A combination of MICs of an N-chloramine, a simple chlorinated amino acid, and diazolidinyl urea gave synergistic activity against bacteria, but not fungi. The two compounds at a higher concentration, 0.1 and 0.3%, respectively, gave synergistic inhibition of fungi; kill times were 1 h for Trichophyton tonsurans, 3 h for Aspergillus niger and Fusarium moniliforme, and 6 h for Aspergillus fumigatus.  相似文献

Twelve strains of filamentous fungi, most of them belonging to the Deuteromycetes class, were isolated from activated sludge adapted to the delignification effluent from a nitrocellulose industry and screened to be used in the treatment of the effluent. The screening experiment was carried out using the effluent without co-substrate, treated for 120 h and pH 5. Aspergillus 2BNL1, Aspergillus 1AAL1 and Lentinus edodes UEC 2019 showed the highest effluent color reduction rates between 83% and 95%. The white-rot fungus L. edodes UEC 2019 was used as the control for the decolorization. In addition to color reduction, total phenol was also reduced in 56% and 79% by Aspergillus 2BNL1 and L. edodes UEC 2019, respectively. A kinetic experiment showed that Aspergillus 2BNL1 and Aspergillus 1AAL1 reduced the effluent color in the range of 81-95% at the first 24 h while L. edodes required 72 h to achieve a similar result. UV/Visible spectra revealed that all fungi treatments were able to decrease the chromophore compounds present in the effluent, except Aspergillus 1AAL1 that increased the UV absorptions. The molar weight distribution analysis showed that the three fungi were able to change the pattern of the effluent chromatogram, probably by degradation of the high molecular weight compounds.  相似文献

Fluorescein-labeled antiserum prepared with Aspergillus flavus strain CS was tested for specificity by staining fungi grown in soil in the vicinity of buried slides. All 14 strains of A. flavus fluoresced as intensely or nearly as intensely as the antigen control. Among 21 isolates of species of Aspergillus other than A. flavus, 17 reacted with moderate to low fluorescence at intensities readily distinguishable from that of A. flavus. The fluorescence of the remaining four cultures, and particularly A. sydowi, was indistinguishable from that of A. flavus. Fungi other than aspergilli were generally nonreactive. Interfering cross-reactions were encountered for one strain of Spicaria and one strain of Stemphylium; three isolates could not be evaluated because of interfering autofluorescence. An additional 22 isolates were either wholly negative or had a low order of fluorescence. Agglutination tests between each of the fungi and A. flavus CS serum revealed close agreement between agglutination titer and fluorescent-staining reaction. Unknown fungi freshly isolated from soil were checked for reaction to the A. flavus labeled antiserum; only one isolate gave a pronounced staining reaction, and that one proved to be a strain of A. flavus. In a simplified ecological model, the fluorescent-antibody technique was used to follow the development of A. flavus in mixed culture in soil with five other soil fungi.  相似文献

The effect of fungi–termite interaction on three rice varieties was conducted in a screen house at the Africa Rice Center (AfricaRice) Ibadan, Nigeria. Of the 10 fungi species (Fusarium verticilloides, Trichoderma sp., Aspergillus niger, Aspergillus flavus, Macrophoma sp., Neurospora sp., Botryodiplodia theobromae, Penicillum sp., Rhizopus sp. and Sclerotium rolfsii) isolated from termites, soil and rice plants, F. verticilloides, Trichoderma sp. and B. theobromae were used for the interaction study. Each fungus was inoculated singly and in combinations with termite into the root of each rice variety in potted soil. Leaf samples were taken to measure the chlorophyll content which is a major parameter to estimate effect of termite–fungi interaction. The chlorophyll content of the inoculated rice plants was significantly reduced when compared with the control. Fungi interaction with termite had significant reduction on the chlorophyll content. The synergistic relationship between the fungi and the termite was discussed.  相似文献

