Twenty-five lambs grazing a parasite-contaminated pasture were divided into four groups. One group of seven lambs were left untreated and three groups of six lambs each were treated with levamisole at 2 week, 3 week or 1 month intervals respectively. Body weights, fecal egg counts and packed cell volumes were determined every 2 weeks for 8 weeks. Lambs were then necropsied on the day they would have received another treatment, thus the maximum accumulation of parasites at each treatment frequency was measured. Treatment at 2 and 3 week intervals markedly suppressed fecal egg counts, but egg counts of sheep treated every month rose between treatments and untreated animals continually shed eggs. Treatment at all three frequencies provided only slight protection from blood loss, but it lessened weight loss. There was little difference between treatment groups in burdens of adult H. contortus, but burdens of T. axei and T. colubriformis and arrested larvae of H. contortus increased with decreasing frequency of treatment. The results support the findings of other workers that Trichostrongylus spp. have a long adult life span and continue to accumulate with time, whereas H. contortus may have a short adult life span and a rapid population turnover. In the face of constant reinfection, repeated anthelmintic treatment gives more protection from the effects of the long-lived parasites than from those with a rapid population turnover.  相似文献

Self-cure reactions and immunological responses preventing establishment of Haemonchus contortus in sheep may operate as separate entities. In one experiment, self-cure occurred when challenge infection with 5000 larvae was superimposed on an infection with 5000 larvae given to worm-free sheep 6 weeks previously. Resident worms were rejected and establishment of infection by incoming larvae was impeded. The latter effect was not observed in sheep treated similarly but with resident parasites removed by treatment with oxfendazole before challenge. In another experiment, younger worm-free sheep primed by three infections with 2000 larvae at intervals of 2 weeks or a single infection with 6000 larvae were challenged with 10,000 larvae 6 weeks after the first priming infection. Self-cure was not incited but establishment of infection was impeded in sheep primed with three divided doses of larvae whether or not priming infections had been removed by oxfendazole. Infection regimes used for priming did not influence numbers of arrested fourth-stage larvae derived from challenge infection. However, more arrested larvae were present when challenge was superimposed on extant infections, indicating that resident worms or a factor activated by their presence induced developmental arrest. In a third experiment, large burdens with H. contortus were established in sheep immunosuppressed with the corticosteroid, dexamethasone, at the time of infection. Self-cure was not triggered by a challenge infection given 32 days later either in these sheep, or in sheep with a smaller worm burden derived from infection given without immunosuppression. Faecal egg counts, however, indicated that development of the challenge infection was prevented in both groups of sheep.

Investigation of self-cure is restricted by lack of a predictable system for reproducing the phenomenon. Self-cure was induced by a single infection with 5000 larvae in mature sheep but not with 6000 larvae in immature sheep. Three infections with 3000 larvae given at intervals of 2 weeks to mature sheep did not prime for self-cure. Procedures aimed at heightening immediate hypersensitivity, i.e. treatment with pertussis vaccine or concurrent infections with Ostertagia circumcincta, did not promote self-cure reactivity in the latter situation.  相似文献

Lambs were infected with 6000 Trichostrongylus colubriformis L3 per week for 18, 12 or 6 weeks, beginning at ages 14, 20 and 26 weeks, respectively. At the end of the primary infection subgroups had geometric mean adult worm burdens of 5000, 15,000 and 18,000, respectively. In the remaining sheep the worm population was removed with anthelmintic, and sheep had no further larval intake until challenged with T. colubriformis 1,6, 12, 18 or 24 weeks later. In the groups given 18 or 12 weeks primary infection, establishment of challenge doses was low (less than 25% of establishment in helminth naive controls) in most animals at all challenge times. However, for the groups given 6 weeks primary infection, establishment was low only at the first two challenge times. Thereafter it had similar mean to control groups, but much greater variance. Other subgroups were challenged with T. colubriformis, Ostertagia circumcincta and Haemonchus contortus 1 week after worm removal. In these animals T. colubriformis establishment was not different to animals challenged at the same time with T. colubriformis alone, however immunity to T. colubriformis afforded little protection against the other species. The results of this experiment were incorporated into a simulation model of the population dynamics of T. colubriformis.  相似文献

