为了从显微结构上进一步探讨虉草(Phalaris arundinacea L.)的抗旱耐涝性及与利用的关系,于2011年采用常规石蜡切片技术,对其根、茎叶3种营养器官进行解剖观察。结果表明,虉草根的结构自外而内依次为表皮、皮层、维管束鞘、初生韧皮部和初生木质部;茎由表皮、基本组织和维管束构成;叶片内部结构可分为表皮、叶肉和叶脉3部分。根皮层大的细胞间隙和气腔,初生木质部的后生大导管和茎基本组织解体形成的髓腔都是虉草良好的通气组织,是其耐水淹的主要显微特征。茎、叶片角质化的表皮和叶表皮所含的丰富泡状细胞组是虉草具有抗旱性的主要解剖结构特征。叶肉细胞排列紧密且只有少量气孔分布于叶片下表皮,这样的结构可减少蒸腾;叶肉细胞富含叶绿体,增强光合作用,获得更多的同化产物,确保了植株在干旱条件下也有足够的光合产物来维持正常的生理活动。茎、叶维管束部分大量的木纤维起到支撑作用。虉草根的皮层和维管柱部分、茎的基本组织和维管束部分、叶的叶脉部分都含有大面积的厚壁细胞,厚壁细胞中含有丰富的粗纤维和木质素。丰富的粗纤维、木质素等成分则是虉草能成为新能源燃料植物的必备条件。  相似文献

采用石蜡切片技术对扇脉杓兰营养器官的解剖结构进行了研究。结果表明:根状茎的薄壁细胞中含丰富的淀粉粒,维管柱中分布着排列紧凑的周木维管束;根的皮层发达,有的皮层细胞中存在真菌菌丝团,木质部与韧皮部呈辐射状相间排列,根和根状茎的内皮层细胞都形成马蹄形加厚结构。茎的表面分布气孔,皮层面积较小,皮层内部的基本组织发达,外韧维管束散生分布其中,茎和叶上都附有非腺性毛;叶为等面叶,叶肉细胞排列疏松,气孔主要分布于远轴面,略外凸,保卫细胞中含有叶绿体,叶缘处的叶肉组织中含有气腔结构。扇脉杓兰营养器官的这些特征与其荫蔽湿润的生境是相适应的。  相似文献

在光学显微镜下,对安徽省贝母属(Fritillaria L.)8种1变种,其中包括安徽贝母(F. anhuiensis)和宁国贝母(F. ningguoensis)各两个居群的植物叶片进行了解剖结构观察,结果发现:叶肉细胞均含有淀粉粒,近上表皮的叶肉细胞多为方形或圆柱状,较大,排列较紧密,近下表皮的叶肉细胞形状不规则,多有分枝,细胞间隙大;叶均无表皮毛等附属物,除铜陵黄花贝母(F. monantha var. tonglingensis)的下表皮细胞类似平行四边形外,其它种叶表皮细胞表面观为长方形或长条形,细胞长轴与叶脉平行,垂周壁为平直、浅波状或深波状;气孔器为Allium型,仅分布于下表皮,椭圆形,长轴与叶脉平行,单个随机分布,部分种保卫细胞两极有“T”形加厚。叶表皮细胞的形状,垂周壁式样,气孔器的类型等特征较为稳定,可以为贝母属内种的划分及系统演化关系的探讨提供实验证据。  相似文献

The morpho-anatomy and histochemistry of the hysteranthous leaf ofUrginea maritima (L.) Baker and its adaptive strategies to the Mediterranean climate were investigated. The leaf ofU. maritima is 714 μm thick and possesses moderate specific leaf mass (8.564 mg cm-2) and low tissue density (136.5 mg cm-3). The epidermal cells are compactly arranged and covered with cuticle. The average density of stomata in lower epidermis is higher than that of the upper one. The mesophyll cells occupy 52.96% of the total volume of the leaf, while the mesophyll intercellular spaces and the air spaces occupy 30.41%. Idioblastic cells containing raphide bundles and different phenotypes of crystalloid inclusions, embedded in polysaccharides, occur in the lower side of the mesophyll. The presence of oil droplets and lipids is evident. Bundle sheath cells are hardly visible with no chloroplasts which are a pronounced C3 plant character. Plastids containing protein crystalloid inclusions are abundant in the protophloem sieve elements.U. maritima, a deciduous plant, possesses leaves with mesophytic characters, in order to optimize its adaptation to the seasonal fluctuation of environmental conditions of the Mediterranean climate.  相似文献

