Two field experiments were conducted to examine the effect of soil solarization on the survival of arbuscular mycorrhizal (AM) fungi and root colonization of three crops. The experiments were carried out in a loamy sand soil (Rehovot) and a silty soil (Bet She'an Valley). For both experiments, assessment of indigenous AM fungal populations by the most probable number (MPN) method indicated that populations were reduced to zero after 2 or 4 weeks of solarization treatment. However, Glomus intraradices inoculum applied to the soil prior to solarization remained viable even after 8 weeks of solarization. After soil fumigation with methyl bromide both indigenous and applied AM fungi were nondetectable. Percentage root colonization by the indigenous AM fungal populations, together with plant-growth parameters, were assessed for three crops: onion and wheat (Rehovot), and carrot (Bet She'an). When sown on solarized field plots, onion and carrot seedlings showed a plant growth retardation, whereas wheat showed an increased growth response. Root colonization by indigenous AM fungi was not evident until 6 weeks after seedling emergence. Fumigation with methyl bromide reduced root colonization by indigenous AM populations, and reduced onion and wheat plant development at early growth stages. In a laboratory experiment, a temperature of 45° C for up to 24 h did not affect AM spore viability, indicating that temperatures reached during the solarization treatment cannot solely account for the reduced AM fungi viability in the field. Apparently, soil solarization temporarily delays root colonization by indigenous AM fungi until 6-8 weeks after plant emergence.

A field experiment was conducted to elucidate the survival of P. verrucosum in infested bulk soil(T1) and infested soil with waste grain (T2). The infested soil and reference soil (T3) was filled into steel cylinders, which were buried and sampled 13 times during a period from October 1994 to March 1996.The abundance of P. verrucosum and indigenous soil fungi were assessed by dilution plating on a selective and diagnostic medium (DYSG). Kernel infection was examined in T2. According to our results, P. verrucosum seems well adapted to survival in arable soil and little affected by indigenous fungi. During the first autumn and winter the grain caused a proliferation of P. verrucosum while its abundance in bulk soil was more constant except for a decrease in February 95, which is ascribed to frost/thaw alternations. In T2, P. verrucosum initially infected more than 50% of the kernels but during the first few months it was ousted by other fungi. A hypothesis regarding waste grain as the natural niche for the fungus in the field was therefore partly rejected. A gradual decrease in the abundance of P. verrucosum in soil during spring, a die-off in the dry summer and aproliferation during the second winter were found in both T1 and T2. Our results cannot provide the reason for the increase during the second winter. On an overall basis, however, they show that P. verrucosum can survive in the field, proliferate on soil organic matter and probably become an integral part of the soil ecosystem. This may constitute a risk of grain contamination when given appropriate environmental conditions. This revised version was published online in June 2006 with corrections to the Cover Date.

Effects of soil extracts from repeated plantation woodlands of Chinese-fir on soil fungi growth, the activities of microbial communities, and rates of net soil nitrogen mineralization were investigated. Soil extracts from replanted woodlands significantly inhibited soil non-pathogenic fungi growth, reduced soil respiration activities, and net soil nitrogen mineralization rates. However, soil extracts from replanted woodland increased the growth of pathogenic fungi. The combination of soil extracts and pathogenic fungi did not significantly reduce the growth of Chinese-fir seedlings when compared to the soil extracts alone. The combination of soil extracts with pathogenic and non-pathogenic fungi significantly increased the growth of Chinese-fir seedlings when compared to the combination of soil extracts and pathogenic fungi. The results suggest that the allelochemicals from soil extracts, rather than pathogenic fungi, are the key factor regulating the productivity and nitrogen cycling in repeated plantation woodlands.

