The concentrations of the main plasma inorganic electrolytes Na+, K+, Ca2+, Mg2+, Cl- and and PO43- have been determined for different orders of marine fishes. For Na+ and Cl- a typical decrease was found when passing from cyclostomes, holocephalans and elasmobranchs to teleosts. The concentrations of K+, Ca2+ and Mg2+ showed a similar trend except that there was a rise in the teleost group, which showed a large range of variation for these three ions. In the case of PO43- no significant differences between groups were found.  相似文献

The haemolytic reagent allowing direct serological detection of He homozygous pigs was obtained by the immunization of a Landrace sow. Another monospecific reagent prepared from immune serum of a Miniature pig made possible the evidence for a new factor of the H system - He. This factor is genetically controlled by the allele H be. Its frequency in Miniature pigs was 0.099.
The H system with alleles H 1=Ha, H2= Hb, H3= Hab, H4=Hcd, H5= Hbd, H6=Hbe and H7= H- continues to be a genetically open system.  相似文献

Low-K+, high-Na+ cells of strain RL21a of Neurospora crassa , in steady state with 25 m M Na+, were used to study K+/Na+ exchanges in the presence or absence of Ca2+ and Mg2+. In the presence of Ca2+ and Mg2+, a low concentration of K+ (0.3 m M ) triggered a rapid exchange, but in the absence of the divalents, a high K+ concentration (30 m M ) was required to initiate the exchange at a rapid rate. In the absence of Ca2+ and Mg2+, K+ uptake did not occur at low K+ concentration, internal K+ did not regulate Na+ influx in the presence of external K+, and the efflux of Na+ proceeded at maximum activity at very low-K+ contents.  相似文献

Pyoverdine (PvdI) is the major siderophore secreted by Pseudomonas aeruginosa PAOI in order to get access to iron. After being loaded with iron in the extracellular medium, PvdI is transported across the bacterial outer membrane by the transporter, FpvAI. We used the spectral properties of PvdI to show that in addition to Fe3+, this siderophore also chelates, but with lower efficiencies, all the 16 metals used in our screening. Afterwards, FpvAI at the cell surface binds Ag+, Al3+, Cd2+, Co2+, Cu2+, Fe3+, Ga3+, Hg2+, Mn2+, Ni2+ or Zn2+ in complex with PvdI. We used Inductively Coupled Plasma-Atomic Emission Spectrometry to monitor metal uptake in P. aeruginosa : TonB-dependent uptake, in the presence of PvdI, was only efficient for Fe3+. Cu2+, Ga3+, Mn2+ and Ni2+ were also transported into the cell but with lower uptake rates. The presence of Al3+, Cu2+, Ga3+, Mn2+, Ni2+ and Zn2+ in the extracellular medium induced PvdI production in P. aeruginosa . All these data allow a better understanding of the behaviour of the PvdI uptake pathway in the presence of metals other than iron: FpvAI at the cell surface has broad metal specificity at the binding stage and it is highly selective for Fe3+ only during the uptake process.  相似文献

Two new erythrocyte antigens have been detected by isoimmune antibodies obtained from Gottingen Miniature pigs. By family studies they proved to belong to the E system. The factor designated En is genetically controlled by alleles E 2= E edghkmn, E 3= E aegln, E 4= E edfhkmn, E 6= E aefln, E 9= E edghjmn, E 12= E aegmno and E edgkln, and the factor designated Eo by the allele E 12= E aegmno.
The En blood factor occurs in all the breeds studied and in frequency it comes close to a related Ee factor. It stands in opposition not only to Eb but also to Ei factor. The Eo factor can be characterized as a rarely occurring Ea subgroup. Besides being antithetic to Ed it is also antithetic to El and Ei factors. For the present, it was found only in Miniature pigs.  相似文献

The potassium ion is an indispensible cytosolic component of living cells and a key osmolyte of plant cells, crossing the plasmalemma to drive physiological processes like cell growth and motor cell activity. K+ transport across the plasmalemma may be passive through channels, driven by the electrochemical gradient, K+ equilibrium potential (EK) – membrane potential (Vm), or secondary active by coupling through a carrier to the inward driving force of H+ or Na+. Known K+ channels are permeable to monovalent cations, a permeability order being K+ > Rb+ > NH4+ > Na+≥ Li+ > Cs+. The macroscopic K+ currents across a cell or protoplast surface commonly show rectification, i.e. a Vm-dependent conductance which in turn, may be controlled by the cytosolic activity of Ca2+, of K+, of H+, or by the K+ driving force. Analysis by the patch clamp technique reveals that plant K+ channels are similar to animal channels in their single channel conductance (4 to 100 pS), but different in that a given channel population slowly activates and may not inactivate at all. Single-channel kinetics reveal a broad range of open times (ms to s) and closed times (up to 100 s). Further progress in elucidating plant K+ channels will critically depend on molecular cloning, and the availability of channel-specific (phyto)toxins.  相似文献