1988. Infection with Haemonchus contortus in sheep and the role of adaptive immunity in selection of the parasite. International Journal for Parasitology 18: 1071–1075. A series of infections with Haemonchus contortus in immune and non-immune sheep gave no indication that successive generations of the parasite were selected for the enhanced ability to cope with host-protective immunity. Separate subpopulations of the parasite were passaged through individual immune sheep and were compared at each generation with infections by the pooled populations in an additional panel of immune sheep. The experiment ceased when infections became too low to allow the production of sufficient infective larvae for reinfection. Results showed that H. contortus is unable to make adjustments to the immune status of a given sheep. Attention is thus diverted from the process through which host identifies self and non-self and from the histocompatibility system as the targets for possible antigen-mimicry by H. contortus.  相似文献

Lines of Merino sheep selected for increased (IRH) and decreased (DRH) resistance to Haemonchus contortus were compared with an unselected (CH) line, after approximately four generations of selection. Measurements were recorded on 69 IRH, 47 DRH and 84 CH animals. Following artificial challenge with H. contortus, the IRH line had significantly (P < 0.001) lower faecal egg counts than the CH and DRH lines (2730,12,720 and 17,400 epg, respectively). Significant differences (P<0.05) were found between all lines in the minimum packed cell volumes during artificial infection (25.7, 22.0 and 20.3%) and in faecal egg counts after natural infection (140, 3590 and 8750 epg). Differences were also recorded (P<0.05) following artificial challenge with Trichostrongylus colubriformis (490, 840 and 1340 epg). On a percentage basis, faecal egg counts in the IRH line deviated less from the CH line following artificial infection with T. colubriformis (42%) than with H. contortus (79%). The reverse was true for the DRH line (60 and 37%, respectively). Differences in egg output of this magnitude should have a marked effect on requirements for anthelmintic treatment, rate of development of drug resistance and level of pasture contamination when the lines are grazed separately.  相似文献

Hammondia hammondi and Toxoplasma gondii are two related coccidian parasites, with cats as definitive hosts and warm-blooded animals as intermediate hosts. It is difficult to differentiate them by morphological and serological parameters. In the present study, primers were designed to specifically amplify the ITS-1 region of H. hammondi to differentiate it from T. gondii. Attempts were made to detect the presence of H. hammondi DNA in the tissues of mice infected with H. hammondi alone, as well as from mixed infections with T. gondii, using the newly designed primers. The de novo primers effectively amplified the H. hammondi-specific target fragment from all samples containing H. hammondi, including those with concomitant T. gondii infection. Further, the primers did not amplify any fragment from the related parasites like T. gondii, Neospora caninum and Hammondia heydorni. The new primers provide simple and efficient means to differentially diagnose H. hammondi from T. gondii even in samples containing both parasites, thus obviating the need for other labourious techniques like mouse bioassay and in vitro cultivation.  相似文献

First stage (L1) larvae of Haemonchus contortus, Trichostrongylus colubriformis and Ostertagia circumcincta can be cryopreserved in the presence of DMSO using a two-step freezing protocol involving an initial period at −80°C prior to transfer to liquid nitrogen. Thawed L1 larvae continue development in vitro producing third stage (L3) larvae that are infective to sheep when dosed per os. Establishment rates for L3 larvae grown from thawed L1 larvae were 40 and 80% for H. contortus and T. colubriformis, respectively. There was no difference in survival or infectivity between benzimidazole (BZ)-susceptible and BZ-resistant H. contortus isolates and cryopreservation caused no shift in their BZ-resistance status as indicated in an in vitro larval development assay. Cryopreservation also had no effect on the sensitivity of these isolates to the avermectins or levamisole in vitro. High survival rates (60–70%), good levels of establishment and the stability of anthelmintic resistance status of isolates indicate that little if any selection occurs during the cryopreservation process. L1 larvae of all 3 species have been successfully recovered after 16 months storage in liquid nitrogen, cultured to the L3 stage and established in sheep.  相似文献

The identification of genes associated with anthelmintic resistance can be facilitated in Haemonchus contortus by the ability of this species to hybridise with Haemonchus placei. Although the hybrid males are sterile, the lines can be rescued by backcrossing the females to either parental species. Resistance genes can be retained in Haemonchus hybrids, while the unwanted contortus background is removed through backcrossing to H. placei and anthelmintic selection of the progeny. Under this selection, genes involved in resistance would retain the H. contortus nucleotide sequence, while those that are not would either be H. placei or a random mixture of both, depending on the amount of backcrossing that had occurred. The first candidate gene to be tested in this system was a Haemonchus P-glycoprotein, hcpgp-1. hcpgp-1 was amplified, cloned and sequenced from H. contortus and H. placei. Two restriction sites were then identified in the sequenced product; one specific to H. contortus hcpgp-1 and the other found only in the H. placei gene. These genes were identified from macrocyclic lactone selected and non-selected worms by restricting PCR products from individual worms. Fitted occurrence of the H. contortus allele was 49% of unselected worms and 69% of macrocyclic lactone selected worms. The probability of this percentage occurring by chance was P=0.006. Thus macrocyclic lactone selection was acting to increase the percentage of hcpgp-1 from macrocyclic-lactone-resistant CAVRS.  相似文献