In this study, the effect of two boric acid concentrations (1% and 2%) and four derivates of tall oil with varying chemical composition were tested separately and in combination. The tall oil derivates were chosen in a way that they consist of different amounts of free fatty, resin acids and neutral compounds. Decay tests using two brown rot fungi (Postia placenta and Coniophora puteana) were performed on both unleached and leached test samples. Boric acid showed a low weight loss in test samples when exposed to fungal decay before leaching, but no effect after leaching. The tall oil derivates gave better efficacy against decay fungi compared to control, but are not within the range of the efficacy needed for a wood preservative. Double impregnation with boric acid and tall oil derivates gave synergistic effects for several of the double treatments both in unleached and leached samples. In the unleached samples the double treatment gave a better efficacy against decay fungi than tall oil alone. In leached samples a better efficacy against brown rot fungi were achieved than in samples with boron alone and a nearly similar or better efficacy than for tall oil alone. Boric acid at 2% concentration combined with the tall oil derivate consisting of 90% free resin acids (TO-III) showed the best performance against the two decay fungi with a weight loss less than 3% after a modified pure culture test.  相似文献

Summary Fungi inhabiting soil recently cleared of the native vegetation have been isolated and compared with those isolated from under established clover pastures, Three methods of isolation of the fungi were used in each situation, and profoundly influenced the population obtained.In newly cleared soils the composite fungal population pattern deduced all three isolation methods comprised species of Mortierella, Phoma, Trichoderma, Penicillium, Fusarium, Sclerotium and non-sporing mycelia. In soil supporting established clover pastures the main rhizosphere fungi were species of Fusarium, Aspergillus, Phoma, Curvularia, Macrophomina, Sclerotium and one Ascomycete.Fusarium oxysporum was the most frequently isolated fungus.Conventional plating techniques suggested that the dominant fungi in the rhizosphere of clover wereFusarium oxysporum andAspergillus versicolor, whereas isolation of fungi from hyphae adhering to the clover roots indicated that species of Macrophomina, Curvularia and an Ascomycete were the most abundantly occurring organisms. In the case of the root residues of the native vegetation in newly cleared soils plating techniques resulted in species of the freely sporing Mortierella, Trichoderma, Aspergillus, Fusarium and Penicillium occurring most frequently, whereas the isolation of fungi from hyphal strands on the residues gave a population pattern dominated by species of Mortierella, Phoma, Sclerotium, and non-sporing mycelia.The study indicated a marked succession of fungal species during the decomposition of the root debris in newly cleared soils, and also that some early members of this changing population might exert an adverse effect on the establishment of clover.  相似文献

Lipases in autolysed cultures of filamentous fungi   总被引:2,自引:0,他引:2  
Fifty-one fungi from different genera and strains were checked in plate to determine lipase activity in protein precipitates from their autolysed cultures. Each of them was then analysed at 3·5, 6·5 and 9·2 pH units and, as a consequence, basic lipases with high activity at 9·2 pH were found after 1 h of incubation. Only 25% of the studied fungi showed this lipase activity, among them the best producers were fungi from genus Fusarium (47% of fungi had lipase activity). In addition to lipase activity, Fusaria showed a low hydrolytic activity on cutin and suberin. The genus Aspergillus produced lipase and cutinase activity to a similar extent. Aspergillus nidulans 2544 also showed suberinase activity in a considerable amount. Penicillium species had very low activities. Other species and strains from genus Trichoderma , order Mucorales and class Basidiomycetes, did not show lipase activity in their degradative processes.  相似文献

Aspergillus nidulans, Neurospora crassa and Escherichia coli were grown on media containing a range of concentrations of nitrate, or ammonia, or urea, or l-glutamate, or l-glutamine as the sole source of nitrogen and the glutamate dehydrogenate and glutamine synthetase of the cells measured. 2. Aspergillus, Neurospora and Escherichia coli cells, grown on l-glutamate or on high concentrations of ammonia or on high concentrations of urea, possessed low glutamate dehydrogenase activity compared with cells grown on other nitrogen sources. 3. Aspergillus, Neurospora and Escherichia coli cells grown on l-glutamate possessed high glutamine synthetase activity compared with cells grown on other nitrogen sources. 4. The hypothesis is proposed that in Aspergillus, Neurospora and Escherichia colil-glutamate represses the synthesis of glutamate dehydrogenase and l-glutamine represses the synthesis of glutamine synthetase. 5. A comparison of the glutamine-synthesizing activity and the gamma-glutamyltransferase activity of glutamine synthetase in Aspergillus and Neurospora gave no indication that these fungi produce different forms of glutamine synthetase when grown on ammonia or l-glutamate as nitrogen sources.  相似文献

