Communities, populations and individuals of arbuscular mycorrhizal fungi   总被引:5,自引:1,他引:4  
Arbuscular mycorrhizal fungi in the phylum Glomeromycota are found globally in most vegetation types, where they form a mutualistic symbiosis with plant roots. Despite their wide distribution, only relatively few species are described. The taxonomy is based on morphological characters of the asexual resting spores, but molecular approaches to community ecology have revealed a considerable unknown diversity from colonized roots. Although the lack of genetic recombination is not unique in the fungal kingdom, arbuscular mycorrhizal fungi are probably ancient asexuals. The long asexual evolution of the fungi has resulted in considerable genetic diversity within morphologically recognizable species, and challenges our concepts of individuals and populations. This review critically examines the concepts of species, communities, populations and individuals of arbuscular mycorrhizal fungi.  相似文献

Brown–black (or dematiaceous) fungi are responsible for a wide variety of infectious syndromes in both immunocompetent and immunocompromised patients, including local infections, allergic disease, pneumonia, brain abscess, and disseminated infection. They are distinct from the more common hyaline molds, Aspergillus and Fusarium, that cause human disease. They are often found in soil and generally distributed worldwide. In recent years, these fungi have been increasingly recognized as important pathogens. Dematiaceous fungi may have unique pathogenic mechanisms owing to the presence of melanin in their cell walls, which imparts the characteristic dark color to their spores and hyphae. Diagnosis rests on careful microscopic and pathologic examination, as there are no specific laboratory tests to reliably identify these fungi. Therapy depends upon the clinical syndrome, although disseminated infection often has high mortality. Triazoles such as voriconazole, posaconazole, and itraconazole are the most active antifungal agents available. Further studies are needed to better understand the pathogenesis and optimal treatment of these uncommon infections.  相似文献

Muscina angustifrons (Diptera: Muscidae) is a mycophagous species that exploits a variety of fungi, including ectomycorrhizal fungi. Larvae of this species have been shown to feed on sporocarps (including spores), and full-grown larvae leave sporocarps and pupate 0–6?cm below the soil surface. In this study, we examined whether M. angustifrons larvae are capable of transporting ectomycorrhizal fungal spores and enhancing ectomycorrhiza growth on host-plant roots. Full-grown larvae were found to move horizontally 10–20?cm from their feeding sites and burrow underground. These wandering larvae retained ectomycorrhizal fungal spores in their intestines, which were excreted following relocation to underground pupation sites. Excreted spores retained germination and infection capacities to form ectomycorrhiza on host-plant roots. In the infection experiments, ectomycorrhizal fungal spores applied in the vicinity of underground host-plant roots were more effective in forming ectomycorrhiza than those applied to the ground surface, suggesting that belowground transportation of spores by M. angustifrons larvae could enhance ectomycorrhizal formation. These results suggested that M. angustifrons larvae act as a short-distance spore transporter of ectomycorrhizal fungi.  相似文献

The appendages of Clostridium bifermentans UK-A 1003 spores were shown to originate from a substance located just exterior to the outer forespore membrane. The dense spore coat develops along the periphery of this material, and, as the appendages develop in the cytoplasm, the coalescing spore coat intervenes between the appendages and their origin. Freeze etching revealed that the appendages are in the form of distinct fibers in proximity to the mature spore body. These fibers form a network around the spore, seemingly encasing it and insuring that the appendages remain attached to the mature, free spore. The inner wall of each appendage tubule is lined with fibers whereas the outer surface is smooth. The developing exosporium contained several layers consisting of small (3 nm) globular subunits; the outer exosporial surface is composed of relatively unstructured material.  相似文献

Trichoduboscqia epeori Léger was found to parasitize nymphs of the mayfly Rhithrogena iridina Kolenati in southwest Germany for a new host record. It was studied by light and electron microscopy. The pansporoblast membrane is evaginated at several points, usually four, to produce long needle-like appendages >20 μm in length with a resilient inner core superficially resembling collagen, which is thought to maintain their orientation. It is suggested that the pansporoblast appendages may play a role in host infection. The structure and ultrastructure of developmental stages are recorded for the first time. Apart from the pansporoblast appendages, the ultrastructure of T. epeori conforms to the general pattern seen in many other pansporoblastic Microspora. Typically 16 spores are produced per pansporoblast but 32-spore pansporoblasts were also found, and the taxonomic significance of this is discussed.  相似文献