Trypanosoma lewisi and Trypanosoma musculi were collected from immunosuppressed infected hosts and extracted with phosphate buffered saline. Antisera were obtained from rats repeatedly infected with T. lewisi or mice repeatedly infected with T. musculi. Cellular antigens (CAg) present in the extracts of the parasites were analyzed by microimmunodiffusion (MID), crossed immunoelectrophoresis (CIE) and tandem crossed immunoelectrophoresis (TCIE). Trypanosome extracts were absorbed with the heterologous hyperimmune antisera to examine shared and unique antigens of the parasites.

Extracts of T. lewisi formed four precipitin lines when reacted with hyperimmune rat antiserum and three precipitin lines were detected by mouse anti-T. musculi serum in MID analyses. T. musculi extract formed two precipitin lines with mouse hyperimmune serum and two precipitin lines with rat anti-T. lewisi serum in the MID tests. When T. lewisi was reacted with the homologous hyperimmune rat antiserum in CIE, 14 precipitin peaks developed, while T. musculi extract formed eight peaks with homologous mouse hyperimmune serum. Seven precipitin peaks developed when T. lewisi extract was reacted with the mouse antiserum and T. musculi extract formed eight peaks during its electrophoretic migration into rat anti-T lewisi serum. TCIE clearly showed that five T. lewisi CAg could not be detected in the T. musculi extract by the rat antiserum, while mouse anti-T. musculi serum formed six precipitin peaks with the T. lewisi extract and seven peaks with the homologous extract. One of the CAg present in the T. musculi extract was not found in the T. lewisi extract. Absorptions of the extracts with heterologous antisera and subsequent CIE against the homologous antisera indicated three of the CAg of T. lewisi were not shared by T. musculi, while a single antigen of T. musculi was not detected in T. lewisi. Although concentrations of antibodies in each of the antisera and CAg in the parasite extracts were not equivalent, the data indicated that a minimum of eight CAg are shared by these rodent trypanosomes and at least three antigens appeared to be unique to T. lewisi and a single antigen to T. musculi.  相似文献

Immunoperoxidase staining of muscle infected with Trichinella spiralis for murine collagen types I and IV provided both qualitative and quantitative evidence of extensive synthesis of both types of collagen by fibroblasts in infected muscle compared to that seen uninfected muscle. Moreover, fibroblasts in muscle infected with T. pseudospiralis, a nonencapsulating species, showed significantly less staining for both types of collagen compared to muscle from mice infected with T. spiralis. Analysis of collagen composition of isolated nurse cells using an ELISA specific for either type I or type IV murine collagen suggested that of these 2 types of collagen, only type IV basement membrane collagen is found in Trichinella capsular collagen. Excretory/secretory products of T. spiralis and T. pseudospiralis induced extensive synthesis of exclusively type IV collagen by 3T3 murine fibroblasts in vitro.  相似文献

The infection of laboratory rodents by cyclically established Trypanosoma vivax infections in ruminants was only successful for a period up to 4 weeks after the first fly feed on the ruminants; rats and mice were equally susceptible. Similar results were obtained when ruminants were inoculated by syringe with infected blood from the early stage of a cyclically transmitted T. vivax infection. The development of antibodies, as measured by fluorescence, in the ruminants had no influence on the outcome of the rodent-inoculations.

When blood was inoculated into susceptible oxen from the later chronic stage of a cyclically and from a mechanically established bovine infection, subsequent rodent infections were achieved only occasionally; parasitaemia in the rodents was low and the patent period short. It was concluded that the infectivity of T. vivax for rodents was predominantly a characteristic of the isolate and not of the donor or recipient mammalian species. The value of blood inoculation of rodents in epidemiological studies for detecting early infections of T. vivax is emphasized.  相似文献