We performed the first field-scale atrazine remediation study in the United States using chemically killed, recombinant organisms. This field study compared biostimulation methods for enhancing atrazine degradation with a novel bioaugmentation protocol using a killed and stabilized whole-cell suspension of recombinant Escherichia coli engineered to overproduce atrazine chlorohyrolase, AtzA. AtzA dechlorinates atrazine, producing non-toxic and non-phytotoxic hydroxyatrazine. Soil contaminated by an accidental spill of atrazine (up to 29 000 p.p.m.) supported significant populations of indigenous microorganisms capable of atrazine catabolism. Laboratory experiments indicated that supplementing soil with carbon inhibited atrazine biodegradation, but inorganic phosphate stimulated atrazine biodegradation. A subsequent field-scale study consisting of nine (0.75 m3) treatment plots was designed to test four treatment protocols in triplicate. Control plots contained moistened soil; biostimulation plots received 300 p.p.m. phosphate; bioaugmentation plots received 0.5% (w/w) killed, recombinant E. coli cells encapsulating AtzA; and combination plots received phosphate plus the enzyme-containing cells. After 8 weeks, atrazine levels declined 52% in plots containing killed recombinant E. coli cells, and 77% in combination plots. In contrast, atrazine levels in control and biostimulation plots did not decline significantly. These data indicate that genetically engineered bacteria overexpressing catabolic genes significantly increased degradation in this soil heavily contaminated with atrazine.

Ultramafic soils at Bandalup Hill (Western Australia) are characterised by high concentrations of Ni and low levels of P. Amongst the plant species that can sustain such hostile conditions, Hakea verrucosa F. Muell from a non-mycorrhizal family (Proteaceae) would be expected to rely on cluster roots to access P. However, the acidification of ultramafic soils by cluster roots might increase the dissolution of soil Ni, and therefore its availability to plants. Symbiosis with mycorrhizal fungi, on the other hand, might help to reduce the uptake of Ni by H. verrucosa. Therefore, the aim of this study was to investigate the mycorrhizal status of H. verrucosa, and assess any contribution from mycorrhizal fungi to its growth and nutrient status. Seedlings of H. verrucosa were first grown in undisturbed ultramafic soil cores from Bandalup Hill for 8 weeks to assess the presence of mycorrhizal fungi in their roots. In a second experiment, H. verrucosa seedlings were grown in the same ultramafic soil that was either steamed or left untreated. Seedlings were inoculated with an arbuscular mycorrhizal (AM) fungal consortium from Bandalup Hill. Fungal hyphae, vesicles, as well as intracellular arbuscules and hyphal coils were observed in the cluster roots of H. verrucosa in both experiments. In the first experiment, 57% of the root length was colonized by AM fungi. Seedlings had high (between 1.4 and 1.9) shoot to root ratios and their roots had very few root hairs, despite growing in P-deficient soil. Steaming of the ultramafic soil increased the growth of seedlings and their nutrient uptake. Inoculation with AM fungi reduced the seedling growth in steamed ultramafic soil; however, it increased their shoot P and K concentration and also the shoot K content. The shoot Ni concentration of seedlings was not affected by the presence of AM fungi.

Cryptic belowground organisms are difficult to observe and their responses to global changes are not well understood. Nevertheless, there is reason to believe that interactions among above- and belowground communities may mediate ecosystem responses to global change. We used grassland mesocosms to manipulate the abundance of one important group of soil organisms, arbuscular mycorrhizal (AM) fungi, and to study community and ecosystem responses to CO2 and N enrichment. Responses of plants, AM fungi, phospholipid fatty acids and community-level physiological profiles were measured after two growing seasons. Ecosystem responses were examined by measuring net primary production (NPP), evapotranspiration, total soil organic matter (SOM), and extractable mineral N. Structural equation modeling was used to examine the causal relationships among treatments and response variables. We found that while CO2 and N tended to directly impact ecosystem functions (evapotranspiration and NPP, respectively), AM fungi indirectly impacted ecosystem functions by influencing the community composition of plants and other root fungi, soil fungi and soil bacteria. We found that the mycotrophic status of the dominant plant species in the mesocosms determined whether the presence of AM fungi increased or decreased NPP. Mycotrophic grasses dominated the mesocosm communities during the first growing season, and the mycorrhizal treatments had the highest NPP. In contrast, nonmycotrophic forbs were dominant during the second growing season and the mycorrhizal treatments had the lowest NPP. The composition of the plant community strongly influenced soil N, and the community composition of soil organisms strongly influenced SOM accumulation in the mesocosms. These results show how linkages between above- and belowground communities can determine ecosystem responses to global change.

