Blood absorption of tetracycline hydrochloride of various dispersity levels from capsules containing tetracycline alone or in combination with additives, such as magnesium carbonate and calcium salts was studied on humans. It was found that higher dispersity levels of tetracycline hydrochloride powder in capsules was not accompanied by increased blood absorption of the antibiotic. Addition of magnesium carbonate and calcium salts to the antibiotic in the process of capsulation markedly retarded the blood absorption. Clear correlation between the antibiotic dissolution rate in vitro and intensity of its blood absorption in volunteers was shown.  相似文献

Difference in the levels of bioavailability of 4 types of tetracycline hydrochloride tablets was shown on rabbits and guinea pigs. The rates of the antibiotic absorption into the blood of the laboratory animals and volunteers were the same. Relation between the rate of disintegration of various types of tablets of tetracycline hydrochloride and the antibiotic blood levels in the rabbits after administration of the same tablets was found.  相似文献

Desintegration and dissolution of capsules and tablets of methacycline hydrochloride were studied. The study on solubility of methacycline hydrochliride capsules filled with methacycline granulate or powder according to the same formula showed that the rate of the antibiotic liberation from the capsules filled with the powder decreased during storage while that from the capsules filled with the granulate did not change. Investigation of the effect of the mass packing value in a drop on the antibiotic liberation from the capsules showed that an increase in the packing coefficient above 1.38 resulted in a marked decrease in the rate of methacycline liberation from the capsules filled with the granulate. No correlation between desintegration and dissolution of methacycline capsules and tablets was found.  相似文献

Modified-release multiple-unit tablets of loratadine and pseudoephedrine hydrochloride with different release profiles were prepared from the immediate-release pellets comprising the above two drugs and prolonged-release pellets containing only pseudoephedrine hydrochloride. The immediate-release pellets containing pseudoephedrine hydrochloride alone or in combination with loratadine were prepared using extrusion–spheronization method. The pellets of pseudoephedrine hydrochloride were coated to prolong the drug release up to 12 h. Both immediate- and prolonged-release pellets were filled into hard gelatin capsule and also compressed into tablets using inert tabletting granules of microcrystalline cellulose Ceolus KG-801. The in vitro drug dissolution study conducted using high-performance liquid chromatography method showed that both multiple-unit capsules and multiple-unit tablets released loratadine completely within a time period of 2 h, whereas the immediate-release portion of pseudoephedrine hydrochloride was liberated completely within the first 10 min of dissolution study. On the other hand, the release of pseudoephedrine hydrochloride from the prolonged release coated pellets was prolonged up to 12 hr and followed zero-order release kinetic. The drug dissolution profiles of multiple-unit tablets and multiple-unit capsules were found to be closely similar, indicating that the integrity of pellets remained unaffected during the compression process. Moreover, the friability, hardness, and disintegration time of multiple-unit tablets were found to be within BP specifications. In conclusion, modified-release pellet-based tablet system for the delivery of loratadine and pseudoephedrine hydrochloride was successfully developed and evaluated.  相似文献

A process for the microencapsulation of mammalian cells in a commercially available water-insoluble polyacrylate (EUDRAGIT RL) is described, and the effects of process parameters are outlined The polymer dissolved in diethyl phthalate was pumped along the annulus formed from two concentric needles, while the cell suspension was pumped inside the inner needle Droplets of polymer solution containing cells were blown off the end of the needles by a coaxial air stream. The droplets fell into a corn oil-mineral oil curing bath, in which the solvent was removed from the nascent capsule causing the polymer to precipitate around the cell suspension core. Capsules were washed free of oils and solvent in a fractionated plasma that allowed for quantitative transfer of capsules from the oil phase to an aqueous medium. By appropriate adjustment of the coaxial air flow rate, capsule size could be varied from 250-1000 mum, although the most convenient size was found to be 400-700 mum. Adding Ficoll 400 to the cell suspension to match the density of the suspension to the polymer solution resulted in capsules with a well-centered core but did not affect capsule strength. It appeared that increasing the polymer solution concentration or the polymer to the cell flow rate ratio resulted in an increased capsule strength, although differences in capsule size made unequivocal conclusions difficult. These capsules are of potential use as an artificial pancreas for the treatment of diabetes (with pancreatic islets) or for large-scale tissue culture and the production of bioactive products (e.g., with fibroblasts).  相似文献

