Increased levels of soluble activity of all three enzymes involved in polyadenylic acid metabolism were measured in PHA-stimulated versus normal lymphocytes. Poly(A)-polymerase and poly(A)-exonuclease values increased significantly (from 25.7 ± 4.2 (S.E.M.) to 53.5 ± 10.6 (S.E.M.), and from 334.6 ± 33.2 (S.E.M.) to 653.2 ± 53.4 (S.E. M.) respectively), while a moderate increase was observed in poly(A)-endonuclease (from 299.2 ± 33.8 (S.E.M.) to 403.0 ± 77.1 (S.E.M.). The above differences persisted after two fractionations of the crude cell extracts by ion exchange chromatography and molecular sieving, and could not be attributed to the competitive action of all three enzymes in the untreated extracts. In a double-blind cross-over trial the effects of nitrazepam (5 and 10 mg.), amylobarbitone sodium (100 and 200 mg.), and placebo were compared in normal healthy young people. Though they reported a good night''s sleep and adjudged themselves to be alert after all four drug treatments, behavioural tests showed their performance to be significantly impaired 13 hours after treatment with nitrazepam or amylobarbitone, and E.E.G. records showed more drowsiness and light sleep 18 hours after treatment with nitrazepam than with amylobarbitone or placebo. E.E.G. fast activity was more plentiful after drugs in either dosage than with placebo.  相似文献

A study of renal biopsy specimens obtained in Senegal from 24 children and six adults with nephrotic syndrome showed two unusual varieties of nephropathy--namely, an extramembranous glomerulonephritis associated with hypocomplementaemia (four cases), a combination previously described only in systemic lupus erythematosus, and a "tropical nephropathy" (16 cases). The latter, though lacking the diffuse glomerular deposits of immunoglobulin described in quartan malarial nephropathy (Q.M.N.), showed a curious progressive and segmental glomerulosclerosis, characterized by a "flaking" or fibrillary splitting of the glomerular capillary wall, seen in Q.M.N. Serological evidence of malaria was lacking in a third of the childhood cases.  相似文献

Spike activity has been recorded, in two young baboons, at the part of the optic tract (O.T.) localized between the chiasma and the lateral geniculate (L.G.) during REM and also slow wave sleep, preceding or not a REM period. These spikes, mono or biphasic, were of 60-200 ms duration and of 30-140 muV amplitude. They occurred generally in bursts (up to 50 spikes); 4 to 10 spikes bursts were the more frequent. These O.T. spikes were assimilated to the L.G. spikes.  相似文献

Morphine-3-glucuronide--a potent antagonist of morphine analgesia   总被引:11,自引:0,他引:11  
In this study, morphine-3-glucuronide (M3G), the major plasma and urinary metabolite of morphine, was shown to be a potent antagonist of morphine analgesia when administered to rats by the intra-cerebroventricular (i.c.v.) route. The antagonism of morphine analgesia was observed irrespective of whether i.c.v. M3G (2.5 or 3.0 micrograms) was administered 15 mins prior to or 15 mins after i.c.v. morphine (20 micrograms). When M3G (10mg) was administered intraperitoneally (i.p.) to rats 30-40 mins prior to morphine (1.5mg i.p.), the analgesic response was significantly reduced compared to administration of morphine (1.5mg i.p.) alone. It was further demonstrated that i.c.v. M3G (2.0 micrograms) antagonized the analgesic effects of subsequently administered i.c.v. morphine-6-glucuronide (0.25 micrograms).  相似文献

Extracorporeal circulation with membrane oxygenator (E.C.M.O.) was performed in 6 dogs and 5 baboons during 4 to 57 hours. Examination of pulmonary samples showed progressive lesions in the two species: at the second hour of E.C., alteration of capillary endothelium, at the sixth, extensive lesions of all the lung structures, worsening later on. We discussed whether these lesions are related to the E.C.M.O., their reversibility and the questionable benefit of E.C.M.O. for management of respiratory distress syndrome.  相似文献

A leaf spot disease of cherry laurel is described. The infected areas eventually become occluded and fall out, leaving irregular holes in the foliage. The disease is caused by Trochila Laurocerasi (Desm.) Fr., the imperfect stage of which is proved to be Gloeosporium-phacidiellum Grove. Infection takes place on the young leaves, most commonly through wounds, and a moist atmosphere is essential for the development and spread of the disease. Control can be effected by spraying with colloidal sulphur during spring and early summer, and by cutting back the bushes only in the autumn, not at short intervals during the summer as is frequently the practice.