Five varieties ofPisum sativum L. were tested for seedborne fungi. Tests were conducted by standard International Seed Testing methods. It was observed that fungi likeAlternaria, Aspergillus, Rhizopus, Mucor andFusarium were associated with all the five varieties tested.Fusarium andRhizopus were dominant in all the varieties. They were also responsible for reduction in germination percentage of seeds. Early December was selected for testing the efficacy of certain fungicides (namely, Agrosan G.N., Ceresan, Copper carbonate, Tafasan, Tillex, Stardex and Sulphur) against seedborne fungi ofPisum sativum L., Agrosan G.N. and Ceresan gave the better control of seedborne pathogens without any adverse effect on germination.  相似文献

Culture filtrates of selected soil fungi, namely Aspergillus flavus, Aspergillus fumigatus, Aspergillus niger, Fusarium oxysporum, Penicillium vermiculatum and Rhizopus nigricans exhibited variable response to egg hatching and mortality of the root-knot nematode, Meloidogyne incognita. Higher concentrations of the culture filtrates of all the fungi inhibited egg hatching and proved to be toxic to the juveniles of M. incognita. In addition, development of the gall and multiplication of M. incognita were also found adversely affected in varying degrees on all the plants of Vigna radiata treated with the filtrates. The culture filtrate of A. niger showed highest toxicity to the nematode than those of any other fungus tested. Soil drench application of the culture filtrates gave better seedling growth and least nematode multiplication in comparison to seed soaking treatment.  相似文献

We describe a general, simple and inexpensive method for the isolation of DNA from filamentous fungi. Starting from freeze-dried mycelium 01–015% by weight can be isolated as high molecular weight DNA suitable for restriction and ligation in 2 h. The preparation can be done in Eppendorf tubes and allows the processing of many samples in parallel. We have used the method with the basidiomycetes Phanerochaete chrysosporium, Coprinus cinereus and the ascomycete Aspergillus nidulans and others have used it with Trichoderma reesei, Aspergillus niger and for the isolation of DNA from tomato plants.  相似文献

Street marijuana, commercial cigarettes and pipe tobaccos were studied for the presence of fungi and actinomycetes associated with hypersensitivity pneumonitis. Aspergillus species and thermophilic actinomycetes were isolated from the smoking materials. In addition, Aspergillus fumigatus spores were isolated from marijuana smoke, indicating the potential hazard involved in developing serious disease. Precipitin antibodies against fungi, particularly Aspergillus, showed a higher prevalence in marijuana smokers, whereas only very few cigarette smokers and nonsmokers demonstrated antibodies to fungi. Cigarette smokers and nonsmokers showed more or less similar prevelance of antibodies against thermophilic actinomycetes.  相似文献

This work used three fungi, Rhizopus oligosorus BCRC 31996, Monascus pilosus BCRC31527, and Aspergillus sp. BCRC31742, to produce glucosamine by using submerged fermentation and flask cultures. The reaction of glucosamine with 1-naphthyl isothiocyanate as derivatizing agent was carried out in pyridine at 50 degrees C for 1 h. The derivative was accurately analyzed and quantified by using high performance liquid chromatography. The relative standard deviation of glucosamine determined between experimental and real values were less than 2%. The kinetic and strategy of producing glucosamine in a flask culture was investigated to achieve an optimum yield of glucosamine under different conditions including three kinds of fungi, medium, and pH values. The descending ability of producing glucosamine for the three fungi was Aspergillus sp. BCRC31742 > Monascus pilosus BCRC31527 > Rhizopus oligosorus BCRC 31996 under the conditions studied. The experimental result shows that the glucosamine concentration had an optimum value and was 3430 mg/L by using Aspergillus sp. BCRC31742 culture in glucose and peptone (GP) medium, the yield of which was the best amount using wild-type microorganisms in the past. The generation culture of fungi and the pH control played important roles in enhancing the yield of glucosamine. The specific growth rate of the microorganism and the biomass, content, yield, and productivity of glucosamine were calculated as well.  相似文献

