The physical structure and activity of aerobic granules, and the succession of bacterial community within aerobic granules under constant operational conditions and shock loading were investigated in one sequencing batch reactor over ten months. While the maturation phase of the granulation process began on day 30, the structure of microbial community changed markedly until after three months of reactor operation under constant conditions with a loading rate of 1.5 g phenol L−1 day−1. A shock loading of 6.0 g phenol L−1 day−1 from days 182–192 led to divergence of bacterial community, an inhibition of the biomass activity, and a decrease in phenol removal rate in the reactor. However, phenol was still completely removed under this disturbance. After the shock loading, the mean sizes of aerobic granules increased, and the activity of the microbial population within the granules decreased, although there appeared highly resilient for the dominant bacterial community of aerobic granules which mainly included β-Proteobacteria. Correlation analysis suggested that biomass concentration and biomass loading were significantly related to the community composition of aerobic granules during the whole operational period. The development of a relatively stable bacterial community in aerobic granules implied that those distinct dominant microbes in aerobic granules were favorably selected and proliferated under the operational conditions.  相似文献

Aerobic sludge granules are compact, strong microbial aggregates that have excellent settling ability and capability to efficiently treat high-strength and toxic wastewaters. Aerobic granules disintegrate under high organic loading rates (OLR). This study cultivated aerobic granules using acetate as the sole carbon and energy source in three identical sequencing batch reactors operated under OLR of 9–21.3 kg chemical oxygen demand (COD) m−3 day−1. The cultivated granules removed 94–96% of fed COD at OLR up to 9–19.5 kg COD m−3 day−1, and disintegrated at OLR of 21.3 kg COD m−3 day−1. Most tested isolates did not grow in the medium at >3,000 mg COD l−1; additionally, these strains lost capability for auto-aggregation and protein or polysaccharide productivity. This critical COD regime correlates strongly with the OLR range in which granules started disintegrating. Reduced protein quantity secreted by isolates was associated with the noted poor granule integrity under high OLR. This work identified a potential cause of biological nature for aerobic granules breakdown.  相似文献

Waste rice straw (RS) is generated in massive quantities around the world and is often burned, creating greenhouse gas and air quality problems. Anaerobic digestion (AD) may be a better option for RS management, but RS is presumed to be comparatively refractory under anaerobic conditions without pre-treatment or co-substrates. However, this presumption assumes frequent reactor feeding regimes but less frequent feeding may be better for RS due to slow hydrolysis rates. Here, we assess how feeding frequency (FF) and organic loading rate (OLR) impacts microbial communities and biogas production in RS AD reactors. Using 16S rDNA amplicon sequencing and bioinformatics, microbial communities from five bench-scale bioreactors were characterized. At low OLR (1.0 g VS l−1 day−1), infrequently fed units (once every 21 days) had higher specific biogas yields than more frequent feeding (five in 7 days), although microbial community diversities were statistically similar (P > 0.05; ANOVA with Tukey comparison). In contrast, an increase in OLR to 2.0 g VS l−1 day−1 significantly changed Archaeal and fermenting Eubacterial sub-communities and the least frequency fed reactors failed. ‘Stable’ reactors were dominated by Methanobacterium, Methanosarcina and diverse Bacteroidetes, whereas ‘failed’ reactors saw shifts towards Clostridia and Christensenellaceae among fermenters and reduced methanogen abundances. Overall, OLR impacted RS AD microbial communities more than FF. However, combining infrequent feeding and lower OLRs may be better for RS AD because of higher specific yields.  相似文献

