Oocytes of certain insects contain peculiar organelles termed accessory nuclei (AN). These organelles originate by budding off from the envelope of the oocyte nucleus and contain 1-2 dense inclusions immersed in a translucent ground substance. We have demonstrated that in the wasp Vespula germanica each inclusion consists of two elements: a spherical body, and a hemispherical structure composed of numerous 20-30 nm particles. Immunoelectron microscopy and whole-mount in situ hybridization have shown that the inclusions contain AgNOR-staining proteins, p80-coilin, Sm proteins, and small nuclear RNAs (snRNAs). These results indicate that the inclusions and hemispherical structures are homologous to Cajal bodies and B-snurposomes of Xenopus germinal vesicles, respectively. During previtellogenesis, AN (together with their Cajal bodies) migrate to the cortical ooplasm of the oocyte where they reside at least until the onset of embryogenesis. We suggest that AN are vehicles for the transport and localization of snRNPs to the periphery of the oocyte, i.e., to the region where the blastoderm of the embryo develops and where there is a requirement for a high concentration of RNA-processing factors.  相似文献

Light signals are perceived by multiple photoreceptors that converge to suppress the RING E3 ubiquitin ligase CONSTITUTIVE PHOTOMORPHOGENIC1 (COP1) for the regulation of stomatal development. Thus, COP1 is a point of integration between light signaling and stomatal patterning. However, how light signaling is collected into COP1 for the production and spacing of stomata is still unknown. Here, we report that the loss‐of‐function mutant of ANGUSTIFOLIA3 (AN3) delays asymmetric cell division, which leads to decreased stomatal index. Furthermore, overexpression of AN3 accelerates asymmetric cell division, which results in clusters of stomata. In addition, the stomatal development through AN3 regulation is mediated by light signaling. Finally, we find that an3 is a light‐signaling mutant, and that AN3 protein is light regulated. Self‐activation by AN3 contributes to the control of AN3 expression. Thus, AN3 is a point of collection between light signaling and stomatal patterning. Target‐gene analysis indicates that AN3 is associated with COP1 promoter for the regulation of light‐controlling stomatal development. Together, these components for regulating stomatal development form an AN3–COP1–E3 ubiquitin ligase complex, allowing the integration of light signaling into the production and spacing of stomata.  相似文献

Sound processing properties for calling song (CS) models, as described for the prothoracic L3 auditory neurone in Acheta domesticus, are investigated for the homologous auditory neurone 2 (AN2) in female Gryllus bimaculatus De Geer. AN2 of G. bimaculatus responds selectively to the syllable period (SP) of models of a male CS. The selectiveness of this response parallels the selectivity of phonotaxis females perform in response to the same SPs. Both, the responses of AN2 and female behaviour show clear interindividual variability. The SP‐selective responses of AN2 result from an SP‐dependent reduction in the spiking to subsequent syllables of the model CSs, measured as the percentage decrement. This SP‐dependent response does not primarily result from inbuilt properties of the AN2 membrane. Rather, it is dependent on inhibitory input to the AN2. However, clear inhibitory postsynaptic potentials in dendritic recordings of the AN2 are not encountered. This immediate response of AN2 to CSs is followed by an increased rate of tonic firing between stimulus CSs, which is termed the prolonged response, and is dependent on the carrier frequencies that make up the male CSs. With stimulation on the contralateral side of the soma of AN2s, more than 50% of AN2s exhibit a prolonged response. However, with stimulation from the ipsilateral side of the soma, most AN2s exhibit a prolonged response. The prolonged response of AN2 at 5 kHz may be even more sensitive than the immediate response. Thus, the AN2 neurone could provide a basis for phonotaxis that is selective for both the SPs and the carrier frequencies of potentially attractive calling songs.  相似文献

