Summary The concentrations of zinc, cadmium, lead and copper have been determined in the hepatopancreas, hindgut and rest of the body tissues of Oniscus asellus collected from eight sites in the U.K. The hepatopancreas is by far the most important storage organ of heavy metals, particularly cadmium, and at each site, contains a mean of at least 89% of the total body load of this element. Specimens of Oniscus asellus from contaminated sites may contain concentrations of zinc, cadmium, lead and copper in the hepatopancreas of about 1%, 0.5%, 2.5% and 3% of the dry weight respectively, which are among the highest so far recorded in the soft tissues of any animal.There is a significant positive correlation between the mean relative dry weight of the hepatopancreas of Oniscus asellus and the concentrations of zinc or cadmium in leaf litter from all eight sites. It is suggested that animals from sites which are contaminated heavily with zinc or cadmium have a large hepatopancreas because this enables them to de-toxify a greater amount of the metal.  相似文献

The effects of cadmium and zinc toxicity on orientation behaviour (photo- and geo-taxis) of Echinoparyphium recurvatum cercariae was investigated at concentrations ranging from 10 to 1000 microg l(-1). Exposure to the toxicants at all metal concentrations caused a change in orientation to negative phototaxis and positive geotaxis during the submaximal dispersal phase (0.5 h cercarial age). Autometallography staining of cercariae exposed to 1000 microg l(-1) cadmium or zinc showed selective binding of heavy metals to tegumental surface sites associated with sensory receptors. The significance to parasite transmission of changes in cercarial orientation behaviour in metal polluted environments is discussed.  相似文献

Environmental metal pollution is one of the major problems faced by humankind. This type of pollution affects aquatic systems (estuaries, coastal lagoons, etc.), which are very dynamic systems, therefore making the study of the effects on the organisms that inhabit them an essential issue. In this study, the capacity of metal regulation by decapod crustacean Penaeus vannamei juveniles was determined. The effects of zinc, lead, and cadmium were tested individually and as a metal mixture exposure to determine possible synergism. The results showed that juvenile shrimps were capable of regulating zinc and lead, whereas cadmium was accumulated without any excretion, at least within the concentrations studied. It was also proved that under the estuarine conditions tested here, P. vannamei juveniles showed capacity to act as a bioindicator for cadmium.  相似文献

Effects of zinc, copper, and selenium on placental cadmium transport   总被引:1,自引:0,他引:1  
The objective of the present study was to evaluate the potential effects of zinc, copper, and selenium on placental cadmium transport. From November 2002 through January 2003, a total of 47 healthy pregnant women from Da-Ye City, Hubei Province in Central China participated in the study. Their age, parity, gestational age, pregnancy history, and lifestyle data were obtained by questionnaire interview. The placental, whole-blood, and cord blood levels of cadmium were determined by inductively coupled plasma mass spectrometer (ICP-MS), whole-blood zinc was measured by flame atomic absorption spectrometry (F-AAS), whole-blood copper by ICP-MS, and selenium was by atomic fluorescence spectrophotometry (AFS). The cord blood cadmium concentration (0.020-1.48 microg/L) was significantly lower than in maternal blood (0.80-25.20 microg/L, p<0.01). The placental cadmium concentration was from 0.082 to 3.97 microg/g dry weight. Multiple linear regression analysis indicated that lower levels of maternal blood copper were significantly associated with higher cadmium concentrations in cord blood. Placental cadmium in women with lower levels of maternal blood zinc was significantly higher than in those with normal zinc levels. The placental cadmium level in women with lower whole-blood selenium was significantly lower than in subjects with normal selenium levels. It was concluded that the essential elements copper, selenium, and zinc might significantly affect placental cadmium transport.  相似文献

采用体外暴露染毒法,研究了不同浓度与时间条件下,镉诱导河南华溪蟹(Sinopotamon henanense)金属硫蛋白(metallothionein,MT)在肝胰腺、肌肉、鳃和卵巢中的表达差异。镉浓度分别为0、14·5mg/L、29mg/L和58mg/L;处理时间依次为1d、3d和5d。利用镉血红蛋白饱和法和火焰原子吸收分光光度法(AAS)测定MT的蛋白含量。结果显示,用不同的染毒浓度和处理时间,镉在组织中诱导产生MT的含量有较大差异,其中肝胰腺MT的诱导量最大,变化规律也最明显;肌肉中也有较大量MT的表达;而鳃和卵巢MT的诱导量均较低。此外,本文分析了镉的浓度与时间梯度对诱导MT表达的影响与毒性效应机制。结论:组织不同,染毒浓度及时间不同,镉诱导MT的表达也不同,具有一定的组织差异性和规律性。  相似文献

