Arbuscular mycorrhizal fungal (AMF) spore communities were surveyed in a long-term field fertilization experiment in Switzerland, where different amounts of phosphorus (P) were applied to soil. Plots receiving no P as well as plots systematically fertilized in excess to plant needs for 31 years were used to test the hypothesis that application of P fertilizer changes the composition and diversity of AMF communities. AMF spores were isolated from the field soil, identified, and counted so as to quantify the effect of P fertilization on AMF spore density, composition, and diversity. Trap cultures were established from field soil with four host plants (sunflower, leek, maize, and Crotalaria grahamiana), and the spore communities were then analyzed in substrate samples from the pots. Altogether, nine AMF species were detected in the soil. No evidence has been acquired for effect of P fertilization on spore density, composition, and diversity of AMF in both the field soil and in trap cultures. On the other hand, we observed strong effect of crop plant species on spore densities in the soil, the values being lowest under rapeseed and highest under Phacelia tanacetifolia covercrop. The identity of plant species in trap pots also significantly affected composition and diversity of associated AMF communities, probably due to preferential establishment of symbiosis between certain plant and AMF species. AMF spore communities under mycorrhizal host plants (wheat and Phacelia in the fields and four host plant species in trap pots) were dominated by a single AMF species, Glomus intraradices. A wide range of methods are commonly used for preserving environmental samples prior to molecular analyses. However, the effect of these preservation methods on fungal DNA is not understood. The objective of this study was to test the effect of eight different preservation methods on the quality and yield of DNA extracted from Bromus inermis and Daucus carota roots colonized by the arbuscular mycorrhizal (AM) fungus, Glomus intraradices. The total DNA concentration in sample extracts was quantified using spectrophotometry. Samples that were frozen (− 80 ºC and − 20 ºC), stored in 95% ethanol, or silica gel dried yielded total (plant and fungal) DNA concentrations that were not significantly different from fresh samples. In contrast, samples stored in CTAB solution or freeze-dried resulted in significantly reduced DNA concentrations compared with fresh samples. The preservation methods had no effect on the purity of the sample extracts for both plant species. However, the DNA of the dried samples (silica gel dried, freeze-dried, heat dried) appeared to be slightly more degraded compared with samples that remained hydrated (frozen, stored in ethanol or CTAB solutions) during storage when visualized on a gel. The concentration of AM fungal DNA in sample extracts was quantified using TaqMan real time PCR. Methods that preserved samples in hydrated form had similar AM fungal DNA concentrations as fresh samples, except D. carota samples stored in ethanol. In contrast, preservation methods that involved drying the samples had very low concentrations of AM fungal DNA for B. inermis, and nearly undetectable for D. carota samples. The drying process appears to be a major factor in the degradation of AM fungal DNA while having less of an impact on plant DNA. Based on these results, samples that need to be preserved prior to molecular analysis of AM fungi should be kept frozen to minimize the degradation of plant and AM fungal DNA.  相似文献