Microcapsules composed of synthetic (sodium polystyrene sulfonate and polyallylamine hydrochloride) and biodegradable polyelectrolytes (dextran sulfate and polyarginine hydrochloride) deposited on carbonate microparticles have been obtained. The ultrastructural organization of biodegradable microcapsules has been studied by transmission electron microscopy. The shell of biodegradable microcapsules is well formed even after the deposition of six polyelectrolyte layers and has an average thickness of 44 ± 3.0 nm; their inner polyelectrolyte matrix is less branched than that of synthetic microcapsules. By using spectroscopy, the efficiency of the encapsulation of FITC-labeled BSA by adsorption depending on the number of PE layers in the capsule has been estimated. It has been shown that the maximum amount of the protein is incorporated into capsules comprising six and seven polyelectrolyte layers (4 and 2 pg/capsule, respectively). It has been concluded that the adsorption of proteins into preformed polyelectrolyte capsules enables one to avoid protein losses that occur with the method in which biomineral cores obtained by coprecipitation are used for encapsulation.  相似文献

The diameter, membrane thickness, and compression intensity of hollow Ca-alginate capsules were measured at different gelation conditions, such as the reactant concentration, dropping velocity, and gelation time. The optimum operation conditions for preparing capsules were determined at 100 g/L CaCl(2), 10 g/L sodium alginate (Na-alginate), a dropping velocity of 150 droplets/min, and a gelation time of 10 min. Diffusion of some saccharide and amino acid from bulk solution into capsules was investigated, and the diffusion coefficients were calculated by the developed mathematical model. All the tested substances can diffuse easily into the capsules. The combined diffusion coefficients of the capsule D(m) are 92-99% as large as their diffusion coefficients in pure water, while the diffusion coefficients in the capsule membrane D(1) are 60-95% as large as those. By employing polyethylene glycol (PEG) and bovine serum albumin (fraction V) (BSA(V)), the molecular weight cut-off of the capsule was determined. For linear macromolecules, hollow Ca-alginate capsules have a molecular weight cut-off of 4000. No diffusion of BSA(V) into the capsules was observed.  相似文献

The results of the experiments on determination of the effect of aeration and agitation conditions on biosynthesis of tetracycline in the apparatus of semi-production capacity are discussed. It was shown that the antibiotic production level was not connected with the rate of oxygen solution expressed in the sulphite numbers, i.e. this parameter cannot be used as a scaling-up criterion. Accumulation of the antibiotic in the fermentation broth depended on the volume of the air supplied for aeration. It was determined that the level of CO2 dissolved in the fermentation broth did not reach the values having an inhibitory effect on the biosynthetic process.  相似文献

The quantitative indices and regularities of tetracycline hydrochloride binding with colelagen in the process of complex formation were studied in vitro. It was found that tetracycline-interacted with collagen. The amount of the antibiotic bound depended on its initial concentration in the mixture and the aggregate state of the protein: the amount of tetracycline hydrochloride absorbed by dissloved collagen was 10 times higher than that absorbed by polymerized collagen. The antibiotic bound with collagen did not lose its specific antibacterial activity.  相似文献

A Psychrobacter psychrophilus strain resistant to tetracycline and streptomycin was isolated from a 15 000–35 000-year-old permafrost subsoil sediment sampled from the coast of the Eastern-Siberian Sea. The genes conferring antibiotic resistance were localized on an c . 30-kb pKLH80 plasmid. It was shown that the antibiotic resistance region of this plasmid has a mosaic structure and contains closely linked streptomycin resistance ( strA-strB ) and tetracycline resistance [ tetR-tet (H)] genes, followed by a novel IS element (IS Ppy1 ) belonging to the IS 3 family. Both the strA-strB and tetR-tet (H) genes of pKLH80 were highly similar to those found in modern clinical bacterial isolates. It was shown that the IS Ppy1 element of pKLH80 can direct translocation of the adjacent antibiotic resistance genes to different target plasmids, either by one-ended transposition or by formation of a composite transposon resulting from the insertion of the IS Ppy1 second copy at the other side of the antibiotic resistance region. Thus, our data demonstrate that clinically important antibiotic resistance genes originated long before the introduction of antibiotics into clinical practice and confirm an important role of horizontal gene transfer in the distribution of these genes in natural bacterial populations.  相似文献