The distribution of metals within the hepatopancreas of Oniscus asellus (Crustacea, Isopoda) from two uncontaminated sites, and two sites contaminated with zinc, cadmium and lead, has been studied by atomic absorption spectrophotometry, light microscopy, transmission and scanning electron microscopy and X-ray microanalysis. The hepatopancreas contains two types of intracellular granule. The first type, in the S cells, are spherical granules which contain copper, sulphur and calcium. In woodlice from contaminated sites, these ‘copper’ granules, also contain zinc, cadmium and lead. The second type, in the B cells, are flocculent deposits which contain iron. In woodlice from contaminated sites, these ‘iron’ granules also contain zinc and lead. Moribund woodlice from contaminated sites have large numbers of ‘copper’ and ‘iron’ granules in the hepatopancreas and a fine deposit of zinc and lead on the membranes of the cells. There are numerous microorganisms in close association with the microvillous border of the hepatopancreas of woodlice from all four sites. Within the microorganisms of Oniscus asellus from contaminated sites, there are deposits of material which contain zinc, lead, calcium and phosphorus ‘Copper’ and ‘iron’ granules could have evolved as storage sites for essential metals to be utilized when demand from the body exceeds uptake from the food. Woodlice in contaminated sites may be able to ‘detoxify’ potentially harmful amounts of essential and non-essential metals by storing them in a relatively insoluble form within these granules.  相似文献

Studies have been made on the content and distribution of Cd, K, Mg, Ca, S and P in organs and cells of mussels and scallops before and after immersion in water containing cadmium chloride. In normal molluscs, the lowest content of Cd was found in the gonads; cadmium content of the hepatopancreas and kidney in the scallop is correspondingly 20 and 40 times higher than in the mussel. After immersion in water with cadmium (250 micrograms/l in summer experiments and 500 micrograms/l in spring ones), cadmium content of the hepatopancreas and kidney of the scallop was found to be also higher than that in the mussel, although the difference is less significant, being dependent on the season of year. The highest accumulation of cadmium (9.760 +/- 790 micrograms/g dry weight) was observed in the scallop kidney in summer period. After immersion in cadmium-containing water, the content of sulphur in the kidney and hepatopancreas increases, positive correlation being observed between the distribution of Cd and S. Exposure to cadmium results in the increase of Ca and Mg content in the gonads of scallops, as well as in the increase in Ca content of the gonads of mussels. Accumulation of cadmium in the gonads is rather small, maximum density of the material being observed histochemically in the gonadal wall. Morphological changes in the gonadal cells of mussels in summer period may be associated with the increase in cellular calcium.  相似文献

Studies were performed regarding the effect of cadmium accumulation on the levels of essential elements (copper, zinc and iron) in the tissues of a marine bivalve mollusc, Mizuhopecten yessoensis, exposed to cadmium at 250 ppb during 2 weeks. It was found that the concentration of cadmium in the tissues increased in the order gonads < gills < hepatopancreas < kidney during exposure time. However, the highest value of concentration factor was recorded in the gills. Our data demonstrate that cadmium accumulation in all mollusc tissues is followed by the alterations in copper, zinc and iron concentration, but that the pattern of these changes varies with each tissue. Cadmium had the most pronounced effects on essential trace element homoeostasis in the kidney. The present study suggests that levels of the essential metals in a particular tissue can be modified depending on the level of cadmium accumulation. The possible mechanisms of the effects of cadmium on the essential trace elements are discussed.  相似文献

Variations of the total free amino acid (FAA) pool and the content of specific amino acids have been measured in the muscle and hepatopancreas of adult shrimps, Penaeus japonicus, acclimatized at five water salinities: 38, 32, 26, 20 and 14%‰ The FAA content is always higher in muscle than in hepatopancreas at all tested salinites. On the other hand, the hepatopancreas exhibits the highest concentrations of essential amino acids. Two steps in the evolution of FAA content can be observed, the first one regarding decrease in salinity from 38 to 20%‰ and the second one, when salinity goes below 20%°. The first step can be characterized by a 16% decrease of total FAA content in the muscle and a 36% increase in the hepatopancreas. In muscle, the variations are mainly due to changes in non-essential FAA content, whereas in the hepatopancreas, they are linked to variations in essential FAA content. The other step is characterized by a drastic increase in moisture and decrease in FAA content in both studied organs when water salinity is 14%‰ The total FAA content is about 40% lower in shrimps at 14%° compared to 38%‰ seawater salinity. During adaptation, the FAA pool (mainly NEFAAs) of muscle seems to be directly related to osmoregulation, whereas in the hepatopancreas, its evolution seems to be linked with energy expenditure and protein synthesis. The results are evaluated in order to elucidate the role of FAA in intracellular osmoregulation and in relation to animal ecology.  相似文献