The 16S rRNA gene from the Thermococcus New Zealand isolate AN1 was cloned and sequenced. Analysis of the gene revealed the presence of signature sequences, indicating that strain AN1 represents a new species of the genus Thermococcus. Since the isolate AN1 differed from other thermococci in both its lower optimal NaCl concentration and generally lower optimal temperature for growth, in its unusual lipid membrane composition, and in its sensitivity to antibiotics, we propose that strain AN1 represents a new species of Thermococcus. We have carried out a quantitative study of the state of a neuropeptide Y (NPY)-ergic system of the hypothalamic arcuate nucleus (AN) in rats in the norm and in streptozotocin (STZ)-induced diabetes mellitus. The NPY-containing objects were identified using an indirect immunofluorescent technique; a system of digital analysis of images and a special software were used. Analysis of the morphometric parameters of the NPY-immunopositive objects within the AN sections allowed us to classify in an automatic mode such objects as neurons of various sizes, fibers, and terminals, as well as to count the number of objects of each class. In addition, the fluorescent intensity of the objects under study (directly proportional to the concentration and amounts of NPY in these objects) was determined. On the basis of these parameters, criteria of the state of synthesis and secretion of NPY and NPY-ergic innervation in the AN subnuclei were proposed. In control animals, the greatest number of NPY-immunopositive neurons and total content of NPY in the neurons and terminals were observed in the ventrolateral and dorsomedial subnuclei of the AN (vl- and dmAN, respectively); this fact points to the high intensities of NPY synthesis and secretion in these structures. The greatest content of NPY in the nerve terminals within the vlAN zone is indicative of the highest intensity of NPY-ergic afferent influences on this subnucleus. It is concluded that the approach we proposed is adequate enough for integral quantitative estimation of the state of peptidergic systems. The development of STZ-induced diabetes mellitus caused changes in the NPY-ergic system in different AN subnuclei; such changes demonstrated certain specificity. In particular, an increase in the number of immunopositive neurons and the total content of NPY in the neurons of vlAN and the ventromedial AN subnucleus (vmAN), which is evidence in favor of intensification of synthesis of the neuropeptide, was not accompanied by an adequate rise in the NPY content in the terminals located in the subnucleus under study. The content of NPY in the eminentia medianus also decreased. The above data allow us to hypothesize that the NPY-ergic system of the AN under conditions of diabetes mellitus is functionally insufficient.  相似文献

Summary Acrylamide was enzymatically produced from acrylonitrile (AN) in a two-stage packed bed reactor with immobilizedBrevibacterium sp. CH2 cells. Continuous production became possible because of higher tolerance of CH2 nitrile hydratase with respect to 6% AN than either the CH1 enzyme or theCorynebacterium enzyme of Nitto Chemical Industry which rapidly deactivated in a solution of 3% AN. The maximum volumetric productivity of 93.2 g/(L.h) was obtained when the reactor was fed with 6% AN at 4 °C, which compares that of 8.3 g/(L.h) with CH1 cells.  相似文献

黑果枸杞(Lycium ruthenicum)富含花青素,AN2基因是调控黑果枸杞花青素合成代谢的主效基因。为解析AN2基因启动子的活性差异,采用Tail-PCR方法分别克隆了黑果枸杞和红果枸杞(L. barbarum) AN2基因起始密码子上游约1 686 bp (LrAN2p)和1 495 bp (LbAN2p)的序列。Plant CARE预测表明,LbAN2pLrAN2p中分别有133和137个的顺式作用元件, 其中,参与光调控的顺式元件分别有11和15个;参与激素响应相关的顺式元件分别有13和16个。构建AN2启动子植物表达载体pKGWFS7:LbAN2p和pKGWFS7:LrAN2p,利用农杆菌介导的烟草遗传转化体系获得转基因烟草。GUS染色结果表明,LrAN2p能够驱动GUS在烟草中的表达,叶片呈现蓝色,具有较LbAN2p更强的启动活性,qRT-PCR结果表明,LrAN2p转基因烟草中GUS基因具有更高的转录水平,这可能会使AN2基因在黑果枸杞中具有更高的表达,激活黑果枸杞花青素合成代谢通路。这为解析枸杞果色形成及AN2基因的表达调控机制奠定了理论基础。  相似文献

