The role of organic acids in the mobilization of plant nutrients from the rhizosphere was assessed in seven contrasting soil types. The results indicated that malate was poor at mobilizing micronutrients from all the test soils, whilst citrate was capable of mobilizing significant quantities. Citrate was also capable of mobilizing P from one soil which possessed a large Ca-P fraction. This mobilization of P was due to both the complexing action of the citrate anion and due to the dissolution properties of the protons released from citric acid upon equilibrium with the soil solution. The reaction of citrate with cations was found to be near instantaneous with significant absorption to the solid phase in some soils at low concentrations. Soil decomposition studies indicated that citrate was rapidly broken down in organic soils but was more resistant to degradation in subsoil horizons. It was concluded that organic acids can be expected to be of little consequence in nutrient mobilization from high pH soils, whilst in acid soils they may be involved both in a more general mechanism for micronutrient uptake or as a potential Al detoxification mechanism.Abbreviations C6H8O7 Citric acid and H-citrate indicates - C6H5O7Na3 whilst Na-citrate indicates  相似文献

The present work highlights metal-organic acid interactions with special reference to their plant availability. Pot experiments were conducted to investigate the effect of various organic (carboxylic and amino) acids on the uptake and translocation of root-absorbed Cd by maize (Zea mays) plants grown in sand and soil culture. Statistically significant increases in Cd accumulation from Cd-treated plants in the presence of increasing concentrations of organic acids, suggest the existence of Cd-organic acid interactions in the soil-plant system. In order to support the above hypothesis of formation of organically bound Cd, separate experiments were performed to synthesize and estimate its various forms viz. cationic, anionic and neutral. The chemical nature of the organically bound forms was ascertained by electrophoretic experiments. Amino acids have been found to be less effective in the mobilisation of cadmium compared to carboxylic acids. The results are discussed on the basis of the potential of organic acids to form complexes with Cd.  相似文献

The importance of airborne plant litterfall as a source of organic material was evaluated for a small oligotrophic lake. The airborne plant litter input was estimated to be 100 kg yr–1 (45 kg C. yr–1), which represents approximately 5% of the phytoplankton productivity. Contribution of amino compounds to dissolved organic nitrogen in forest soils   总被引:16,自引:2,他引:16  
Dissolved organic nitrogen (DON) may play an important role in plantnutrition and nitrogen fluxes in forest ecosystems. In spite of the apparentimportance of DON, there is a paucity of information concerning its chemicalcomposition. However, it is exactly this chemical characterization that isrequired to understand the importance of DON in ecosystem processes. Theprimaryobjective of this study was to characterize the distribution of free aminoacidsand hydrolyzable peptides/proteins in the DON fraction of Oa horizon leachatesalong an extreme edaphic gradient in northern California. Insitu soil solutions were extracted by centrifugation from Oahorizonscollected beneath Pinus muricata (Bishop pine) andCupressus pygmaea (pygmy cypress) on slightlyacidic/fertile and highly acidic/infertile sites. DON accounted for 77 to99% of the total dissolved nitrogen in Oa horizon leachates. Nitrogen infree amino acids and alkyl amines ranged from 0.04–0.07 mgN/L on the low fertility site to 0.45–0.49 mg N/L onthe high fertility site, and accounted for 1.5 to 10.6% of the DON fraction.Serine, glutamic acid, leucine, ornithine, alanine, aspartic acid andmethylamine were generally the most abundant free amino compounds. Combinedamino acids released by acid hydrolysis accounted for 48 to 74% of theDON, suggesting that proteins and peptides were the main contributor to DON inOa horizon leachates. Together, nitrogen from free andcombined amino compounds accounted for 59 to 78% of the DON. Most of theDON was found in the hydrophobic fraction, which suggests the presence ofprotein/peptide-polyphenol complexes or amino compounds associated withhumic substances. Because free and combined amino acids can be an importantnitrogen source for some plants, soil DON may play an important role in plantnutrition and ecosystem function.  相似文献

