We extend on the work developed by R.R. Poznanski and J. Bell from a linearized somatic persistent sodium current source to a non-linear representation of the dendritic Na(+)P current source associated with a small number of persistent sodium channels. The main objective is to investigate the modulation in the amplification of excitatory postsynaptic potentials (EPSPs) in dendrites studded with persistent sodium channels. The relation between membrane potential (V) and persistent sodium current density (I(NaP)) is approximated heuristically with a sigmoidal function and the resultant cable equation is solved analytically using a regular perturbation expansion and Green's function techniques. The transient simulated (non-evoked) response is found as a result of current injection in the form of synaptically induced voltage change located at a distance from the recording site in a cable with a uniform distribution of ion channel densities per unit length of cable (the so-called 'hot-spots') and with the conductance of each hot-spot (i.e., number of channels per hot-spot) assumed to be a constant. The results show an amplification in the observed EPSPs to be compatible with the experimentally derived estimates, and in addition a saturation in the amplification is observed indicating an optimum number of ionic channels.  相似文献

Layer five pyramidal neurons of rat and cat neocortex have numerous ionic conductance mechanisms. The presence of these voltage-dependent conductances in the dendrites has a significant effect on the transmission of current from synaptic sites to the spike generating region in the proximal axon. Here we show such threshold activation of persistent sodium channels markedly amplifies current flowing through glutamate activated dendritic channels.  相似文献

The present study compares the structure and function of retinal ganglion and amacrine cell dendrites. Although a superficial similarity exists between amacrine and ganglion cell dendrites, a comparison between the branching pattern of the two cell types reveals differences which can only be appreciated at the microscopic level. Whereas decremental branching is found in ganglion cells, a form of non-decremental or "trunk branching" is observed in amacrine cell dendrites. Physiological differences are also observed in amacrine vs ganglion cells in which many amacrine cells generate dendritic impulses which can be readily distinguished from those of the soma, while separate dendritic impulses in ganglion cell dendrites have not been reported. Despite these differences, both amacrine and ganglion cell dendrites appear to contain voltage-gated ion channels, including TTX-sensitive sodium channels. One way to account for separate dendritic impulses in amacrine cells is to have a higher density of sodium channels and we generally find in modeling studies that a dendritic sodium channel density that is more than about 50% of that in the soma is required for excitatory, synaptic currents to give rise to local dendritic spike activity. Under these conditions, impulses can be generated in the dendrites and propagate for some distance along the dendritic tree. When the soma generates impulse activity in amacrine cells, it can activate, antidromically, the entire dendritic tree. Although ganglion cell dendrites do not appear to generate independent impulses, the presence of voltage-gated ion channels in these structures appears to be important for their function. Modeling studies demonstrate that when dendrites lack voltage-gated ion channels, impulse activity evoked by current applied to the cell body is generated at rates that are much higher than those observed physiologically. However, by placing ion channels in the dendrites at a reduced density compared to those of amacrine cells, the firing rate of ganglion cells becomes more physiological and the relationship between frequency and current (F/I relationship) can be precisely matched with physiological data. Recent studies have demonstrated the presence of T-type calcium channels in ganglion cells and our analysis suggests that they are found in higher density in the dendrites compared to the soma. This is the first voltage-gated ion channel which appears more localized to the dendrites than other cell copartments and this difference alone cries for an interpretation. The presence of a significant T-type calcium channel density in the dendrites can influence their integrative properties in several important ways. First, excitatory synaptic currents can be augmented by the activation of T-type calcium channels, although this is more likely to occur for transient rather than sustained synaptic currents because T-type currents show strong inactivation properties. In addition, T-type calcium channels may serve to limit the electrical load which dendrites impose on the spike initiation process and thus enhance the speed with which impulses can be triggered by the impulse generation site. Dendrites of many types of neurons contain voltage-dependent conductances that are active at subthreshold membrane potentials. To understand the computations neurons perform it is key to understand the role of active dendrites in the subthreshold processing of synaptic inputs. We examine systematically how active dendritic conductances affect the time course of postsynaptic potentials propagating along dendrites, and how they affect the interaction between such signals. Voltage-dependent currents can be classified into two types that have qualitatively different effects on subthreshold input responses: regenerative dendritic currents boost and broaden EPSPs, while restorative currents attenuate and narrow EPSPs. Importantly, the effects of active dendritic currents on EPSP shape increase as the EPSP travels along the dendrite. The effectiveness of active currents in modulating the EPSP shape is determined by their activation time constant: the faster it is, the stronger the effect on EPSP amplitude, while the largest effects on EPSP width occur when it is comparable to the membrane time constant. We finally demonstrate that the two current types can differentially improve precision and robustness of neural computations: restorative currents enhance coincidence detection of dendritic inputs, whereas direction selectivity to sequences of dendritic inputs is enhanced by regenerative dendritic currents.  相似文献