The bacterial communities in three continuously stirred tank reactors co-digesting cow manure with grass silage, oat straw, and sugar beet tops, respectively, were investigated by 16S rRNA gene-based fingerprints and clone libraries. The analyses revealed both clearly distinct and similar phylotypes in the bacterial communities between the reactors. The major groups represented in the three reactors were Clostridia, unclassified Bacteria, and Bacteroidetes. Phylotypes affiliated with Bacilli or Deltaproteobacteria were unique to the sugar beet and straw reactor, respectively. Unclassified Bacteria dominated in sugar beet reactor while in the straw and grass reactor Clostridia was the dominant group. An increase in organic loading rate from 2 to 3 kg volatile solids m?3 d?1 resulted in larger changes in the bacterial community in the straw compared to grass reactor. The study shed more light on the evolution of bacterial community during anaerobic co-digestion of different crops and manure to methane.  相似文献

Herein batch and continuous mesophilic anaerobic digestion of grass silage liquor was studied. The continuous process was carried out in Armfield digesters with an OLR ranging from 0.851 to 1.77 kg COD m−3 day−1. The effect of recirculation of effluent from the digester was investigated using different OLRs of grass silage liquor feed. These results showed that as the OLR increased, the methane yield decreased for the reactor with no recycle and increased for the reactor with recycle. However, the COD removal for both digesters was nearly the same at the same OLR. Overall these studies show that grass silage liquor can produce a high quality methane steam between 70% and 80% and achieve methane yields of 0.385 m3 kg−1 COD.  相似文献

The processes driving primary productivity and its impacts on fish production were investigated in field trials in eight seasonal earthen wetland ponds ‘Fingerponds’ (192 m2) in Uganda between 2003 and 2005. The ponds were stocked by the seasonal flood with predominantly Oreochromis spp. at densities ranging from 0.1 to 0.5 fish m−2. Chicken manure (521, 833 or 1,563 kg ha−1) was applied fortnightly. Results showed that primary productivity was enhanced with maximum average net primary productivity (±Standard Error) of 11.7 (±2.5) g O2 m−2 day−1 at the Gaba site and 8.3 (±1.5) g O2 m−2 day−1 at the Walukuba site. Net fish yields were higher in manured ponds with up to 2,670 kg ha−1 yield for a 310 day growth period compared to less than 700 kg ha−1 in unmanured ponds. Fish production was limited mainly by high recruitment, falling water levels, light limitation from high suspended solids and turbidity, and low zooplankton biomass. It was concluded that Fingerponds have a high potential for sustainable fish production and can contribute to the alleviation of protein shortages amongst the riparian communities around Lake Victoria. Production can be enhanced further with improved stock management.  相似文献

An alternative to land spreading of manure effluents is to mass-culture algae on the N and P present in the manure and convert manure N and P into algal biomass. The objective of this study was to determine how the fatty acid (FA) content and composition of algae respond to changes in the type of manure, manure loading rate, and to whether the algae was grown with supplemental carbon dioxide. Algal biomass was harvested weekly from indoor laboratory-scale algal turf scrubber (ATS) units using different loading rates of raw and anaerobically digested dairy manure effluents and raw swine manure effluent. Manure loading rates corresponded to N loading rates of 0.2 to 1.3 g TN m−2 day−1 for raw swine manure effluent and 0.3 to 2.3 g TN m−2 day−1 for dairy manure effluents. In addition, algal biomass was harvested from outdoor pilot-scale ATS units using different loading rates of raw and anaerobically digested dairy manure effluents. Both indoor and outdoor units were dominated by Rhizoclonium sp. FA content values of the algal biomass ranged from 0.6 to 1.5% of dry weight and showed no consistent relationship to loading rate, type of manure, or to whether supplemental carbon dioxide was added to the systems. FA composition was remarkably consistent among samples and >90% of the FA content consisted of 14:0, 16:0, 16:1ω7, 16:1ω9, 18:0, 18:1ω9, 18:2 ω6, and 18:3ω3.  相似文献