5-Aminolevulinic acid (ALA), a heme precursor that accumulates in acute intermittent porphyria (AIP) and lead poisoning, undergoes enolization and subsequent iron-catalyzed oxidation at neutral pH. Iron is released from horse spleen ferritin (HoSF) by both ALA-generated O2•− and enoyl radical (ALA√), which amplifies the chain of ALA oxidation (autocatalysis). Iron chelators such as EDTA, ATP, but not citrate, and phosphate accelerate this process and ALA-promoted iron release from HoSF is faster in horse spleen isoferritins containing larger amounts of phosphate in the core. ALA (+0.377 V versus standard hydrogen electrode) is less effective in releasing iron from ferritin than are thioglycollic acid, 6-hydroxydopamine, and N,N,N′,N′-tetramethyl-p-phenylenediamine. During electrochemical one electron oxidation of ALA in a nitrogen atmosphere, spin trapping experiments with 3,5-dibromo-4-nitrosobenzenesulfonic acid demonstrated the formation of a spin adduct characterized by a six line signal, indicating a secondary carbon-centered radical and attributed to a resonant ALA√ radical. Iron is also released in such anaerobic electrochemical oxidations of ALA in the presence of ferritin, suggesting that, in addition to O2•−, ALA√ can promote iron mobilization from ferritin. Hence, ALA√ may amplify the metal-catalyzed oxidation of ALA, damaging ALA-accumulating cells and possibly contributing to the symptoms of porphyria.  相似文献

Assessment of host plant specificity is a critical step in the evaluation of classical biological control agents of weeds which is necessary for avoiding possible damage to non-target plants. Volatile organic compounds (VOCs) emitted by plants likely play an important role in determining which plants attract and are accepted by a prospective arthropod agent. However, current methods to evaluate host plant specificity usually rely on empirical choice and no-choice behavioural experiments, with little knowledge about what chemical or physical attributes are stimulating the insect. We conducted experiments to measure the quantitative and qualitative effects on emission of VOCs caused by simple mechanical damage to leaves of plants known to differ in suitability and attractiveness to a prospective agent. More VOCs were detected from damaged than from undamaged leaves for all three species tested. Discriminant analysis was able to correctly distinguish the taxonomic identity of all plants based on their VOC profiles; however, the VOCs that discriminated species among undamaged leaves were completely different from those that discriminated among damaged leaves. Thus, damaged and undamaged plants present different VOC profiles to insects, which should be considered when conducting host plant specificity experiments. An unacceptable non-target plant, Centaurea cineraria, emitted all except one of the VOCs that were emitted by its preferred host plant, Centaurea solstitialis, indicating the importance of compounds that are repellant in host plant specificity. Centaurea cyanus emitted fewer VOCs than C. solstitialis, which suggests that it lacked some VOCs important for host plant recognition.  相似文献

134Caesium chloride solution was injected into the surface of a peaty podzol at 3 cm depth with 5 cm spacing over a M2 at two upland sites, one of which had been fertilized and reseeded. The movement of radio caesium in the soil was subsequently followed by coring and/or taking out 10-cm square blocks at monthly intervals over a period of 2 years. There was very little movement of the caesium down the profile, with more than 95% remaining at the point of application. Lateral movement was also minimal. The caesium did not concentrate in the roots. Samples of herbage collected from the surface during the growing season showed a progressively decreasing concentration of caesium over the period. Total caesium removed in herbage amounted to 3.5 and 0.6% of the original present for the improved and umimproved soils, respectively.The extractability of the caesium from the 134Cs-amended peaty soil was compared with that from a low-ash peat which had been treated with the radionuclide for 2 months. Initially 1 M ammonium acetate (pH 7.2) extracted 30 and 56% of the Cs from 0–3 and 3–6 horizous of the improved peaty podzol, respectively. After 5 months only about 12 and 7% of the Cs was extracted by 1 M ammonium acetate and 4 and 1% by 0.1 M sodium pyrophosphate from the 0–3 and 3–6 cm horizons, respectively. After 24 months 1 M ammonium acetate extracted 1.6 and 3.3% of the Cs from the 0–3 and 3–6 cm horizons, respectively. In contrast, the values for the peat after 2 months were 100 and 80% for 1 M ammonium acetate and 0.1 M pyrophosphate, respectively.  相似文献