Species of the Heteropogon-Themeda clade are ecologically important grasses distributed across the tropics, including widespread species, such as the pantropical Heteropogon contortus and Themeda triandra, and range-restricted species such as Heteropogon ritchiei and Themeda anathera. Here, we examine habitat preferences of the grassland/savanna and wetland species by describing bioclimatic niche characteristics, characterizing functional traits, and investigating the evolution of functional traits of 31 species in the Heteropogon-Themeda clade in relation to precipitation and temperature. The climatic limits of the clade are linked to mean annual precipitation and seasonality that also distinguish seven wetland species from 24 grassland/savanna species. Tests of niche equivalency highlighted the unique bioclimatic niche of the wetland species. However, climatic factors do not fully explain species geographic range, and other factors are likely to contribute to their distribution ranges. Trait analyses demonstrated that the wetland and grassland/savanna species were separated by culm height, leaf length, leaf area, awn length, and awn types. Phylogenetic analyses showed that the wetland species had tall stature with long and large leaves and lack of hygroscopic awns, which suggest selective pressures in the shift between savanna/grassland and wetland. The two most widespread species, H. contortus and T. triandra, have significantly different bioclimatic niches, but we also found that climatic niche alone does not explain the current geographic distributions of H. contortus and T. triandra. Our study provides a new understanding of the biogeography and evolutionary history of an ecologically important clade of C4 tropical grasses.  相似文献

When mice inoculated with five cysticercoids of Hymenolepis microstoma were challenged with H. nana, they showed strong resistance to challenges with both eggs and cysticercoids of H. nana from day 20. The immunity became complete from day 30 onward: no tissue cysticercoids or lumenal adults of H. nana were established in these mice. However, when mice were challenged with H. nana 10 or 20 days after 10-day old immature H. microstoma were removed by an anthelmintic, the immunity evoked was directed exclusively against the lumenal phase of the cysticercoid challenge but not the tissue cysticercoids of the egg challenge. When mice experienced the prepatent infection with H. microstoma twice, the immunity evoked was also against the lumenal phase of the egg challenge: the oncospheres developed into tissue cysticercoids but thereafter completely failed to develop into lumenal adult tapeworms. Infection with a single cysticercoid of H. microstoma was shown to be sufficient to evoke immunity against H. nana cysticercoid infections in two strains of mice. Sera from mice which experienced a patent infection with H. microstoma revealed that IgG, IgA, IgM isotypes reacted against oncospheres and cysticercoids of both species, while sera from mice which experienced two prepatent infections reacted with cysticercoids only. Sera from H. microstoma infected mice resistant to H. nana caused precipitations on 4-day-old H. nana in vitro. A correlation exists between the presence of stage specific antibodies and resistance to the different stages.  相似文献

In the intestine of two wolves (Canis lupus) from Germany 33 adult tapeworms were found. The cestodes were determined as Taenia krabbei Moniez, 1879 (Syn. T. cervi Christiansen, 1931). Adult parasites were compared with cysticerci from thigh muscles of roe deer (Capreolus capreolus) as well as cysticerci from cardiac muscles of sheep (Ovis ammon). It is postulated that T. krabbei from wolf and roe deer is a valid and distinct species which uses Cervidae as original intermediate hosts. T. krabbei is highly similar in all morphological criteria to T. ovis. However, the life history of T. ovis includes Bovidae, preferably sheep as intermediate hosts.  相似文献

Seroprevalence of equine babesiosis in the Black Sea region of Turkey   总被引:1,自引:0,他引:1  
The prevalence of Theileria equi and Babesia caballi was determined in equid blood samples in five provinces of the Black Sea region of Turkey by using the indirect fluorescent antibody test (IFAT). Of 153 samples, 53 (34.6%) and 33 (21.5%) were seropositive to B. caballi and T. equi, respectively. In addition, 8 (5.2%) of samples were seropositive to both T. equi and B. caballi. Anti T. equi and B. caballi antibodies were detected in all five regions. The prevalence of B. caballi was higher than T. equi in all counties. Antibodies to T. equi and B. caballi were detected in horses of all ages, and there were no significant differences among age groups. Out of 84 horses, 32 (38.0%) were positive for B. caballi infection and 20 (23.8%) were positive for T. equi infection. Five horses (5.6%) were found to be seropositive to both B. caballi and T. equi. Of 38 donkeys, 14 (36.8%) were found to be positive for B. caballi infection and 5 (13.1%) positive for T. equi infection. In addition, 2 (5.2%) samples were seropositive for both T. equi and B. caballi infections. Out of 31 mules, 8 (25.8%) were positive for B. caballi infection and 8 (25 8%) positive for T. equi infection. One (3.2%) sample was seropositive for both T. equi and B. caballi infections. Of all the animals in this study, only 3 horses were infected by Rhipicephalus turanicus and Hyalomma detritum, and no haemoparasites were detected by microscopic examination.  相似文献

