The presence of strong sinks of photoassimilates is thought to stimulate photosynthesis by minimizing photosynthetic end product accumulation in leaves. This hypothesis was examined in soybeans (Glycine max [L] Merr.) with treatments designed to alter the phloem translocation of photoassimilates out of source leaves. Pod removal and petiole girdling resulted in 70 and 90% reductions, respectively, in leaf CO2 exchange rate. Reductions of similar magnitude also were observed in stomatal diffusive conductivity.  相似文献

BACKGROUND AND AIMS: In crops other than sugarcane there is good evidence that the size and activity of carbon sinks influence source activity via sugar-related regulation of the enzymes of photosynthesis, an effect that is partly mediated through coarse regulation of gene expression. METHODS: In the current study, leaf shading treatments were used to perturb the source-sink balance in 12-month-old Saccharum spp. hybrid 'N19' (N19) by restricting source activity to a single mature leaf. Changes in leaf photosynthetic gas exchange variables and leaf and culm sugar concentrations were subsequently measured over a 14 d period. In addition, the changes in leaf gene response to the source-sink perturbation were measured by reverse northern hybridization analysis of an array of 128 expressed sequence tags (ESTs) related to photosynthetic and carbohydrate metabolism. KEY RESULTS: Sucrose concentrations in immature culm tissue declined significantly over the duration of the shading treatment, while a 57 and 88% increase in the assimilation rate (A) and electron transport rate (ETR), respectively, was observed in the source leaf. Several genes (27) in the leaf displayed a >2-fold change in expression level, including the upregulation of several genes associated with C(4) photosynthesis, mitochondrial metabolism and sugar transport. Changes in gene expression levels of several genes, including Rubisco (EC 4.1.1.39) and hexokinase (HXK; EC 2.7.1.1), correlated with changes in photosynthesis and tissue sugar concentrations that occurred subsequent to the source-sink perturbation. CONCLUSIONS: These results are consistent with the notion that sink demand may limit source activity through a kinase-mediated sugar signalling mechanism that correlates to a decrease in source hexose concentrations, which, in turn, correlate with increased expression of genes involved in photosynthesis and metabolite transport. The signal feedback system reporting sink sufficiency and regulating source activity may be a potentially valuable target for future genetic manipulation to increase sugarcane sucrose yield.  相似文献

Bean plants, trimmed to a simplified “double source, double sink” translocation system (the paired primary leaves serving as the double source and the paired lateral leaflets of the immature first trifoliate leaf as the double sink) were used to study the magnitude and short-term time course of change in the allocation ratio (partition ratio) of assimilates translocated from the labeled primary leaf to its respective “near” and “far leaflet” sinks in response to an increase or decrease in the source strength of the opposite primary leaf (the “control” leaf). If the rates of net photosynthesis in the two primary leaves were similar, assimilates from the labeled source leaf partitioned to the leaflet sinks in the ratio of 5:1 or higher, the dominant sink being the leaflet “nearer” to the labeled source leaf. If the rate of net photosynthesis in the control leaf was increased substantially above that of the labeled source leaf, the rate of translocation from the labeled source to either the near leaflet sink or far leaflet sink remained unaffected, despite, presumably, a higher translocation rate from the control leaf, and hence a higher phloem pressure gradient (or increased cross-sectional area) in the transport pathway from the control leaf to the leaflet sinks. If the control leaf was excised, thus reducing the source leaf area by about a half, the translocation rate from the remaining source leaf rapidly doubled, the partition ratio becoming equal to unity. If the control leaf was darkened, the partition ratio adjusted to an intermediate value. Although export rates from the labeled source leaf were increased either by excising or darkening the control leaf, the rate of net photosynthesis in the labeled leaf remained constant.  相似文献