Leaves of the Princeton and a variegated clone of Coleus blumei Benth. were examined with the light microscope to determine the course of their vasculature and the spatial relationship between the mesophyll, bundle sheath, and vascular tissues. In Princeton clone leaves two leaf traces enter the petiole at the node and quickly branch to form an arc of bundles which undergo further divisions as well as fusions in the distal half of the petiole. The anastomosing arc of bundles reaches its greatest complexity in the base of the midvein, where its lateral-most bundles unite and diverge outward to form secondary veins. As the midvein bundles continue acropetally, they gradually fuse more and divide less until only a single bundle remains, from which secondaries and smaller veins branch. Major (ribbed) veins include not only the midvein and secondaries but also tertiary and quaternary veins. Decreasing vein size is accompanied by increasing direct contact between vascular and photosynthetic tissues. Minor veins, which make up 86% of the total vein length, are completely surrounded by photosynthetic bundle sheaths and mesophyll consisting of palisade and spongy parenchyma. Statoliths occur in a layer of cells just outside the phloem of the petiole-midrib axis and secondary veins. Functional hydathodes are present at the apices of the marginal teeth. The overall organization of tissues in variegated leaves differs little in either the green or albuminous areas from corresponding (but always green) regions of Princeton leaves. Chloroplasts are lacking in mesophyll, bundle-sheath, and most guard cells of the albuminous region but are present in guard cells which are within 1 mm of green areas.  相似文献

Huanglian (Coptis chinensis Franch.) is a slow-growing perennial medicinal herb with considerable economic value. This study aimed to determine the structural characteristics and the levels of berberine deposits in the organs and tissues of Huanglian using light and epifluorescence microscopy. The adventitious roots are composed of primary and secondary structures with endodermis, exodermis, and phellem. The rhizome structures are composed of primary and secondary structures with cuticle and phellem. The leaves are composed of sclerenchymatous rings, isolateral mesophyll, and thin cuticles. We detected berberine in the xylem walls of the roots and rhizomes as well as in the sclerenchymatous rings of the petioles. We postulate that as the exodermis is developed, the deposition of berberine in the xylem closest to the root tips may affect water and nutrient absorption and transfer. Leaf blades had a thin cuticle and isolateral mesophyll, suggesting shade tolerance. These structural and histochemical features suggest that Huanglian is adapted to the slow growing nature of a shady environment.  相似文献

Temporal and spatial patterns of palisade mesophyll cell expansion in Zinnia elegans were characterized as a basis for developing a suspension culture model for mesophyll cell expansion. Our objectives were to 1) identify the leaf regions from which cells in various stages of expansion could be selectively isolated for culture, and 2) develop a basis for comparison of rate and extent of mesophyll cell expansion in culture with that in the leaf. Palisade mesophyll cells were isolated from expanding leaves by gentle physical maceration without the use of enzymes. Isolated cells from leaves in different stages of expansion were then measured by computer image analysis. Analysis of size frequency distributions showed that unexpanded cells can be isolated from the entire blade of small leaves or the basal regions of partially expanded leaves. Fully expanded cells can be obtained from the apical and middle regions of partially expanded leaves. Within the leaf, Zinnia mesophyll cells expanded from about 400 μm2 to about 2.300 μm2 at an estimated rate of 160 μm2 d-1. The percent increase in cell length exceeded the percent increase in cell width. Expansion of mesophyll cells continued for 6–8 d after epidermal expansion ceased. This difference in the timing of cell expansion in epidermal and mesophyll cells indicates that different regulatory factors may be operating in these adjacent tissues and underscores the importance of investigating the regulation of mesophyll cell expansion at the cellular level.  相似文献