Many neurons receive excitatory glutamatergic input almost exclusively onto dendritic spines. In the absence of spines, the amplitudes and kinetics of excitatory postsynaptic potentials (EPSPs) at the site of synaptic input are highly variable and depend on dendritic location. We hypothesized that dendritic spines standardize the local geometry at the site of synaptic input, thereby reducing location-dependent variability of local EPSP properties. We tested this hypothesis using computational models of simplified and morphologically realistic spiny neurons that allow direct comparison of EPSPs generated on spine heads with EPSPs generated on dendritic shafts at the same dendritic locations. In all morphologies tested, spines greatly reduced location-dependent variability of local EPSP amplitude and kinetics, while having minimal impact on EPSPs measured at the soma. Spine-dependent standardization of local EPSP properties persisted across a range of physiologically relevant spine neck resistances, and in models with variable neck resistances. By reducing the variability of local EPSPs, spines standardized synaptic activation of NMDA receptors and voltage-gated calcium channels. Furthermore, spines enhanced activation of NMDA receptors and facilitated the generation of NMDA spikes and axonal action potentials in response to synaptic input. Finally, we show that dynamic regulation of spine neck geometry can preserve local EPSP properties following plasticity-driven changes in synaptic strength, but is inefficient in modifying the amplitude of EPSPs in other cellular compartments. These observations suggest that one function of dendritic spines is to standardize local EPSP properties throughout the dendritic tree, thereby allowing neurons to use similar voltage-sensitive postsynaptic mechanisms at all dendritic locations.  相似文献

Dendritic ion channels play a critical role in shaping synaptic input and are fundamentally important for synaptic integration and plasticity. In the hippocampal region CA1, somato-dendritic gradients of AMPA receptors and the hyperpolarization-activated cation conductance (Ih) counteract the effects of dendritic filtering on the amplitude, time-course, and temporal integration of distal Schaffer collateral (SC) synaptic inputs within stratum radiatum (SR). While ion channel gradients in CA1 distal apical trunk dendrites within SR have been well characterized, little is known about the patterns of ion channel expression in the distal apical tuft dendrites within stratum lacunosum moleculare (SLM) that receive distinct input from the entorhinal cortex via perforant path (PP) axons. Here, we measured local ion channels densities within these distal apical tuft dendrites to determine if the somato-dendritic gradients of Ih and AMPA receptors extend into distal tuft dendrites. We also determined the densities of voltage-gated sodium channels and NMDA receptors. We found that the densities of AMPA receptors, Ih, and voltage-gated sodium channels are similar in tuft dendrites in SLM when compared with distal apical dendrites in SR, while the ratio of NMDA receptors to AMPA receptors increases in tuft dendrites relative to distal apical dendrites within SR. These data indicate that the somato-dendritic gradients of Ih and AMPA receptors in apical dendrites do not extend into the distal tuft, and the relative densities of voltage-gated sodium channels and NMDA receptors are poised to support nonlinear integration of correlated SC and PP input.  相似文献