The reduction of photosynthetic capacity in many plants grown at elevated CO2 is thought to result from a feedback effect of leaf carbohydrates on gene expression. Carbohydrate feedback at elevated CO2 could result from limitations on carbohydrate utilization at many different points, for example export of triose phosphates from the chloroplast, sucrose synthesis and phloem loading, transport in the phloem, unloading of the phloem at the sinks, or utilization for growth of sinks. To determine the relative importance of leaf versus whole plant level limitations on carbohydrate utilization at elevated CO2, and the possible effects on the regulation of photosynthetic capacity, we constructed a treatment system in which we could expose single, attached, soybean leaflets to CO2 concentrations different from those experienced by the rest of the plant. The single leaflet treatments had dramatic effects on the carbohydrate contents of the treated leaflets. However, photosynthetic capacity and rubisco content were unaffected by the individual leaflet treatment and instead were related to the whole plant CO2 environment, despite the fact that the CO2 environment around the rest of the plant had no significant affect on the total non-structural carbohydrate (TNC) contents of the treated leaflets. These results necessitate a re-evaluation of the response mechanisms to CO2 as well as some of the methods used to test these responses. We propose mechanisms by which sink strength could influence leaf physiology independently of changes in carbohydrate accumulation.  相似文献

In citrus, the occurrence of a sink effect on photosynthesis (A) is controversial. Leaf carbohydrates and photosynthetic rates in field-grown trees of Satsuma mandarin ( Citrus unshiu [Mak.] Marc.) cv. Okitsu, were measured to elucidate whether or not the demand for photoassimilates regulates A. The data indicated that the source-sink imbalances induced by different treatments altered both soluble (sucrose, glucose and fructose) and insoluble carbohydrates in leaves, as well as photosynthetic rates. In general, girdling and defruiting increased starch and reduced photosynthesis, whereas source-limiting conditions imposed through partial defoliations had the opposite effect. These results are compatible with the assumption that a lack of sink activity leads to carbohydrate accumulation and feedback inhibition of A, and vice versa. Further evidence supporting a source-sink effect on A was provided by measurements of the dry matter:leaf area ratio, since defoliations, for example, increased this ratio. The in vivo sucrose supplementation to plants with different source:sink ratios (control, defoliated, girdled and defruited plants) increased carbohydrates and reduced photosynthesis. This suggests that sugars may have, per se, the potential to repress photosynthetic rates in intact plants with active sinks. Based on these results we propose that sugar accumulation in citrus leaves causes a feedback inhibition of A.  相似文献

Seed development largely depends on the long‐distance transport of sucrose from photosynthetically active source leaves to seed sinks. This source‐to‐sink carbon allocation occurs in the phloem and requires the loading of sucrose into the leaf phloem and, at the sink end, its import into the growing embryo. Both tasks are achieved through the function of SUT sucrose transporters. In this study, we used vegetable peas (Pisum sativum L.), harvested for human consumption as immature seeds, as our model crop and simultaneously overexpressed the endogenous SUT1 transporter in the leaf phloem and in cotyledon epidermal cells where import into the embryo occurs. Using this ‘Push‐and‐Pull’ approach, the transgenic SUT1 plants displayed increased sucrose phloem loading and carbon movement from source to sink causing higher sucrose levels in developing pea seeds. The enhanced sucrose partitioning further led to improved photosynthesis rates, increased leaf nitrogen assimilation, and enhanced source‐to‐sink transport of amino acids. Embryo loading with amino acids was also increased in SUT1‐overexpressors resulting in higher protein levels in immature seeds. Further, transgenic plants grown until desiccation produced more seed protein and starch, as well as higher seed yields than the wild‐type plants. Together, the results demonstrate that the SUT1‐overexpressing plants with enhanced sucrose allocation to sinks adjust leaf carbon and nitrogen metabolism, and amino acid partitioning in order to accommodate the increased assimilate demand of growing seeds. We further provide evidence that the combined Pushand‐Pull approach for enhancing carbon transport is a successful strategy for improving seed yields and nutritional quality in legumes.  相似文献