Experimental American leishmaniasis and Chagas' disease in the Brazilian squirrel monkey: cross immunity and electrocardiographic studies of monkeys infected with Leishmania braziliensis and Trypanosoma cruzi. Adult, laboratory-bred squirrel monkeys (Saimiri sciureus) previously infected with either Leishmania braziliensis braziliensis or L. b. panamensis were challenge infected with blood-form trypomastigotes of Trypanosoma cruzi (Brazil strain). Monkeys previously infected with T. cruzi were challenged with stationary phase promastigote forms of L. b. braziliensis. Monkeys were examined during the course of challenge for evidence of infection, electrocardiographic alterations and parasite-specific antibody responses. T. cruzi epimastigotes were cultured from the blood of monkeys up to 3 months after challenge with this parasite. Unulcerated cutaneous lesions appeared and persisted in monkeys challenged with L. b. braziliensis. The formation of satellite lesions was observed in one monkey. Increased QRS intervals were not observed in T. cruzi challenged monkeys without prior cardiac irregularities and QRS left axis shifts were observed in only two of these monkeys. Elevated titers of parasite binding IgM and IgG specific for both T. cruzi and L. braziliensis were observed in all monkeys following challenge. These results indicate that prior infection with T. cruzi or L. braziliensis does not protect against heterologous challenge infection with these organisms. However, prior infection with Leishmania parasites may provide some protection against chagasic cardiopathies.  相似文献

Primary amoebic meningo-encephalitis (PAME) in mouse can be easily produced by intranasal inoculation of Naegleria aerobia Singh & Das, 1970 and Hartmannella culbertsoni Singh & Das, 1970. The ultrastructural characteristics of N. aerobia and H. culbertsoni from infected mouse brain tissues showed many similar characteristics. The distinctive features between the two were the retention of phagocytic activity by N. aerobia as well as pinocytic activity in the ingestion of food as compared to H. culbertsoni where only pinocytic activity was observed. Staining characteristics of mitochondria suggested that N. aerobia was more active in the brain tissues than H. culbertsoni. Membrane-bound darkly stained bodies of unknown function were found in the brains infected with both N. aerobia and H. culbertsoni. They may have some bearing on the pathogenicity of these amoebae. Human cases of PAME so far reported are mostly due to N. aerobia and rarely with H. culbertsoni. This may be due to certain inherent characteristics present in N. aerobia which help them to survive on human brain tissue. On the other hand, the same may be lacking in H. culbertsoni. These ultrastructural differences in both N. aerobia and H. culbertsoni, as revealed by electron microscope studies of the infected brain tissues of mice, are presented in this paper.  相似文献

A comparison of worm burdens of grazing Merino lambs that were untreated, treated once and allowed to graze for 2 weeks, or given suppressive anthelmintic treatment until their final 2 weeks on pasture, showed that resistance to reinfection with Ostertagia circumcincta and Trichostrongylus colubriformis did not develop until the lambs were about 8 months old. By contrast, there was clear evidence that untreated lambs, but not lambs given one or more treatments, had acquired substantial resistance to infection with Haemonchus contortus by an age of 4 months.  相似文献

Cattle immunized against Theileria parva (Muguga) showed little resistance to Theileria lawrencei (Serengeti) stabilate challenge, while cattle immune to T. lawrencei (Serengeti) were fully resistant to challenge with T. parva (Muguga) stabilate. Cattle inoculated with cultured lymphoid cells infected with T. lawrencei (Serengeti) macroschizonts survived a subsequent T. lawrencei (Serengeti) stabilate challenge.  相似文献

Ray flower and leaf flavonoids were investigated for the three species of Helianthus series Microcephali. Ray flowers of all species contain coreopsin, sulphurein, and quercetin 7-O-glucoside; those of H. microcephalus also contain quercetin 3-O-glucoside. A mixture of flavonoid aglycones, mostly methoxylated flavones, occurs in leaves of H. microcephalus, but not in H. glaucophyllus or H. laevigatus which also lack the glandular trichomes that in Helianthus are typically associated with flavonoid aglycones. The presence of compounds with the 6,8,4′ pattern of methoxylation in H. microcephalus suggests that the series is more similar in flavonoids to series Angustifolii than to series Corona-solis.  相似文献