The temporal distribution of the horizontal rapid eye movements and the related monophasic potentials recorded from the ascending MLF following intravenous injection of o.i mg/kg of anticholinesterase has been investigated in precollicular decerebrate animals. In particular the intervals between individual MLF potentials occurring during successive REM episodes have been evaluated over a total period of 2000 sec on each experiment. 2. There was a bimodal distribution of intervals due to the fact that all the rapid eye movements and the related MLF potentials were grouped in bursts which occurred at quite regular intervals. 3. During the cholinergically induced episodes of REM, there were usually bursts of REM in one direction followed by bursts of REM in the opposite direction. The mean number of individual eye movements within each burst was 4.67 +/- 0.84, S.D., while the average interval between the individual eye movements corresponded to 167 +/- 36 msec, S.D. 4. There was a great regularity in the periodic occurrence of the bursts of REM. In particular the mean interval between the beginning of a burst of REM in one direction (i.e., towards the left side) and that of the next train oriented in the opposite direction (i.e., towards the right side) was 1.97 +/- 0.47 sec, S.D., while the mean interval between the beginning of this last train and that of the successive train oriented in the former direction corresponded to 2.97 +/- 0.48 sec, S.D. Moreover, the duration of the whole period corresponding to the interval between two successive bursts of REM oriented in the same direction (i.e., towards left or towards right) corresponded on average to 4.94 +/- 0.55 sec S.D. and 4.99 +/- 0.52 sec, S.D. respectively. 5. In addition to these "simple bursts" of rapid eye movements oriented in one direction, there were "complex bursts" in which an alternation of the individual eye movements within each burst was observed. In these instances the mean number of spikes was greater (5.35 +/- 1.20, S.D.) and the mean interval shorter 119 +/- 44 msec, S.D.) than those observed in the "simple bursts", About 10-15% of the bursts occurring during the cholinergically induced REM episodes were of the complex type. 6. These findings obtained from an individual experiment were confirmed in all the decerebrate animals treated with the same dose of anticholinesterase; only slight quantitative differences were detected from case to case. 7. Since the bursts of REM induced by the anticholinesterase depend upon the activity of the vestibular nuclei, it is postulated that cholinergic reticular neurons activate structures which show waxing and waning in their activity before acting on the vestibulo-oculomotor system. This system probably contains the inhibitory interneurons which transform the regularly modulated input into a rhythmic vestibular output...  相似文献

Motor imagery, i.e., a mental state during which an individual internally represents an action without any overt motor output, is a potential tool to investigate action representation during development. Here, we took advantage of the inertial anisotropy phenomenon to investigate whether children can generate accurate motor predictions for movements with varying dynamics. Children (9 and 11 years), adolescents (14 years) and young adults (21 years) carried-out actual and mental arm movements in two different directions in the horizontal plane: rightwards (low inertia) and leftwards (high inertia). We recorded and compared actual and mental movement times. We found that actual movement times were greater for leftward than rightward arm movements in all groups. For mental movements, differences between leftward versus rightward movements were observed in the adults and adolescents, but not among the children. Furthermore, significant differences between actual and mental times were found at 9 and 11 years of age in the leftward direction. The ratio R/L (rightward direction/leftward direction), which indicates temporal differences between low inertia and high inertia movements, was inferior to 1 at all ages, except for the mental movements at 9 years of age, indicating than actual and mental movements were shorter for the rightward than leftward direction. Interestingly, while the ratio R/L of actual movements was constant across ages, it gradually decreased with age for mental movements. The ratio A/M (actual movement/mental movement), which indicates temporal differences between actual and mental movements, was near to 1 in the adults'' groups, denoting accurate mental timing. In children and adolescents, an underestimation of mental movement times appeared for the leftward movements only. However, this overestimation gradually decreased with age. Our results showed a refinement in the motor imagery ability during development. Action representation reached maturation at adolescence, during which mental actions were tightly related to their actual production.  相似文献

The study was targeted at revealing Mycoplasma pneumoniae (M.p.) and Mycoplasma hominis (M.h.) antigens in blood samples of children with bronchial asthma (BA),both in a free state and those included in circulating immune complexes (CIC). The mycoplasma antigens of one or both species have been detected in one third out of 62 patients with BA. In this group of patients the frequency of detection of specific antibodies twice exceeded that of mycoplasma antigens. Testing paired blood samples of children with BA (n=26) showed, that at receipt in a hospital and a month after primary examination the mycoplasma antigens were detected in 16 and 12 patients, respectively, the association of M.p. and M.h. antigens being more frequent. Data on distribution of antibodies according to immunoglobulin classes testify that basically two (M, G), or four (M, G, A, E) classes were registered in children, the M class antibodies in high percentage of cases (from 36.6 up to 50.0%)being detected in every term of examination. These data indirectly testify that the antigens can be partly included in the CIC structure. The level of the total CIC content in BA children's blood sera one month after hospitalization twice exceeded the value detected at primary examination. Three months later later this parameter decreased not reaching the control value. The differential analysis of the precipitated CIC within the whole period of examination showed that mycoplasma antigens were present in the CIC structure in 87.4 - 65.0% of cases. The data obtained precondition future studies on the role of mycoplasma and M.p. and M.h. antigens included in the CIC in the pathogenesis of BA.  相似文献