Despite the large amount of data regarding sucrose-binding proteins (SBP), their functions remain largely unknown and controversial. In this investigation we performed a detailed temporal and spatial characterization of the phenotypes related to photosynthesis, sucrose exudation and carbohydrate metabolism in SBP antisense plants to gain insights into the physiological role of SBP. Significant reductions in net photosynthesis and in stomatal conductance were observed in the SBP antisense lines but were restricted to the vegetative phase, and persisted during a daily time course at this phase. Photosynthesis was saturated at a substantially lower irradiance in source leaves of the antisense lines, suggesting that light utilization is decreased in these plants. A slight reduction in soluble sugars was observed throughout the development of source leaves, partially overlapping a decrease in sucrose synthase activity (EC 2.4.1.13); whereas a transient increase in starch and adinosine diphosphate (ADP)-glucose pyrophosphorylase activity (EC 2.7.7.27) as well as decreased leaf sucrose exudation were detected in the beginning of the vegetative phase. These changes in source leaves were accompanied by reductions in sucrose and starch in sink leaves, hexoses and sucrose in roots and hexoses in shoot apex, which were observed before the occurrence of a significant reduction in height and in leaf number in the transgenic lines. These alterations in growth parameters did not persist throughout the development, but were associated with a delay in flowering time and leaf senescence in the SBP antisense lines. A likely involvement of SBP in sink strength is discussed.  相似文献

The turnover and transport of sugars are described in peach (Prunus persica L. Batsch), a species exporting both sucrose and sorbitol. Apparent export rate was slower in peach leaves than in leaves of herbaceous species. Sorbitol was the major soluble end product of photosynthesis and the major soluble carbohydrate in the leaf (higher than sucrose). Carbon fluxes were described using 14C labeling, radioactivity loss curves, and compartmental analysis during the second half of the photoperiod when chemical steady state was reached for soluble carbohydrates. The measured specific radioactivity of sucrose was typical of a primary product. The delayed decrease in specific radioactivity of sorbitol indicated that part of it was secondarily synthesized. Sucrose is proposed to be the carbon source for the delayed synthesis of sorbitol in the light. The sorbitol to sucrose ratio was higher in the petiole than in the leaf tissues. In phloem sap, obtained using stylectomy of aphids and collected from the main stem between source leaves and apex, this ratio was lower than in the petiole, suggesting a preferential sorbitol demand by sinks.  相似文献

The development of sink organs such as fruits and seeds strongly depends on the amount of nitrogen that is moved within the phloem from photosynthetic‐active source leaves to the reproductive sinks. In many plant species nitrogen is transported as amino acids. In pea (Pisum sativum L.), source to sink partitioning of amino acids requires at least two active transport events mediated by plasma membrane‐localized proteins, and these are: (i) amino acid phloem loading; and (ii) import of amino acids into the seed cotyledons via epidermal transfer cells. As each of these transport steps might potentially be limiting to efficient nitrogen delivery to the pea embryo, we manipulated both simultaneously. Additional copies of the pea amino acid permease PsAAP1 were introduced into the pea genome and expression of the transporter was targeted to the sieve element‐companion cell complexes of the leaf phloem and to the epidermis of the seed cotyledons. The transgenic pea plants showed increased phloem loading and embryo loading of amino acids resulting in improved long distance transport of nitrogen, sink development and seed protein accumulation. Analyses of root and leaf tissues further revealed that genetic manipulation positively affected root nitrogen uptake, as well as primary source and sink metabolism. Overall, the results suggest that amino acid phloem loading exerts regulatory control over pea biomass production and seed yield, and that import of amino acids into the cotyledons limits seed protein levels.  相似文献

The photocontrol of the functional coupling between photosynthesis and stomatal conductance in the leaf was investigated in gas exchange experiments using monochromatic light provided by lasers. Net photosynthesis and stomatal conductance were measured in attached leaves of Malva parviflora L. as a function of photon irradiance at 457.9 and 640.0 nanometers.

Photosynthetic rates and quantum yields of photosynthesis were higher under red light than under blue, on an absorbed or incident basis.