Dendritic and axonal processes of nerve cells, along with the soma itself, have membranes with spatially distributed densities of ionic channels of various kinds. These ionic channels play a major role in characterizing the types of excitable responses expected of the cell type. These densities are usually represented as constant parameters in neural models because of the difficulty in experimentally estimating them. However, through microelectrode measurements and selective ion staining techniques, it is known that ion channels are non-uniformly spatially distributed. This paper presents a non-optimization approach to recovering a single spatially non-uniform ion density through use of temporal data that can be gotten from recording microelectrode measurements at the ends of a neural fiber segment of interest. The numerical approach is first applied to a linear cable model and a transformed version of the linear model that has closed-form solutions. Then the numerical method is shown to be applicable to non-linear nerve models by showing it can recover the potassium conductance in the Morris-Lecar model for barnacle muscle, and recover the spine density in a continuous dendritic spine model by Baer and Rinzel.  相似文献

The dendrites of CA1 pyramidal neurons in the hippocampus express numerous types of voltage-gated ion channel, but the distributions or densities of many of these channels are very non-uniform. Sodium channels in the dendrites are responsible for action potential (AP) propagation from the axon into the dendrites (back-propagation); calcium channels are responsible for local changes in dendritic calcium concentrations following back-propagating APs and synaptic potentials; and potassium channels help regulate overall dendritic excitability. Several lines of evidence are presented here to suggest that back-propagating APs, when coincident with excitatory synaptic input, can lead to the induction of either long-term depression (LTD) or long-term potentiation (LTP). The induction of LTD or LTP is correlated with the magnitude of the rise in intracellular calcium. When brief bursts of synaptic potentials are paired with postsynaptic APs in a theta-burst pairing paradigm, the induction of LTP is dependent on the invasion of the AP into the dendritic tree. The amplitude of the AP in the dendrites is dependent, in part, on the activity of a transient, A-type potassium channel that is expressed at high density in the dendrites and correlates with the induction of the LTP. Furthermore, during the expression phase of the LTP, there are local changes in dendritic excitability that may result from modulation of the functioning of this transient potassium channel. The results support the view that the active properties of dendrites play important roles in synaptic integration and synaptic plasticity of these neurons.  相似文献

在麻醉Wistar大鼠上,结合脑室给药,应用双电极刺激技术刺激海马独立的两条侧枝/联合纤维通路、TA通路,并在CA1区放射层记录兴奋性突触后电位(EPSP),对海马CA1区锥体细胞近、远端树突EPSP的空间整合进行了初步探讨。结果表明,海马CA1区锥体细胞近、远端树突的空间整合都是亚线性的;近端树突的空间整合不受期望值大小的影响,但远端树突的空间整合随期望值增加而减小(更趋于亚线性)。此外,荷包牡丹碱没有影响EPSP的空间整合;但瞬时A型钾通道(IAK^+)的拮抗剂氨基吡啶-4却使得近端树突的空间整合趋于线性发展。本研究表明,海马CA1锥体细胞近、远端树突不同的被动、主动特征使它们具有了不同的空间整合特性。由于近端树突接受海马内部侧枝/联合纤维投射的信息,远端树突通过TA通路接受内嗅皮层投射的信息,由此提示,CA1区锥体细胞对来自海马内部和直接来自皮层的信息输入采用了不同的整合方式。  相似文献

A longstanding hypothesis is that ion channels are present in the membranes of synaptic vesicles and might affect neurotransmitter release. Here we demonstrate that TRPM7, a member of the transient receptor potential (TRP) ion channel family, resides in the membrane of synaptic vesicles of sympathetic neurons, forms molecular complexes with the synaptic vesicle proteins synapsin I and synaptotagmin I, and directly interacts with synaptic vesicular snapin. In sympathetic neurons, changes in TRPM7 levels and channel activity alter acetylcholine release, as measured by EPSP amplitudes and decay times in postsynaptic neurons. TRPM7 affects EPSP quantal size, an intrinsic property of synaptic vesicle release. Targeted peptide interference of TRPM7's interaction with snapin affects the amplitudes and kinetics of postsynaptic EPSPs. Thus, vesicular TRPM7 channel activity is critical to neurotransmitter release in sympathetic neurons.  相似文献

