Sodium-induced calcium deficiency in salt-stressed corn
Abstract The effect of the Na+/Ca2+ ratio in the root media on salt-stressed corn (Zea mays L. cvs DeKalb XL-75 and Pioneer 3906) was determined in greenhouse experiments. Plants grown in a complete nutrient solution salinized with 86.5 mol m?3 NaCl exhibited severe Ca2+ deficiency symptoms at the four-leaf stage. The symptoms disappeared when part of the NaCl was replaced with 10 mol m?3 CaCl2 (Na+/Ca2+ molar ratio = 5.7). Salt stress at an iso-osmotic potential of ?0.4 MPa substantially decreased shoot growth at all solution Na+/Ca2+ ratios from 34.6 to 0.26. However, the dry weights of blades at 26 d of age were much less when plants were salinized with NaCl alone, particularly that of DeKalb XL-75 which was more susceptible to Na-induced Ca2+ deficiency than was Pioneer 3906. The growth of sheaths was similarity reduced by sail stress at all Na+/Ca2+ ratios. The symptoms of Ca2+ deficiency were correlated with low Ca2+ concentrations in the leaf tissue. Ca2+ concentrations in the developing blades of NaCl-stressed plants were much lower than in control plants. As the Na+/Ca2+ ratio in the solution was decreased, Ca2+ levels increased in both the blades and sheaths while Na+ concentrations greatly decreased. DeKalb XL-75 was much less effective than Pioneer 3906 in restricting the uptake of Na+. The results clearly indicate that NaCl stress may cause lesions and unique plant responses that are not manifested on agronomic plants grown on saline soils.

Sodium exclusion mechanisms at the root surface of two maize cultivars
Sodium exclusion at the root surface of intact plants was investigated in two maize cultivars (Zea mays L. cv. Pioneer 3906 and cv. DeKalb XL75). The strong sodium exclusion by Pioneer 3906 relative to DeKalb XL75 was not caused by larger efflux rates. In contrast, this cultivar showed reduced influx rates resulting in about half the net uptake in comparison with DeKalb XL75. It is concluded that the cultivar difference in sodium exclusion at the root surface is based on a relatively low passive sodium permeability of the epidermal and cortex plasmalemma of Pioneer 3906. For both cultivars, the flux ratio equation indicated an active component of sodium efflux.

A greenhouse study was designed to test the effects of sodium sulphate (Na2SO4) on red-osier dogwood (Cornus stolonifera Michx) seedlings in the presence and absence of additional calcium (Ca2+). Changes in growth parameters, ion and carbohydrate accumulation and cell wall composition were examined. Calcium alleviated the effect of Na2SO4 on shoot height; however, it did not affect the reduction in shoot and root dry weights. An increased level of sodium (Na+) in roots of plant exposed to Na2SO4 was recorded in the presence of supplemental Ca2+ whereas there was no change in potassium (K+) and Ca2+ levels. In shoots of seedlings treated with Na2SO4, the addition of Ca2+ did not affect Na+, K+ and Ca2+ levels. The amount of soluble carbohydrates was increased in leaves of seedlings treated with Na2SO4 both in the absence and presence of supplemental Ca2+. The decrease in cell wall material in response to salt stress was alleviated by Ca2+ in stem tissues although Ca2+ did not alter the changes in hemicellulose and cellulose. Sugar composition of pectins and hemicellulose were modified in stems and leaves by Na2SO4 and/or Ca2+. The results of this study showed that calcium was able to alleviate the effects of salt stress on shoot height and cell wall content of red-osier dogwood stems. Furthermore, changes occurred in cell wall composition of red osier seedlings treated with Na2SO4.

The inherent differences for salt tolerance in two maize cultivars (Agatti-2002 and Sahiwal-2002) were evaluated in pot experiments. Plants were grown in half-strength of Hoagland nutrient solution added with 0, 80, 100, 120, 140 and 160 mM of NaCl. Salt stress markedly reduced the shoot and root lengths and fresh and dry masses. Reduction in growth attributes was more pronounced in cv. Agatti-2002 than cv. Sahiwal-2002. Both maize cultivars exhibited significant perturbations in important biochemical attributes being employed for screening the crops for salt tolerance. Cultivar Sahiwal-2002 was found salt tolerant as compared to cv. Agatti-2002 because it exhibited lower levels of H2O2, malondialdehyde (MDA) and higher activities of antioxidant enzymes. In addition, cultivar Sahiwal-2002 exhibited less salt-induced degradation of photosynthetic pigments, lower levels of toxic Na+ and Cl and higher endogenous levels of K+ and K+/Na+ ratio. The results indicate that salt stress induced a marked increase in MDA, H2O2, relative membrane permeability, total soluble proteins and activities of antioxidant enzymes (superoxide dismutase, peroxidase, catalase andascorbate peroxidase). Moreover, increase in endogenous levels of Na+ and Cl and decrease in K+ and K+/Na+ ratio and photosynthetic pigments were recorded in plants grown under salinity regimes.

