Summary The genetic control of Endosperm Balance Number (EBN) was investigated by a complete diallel of four exceptional diploid Solanum commersonii-S. chacoense hybrids (1 1/2 EBN) and backcrosses to their species parents, S. commersonii (1 EBN) and S. chacoense (2 EBN). Crosses in which the female parent had a higher EBN value than the male, S. chacoense (2 EBN)XF1 (11/2 EBN) and F1 (11/2 EBN)X S. commersonii (1 EBN), produced viable seed to aborted seed ratios of 11.1 and 11.3, respectively, and had average to small sized viable seed. Crosses in which the female parent had a lower EBN value than the male, S. commersonii (1 EBN)XF1 (11/2 EBN) and F1 (11/2 EBN)XS. chacoense (2 EBN), produced viable seed to aborted seed ratios of 1 7.9 and 1 6.7, respectively, and had average to large sized viable seeds. The results of these crosses appear to be consistent with the relative EBN values of the male and female parent. A model is proposed for the system regulating endosperm development. The assumptions of this model are: (1) three unlinked loci control the system; (2) the loci are homozygous within a species; (3) the genes have additive effects and are of equal strength within a species; (4) the genes within S. chacoense have twice the effect with respect to endosperm regulation as those within S. commersonii; and (5) a slight excess maternal dosage will produce the qualitative effect of small but viable seed. This model, in which quantitative genes operate in a dosage dependent system bears many similarities to classical, threshold-type genetic models.  相似文献

Hexaploid bread wheat was derived from a hybrid cross between a cultivated form of tetraploid Triticum wheat (female progenitor) and a wild diploid species, Aegilops tauschii Coss. (male progenitor). This cross produced a fertile triploid F1 hybrid that set hexaploid seeds. The identity of the female progenitor is unknown, but various cultivated tetraploid Triticum wheats exist today. Genetic and archaeological evidence suggests that durum wheat (T. turgidum ssp. durum) may be the female progenitor. In previous studies, however, F1 hybrids of durum wheat crossed with Ae. tauschii consistently had low levels of fertility. To establish an empirical basis for the theory of durum wheat being the female progenitor of bread wheat, we crossed a durum wheat cultivar that carries a gene for meiotic restitution with a line of Ae. tauschii. F1 hybrids were produced without using embryo rescue techniques. These triploid F1 hybrids were highly fertile and spontaneously set hexaploid F2 seeds at the average selfed seedset rate of 51.5%. To the best of our knowledge, this is the first example of the production of highly fertile F1 hybrids between durum wheat and Ae. tauschii. The F1 and F2 hybrids are both similar morphologically to bread wheat and have vigorous growth habits. Cytological analyses of F1 male gametogenesis showed that meiotic restitution is responsible for the high fertility of the triploid F1 hybrids. The implications of these findings for the origin of bread wheat are discussed.  相似文献

The reproductive isolation barriers and the mating patterns among Pinus pumila, P. parviflora var. pentaphylla and their hybrids were examined by flowering phenology and genetic assays of three life stages: airborne‐pollen grains, adults and seeds, in a hybrid zone on Mount Apoi, Hokkaido, Japan. Chloroplast DNA composition of the airborne‐pollen was determined by single‐pollen polymerase chain reaction. Mating patterns were analysed by estimating the molecular hybrid index of the seed parent, their seed embryos and pollen parents. The observation of flowering phenology showed that the flowering of P. pumila precedes that of P. parviflora var. pentaphylla by about 6 to 10 days within the same altitudinal ranges. Although this prezygotic isolation barrier is effective, the genetic assay of airborne‐pollen showed that the two pine species, particularly P. pumila, still have chances to form F1 hybrid seeds. Both parental species showed a strong assortative mating pattern; F1 seeds were found in only 1.4% of seeds from P. pumila mother trees and not at all in P. parviflora var. pentaphylla. The assortative mating was concluded as the combined result of flowering time differentiation and cross‐incompatibility. In contrast to the parental species, hybrids were fertilized evenly by the two parental species and themselves. The breakdown of prezygotic barriers (intermediate flowering phenology) and cross‐incompatibility may account for the unselective mating. It is suggested that introgression is ongoing on Mount Apoi through backcrossing between hybrids and parental species, despite strong isolation barriers between the parental species.  相似文献

