Summary A number of culture conditions for protease production by Aspergillus oryzae NRRL 2160 on solid substrates were investigated. The pH of the medium and the substrate markedly affected protease production. High protease yield was obtained when the fungus was cultivated for 72–96 h on rice hulls: rice bran (7:3), at an initial pH of 7.0. Maximal protease production was achieved at an initial moisture content of 35–40%, corresponding to a water activity range of 0.982–0.986. Casein and gluten were effective inducers. Metarhizium anisopliae was grown by solid-state fermentation on a series of compositions of rice, rice bran, or rice husk media for the production of cyclodepsipeptides, destruxins A and B. The best yield for destruxin A and destruxin B were 2.9 mg kg–1 substrate and 227 mg kg–1 substrate, respectively, after 14 days cultivation with rice/bran/husk medium at 71% water content together with water activity of 0.921.  相似文献

The physiology of spore-negative and spore-positive root nodules was investigated inMyrica gale L. grown in water culture in a growth chamber. Spore(–) nodules were induced withFrankia cultures and spore(+) nodules with crushed nodules. Gas exchange was measured in a flow-through system.The time course of acetylene reduction following addition of acetylene was essentially the same in both spore(–) and spore(+) nodules with a stable maximum between 2 and 4 minutes followed by a steep decline to a minimum (37% of the maximum) between 9 and 30 minutes depending on the plant. The minimum was followed by a partial recovery. Nodule CO2 evolution showed a similar pattern but the minimum rate (83% of the maximum) was not nearly as low.Plants nodulated with one spore(–) and one spore(+) strain were compared at 6, 8 and 10 weeks after inoculation. At 6 weeks the spore(–) plants had 52% greater specific nitrogenase activity and 46% more biomass than the spore(+) plants. At 8 and 10 weeks, however, the differences between plants with spore(–) and spore(+) nodules became smaller.Plants nodulated with 4 spore(–) and 5 spore(+) strains were compared at 8 weeks after inoculation. Collectively the spore(–) plants exhibited a 32% greater specific nitrogenase activity, a 15% lower energy cost of nitrogenase activity (CO2/C2H4), and invested 31% less biomass in nodules than the spore(+) plants. The spore(–) plants also produced 16% more biomass indicating that spore(–) strains are generally more desirable than spore(+) strains. However, two spore(+) strains were as effective as the spore(–) strains.  相似文献

Thirty four samples of rice bran, of which 9 were from raw (untreated) rice (RR) and 25 from parboiled rice (PbR) were collected from commercial rice mills in and around Madras and analysed for storage mycoflora and mycotoxins. Fungi of the Aspergillus flavus group were present in 29 of the 34 samples (8 from RR and 21 from PbR) in quantities ranging from <1–432 thousand propagules/g, though not always as the dominant mycoflora. Fungal numbers were usually higher in RR than in PbR samples.Five of the 9 RR samples and 6 of the 25 PbR samples were positive for aflatoxins. Among 29 isolates of A. flavus obtained from the bran samples, 16 isolates –6 from RR bran and 10 from PbR bran — were found to be toxigenic in vitro. Some isolates of A. candidus also seemed to produce aflatoxin and other fluorescent substances.  相似文献

An obligatory alkalophilic Bacillus sp. P-2, which produced a thermostable alkaline protease was isolated by selective screening from water samples. Protease production at 30 °C in static conditions was highest (66 U/ml) when glucose (1% w/v) was used with combination of yeast extract and peptone (0.25% w/v, each), in the basal medium. Protease production by Bacillus sp. P-2 was suppressed up to 90% when inorganic nitrogen sources were supplemented in the production medium. Among the various agro-byproducts used in different growth systems (solid state, submerged fermentation and biphasic system), wheat bran was found to be the best in terms of maximum enhancement of protease yield as compared to rice bran and sunflower seed cake. The protease was optimally active at pH 9.6, retaining more than 80% of its activity in the pH range of 7–10. The optimum temperature for maximum protease activity was 90 °C. The enzyme was stable at 90 °C for more than 1h and retained 95 and 37% of its activity at 99 °C and 121 °C, respectively, after 1 h. The half-life of protease at 121 °C was 47 min.  相似文献