Summary Ascending auditory interneurons of the cricket,Teleogryllus commodus (Walker), were investigated using simultaneous intracellular and extracellular recording in order to identify units which had previously been characterized only by extracellular recording. The morphology and physiology of the large adapting unit (LAU: Fig. 1) and of the small tonic unit (STU: Fig. 2) ofTeleogryllus correspond well to those of the ascending neuron 2 (AN2) and the ascending neuron 1 (AN1) ofGryllus (Figs. 1, 2), respectively.A summary of the ascending auditory interneurons described by various authors in 5 species of crickets is presented in order to establish common identities.Physiological evidence for direct connections between auditory afferents and the ascending auditory interneurons AN1 (STU) and AN2 (LAU) is presented. Twin studies suggest that genetic factors play a substantial role in anorexia nervosa (AN) and self‐induced vomiting (SV), a key symptom that is shared among different types of eating disorders (EDs). We investigated the association of 25 single nucleotide polymorphisms (SNPs), capturing 71–91% of the common variance in candidate genes, stathmin (STMN1), serotonin receptor 1D (HTR1D), tryptophan hydroxylase 2 (TPH2) and brain‐derived neurotrophic factor (BDNF), with AN and EDs characterized by regular SV. The first allele frequencies of all the SNPs were compared between a Dutch case group (182 AN, 149 EDs characterized by SV) and 607 controls. Associations rendering P‐values < 0.05 from this initial study were then tested for replication in a meta‐analysis with two additional independent ED case–control samples, together providing 887 AN cases, 306 cases with an ED characterized by SV and 1914 controls. A significant effect for the minor C‐allele of tryptophan hydroxylase 2 rs1473473 was observed for both AN [odds ratio (OR) = 1.30, 95% CI 1.08–1.57, P < 0.003] and EDs characterized by SV (OR = 1.52, 95% CI 1.28–2.04, P < 0.006). In the combined case group, a dominant effect was observed for rs1473473 (OR = 1.38, 95% CI 1.16–1.64, P < 0.0003). The meta‐analysis revealed that the tryptophan hydroxylase 2 polymorphism rs1473473 was associated with a higher risk for AN, EDs characterized by SV and for the combined group.  相似文献

以西藏东南部色季拉山海拔3950—4350 m为研究区,采用30×50 m网格采样法,以地统计学半变异函数为工具,研究了色季拉山森林生态系统沟谷与坡面上土壤氮素空间变异特征及模型。结果表明:土壤全氮、硝态氮和铵态氮含量均表现为0—10 cm10—20 cm,两个层次上空间变异性表现为全氮和铵态氮0—10 cm10—20 cm,而硝态氮表现为10—20 cm0—10cm;不同海拔高度土壤氮含量表现为随着海拔高度的升高而增加,但这种海拔梯度效应并未达显著水平(P0.05);沟谷区土壤氮含量高于坡面,这可能与植被残体在沟谷区的堆积分解促进氮循环有关;土壤全氮、铵态氮和硝态氮均具有中等程度的空间依赖性,其中土壤全氮空间变异符合指数模型,块金值/基台值为50%;土壤铵态氮和硝态氮含量空间变异分布均符合高斯模型,块金值/基台值分别为70.91%和37.45%;该区域土壤全氮、铵态氮和硝态氮含量空间依赖性表现为:硝态氮全氮铵态氮,即土壤硝态氮更易受到空间结构因素的影响,而铵态氮含量空间变化则主要受随机因素的影响。  相似文献

Bats that glean prey (capture them from surfaces) produce relatively inconspicuous echolocation calls compared to aerially foraging bats and could therefore be difficult predators to detect, even for insects with ultrasound sensitive ears. In the cricket Teleogryllus oceanicus, an auditory interneuron (AN2) responsive to ultrasound is known to elicit turning behaviour, but only when the cricket is in flight. Turning would not save a cricket from a gleaning bat so we tested the hypothesis that AN2 elicits more appropriate antipredator behaviours when crickets are on the ground. The echolocation calls of Nyctophilus geoffroyi, a sympatric gleaning bat, were broadcast to singing male and walking female T. oceanicus. Males did not cease singing and females did not pause walking more than usual in response to the bat calls up to intensities of 82 dB peSPL. Extracellular recordings from the cervical connective revealed that the echolocation calls elicited AN2 action potentials at high firing rates, indicating that the crickets could hear these stimuli. AN2 appears to elicit antipredator behaviour only in flight, and we discuss possible reasons for this context-dependent function.  相似文献

