线粒体是细胞物质代谢与能量代谢的中心,在多种生理和病理过程中扮演着重要角色。表观遗传修饰是一种独立于DNA序列并在建立与维持特定基因表达谱中发挥主要作用的遗传调控模式。近年来的研究表明,线粒体能量代谢通过中间产物,介导线粒体–核信号的传递,调节染色质的表观修饰状态,进而影响基因表达。线粒体代谢紊乱可以诱导表观遗传重编程,进而启动衰老表型及退行性疾病的发生。本文综述了线粒体代谢与染色质表观遗传修饰关系的研究进展,探讨了线粒体应激在染色质重组中发挥的作用,展望了其在认知功能障碍等衰老相关性疾病研究中的前景。  相似文献

衰老是多因素作用下的组织、器官功能性衰退进程。自噬是真核细胞溶酶体介导而降解细胞质成分的过程,参与衰老及相关的各种生理病理过程。环境因素所致衰老则大多数伴随着表观遗传修饰的改变。热量限制被认为是调节衰老、延长寿命的干预措施。本综述围绕自噬与表观遗传修饰,阐述了热量限制引发自噬、通过表观遗传修饰延缓衰老的分子机制,以及两者在热量限制中的交互作用。以期为衰老与延缓衰老的机制研究提供新思路。  相似文献

衰老是机体随着时间推移而表现的一种各组织器官退化,最终导致细胞乃至个体死亡的进程。干细胞的衰竭、功能缺失及表观遗传的改变被认为是衰老的驱动力。对模式生物及人类衰老的研究发现,相对于基因决定,表观遗传改变在寿命影响中起到更为重要的作用,饮食、运动等环境因素会引起细胞表观遗传的改变,包括DNA甲基化水平改变、组蛋白翻译后修饰改变、染色质重塑、非编码RNA调控等,最终会对细胞乃至个体寿命产生影响。该综述主要是对表观遗传改变影响细胞衰老的最新发现进行概述,并且尝试对可能的能够延缓或者实现健康衰老的策略进行总结探讨,期望为以后尝试延缓衰老及实现健康衰老提供潜在的干预靶点。  相似文献

表观遗传修饰异常见于人类的多种疾病(如肿瘤、老年性疾病、发育源性疾病等),影响着这些疾病的发生发展。已有的研究表明,异常表观遗传改变可以作为疾病状态和疾病预测的生物标志物。表观遗传修饰改变的可逆性和可控性也为疾病早期的预防和治疗提供了新策略。本文对DNA甲基化修饰、组蛋白共价修饰、非编码RNA等三种表观遗传方式在肿瘤、老年性疾病和发育源性疾病的研究,以及三者作为表遗传生物标志物在疾病早期诊断和治疗的应用展开介绍,以期为肿瘤、老年性和发育源性相关疾病的诊断与治疗提供借鉴和 参考。  相似文献

叶片是植物重要的光合器官, 它的衰老由外界环境刺激和内源发育信号所启动, 复杂的基因调控网络参与衰老过程的精确调控。最新研究表明, 植物通过对基因表达的重编程, 在表观遗传水平上调节着叶片衰老过程。该文简要介绍了表观遗传的分子机制, 在此基础上重点综述了组蛋白修饰、染色质重塑、DNA甲基化及小RNAs途径对叶片衰老调控的最新研究进展, 同时讨论了该领域存在的问题和未来研究方向。  相似文献

On April 18, 2007 an international meeting on Pathophysiology of Ageing, Longevity and Age-Related Diseases was held in Palermo, Italy. Several interesting topics on Cancer, Immunosenescence, Age-related inflammatory diseases and longevity were discussed. In this report we summarize the most important issues. However, ageing must be considered an unavoidable end point of the life history of each individual, nevertheless the increasing knowledge on ageing mechanisms, allows envisaging many different strategies to cope with, and delay it. So, a better understanding of pathophysiology of ageing and age-related disease is essential for giving everybody a reasonable chance for living a long and enjoyable final part of the life.  相似文献