Degenerate primers corresponding to the consensus sequences of the copper-binding regions in the N-terminal domains of known basidiomycete laccases were used to isolate laccase gene-specific sequences from strains representing nine genera of wood rot fungi. All except three gave the expected PCR product of about 200 bp. Computer searches of the databases identified the sequence of each of the PCR products analyzed as a laccase gene sequence, suggesting the specificity of the primers. PCR products of the white rot fungi Ganoderma lucidum, Phlebia brevispora, and Trametes versicolor showed 65 to 74% nucleotide sequence similarity to each other; the similarity in deduced amino acid sequences was 83 to 91%. The PCR products of Lentinula edodes and Lentinus tigrinus, on the other hand, showed relatively low nucleotide and amino acid similarities (58 to 64 and 62 to 81%, respectively); however, these similarities were still much higher than when compared with the corresponding regions in the laccases of the ascomycete fungi Aspergillus nidulans and Neurospora crassa. A few of the white rot fungi, as well as Gloeophyllum trabeum, a brown rot fungus, gave a 144-bp PCR fragment which had a nucleotide sequence similarity of 60 to 71%. Demonstration of laccase activity in G. trabeum and several other brown rot fungi was of particular interest because these organisms were not previously shown to produce laccases.  相似文献

A novel series of thiazolidinone derivatives, namely 4-{4-dimethylamino-6-[4-oxo-2-phenyl-5-(4-pyridin-2-yl-piperazin-1-ylmethyl)-thiazolidin-3-yl]-[1,3,5]-triazin-2-yloxy}-1-methyl-1H-quinolin-2-ones, have been synthesized from the key intermediate 4-(4-amino-6-dimethylamino-[1,3,5]-triazin-2-yloxy)-1-methyl-1H-quinolin-2-one (5). Compound 5 was condensed with various aldehydes to give Schiff base derivatives, which after cyclization gave thiazolidinones that were linked with 1-pyridin-2-yl-piperazine to obtain the target compounds. The newly synthesized compounds were evaluated for their antimicrobial activity against eight bacteria (Staphylococcus aureus, Bacillus cereus, Escherichia coli, Pseudomonas aeruginosa, Klebsiella pneumoniae, Salmonella typhi, Proteus vulgaris, Shigella flexneri) and four fungi (Aspergillus niger, Candida albicans, Aspergillus fumigatus, Aspergillus clavatus).  相似文献

Cdc42/Rho GTPases are universally important regulators of cellular morphogenesis. Whereas their functions are well characterized in budding yeast, they are only beginning to be understood in filamentous fungi. The recent systematic analysis of Cdc42, Rac1 and Rho function in Aspergillus niger provides the first global perspective on their respective roles in hyphal morphogenesis. Surprisingly, the partitioning of these roles between Cdc42 and Rac1 seems to vary even among related fungi. These observations highlight the variable use of a common signalling module in filamentous fungi.  相似文献

Filamentous fungi have long been used for production of a range of valuable products; with the advent of molecular biology, it became apparent that these fungi possess considerable potential as expression hosts for the production of heterologous proteins and small molecules. Aspergillus is an important genus, including well known species of economically significant molds, and widely used for basic genetic research. The development of a genetic engineering "toolkit" for Aspergillus, such as those existing for the simpler yeasts and bacteria, was delayed due to the added complexity of the filamentous fungi, and also to the lesser resources devoted to their study. History of the development of Aspergillus as an expression host, current state of the art and future directions are reviewed, touching on related research in other fungi when discussing the areas of greatest potential for future biotechnological applications, focusing on the large and diverse families of fungal secondary metabolites.  相似文献
