in Blasia is shown to be closely comparable to that of Marchantia and Pellia. It is concluded that Sharp's report of blepharoplast development in Blasia by fragmentation and coalescence was based on anomalous plant material. 相似文献

Chromatin condensation begins as the multilayered structure (MLS) in Phaeoceros reaches its maximally structured differentiation. As nuclear elongation and chromatin condensation proceed, the S2–4 strata disappear, and the nuclear beak extends between the S1 and the nearly spherical anterior mitochondrion. In a mature sperm the mitochondrion is elongate and lies completely anterior to the blunt front end of the nucleus. The 12 S1 tubules extend over the anterior mitochondrion and nucleus, but their number becomes reduced to five at the level where the nucleus' midportion is constricted. The anterior ends of the S1 tubules lie embedded in a rather conical osmiophilic crest. Flagellar insertion is restricted to the extreme anterior tip of the S1 tubules. The locomotory apparatus in Phaeoceros is compared with that of other bryophytes. 相似文献

Electron microscopic examination of thin sections showed that the blepharoplast of a young spermatid of Phaeoceros consists of two side-by-side centrioles and an accumulation of osmiophilic, granular matrix at their proximal ends. Lying between these nearly parallel organelles is a dark-staining body that will later disappear at the onset of flagellogenesis. For a brief period the centrioles are oriented perpendicular to the nuclear surface so that the granular matrix at their proximal ends is confluent with the nuclear envelope; furthermore, the nucleoplasm immediately in front of the centrioles becomes densely staining. The multilayered structure (MLS) develops directly under the centrioles. It comprises a band of 12 microtubules (the S1 stratum) and three lower strata (S2–4) whose constitutent lamellae are oriented at an oblique angle to the S1 axis. While the S1 tubules grow rearward over the nucleus which forms a beak adjacent to the posterior end of the lamellar strata, the centrioles are transformed into basal bodies with the distal growth of the axonemes and the proximal growth of the central cartwheels and lowermost triplets. The proximal ends of the basal bodies and the S1 tubules overlying the lamellar strata are invested with osmiophilic matrix that extends down to the S2 layer and may temporarily occlude the lamellar plates. At the onset of nuclear elongation an anterior mitochondrion becomes situated close beneath the lamellar strata which extend laterally beyond the S1 tubules. 相似文献

Immature sporophytes of Blasia pusilla L. collected in the field were excised from the protective gametophytic tissues and cultured on a slightly modified Knop's agar substrate in microphytotrons. Under the experimental conditions the setae elongated and after 33½ to 6 weeks many began to give rise to from 1 to 20 or more aposporous outgrowths. These subsequently developed into young gametophytes. The apices were then cut off, sterilized, and grown on glucose-mineral agar in aseptic culture. The resulting gametophytes were similar to haploid plants derived from spores in pattern and rate of growth, and in possession of rhizoids, ventral scales, lateral lobes, auricles, and stellate and discoid gemmae. They produced archegonia but no antheridia. The chromosome number of the aposporous plants was 18 in contrast to the normal haploid number of 9. 相似文献