Abstract: The role of the Na+/Ca2+ exchanger and intracellular nonmitochondrial Ca2+ pool in the regulation of cytosolic free calcium concentration ([Ca2+]i) during catecholamine secretion was investigated. Catecholamine secretion and [Ca2+]i were simultaneously monitored in a single chromaffin cell. After high-K+ stimulation, control cells and cells in which the Na+/Ca2+ exchange activity was inhibited showed similar rates of [Ca2+]i elevation. However, the recovery of [Ca2+]i to resting levels was slower in the inhibited cells. Inhibition of the exchanger increased the total catecholamine secretion by prolonging the secretion. Inhibition of the Ca2+ pump of the intracellular Ca2+ pool with thapsigargin caused a significant delay in the recovery of [Ca2+]i and greatly enhanced the secretory events. These data suggest that both the Na+/Ca2+ exchanger and the thapsigargin-sensitive Ca2+ pool are important in the regulation of [Ca2+]i and, by modulating the time course of secretion, are important in determining the extent of secretion.  相似文献

The nitrogen-related phosphotransferase system (Ntr-PTS) is a paralogous system working in parallel to the well-known carbohydrate:PTS. In a chain of phosphotransfer reactions, EINtr and NPr (PtsO) deliver phosphoryl groups to the EIIANtr (PtsN) protein. EIIANtr is implicated in important regulatory processes such as the σE-dependent cell envelope stress response and regulation of K+ uptake. Phosphorylation is believed to trigger the output of EIIANtr in these regulations. EIIANtr is encoded within the gene cluster ptsN–yhbJ–ptsO , which is highly conserved in Proteobacteria . In this study, we investigated the phosphorylation of the Escherichia coli EIIANtr protein in vivo by 32P-labeling. We show that EIIANtr is readily phosphorylated in wild-type cells. This phosphorylation occurs at a single site, the histidine 73 in EIIANtr. YhbJ and NPr are dispensable for this phosphorylation. A detailed analysis revealed that both the energy coupling phosphotransferases of the Ntr-PTS as well as the 'sugar'-PTS contribute to the phosphorylation of EIIANtr, suggesting cross talk between both systems.  相似文献

Using excised roots of Atriplex hortensis L., cv. Gelbe Gartenmelde, the uptake, accumulation and xylem transport of K+ and Na+ have been measured. Influx as well as xylem transport proved to discriminate little between K+ and Na+, when considered in relation to the external solution. Both K+ and Na+ inhibited the uptake and xylem transport of each other to about the same degree. Measurements of intracel-lular Na+ fluxes by means of compartment analysis indicated that the low degree of K/Na discrimination during uptake was due to low influx selectivity. Moreover, K+/Na+ exchange at the plasmalemma was not very efficient in Atriplex roots. In order to establish the basis of the low K/Na discrimination in xylem transport, the rates of K+ and Na+ transport were related to the cytoplasmic K+ and Na+ concentrations to yield the selectivity ratio of transport, S(transport) = (φcx(K) × [Na+]c)/(φcx(Na) × [K+]c). Under all conditions this ratio was far below one indicating that Na+ was favoured during xylem release in excised roots of Atriplex at low external concentrations. The implications of this discrimination in favour of Na+ are discussed with respect to salt tolerance of A. hortensis .  相似文献

Effects of microinjected cations on the early events of fertilization were examined using eggs of Oryzias latipes . Microinjection of either Ca2+, Ba2+ or Sr2+ into the thin cortical cytoplasm induced breakdown of cortical alveoli (vesicles) (CABD) under Ca-Mg-free conditions, but microinjection of Mg2+, Mn2+ or Co2+ prevented CABD at the injected region when the eggs were inseminated in regular saline. Under Ca-Mg-free conditions, CABD could also be induced by microinjection of various solutions (NaCl, choline chloride, sucrose, pH buffer) without any divalent cations or ionophore A23187. Ca2+ microinjected into the cortical cytoplasm did not play a role in sperm penetration. Upon microinjection with either Ca2+, Mg2+ or K+, the resting membrane potential leakage was transiently observed. However, depolarization of the membrane followed by slow hyperpolarization was observed only upon microinjection of Ca2+. From these experiments, it was inferred that microinjected divalent cations such as Ca2+, Ba2+ or Sr2+ do not act directly upon the cortical alveolus membrane, but trigger the induction of CABD via depolarization of the membrne and increase in intracellular Ca2+.  相似文献