Symbiotic associations between plants and arbuscular mycorrhizal (AM) fungi are ubiquitous in many herbaceous plant communities and can have large effects on these communities and ecosystem processes. The extent of species-specificity between these plant and fungal symbionts in nature is poorly known, yet reciprocal effects of the composition of plant and soil microbe communities is an important assumption of recent theoretical models of plant community structure. In grassland ecosystems, host plant species may have an important role in determining development and sporulation of AM fungi and patterns of fungal species composition and diversity. In this study, the effects of five different host plant species [Poa pratensis L., Sporobolus heterolepis (A. Gray) A. Gray, Panicum virgatum L., Baptisia bracteata Muhl. ex Ell., Solidago missouriensis Nutt.] on spore communities of AM fungi in tallgrass prairie were examined. Spore abundances and species composition of fungal communities of soil samples collected from patches within tallgrass prairie were significantly influenced by the host plant species that dominated the patch. The AM fungal spore community associated with B. bracteata showed the highest species diversity and the fungi associated with Pa. virgatum showed the lowest diversity. Results from sorghum trap cultures using soil collected from under different host plant species showed differential sporulations of AM fungal species. In addition, a greenhouse study was conducted in which different host plant species were grown in similar tallgrass prairie soil. After 4 months of growth, AM fungal species composition was significantly different beneath each host species. These results strongly suggest that AM fungi show some degree of host-specificity and are not randomly distributed in tallgrass prairie. The demonstration that host plant species composition influences AM fungal species composition provides support for current feedback models predicting strong regulatory effects of soil communities on plant community structure. Differential responses of AM fungi to host plant species may also play an important role in the regulation of species composition and diversity in AM fungal communities. 采用Illumina MiSeq高通量测序技术,研究江西鄱阳湖周边平原岗地的泡桐纯林及桐-药复合经营模式(泡桐-玉竹、泡桐-麦冬和泡桐-射干)下泡桐丛枝菌根真菌(arbuscular mycorrhizae fungi, AMF)群落结构特征。研究发现,泡桐AMF群落主要由球囊霉科、巨孢囊霉科、无梗囊霉科和多孢囊霉科组成,其中球囊霉科真菌占绝对优势,但不同科的相对丰度在不同经营模式下仍存在差异。与泡桐纯林相比,桐-药复合经营模式会降低泡桐菌根侵染率及AMF群落多样性。只有泡桐-射干经营模式中的泡桐含有多孢囊霉科真菌,且相对多度占2.73%。研究结果表明桐-药复合经营模式下中药材种类的差异会不同程度地改变泡桐AMF的群落结构。这为进一步研究桐-药复合经营模式下泡桐AMF的生态功能和资源利用提供了科学依据。  相似文献

On the basis of preliminary experiments, some Crotalaria species from Senegal were investigated to determine (1) their susceptibility to Meloidogyne javanica and M. incognita compared to a sensitive host (tomato), (2) their mycorrhizal and rhizobial responses, and (3) the effect of their cultivation on the mycorrhizal soil infectivity. The nematode invasion rates on Crotalaria spp. ranked from 0.17 to 7.17% and from 0.58 to 5.25%, respectively, for M. incognita and M. javanica, vs. 97% and 77% on tomato. Moreover, the inoculated J2 which invaded tomatoes developed into adult females, while those on Crotalaria spp. rarely developed beyond the third stage, confirming that all Crotalaria spp. evaluated are non hosts or poor hosts. In two other experiments, Crotalaria spp. were inoculated with an arbuscular mycorrhizal fungus (Glomus intraradices). Mycorrhization was generally well developed among Crotalaria species, and mycorrhizal colonization enhanced mainly phosphorus content of shoot tissues and always significant plant growth. Inoculation with both rhizobial isolates and Glomus intraradices enhanced growth and nodule formation on some Crotalaria species. The data recorded in both experiments showed, for the first time, that Crotalaria spp. are highly mycorrhiza dependent, some of them reaching more than 90% mycorrhizal dependency. Among Crotalaria species, twelve were used in two different experiments. A significant correlation was obtained between their mycorrhizal dependencies calculated on the shoot dry mass recorded in each experiment. Crotalaria spp. could be used as pre-crops for providing green manure while at the same time decreasing the level of detrimental nematodes and increasing the level of beneficial mycorrhizal fungi.  相似文献

We studied the production of xyloglucanase enzymes of pea and lettuce roots in the presence of saprobe and arbuscular mycorrhizal (AM) fungi. The AM fungus Glomus mosseae and the saprobe fungi Fusarium graminearum, Fusarium oxysporum-126, Trichoderma harzianum, Penicillium chrysogenum, Pleurotus ostreatus and Aspergillus niger were used. G. mosseae increased the shoot and root dry weight of pea but not of lettuce. Most of the saprobe fungi increased the level of mycorrhization of pea and lettuce, but only P. chrysogenum and T. harzianum inoculated together with G. mosseae increased the dry weight of pea and lettuce respectively. The AM and saprobe fungi increased the production of xyloglucanases by plant roots. The level of xyloglucanase activities and the number of xyloglucanolytic isozymes in plants inoculated with G. mosseae and most of the saprobe fungi tested were higher than when both microorganisms were inoculated separately. The possible relationship between xylogucanase activities and the ability of AM and saprobe fungi to improve the dry weight and AM root colonization of plants was discussed.  相似文献