为了探讨外来植物无瓣海桑的潜在危害,采用石蜡切片法对海桑(Sonneratia caseolaris Engl.)、无瓣海桑(S.apetala B.Ham)叶片进行了解剖学研究。实验结果显示,两种植物的叶片均为等面叶;中脉维管束为周韧维管束;具4级侧脉,第1级侧脉为半周韧维管束;成熟叶片叶肉组织具发达的贮水薄壁细胞,具含单宁成分的薄壁细胞,具晶体细胞和石细胞;盐腺由表皮细胞发育而成,可分为3个发育阶段。作为外来植物的无瓣海桑,其中脉维管束具微弱形成层,叶脉维管组织比海桑更发达;贮藏组织中含单宁细胞、晶体细胞较多;栅栏组织含叶绿体多于海桑等特点,使其比海桑对环境具有更大的适应性。因此,无瓣海桑有可能成为入侵植物。  相似文献

Leaf discs obtained from mature leaves of Xerosicyos danguyi were found to contain appreciable levels of stachyose throughout an 8-h nocturnal period during which this plant performs Crassulacean acid metabolism (CAM). In contrast, in mesophyll tissues obtained from paradermal sections of these same leaf discs and which were devoid of vascular tissues, stachyose pools were rapidly depleted during the nocturnal phase. The pattern of this depletion followed closely the depletion pattern observed for starch, indicating that mesophyll stachyose was possibly involved in nocturnal CAM processes and was not necessarily being used for export. Pulse-labelling of intact X. danguyi leaves prior to excision of leaf discs and mesophyll samples also indicated that, while labelled stachyose had turned-over completely in the mesophyll tissues by the end of the nocturnal period, substantial levels of labelled stachyose were always recovered from the leaf discs from which these mesophyll samples were derived. The data indicate the existence of two separate pools of stachyose in the X. danguyi leaf, one a mesophyll pool which turns over rapidly at night and which may be involved to a small extent in nocturnal CAM processes, and the other a pool associated with and possibly synthesized by the vascular tissues and which presumably represents the phloem-transport pool.Abbreviation CAM Crassulacean acid metabolism This work was supported by National Science Foundation Grant DCB 8901785.  相似文献

Anatomical observations of leaves infected by Taphrina deformans were studied in tolerant peach trees (TPT) and in very susceptible (VSPT) ones. Leaves from the first sampling (2nd April) showed hyphae penetrating through the stomata or into the cuticle of the host tissue; anatomical structures of leaf sections were similar for both TPT and VSPT. The ultrastructure of the leaves of TPT showed seemingly normal mesophyll cells. In contrast, mesophyll cells of the VSPT showed important signs of degradation. Cells were organelle‐free and the middle lamella was expanded and invaded by hyphae of T. deformans. In some samples, the leaves of TPT showed deformed epidermal cells, loss of some spongy cells and increase of the intercellular spaces and division of the palisade cells. The pathogen proliferation in the leaves of the VSPT was considerably superior. In this case, stimulation of cell division occurred in the abaxial epidermis. Cells showed periclinal and oblique divisions, with an increased number of plasmodesmata; palisade or spongy cells were not differentiable. Leaves from TPT collected on 26th April showed hyphae with a non‐cylindrical section and with a squashed aspect. The hyphae were very evident in the intercellular spaces, showing abundant endoplasmic reticulum of rough type (RER) in the cytoplasm. On the other hand, epidermis of the leaves of the VSPT had numerous hyphae under the cuticle, which were growing in a thick pectin matrix. Leaves from TPT and VSPT collected on 6th May showed relevant differences. The leaves of TPT had a palisade mesophyll with fewer cells but with active chloroplasts. In contrast, the leaves from VSPT showed empty mesophyll cells, the cytoplasm was collapsed and the adaxial epidermis was covered with the fungus fructification. The observed anatomical and ultrastructural differences of leaves from TPT and VSPT confirm a different behaviour in plant‐host reaction at early stages of infection.  相似文献