Salinity causes changes in cytosolic Ca2+, [Ca2+]cyt, Na+, [Na+]cyt and pH, pHcyt, which induce specific reactions and signals. Reactions causing a rebalancing of the physiological homeostasis of the cytosol could result in plant resistance and growth. Two wheat cultivars, Triticum aestivum, Seds1 and Vinjett, were grown in nutrient solution for 7 days under moderate salinity (0 and 50 mM NaCl) with and without extra addition of 5 mM CaSO4 to investigate the seedling‐ion homeostasis under salinity. In the leaf protoplasts [Ca2+]cyt, [Na+]cyt and pHcyt were detected using acetoxymethyl esters of the ion‐specific dyes, Fura 2, SBFI and BCECF, respectively, and fluorescence microscopy. In addition, both cultivars were grown for 3 weeks at 0, 50 and 125 mM NaCl with, or without, extra addition of 5 mM CaSO4 to detect overall Na+ and Ca2+ concentrations in leaves and salinity effects on dry weights. In both cultivars, salinity decreased [Ca2+]cyt, while at extra Ca2+ supplied, [Ca2+]cyt increased. The [Ca2+]cyt increase was accompanied by increase in the overall Ca2+ concentrations in leaves and decrease in the overall Na+ concentration. Moreover, irrespective of Ca2+ treatment under salinity, the cultivars reacted in different ways; [Na+]cyt significantly increased only in cv. Vinjett, while pHcyt increased only in cv. Seds1. Even at rather high total Na+ concentrations, the cytosolic concentrations were kept low in both cultivars. It is discussed whether the increase of [Ca2+]cyt and pHcyt can contribute to salt tolerance and if the cytosolic changes are due to changes in overall Ca2+ and Na+ concentrations.

Using radio-tracers, we measured Na+ and K+ accumulation in roots and transport to shoots in Zea mays (cv Pioneer 3906) as a function of NaCl concentration and O2 partial pressure in the nutrient solution. Under fully aerobic conditions, roots partially excluded Na+ from the shoots over a wide range of NaCl concentration (0.2-200 millimolar). With root anoxia, the exclusion mechanism broke down so that much greater amounts of Na+ reached the shoots, with simultaneous inhibition of K+ transport. The ratio Na+/K+ entering the shoot consequently increased 90 to 200 times. Increases in Na+ transport were first detected when the O2 partial pressure was reduced from ambient (21% v/v) to 15%, whereas K+ transport was not inhibited until O2 concentrations were <5%. Since soil O2 deficiency can often accompany high salinity in irrigation agriculture, failure of the Na+ exclusion mechanism may be a contributory factor in salinity damage of salt-sensitive glycophytes.

To assess whether foliar application of K+S as potassium sulfate (K2SO4) could alleviate the adverse effects of salt on sunflower (Helianthus annuus L. cv. SF-187) plants, a greenhouse experiment was conducted. There were two NaCl levels (0 and 150 mM) applied to the growth medium and six levels of K+S as K2SO4 (NS (no spray), WS (spray of water+0.1% Tween 20 solution), 0.5% K+0.21% S, 1.0% K+0.41% S, 1.5% K+0.62% S, and 2.0% K+0.82% S in 0.1% Tween-20 solution) applied two times foliarly to non-stressed and salt-stressed sunflower plants. Salt stress markedly repressed the growth, yield, photosynthetic pigments, water relations and photosynthetic attributes, quantum yield (Fv/Fm), leaf and root K+, Mg2+, P, Ca2+, N as well as K+/Na+ ratios, while it enhanced the cell membrane permeability, and leaf and root Na+ and Cl concentrations. Foliar application of potassium sulfate significantly improved growth, achene yield, photosynthetic and transpiration rates, stomatal conductance, water use efficiency, leaf turgor and enhanced shoot and leaf K+ of the salt-stressed sunflower plants, but it did not improve leaf and root Na+, Cl, Mg2+, P, Ca2+, N as well as K+/Na+ ratios. The most effective dose of K+S for improving growth and achene yield was found to be 1.5% K+0.62% S and 1% K+0.41% S, respectively. Improvement in growth of sunflower plants due to exogenously applied K2SO4 was found to be linked to enhanced photosynthetic capacity, water use efficiency, leaf turgor and relative water content.