At night, honey bees pass through a physiological state that is similar to mammalian sleep. Like sleep in mammals, sleep-like behaviour in honey bees is an active process. This is expressed most clearly in these insects by spontaneous antennal movements which appear at irregular intervals throughout the night and interrupt episodes of antennal immobility. Here we present a newly developed video technique for the continuous recording of the position and movements of the bee's antennae. The same technique was used to record head inclination and ventilatory movements. Despite the constancy of the ambient temperature, the magnitudes of antennae-related parameters, as well as head inclination and ventilatory cycle duration, displayed dynamic unimodal time-courses which exhibited a high degree of temporal covariance. The similarity between these time-courses and the nightly time-course of the reaction threshold for a sensory stimulus, investigated previously, indicates that, in honey bees, deepest "sleep" and least ventilatory activity occur at the same time (in the 7th hour of the rest phase).Abbreviations DD continuous darkness - EMG electromyogram - LD periodic alternation between light (L) and darkness (D) - MEST Middle European Summer Time (UTC+2 h); - UV ultraviolet This paper is dedicated to Professor Martin Lindauer, who—to our knowledge—was the first to meticulously record the nightly behaviour of honey bees (Lindauer 1952) and who also inspired one of us (W.K.) to investigate antennal motility during nightly rest in these animals.  相似文献

A young male presented with incessant narrow QRS tachycardia and left ventricular dysfunction. 24-Holter monitoring revealed multiple episodes of sustained and nonsustained episodes of tachycardia with prolonged sinus pauses at termination. The analysis of the electrocardiogram, followed by an invasive electrophysiological study, suggested an unusual mechanism for this tachy-brady syndrome.  相似文献

The mammary cancer cell line CAMA-1 synchronized at the G1/S boundary by thymidine block or at the G1/M boundary by nocodazole was used to evaluate 1) the sensitivity of a specific cell cycle phase or phases to 17 beta-estradiol (E2), 2) the effect of E2 on cell cycle kinetics, and 3) the resultant E2 effect on cell proliferation. In synchronized G1/S cells, E2-induced 3H-thymidine uptake, which indicated a newly formed S population, was observed only when E2 was added during, but not after, thymidine synchronization. Synchronized G2/M cells, enriched by Percoll gradient centrifugation to approximately 90% mitotic cells, responded to E2 added immediately following selection; the total E2-treated population traversed the cycle faster and reached S phase approximately 4 hr earlier than cells not exposed to E2. When E2 was added during the last hour of synchronization (ie, at late G2 or G2/M), or for 1 hr during mitotic cell enrichment, a mixed response occurred: a small portion had an accelerated G1 exit, while the majority of cells behaved the same as controls not incubated with E2. When E2 addition was delayed until 2 hr, 7 hr, or 12 hr following cell selection, to allow many early G1 phase cells to miss E2 exposure, the response to E2 was again mixed. When E2 was added during the 16 hr of nocodazole synchronization, when cells were largely at S or possibly at early G2, it inhibited entry into S phase. The E2-induced increase or decrease of S phase cells in the nocodazole experiments also showed corresponding changes in mitotic index and cell number. These results showed that the early G1 phase and possibly the G2/M phase are sensitive to E2 stimulation, late G1, G1/S, or G2 are refractory; the E2 stimualtion of cell proliferation is due primarily to an increased proportion of G1 cells that traverse the cell cycle and a shortened G1 period, E2 does not facilitate faster cell division; and estrogen-induced cell proliferation or G1/S transition occurs only when very early G1 phase cells are exposed to estrogen. These results are consistent with the constant transition probability hypothesis, that is, E2 alters the probability of cells entering into DNA synthesis without significantly affecting the duration of other cell cycle phases. Results from this study provide new information for further studies aimed at elucidating E2-modulated G1 events related to tumor growth.  相似文献