Illus. 1961.—Internal barriers restricting development of F1 hybrids in crosses among closely related diploid species of Solanum belonging to the series Tuberosa and Commersoniana were studied to determine the early stages in development of barriers to gene exchange. Both of the parental series (species complexes) are native to South America, the Tuberosa occurring along the length of the Andes, while the Commersonia occur at lower altitudes in Argentina, Paraguay, and Bolivia. They are distinguished by a fair number of morphological characters. Following crossings between the species of each series and between species of the different series comparisons were made of the number of berries formed per attempted cross, seed per berry, percentage seed germination, and F1 vigor and pollen fertility. Although the species used were closely enough related that all could readily be hybridized, berry and seed set following crosses between species of different series were on the average lower than they were following crosses between species of the same series. These barriers are just forming and are expressed in a varying manner, so that there were significant differences between species of the same series, and even between clones of the same species in barrier expression. The barriers of some of the species were expressed only when they were tested as female or as male parent. The inter-series F1 seeds germinated as well as did those of the parent species and the hybrids had a normal vigor and pollen fertility.  相似文献

Somatic fusion of mesophyll protoplasts was used to produce hybrids between the frost-tolerant species Solanum commersonii (2n=2x=24) and dihaploid S. tuberosum (2n=2x=24). This is a sexually incompatible combination due to the difference in EBN (Endosperm Balance Number, Johnston et al. 1980). Species with different EBNs as a rule are sexually incompatible. Fifty-seven hybrids were analysed for variation in chromosome number, morphological traits, fertility and frost tolerance. About 70% of the hybrids were tetraploid, and 30% hexaploid. Chloroplast counts in stomatal guard cells revealed a low frequency of cytochimeras. The frequency of aneuploids was relatively higher at the hexaploid level (hypohexaploids) than at the tetraploid level (hypotetraploids). The somatic hybrids were much more vigorous than the parents, and showed an intermediate phenotype for several morphological traits and moderate to profuse flowering. Hexaploid hybrid clones were less vigorous and had a lower degree of flowering than the tetraploid hybrid clones. All of the hybrids were female fertile but male sterile except for one, which was fully fertile and self-compatible. Many seeds were produced on the latter clone by selfing and on the male-sterile clones by crossing. The somatic hybrid plants showed an introgression of genes for frost tolerance and an adaptability to cold from S. commersonii. Therefore, the use of these somatic hybrids in breeding for and in genetic esearch on frost tolerance and cold-hardening is suggested.  相似文献

The experimental study was conducted during the period of 2008–2010 at the experimental field of the Institute of Forage Crops in Pleven. The hybridization scheme included direct and back crosses covering four varieties of forage pea (Pisum sativum L.), namely two spring ones, Usatii 90 and Kamerton from Ukraine, and a winter one from Bulgaria, Pleven 10. There was analyzed the inheritance of quantitative traits such as plant height, height to first pod, pod number per plant, seed number per plant, seed number per pod, seed weight per plant and number of fertile nodes per plant of parental components (P1 and P2) and both first (F1) and second (F2) hybrid generations. The cross Usatii 90 × Pleven 10 showed the highest real heterosis effect for plant height (8.26%), pods per plant (158.79%), seeds per plant (272.16%), seeds per pod (42.09%), seed weight per plant (432.43%) and number of fertile nodes per plant (117.14%). The cross Pleven 10 × Usatii 90 had the highest real heterosis effect height to first pod (11.06%). In F2 plants, the strongest depression for plant height (5.88%), seeds per plant (57.88%), seeds per pod (55.93%) and seed weight per plant (55.99%) was in the cross Usatii 90 × Pleven 10, for height to first pod (1.47%) in the cross Kamerton × Pleven 10 and for number of fertile nodes per plant (15.91%) in the cross Pleven 10 × Usatii 90. The highest positive degree of transgression for number of fertile nodes per plant (165.64%) and seed weight per plant (162.10%) was in the cross Pleven 10 × Kamerton and for pod number per plant (102.54%) and seeds per plant (99.13%) in Kamerton × Pleven 10. The stability of the characters was determined. Low variability in F1 and F2 was found in plant height (3.97–6.85%). Variability of number seeds per plant in F1 was highest (11.86–33.23%). For all other traits, the variability varied from average to high. A lower narrow-sense heritability coefficient was observed for plant height, height to first pod, pods per plant, seeds per plant and seed weight per plant (from 0.001 to 0.230). In few cases, such as in fertile nodes per plant (0.39 and 0.81) and seeds per pod (0.44), the coefficients of broad-sense heritability were higher.  相似文献