This work conducted a denitrifying sulfide removal (DSR) test in an expanded granular sludge bed (EGSB) reactor at sustainable loadings of 6.09 kg m−3 day−1 for sulfide, 3.11 kg m−3 day−1 for nitrate–nitrogen, and 3.27 kg m−1 day−1 for acetate–carbon with >93% efficiency, which is significantly higher than those reported in literature. Strains Pseudomonas sp., Nitrincola sp., and Azoarcus sp. very likely yield heterotrophs. Strains Thermothrix sp. and Sulfurovum sp. are the autotrophs required for the proposed high-rate EGSB-DSR system. The EGSB-DSR reactor experienced two biological breakdowns, one at loadings of 4.87, 2.13, and 1.82 kg m−3 day−1; reactor function was restored by increasing nitrate and acetate loadings. Another breakdown occurred at loadings of up to 8.00, 4.08, and 4.50 kg m−1 day−1; the heterotrophic denitrification pathway declined faster than the autotrophic pathway. The mechanism of DSR breakdown is as follows. High sulfide concentration inhibits heterotrophic denitrifiers, and the system therefore accumulates nitrite. Autotrophic denitrifiers are then inhibited by the accumulated nitrite, thereby leading to breakdown of the DSR process.  相似文献

Efficient nitrification and denitrification of wastewater containing 1,700 mgl−1 of ammonium-nitrogen was achieved using aerobic granular sludge cultivated at medium-to-high organic loading rates. The cultivated granules were tested in a sequencing batch reactor (SBR) fed with 6.4 or 10.2 kg NH4+-N m−3 day−1, a loading significantly higher than that reported in literature. With alternating 2 h oxic and 2 h anoxic operation (OA) modes, removal rate was 45.5 mg NH4+-N g−1 volatile suspended solids−1 h−1 at 6.4 kg NH4+-N m−3 day−1 loading and 41.3 ± 2.0 at 10.2 kg NH4+-N m−3 day−1 loading. Following the 60 days SBR test, granules were intact. The fluorescence in situ hybridization and confocal laser scanning microscopy results indicate that the SBR-OA granules have a distribution with nitrifers outside and heterotrophs outside that can effectively expose functional strains to surrounding substrates at high concentrations with minimal mass transfer limit. This microbial alignment combined with the smooth granule surface achieved nitrification–denitrification of wastewaters containing high-strength ammonium using aerobic granules. Conversely, the SBR continuous aeration mode yielded a distribution with nitrifers outside and heterotrophs inside with an unsatisfactory denitrification rate and floating granules as gas likely accumulated deep in the granules.  相似文献

The activated sludge process (ASP) has high operational costs due to the need for aeration at dissolved O2 (DO) levels of ≥2 mg l−1 and high capital costs to construct large reactors due to a low organic loading [typically 1 kg chemical oxygen demand (COD) m−3 day−1]. A novel method for improving the energy use and treatment efficiency of the ASP via limited oxygenation (0.4 mg DO l−1) and high organic loading (6.2 kg COD m−3 day−1) is proposed based on a laboratory-scale ASP for ammonia-rich industrial wastewaters. The sludge blanket phenomenon and granulation occurred simultaneously in the upflow microaerobic reactor.  相似文献

In this study, enumeration and identification of total aerobic heterotrophic bacteria and petroleum-utilizing bacteria as well as the degradative potential of petroleum-utilizing bacterial isolates were carried out. The average counts of total aerobic heterotrophic bacteria in cow dung and poultry manure were 74.25 × 105 c.f.u. g−1 and 138.75 × 105 c.f.u. g−1 respectively. Acinetobacter sp, Bacillus sp, Pseudomonas sp, and Serratia spp. occurred as aerobic heterotrophs in both cow dung and poultry manure. However, Alcaligenes spp. occurred only in cow dung while, Flavobacterium sp, Klebsiella sp, Micrococcus sp, and Nocardia spp. occurred only in poultry manure as aerobic heterotrophs. The average counts of petroleum-utilizing bacteria in cow dung and poultry manure were 9.25 × 105 c.f.u. g−1 and 17.25 × 105 c.f.u. g−1 respectively. Pseudomonas spp. occurred as petroleum utilizer in both cow dung and poultry manure. However, Bacillus spp. occurred only in cow dung while Acinetobacter spp. and Micrococcus spp. occurred only in poultry manure as petroleum utilizers. Relative abundance of petroleum utilizers in total aerobic heterotrophs ranged from 6.38% to 20.00% for cow dung and from 9.38% to 17.29% for poultry manure. Introduction of pure cultures of petroleum-utilizing bacteria from cow dung and poultry manure into sterile oil-polluted soil revealed oil degradation in one week period.  相似文献