Accurate methods for assessing body composition in subjects with obesity and anorexia nervosa (AN) are important for determination of metabolic and cardiovascular risk factors and to monitor therapeutic interventions. The purpose of our study was to assess the accuracy of dual‐energy X‐ray absorptiometry (DXA) for measuring abdominal and thigh fat, and thigh muscle mass in premenopausal women with obesity, AN, and normal weight compared to computed tomography (CT). In addition, we wanted to assess the impact of hydration on DXA‐derived measures of body composition by using bioelectrical impedance analysis (BIA). We studied a total of 91 premenopausal women (34 obese, 39 with AN, and 18 lean controls). Our results demonstrate strong correlations between DXA‐ and CT‐derived body composition measurements in AN, obese, and lean controls (r = 0.77–0.95, P < 0.0001). After controlling for total body water (TBW), the correlation coefficients were comparable. DXA trunk fat correlated with CT visceral fat (r = 0.51–0.70, P < 0.0001). DXA underestimated trunk and thigh fat and overestimated thigh muscle mass and this error increased with increasing weight. Our study showed that DXA is a useful method for assessing body composition in premenopausal women within the phenotypic spectrum ranging from obesity to AN. However, it is important to recognize that DXA may not accurately assess body composition in markedly obese women. The level of hydration does not significantly affect most DXA body composition measurements, with the exceptions of thigh fat.  相似文献

The mechanism of toxicity of acrylonitrile (AN) has not been fully defined. The research described herein was undertaken to investigate the possible effects of AN on the levels of metallic elements in liver and brain of mice. Thirty-two mice were randomly assigned to four separate groups and treated intraperitoneal (i.p.) once daily for 1 week. Mice in the control group received normal saline, and mice in the three exposure groups received 5, 10, or 20 mg AN/kg b.w. Samples of brain and liver were collected immediately after decapitation. Tissue levels of trace elements (zinc, copper, iron) were determined with flame atomic absorption spectrophotometer or double channel atomic fluorescence absorption spectrophotometer (selenium). Mean brain weights of AN-treated mice were increased as a function of dose compared to controls, but there was no significant change in the ratio of liver/body weight in the four groups. While mean brain zinc decreased as a function of AN dosage, mean liver zinc of the low-dose group significantly increased (p < 0.05); mean liver copper in the medium-dose AN group was significantly higher compared to controls (p < 0.05); however, mean brain copper was increased, but the difference did not attain statistical significance in the three AN groups when compared with the controls (p > 0.05). Mean brain iron levels were significantly decreased in the middle-dose AN group (p < 0.05), but there were no consistent changes in liver iron. Tissue levels of selenium in brain and liver were similar for the control and AN treatment groups. AN induces significant and differential changes in the levels of zinc, copper, and iron in brain and liver. These changes likely play a pivotal role in mediating AN toxicity, most likely via changes in cellular redox status.  相似文献