The seed fertility of 172 interspecific and 32 intraspecific F1 hybrids in the genus Tragopogon was determined. Intrageneric barriers to interbreeding were variable; some were weak and others quite strong. Maternal influences on fertility were found in 18 pairs of reciprocal interspecific crosses. It was postulated that cytoplasmic and nuclear factors interacted to determine the level of seed fertility. Although F1's were generally sterile or semi-sterile, the proportion of fertile seeds produced in most crosses increased in the F2 generation. In crop plants the shift from being annuals to perennials may allow future agricultural systems requiring less energy inputs. The practicability of this was tested for Solanum melongena. Leaf protoplasts of S. melongena (2n = 2x = 24) and one of the related arborescent species Solanum marginatum (2n = 2x = 24) were electrofused and fertile somatic hybrids with arborescent habit regenerated. The magnetic cell sorter (MACS) technique was used for the selection of heterokaryons. The hybrid nature of 18 regenerated plants was assessed on the banding patterns generated by inter-simple sequence repeat PCR. When taken to maturity in the greenhouse, hybrids grew more vigorously compared to the parental species. Their morphological traits were intermediate between those of S. melongena and S. marginatum. Hybrids flowered and produced an average of 85% stainable viable pollen and fertile fruits. The somatic hybrids were maintained in the greenhouse for more than 3 years and continued to produce flowers developing into two types of fruits with plentiful seeds. Fruits were either striated green containing non-germinable seeds or yellow with fully germinable seeds. Their S1 progenies showed common features with S0 hybrids, including fertility and arborescent habit. Cytologically, somatic hybrids exhibited the expected chromosome number of 2n = 4x = 48, while chromosome pairing during microsporogenesis was associated with a low frequency of intergenomic pairing. It is concluded that an arborescent perennial species has been obtained by somatic hybridization. The usefulness of this species per se or in eggplant breeding will depend not only on the transmission of the arborescent habit to cultivated eggplant varieties, but also on the variability that should be created from backcrossing the S. melongena + S. marginatum hybrids to S. melongena.  相似文献

Observations were made on the morphology, seed fertility, pollen viability and meiotic configurations of 22 different interspecific hybrids of 12 North American taxa representative of 3 sections of Ruellia. The hybrids were classified as (1) fully vigorous and fertile; (2) vigorous but with reduced fertility; (3) reduced vigor and fertility; (4) weak and sterile. Hybrids showed meiotic irregularities in microsporogenesis: univalents, chains, precocious separation of bivalents, nondisjunction, and micronuclei were commonly observed. Interspecific pollinations within sect. Dipteracanthus resulted in fertile or partially sterile F1 hybrids. Intersectional crosses produced partially or completely sterile hybrids or failed. Assuming that crossability is correlated with degree of evolutionary relationship, the parental species are interpreted in genecological terms to be grouped in polytypic ecospecies. Hybridization experiments doubtless would be of value in clarifying relationships in tropical and subtropical species of Ruellia.  相似文献