Mean values of the composition of the seminal fluid of 30 turbots were: osmotic pressure =306 mosml−1, pH =7–31, total protein =8.8mg ml1, Na+=133.0 mmoll1, K+=3.80mmoll−1, Cl=129mmol l−1. The rounded nucleus, the reduced middle piece and the typical '9 + 2' structure of the flagellum mean that the spermatozoon of turbot can be considered to be of a primitive type.  相似文献

Several alleles were found to determine the colour of the dorsal pronotum in Chorthippus brunneus; there was evidence for at least two loci ( C and V ). Brown ( CB )was the universal recessive and green ( CC ) was dominant to all other colours. The white allele ( CW )was codominant with green( CG )and purple ( CP ). Wing-patterns were determined by a separate, probably linked locus ( W ). A dominant plain wing-pattern ( WP ) was associated with colours other than brown. Striped( WS )and mottled( Wmo ) were codominant and a plain recessive allele ( WP ) was also found. All three alleles were associated with the brown phenotype. A purple-sided allele ( SPu ) was sometimes obmd with Cpu.. S Pu was dominant to brown sides ( SB ), A series of markings on the dorsal and lateral pronotum ( linea intermedia, fascia postocularis, linea media, carina media and zona lateralis ) were investigated and found to be controlled at separate loci which may be linked to W . These characters were expressed by dominant alleles.
Epistatic effects by modifier loci were shown to have an important effect on the determination of wing phenotype. Allele Wo+ , for example, suppressed the stripe-wing pattern, linea media, carina media and zona lateralis .
It was concluded that colour patterns appear to be under genetic control and that dominant alleles were rare in the wild. Changes in shades of colours were shown to be age-dependent and minor.  相似文献

Abstract: The effects of alcohol and Ca2+ transport inhibitors on depolarization-induced stimulation of oxidative phosphorylation and free-Ca2+ concentrations in rat synaptosomes were investigated. Glucose oxidation was stimulated by depolarization with K+ or veratridine and by the Ca2+ ionophore ionomycin. The stimulation by K+, veratridine, and ionomycin was correlated with elevation of synaptosomal free Ca2+. Depolarization-stimulated respiration was inhibited by verapamil, Cd2+, and ruthenium red but not by diltiazem. Synaptosomal Ca2+ elevation was inhibited by verapamil but not by ruthenium red. These results indicate that the stimulation depends on elevation of mitochondrial free Ca2+. Ethanol, at pharmacological concentrations (50–200 m M ), inhibited the Ca2+-dependent stimulation of oxidative phosphorylation. This inhibition resulted, in part, from the inhibition of voltage-gated Ca2+ channels, which inhibited the elevation of synaptosomal free Ca2+, and, in part, from the stimulation of the mitochondrial Ca2+/Na+ antiporter, which inhibited the elevation of the mitochondrial matrix free Ca2+. The inhibition by ethanol of the excitation-induced stimulation of oxidative phosphorylation in the synapse may contribute to the depressant and narcotic effects of alcohol and enhance excitotoxicity.  相似文献

The appearance of soil NO3 after forest disturbance is commonly ascribed to a higher availability of NH4+ to autotrophic nitrifiers, or to a reduction in available-C resulting in lower microbial assimilation of NO3. Alternatively, it has been proposed that increasing NH4+ pools following disturbance could increase net nitrification by reducing microbial assimilation of NO3. Forest floor material was collected from shelterwood harvest plots which displayed both low available-C and low NH4+ pools, and where previous experiments had suggested the prevalence of heterotrophic nitrification. Subsamples were amended with incremental rates of glucose-C or NH4+, and gross NO3 transformation rates were measured by isotope dilution. Glucose-C additions had little effect on the net difference between gross NO3 production and consumption rates. On the other hand, NH4+ additions caused gross NO3 consumption processes to decrease sharply, while gross NO3 production processes remained constant. The results suggest that NH4+ can have an immediate positive effect on net nitrification rates by suppressing NO3 assimilation and uptake systems.  相似文献

