 Following artificial inoculation of nonhost Populus balsamifera with Ophiostoma ulmi, structural defensive tissues were formed in the xylem. Among these tissues there was a perimedullary sheath of cells, located adjacent to the invaded xylem, that originated from the dedifferentiation of perimedullary and xylem parenchyma cells. Histochemical tests revealed that this sheath was intensively suberized. A band of lignified cells was frequently detected on both sides of this suberized tissue. The formation of such a tissue at the pith margin represents a new type of anatomical barrier in relation to compartmentalization processes described for trees. Ultrastructural examination showed that the wall of cells forming this zone was generally composed of a compound middle lamella, a suberized secondary wall and a tertiary wall layer. Using colloidal gold conjugated to monoclonal antibodies against pectin and to an exoglucanase for cellulose, only limited labelling was obtained for pectin whereas labelling for cellulose was abundant in the compound middle lamella and the tertiary wall layer. In a few fibres close to this suberized zone, the latter probe also made it possible to distinguish the occasional presence of several alternating wall layers mainly composed of either suberin or cellulose. In Salix sp., another tree species belonging to the Salicaceae, this type of suberized reaction zone was also observed. The new reaction zone is similar in structure and location to a suberized barrier formed in nonwoody carnation (Dianthus caryophyllus) plants in the defense against vascular fungi. Shoot pruning could cause short-term damages to vines. In response to damage, tyloses develop in shoot xylem vessels interfering free water and mineral transportation. In this study, the tylosis development at different nodes of the current-year and perennial shoots of sixty three-year-old grapevines (Vitis vinifera L.) after pruning was investigated. The results showed that tyloses at the nodes closest to the trunk developed rapidly; and tylosis development initiated at the time when the size of vessel-ray pit was greater than that of parenchyma cell-parenchyma cell pit. In current-year shoots, tyloses were formed in up to 87% of the vessels, and 40% of the vessels were completely blocked by tyloses. In wound-induced perennial shoots, 30% of the vessels were completely blocked by tyloses. When few vessels were blocked by tyloses, new vessels could differentiate, and water transportation system may be restored. However, when tyloses developed in a large number of vessels and the large number of the vessels were blocked, the original capability of water transport was decreased (the largest decrease was 21.1% in this study), resulting in dehiscence or shrinkage cracking in this area. The study proved that the tylosis formation in functional vessels limited the water transport efficiency.  相似文献

Summary The cell wall in laticifers of theConvolvulaceae, Calystegia silvatica, C. soldanella, C. tuguriorum, Convolvulus cneorum, C. verecundus, C. sabaticus subsp.mauritanicus, andIpomoea indica, contains an impregnated layer that surrounds the cells. The impregnated layer lies inside the primary wall of the laticifer, separated from the protoplast by a third (tertiary) layer of variable thickness. Histochemical and cytochemical staining give a positive reaction for suberin. The layer is often differentiated into dark and translucent regions, the latter frequently being composed of lamellae. The ultrastructure of this layer and its position within the cell wall of the laticifer is comparable to the condition found in oil cells where the walls contain a suberized layer. A suberized layer within the wall is unique for a laticifer system.  相似文献

Cell walls of the periderm of native potato tuber (Solanum tuberosum L. cv. Primura) consist of a primary wall, a suberized secondary wall and a tertiary wall. With a mixture of pectinase and cellulase intact periderm membranes can be isolated. Isolation does not affect fine structure. It is suggested that the lignin in the middle lamellae and primary walls prevents the enzymes from digesting pectinaceous materials and cellulose. In specimens fixed with OsO4, the suberized walls appear as alternating electrondense and electron-lucent lamellae. This lamellar architecture is not altered by extraction with chloroform. Therefore, the current view that the electronlucent lamellae consist of soluble lipids (waxes) can no longer be maintained. It is argued that the lamellation is a property of the suberin itself, and the suberized wall consists of alternating layers of suberins differing in polarity. A hypothesis of suberin assembly from sub-units is advanced and the subunits are shown for the first time.  相似文献