Bacterial wilt, caused by soil-borne pathogen Ralstonia solanacearum, is a serious disease in many plants such as Solanaceae. To investigate the effects of accumulated nitrogen in soil on the phenotype and pathogenicity of the R. solanacearum, a serial passage experiment (SPE) was designed. Specifically speaking, minimal medium supplied with a slight excess of ammonium sulphate (AS) or ammonium nitrate (AN) was used to simulate the nutrition of soil containing excess nitrogen. During the period of 30 SPE, the phenotype, pathogenicity and relative expression of nitrogen metabolism genes in R. solanacearum were monitored. Phenotypic analysis results illustrated that the colony morphology of R. solanacearum changed after long-term culture, from high virulence colonies with strong fluidity to small, round non-mucoid colonies; The strain after prolonged stress of excessive exogenous nitrogen was a no-virulence phenotype conversion type (PC-type). The time for a change in colony morphology to occur after exposure to exogenous AS or AN was significantly less than the untreated samples, which treated without exogenous nitrogen. The results of pathogenicity also demonstrated that the cultures treated with exogenous AN or AS reduced virulence more quickly than the control. The disease index of 10 SPE with AN treatment or AS treatment was 89% or 68% lower than that of the control, respectively. In addition, as the incubation time increased, the swimming motility and the number of biofilms formation of the cultures were significantly changed under both treatments in comparison to the untreated samples. Furthermore, the relative expression of the nitric oxide reductase norB gene in the cultures treated with AN was 1.51-fold higher compared with the control after 30 SPE. These results indicated that excessive nitrogen supply in the environment could accelerate the transformation of R. solanacearum from high virulence wild-type into a PC-type, probably for the purpose of adapting to the adverse environment.  相似文献

The G- and C-banded karyotype ofHolochilus brasiliensis collected in central Brazil (2n=55, AN=56, acrocentric X and Y) can be regarded as the most representative of the ancestral form of the genus.H. magnus (2n=52, AN=58, acrocentric X and Y) andH. brasiliensis vulpinus (2n=40, AN=56), both living in southern Brazil, would be in different phases of chromosome number reduction due to centric fusions.H. magnus, in addition, shows an X of variable morphology and a metacentric Y. This fact, coupled with its distinct morphology and restricted distribution, suggests that it may represent a distinct trend in the genus' main evolutionary line.  相似文献

Bemisia tabaciis a complex of closely related genetic types of whiteflies, few of which are invasive. One of these, B biotype, has proven to be particularly adapted to invading new areas, but the underlying reasons as to why it has a well-developed capacity to invade is not known. To develop an understanding of factors that may be contributing to B’s invasive capacity, inter-biotype mating interactions and host plant suitability for the exotic B (B. tabaci Mediterranean/Asia Minor/Africa) and the indigenous Australian (AN) biotype (B. tabaci Australia) were examined. The results suggest that when confined to a mutually acceptable host, B cannot establish when the ratio of AN : B exceeds 20 : 1. However, when simultaneously provided with a host that only it prefers, B is able to establish even at 50 : 1 (AN : B). Further, when both biotypes occur together the number of progeny per female increases (relative to the number produced when only one biotype is present). The response is observed for both biotypes, but is considerably greater in the case of B. In addition, B performs better in the presence of the AN biotype B. tabaci Australia while AN perform worse in coexistence with B, but only if the demographics allow B to mate without significant interference. This leads to the prediction that B will invade in circumstances where its unique hosts are of sufficient number to escape the full negative impact of inter-biotype mating interactions and reduced competitiveness in terms of reproductive rate, while exposing the indigenous biotype to the full effects of the interaction.  相似文献

Objective: To test a newly developed dual energy X‐ray absorptiometry (DXA) method for abdominal fat depot quantification in subjects with anorexia nervosa (AN), normal weight, and obesity using CT as a gold standard. Design and Methods: 135 premenopausal women (overweight/obese: n = 89, normal‐weight: n = 27, AN: n = 19); abdominal visceral adipose tissue (VAT), subcutaneous adipose tissue (SAT), and total adipose tissue (TAT) areas determined on CT and DXA. Results: There were strong correlations between DXA and CT measurements of abdominal fat compartments in all groups with the strongest correlation coefficients in the normal‐weight and overweight/obese groups. Correlations of DXA and CT VAT measurements were strongest in the obese group and weakest in the AN group. DXA abdominal fat depots were higher in all groups compared to CT, with the largest % mean difference in the AN group and smallest in the obese group. Conclusion: A new DXA technique is able to assess abdominal fat compartments including VAT in premenopausal women across a large weight spectrum. However, DXA measurements of abdominal fat were higher than CT, and this percent bias was most pronounced in the AN subjects and decreased with increasing weight, suggesting that this technique may be more useful in obese individuals.  相似文献