The effect of cadmium, zinc, and copper accumulation on metallothionein content in the selected tissues of bank voles and yellow-necked mice trapped near the Sendzimir Steelworks in Krakow and the zinc smelter in Bukowno were analysed. The Borecka Forest was chosen as a control area. The highest cadmium concentration, 32.98 microg g(-1) dry weight, was detected in the kidneys of the bank voles caught in the Bukowno area. Zinc and copper concentrations in the tissues did not exceed the critical values. Metallothionein content in the liver and kidneys was associated with heavy metal accumulation in the tissues. The highest content of sulphydryl groups was detected in the livers of the bank voles trapped within the neighbourhood of the zinc smelter in Bukowno. The highest level of disulphide bonds was found in the kidneys of the bank voles from the same area.  相似文献

The effect of cadmium (Cd) on mortality, resistance and bioaccumulation in the tropical freshwater prawn Macrobrachium rosenbergii from Egypt were studied. Survival of prawns exposed to Cd doses over 60 μg l?1 were significantly lower than that of prawns exposed to lower doses. After 96 h prawns exposed to >40 μg l?1 of Cd had a greater reduction in total haemocyte count and phagocytic activity than those exposed to lower concentrations. Bioaccumulation of Cd in the gills, hepatopancreas and muscles was variable. Cadmium accumulated significantly in the gills and hepatopancreas, but Cd accumulation in the muscles increased only marginally. Macrobrachium rosenbergii manifested histopathological alterations in the gills, hepatopancreas and muscles when exposed to various concentrations of cadmium.  相似文献

Variation in Esterase 2 C activities, involving the hydrolysis of 2-carboxylic esters, α-glucosidase acetyl-glucosaminidase and alkaline and acid phosphotases, in the hepatopancreas and the abdominal muscle of Palaemon serratus was examined by polyacrylamide gradient gel electrophoresis. Soluble proteins were measured in the hepatopancreas and the abdominal muscle, and trypsin and chymotrypsin activities in the hepatopancreas. The activities and the isoenzymatic variations in shrimps acclimated at 5 different temperatures (between 14 and 30°) were compared and the molecular weight of each isozyme evaluated. It was found that: (a) the concentrations of soluble proteins decrease in the hepatopancreas between 18 and 30°, but remain unchanged in the abdominal muscle; (b) esterase and phosphatase activities increase with temperature but in a more or less random manner, according to the isozyme under consideration; (c) glycosidase activities increase with temperature; and (d) trypsin activity varies in an inverse relation to chymotrypsin activity.  相似文献

The regulation of metallothionein (MT) gene expression in a cadmium resistant CHO cell line which overproduces MT was examined in this study. Our results show that MT mRNA levels reach a maximum 24-30 h after a primary zinc exposure and, subsequently, MT mRNA concentrations decline. This decrease in MT mRNA levels can be correlated with the accumulation of metallothionein and decreased rates of zinc and cadmium uptake.  相似文献

SUMMARY. The toxicity of zinc to Hormidium rivulare Kütz. in laboratory culture media is decreased by rises in the levels of magnesium, calcium and phosphate, and increased by rises in pH and cadmium. The effects of all these are sufficiently marked that they may be expected to have considerable importance in the field. In contrast, assays with sodium, chloride and sulphate showed no detectable influence of these ions on zinc toxicity.
When applied at higher concentrations, calcium was always more effective than magnesium, but the reverse was sometimes true at lower concentrations. Both magnesium and phosphate were more effective in reducing zinc toxicity with zinc-tolerant populations than with zinc-sensitive ones. Cadmium was highly toxic, either alone or in combination with zinc; however, calcium had a proportionately greater effect in reducing cadmium toxicity than zinc toxicity. The toxic effects of zinc and cadmium were synergistic, and it seems probable that the presence of cadmium at levels of 0.01 mgl−1 and above will usually lead to a significant increase in the toxicity of any zinc also present. Cadmium (in the absence of zinc) was 34 times more toxic than zinc to a zinc + cadmium sensitive population, and 15.5 times more toxic to a zinc + cadmium tolerant population. Because of the synergistic response, cadmium had an even greater effect in the presence of zinc.  相似文献