During a study of transpiration and embolism-formation in petiolesof sunflower, tyloses were frequently observed in early metaxylemvessels. Tyloses were confined to the inner ends of the xylemarcs, remote from the phloem. Vessels in this position are especiallyvulnerable to embolism. All stages of the invasion of vessellumens by xylem parenchyma cells were observed, from the earlyprotuberance of a cell through a pit to the complete occlusionof the lumen by one to several cells. The lumen space not occupiedby tyloses was seen both filled with xylem sap, or embolizedand gas-filled. Thus, during the early stages of tylosis formationthe vessel remained active in carrying the transpiration stream.Thin-walled vessels of the protoxylem or early metaxylem werenot tylosed, but were squashed and disappeared. These observationsare interpreted as evidence that vessels vulnerable to embolismare decommissioned and replaced by parenchyma tissue, whilenew and less vulnerable vessels are added to the xylem arcsat the cambial side. It is proposed that tylosis formation istriggered by the frequent embolization of the vulnerable vesselsto give, ultimately, an incompressible tissue. Then tyloseswould be necessary to preserve the tissue pressure which expresseswater to refill embolisms in the remaining vessels, and maintaintranspiration, as explained by the compensating pressure theoryof water transport. Summary Styloid calcium oxalate crystal idioblasts of Agave americana L. are suberized. Where the crystals do not touch the cell wall directly they are enclosed in a suberinic sheath which is connected with the suberinic wall layer. No polysaccharides are laid down as a tertiary wall layer, nor could any polysaccharides be found in the crystal sheath. These results contradict those of Arnott (1973) but agree fully with those of Rothert and Zalenski (1899).  相似文献

Styloid-calcium-oxalate-crystal-containing idioblasts possess an interior cell-wall layer which has a lamellar ultrastructure. Idioblasts were isolated by centrifugation of an Agave americana leaf homogenate through 2M sucrose. The aliphatic monomers of the polymeric material from an idioblast fraction were primarily -hydroxy acids (32%) and dicarboxylic acids (35%), with C18:1 dicarboxylic acid being the most dominant monomer (25%). Nitrobenzene oxidation of the idioblasts yielded syringaldehyde and vanillin in a ratio of 0.46:1. The major class of wax associated with the idioblasts was free fatty acids (34%). A major homologue of both the fatty acid and fatty alcohol fractions of this wax was C22. The hydrocarbon fraction of the wax had a broad chainlength distribution with a large amount of even-numbered (47%) and shorter-chain homologues. The ultrastructure, the composition of the aliphatic and aromatic components of the polymeric material as well as the composition of the wax show that the idioblast cell wall is suberized. As a first step towards studying the biochemical relationship between Golgi vesicles (GVs) and tube wall components, isolation of GVs from the growing pollen tubes ofCamellia japonica was attempted using a centrifugation method with mannitol. The isolated GV was identified ultrastructurally and immunocytochemically. The main components of the GV were proteins and carbohydrates. The main monosaccharides of GV polysaccharides were galactose, arabinose and uronic acid, and pectins and arabinogalactan proteins also were detected immunochemically. An antiserum against the isolated GVs reacted with the outer layer of the pollen tube wall and the intine layers of the grain wall as well as thein situ GVs in the pollen tube and the grain cytoplasm. We have thus successfully isolated GVs and shown that they contain pectic substances and arabinogalactan proteins which contribute to formation of the pollen tube primary wall.  相似文献