Soil solarization, the process of heating soil by covering fields with clear plastic, is a promising method to reduce populations of soilborne pests and weeds without the use of pesticides. However, the destruction of beneficial organisms such as arbuscular mycorrhizal (AM) fungi also may occur, thereby reducing positive effects of solarization. We compared the effects of solarization and chemical fumigants on the survival of indigenous AM fungi in 1995 and 1996. The infectivity of AM fungi was monitored before and after solarization using a greenhouse bioassay with Sorghum bicolor L. for both years. AM colonization of roots was also monitored in the field 8 months after solarization in 1995. Weed densities were measured 8 months after treatment in 1996. Solarization increased the average daily soil temperature 6-10°C and the maximum soil temperature reached by 10-16°C (5-20 cm depth). Solarization did not reduce the infectivity of AM fungi immediately after the solarization period in either year, as determined by the greenhouse bioassay. Infectivity was greatly reduced in solarized plots 8 months after solarization (over winter) in both years as assessed in the field (1995) or with the greenhouse bioassay (1996). Fumigation with metam sodium at 930 l ha-1 (350 kg active ingredient ha-1) reduced the infectivity of AM fungi in both years, and fumigation with methyl bromide at 800 kg ha-1 eliminated infection by AM fungi. Solarization was as effective as methyl bromide and metam sodium at 930 l ha-1 in controlling winter annual weeds measured 8 months after treatment. Solarization apparently reduced AM fungi in soil indirectly by reducing weed populations that maintained infective propagules over the winter. Fumigation with metam sodium or methyl bromide directly reduced AM fungi in soil.

Summary

The identity of a fairy-club fungus commonly associated with glasshouse grown conifers belonging to families other than the Pinaceae was confirmed as Clavaria acuta. In studying the relationship between this fungus and plants with which it grows, endogonaceous fungi were found to be associated with root nodules formed on conifers over a wider range than previously reported.

American chestnut (Castanea dentata) was once a dominant overstory tree in eastern USA but was decimated by chestnut blight (Cryphonectria parasitica). Blight-resistant chestnut is being developed as part of a concerted restoration effort to bring this heritage tree back. Here, we evaluate the potential of field soils in the northern portion of the chestnut's former range to provide ectomycorrhizal (EM) fungus inoculum for American chestnut. In our first study, chestnut seedlings were grown in a growth chamber using soil collected from three sites dominated by red oak (Quercus rubra) as inoculum and harvested after 5 months. Of the 14 EM fungi recovered on these seedlings, four species dominated in soils from all three sites: Laccaria laccata, a Tuber sp., Cenococcum geophilum, and a thelephoroid type. Seedlings grown in the nonsterilized soils were smaller than those growing in sterilized soils. In the second study, chestnut seedlings were grown from seed planted directly into soils at the same three sites. Seedlings with intermingling roots of established trees of various species were harvested after 5 months. Seventy-one EM fungi were found on the root tips of the hosts, with 38 occurring on chestnut seedlings. Multiple versus single host EM fungi were significantly more abundant and frequently encountered. The fungi observed dominating on seedlings in the laboratory bioassay were not frequently encountered in the field bioassay, suggesting that they may not have been active in mycelial networks in the field setting but were in the soils as resistant propagules that became active in the bioassay. These results show that soil from red oak stands can be used to inoculate American chestnut with locally adapted ectomycorrhizal fungi prior to outplanting, a relatively cost effective approach for restoration efforts.