Transmission electron microscopy of serial thin sections was used to reconstruct several early developmental stages of the blepharoplast in Coleochaete pulvinata spermatids. These were compared to published studies of blepharoplast development in Charales and the closest relatives of charophycean green algae among embryophytes, i.e., hornworts and liverworts. Bicentriolar centrosomes such as occur in bryophytes and fern allies were not observed in Coleochaete. Centriole replication in C. pulvinata was orthogonal as in Charales. The resulting two daughter centrioles were oriented perpendicularly and joined proximally by electron-dense material. Their orthogonal relationship was maintained throughout blepharoplast development by a massive, banded connective which appeared early. In spermatids of hornworts and liverworts, a multilayered structure (MLS) develops in association with two centrioles destined to become flagellar basal bodies. When the MLS of these lower land plants is sectioned at right angles to the long axis of the microtubular layer, the MLS is observed to lie beneath cross sections of both centrioles. In contrast, when developing MLSs of C. pulvinata and Charales are similarly sectioned, they occur beside a cross section of just one of the two centrioles. In C. pulvinata (as in other charophytes), MLS lamellae are oriented at a 90-degree angle to the long axis of the S1 microtubules from the beginning. This contrasts with the 40–45 degree angle between the MLS lamellae and S1 microtubules universally reported for archegoniates. In early C. pulvinata spermatids, spline microtubules are closely associated with an anterior mitochondrion having a low stromal density and few cristae. An anterior mitochondrion is typically associated with blepharoplast development in hornworts and liverworts, but has not previously been reported to occur in Coleochaete or any other charophycean alga. In Coleochaete, as in hornworts and liverworts, but unlike Charales, structure of mature blepharoplasts reflects early blepharoplast ontogeny. Very little change in positional relationships among blepharoplast components (flagella, connective, MLS) occurs during development. Spermatids of the liverwort Marchantia polymorpha were shown by analysis of thin-sections to contain a four-layered organelle, the Vierergruppe, lying between the flagellar bases above and a subjacent mitochondrial body. The uppermost layer consists of parallel tubules structurally similar to those of the axoneme. Some of the tubules extend beyond the four-layered region to form a long ribbon-like appendage. The second layer consists of low, vertical, membranous fins connected by numerous short links to the tubules above. The third layer, limited above and below by two horizontal membranes, consists of vertical partitions, apparently continuous with the fins. The fourth and lowermost layer consists of narrow parallel tubules aligned beneath alternate partitions. The long axes of the lower three layers are parallel to each other and lie at an angle of 48° to the tubule axis of the uppermost stratum. A graphic representation of a three-dimensional scale model interpreting organelle structure is given. The Vierergruppe supersedes Dreiergruppe interpretation in most if not all of the bryophytes studied to date. 相似文献

Requirements for spore germination in the rare and native New Jersey fern, Schizaea pusilla Pursh., were examined. Spores did not germinate in darkness and gibberellins (GA) did not induce germination in the dark. However, a dark pretreatment promoted germination in a subsequent light treatment and low temperatures during the dark pretreatment greatly enhanced germination in culture. Three wks of dark pretreatment were required for maximum germination. GA3 promoted germination in red light more effectively than GA4+7. Greater than ten days of continuous illumination was necessary for germination. Spores given red light reached half-maximum germination six days earlier than spores under white light. Red light promoted germination while blue light did not. Far-red light alone could stimulate germination and enhanced the promotive effect of red light; typical phytochrome photoreversibility was not observed. 运用光镜和电镜以及细胞化学方法 ,对中国圆田螺Cipangopaludinachinensis典型精子及其发生的超微结构进行了研究。根据染色质和细胞形态的变化 ,可将其典型精子发生分为五个时期 :精原细胞、初级精母细胞、次级精母细胞、精细胞和成熟精子。精子发生过程中 ,线粒体逐渐增大 ,并随着轴的增生而不断发展为螺旋化线粒体系 ;染色质逐渐凝聚 ,经过颗粒化和纤维化而终呈均质 ;随着染色质的变化 ,细胞核先变小再拉长并螺旋化。精细胞期细胞核基部发生内陷 ,伴随有中心粒的移入 ,并向后方形成“9 2”式的轴。成熟典型精子单鞭毛 ,全长约 40 μm ,包括头、中、尾三段 :头段螺旋状 ,主要由细胞核构成 ,旋束单股 ;中段螺旋状 ,主要由线粒体和轴丝构成 ,旋束四股 ;尾段非螺旋状 ,主要由糖原颗粒和轴丝构成 相似文献