菰(Zizania latifolia)是一种多年生挺水植物,为了探讨该植物根、茎和叶的解剖结构、组织化学及其质外体屏障的通透性生理。该文利用光学显微镜和荧光显微镜,对菰的根、茎、叶进行了解剖学和组织化学研究。结果表明:(1)菰不定根解剖结构由外而内分别为表皮、外皮层、单层细胞的厚壁机械组织层、皮层、内皮层和维管柱;茎结构由外而内分别为角质层、表皮、周缘厚壁机械组织层、皮层、具维管束的厚壁组织层和髓腔。叶鞘具有表皮和具维管束皮层,叶片具有表皮,叶肉和维管束。(2)不定根具有位于内侧的内皮层及其邻近栓质化细胞和外侧的外皮层组成的屏障结构;茎具内侧厚壁机械组织层,外侧的角质层和周缘厚壁机械组织层组成的屏障结构,屏障结构的细胞壁具凯氏带、木栓质和木质素沉积的组织化学特点,叶表面具有角质层。(3)菰通气组织包括根中通气组织,茎、叶皮层的通气组织和髓腔。(4)菰的屏障结构和解剖结构是其适应湿地环境的重要特征,但其茎周缘厚壁层和厚壁组织层较薄。由此推测,菰适应湿地环境,但在旱生环境中分布有一定的局限性。  相似文献

Summary The structure of the phloem was studied in stem and leaf ofArtemisia afra Jacq., with particular attention being given to the sieve element walls. Both primary and secondary sieve elements of stem and midvein have nacreous walls, which persist in mature cells. Histochemical tests indicated that the sieve element wall layers contained some pectin. Sieve element wall layers lack lignin. Sieve elements of the minor veins (secondary and tertiary veins) lack nacreous thickening, although their walls may be relatively thick. These walls and those of contiguous transfer cells are rich in pectic substances. Transfer cell wall ingrowths are more highly developed in tertiary than in secondary veins.  相似文献

As the density and species composition of insects may change in relation to distance from the forest edge, the role of herbivory in tree establishment may also change across edges. To determine the importance of insect herbivory in tree establishment, insect densities were experimentally altered at different distances from the forest edge. Plots were established at three distances from the edge, with plots located in forest, edge, and field habitats. In half of each plot, insect densities were reduced by insecticide application. Seeds of two tree species, Acer rubrum and Fraxinus americana, were planted into each plot in 1995. The experiment was repeated in 1996 with the addition of Quercus palustris and Quercus rubra.Distance from the forest edge was the most important factor in determining seedling emergence and mortality. Overall seedling performance increased from field to edge to woods, although responses varied among species. In 1995, a drought year, insect removal increased emergence and decreased mortality of tree seedlings. In 1996, a year with normal precipitation, insect removal had much less effect on A. rubrum and F. americana. For the two Quercus species, mortality was reduced by insect removal. The tree species differed in their susceptibility to insect herbivory, with Acer rubrum the most susceptible and Fraxinus americana the least. Herbivory by insects was shown to have the potential to affect both the composition and spatial pattern of tree invasions. Herbivore importance differed greatly between the two years of the study, making the interaction between insects and tree seedlings variable both in space and time.  相似文献

Summary The development of mestome sheath cells ofAegilops comosa var.thessalica was studied by electron microscopy. Anatomical and cytological observations show that this grass belongs to the C3 or non-Kranz plants. In the asymmetrically thickened walls of mestome sheath cells a suberized lamella is present. This lamella is deposited asynchronously. In the midrib and the large lateral bundles it appears first in the outer and inner walls and usually later in the radial walls. In the small lateral bundles its appearance is delayed in the inner walls of those cells situated on the xylem side. At maturity the suberized lamella is observed in all cell walls; however, in the small lateral bundles it is partly or totally absent from the walls of some cells situated on the xylem side. Tertiary wall formation is asynchronous as well, for it generally follows the deposition pattern of the suberized lamella.During the development of the mestome sheath cells microtubules show marked changes in their number and orientation, being fewer and longitudinal during suberin deposition. Dictyosomes are very active and may be involved in primary and tertiary wall formation. Endoplasmic reticulum cisternae are abundant and partly smooth, while plasmalemmasomes may function to reduce the plasmalemma extension. However, cytoplasmic structures that are clearly involved in suberin synthesis could not be identified.Suberized lamellae react strongly with silver hexamine. This is probably due to post-fixation with osmium tetroxide.On the basis of structural characteristics the mestome sheath may be regarded as an endodermis (cf., alsoFahn 1974). The significance of this view for water and assimilate exchange between the mesophyll and the bundle is discussed.This report represents a portion of a doctoral dissertation.  相似文献