The microbial ecology associated with siliceous sinters was studied in five geochemically diverse Icelandic geothermal systems. Bacterial 16S rRNA clone libraries were constructed from water-saturated precipitates from each site resulting in a total of 342 bacterial clone sequences and 43 species level phylotypes. In near-neutral, saline (2.6–4.7% salinity) geothermal waters where sinter growth varied between 10 and ~300 kg year−1 m−2, 16S rRNA gene analyses revealed very low (no OTUs could be detected) to medium (9 OTUs) microbial activity. The most dominant phylotypes found in these waters belong to marine genera of the Proteobacteria. In contrast, in alkaline (pH = 9–10), meteoric geothermal waters with temperature = 66–96°C and <1–20 kg year−1m−2 sinter growth, extensive biofilms (a total of 34 OTUs) were observed within the waters and these were dominated by members of the class Aquificae (mostly related to Thermocrinis), Deinococci (Thermus species) as well as Proteobacteria. The observed phylogenetic diversity (i.e., number and composition of detected OTUs) is argued to be related to the physico-chemical regime prevalent in the studied geothermal waters; alkaliphilic thermophilic microbial communities with phylotypes related to heterotrophic and autotrophic microorganisms developed in alkaline high temperature waters, whereas halophilic mesophilic communities dominated coastal geothermal waters.  相似文献

It is possible to cultivate aerobic granular sludge at a low organic loading rate and organics-to-total nitrogen (COD/N) ratio in wastewater in the reactor with typical geometry (height/diameter = 2.1, superficial air velocity = 6 mm/s). The noted nitrification efficiency was very high (99%). At the highest applied ammonia load (0.3 ± 0.002 mg NH4+–N g total suspended solids (TSS)−1 day−1, COD/N = 1), the dominating oxidized form of nitrogen was nitrite. Despite a constant aeration in the reactor, denitrification occurred in the structure of granules. Applied molecular techniques allowed the changes in the ammonia-oxidizing bacteria (AOB) community in granular sludge to be tracked. The major factor influencing AOB number and species composition was ammonia load. At the ammonia load of 0.3 ± 0.002 mg NH4+–N g TSS−1 day−1, a highly diverse AOB community covering bacteria belonging to both the Nitrosospira and Nitrosomonas genera accounted for ca. 40% of the total bacteria in the biomass.  相似文献

The biomass of industrially grown Phaeodactylum tricornutum was subjected in a novel way to bio-methanation at 33°C, i.e., in an anaerobic membrane bioreactor (AnMBR) at a hydraulic retention time of 2.5 days, at solid retention times of 20 to 10 days and at loading rates in the range of 2.6–5.9 g biomass-COD L−1 day−1 with membrane fluxes ranging from 1 to 0.8 L m−2 h−1. The total COD recovered as biogas was in the order of 52%. The input suspension was converted to a clear effluent rich in total ammonium nitrogen (546 mg TAN L−1) and phosphate (141 mg PO4-P L−1) usable as liquid fertilizer. The microbial community richness, dynamics, and organization in the reactor were interpreted using the microbial resource management approach. The AnMBR communities were found to be moderate in species richness and low in dynamics and community organization relative to UASB and conventional CSTR sludges. Quantitative polymerase chain reaction analysis revealed that Methanosaeta sp. was the dominant acetoclastic methanogen species followed by Methanosarcina sp. This work demonstrated that the use of AnMBR for the digestion of algal biomass is possible. The fact that some 50% of the organic matter is not liquefied means that the algal particulates in the digestate constitute a considerable fraction which should be valorized properly, for instance as slow release organic fertilizer. Overall, 1 kg of algae dry matter (DM) could be valorized in the form of biogas (€2.07), N and P in the effluent (€0.02) and N and P in the digestate (€0.04), thus totaling about €2.13 per kilogram algae DM.  相似文献