The hypothesis that inoculation of transplants with vesicular-arbuscular mycorrhizal (VAM) fungi before planting into saline soils alleviates salt effects on growth and yield was tested on lettuce (Lactuca sativa L.) and onion (Allium cepa L.). A second hypothesis was that fungi isolated from saline soil are more effective in counteracting salt effects than those from nonsaline soil. VAM fungi from high- and low-salt soils were trap-cultured, their propagules quantified and adjusted to a like number, and added to a pasteurized soil mix in which seedlings were grown for 3–4 weeks. Once the seedlings were colonized by VAM fungi, they were transplanted into salinized (NaCl) soil. Preinoculated lettuce transplants grown for 11 weeks in the saline soils had greater shoot mass compared with nonVAM plants at all salt levels [2 (control), 4, 8 and 12 dS m–1] tested. Leaves of VAM lettuce at the highest salt level were significantly greener (more chlorophyll) than those of the nonVAM lettuce. NonVAM onions were stunted due to P deficiency in the soil, but inoculation with VAM fungi alleviated P deficiency and salinity effects; VAM onions were significantly larger at all salt levels than nonVAM onions. In a separate experiment, addition of P to salinized soil reduced the salt stress effect on nonVAM onions but to a lesser extent than by VAM inoculation. VAM fungi from the saline soil were not more effective in reducing growth inhibition by salt than those from the nonsaline site. Colonization of roots and length of soil hyphae produced by the VAM fungi decreased with increasing soil salt concentration. Results indicate that preinoculation of transplants with VAM fungi can help alleviate deleterious effects of saline soils on crop yield.

Soil crusts of rosemary scrubs in south-central Florida were examined for effects on seed germination of four herbs that are killed by fire and must recruit from seed: Eryngium cuneifolium (Apiaceae), Hypericum cumulicola (Hypericaceae), Polygonella basiramia (Polygonaceae), and Paronychia chartacea ssp. chartacea (Caryophyllaceae). Biological soil crusts in these sites are dominated by algae, cyanobacteria, fungi, and bacteria. Because crusts can change soil stability, water, and nutrients, they can affect seed germination. A series of greenhouse and field experiments were designed to first examine the effects of crusts in isolation and then to determine their role in the context of other environmental factors – time since fire and distance to the dominant shrub in this system, Ceratiola ericoides. In the greenhouse experiment, germination in autoclaved crusts was dramatically reduced relative to germination in living crusts for all but P. basiramia. In four field experiments where crusts were left intact, disturbed (mechanically or by flaming), or completely removed, the effects of crusts were variable and species-specific, but were significant enough to impact aboveground population sizes. More germination was consistently observed in recently burned sites away from C. ericoides shrubs. Overall rates of germination were generally very low during this study, possibly as a result of seasonal droughts that could have reduced germination, increased seed dormancy, and/or decreased seed viability. The importance of water for germination was confirmed in an experiment with two watering regimes and three crust treatments designed to create a gradient of soil water availability. Germination was significantly greater in the high water treatment and unaffected by soil crust moisture. Dry years are not uncommon in scrub and the results of this study help us to understand how scrub herbs fare during drought and what role biological soil crusts play in germination.