Stomatal conductance was higher under blue than under red light at all intensities. Based on a calculated apparent photon efficiency of conductance, blue and red light had similar effects on conductance at intensities higher than 0.02 millimoles per square meter per second, but blue light was several-fold more efficient at very low photon irradiances. Red light had no effect on conductance at photon irradiances below 0.02 millimoles per square meter per second. These observations support the hypothesis that stomatal conductance is modulated by two photosystems: a blue light-dependent one, driving stomatal opening at low light intensities and a photosynthetically active radiation (PAR)-dependent one operating at higher irradiances.

When low intensity blue light was used to illuminate a leaf already irradiated with high intensity, 640 nanometers light, the leaf exhibited substantial increases in stomatal conductance. Net photosynthesis changed only slightly. Additional far-red light increased net photosynthesis without affecting stomatal conductance. These observations indicate that under conditions where the PAR-dependent system is driven by high intensity red light, the blue light-dependent system has an additive effect on stomatal conductance.

The wavelength dependence of photosynthesis and stomatal conductance demonstrates that these processes are not obligatorily coupled and can be controlled by light, independent of prevailing levels of intercellular CO2. The blue light-dependent system in the guard cells may function as a specific light sensor while the PAR-dependent system supplies a CO2-modulated energy source providing functional coupling between the guard cells and the photosynthesizing mesophyll.

植物器官(如叶、叶鞘、绿色的茎等)可以通过光合作用将CO2合成为碳水化合物, 并经过长距离运输到达库组织(如新生组织、花粉、果实等)中进行贮存或利用。蔗糖是高等植物长距离运输碳水化合物的主要形式。蔗糖分子从源到库的运输包括源组织韧皮部的装载、维管束的运输和库组织韧皮部的卸载3个步骤。遗传学和分子生物学研究证明, 蔗糖转运蛋白、转化酶和单糖转运蛋白在糖分子的装载和卸载过程中发挥重要作用。该文综述了目前对光合产物运输过程及其调控分子机制的最新研究进展。  相似文献

Combined δ(13) C and δ(18) O analyses of water-soluble leaf and twig phloem material were used to determine intrinsic water-use efficiency (iWUE) and variability of stomatal conductance at different crown positions in adult European beech (Fagus sylvatica) and Douglas-fir (Pseudotsuga menziesii) trees. Simultaneous gas exchange measurements allowed evaluation of the differences in calculating iWUE from leaf or phloem water-soluble compounds, and comparison with a semi-quantitative dual isotope model to infer variability of net photosynthesis (A(n) ) between the investigated crown positions. Estimates of iWUE from δ(13) C of leaf water-soluble organic matter (WSOM) outperformed the estimates from phloem compounds. In the beech crown, δ(13) C of leaf WSOM coincided clearly with gas exchange measurements. The relationship was not as reliable in the Douglas-fir. The differences in δ(18) O between leaf and phloem material were found to correlate with stomatal conductance. The semi-quantitative model approach was applicable for comparisons of daily average A(n) between different crown positions and trees. Intracanopy gradients were more pronounced in the beech than in the Douglas-fir, which reached higher values of iWUE at the respective positions, particularly under dry air conditions.  相似文献

The hypothesis was tested that, in plants of the alpine meadow grass (Poa alpina L.) exposed to elevated CO2, net photosynthesis and export from source leaves is reduced as a result of feedback from sinks. Nutrient supply was used as one way of reducing photosynthesis and export. Single plants were grown in sand culture under specified controlled environmental conditions for a period of 50 d at two levels of nitrogen and phosphorus ('low': 0.2 mol m-3 N, 0.04 mol m-3 P; 'high': 2.5 mol m-3 N, 0.5 mol m-3 P). Compartmentation within, and export of carbon from, individual youngest fully expanded leaves of acclimated plants was determined using 14C feeding and efflux plus mass balance calculations of carbohydrate export. Independent of treatment, the bulk of soluble carbohydrate (65-75%) was present as fructan, with most of the remainder being sucrose. Depending on nutrient supply, CO2 could alter export from source leaves either by a reduction in the amount of sucrose present in a readily available pool for transport, or by altering the rate constant describing phloem loading.Key words: Poa alpina L., phloem transport, carbohydrate, compartmentation, export, elevated CO2, nutrients.   相似文献