The inheritance of endosperm balance number (EBN), a genetic, dose-dependent crossability system functioning in tuber-bearing Solanum (potato) species, was investigated for certain wild potato species having an EBN equal to one half of their ploidy. The EBN of Solanum acaule, a disomic 4(2EBN) South American species, was investigated by producing F1 and F2 hybrids with artificial 4x(2EBN) S. commersonii. This allowed assessment of recombination among the two genomes of disomic S. acaule and that of S. commersonii. When crossability of the hybrids with 1EBN, 2EBN and 4EBN standards was tested, no variation for EBN was detected. The apparent lack of recombination and segregation for EBN in these hybrids indicates that the genomes of S. acaule and S. commersonii carry EBN in a genetically-similar way. Combined with previous reports, these data indicate that the inheritance of EBN is similar in widely-separated taxa from South America and Mexico.  相似文献

Pollen-tube growth and seed siring ability were measured in crosses between the Louisiana iris species Iris fulva and Iris hexagona and their F1 and F2 hybrids. Flowers of the parental species were pollinated with self, outcross intraspecific, and interspecific pollen. Pollen-tube lengths were similar for all three pollen types in I. fulva, but in I. hexagona interspecific pollen tubes were longer than intraspecific pollen tubes. Pollen-tube lengths also differed for F1 and F2 flowers pollinated with I. fulva, I. hexagona, and hybrid pollen. For both hybrid classes I. fulva pollen tubes were the shortest while pollen tubes from I. hexagona and hybrids grew the furthest. Mixtures of genetically marked pollen were used to assess the seed siring ability of intra- and interspecific pollen in the parental species by varying the proportion of each pollen type in a replacement series design. For both species, the observed proportions of hybrid seeds were lower than the expected based on the frequency of each pollen type in the mixtures across all treatments. Flowers of I. fulva produced less than 10% hybrid progeny even when 75% of the pollen applied to stigmas was derived from interspecific flowers. The frequency of hybrid seed formation was somewhat greater in I. hexagona, but was still significantly lower than expected across all mixture treatments. Seed set per fruit remained constant across the mixture treatments for both species, but in I. fulva fruit set decreased with an increase in the proportion of interspecific pollen. The data indicate that both pre- and postfertilization processes contribute to discrimination against hybrid seed formation.  相似文献