Land snails are highly tolerant to cadmium exposure and are able to accumulate soil cadmium independently of food ingestion. However, little information exists on the kinetics of cadmium retention in terrestrial gastropods exposed to an increase in the soil cadmium content, over time. There is also little knowledge about how exposure to cadmium-polluted soils influences shell growth and architecture. In this context, we examined cadmium accumulation in the hepatopancreas and shell of juvenile Cantareus aspersus exposed to elevating high levels of cadmium in soil. Also, the toxicity of cadmium to snails was assessed using a range of conchological endpoints, including shell height, width, volume, allometry and integrity. Test snails, aged three months, were reared under semi-field conditions, fed an uncontaminated diet and exposed first, for a period of 30 days, to a series of soil cadmium concentrations, and then, for a second period of 30 days, to soils with higher cadmium content. Cadmium showed a dose-dependent accumulation in both the hepatopancreas and shell. The kinetics of cadmium retention in the hepatopancreas of snails previously exposed to cadmium-spiked soils was significantly influenced by a new exposure event. The shell was not a relevant bioaccumulator for soil cadmium. Under the present experimental conditions, only high cadmium exposure significantly affected either the shell growth or snail survival. There was no consistent effect on shell allometry, but the shell integrity, especially in rapidly growing parts, appeared to be affected by high cadmium exposure. Our results attest to the value of hepatopancreas for describing cadmium retention in land snails and to the difficulty of using conchological parameters in field surveys for estimating the environmental hazard of soil cadmium.  相似文献

The induction of micronuclei (MN) in mitotically active cells has been widely used and promoted as a biological marker of exposure to environmental toxins. In our study the effect of zinc on cadmium genotoxicity was investigated in V 79 cells. The results indicate that cadmium chloride exposure for 24 h increased micronucleus frequency and the percentage of binucleated cells in dose-dependent manner. At the highest concentration of cadmium (50 microM Cd) 23 MN were found in 1000 cells. The protective effect of zinc on cadmium genotoxicity was investigated at lower concentrations (5-25 microM CdCl2). At 50 microM Cd, the number of MN increased significantly (16 MN).  相似文献

The distributions of cadmium, chromium, lead, and zinc in eelgrass were studied in samples collected from the field, and the loss/accumulation of the metals during decomposition of eelgrass leaves was studied in laboratory experiments.Concentrations of heavy metals in the below grounds parts were greater in the roots than in the different age groups of the rhizomes. In the rhizomes, the highest concentrations of lead were recorded in the oldest parts, whereas highest chromium and zinc concentrations were found in the youngest parts. The concentration of cadmium did not vary. In the above ground parts, the concentrations of cadmium, lead, and zinc increased with age of the leaves, and concentrations in the leaves were greater than in the stem fraction. The concentrations of chromium decreased with age of the leaves.In the laboratory study of decomposition of leaf material, the concentrations of chromium, lead and zinc increased significantly and a net absorption from the surrounding water was recorded. Cadmium concentrations were relatively constant and a loss of cadmium was proportional to the release of soluble organic compounds indicating an association of cadmium with the soluble phase.The investigation demonstrated the utility of compositional analyses and decomposition experiments in assessing the significance of eelgrass in the heavy metal cycling in coastal areas. Furthermore, significant differences in the fate of heavy metals associated with eelgrass detritus are discussed.  相似文献

Lead and zinc tolerant genotypes ofHolcus lanatus L. were grown in culture solution at different cadmium, lead and zinc concentrations, and combinations. In all treatments, an increased inhibition of root length with increasing concentrations of heavy metals was observed. Growth of genotype 1 was better than that of genotype 2 in all treatments, suggesting that genotype 1 is more tolerant. The better root growth of genotype 1, at different cadmium concentrations, than that of genotype 2, indicated the existence of a co-tolerance or greater tolerance of genotype 1 to cadmium. Heavy metal combinations resulted in increased lead or zinc uptake by plants, while cadmium was decreased. In a lead-zinc combination, decreased lead and increased zinc uptake were detected. The different interactive effects of heavy metals on root growth of genotype 1 (additive or synergistic) and genotype 2 (additive or antagonistic) may suggest their differential susceptibility to the above metals.  相似文献