Abstract— The degradation of taurine and GABA in mammalian brain was studied in vivo and in vitro. Small amounts of [35 S]isethionate (10–20 pmol/g brain wet weight) and [35 S]sulphate (about 2 pmol/g) were detected in mouse brain after intramuscular injection of [35 S]taurine. Taurine also produced isethionate in rat brain homogenates (about 20 nmol/h/g protein) and subcellular fractions (about 40 nmol/h/g protein in synaptosomes and about 300 nmol/h/g in mitochondria), but the reaction was not stimulated either by external electrical pulses or by the addition of various cofactors (NAD and NADP in both oxidized and reduced forms, riboflavin, glutathione. pyridoxal-5'-phosphate, ATP) to the incubation medium. [14 C]GABA was readily metabolized to [14 C]succinate both in vivo and in vitro. Isethionate formation activity was concentrated in the mitochondrial fraction, as was also GABA-T activity. Partially purified GABA-T from calf brain also slightly catalysed the formation of [35 S]isethionate (about 1.3 μmol/min/g protein) from [35 S]taurine. It appears that the slight formation of isethionate from taurine is coupled to GABA-T activity. The formation of isethionate from taurine is so small, that it apparently has no role in the control of the brain taurine pool. 相似文献

The pyrenoid structure in 15 species of the Monostroma complex is very diverse us revealed by a study of the morphology of the pyrenoid matrix, associated starch shell, and pattern of intrapyrenoidalthylakoid bands. From these characteristics 8 types of pyrenoid structure were classified. The variation of pyrenoid structure was shown not only among the species studied, but also between the alternation of generations (M. angicava and M. nitidum). In M. fuscum var. splendens, M. groenlandicum, M. undulatum, and M. zostericola pyrenoid structure is the same throughout the life cycle. The pyrenoid matrix of M. zostericola is surrounded by a double membrane that prevents the direct connection of the pyrenoid matrix with chloroplast thylakoids. The pyrenoid also lacks a starch shell. These findings support the establishment of a new genus Kornmannia by Bliding to include M. zostericola. In addition, similarities in pyrenoid ultrastructure suggest an affinity of Capsosiphon fulvescens with M. groenlandicum. 相似文献

本文用免疫电镜方法对脊髓胶状质内GABA能神经元的突触联系进行了超微结构研究。结果表明;脊髓胶状质内有许多GABA能神经元胞体和末梢分布;标记的GABA能神经末梢可作为突触前成分与未标记的GABA形成输一树突触。未标记的末梢可与标记的GABA末梢形成输一轴突触。此外,标记的GABA能神经末梢还可作为突触前成分与标记的GABA能轴突、树突或胞体形成输-轴、轴-树或轴-体突触,即自调节突触。上述结果揭示:GABA能末梢可对脊髓胶状质内其它神经元产生抑制或脱抑制作用。值得注意的是胶状质内含GAnA的神经结构可形成各种形式的自调节突触,并借此实现其对脊髓功能的复杂调节。 相似文献

The vascular bundles in the uppermost 1-4 mm of the coleoptiles of 9 varieties of Avena sativa, and also of Avena fatua L., all terminate essentially vertically with a small “cap” of tracheary elements. In Triticum vulgare Vill., by contrast, they terminate with a horizontal or downward-pointing section. This brings the two bundles more or less together and may result in their complete fusion, usually with a short vascular extension. In both genera the bundles contain one or more series of apparently active, undifferentiated cells. In the mature embryos the bundles are entirely procambial in nature, but xylem differentiation begins rapidly upon germination and proceeds towards the tip, which is reached by the time the coleoptile is 1.5 mm long; thereafter it proceeds basipetally and it may continue at the base after elongation has ceased there. The differentiation of stomata also appears to proceed basipetally. It may be deduced that the coleoptile cannot form lignin while in the embryo but begins to do so upon germination. Parallels are brought out between auxin production first by the endosperm and then by the tip, on the one hand, and lignification in the xylem and in the stomata, on the other. 相似文献