The dynamics of leaf chlorophyll level, nitrogen content, photosynthesis and stomatal conductance were followed in detail in two cultivars of maize (Zea mays) during a short period of water stress, applied at tasseling, and during the subsequent recovery phase. Plants used in the experiment were grown in sand-nutrient solution culture under field weather conditions. Water stress reduced chlorophyll levels, stomatal conductance and photosynthesis, but the nitrogen content of the leaves was not affected. It is concluded that the stress-induced loss of chlorophyll is not mediated by a lack of nitrogen. Considerable differences were observed between genotypes in the rate of post-stress recovery of chlorophyll level. Recovery, upon rewatering, of stomatal conductance and photosynthesis preceded that of chlorophyll level. Losses of up to 40% of leaf chlorophyll content were insufficient to affect rates of photosynthesis measured at mid-day.  相似文献

The hypothesis that electric and hydraulic long-distance signals modify photosynthesis and stomatal aperture upon re-irrigation in intact drought-stressed plants was examined. Maize plants (Zea mays L.) were exposed to drought conditions by decreasing the soil water content to 40-50% of field capacity. The decrease in water content resulted in a decline in stomatal conductance to 50-60% of the level in well-watered plants. Re-irrigation of the plants initiated both hydraulic and electric signals, followed by a two-phase response of the net CO2 uptake rate and stomatal conductance of leaves. The transitional first phase (phase 1) is characterized by a rapid decrease in both levels. In the second phase (phase 2), both parameters gradually increase to levels above those of drought-stressed plants. Elimination of either the hydraulic signal by compensatory pressure application to the root system, or of the electric signal by cooling of the leaf blade gave evidence that the two signals (1) propagated independently from each other and (2) triggered the two-phase response in leaf gas exchange. The results provided evidence that the hydraulic signal initiated a hydropassive decrease in stomatal aperture and for the involvement of electric signals in the regulation of photosynthesis of drought-stressed plants.  相似文献

Metabolite transport processes and primary metabolism are highly interconnected. This study examined the importance of source-to-sink nitrogen partitioning, and associated nitrogen metabolism for carbon capture, transport and usage. Specifically, Arabidopsis aap8(AMINO ACID PERMEASE 8) mutant lines were analyzed to resolve the consequences of reduced amino acid phloem loading for source leaf carbon metabolism,sucrose phloem transport and sink development during vegetative and reproductive growth phase. Results showed that decreased amino acid transport had a negative effect on sink development of aap8 lines throughout the life cycle, leading to an overall decrease in plant biomass. During vegetative stage, photosynthesis and carbohydrate levels were decreased in aap8 leaves, while expression of carbon metabolism and transport genes, as well as sucrose phloem transport were not affected despite reduced sink strength.However, when aap8 plants transitioned to reproductive phase, carbon fixation and assimilation as well as sucrose partitioning to siliques were strongly decreased. Overall,this work demonstrates that phloem loading of nitrogen has varying implications for carbon fixation, assimilation and source-to-sink allocation depending on plant growth stage. It further suggests alterations in source-sink relationships, and regulation of carbon metabolism and transport by sink strength in a development-dependent manner.  相似文献