While several recent studies have described changes in microbial communities associated with exotic plant invasion, how arbuscular mycorrhizal fungi (AMF) communities respond to exotic plant invasion is not well known, despite the salient role of this group in plant interactions. Here, we use molecular methods (terminal restriction fragment length polymorphism analyses based on the large subunit of the rRNA gene) to examine AMF community structure in sites dominated by the invasive mycorrhizal forb, Centaurea maculosa Lam. (spotted knapweed), and in adjacent native grassland sites. Our results indicate that significant AMF community alteration occurs following C. maculosa invasion. Moreover, a significant reduction in the number of restriction fragment sizes was found for samples collected in C. maculosa-dominated areas, suggesting reduced AMF diversity. Extraradical hyphal lengths exhibited a significant, on average 24%, reduction in C. maculosa-versus native grass-dominated sites. As both AMF community composition and abundance were altered by C.maculosa invasion, these data are strongly suggestive of potential impacts on AMF-mediated ecosystem processes. Given that the composition of AMF communities has the potential to differentially influence different plant species, our results may have important implications for site restoration after weed invasion.  相似文献

31P nuclear magnetic resonance (NMR) spectroscopy was used to study phosphate (P) metabolism in mycorrhizal and nonmycorrhizal roots of cucumber (Cucumis sativus L) and in external mycelium of the arbuscular mycorrhizal (AM) fungus Glomus intraradices Schenck & Smith. The in vivo NMR method allows biological systems to be studied non-invasively and non-destructively. 31P NMR experiments provide information about cytoplasmic and vacuolar pH, based on the pH-dependent chemical shifts of the signals arising from the inorganic P (Pi) located in the two compartments. Similarly, the resonances arising from α, β and γ phosphates of nucleoside triphosphates (NTP) and nucleoside diphosphates (NDP) supply knowledge about the metabolic activity and the energetic status of the tissue. In addition, the kinetic behaviour of P uptake and storage can be determined with this method. The 31P NMR spectra of excised AM fungi and mycorrhizal roots contained signals from polyphosphate (PolyP), which were absent in the spectra of nonmycorrhizal roots. This demonstrated that the Pi taken up by the fungus was transformed into PolyP with a short chain length. The spectra of excised AM fungi revealed only a small signal from the cytoplasmic Pi, suggesting a low cytoplasmic volume in this AM fungus. Arbuscular mycorrhizal fungi are able to alleviate the stress for plants caused by heavy metal contamination of soil. To analyze the molecular response of arbuscular mycorrhizal fungi to these pollutants, a subtractive cDNA library was constructed using RNA from Glomus intraradices extraradical hyphae of a root organ culture treated with a mixture of Cd, Zn, and Cu. Screening by reverse Northern blot analysis indicated that, among 308 clones, 17% correspond to genes up-regulated by heavy metals. Sequence analysis of part of the clones resulted, amongst others, in the identification of six genes putatively coding for glutathione S-transferases belonging to two different classes of these enzymes. Expression analyses indicated that the genes are differentially expressed during fungal development and that their RNA accumulation dramatically increases in extraradical hyphae grown in a heavy metal-containing solution.  相似文献

The water relations of arbuscular mycorrhizal plants have been compared often, but virtually nothing is known about the comparative water relations of mycorrhizal and nonmycorrhizal soils. Mycorrhizal symbiosis typically affects soil structure, and soil structure affects water retention properties; therefore, it seems likely that mycorrhizal symbiosis may affect soil water relations. We examined the water retention properties of a Sequatchie fine sandy loam subjected to three treatments: seven months of root growth by (1) nonmycorrhizal Vigna unguiculata given low phosphorus fertilization, (2) nonmycorrhizal Vigna unguiculata given high phosphorus fertilization, (3) Vigna unguiculata colonized by Glomus intraradices and given low phosphorus fertilization. Mycorrhization of soil had a slight but significant effect on the soil moisture characteristic curve. Once soil matric potential (m) began to decline, changes in m per unit change in soil water content were smaller in mycorrhizal than in the two nonmycorrhizal soils. Within the range of about –1 to –5 MPa, the mycorrhizal soil had to dry more than the nonmycorrhizal soils to reach the same m. Soil characteristic curves of nonmycorrhizal soils were similar, whether they contained roots of plants fed high or low phosphorus. The mycorrhizal soil had significantly more water stable aggregates and substantially higher extraradical hyphal densities than the nonmycorrhizal soils. Importantly, we were able to factor out the possibly confounding influence of differential root growth among mycorrhizal and nonmycorrhizal soils. Mycorrhizal symbiosis affected the soil moisture characteristic and soil structure, even though root mass, root length, root surface area and root volume densities were similar in mycorrhizal and nonmycorrhizal soils.  相似文献