Carnitine is a zwitterion essential for the beta-oxidation of fatty acids. We report novel localization of the organic cation/carnitine transporter, OCTN1, to mitochondria. We made GFP- and RFP-human OCTN1 cDNA constructs and showed expression of hOCTN1 in several transfected mammalian cell lines. Immunostaining of GFP-hOCTN1 transfected cells with different intracellular markers and confocal fluorescent microscopy demonstrated mitochondrial expression of OCTN1. There was striking co-localization of an RFP-hOCTN1 fusion protein and a mitochondrial-GFP marker construct in transfected MEF-3T3 and no co-localization of GFP-hOCTN1 in transfected human skin fibroblasts with other intracellular markers. L-[(3)H]Carnitine uptake in freshly isolated mitochondria of GFP-hOCTN1 transfected HepG2 demonstrated a K(m) of 422 microM and Western blot with an anti-GFP antibody identified the expected GFP-hOCTN1 fusion protein (90 kDa). We showed endogenous expression of native OCTN1 in HepG2 mitochondria with anti-GST-hOCTN1 antibody. Further, we definitively confirmed intact L-[(3)H]carnitine uptake (K(m) 1324 microM), solely attributable to OCTN1, in isolated mitochondria of mutant human skin fibroblasts having <1% of carnitine acylcarnitine translocase activity (alternate mitochondrial carnitine transporter). This mitochondrial localization was confirmed by TEM of murine heart incubated with highly specific rabbit anti-GST-hOCTN1 antibody and immunogold labeled goat anti-rabbit antibody. This suggests an important yet different role for OCTN1 from other OCTN family members in intracellular carnitine homeostasis.  相似文献

有机酸不仅是碳代谢的中间体,而且在一些植物应对养分缺乏、金属胁迫以及根-土界面中操纵植物-微生物间的交互作用方面都发挥着关键作用.从植物营养学角度,对最近关于植物体内有机酸的形成与生理,及其与氮素代谢,磷和铁的吸收,铝的耐受以及土壤生态之间的关系等方面进行了总结,并对有机酸的跨膜运输、转基因模型中有机酸调控的生物技术操作的最新发现进行了讨论,以期为理解有机酸代谢的植物营养学基本原理提供基础.最后,还提出了通过生物技术,培育作物新品种,以更好地适应环境与金属胁迫.  相似文献

Summary Changes in the composition of organo-mineral particle size fractions as a result of cultivation of a grassland soil are discussed with reference to models of soil organic matter formation and turnover. The data presented indicated that physically stabilized organic matter is an important reservoir, with an intermediate turnover time, which is responsible for nutrient supply in agricultural soils. Possible mechanisms of stabilization and mobilization of organic matter are presented in the light of the arrangement of organic and inorganic components of the soil.  相似文献

Many parasitic wasps (parasitoids) exploit volatile organic compounds (VOCs) emitted by herbivore-infested plants in order to locate their hosts, but it remains largely unknown which specific compounds within the volatile blends elicit the attractiveness to parasitoids. One way of studying the importance of specific VOCs is to test the attractiveness of odor blends from which certain compounds have been emitted. We used this approach by testing the attraction of naive and experienced females of the two parasitoids Cotesia marginiventris and Microplitis rufiventris to partially altered volatile blends of maize seedlings (Zea mays var. Delprim) infested with Spodoptera littoralis larvae. Adsorbing filter tubes containing carbotrap-C or silica were installed in a four-arm olfactometer between the odor source vessels and the arms of the olfactometer. The blends breaking through were tested for chemical composition and attractiveness to the wasps. Carbotrap-C adsorbed most of the sesquiterpenes, but the breakthrough blend remained attractive to naive C. marginiventris females. Silica adsorbed only some of the more polar VOCs, but this essentially eliminated all attractiveness to naive C. marginiventris, implying that among the adsorbed compounds there are some that play key roles in the attraction. Unlike C. marginiventris, M. rufiventris was still attracted to the latter blend, showing that parasitoids with a comparable biology may employ different strategies in their use of plant-provided cues to locate hosts. Results from similar experiments with modified odor blends of caterpillar-infested cowpea (Vigna unguiculata) indicate that key VOCs in different plant species vary greatly in quality and/or quantity. Finally, experienced wasps were more strongly attracted to a specific blend after they perceived the blend while ovipositing in a host. Considering the high number of distinct adsorbing materials available today, this in situ modification of complex volatile blends provides a new and promising approach pinpointing on key attractants within these blends. Advantages and disadvantages compared to other approaches are discussed.  相似文献