During prolonged depolarization of excitable cells, some voltage-activated, tetrodotoxin-sensitive sodium channels are resistant to inactivation and can continue to open for long periods of time, generating a "persistent" sodium current ( I(NaP)). The amplitude of I(NaP) is small [generally less than 1% of the peak amplitude of the transient sodium current ( I(NaT))], activates at potentials close to the resting membrane potential, and is more sensitive to Na channel blocking drugs than I(NaT). It is thought that persistent Na channels are generated by a change in gating of transient Na channels, possibly because of a change in phosphorylation or protein structure, e.g. loss of the inactivation gate. Drugs that block Na channels can prevent the increase in [Ca(2+)](i) in cardiac cells during hypoxia. Hypoxia increases the amplitude of I(NaP). Paradoxically, NO causes a similar increase in I(NaP) and the effects of both can be inhibited by reducing agents such as dithiothreitol and reduced glutathione. It is proposed that an increased inflow of Na(+) during hypoxia increases [Na(+)](i), which in turn reverses the Na/Ca exchanger so that [Ca(2+)](i) rises. An increase in I(NaP) and [Ca(2+)](i) could cause arrhythmias and irreversible cell damage.  相似文献

Understanding single-neuron computations and encoding performed by spike-generation mechanisms of cortical neurons is one of the central challenges for cell electrophysiology and computational neuroscience. An established paradigm to study spike encoding in controlled conditions in vitro uses intracellular injection of a mixture of signals with fluctuating currents that mimic in vivo-like background activity. However this technique has two serious limitations: it uses current injection, while synaptic activation leads to changes of conductance, and current injection is technically most feasible in the soma, while the vast majority of synaptic inputs are located on the dendrites. Recent progress in optogenetics provides an opportunity to circumvent these limitations. Transgenic expression of light-activated ionic channels, such as Channelrhodopsin2 (ChR2), allows induction of controlled conductance changes even in thin distant dendrites. Here we show that photostimulation provides a useful extension of the tools to study neuronal encoding, but it has its own limitations. Optically induced fluctuating currents have a low cutoff (~70Hz), thus limiting the dynamic range of frequency response of cortical neurons. This leads to severe underestimation of the ability of neurons to phase-lock their firing to high frequency components of the input. This limitation could be worked around by using short (2 ms) light stimuli which produce membrane potential responses resembling EPSPs by their fast onset and prolonged decay kinetics. We show that combining application of short light stimuli to different parts of dendritic tree for mimicking distant EPSCs with somatic injection of fluctuating current that mimics fluctuations of membrane potential in vivo, allowed us to study fast encoding of artificial EPSPs photoinduced at different distances from the soma. We conclude that dendritic photostimulation of ChR2 with short light pulses provides a powerful tool to investigate population encoding of simulated synaptic potentials generated in dendrites at different distances from the soma.  相似文献

The persistent Na+ current, INaP, is known to amplify subthreshold oscillations and synaptic potentials, but its impact on action potential generation remains enigmatic. Using computational modeling, whole-cell recording, and dynamic clamp of CA1 hippocampal pyramidal cells in brain slices, we examined how INaP changes the transduction of excitatory current into action potentials. Model simulations predicted that INaP increases afterhyperpolarizations, and, although it increases excitability by reducing rheobase, INaP also reduces the gain in discharge frequency in response to depolarizing current (f/I gain). These predictions were experimentally confirmed by using dynamic clamp, thus circumventing the longstanding problem that INaP cannot be selectively blocked. Furthermore, we found that INaP increased firing regularity in response to sustained depolarization, although it decreased spike time precision in response to single evoked EPSPs. Finally, model simulations demonstrated that I(NaP) increased the relative refractory period and decreased interspike-interval variability under conditions resembling an active network in vivo.  相似文献