桉树(Eucalyptus spp.)种植所产生的生态争议已受到广泛关注。丛枝菌根真菌(Arbuscular Mycorrhizal Fungi, AM真菌)是土壤微生物的重要组成部分,与根系共生后可促进植物的养分运输、提高植物逆境生存能力等。然而,桉树种植对土壤AM真菌群落结构和功能的影响尚不清楚。研究比较了次生林改种桉树后不同年限(2年/5年/10年)的土壤理化性质、AM真菌种类、群落组成及多样性的变化。结果表明,桉树种植显著改变了土壤理化性质,具体表现为2年生和10年生桉树林中土壤pH和有机碳显著降低(P0.05);AM真菌的孢子密度在桉树林土壤中显著低于次生林土壤(其中在5年生土壤中最低);AM真菌的种丰度随种植年份的增加逐渐下降,在10年生桉树林土壤中有所恢复; 5年生桉树的菌根侵染率最高,10年生桉树侵染率最低。AM真菌群落组成和结构发生显著变化,优势种Funneliforms geosporus的丰度在2年生和10年生林地中显著降低;而Septoglomus deserticola的丰度在2年生和10年生林地中显著增加。另外,冗余分析(Redundancy analysis, RDA)结果表明,土壤AM真菌群落结构主要受土壤pH值(解释率:89.88%)变化的影响。研究揭示了次生林改种桉树林后土壤AM真菌的群落变化特征,为桉树林的栽培管理和环境修复提供了数据支撑。  相似文献

Work on glomalin-related soil protein produced by arbuscular mycorrhizal (AM) fungi (AMF) has been limited because of the unknown identity of the protein. A protein band cross-reactive with the glomalin-specific antibody MAb32B11 from the AM fungus Glomus intraradices was partially sequenced using tandem liquid chromatography-mass spectrometry. A 17 amino acid sequence showing similarity to heat shock protein 60 (hsp 60) was obtained. Based on degenerate PCR, a full-length cDNA of 1773 bp length encoding the hsp 60 gene was isolated from a G. intraradices cDNA library. The ORF was predicted to encode a protein of 590 amino acids. The protein sequence had three N-terminal glycosylation sites and a string of GGM motifs at the C-terminal end. The GiHsp 60 ORF had three introns of 67, 76 and 131 bp length. The GiHsp 60 was expressed using an in vitro translation system, and the protein was purified using the 6xHis-tag system. A dot-blot assay on the purified protein showed that it was highly cross-reactive with the glomalin-specific antibody MAb32B11. The present work provides the first evidence for the identity of the glomalin protein in the model AMF G. intraradices, thus facilitating further characterization of this protein, which is of great interest in soil ecology.  相似文献

Arbuscular mycorrhizal fungi (AMF) are important symbionts of plants that improve plant nutrient acquisition and promote plant diversity. Although within-species genetic differences among AMF have been shown to differentially affect plant growth, very little is actually known about the degree of genetic diversity in AMF populations. This is largely because of difficulties in isolation and cultivation of the fungi in a clean system allowing reliable genotyping to be performed. A population of the arbuscular mycorrhizal fungus Glomus intraradices growing in an in vitro cultivation system was studied using newly developed simple sequence repeat (SSR), nuclear gene intron and mitochondrial ribosomal gene intron markers. The markers revealed a strong differentiation at the nuclear and mitochondrial level among isolates. Genotypes were nonrandomly distributed among four plots showing genetic subdivisions in the field. Meanwhile, identical genotypes were found in geographically distant locations. AMF genotypes showed significant preferences to different host plant species (Glycine max, Helianthus annuus and Allium porrum) used before the fungal in vitro culture establishment. Host plants in a field could provide a heterogeneous environment favouring certain genotypes. Such preferences may partly explain within-population patterns of genetic diversity.  相似文献