Verticillium lecanii has been recognized as an entomopathogen with high potential in biological control of pests. Two types of cultivation methods, the solid-state fermentation (SSF) and the liquid-state fermentation (LSF), were examined for V. lecanii. In SSF, the substrate types including rice, rice bran, rice husk, and the mixtures of these components were tested. The results showed that both cooked rice with appropriate water addition and rice bran gave significantly higher spore production of 1.5 2 109 spores/g substrate and 1.4 2 109 spores/g substrate, respectively. In LSF, SMAY liquid medium was used as a base, and the effects of environmental conditions on the spore production of V. lecanii were investigated. From the time course study, on the 9th day the spore yield reached 1.2 2 109 spores/ml of broth at 24v°C, 150 rpm for this strain. A series of medium volumes in the shaker-flask have been tested for the requirement of aeration. The largest surface aeration test, one tenth of the medium volume in the shaker-flask for cultivation, gave the highest spore count. The optimal pH value was tested and the initial pH 5 in the SMAY medium produced a high spore density. Finally, V. lecanii spores from SSF and LSF were different in size, shape, and size distribution; while mean spore length from SSF was 6.1 7m, and mean spore length from LSF was 5.0 7m.  相似文献

The entomopathogenic fungusVerticillium lecanii was cultivated in different solid-state cultures using wheat bran, sugar-beet pulp, and mixtures of both. The optimum value of water activity for growth and sporogenesis was 0.990–0.995. The use of water for medium moistening was more efficient in supporting spore formation than the use of dilute Czapek-Dox mineral medium. The yield of spores (per 1 g of the medium dry matter) was 3.0×109, 1.6×109, and 3.2×109 in the wheat bran medium, sugar-beet medium, and mixed medium, respectively. Conidial germination in soil extract (80×dilution) was independent of the spore origin and was about 43%.  相似文献

Major parameters affecting the production of chitinase by Beauveria felinaRD 101 under solid substrate fermentation conditions have been optimized. Wheat bran moistened with 100 MS-HCl medium adjusted to pH 5.0, inoculated with 1 × 1010 conidia g–1 initial dry bran and incubated at 28 °C for 6 days produced maximum chitinase activity of 6.34 U g–1 initial dry substrate.  相似文献

Summary Solid-state cultures of a pectinase-producing fungus (Aspergillus foetidus NRRL 341) were performed under different acidic conditions. Glass bottles containing 5 g of wheat bran and 7.5 mL of 0.2, 0.3, 0.4, or 0.5 N HCl were autoclaved (15 min, 121°C), inoculated with a spore suspension appropriately diluted to achieve an initial concentration of 4 × 104 spores per gram of wet substrate (with a 60 % humidity, on wet basis) and incubated at 30°C.Time course of pH and of different pectinase activities were determined in culture extracts. Total pectinase activity (TPA), expressed in terms of viscosimetric units per gram of wet substrate (VU.g–1), was affected by the initial culture acidity. The higher the HCl concentration used, the higher the TPA achieved, but after longer cultivation times. On the other hand, when 0.5 HCl was used, no fungal growth was observed. Nevertheless, enzyme productivity increased with culture acidity. When 0.4 HCl was used, TPA reached its maximum after 36 h of cultivation (2,535 VU.g–1). With 0.2 and 0.3 N HCl, TPA was the highest at 24 h (733 VU.g–1) and at 30 h (1,860 VU.g–1) respectively.The composition of the pectinase pool was also affected by culture acidity. The effect of inoculum level on xylitol production byCandida guilliermondii was evaluated in a rice straw hemicellulose hydrolysate. High initial cell density did not show a positive effect in this bioconversion since increasing the initial cell density from 0.67 g L–1 to 2.41 g L–1 decreased both the rate of xylose utilization and xylitol accumulation. The maximum xylitol yield (0.71 g g–1) and volumetric productivity (0.56 g L–1 h–1) were reached with an inoculum level of 0.9 g L–1. These results show that under appropriate inoculum conditions rice straw hemicellulose hydrolysate can be converted into xylitol by the yeastC. guilliermondii with efficiency values as high as 77% of the theoretical maximum.  相似文献