Identifying the contribution of pre‐ and postzygotic barriers to gene flow is a key goal of speciation research. The widespread dung fly species Sepsis cynipsea and Sepsis neocynipsea offer great potential for studying the speciation process over a range of opportunities for gene exchange within and across sister species (cross‐continental allopatry, continental parapatry and sympatry). We examined the role of postcopulatory isolating barriers by comparing female fecundity and egg‐to‐adult viability of F1 and F2 hybrids, as well as backcrosses of F1 hybrids with the parental species, via replicated crosses of sym‐, para‐ and allopatric populations. Egg‐to‐adult viability was strongly but not totally suppressed in hybrids, and offspring production approached nil in the F2 generation (hybrid breakdown), indicating yet unspecified intrinsic incompatibilities. Viable F1 hybrid offspring showed almost absolute male (the heterogametic sex) sterility while females remained largely fertile, in accordance with Haldane's rule. Hybridization between the two species in European areas of sympatry (Swiss Alps) indicated only minor reinforcement based on fecundity traits. Crossing geographically isolated European and North American S. neocynipsea showed similar albeit weaker isolating barriers that are most easily explained by random genetic drift. We conclude that in this system with a biogeographic continuum of reproductive barriers, speciation is mediated primarily by genetic drift following dispersal of flies over a wide (allopatric) geographic range, with some role of natural or sexual selection in incidental or direct reinforcement of incompatibility mechanisms in areas of European sympatry. F1 and backcross hybrids between sexually incompatible species Solanum commersonii and Solanum tuberosum were characterized for glycoalkaloid content and capacity to cold acclimate. Glycoalkaloid (GA) analysis revealed that F1 triploids and BC1 pentaploids contained the glycoalkaloids of both parents. In BC2 (near) tetraploids the situation was different, in that some hybrids produced the GAs of both parents, whereas others contained only the GAs of S. tuberosum. This suggested that the GAs from S. commersonii may be lost rapidly, and that they may have a simple genetic control. The total tuber GA content of BC1 and BC2 groups averaged quite acceptable levels (165.9 mg/kg in BC1 and 192.8 mg/kg in BC2), with six genotypes having a GA content <200 mg/kg fresh weight. The F1 triploid hybrids expressed a capacity to cold acclimate similar to S. commersonii, whereas BC1 and BC2 genotypes generally displayed an acclimation capacity higher than the sensitive parent but lower than S. commersonii. However, one BC1 and two BC2 genotypes with an acclimation capacity as high as S. commersonii were identified. The polar lipid fatty acid composition in S. commersonii and its hybrid derivatives showed that, following acclimation, there was a significant increase in 18:3. Correlation analysis between the capacity to cold acclimate and the increase in 18:3 was significant, suggesting that the increase in 18:3 can be used as a biochemical marker for the assisted selection of cold-acclimating genotypes in segregating populations.Communicated by G.  The wild potato relative Solanum commersonii is reported to carry resistance to bacterial wilt disease caused by Ralstonia solanacearum. To overcome sexual incompatibilites due to differences in ploidy and endosperm balance numbers, somatic hybrids were made that combine the S. tuberosum and S. commersonii genomes. The resulting somatic hybrid plants are vigorous, but their disease resistance level and their fertility was unknown. We therefore tested the S. commersonii and S. tuberosum source material cv Superior, potato cv Atlantic and six somatic hybrid lines for resistance to a virulent strain of R. solanacearum (race 3, biovar 2) at 28°C. As expected, S. commersonii was significantly more wilt-resistant than the cultivated potatoes. In five of the six somatic hybrid lines, disease resistance levels were similar to that of the resistant S. commersonii parent. The resistance level of the sixth somatic hybrid was intermediate, significantly different from both S. commersonii and S. tuberosum. In controlled crosses, the somatic hybrids in this study proved both to be male- and female-fertile and were self-compatible. Summary Crosses were made to obtain interspecific hybrids between B. fruticulosa (wild species , 2n = 16) × B. campestris (cultivar , 2n = 20). Although many pollen grains germinated and their tubes entered the style, only about 30% of the ovules received pollen tubes. Fertilized ovules aborted at various stages of development. A few hybrid seeds resulted from hand pollinations in the field, and they showed poor germination and seedling establishment. The in vitro culture of ovaries, ovules, and seeds increased the frequency of obtaining hybrid seeds and plants: the most effective method was ovary culture followed by ovule culture. The hybrid nature of the plants was confirmed through morphological, cytological, and electrophoretic studies. A meiotic analysis of F1 hybrids (2n = 18) showed that they had 0–5 bivalents and were completely pollen sterile. Electrophoretic analysis of leaf esterases and acid phosphatases of F1 hybrids revealed bands derived from each parent. Induced amphidiploids of F1 hybrids contained mostly bivalents, and had about 50% fertile pollen.  相似文献

Summary Successful crossing is reported between L. atlanticus Gladst. (2n = 38) and L. cosentinii Guss. (2n = 32), using lines of both species selected for crossability followed by selection of relatively fertile progenies. In one cross, 82E75, from a single F2 segregating plant, 22 F3 seeds were obtained. Some other less crossable combinations were completely sterile in the f1 or F2. Backcrossing to both parent species was successful, but some crosses gave relatively more seed by using F2 plants for backcrossing rather than F2's. It is concluded that potential exists for introgression of useful genes in both directions.  相似文献

Hybrid fitness is an important parameter to predict the evolutionary consequences of a hybridization event and to characterize hybrid zones. We studied fitness parameters of F1 and later‐generation hybrids between the lowland species Salix purpurea and the alpine S. helvetica that have recently emerged during colonization of an alpine glacier forefield. Fruit production (number of capsules per catkin and fruit set) did not differ between hybrids and parents, but the number of seeds per capsule of F1 hybrids was slightly lower than that of later‐generation hybrids and of the parents. Germination rates and seedling growth were tested on three substrates (pH 4.5, 7.0, and 8.0). Germination rates of seeds collected from F1 hybrids were lower on acid and neutral substrates, but equal at pH 8.0 compared to all other groups, while the seeds from later‐generation hybrids performed as well as the parents on all three substrates. In seedling growth, the colonizer S. purpurea performed better than all other taxa on all three substrates, while hybrids resembled the subalpine species S. helvetica. Results suggest that endogenous selection acts against F1 hybrids, but favors fitter genotypes in later‐generation hybrids. Exogenous selection via soil pH appears to be weak during seedling establishment. The pioneer vegetation on the glacier forefield may offer sufficient niche space for hybrid seedlings. Owing to the relatively high fitness of the hybrids and the scattered distribution of hybrids and parental individuals on the glacier forefield, this hybrid zone can be assigned to a mosaic model, probably facilitating gene flow and introgression between the parental species. As establishment of the hybrid zone appears to be linked to a colonization process, we propose to call it a pioneer mosaic hybrid zone.  相似文献