Transgenic tobacco plants were generated carrying a rice homeobox gene,OSH1, controlled by the promoter of a gene encoding a tobacco pathogenesis-related protein (PR1a). These lines were morphologically abnormal, with wrinkled and/or lobed leaves. Histological analysis of shoot apex primordia indicated arrest of lateral leaf blade expansion, often resulting in asymmetric and anisotropic growth of leaf blades. Other notable abnormalities included abnormal or arrested development of leaf lateral veins. Interestingly,OSH1 expression was undetectable in mature leaves with the aberrant morphological features. Thus,OSH1 expression in mature leaves is not necessary for abnormal leaf development. Northern blot and in situ hybridization analyses indicate thatPR1a-OSH1 is expressed only in the shoot apical meristem and in very young leaf primordia. Therefore, the aberrant morphological features are an indirect consequence of ectopicOSH1 gene expression. The only abnormality observed in tissues expressing the transgene was periclinal (rather than anticlinal) division in mesophyll cells during leaf blade initiation. This generates thicker leaf blades and disrupts the mesophyll cell layers, from which vascular tissues differentiate. TheOSH1 product appears to affect the mechanism controlling the orientation of the plane of cell division, resulting in abnormal periclinal division of mesophyll cell, which in turn results in the gross morphological abnormalities observed in the transgenic lines.  相似文献

Agrobacterium 6b oncogenes induce tumours and modify plant growth in various ways. Here we show that the AB-6b gene from strain AB4 placed under 2x35S promoter control (2x35S-AB-6b) induces a complex enation syndrome in transgenic Nicotiana tabacum plants, that also occurs in a few rare cases of genetic enations. In Arabidopsis thaliana, 2x35S-AB-6b induced radially symmetrical tubes on the abaxial side of the leaves, which must therefore be considered as the Arabidopsis equivalents of enations on other plant species. Tobacco and Arabidopsis 2x35S-AB-6b leaves contained small, supernumerary densely packed cells between the spongy mesophyll and the abaxial epidermis, close to vascular strands arising at an early stage of leaf development. On tobacco, the 2x35S-AB-6b enation syndrome could be transmitted across graft junctions to growing tissues of untransformed plants, both acropetally and basipetally. We propose that the AB-6b gene encodes the synthesis of one or more enation factor(s) that are transported by the phloem and modify the growth of developing tissues.  相似文献

Mutants with altered leaf morphology are useful as markers for the study of genetic systems and for probing the leaf differentiation process. One such mutant with deficient greening and altered development of the leaf mesophyll appeared in an inbred line of sunflower (Helianthus annuus L.). The objectives of the present study were to determine the inheritance of the mutant leaf trait and its morphological characterisation. The mutation, named mesophyll cell defective1 (mcd1), has pleiotropic effects and it is inherited as a monogenic recessive. The structure and tissue organization of mcd1 leaves are disrupted. In mcd1 leaves, the mesophyll has prominent intercellular spaces, and palisade and spongy tissues are not properly shaped. The mutant palisade cells also appear to be more vacuolated and with a reduced number of chloroplasts than the wild type leaves of equivalent developmental stage. The lamina thickness of mcd1 leaves is greatly variable and in some areas no mesophyll cells are present between the adaxial and abaxial epidermis. The leaf area of the mcd1 mutant is extremely reduced as well as the stem height. A deficient accumulation of photosynthetic pigments characterizes both cotyledons and leaves of the mutant. In mcd1 leaves, chlorophyll (Chl) fluorescence imaging evidences a spatial heterogeneity of leaf photosynthetic performance. Little black points, which correspond to photosystem II (PSII) maximum efficiency (Fv/Fm) values close to zero, characterize the mcd1 leaves. Similarly, the lightadapted quantum efficiency (ΦPSII) values show a homogeneous distribution over wild type leaf lamina, while the damaged areas in mcd1 leaves, represented by yellow zones, are prominent. In conclusion, the loss of function of the MCD1 gene in Helianthus annuus is correlated with a variegated leaf phenotype characterized by a localized destruction of mesophyll morphogenesis and defeat of PSII activity.  相似文献