Recent studies have shown that the dendrites of several neurons are not simple translators but are crucial facilitators of excitatory postsynaptic potential (EPSP) propagation and summation of synaptic inputs to compensate for inherent voltage attenuation. Granule cells (GCs)are located at the gateway for valuable information arriving at the hippocampus from the entorhinal cortex. However, the underlying mechanisms of information integration along the dendrites of GCs in the hippocampus are still unclear. In this study, we investigated the input integration around dendritic branches of GCs in the rat hippocampus. We applied differential spatiotemporal stimulations to the dendrites using a high-speed glutamate-uncaging laser. Our results showed that when two sites close to and equidistant from a branching point were simultaneously stimulated, a nonlinear summation of EPSPs was observed at the soma. In addition, nonlinear summation (facilitation) depended on the stimulus location and was significantly blocked by the application of a voltage-dependent Ca2+ channel antagonist. These findings suggest that the nonlinear summation of EPSPs around the dendritic branches of hippocampal GCs is a result of voltage-dependent Ca2+ channel activation and may play a crucial role in the integration of input information.  相似文献

In addition to a fast activating and immediately inactivating inward sodium current, many types of excitable cells possess a noninactivating or slowly inactivating component: the persistent sodium current (I(NaP)). The I(NaP) is found in normal primary sensory neurons where it is mediated by tetrodotoxin-sensitive sodium channels. The dorsal root ganglion (DRG) is the gateway for ectopic impulses that originate in pathological pain signals from the periphery. However, the role of I(NaP) in DRG neurons remains unclear, particularly in neuropathic pain states. Using in vivo recordings from single medium- and large-diameter fibers isolated from the compressed DRG in Sprague-Dawley rats, we show that local application of riluzole, which blocks the I(NaP), also inhibits the spontaneous activity of A-type DRG neurons in a dose-dependent manner. Significantly, riluzole also abolished subthreshold membrane potential oscillations (SMPOs), although DRG neurons still responded to intracellular current injection with a single full-sized spike. In addition, the I(NaP) was enhanced in medium- and large-sized neurons of the compressed DRG, while bath-applied riluzole significantly inhibited the I(NaP) without affecting the transient sodium current (I(NaT)). Taken together, these results demonstrate for the first time that the I(NaP) blocker riluzole selectively inhibits I(NaP) and thereby blocks SMPOs and the ectopic spontaneous activity of injured A-type DRG neurons. This suggests that the I(NaP) of DRG neurons is a potential target for treating neuropathic pain at the peripheral level.  相似文献

Although the supralinear summation of synchronizing excitatory postsynaptic potentials (EPSPs) and backpropagating action potentials (APs) is important for spike-timing-dependent synaptic plasticity (STDP), the spatial conditions of the amplification in the divergent dendritic structure have yet to be analyzed. In the present study, we simulated the coincidence of APs with EPSPs at randomly determined synaptic sites of a morphologically reconstructed hippocampal CA1 pyramidal model neuron and clarified the spatial condition of the amplifying synapses. In the case of uniform conductance inputs, the amplifying synapses were localized in the middle apical dendrites and distal basal dendrites with small diameters, and the ratio of synapses was unexpectedly small: 8-16% in both apical and basal dendrites. This was because the appearance of strong amplification requires the coincidence of both APs of 3-30 mV and EPSPs of over 6 mV, both of which depend on the dendritic location of synaptic sites. We found that the localization of amplifying synapses depends on A-type K+ channel distribution because backpropagating APs depend on the A-type K+ channel distribution, and that the localizations of amplifying synapses were similar within a range of physiological synaptic conductances. We also quantified the spread of membrane amplification in dendrites, indicating that the neighboring synapses can also show the amplification. These findings allowed us to computationally illustrate the spatial localization of synapses for supralinear summation of APs and EPSPs within thin dendritic branches where patch clamp experiments cannot be easily conducted.  相似文献

The various properties of neuronal dendrites--their morphology, active membrane and synaptic properties--all play important roles in determining the functional capabilities of central nervous system neurons. Because of their fundamental involvement in both synaptic integration and synaptic plasticity, the active dendritic properties are important for both neuronal information processing and storage. The active properties of dendrites are determined by the densities of voltage-gated ion channels located within the dendrites in addition to the biophysical characteristics of those channels. The real power of this system resides in the level of plasticity that is provided by the many forms of channel modulation known to exist in neurons. Indeed, voltage gated ion channel modulation shapes the active properties of neuronal dendrites to specific conditions, thus tailoring the functional role of the single neuron within its circuit.  相似文献