AIMS: Further characterization and comparison of spore appendages from Bacillus cereus strains. METHODS AND RESULTS: Appendages were isolated from 10 B. cereus strains from the food industry and food-borne outbreaks. The appendage proteins were dissolved in sample buffer containing 2% SDS and 5% mercaptoethanol at 100 degrees C, and subjected to SDS-PAGE. None of the appendages showed identical protein patterns. Western blots, using antibodies raised against a 3.5 kDa appendage protein, showed that the majority of the appendage proteins reacted with the antibody. Removal of the appendages by sonic treatment of the spores did not alter their heat resistance. The appendages were digested by proteinase K, pepsin, and the enzymes in the detergent Paradigm 10, but not by trypsin or chymotrypsin. Spore adhesion to stainless steel was scarcely affected by removal of the appendages. Digestion of adhered intact spores (with appendages) with Paradigm 10 showed a high degree of variation. CONCLUSIONS: Spore appendages from B. cereus are complex proteinaceous structures that differ among strains. SIGNIFICANCE AND IMPACT OF THE STUDY: Information about spore appendages and their involvement in spore adhesion is crucial for improving cleaning methods used for control of bacterial spores in the food industry.  相似文献

Birds are important biotic dispersers of a wide range of propagules. Fungi spores are mainly dispersed by wind. Nevertheless there are several animals known to disperse fungi spores, which might be particularly important if spores are delivered to particularly favourable sites i.e. directed dispersal. This may be especially important for fungi that require specific microsites such as flowers. We sampled birds for the presence of fungi spores and pollen grains during one year at two forest sites in central Portugal. We found that out of the 894 birds sampled, 131 individuals from 11 species carried spores from at least 6 morphological types, mainly during winter. The great majority of birds found to carry fungi spores was also found to carry pollen grains, suggesting that they were feeding on flowers which are the main origin of the spores. This co‐dispersion of pollen and fungi spores suggest that the latter are not randomly dispersed on the environment, but are likely to have an increased probability of being deposited on flowers propitious to fungi development. Our results suggest that directed dispersal of fungi by flower‐visiting birds might by a common and under‐appreciated phenomenon with potentially important ecological, biogeographic and even economic outcomes.  相似文献

Laccases occur widely in fungi; they have been characterized less frequently in higher plants. Here we have focused on more recent reports on the occurrence of laccase and its functions in physiological development and industrial utility. The reports of molecular weights, pH optima, and substrate specificity are extremely diverse. Conclusive proof of the occurrence of laccase in a tissue must demonstrate that the enzyme be able to oxidize quinol with concomitant uptake of oxygen. Laccase is involved in the pigmentation process of fungal spores, the regeneration of tobacco protoplasts, as fungal virulence factors, and in lignification of cell walls and delignification during white rot of wood. Commercially, laccases have been used to delignify woody tissues, produce ethanol, and to distinguish between morphine and codeine. A very wide variety of bioremediation processes employ laccase in order to protect the environment from damage caused by industrial effluents. Research in recent years has been intense, much of it elicited by the wide diversity of laccases, their utility and their very interesting enzymology.  相似文献

Patterns of fungal spore dispersal affect gene flow, population structure and fungal community structure. Many Basidiomycota produce resupinate (crust-like) basidiocarps buried in the soil. Although spores are actively discharged, they often do not appear to be well positioned for aerial dispersal. We investigated the potential spore dispersal mechanisms of one exemplar of this growth form, Tomentella sublilacina. It is a widespread ectomycorrhizal fungus that sporulates in the soil organic horizon, can establish from the spore bank shortly after disturbance, but also can be a dominant species in mature forest stands. We investigated whether its spores could be dispersed via spore-based food webs. We examined external surfaces, gut contents and feces from arthropod fungivores (mites, springtails, millipedes, beetles, fly larvae) and arthropod and vertebrate predators (centipedes, salamanders) from on and around T. sublilacina sporocarps. Spore densities were high in the guts of many individuals from all fungivore groups. Centipede gut contents, centipede feces and salamander feces contained undigested invertebrate exoskeletons and many apparently intact spores. DAPI staining of spores from feces of fungivores indicated that 7-73% of spores contained intact nuclei, whereas spores from predators had lower percentages of intact nuclei. The spiny spores often were lodged on invertebrate exoskeletons. To test the viability of spores that had passed through invertebrate guts we used fecal droppings of the millipede Harpaphe haydeniana to successfully inoculate seedlings of Pinus muricata (Bishop pine). These results indicate the potential for T. sublilacina spore dispersal via invertebrates and their predators in soil food webs and might help to explain the widespread distribution of this species. It is likely that this is a general mechanism of dispersal for fungi producing resupinate sporocarps, indicating a need to develop a fuller understanding of the linkages of soil food webs and spore dispersal.  相似文献