Abstract Transport of Mn2+ was repressed in Candida utilis cells grown in continuous culture in high-Mn2+ (100 μM Mn2+) medium as compared to cells grown in basic (0.45 μM Mn2+) and low-Mn2+ (< 0.05 μM Mn2+) media. In contrast, no repression of Cu2+ uptake occurred in high-Cu2+-grown (25 μM Cu2+) cells as compared to cells grown in basic medium (0.54 μM Cu2+). Cu2+-limited cells did not hyperaccumulate Cu2+ and there was not significant difference in initial uptake rates for all 3 Cu2+ conditions. Mn2+ uptake appears to be regulated by a mechanism sensitive to the external Mn2+ concentration, whereas Cu2+ transport is not governed in this way by the external Cu2+.  相似文献

SUMMARY. We have analysed data from ninety-nine Scottish freshwater lochs, to explore the relationship between water chemistry and phytoplankton assemblages. Our results confirm that there are strong correlations both between the phytoplankton quotient and divalent cation concentrations, particularly Ca++, and also between the phytoplankton quotient and the (Na++ K+)/(Ca+++ Mg++) ratio. However, the latter is evidently a spurious relationship, arising from the former association, together with an association between Ca++ and the (Na++K+)/ (Ca+++ Mg++) ratio. The observed correlation does not persist if account is taken of concomitant variations in the Ca++concentration. This conclusion is suggested both by relatively informal analyses (median polish, partial correlation coefficients) and by more formal modelling and testing (stepwise regression, all-subsets regression).  相似文献

Abstract The Escherichia coli ptsI and ptsH genes code for the synthesis of two proteins of the phosphoenolpyruvate-dependent phosphotransferase system (PTS), namely enzyme I and protein HPr. A number of ptsI + ptsH +/F' ptsI + ptsH merodiploids was obtained. It was shown in experiments in vivo that ptsH mutations in the transposition are dominant. Bacterial extracts from these merodiploids supported [14C]methyl glucoside (MG) phosphorylation at the expense of phosphoenolpyruvate only half as much as extracts from the pts + cells. ptsI + ptsH /F' ptsI + ptsH + merodiploids appeared to be non-viable; the reason for this lack of viability is discussed.  相似文献

The effect of several metal ions on NADP+-malic enzyme (EC 1.1.1.40) purified from Zea mays L. leaves was studied Mg2+, Mn2+, Co2+ and Cd2+ were all active metal cofactors. The malic enzyme from maize has a moderately high intrinsic preference for Mn2+ relative to Mg2+ at pH 7.0 and 8.0 Negative cooperativity detected in the binding of Mg2+ at pH 7.0 and 8.0 and in the binding of Mn2+ at pH 7.0 suggests the existence of at least two binding sites with different affinity. All of the activating metal ions have preference for octahedral coordination geometry and have ionic radii of 0.86–1.09 Å. The ions that act as inhibitors are outside this range and/or are incapable of octahedral coordination. Ba2+, Sr2+, Cd2+, Ca2+, Be2+, Ni2+, Cu2+, Zn2+, Co2+, Hg2+ showed mixed-type inhibition. The reciprocal of their K1 values follow the order of their apparence in the Irving-Williams series of stability that derives in part from size effects. It is suggested that the size of the ions may play a partial role in determining the strength of the metal interaction.  相似文献

Abstract: The present study reports the ion dependency of 2β-carbomethoxy-3β-(4-fluorophenyl)[3H]tropane ([3H]- CFT) binding to the dopamine transporter in the rat striaturn. The results indicate that [3H]CFT binding to synaptosomal P2 membranes requires low concentrations of Na+ (peak binding between 20 and 50 m M Na+), is stimulated by phosphate anion or l-, but is unaffected or only slightly affected by F-, Cl-, Br-, NO3-, or SO42-, Concentrations of Na+ of >50 m M become inhibitory except in the presence of l-, which shifts peak binding levels toward higher Na+ concentrations and also elevates the peak binding level. K+ strongly decreased [3H]CFT binding with a shallow inhibition curve, and Na+ could not overcome this effect. Saturation analysis of [3H]CFT binding revealed a single binding site changing its affinity for CFT depending on the concentration of sodium phosphate buffer (6, 10, 30, 50, 130, or 200 m M ; 1 mM plus 49 mM NaCIversus 10 m M plus 40 m M NaCI; or 1 mM plus 129 m M Nal versus 10 m M plus 120 m M Nal). No differences were observed in the density of CFT binding sites between any of the conditions examined.  相似文献