Crosses were performed within and between 19 Hawaiian populations of the mostly dioecious woody shrub Wikstroemia. These populations are from a variety of habitats on the six major islands and are morphologically diverse. The populations are referrable to eight of the 12 species recognized by Peterson. Almost all hybrid combinations produced seeds, and seed-set was not significantly different among three categories of hybrids (within population, between populations on the same island, and between populations on different islands). F1s from many of the crosses were vigorous and fertile, leading to the conclusion that these populations are not reproductively isolated from each other and the assumption that they are genetically very similar. Natural hybridization was difficult to detect because the species described by Peterson intergrade morphologically and ecologically. The crossability and interfertility of populations of Wikstroemia may be explained by one or more of the following hypotheses: 1) recency of colonization, 2) the founder effect, 3) lack of selection for barriers to interbreeding, and 4) gross morphological changes without associated large genetic changes.  相似文献

Crosses were made in all combinations of the six diploid species of the Solarium nigrum complex that occur in North America. Some interspecific pollinations failed to yield viable seed; successful crosses gave rise to moderately to highly sterile F1 hybrids. Results of interspecific crosses suggested phylogenetic relationships that were not completely in accord with those suggested by morphology. Interspecific crosses also gave varied results. All interpopulational crosses within S. interius and S. sarachoides produced fully fertile hybrids. In contrast, hybrids within S. americanum and S. douglasii varied from fully fertile to almost completely sterile. Populations of S. pseudogracile could be divided into two groups which are geographically separated but not morphologically differentiated. Fully fertile hybrids resulted from crosses within a group, whereas crosses between groups gave hybrids with reduced fertility. Four crossing groups were observed within 5. nodiflorum; three of the parental groups are sympatric and are morphologically differentiated. Although hybrid sterility in interspecific crosses is sometimes used to support delimitation of species, the presence of sterility in intraspecific crosses suggests that such an interpretation is unwarranted for the S. nigrum complex. Hybrid sterility, therefore, is not considered to have special taxonomic significance in this complex.  相似文献

North American Elymus canadensis L., 2n = 28, and Asian Agropyron libanoticum Hack., 2n = 14, crossed with ease and yielded vigorous but sterile F1 hybrids, 2n = 21. Chromosome pairing in the hybrids averaged 9.47I, 5.38II, and 0.26III in 150 metaphase-I cells. One genome of E. canadensis is more or less homologous with the A. libanoticum genome. Treatment of the F1 hybrids with colchicine produced 42-chromosome amphiploids, C0, which were advanced through two seed generations, C1 and C2. More than half of the metaphase-I cells in the C0 amphiploids contained 21II; and average associations were 1.09I, 20.16II, 0.07III, and 0.09IV in 116 cells. Meiosis became increasingly irregular beyond metaphase-I; nevertheless, the C0 amphiploids produced 68% stainable pollen and averaged 0.75 seed per spikelet. Multivalent frequencies increased in advanced generations, and the C2 amphiploids averaged 1.11I, 19.00II, 0.23III, and 0.55IV in 100 metaphase-I cells. Meiosis was essentially regular in the C1 and C2 amphiploids beyond metaphase I, and the C2 amphiploids averaged 73% stainable pollen and 2.28 seeds per spikelet. The amphiploids have an excellent chance of developing into a meiotically stable, fertile, new species. Forage characteristics of the amphiploids indicate that they have considerable economic potential as a forage grass.  相似文献