Fructosyl transferase (FTase) production by Aspergillus oryzae CFR 202 was carried out by solid-state fermentation (SSF), using various agricultural by-products like cereal bran, corn products, sugarcane bagasse,cassava bagasse (tippi) and by-products of coffee and tea processing. The FTase produced was used for the production of fructo-oligosaccharides (FOS), using 60% sucrose as substrate. Among the cereal bran used, rice bran and wheat bran were good substrates for FTase production by A. oryzae CFR 202. Among the various corn products used, corn germ supported maximum FTase production, whereas among the by-products of coffee and tea processing used, spent coffee and spent tea were good substrates, with supplementation of yeast extract and complete synthetic media. FTase had maximum activity at 60°C and pH 6.0. FTase was stable up to 40°C and in the pH range 5.0–7.0. Maximum FOS production was obtained with FTase after 8 h of reaction with 60% sucrose. FTase produced by SSF using wheat bran was purified 107-fold by ammonium sulphate precipitation (30–80%), DEAE cellulose chromatography and Sephadex G-200 chromatography. The molecular mass of the purified FTase was 116.3 kDa by SDS-PAGE. This study indicates the potential for the use of agricultural by-products for the efficient production of FTase enzyme by A. oryzae CFR 202 in SSF, thereby resulting in value addition of those by-products.  相似文献

Fourteen assays were conducted to study the influence of different variables, namely xylose concentration, inoculum level, agitation speed and nutrient supplementation, on xylitol biosynthesis by Candida guilliermondii FTI 20037. The maximum predicted values for xylitol yield (0.65 g g–1) and xylitol productivity (0.66 g l–1 h–1) can be attained with rice straw hydrolysate containing 60 g xylose l–1 without supplementation of ammonium sulfate, calcium chloride and rice bran extract, using 5 g inoculum l–1, at 250 rpm. The production of extracellular α-amylase by thermotolerant Bacillus subtilis was studied in solid state fermentation (SSF). The effect of wheat bran (WB) and rice husk (RH) was examined. The appropriate incubation period, moisture level, particle size and inoculum concentration was determined. Maximum yields of 159,520 and 21,760 U g−1 were achieved by employing WB and RH as substrates in 0.1 M phosphate buffer at pH 7 with 30% initial moisture content at 24 and 48 h. Particle size and inoculum concentration were found to be 1000 μm, 20% and 500 μm, 15% for WB and RH, respectively. Enzyme yield was 7.3-fold higher with WB medium compared with RH.  相似文献

Rice bran was superior to other proteinaceous substrates for protease production by Rhizopus oligosporus ACM 145F in solid-state fermentation. Maximum protease yield was after 72 h. The optimal initial moisture content was 47% (a w=0.97). Dried, ground and resuspended fermented rice was the most pratical and effective inoculum preparation, although, in the laboratory, spore suspensions prepared directly from agar slants were more convenient. Bjerkandera adusta produced aromatic compounds such as benzaldehyde (bitter almond aroma), benzyl alcohol and benzoic acid from L-phenylalanine (3 g kg–1). Two supports for the fungus, wheat bran (organic support) and Perlite (mineral support), gave optimal production with water contents of 66% and 60%, respectively. Benzyl alcohol (4.53 g kg–1) and benzaldehyde (1.56 g kg–1) were produced after 4 days on wheat bran respectively with 20 and 30 g L-phenylalanine kg–1. Aryl alcohol oxidase activity, which oxidises benzyl alcohol to benzaldehyde, was only detected when the fungus was grown on wheat bran, the support which promotes the most benzaldehyde production. Results are compared with those obtained in submerged liquid cultures.  相似文献