The cleavage specificities of the RNase P holoenzymes from Escherichia coli and the yeast Schizosaccharomyces pombe and of the catalytic M1 RNA from E. coli were analyzed in 5'-processing experiments using a yeast serine pre-tRNA with mutations in both flanking sequences. The template DNAs were obtained by enzymatic reactions in vitro and transcribed with phage SP6 or T7 RNA polymerase. The various mutations did not alter the cleavage specificity of the yeast RNase P holoenzyme; cleavage always occurred predominantly at position G + 1, generating the typical seven base-pair acceptor stem. In contrast, the specificity of the prokaryotic RNase P activities, i.e. the catalytic M1 RNA and the RNase P holoenzyme from E. coli, was influenced by some of the mutated pre-tRNA substrates, which resulted in an unusual cleavage pattern, generating extended acceptor stems. The bases G - 1 and C + 73, forming the eighth base pair in these extended acceptor stems, were an important motif in promoting the unusual cleavage pattern. It was found only in some natural pre-tRNAs, including tRNA(SeCys) from E. coli, and tRNAs(His) from bacteria and chloroplasts. Also, the corresponding mature tRNAs in vivo contain an eight base pair acceptor stem. The presence of the CCA sequence at the 3' end of the tRNA moiety is known to enhance the cleavage efficiency with the catalytic M1 RNA. Surprisingly, the presence or absence of this sequence in two of our substrate mutants drastically altered the cleavage specificity of M1 RNA and of the E. coli holoenzyme, respectively. Possible reasons for the different cleavage specificities of the enzymes, the influence of sequence alterations and the importance of stacking forces in the acceptor stems are discussed.  相似文献

In vivo patterns of circulating testosterone (T) were investigated in stock fed controls and parenterally nourished (TPN) rats. Rats were sampled at 2 minute intervals for 30 minutes via a jugular cannula. Both groups exhibited a rapid oscillatory T pattern. In the control group, T concentrations at any specific time interval exhibited large differences with coefficients of variation (17-88%). In TPN rats this variation was 34-79%. Moreover, the mean T concentration of all samples obtained during the 30 minute period for each individual animal ranged from 1.3 +/- 0.1 (S.E.M.) to 3.5 +/- 0.3 (S.E.M.) ng/ml in controls and 1.3 +/- 0.1 (S.E.M.) to 2.0 +/- 0.2 (S.E.M.)ng/ml in controls and 1.3 +/- 0.1 (S.E.M.) to 2.0 +/- 0.2 (S.E.M.) ng/ml for TNP rats respectively. The mean coefficient of variation in control animals, however, was twice that of TPN rats, indicating that variation in basal T may be minimized during intravenous feeding. The occurrence of rapid oscillations in T of both stock and parenterally fed animals shows that nutritional regimen does not affect this phenomenon.  相似文献

The formation of spontaneous rosettes by peripheral blood or spleen mononuclear cells when incubated with sheep red blood cells has proved a useful way of assessing the potency of immunosuppressive drugs and antilymphocyte sera in vitro. A test employing the inhibition by antilymphocyte globulin (A.L.G.) of spontaneous rosette formation around peripheral blood mononuclear cells is described. This has been used to assess the degree of immunosuppression in patients with renal allografts and uraemic patients on regular haemodialysis.Twenty-three patients with renal allografts had 21 clinically diagnosed episodes of rejection. In none of these rejection episodes was the minimal inhibitory concentration (M.I.C.) of A.L.G. (that necessary to reduce the spontaneous rosette formation of peripheral cells by 75%) less than 1/50,000. Nineteen patients had no rejection episodes during 57 patient/months of continuous observation while the M.I.C. was at a greater dilution than 1/50,000. The test has therefore been of great value in suggesting when an individual is capable of rejecting his graft, and allows the dose of immunosuppressive drugs to be adjusted to a minimum in a controlled fashion. It has been of use in diagnosing rejection in the anuric patient, when the distinction between rejection, urinary tract obstruction, and infection is particularly difficult.Fifteen patients maintained on regular haemodialysis for more than a year had, as judged by this technique, less reactive lymphocytes than normal healthy controls. The degree of immunosuppression was not as great as in the patients on full immunosuppressive regimens.  相似文献

Differences between males and females in the effects of regular physical training on the heart were investigated by echocardiography in 579 male (nonathletes: 122, athletes: 457) and in 336 (nonathletes: 84, athletes: 252) female subjects of variable age. The age groups were: children (<15 yr.), adolescent-young (15-18 yr.), young adult (19-30 yr.), adult (31-45 yr.) and people elder than 45 yr older (45<). Hypertrophy characterised by relative left ventricular muscle mass (rel.LVMM) of the athletic heart was manifest also in the females, but in comparison with the males female values were much lower both in the unconditioned and in the conditioned groups. In people belonging to the oldest groups no differences were seen either between the males and females or between athletes and nonathletes. In comparing nonathletic subjects, females showed smaller left ventricular contractility as reflected by a higher LVET/QT ratio and an increased ability of diastolic relaxation as indicated by a higher E/A ratio. Physical training induced a more marked development when the initial level was lower, namely, in the contractility of the females, and in the compliance of the males. No intersex difference was seen in the modified regulation of the athletic heart, characterised by a lower resting heart rate and slower circumferential shortening velocity (VCF).  相似文献