Among the age-related diseases, the development of cognitive impairments, in particular dementia, is the most devastating for the individual and has great social and healthcare costs. Accurate information is needed about the prevalence and incidence of cognitive disorders and the physiology of the ageing brain. In particular, only scant data are available about the relationship between ageing, cognitive status and nutritional factors. In order to address these issues we planned the Conselice Study of Brain Ageing, a longitudinal study of physiologic and pathologic brain ageing. The center involved in the study was the municipality of Conselice, Ravenna province, in the Northern Italian region Emilia-Romagna. A total of 1016 subjects aged 65 and over was enrolled at baseline. Information about cognitive status at 4-years of follow-up was collected for 940 of them. These data have been used to estimate prevalence and incidence of dementia in the elderly Italian population and to investigate the possible role of baseline blood homocysteine as risk factors for dementia.  相似文献

ABSTRACT: The demographic and social changes of the past decades have determined improvements in public health and longevity. So, the number of centenarians is increasing as a worldwide phenomenon. Scientists have focused their attention on centenarians as optimal model to address the biological mechanisms of "successful and unsuccessful ageing". They are equipped to reach the extreme limits of human life span and, most importantly, to show relatively good health, being able to perform their routine daily life and to escape fatal age-related diseases, such as cardiovascular diseases and cancer. Thus, particular attention has been centered on their genetic background and immune system. In this review, we report our data gathered for over 10 years in Sicilian centenarians. Based on results obtained, we suggest longevity as the result of an optimal performance of immune system and an over-expression of anti-inflammatory sequence variants of immune/inflammatory genes. However, as well known, genetic, epigenetic, stochastic and environmental factors seem to have a crucial role in ageing and longevity. Epigenetics is associated with ageing, as demonstrated in many studies. In particular, ageing is associated with a global loss of methylation state. Thus, the aim of future studies will be to analyze the weight of epigenetic changes in ageing and longevity.  相似文献

Ageing is associated with learning and memory impairments. Data are reviewed that suggest that age-related impairments of hippocampal-dependent forms of memory, may be caused, in part, by altered synaptic plasticity mechanisms in the hippocampus, including long-term potentiation (LTP). To the extent that the mechanisms responsible for LTP can be understood, it may be possible to develop therapeutic approaches to alleviate memory decline in normal ageing.  相似文献

Ageing is a crucial risk factor for the development of age-related cardiovascular diseases. Therefore, the molecular mechanisms of ageing and novel anti-ageing interventions need to be deeply studied. Alginate oligosaccharide (AOS) possesses high pharmacological activities and beneficial effects. Our study was undertaken to investigate whether AOS could be used as an anti-ageing drug to alleviate cardiac ageing. D-galactose (D-gal)-induced C57BL/6J ageing mice were established by subcutaneous injection of D-gal (200 mg·kg-1·d-1) for 8 weeks. AOS (50, 100 and 150 mg·kg-1·d-1) were administrated intragastrically for the last 4 weeks. As a result, AOS prevented cardiac dysfunction in D-gal-induced ageing mice, including partially preserved ejection fraction (EF%) and fractional shortening (FS%). AOS inhibited D-gal-induced up-regulation of natriuretic peptides A (ANP), brain natriuretic peptide (BNP) and ageing markers p53 and p21 in a dose-dependent manner. To further explore the potential mechanisms contributing to the anti-ageing protective effect of AOS, the age-related mitochondrial compromise was analysed. Our data indicated that AOS alleviated D-gal-induced cardiac ageing by improving mitochondrial biogenesis, maintaining the mitochondrial integrity and enhancing the efficient removal of impaired mitochondria. AOS also decreased the ROS production and oxidative stress status, which, in turn, further inhibiting cardiac mitochondria from being destroyed. Together, these results demonstrate that AOS may be an effective therapeutic agent to alleviate cardiac ageing.  相似文献