It is shown that kinurenine derivatives, harmane (beta-carboline) and tetracycline hydrochloride known as photosensitizers of cataractogenesis in lens produce luminescence of singlet molecular oxygen (1O2) under photoexcitation in air saturated aqueous (D2O) solution. The quantum yields of the 1O2 generation by these substances are determined. The data obtained by this direct 1O2 determination technique suggest that 1O2 might take part in cataractogenesis.  相似文献

The method of back turbidimetry was used for determination of the biological activity of the antibiotics, since high turbidity of the nutrient medium with Staph. aureus as the testculture prevented from direct measurements. Broth containing phosphate buffer, Staph. aureus and definite concentrations of the antibiotic was used as the reference solution. The experiments showed that the differences in the biological activities of tetracycline hydrochloride and morphocycline may be found with the method of back turbidimetry 6-8 hours after the microbe cultivation on media with the antibiotics.  相似文献

Ludwigia sect. Microcarpium is a polyploid complex of 15 taxa that occur mainly in wet habitats of the southeastern United States. In previous treatments, plants of this section were described as having capsules that dehisced by the separation of the walls from the indurate disc (ring dehiscence). Two additional types of dehiscence are reported in this study. The capsules of some species dehisce by the irregular disintegration of the outer capsule wall (peeled dehiscence), while in another group, they dehisce by four longitudinal, lenticular slits and finally into four basally united parts (loculicidal dehiscence). Each of the different patterns of capsular dehiscence was found to have a distinctive anatomical basis. The loculicidal-dehiscent and peeled-dehiscent capsules apparently evolved from the ring-dehiscent type of capsule independently, and each of these derived types appears to delimit a natural evolutionary unit.  相似文献

Three different concentrations of the antibiotic tetracycline in honey were tested for their influence on the offspring production and longevity of the parasitoid wasp Encarsia formosa. Several earlier publications did not provide a conclusive answer on the effect that the Wolbachia have on these wasps. The results of our experiments show that at high tetracycline hydrochloride concentrations in honey (50mg/ml) the antibiotic is toxic to the females, all females died within three days after the antibiotic treatment. The concentration 5mg/ml was less toxic although the treated females also lived shorter and produced less offspring than the control females. At the lowest tested concentration of 1mg/ml there was no significant difference either in offspring production or in longevity between the control and the treated females. The antibiotic treatment at both 5 and 1mg/ml resulted in exclusively male progeny after the first two days of oviposition. These results are consistent with the theory that in species in which all individuals are infected the Wolbachia should not impose a large fitness cost.  相似文献

A novel perstraction system using liquid-core microcapsules for pesticide and herbicide removal from aqueous environments is proposed. The microcapsules contain an oil, dibutyl sebacate, surrounded by a hydrogel membrane. The extraction efficiency of the capsules was demonstrated with atrazine, methylparathion, ethylparathion, and 2,4-dichloro-phenoxyacetic acid. The results show that all of the tested compounds could be rapidly extracted, typically 75% extraction within 10 minutes using a capsule: liquid volume ratio of only 3.5% for ethylparathion, and that the rate of extraction increased with increasing hydrophobicity of the compound to be extracted. Higher rates of extraction could be achieved by changing the capsule: liquid volume ratio. The effect of different liquid core solvents, size of capsules, agitation rate, and treatment with complexing agents on the properties of the microcapsules and extraction rate were studied. Capsules of a diameter smaller than 0.800 mm show little external resistance to mass transfer. The main resistance to mass transfer of the pesticides/herbicides was found to reside in the hydrogel membrane composed of cross-linked alginate/polyacrylamide. Removal of divalent cations from the membrane by the addition of citrate, resulted in a 50% increase in the mass transfer coefficient, probably as a result of solubilization and exo-diffusion of alginate.  相似文献