Structural aspects of the leaves of two common festucoids,Festuca ovina andPoa sphondylodes, have been examined employing the electron microscopy. The nature of vascular bundles and of sheaths that surround vascular tissues was discussed in the study. The festucoids exhibited a non-Kranz C-3 anatomy with more than four mesophyll cells separating the bundle sheaths of a leaf blade. Vascular tissues in theseFestuca andPoa leaves were surrounded by a double sheath: an inner distinct mestome sheath (MST) and an outer indistinctive layer of parenchymatous bundle sheath (PBS) cells. The PBS cells were much larger than the MST and had thin walls. The MST cells were relatively small and rectangular inP. sphondylodes and more or less hexangular in transverse sections ofF. ovina. InP. sphondylodes, MST had conspicuously thickened inner tangential walls with asymmetrically uninterrupted suberized lamellae in radial and tangential walls. In most differentiated MST cells, all walls were highly suberized. During suberin deposition, MST cells were quite vacuolated and most of the cytoplasm was present as a thin peripheral layer. However, MST walls inF. ovina revealed very thin suberized lamellae with translucent striations. No chloroplasts were detected inP. sphondylodes, whereas the MST inF. ovina contained small chloroplasts. Plasmodesmata were well developed in the primary pit fields of walls between MST and vascular cells, and between adjacent MST cells. Plasmodesmata were less frequent in the walls between the inner and outer sheath cells. Suberized lamellae were totally absent from the PBS cell walls in all veins. External to the PBS, the mesophyll comprised thin walled cells with abundant intercellular spaces. Peripherally arranged chloroplasts in the mesophyll were numerous and often larger than those of PBS and MST cells. Characteristics associated with C-3 and other ultrastructural features were also discussed in the study.  相似文献

Two genomic variants of a chickpea (Cicer arietinum L.) parental line have been developed which exhibit gigas characters. The two genotypes were the result of a single-gene mutation (gigas) and induced tetraploidy of a single parental line. The two genotypes plus parental strain were investigated to determine the similarity-of-effect of polyploidy and this single-gene mutation on leaf anatomy and morphology. Leaves consisted of two rows of alternatively arranged leaflets. Both the tetraploid and parental lines had the same mean number of leaflets per leaf while the gigas plants had fewer, but mean total leaf surface area was greater in the gigas plants. Quantitative comparison of mesophyll and vascular tissue and air space volume density (Vv) showed that leaves of the tetraploid plants had the greatest mesophyll cell density (Vvm) and least air space density. Mesophyll cell density was equal in the parental and single-gene mutant while parental leaves had the greatest vascular tissue density. The greater mesophyll cell density values of the polyploid were due to larger mean mesophyll cell size, not to greater cell numbers per unit area. Leaf models based on tissue density and leaflet size showed tetraploid plants had the greatest productivity potential per unit of leaflet surface area. However, if models were based on a whole leaf, gigas plants had the greatest productivity potential since they had larger total leaf area. The effectiveness of using structural models to predict physiological potential in plant tissues will be tested in future studies.  相似文献

Summary Mesophyll containing the minor veins from leaves ofTetragonia expansa Murr. was examined in preparation for a study of effects of beet yellows virus on the leaf tissues of this plant. The sieve elements throughout the minor veins exhibit the characteristics commonly found in this type of cell in dicotyledons. The cells are connected with one another by sieve plates and with contiguous parenchyma cells by branched plasmodesmata. Mature sieve elements are enucleate and lack ribosomes. No tonoplast was discerned in these cells. Mitochondria, plastids, and sparse endoplasmic reticulum are retained in mature cells. The plastids, which contain a massive fibrous ring of proteinaceous material, resemble the sieve element plastids ofBeta. The P-protein in the sieve elements is occasionally composed of tubules; more commonly it is represented by loose helices. The tracheary elements have scalariform secondary thickenings. In regions free of these thickenings, the primary wall largely disintegrates; only some loosely arranged fibrils remain. The mesophyll and vascular parenchyma cells contain the various organelles characteristic of living plant cells but vary in degree of vacuolation and in density of cytoplasm. Some vascular parenchyma cells have particularly dense protoplasts. They contain numerous ribosomes and their background matrix consists of a dense population of fine fibrils. The relative importance that biomechanical and biochemical leaf traits have on photosynthetic capacity would depend on a complex interaction of internal architecture and physiological differences. Changes in photosynthetic capacity on a leaf area basis and anatomical properties during leaf development were studied in a deciduous tree, Prunus persica, and an evergreen shrub, Olea europaea. Photosynthetic capacity increased as leaves approached full expansion. Internal CO2 transfer conductance (g i) correlated with photosynthetic capacity, although, differences between species were only partially explained through structural and anatomical traits of leaves. Expanding leaves preserved a close functional balance in the allocation of resources of photosynthetic component processes. Stomata developed more rapidly in olive than in peach. Mesophyll thickness doubled from initial through final stages of development when it was twice as thick in olive as in peach. The surface area of mesophyll cells exposed to intercellular air spaces per unit leaf area tended to decrease with increasing leaf expansion, whereas, the fraction of mesophyll volume occupied by the intercellular air spaces increased strongly. In the sclerophyllous olive, structural protection of mesophyll cells had priority over efficiency of photochemical mechanisms with respect to the broad-leaved peach. The photosynthetic capacity of these woody plants during leaf development relied greatly on mesophyll properties, more than on leaf mass per area ratio (LMA) or nitrogen (N) allocation. Age-dependent changes in diffusion conductance and photosynthetic capacity affected photosynthetic relationships of peach versus olive foliage, evergreen leaves maturing functionally and structurally a bit earlier than deciduous leaves in the course of adaptation for xeromorphy.  相似文献