The performance of a laboratory-scale sewage treatment system composed of an up-flow anaerobic sludge blanket (UASB) reactor and a moving bed biofilm reactor (MBBR) at a temperature of (22–35 °C) was evaluated. The entire treatment system was operated at different hydraulic retention times (HRT’s) of 13.3, 10 and 5.0 h. An overall reduction of 80–86% for CODtotal; 51–73% for CODcolloidal and 20–55% for CODsoluble was found at a total HRT of 5–10 h, respectively. By prolonging the HRT to 13.3 h, the removal efficiencies of CODtotal, CODcolloidal and CODsoluble increased up to 92, 89 and 80%, respectively. However, the removal efficiency of CODsuspended in the combined system remained unaffected when increasing the total HRT from 5 to 10 h and from 10 to 13.3 h. This indicates that, the removal of CODsuspended was independent on the imposed HRT. Ammonia-nitrogen removal in MBBR treating UASB reactor effluent was significantly influenced by organic loading rate (OLR). 62% of ammonia was eliminated at OLR of 4.6 g COD m−2 day−1. The removal efficiency was decreased by a value of 34 and 43% at a higher OLR’s of 7.4 and 17.8 g COD m−2 day−1, respectively. The mean overall residual counts of faecal coliform in the final effluent were 8.9 × 104 MPN per 100 ml at a HRT of 13.3 h, 4.9 × 105 MPN per 100 ml at a HRT of 10 h and 9.4 × 105 MPN per 100 ml at a HRT of 5.0 h, corresponding to overall log10 reduction of 2.3, 1.4 and 0.7, respectively. The discharged sludge from UASB–MBBR exerts an excellent settling property. Moreover, the mean value of the net sludge yield was only 6% in UASB reactor and 7% in the MBBR of the total influent COD at a total HRT of 13.3 h. Accordingly, the use of the combined UASB–MBBR system for sewage treatment is recommended at a total HRT of 13.3 h.  相似文献

Air-side stripping without a prior solid–liquid phase separation step is a feasible and promising process to control ammonia concentration in thermophilic digesters. During the process, part of the anaerobic biomass is exposed to high temperature, high pH and aerobic conditions. However, there are no studies assessing the effects of those harsh conditions on the microbial communities of thermophilic digesters. To fill this knowledge gap, the microbiomes of two thermophilic digesters (55°C), fed with a mixture of pig manure and nitrogen-rich co-substrates, were investigated under different organic loading rates (OLR: 1.1–5.2 g COD l−1 day−1), ammonia concentrations (0.2–1.5 g free ammonia nitrogen l−1) and stripping frequencies (3–5 times per week). The bacterial communities were dominated by Firmicutes and Bacteroidetes phyla, while the predominant methanogens were Methanosarcina sp archaea. Increasing co-substrate fraction, OLR and free ammonia nitrogen (FAN) favoured the presence of genera Ruminiclostridium, Clostridium and Tepidimicrobium and of hydrogenotrophic methanogens, mainly Methanoculleus archaea. The data indicated that the use of air-side stripping did not adversely affect thermophilic microbial communities, but indirectly modulated them by controlling FAN concentrations in the digester. These results demonstrate the viability at microbial community level of air side-stream stripping process as an adequate technology for the ammonia control during anaerobic co-digestion of nitrogen-rich substrates.  相似文献