Controls on root colonization by arbuscular mycorrhizal fungi (AMF) include host nutrient status, identity of symbionts and soil physico-chemical properties. Here we show, in the field, that the subset of the AMF community colonizing the roots of a common grass species, Dactylis glomerata, was strongly controlled by neighboring roots of a different plant species, Centaurea maculosa, an invasive forb, thus adding a biological spatial component to controls on root colonization. Using an AMF-specific, 18s rDNA-based terminal restriction fragment length polymorphism (T-RFLP) analysis method, significant differences were found between AMF community fingerprints of samples derived from roots of grasses with (GCm) and without (G0) neighboring C. maculosa. There were also significant differences between samples derived from C. maculosa roots (Cmac) and both GCm and G0 roots. Sample ordination indicated three generally distinct groups consisting of Cmac, GCm and G0, with GCm samples being of intermediate distance between G0and Cmac. Our results indicate that, with the presence of C. maculosa, AMF communities of D. glomerata shift to reflect community composition associated with C. maculosa roots. These results highlight the importance of complex spatial distributions of AMF communities at the scale of a root system. An additional dimension to our study is that C. maculosa is an aggressively invasive plant in the intermountain West. Viewed in this light, these results suggest that pervasive influences of this plant on AMF communities, specifically in roots of its competitors, may represent a mechanism contributing to its invasive success. However, further work is clearly required to determine the extent to which AMF genotypic alteration by neighboring plants influences competitive relationships.  相似文献

Because arbuscular mycorrhizal fungal (AMF) species differ in stimulating the growth of particular host plant species, AMF species may vary in their effects on plant intra-specific competition and the self-thinning process. We tested this hypothesis using a microcosm experiment with Medicago sativa L. as a model plant population and four AMF species. Our results showed that the AMF species Glomus diaphanum stimulated host plant growth more than the other three AMF species did when the plants were grown individually. Glomus diaphanum also induced the highest rate of mortality in the self-thinning plant populations. We also found a positive correlation between mortality and growth response to colonization. Our results demonstrate that AMF species can affect plant mortality and the self-thinning process by affecting plant growth differently.  相似文献

Two indigenous arbuscular mycorrhizal (AM) fungi from the Mediterranean wine growing area in the Northeast of Spain were isolated and classified as Glomus intraradices Schenck & Smith. Both native fungi were found to increase the growth of the vine rootstock 110 Richter under greenhouse conditions compared with G. intraradices (BEG 72) and a phosphorus (P) fertilization treatment. The effectivity of field inoculation of Cabernet Sauvignon plants grafted on Richter 110 with the former native fungi and with G. intraradices BEG 72 in a replant vineyard severely infested by the root-rot fungus Armillaria mellea (Vahl ex Fr.) Kummer was assessed. The native fungi were not effective at enhancing plant development, and only G. intraradices BEG 72, resulted in a positive response. Field inoculation with this selected fungus increased plant shoot dry weight at the end of the first growing season.  相似文献

丛枝菌根真菌(AMF)在自然界分布广泛,能与大部分维管植物的根系形成菌根共生体。它们在调节植物群落结构和全球的碳、氮、磷循环等方面发挥着重要的生态功能,也是农林、环境领域最具应用前景的微生物类群。受限于培养方法、研究手段等,长期以来对AMF基因组、转录组特征的认识非常有限。最近10年,AMF基因组和转录组的相关研究在高通量测序技术的推动下取得了较快发展;研究结果也显著提高了对AMF遗传发育、代谢生理、共生机制等的认识。本文综述了目前已完成测序的AMF种类的基因组、转录组信息。结果发现,已测序的AMF种类普遍具有基因组大、转座子丰富、AT碱基含量高、含大量未知功能基因与特异性基因、缺少部分共生相关基因等特点。在转录层面,总结了不同AMF种类、AMF不同共生结构、共生阶段以及与不同寄主植物共生时的转录本特征。结果发现,不同种类AMF的转录本大小差异明显。不同共生阶段或不同共生结构中的AMF转录本也具有较大的差异,且差异表达的基因大部分与养分交易、信号转导等密切相关。相比之下,同种AMF与不同寄主植物共生时的转录本表现出较高的保守性。最后,本文提出了本领域需要重点关注的研究方向,包括AMF纯培养技术的革新、AMF基因功能的解析、非模式AMF类群的研究以及对AMF蛋白组的研究。  相似文献