Arbuscular mycorrhizal fungi is an important group of soil microorganisms which form beneficial symbiotic associations with roots with a wide range of plants thus improving plant growth, nutrition and health. This paper reviews the current status of preparation and formulation of mycorrhizal inoculum applying polymer materials with determined characteristics. The most widely used methods are based on the entrapment of fungal materials in natural polysaccharide gels. The potential of such inoculant preparations is illustrated by various studies which include immobilization of mycorrhized root pieces, vesicles and spores, in some cases co-entrapped with other plant beneficial microorganisms. Suggestions for further research in this field are also discussed.  相似文献

Filamentous fungi comprise an industrially very important collection of microorganisms, since they are used for the production of a wide variety of products ranging from primary metabolites to secondary metabolites and further on to industrial enzymes (such as proteases, lipases and antibiotics). It is known that fungal morphology is often considered as one of the key parameters in industrial production. For the production of fungal metabolite products, the desired morphology varies from one product to another. Many parameters affect the morphology of fungi during the process of fermentation, among them speed of agitation, specific growth rate, dissolved oxygen, number of spores or conidia per liter of fermentation broth are important and should be considered when higher yield is desired in the process. It is, therefore, of considerable importance to understand the mechanism underlying the morphology of the cell, its growth and product formation by filamentous fungi. Such knowledge may be used in the optimization of the microbial process. Several literatures with various fungi to study their morphology, relating enzyme or product production to the character of the fungi in the study is reviewed. It is also considered that how the process parameters affects the morphology. The aim of this communication is to review the relevant literature to understand the morphology of filamentous fungi.  相似文献

Hydrophobins are surface active proteins produced by filamentous fungi. They have a role in fungal growth as structural components and in the interaction of fungi with their environment. They have, for example, been found to be important for aerial growth, and for the attachment of fungi to solid supports. Hydrophobins also render fungal structures, such as spores, hydrophobic. The biophysical properties of the isolated proteins are remarkable, such as strong adhesion, high surface activity and the formation of various self-assembled structures. The first high resolution three dimensional structure of a hydrophobin, HFBII from Trichoderma reesei, was recently solved. In this review, the properties of hydrophobins are analyzed in light of these new data. Various application possibilities are also discussed.  相似文献

Some physical, chemical, and immunological properties of filamentous appendages and the exosporium on the spores of Bacillus cereus were examined for the purpose of elucidating the origin of filamentous appendages. The main components of both filamentous appendages and the exosporium were protein and their amino acid compositions were similar in point of a high content of glycine, alanine, threonine, valine, and acidic amino acids and a low content of basic and sulphur-containing amino acids. Treatment with 1 N NaOH at 50 C solubilized the isolated appendages completely and the isolated exosporia partially. In both preparations the solubilized proteins consisted of highly acidic monomeric subunits with molecular weights between 2,000 and 5,000. Treatment of the spores with 2% 2-mercaptoethanol at 37 C resulted in the isolation of long filamentous appendages without segmentation. When the spores were treated with 10% 2-mercaptoethanol, there was partial destruction of the exosporium as well as detachment of the filamentous appendages. There was a common antigenic component in the exosporium and the tips of the filamentous appendages. Five strains of B. cereus having a common appendage antigen also had a common exosporium antigen, whereas six other strains had neither a common appendage antigen nor a common exosporium antigen. From these facts it was concluded that the filamentous appendages arose from the exosporium.  相似文献

The atmosphere in Tulsa, Oklahoma was monitored for the presence of smut teliospores using a Burkard Volumetric Spore Trap. Smut spores were identified in the atmospheric samples every day from May to October during 1991 and 1992 at concentrations that were normally below 1000 spores/m3. The peak concentration observed during this study was almost 6000 spores/m3. Daily concentrations fluctuate due to a variety of factors such as precipitation, relative humidity, percent sunshine, and the phenology of fungi in relation to their hosts. In northeastern Oklahoma, the most prevalent species of smuts in the atmosphere during the spring includeSphacelotheca occidentalis, Ustilago tritici, andU. kolleri. Twenty-five strains of Clostridium sordellii were divided into two groups on the basis of spore fine structure. Sixteen strains formed spores with smooth tubular appendages, and nine strains formed spores which lacked appendages. The other properties of the 25 strains were relatively constant. Since the minor strain variability which was encountered did not correlate with spore appendage status, fragmentation of this species on the basis of spore appendage status is not advocated.  相似文献