Arbuscular mycorrhizal (AM) fungi occur in all agricultural soils but it is not easy to assess the contribution they make to plant growth under field conditions. Several approaches have been used to investigate this, including the comparison of plant growth in the presence or absence of naturally occurring AM fungi following soil fumigation or application of fungicides. However, treatments such as these may change soil characteristics other than factors directly involving AM fungi and lead to difficulties in identifying the reason for changes in plant growth. In a glasshouse experiment, we assessed the contribution of indigenous AM fungi to growth of subterranean clover in undisturbed cores of soil from two agricultural field sites (a cropped agricultural field at South Carrabin and a low input pasture at Westdale). We used the approach of estimating the benefit of AM fungi by comparing the curvature coefficients ( C) of the Mitscherlich equation for subterranean clover grown in untreated field soil, in field soil into which inoculum of Glomus invermaium was added and in soil fumigated with methyl bromide. It was only possible to estimate the benefit of mycorrhizas using this approach for one soil (Westdale) because it was the only soil for which a Mitscherlich response to the application of a range of P levels was obtained. The mycorrhizal benefit ( C of mycorrhizal vs. non-mycorrhizal plants or C of inoculated vs. uninoculated plants) of the indigenous fungi corresponded with a requirement for phosphate by plants that were colonised by AM fungi already present in the soil equivalent to half that required by non-mycorrhizal plants. This benefit was independent of the plant-available P in the soil. There was no additional benefit of inoculation on plant growth other than that due to increased P uptake. Indigenous AM fungi were present in both soils and colonised a high proportion of roots in both soils. There was a higher diversity of morphotypes of mycorrhizal fungi in roots of plants grown in the Westdale soil than in the South Carrabin soil that had a history of high phosphate fertilizer use in the field. Inoculation with G. invermaium did not increase the level of colonisation of roots by mycorrhizal fungi in either soil, but it replaced approximately 20% of the root length colonised by the indigenous fungi in Westdale soil at all levels of applied P. The proportion of colonised root length replaced by G. invermaium in South Carrabin soil varied with the level of application of P to the soil; it was higher at intermediate levels of recently added soil P.

Eight biocides were chosen to determine whether they had any effects on nontarget organisms in soil and to what extent they would reduce their target populations under laboratory experimental conditions. A simplified microcosm system was utilized in which reduced species arrays that included field populations of either only bacteria and fungi, or bacteria, fungi, and protozoa (no nematodes, arthropods, or plants) were inoculated into sterilized soil. In a second set of experiments, plants were grown in sterilized soil. A bactericide-streptomycin-four fungicides-cycloheximide, Fungizone (amphotericin B), captan, and PCNB (quintozene)-an acaricide-cygon-an insecticide-nematicide-carbofuran-and an insecticide-diazinon-were used. Each biocide had effects on nontarget organisms although the increases or decreases, with respect to the control, were of only limited duration. Reductions in target groups were typically of longer duration. Streptomycin, applied at 1 mg·g–1 soil, did not decrease bacterial populations during the experimental incubation. At 3 mg·g–1 soil, streptomycin decreased the numbers of bacteria that grew on tryptone agar, but also reduced active hyphae. Fungizone was the most effective of the 4 fungicides tested in reducing active hyphae. Increased bacterial populations were usually observed following fungal reductions. Carbofuran had the fewest effects on the test organisms (bacteria, fungi, and protozoa). Only an initial stimulation of bacterial and fungal populations was observed with cygon although it also increased NH4 +-N concentrations in soil during most of the incubation, as did streptomycin and cycloheximide. A transitory increase in fungal populations following a decrease in ciliate numbers was observed in the cygon with grazers treatments. Diazinon reduced all microbial populations and inorganic nitrogen concentrations measured. Cygon and PCNB decreased growth of blue grama plants, while streptomycin reduced shoot weights of blue grama. These results should be useful in assessing the effects of these biocides when applied to more complex systems.

Effects of long-term mineral fertilization and manuring on the biomass of arbuscular mycorrhizal fungi (AMF) were studied in a field experiment. Mineral fertilization reduced the growth of AMF, as estimated using both measurements of hyphal length and the signature fatty acid 16:1ω5, whereas manuring alone increased the growth of AMF. The results of AMF root colonization followed the same pattern as AMF hyphal length in soil samples, but not AMF spore densities, which increased with increasing mineral and organic fertilization. AMF spore counts and concentration of 16:1ω5 in soil did not correlate positively, suggesting that a significant portion of spores found in soil samples was dead. AMF hyphal length was not correlated with whole cell fatty acid (WCFA) 18:2ω6,9 levels, a biomarker of saprotrophic fungi, indicating that visual measurements of the AMF mycelium were not distorted by erroneous involvement of hyphae of saprotrophs. Our observations indicate that the measurement of WCFAs in soil is a useful research tool for providing information in the characterization of soil microflora.