The principal target of lateral geniculate nucleus in the cat visual cortex is the stellate neurons of layer 4. In previously reported work with intracellular recording and extracellular stimulation in slices of visual cortex, three general classes of fast excitatory synaptic potentials (EPSPs) in layer 4a spiny stellate neurons were identified. One of these classes, characterized by large and relatively invariant amplitudes (mean 1.7 mV, average coefficient of variation (CV) 0.083) were attributed to the action of geniculate axons because, unlike the other two classes, they could not be matched by intracortical inputs, using paired recording. We have examined in detail the properties of this synaptic input in twelve examples, selecting for study those EPSPs where there was secure extracellular stimulation of the single fibre input to a pair of stimuli 50 ms apart. In our analysis, we conclude that the depression that these inputs show to the second stimulus is entirely postsynaptic, since the evidence strongly suggests that the probability of transmitter release at the synaptic site(s) remains 1.0 for both stimuli. We argue that the most plausible explanation for this postsynaptic depression is a reduction in the average probability of opening the synaptic channels. Using a simple biochemical analysis (c.f. Sigworth plot), it is then possible to calculate the number of synaptic channels and their probability of opening, for each of the 12 connections. The EPSPs had a mean amplitude of 1.91 mV (+/- 1.3 mV SD) and a mean CV of 0.067 (+/- 0.022). The calculated number of channels ranged from 20 to 158 (59.4 +/- 48.7) and their probability of opening to the first EPSP had an average of 0.83 (+/- 0.09), with an average depression of the probability to 0.60 for the second EPSP. Geniculate afferents also terminate in layer 6. Intracellular recordings were also made in the upper part of this layer and a total of 51 EPSPs were recorded from pyramidal cells of three principal types. Amongst this dataset we sought EPSPs with similar properties to those characterized in layer 4a. Three examples were found, which is a much lower percentage (6%) than the incidence of putative geniculate EPSPs found in layer 4a (42%).  相似文献

HCN1 hyperpolarization-activated cation channels act as an inhibitory constraint of both spatial learning and synaptic integration and long-term plasticity in the distal dendrites of hippocampal CA1 pyramidal neurons. However, as HCN1 channels provide an excitatory current, the mechanism of their inhibitory action remains unclear. Here we report that HCN1 channels also constrain CA1 distal dendritic Ca2+ spikes, which have been implicated in the induction of LTP at distal excitatory synapses. Our experimental and computational results indicate that HCN1 channels provide both an active shunt conductance that decreases the temporal integration of distal EPSPs and a tonic depolarizing current that increases resting inactivation of T-type and N-type voltage-gated Ca2+ channels, which contribute to the Ca2+ spikes. This dual mechanism may provide a general means by which HCN channels regulate dendritic excitability.  相似文献

Stretching of the m. triceps surae (TS) of decerebrated cats evokes reflex shifts of the membrane potential (MP) in spinal motoneurons resulting from summation of EPSPs. We carried out model analysis of summation of a great number of EPSPs and compared the respective results with changes in the MP observed in real experiments using intrasomatic microelectrode recording. Simulation was based on the supposition of the proximity of the time course of an excitatory postsynaptic current to the positive part of the EPSP derivative. Transformation of EPSPs was performed using low-frequency filtration with two values of the time constant, 7 and 20 msec, (models M1 and M2, respectively). The models obtained provided sufficiently adequate reflection of the ascending phase of the real EPSP but inadequately reflected the rate of its decline and slow components of the MP changes. The disagreement of simulations with the real MP shifts shows that, most probably, final postsynaptic effects are to a considerable extent provided by summation of a great number of EPSPs generated in distal dendrites, and EPSPs immediately recorded in trasomatically cannot provide one with adequate information on the entire pattern of natural synaptic activation of the neuron. In addition, simulation analysis demonstrated a high probability of the contribution of active inhibitory processes to the formation of resulting MP changes under conditions of the stretch reflex.