The aim of this study is to assess the mycorrhizal status of Eucalyptus plantations in south China and to determine the need for inoculation. In four provinces in south China, 155 plantations were sampled for sporocarps of ectomycorrhizal (ECM) fungi, spores of arbuscular mycorrhizal (AM) fungi, and mycorrhizas over 2 years. This study revealed a low above-ground diversity of ECM fungi consisting of 15 taxa fruiting beneath Eucalyptus plantations. The most common ECM genera were Scleroderma and Pisolithus, but they were infrequent. A total of 21 AM fungi, mostly Glomus species, were recognized from spores collected from eucalypt plantations. Four Glomus species were frequently present in soils, but spore density and relative abundance of AM fungi were generally low. Eucalypt roots from all plantation sites were poorly colonized by either ECM fungi or AM fungi. A bioassay with E. urophylla as a bait host, using soils collected from 11 eucalypt plantations, confirmed low levels of inoculum of both ECM and AM fungi in field soil. This is the first integrated study on the mycorrhizal status of eucalypt plantations in China. Findings from this research can be used to encourage adoption of mycorrhizal technology by eucalypt nurseries in the region. The potential of using spores of compatible ECM fungi or collections for forest nurseries is discussed.  相似文献

Bacterial spores are resistant to a wide range of chemical and physical insults that are normally lethal for the vegetative form of the bacterium. While the integrity of the protein coat of the spore is crucial for spore survival in vitro, far less is known about how the coat provides protection in vivo against predation by ecologically relevant hosts. In particular, assays had characterized the in vitro resistance of spores to peptidoglycan-hydrolyzing enzymes like lysozyme that are also important effectors of innate immunity in a wide variety of hosts. Here, we use the bacteriovorous nematode Caenorhabditis elegans, a likely predator of Bacillus spores in the wild, to characterize the role of the spore coat in an ecologically relevant spore-host interaction. We found that ingested wild-type Bacillus subtilis spores were resistant to worm digestion, whereas vegetative forms of the bacterium were efficiently digested by the nematode. Using B. subtilis strains carrying mutations in spore coat genes, we observed a correlation between the degree of alteration of the spore coat assembly and the susceptibility to the worm degradation. Surprisingly, we found that the spores that were resistant to lysozyme in vitro can be sensitive to C. elegans digestion depending on the extent of the spore coat structure modifications.  相似文献

Ammonia fungi invade forest floors immediately after a enrichment disturbance by a large input of ammonium-nitrogen. Latent form(s) of the ammonia fungi are spores and/or mycelium fragments. Ammonia fungi are characterized by their rapid germination stimulated by the presence of ammonium-nitrogen under neutral to weakly alkaline conditions. Each ammonia fungus establishes its territory during suppressed combative abilities of other microbes following ammonium-nitrogen disturbance. Earlyphase ammonia fungi (EP fungi) quickly sporulate before nonammonia fungi colonize with the declining of ammonium-nitrogen concentration associated with descending pH. Ectomycorrhizal species of late-phase ammonia fungi (LP fungi) escape from the L-F horizon as a result of interactions between EP fungi and form mycorrhizae in the H-A horizon when other fungal activities are suppressed by the ammonium-nitrogen disturbance. Ectomycorrhizal ammonia fungi initially use ammonium-nitrogen when the pH rises because of the ammonium-nitrogen disturbance and then gradually utilize both ammonium- and nitratenitrogen when the effects of the ammonium-nitrogen disturbance weaken. Early-stage EP fungi are ruderal stress-tolerant strategists whereas late-stage EP fungi are combative ruderal strategists. LP fungi are combative strategists from the standpoint of the interactions between other ammonia fungi. This classification is based on differences in their respective propagation strategies.  相似文献

A method to identify arbuscular endomycorrhizal fungi based on the amplification of portions of the nuclear gene coding for the small subunit rRNA is presented. By coupling the sensitivity of the polymerase chain reaction and the specificity afforded by taxon-specific primers, a variety of samples can be analyzed, including small amounts of colonized roots. Family-specific primers as well as generic primers are described and can be used to amplify small subunit rRNA fragments from endomycorrhizal fungi by polymerase chain reaction. The amplified products are then subjected to single-strand conformation polymorphism analysis to detect sequence differences. Among the advantages of this approach is the possibility of directly identifying the fungi inside field-collected roots, without having to rely on the fortuitous presence of spores. This technique should have obvious applications in the study of arbuscular endomycorrhizal fungi populations and allow closer examination of their host specificity.  相似文献