In broad bean (Vicia faba L.), an apoplastic phloem loader, the sucrose concentration increases up to approximately 2 mM in the leaf apoplast and up to approximately 150 mM in the guard-cell apoplast during the photoperiod. This high concentration in the guard-cell apoplast results from transpiration and is sufficient osmotically to reduce stomatal aperture size by up to 3 microm or approximately 25% of the maximum aperture size. In this paper, we investigated a parallel and required role for high bulk-leaf apoplastic sucrose concentration, which correlates with high photosynthesis rate. An empirically determined combination of lowered light intensity and lowered CO(2) concentration reduced the photosynthesis rate to nominally one-fifth of the control value without a significant change in transpiration. This reduction in photosynthesis caused the sucrose concentration in the leaf apoplast--the immediate source pool for guard cells--to decrease by 70% (to 0.4 mM). In turn, sucrose concentration in the guard-cell apoplast decreased by approximately 80% (to approximately 40 mM). These results complete the required evidence for a non-exclusive, transpiration-linked, photosynthesis-dependent passive mechanism for the modulation of stomatal aperture size. In an ancillary investigation, hexoses in the bulk-leaf apoplast decreased when photosynthesis was lowered, but their concentrations in the guard-cell apoplast of control plants indicated that their osmotic contribution was negligible.  相似文献

Photosynthesis was studied in relation to the carbohydrate status in intact leaves of the C4 plant Amaranthus edulis. The rate of leaf net CO2 assimilation, stomatal conductance and intercellular partial pressure of CO2 remained constant or showed little decline towards the end of an 8-h period of illumination in ambient air (340 bar CO2, 21% O2). When sucrose export from the leaf was inhibited by applying a 4-h cold-block treatment (1°C) to the petiole, the rate of photosynthesis rapidly decreased with time. After the removal of the cold block from the petiole, further reduction in photosynthetic rate occurred, and there was no recovery in the subsequent light period. Although stomatal conductance declined with time, intercellular CO2 partial pressure remained relatively constant, indicating that the inhibition of photosynthesis was not primarily caused by changes in stomatal aperture. Analysis of the leaf carbohydrate status showed a five- to sixfold increase in the soluble sugar fraction (mainly sucrose) in comparison with the untreated controls, whereas the starch content was the same. Leaf osmotic potential increased significantly with the accumulation of soluble sugars upon petiole chilling, and leaf water potential became slightly more negative. After 14 h recovery in the dark, photosynthesis returned to its initial maximum value within 1 h of illumination, and this was associated with a decline in leaf carbohydrate levels overnight. These data show that, in Amaranthus edulis, depression in photosynthesis when translocation is impaired is closely related to the accumulation of soluble sugars (sucrose) in source leaves, indicating feedback control of C4 photosynthesis. Optimal allocation of leaf nitrogen maximizes daily CO2 assimilation for a given leaf nitrogen concentration. According to the hypothesis of optimization, this condition occurs when the partial derivative of assimilation rate with respect to leaf nitrogen concentration is constant. This hypothesis predicts a linear increase of assimilation rate with leaf nitrogen concentration under constant conditions. Plants of Amaranthus powellii Wats. were grown at 1, 5, 10, or 45 millimolar nitrate to obtain leaves with different nitrogen concentrations. Assimilation rate at 340 microbar CO2/bar, stomatal conductance, CO2- and light-saturated net photosynthetic rate, the initial slope of the CO2 response of photosynthesis, ribulose-1,5′-bisphosphate carboxylase activity, and phosphoenolpyruvate carboxylase activity were linearly related to estimated or actual leaf nitrogen concentration. The data are consistent with the optimal use of leaf nitrogen. This hypothesis and the hypothesis of optimal stomatal conductance were combined to determine the relationship between conductance and leaf nitrogen concentration. The slope of conductance versus leaf nitrogen concentration was not significantly different than the slope predicted by the combination of the two hypotheses. Stomatal conductance was linearly related to leaf nitrogen in the field and the slope decreased with lower xylem pressure potentials in a manner consistent with the hypotheses. Finally, apparent maximum stomatal aperture of isolated abaxial epidermal strips was linearly related to leaf nitrogen suggesting stomatal conductance and assimilation rate are controlled in parallel by leaf nitrogen concentration or some factor correlated with leaf nitrogen.  相似文献