Vertical distribution of organic constituents, i.e. total organic carbon (TOC), extractable organic carbon with ethyl acetate (EOC), hydrocarbons, phytol, sterols, fatty acids and phenolic acids in Lake Vanda was studied to elucidate their features in relation to the stratification of lake water and the distribution of lake organisms. The concentrations of TOC, EOC and sterols increased with depth and attained the maximum values of 25 and 1.5 mgC l–1 and 1.4 g l–1 in the bottom, respectively, while those of fatty acids showed the maximum value of 61 g l–1 at a depth of 55.4 m, along with the highest value of the ratio of unsaturated (UC16, uC18) to saturated (C16, C18) acids (8.5) and with the highest carbon preference index (35). Hydrocarbons were only found in the bottom layers (60.4 and 65.9 m) and bottom sediment. These results suggest strongly that the vertical distribution of lake organisms and their activity are quite different due to depth. In the bottom warm anoxic layers the degradation of organic materials must have occurred significantly and thus refractory organic materials should be concentrated.  相似文献

Bioreactors are often an integral part of dairy factory efforts to reduce the biological oxygen demand of their wastewater. In this study, infeed, mixed liquor and supernatant samples of an aerobic bioreactor used by a dairy factory in South-Eastern Australia were analyzed for nutrients and organic compounds using gas chromatography-mass spectrometry and physicochemical analyses. Despite different concentrations of organic inputs into the bioreactor, nutrients and trace organic compounds were reduced significantly (i.e. average concentration of trace organic compounds: infeed = 1681 μg/L; mixed liquor = 257 μg/L; supernatant = 23 μg/L). However, during one sampling period the bioreactor was adversely affected by the organic loading. Trace organic compounds in the samples were predominantly fatty acids associated with animal products. The analyses suggest that it is possible to trace a disruptive input (i.e. infeed with high organic carbon concentrations) into an aerobic bioreactor by measuring concentrations of fatty acids or ammonia.  相似文献

Bacterial utilization of dissolved organic matter (DOM) was studied in water from a humic and a clearwater oligotrophic lake. Indigenous bacteria were inoculated into either 0.2 m natural filtered lake water, or lake water enriched fivefold with colloidal DOM >100 kD but below 0.2 m. Consumption of DOM was followed from changes in concentrations of total dissolved organic carbon (DOC), dissolved combined and free carbohydrates and amino acids (DCCHO and DFCHO, and DCAA and DFAA, respectively) and by uptake of monosaccharide and amino acid radioisotopes. DCCHO and DCAA made up 8% (humic lake) to 33–44% (clear-water lake) of the natural DOC pools, while DFCHO and DFAA contributed at most 1.7% to the DOC pools. Addition of >100 kD DOM increased the DOC concentrations by 50% (clearwater lake) to 92% (humic lake), but it only resulted in a higher bacterial production (by 63%) in the humic lake. During the incubations 13 to 37% of the DOC was assimilated by the bacteria, at estimated growth efficiencies of 4–8%. Despite the measured reduction of DOC, statistically significant changes of specific organic compounds, especially of DCCHO and DCAA, generally did not occur. Probably the presence of high molecular weight DOC interfered with the applied analytical procedures. Addition of radiotracers indicated, however, that DFAA sustained 17–58% and 29–100% of the bacterial carbon and nitrogen requirements, respectively, and that glucose met 1–3% of the bacterial carbon requirements. Thus, our experiments indicate that radiotracers, rather than measurements of concentration changes, should be used in studies of bacterial utilization of DOC in freshwaters with a high content of humic or high molecular weight organic matter.  相似文献

Fitting a double negative exponential function to N mineralization data can be used to characterize two organic nitrogen pools; an easily decomposable (Ndpm) and a resistant one (Nrpm). The relevance of those two calculated N mineralization pools was investigated by adding easily decomposable organic material to soils. Soil amended with crop residues of sugar-beet or bean was mixed with an equal amount of coarse sand, incubated at 35 °C and leached at specific time-intervals. Upon leaching, NH4 + and NO3 - were measured in the extracts. A double negative exponential function was fitted to the data and two organic N pools were defined. Fitting a double negative exponential function to N mineralization data to characterize an active and resistant organic N pool was sometimes impossible; the N mineralization data did not always resemble a negative exponential function. Additionally, the size of the two pools calculated were not constant with time and were often meaningless; the Nrpm pool was greater than the soil organic N content, the size of the Nrpm pool was smaller than the Ndpm pool or one of the N pools was negative. Relevant values for both Nrpm and Ndpm which were consistent with incubation time were only obtained when excessive amounts of organic material, normally not dealt with in the field, were applied.  相似文献