Summary Wound responses of xylem parenchyma by suberization were investigated in some hardwoods by light and electron microscopy. Suberized ray and axial parenchyma cells form a distinct boundary around the wound in all investigated species. Vessels and fibres within and close behind the suberized area appeared more or less occluded; vessels in Fagus, Quercus, and Populus contained suberized tyloses, those in Betula and Tilia contained amorphous and fibrillar deposits. A common mechanism for suberin deposition in the parenchyma cells became evident. Cisternae of the endoplasmic reticulum were apparently involved in suberization. Suberin compounds are extruded by cytoplasmic vesicles, which fused with the plasma membrane, in order to release their content. The suberin layer exhibited the typical lamellated structure; cytoplasmic continuity between suberized cells by plasmodesmata was maintained through the suberin layer. Fagus revealed the most intense suberized area as compared with the other species. Within the reaction zone of Fagus and Quercus, some individual ray and axial parenchyma cells exhibited a subdivision into 2 or 3 compartments prior to suberization. Subdivision was achieved by the formation of a primary wall-like layer. Subsequently, the compartments became individually suberized. Wounding during winter did not induce suberization. Also, samples wounded and kept under water during the vegetation period showed no response. The role of suberization in the effectivity of wound-associated compartmentalization is discussed.  相似文献

In vitro culture of hypocotyl explants from Kandelia candel, a common mangrove species, on hormone-free Murashige and Skoog (MS) medium resulted in shoot formation. Since the hypocotyls showed good potential for in vitro shoot multiplication, the process of bud primordium formation was analyzed from a histological viewpoint. A wound periderm first appeared at the top, exposed cut surface of the explants. The wound-induced meristem continued to divide giving rise to suberized cells oriented towards the cut surface. After formation of the suberized cell layers, the meristem and its inner derivatives differentiated into multilayered, uniformly packed parenchyma cells. Bud primordia differentiated from the dense cytoplasmic cells of the wound-induced meristem just beneath the suberized layer near the severed vascular bundles. Each explant produced several visible shoot buds. Furthermore, histological sections revealed that additional bud primordia were present within the explant just underneath the suberized cells and that these bud primordia appeared to be arrested in their development. The fact that additional bud primordia were present within the explant suggests that further manipulation of the explant is helpful to maximize the potential of this system.  相似文献

该研究以云南箭竹不同年龄段的假鞭为实验材料,采用滑动切片法并利用光学显微镜观察,分析云南箭竹假鞭的解剖结构特征及其随年龄的动态变化,为假鞭结构研究提供新的解剖学数据信息。结果显示:(1)云南箭竹假鞭节间的表皮层只有1层细胞,皮下层由3~4层细胞壁加厚的纤维细胞组成,皮层一般有20~25层不规则的薄壁细胞,成熟的皮层细胞会形成皮层气道,髓实心不具髓腔。(2)云南箭竹假鞭纤维壁厚随鞭龄增加而增加,且同一年龄假鞭的内侧韧皮部面积大于外侧;纤维腔径随鞭龄增加而逐渐减小,但同一年龄假鞭内侧纤维腔径大于外侧;韧皮部的面积、维管束和导管的直径均随着鞭龄的增加而增大。(3)假鞭维管束一般不具有原生导管,外部维管通常有2个较大的后生导管,在假鞭中部及内部通常只有1个后生导管,另1个后生导管不发育或发育不全。(4)在0.5年生到2年生的云南箭竹假鞭中,被染成紫红色的木质素在纤维细胞壁、薄壁细胞壁、导管细胞壁中都有分布,且随着假鞭年龄的增加染色逐渐加深,表明云南箭竹假鞭木质素含量随着鞭龄的增长而不断增加,木质化程度随鞭龄的增长逐渐提高。  相似文献