Summary A strain ofFusarium moniliforme, previously used for microbial protein production, excreted lactase (-D-galactosidase, EC.3.2.1 23) when cultivated either in a whey liquid medium or on a wheat bran solid medium. The enzyme produced in both media had pH and temperature optima of 4–5 and 50–60°C respectively and was particularly suitable for processing acid whey.In the whey culture, maximum lactase yield was observed after 95 h of growth at 30°C and whey lactose concentration of 9%. The addition of ammonium, potassium and sodium ions to the growth medium considerably enhanced lactase production. A maximum enzyme yield corresponding to hydrolysis of 3 nmoles o-nitrophenyl--D-galactopyranoside sec–1 ml–1 of growth medium, at pH 5 and 60°C, was obtained.In the wheat bran culture, the maximum enzyme yield was obtained after 140 h of growth at 28–30°C. A marked increase in the enzyme production was observed when nitrate or phosphate was added to the growth medium. Also, the addition of certain agricultural by-products (molasses, whey) enhanced lactase production. The observed maximum yield corresponding to the hydrolysis of 182 nmoles of ONPG sec–1 g–1 of wheat bran, at pH 5 and 60°C, is comparable to that reported for certain microorganisms used commercially for lactase production.  相似文献

Large aliquot water samples (30 ml) were enriched with nutrients, then gelled in the cold with Polycell (wallpapering paste) and incubated to derive Saprolegniaceae colonies and hence deduced spore counts, from water collections of about 1 litre. The Saprolegnia pathogen of fish was a component of the total recoveries; it was recognised on the basis of its secondary zoospore cyst ornamentation. Windermere water entering The Ferry House fish hatchery gave spore assays of 2 to 28 1–1 for the pathogen, a significant component of total Saprolegnia, which was 6–73 spores per litre.  相似文献

Decline in soil fertility accelerated by shorter fallow periods was expected to be a major constraint in slash-and-burn rice production systems in northern Laos. In this paper we describe relationships between fallow period, soil fertility parameters, weeds and rice yield. Soil infertility is not perceived a major yield constraint by the farmers. Of the various soil parameters observed only soil organic matter showed consistent association with rice yield (r=0.42, p<0.01). Fallow period and rice yield showed no association and the relationship between fallow and organic matter was very weak (r=0.16, p<0.01). Rice yield was negatively related to densities of Ageratum conyzoides and Lygodium flexuosum. Soil loss during the cropping period ranged from 300–29.300 kg ha–1. For the same period organic matter, total N, available P and available K content in the top 0–3 cm decreased by 11,12,17, and 17%, respectively, and loss of total N for the soil depth of 0–25 cm was estimated at 400 kg ha–1. Soil physical properties, moisture stress and available N are the most likely detriments to rice yields. Further attempts to relate soil properties to rice yield should include repeated measurements during the cropping season and observations on soil physical properties.The research presented was supported by the Provincial Agriculture Service, Luang Prabang, Laos, and the Swiss Development Cooperation.  相似文献

Long-term use of arsenic contaminated groundwater to irrigate crops, especially paddy rice (Oryza sativaL.) has resulted in elevated soil arsenic levels in Bangladesh. There is, therefore, concern regarding accumulation of arsenic in rice grown on these soils. A greenhouse pot experiment was conducted to evaluate the impact of arsenic-contaminated irrigation water on the growth and uptake of arsenic into rice grain, husk, straw and root. There were altogether 10 treatments which were a combination of five arsenate irrigation water concentrations (0–8 mg As l–1) and two soil phosphate amendments. Use of arsenate containing irrigation water reduced plant height, decreased rice yield and affected development of root growth. Arsenic concentrations in all plant parts increased with increasing arsenate concentration in irrigation water. However, arsenic concentration in rice grain did not exceed the maximum permissible limit of 1.0 mg As kg–1. Arsenic accumulation in rice straw at very high levels indicates that feeding cattle with such contaminated straw could be a direct threat for their health and also, indirectly, to human health via presumably contaminated bovine meat and milk. Phosphate application neither showed any significant difference in plant growth and development, nor in As concentrations in plant parts.  相似文献