Brassavola inhabits a wide altitude range and habitat types from Northern Mexico to Northern Argentina. Classification schemes in plants have normally used vegetative and floral characters, but when species are very similar, as in this genus, conflicts arise in species delimitation, and alternative methods should be applied. In this study we explored the taxonomic and phylogenetic value of the anatomical structure of leaves in Brassavola; as ingroup, seven species of Brassavola were considered, and as an outgroup Guarianthe skinneri, Laelia anceps, Rhyncholaelia digbyana and Rhyncholaelia glauca were evaluated. Leaf anatomical characters were studied in freehand cross sections of the middle portion with a light microscope. Ten vegetative anatomical characters were selected and coded for the phylogenetic analysis. Phylogenetic reconstruction was carried out under maximum parsimony using the program NONA through WinClada. Overall, Brassavola species reveal a wide variety of anatomical characters, many of them associated with xeromorphic plants: thick cuticle, hypodermis and cells of the mesophyll with spiral thickenings in the secondary wall. Moreover, mesophyll is either homogeneous or heterogeneous, often with extravascular bundles of fibers near the epidermis at both terete and flat leaves. All vascular bundles are collateral, arranged in more than one row in the mesophyll. The phylogenetic analysis did not resolve internal relationships of the genus; we obtained a polytomy, indicating that the anatomical characters by themselves have little phylogenetic value in Brassavola. We concluded that few anatomical characters are phylogenetically important; however, they would provide more support to elucidate the phylogenetic relantionships in the Orchidaceae and other plant groups if they are used in conjunction with morphological and/or molecular characters.  相似文献

Although the unique tissue required for C4 photosynthesis in nonsucculent plants is often described as being modified leaf parenchyma sheath, which is positioned meaningfully between the mesophyll externally and the vascular tissues internally, the actual range of locations and known associations make that concept untenable. In origin the Kranz tissue develops from procambium as well as ground parenchyma. It is found in stems as well as leaves. In position Kranz tissue can lie in the parenchyma sheath, in the mestome sheath, isolated in the mesophyll, peripherally in some thick leaves, or within the veins. It can be associated with mesophyll only, mesophyll and colorless parenchyma, mesophyll and sclerenchyma, other Kranz tissue and vascular tissues, mesophyll and mestome sheath, mesophyll and phloem, mesophyll and xylem, epidermis, and, finally, mestome sheath and xylem and phloem. 用石蜡切片和扫描电镜对墨兰试管苗植株的生长发育进行了研究。发现幼叶中脉附近的叶肉细胞类似栅栏组织,随着叶片的不断成熟,叶片基部中脉附近的叶肉细胞逐渐变为近圆形或椭圆形,而叶尖部和叶中部中脉附近的叶肉细胞仍似栅栏组织。茎的发育经历了原球茎、根状茎和假鳞茎3个阶段。原球茎的大部分细胞都含有淀粉粒,根状茎的皮层细胞含淀粉粒,而假鳞茎几乎不含淀粉粒。幼根没有髓,皮层细胞含淀粉粒:成熟根具含淀粉粒的髓。出瓶苗上即带有4个芽,一般只有最外侧叶腋的1个花芽和最内侧叶腋的1个叶芽发育。  相似文献