Phaseolus vulgaris seedlings were grown in light with or without chromium. Changes in cell wall components i.e. pectic polysaccharides and xyloglucan contents were looked into during cell elongation, by two different methods in order to find the most suitable method for isolation of cell wall polysaccharides. The first method was short and easy. It made use of organic solvents for preparation of cell wall components and ammonium oxalate and oxalic acid buffer and high temperature for extracting pectic polysaccharides; 0.7 M and 4.3 M KOH was used for extracting low and high molecular weight xyloglucans respectively. On the other hand, in the second method, cell wall components were fractionated by sequential treatments with different inorganic solvents, chelating agents, sodium lauryl sulphate, etc. KOH (1 M and 4 M) was used for extracting xyloglucans. The advantage of using the second method for extracting cell wall polysaccharides especially pectic polysaccharides is discussed.  相似文献

为揭示海桑次生木质部导管解剖特征随土壤理化因子年内动态变动而变化的适应机制,该研究利用海桑具有生长轮的特点,通过显微技术界定了海桑采样枝条一年内10个不同连续时间段形成的新"生长层"(新形成的次生木质部),观测了10个不同连续时间段新"生长层"的导管解剖特征,并对10个新"生长层"形成阶段所对应的土壤理化因子数量特征进行了测定,用逐步回归法分析了10个不同连续时间段海桑新"生长层"管孔数量解剖特征与对应土壤理化因子数量特征之间的关系。结果显示:(1)10个不同连续时间段海桑形成新"生长层"管孔数量特征指标,除相邻管孔间接触壁长占比无显著差异外,其他8项指标包括管孔径向直径、管孔弦向直径、导管壁厚、导管长度、管孔密度、单孔率、导管聚合度和相邻管孔间接触壁长等均具有显著差异(P0.05);多重比较显示,10个不同连续时间段海桑形成新"生长层"管孔数量特征8项指标具不同程度的变动。(2)海桑新"生长层"形成阶段(10个不同连续时间段)土壤理化因子,包括土壤有机质含量、土壤全氮含量、土壤全磷含量、土壤pH值和土壤全盐量等5项指标均具有显著差异(P0.05);多重比较显示,海桑新"生长层"形成阶段(10个不同连续时间段)土壤理化因子指标均具有不同程度变动。(3)10个不同连续时间段海桑形成新"生长层"的管孔数量解剖特征,与所对应新"生长层"形成阶段土壤理化因子数量特征的逐步回归分析表明,随着土壤全盐含量的升高,海桑导管弦向直径和导管聚合度同时呈显著增大趋势(P0.05)。研究表明,海桑在一年内不同连续时间段,随土壤全盐量增加,土壤渗透势将增大,水分在次生木质部导管中输导的安全性将下降,而海桑导管分子随一年内不同连续时间段土壤全盐量的增加呈增大趋势,根据木材生态解剖学的观点,水分输导的安全性将进一步降低,但导管聚合度随一年内不同连续时间段土壤全盐量的增加而增大,具有增进水分输导安全性的作用,这可能是海桑对土壤盐含量变化的生态适应策略。  相似文献

The internodes of Merremia emarginata plant showed exceptionally high stretchability throughout the development period. Therefore, it provides excellent material to study the changes undergoing during cell elongation. In this study, the influence of the hormone treatments (GA3, PAA and NAA) on the wall component synthesis was analyzed in relation to elongation growth during internode development. A clear increasing trend of wall components was observed with increase in internode length. The non-esterified pectic substances were markedly correlated with internode length while esterified pectic substances showed correlation only in hormone treated internodes. Low molecular weight xyloglucans content showed correlation only in GA3 and NAA treated internodes, while high molecular weight xyloglucans were significantly correlated with length of internodes treated with PAA and NAA.  相似文献

Eight patients with diffuse panbronchiolitis (DPB) who had repeated intractable airway infections were continuously treated withNocardia rubra cell wall skeleton (N-CWS), a biological response modifier. As a result, subjective symptoms were reduced in 6 patients. Antibiotics therapy could be discontinued completely in two patients and the dose of antibiotics could be reduced considerably in two other patients. No adverse reactions in relation to N-CWS were observed.