Measurement of the rate of glucose diffusion from EUDGRAGIT RL and HEMA-MMA microcapsules coupled with a Thiele modulus/Biot number analysis of the glucose utilization rate suggests that pancreatic islets and CHO (Chinese hamster ovary) cells (at moderate to high cell densities) should not be adversely affected by the diffusion restrictions associated with these capsule membranes. The mass transfer coefficients for glucose at 20 degrees C were of the same order of magnitude for both capsules, based on release measurements: approximately 5 x 10(-6) cm/s for EUDRAGIT RL and approximately 2 x 10(-6) for HEMA-MMA. Inulin release from EUDRAGIT RL was slower than for glucose (mass transfer coefficient 14 +/- 4 x 10(-8) cm/s). The Thiele moduli were much less than 1, either for a single islet at the center of a capsule or CHO cells uniformly distributed throughout a capsule at 10(-6) cells/ mL, so that diffusion restrictions within the cells in EUDRAGIT RL or 800 mum HEMA-MMA capsules should be negligible. The ratio of external to internal diffusion resistance (Biot number) was less than 1, so that at most, only a small diffusion effect on glucose utilization should be expected (i.e., the overall effectiveness factors were greater than 0.8). These calculations were consistent with experimental observation of encapsulated islet behavior but not fully with CHO cell behavior. Permeability restricted cell viability and growth is potentially a major limitation of encapsulated cells; further analysis is warranted.  相似文献

Soviet doxycycline capsules in a dose of 0.1 g were studied in vitro (for disintegration and solubility) and in vivo (pharmacokinetics on 25 patients). It was found that Soviet doxycycline capsules provided comparatively rapid absorption of doxycycline. The antibiotic availability may be characterized by the solubility test. The results of the study on bioavailability of Soviet doxycycline capsules were compared with the results of analogous studies published by American authors for doxycycline capsules manufactured by various firms of the USA.  相似文献

适量烟气能促进种子萌发,但对苔藓植物孢子的作用尚不清楚.选取采自长白山区泥炭地的粗叶泥炭藓和中位泥炭藓的孢蒴为试验材料,通过燃烧泥炭地植物产生烟气,制备烟溶液,分别与不同大小(大:直径为2.10~2.50 mm;小:直径为1.50~1.90 mm)以及不同保存时长(旧:4.3和6.3年;新:0.3年)的孢蒴进行两组双因素试验,经不同时长的烟溶液浸泡和萌发试验,模拟研究烟气、孢蒴大小和保存时长对苔藓植物孢子萌发的影响.结果表明: 烟溶液浸泡影响孢子萌发,培养10 d时,不同时长的烟溶液浸泡均可使孢子的萌发率提高5倍以上,小孢蒴孢子的萌发率高;培养21 d时,仅适度浸泡(3 d)表现出促进萌发的作用,孢蒴大小对孢子萌发率无影响;烟溶液浸泡对长时间保存(4.3和6.3年)的孢蒴孢子萌发无促进作用.研究表明,适量烟气可加速新泥炭藓孢子以及小孢蒴孢子的萌发.在存在不定期火烧干扰的泥炭地中,与对种子植物的作用类似,烟气可能在苔藓植物种群的有性更新和种群维持中发挥重要作用.  相似文献

The effect of sucrose on fruiting, seed production, and seed germination of lesser centaury [Centaurium pulchellum (Sw.) Druce] was examined using explants of flowers and flower buds. Sucrose concentrations in the culture medium ranged from 0.003 to 0.3 M. It has been shown that the number of auxiliary buds, capsules dimension, number of viable seeds per capsule and seed dimensions increased with the increase of sucrose concentrations. The highest values were recorded at sucrose concentrations higher than 0.03 M, except for seeds size, which were larger at sucrose concentration ranging from 0.003 to 0.1 M. The germination of in vitro produced seeds was affected by previous culture history: a higher germination percentage was obtained in seeds that were raised from explants originally grown on medium with sucrose concentrations higher than 0.003 M.  相似文献

目的采用动物实验及临床观察,探讨益生菌制剂培菲康治疗肝硬化高氨血症的作用机制。方法采用大鼠肝硬化模型,其中治疗组予以培菲康悬液每天灌胃治疗共8周,检测治疗组及对照组血浆内毒素指标;并选用70例肝硬化Child-Pugh分级为B级的患者,其中治疗组口服培菲康胶囊(每次2粒,每日3次)3周,观察治疗前后血浆内毒素变化。结果培菲康治疗组的肝硬化大鼠及患者血浆内毒素均明显低于对照组(P<0.05)。结论培菲康能明显降低肝硬化患者的血氨水平,可作为肝硬化患者的辅助用药。  相似文献