Four endophytic fungi (Fusarium spp.) isolated from the cortical tissue of surface-sterilised banana as well as from tomato roots were tested for their capacity of biological control towards the burrowing nematode Radopholus similis on banana. The pathogenic and parasitic capacities of endophytic fungi towards R. similis were tested in in vitro experiments. No parasitism of fungi on R. similis was observed. However, nematode activity decreased significantly in the presence of all endophytic fungi in vitro when compared to nematodes in the absence of fungi. The effects of fungi on R. similis activities in the soil were tested in the absence of plants. Nematode activities were reduced significantly by 16-30% by endophytic fungi when compared to untreated soil.

Imazaquin, imazethapyr and pendimethalin showed high toxicity to sorghum plants grown in a greenhouse soil mix. However, mycorrhizal sorghum plants were less affected by herbicide toxicity than non-mycorrhizal ones, at low to moderate herbicide concentrations. VAM herbicide safening effects were more evident on imazaquin-treated plants, than for those treated with the other two herbicides. Applications of imazethapyr and pendimethalin at the two highest concentrations, but not imazaquin, reduced VAM colonization rates in sorghum. Applications of the VAM stimulating isoflavonoids, biochanin A and formononetin, at 5 ppm solutions to a field soil sample containing toxic levels of imazaquin (13 ppb) and indigenous VAM fungi, reduced herbicide-induced injury in corn and sorghum under growth chamber conditions. The benefits of isoflavonoids were reduced when additional propagules of Glomus intraradix were added into field-soil samples, and were eliminated when VAM fungi were inactivated by autoclaving. This indicates that herbicide safening effects of biochanin A, and formononetin are VAM-mediated and also suggests the potential use of these isoflavonoids as herbicide safeners.

Seven commercial 3- to 7-ring (R) polycyclic aromatic hydrocarbons (PAH) as well as PAH derived from lignite tar were spiked into 3 soils (0.8 to 9.7% of organic carbon). The disappearance of the original PAH was determined for the freshly spiked soils, for soils incubated for up to 287 d with their indigenous microflora, and for autoclaved, unsterile and pasteurized soils inoculated with basidiomycetous and ascomycetous fungi. Three to 12 d after spiking, 22 to 38% of the PAH could no longer be recovered from the soils. At 287 d, 88.5 to 92.7%, 83.4 to 87.4%, and 22.0 to 42.1% of the 3-, 4-, and 5- to 7-R PAH, respectively, had disappeared from the unsterile, uninoculated soils. In 2 organic-rich sterile soils, the groups of wood- and straw-degrading, terricolous, and ectomycorrhizal fungi reduced the concentration of 5 PAH by 12.6, 37.9, and 9.4% in 287 d. Five- to 7-R PAH were degraded as efficiently as most of the 3- to 4-R PAH. In organic-rich unsterile soils inoculated with wood- and straw-degrading fungi, the degradation of 3- to 4-R PAH was not accelerated by the presence of fungi. The 5- to 7-R PAH, which were not attacked by bacteria, were degraded by fungi to 29 to 42% in optimum combinations of fungal species and soil type. In organic-poor unsterile soil, these same fungi delayed the net degradation of PAH possibly for 2 reasons. Mycelia of Pleurotus killed most of the indigenous soil bacteria expected to take part in the degradation of PAH, whereas those of Hypholoma and Stropharia promoted the development of opportunistic bacteria in the soil, which must not necessarily be PAH degraders. Contemporarily, the contribution of the fungi themselves to PAH degradation may be negligible in the absence of soil organic matter due to the lower production of ligninolytic enzymes. It is concluded that fungi degrade PAH irrespective of their molecular size in organic-rich and wood chip-amended soils which promote fungal oxidative enzyme production.