A full-scale jet biogas internal loop anaerobic fluidized bed (JBILAFB) reactor, which requires low energy input and allows enhanced mass transfer, was constructed for the treatment of food processing wastewater. This reactor has an active volume of 798 m3 and can treat 33.3 m3 wastewater per hour. After pre-treating the raw wastewater by settling, oil separating and coagulation-air floating processes, the reactor was operated with a relatively shorter start-up time (55 days). Samples for the influent and effluent of the JBILAFB reactor were taken and analyzed daily for the whole process including both the start-up and stable running periods. When the volumetric COD loading fluctuated in the range of 1.6–5.6 kg COD m−3 day−1, the COD removal efficiency, the volatile fatty acid(VFA)/alkalinity ratio, the maximum biogas production and the content of CH4 in total biogas of the reactor were found to be 80.1 ± 5%, 0.2–0.5, 348.5 mday−1 and 94.5 ± 2.5%, respectively. Furthermore, the scanning electron microscope (SEM) results showed that anaerobic granular sludge and microorganism particles with biofilm coexisted in the reactor, and that the bacteria mainly in bacilli and cocci were observed as predominant species. All the data demonstrated that the enhanced mass transfer for gas, liquid and solid phases was achieved, and that the formation of microorganism granules and the removal of inhibitors increased the stability of the system.  相似文献

Changes of microbial characteristics in a full-scale submerged membrane bioreactor system (capacity, 60,000 m3 day−1) treating sewage were monitored over the start-up period (96 days). Fluorescence in situ hybridization analysis showed that the percentages of ammonia-oxidizing bacteria (AOB) and nitrite-oxidizing bacteria (nitrobacter-related population) in total bacteria counted with DAPI staining increased significantly from 1.9% and 0.9% to 4.5% and 2.8%, corresponding to an increase of the specific ammonium oxidizing rate (from 0.06 to 0.12 kg N kg−1 mixed liquor suspended solids (MLSS) per day) and the specific nitrate forming rate (from 0.05 to 0.10 kg N kg−1 MLSS day−1). Both the denaturing gradient gel electrophoresis of polymerase chain reaction and clone library results showed that the AOB was dominated by the genus Nitrosomonas, the diversity of which increased markedly with operational time. Most of the day 2 clones were closely related with the uncultured Nitrosomonas sp. clone Ninesprings-49S amoA gene (AY356450.1) originated from activated sludge, while the day 96 clone library showed a more diverse distribution characterized by the appearance of the oligotrophic nitrifiers like the Nitrosomonas oligotropha- and Nitrosomonas ureae-like bacteria, perhaps due to the interception by membrane and the low food-to-microorganisms ratio environment. The above results show that the membrane bioreactor system was characterized by the increased diversity and percentage of nitrifiers, which made it possible to achieve a stable and high efficient nitrification. Ammonia-oxidizing archaea with the changing population structures were also detected, but their roles for ammonia oxidation in the system need further studies.  相似文献

Biodegradation, kinetics, and microbial diversity of aerobic granules were investigated under a high range of organic loading rate 6.0 to 12.0 kg chemical oxygen demand (COD) m−3 day−1 in a sequencing batch reactor. The selection and enriching of different bacterial species under different organic loading rates had an important effect on the characteristics and performance of the mature aerobic granules and caused the difference on granular biodegradation and kinetic behaviors. Good granular characteristics and performance were presented at steady state under various organic loading rates. Larger and denser aerobic granules were developed and stabilized at relatively higher organic loading rates with decreased bioactivity in terms of specific oxygen utilization rate and specific growth rate (μ overall) or solid retention time. The decrease of bioactivity was helpful to maintain granule stability under high organic loading rates and improve reactor operation. The corresponding biokinetic coefficients of endogenous decay rate (k d), observed yield (Y obs), and theoretical yield (Y) were measured and calculated in this study. As the increase of organic loading rate, a decreased net sludge production (Y obs) is associated with an increased solid retention time, while k d and Y changed insignificantly and can be regarded as constants under different organic loading rates.  相似文献