Salinity tolerance of sugar beet (Beta vulgaris L.) cultivars in terms of growth, proline and soluble sugars concentrations, and Na+/K+ and Na+/Ca2+ ratios were analyzed in this study. Three-week-old seedlings of three sugar beet cultivars, ‘Gantang7’, ‘SD13829’, and ‘ST21916’, differing in salinity tolerance, were treated with 0, 50, 100, and 200 mM NaCl. Plant shoots and roots were harvested at 7 days after treatment and subjected to analysis. Low concentration of NaCl (50 mM) enhanced fresh and dry weights of shoot and root in ‘Gantang7’, whereas high one (200 mM) reduced growth in all cultivars and the less reduction was observed in ‘ST21916’. Shoot proline was strongly induced by salinity stress in both ‘Gantang7’ and ‘SD13829’, while it remained unchanged in ‘ST21916’. The addition of 50 mM NaCl significantly increased shoot soluble sugars concentrations in ‘Gantang7’ while it had no significant effects in the other two cultivars. ‘Gantang7’ also showed a higher level of root soluble sugars concentration as compared to the other two cultivars. At 50 mM NaCl, the lower shoot Na+ concentration, and the higher shoot K+ and root Ca2+ concentration in ‘Gantang7’ resulted in the lower shoot Na+/K+ and root Na+/Ca2+ ratio. However, ‘SD13829’ maintained a lower Na+/K+ ratio in both shoot and root when subjected to 200 mM NaCl treatment. According to comprehensive evaluation on salinity tolerance, it is clear that ‘Gantang7’ is more tolerant to salinity than the other two cultivars. Therefore, it is suggested that ‘Gantang7’ should be more suitable for cultivating in the arid and semi-arid irrigated regions.

Thermotolerance of pearl millet (Pennisetum glaucum cv. ICMV-94133) and maize (Zea mays cv. Golden) was assessed at germination and vegetative stage. Final percentage of germinated seeds and rate of germination (number of days to 50 % germination) decreased due to high temperature (45 °C) similarly in the both species. In contrast, at the vegetative stage, high temperature (38/27 °C) caused a significant reduction in shoot dry mass of maize, whereas this attribute remained almost unchanged in pearl millet. Relative growth rate and net assimilation rate (NAR) increased significantly in pearl millet due to high temperature, but in contrast, in maize NAR was slightly reduced. Concentrations of N, P, and K in the shoots of both species increased at high temperature, but N accumulation was more pronounced in pearl millet than in maize. High temperature caused a marked increase in both shoot and root Ca2+ concentration in maize, but it did not affect that of pearl millet. S concentration in the shoots of maize decreased significantly due to high temperature, whereas that in pearl millet remained unaffected. Shoot Na+ concentration of both species was not significantly affected by high temperature. High temperature caused a significant increase in uptake of N, P, and K+ in pearl millet, but the uptake of Ca2+, Mg2+, Na+ and S remained unaffected in this species. In contrast, in maize, a significant increase in uptake of K+ and Ca2+, and a decrease in uptake of N, S, Mg2+, and Na+ were found at high temperature. Overall, maize showed lower tolerance to high temperature compared with pearl millet.

Abstract

The mechanism of growth amelioration in salt-stressed maize (Zea mays L. cv., DK 647 F1) by exogenously applied mannitol (M) and thiourea (T) was investigated. Maize seedlings were planted in pots containing perlite and subjected to 0 or 100 mM NaCl in full strength Hoagland's nutrient solution. Two levels of M (15 and 30 mM) or T (3.5 and 7.0 mM) were sprayed to the leaves of maize seedlings 10 days after germination. Salinity stress caused considerable reduction in plant dry biomass, chlorophyll content, and relative water content in the maize plants. However, it increased the activities of catalase (CAT; EC 1.11.1.6), superoxide dismutase (SOD; EC 1.15.1.1), and polyphenol oxidase (PPO; EC 1.10.3.1), and levels of hydrogen peroxide (H2O2) and electrolyte leakage, but it did not change peroxidase (POD; EC 1.11.1.7) activity. Foliar application of M or T was found to be effective in checking salt-induced shoot growth inhibition. Exogenously applied M or T reduced the activities of CAT, SOD, POD, and PPO in the salt-treated maize plants compared to those in the plants not fed with these organic compounds. Salinity increased Na+ contents but decreased those of K+, Ca2 +, and P in the leaves and roots of the maize plants. Foliar-applied M or T increased the contents of K+, Ca2 +, and P, but decreased that of Na+ in the salt-stressed maize plants with respect to those of the salt-stressed plants not supplied with mannitol or thiourea. Mannitol was found to be more effective than thiourea in improving salinity tolerance of maize plants in terms of growth and physiological attributes measured in the present study.

Plants of two wheat (Triticum aestivum L.) cultivars differing in salt tolerance were grown in sand with nutrient solutions. 35-d-old plants were subjected to 5 levels of salinity created by adding NaCl, CaCl2 and Na2SO4. Growth reduction caused by salinity was accompanied by increased Na+ and Cl- concentrations, Na+/K+ ratio, and decreased concentration of K+. The salt tolerant cv. Kharchia 65 showed better ionic regulation. Salinity up to 15.7 dS m-1 induced increased uptake of Na+ and Cl- but higher levels of salinity were not accompanied by further increase in uptake of these ions. Observed increases in Na+ and Cl- concentrations at higher salinities seemed to be the consequence of reduction in growth. Uptake of K+ was decreased; more in salt sensitive cultivar. This was also accompanied by differences in its distribution.

Basil (Ocimum basilicum L., cultivar Genovese) plants were grown in Hoagland solution with or without 50 mM NaCl or 25 mM Na2SO4. After 15 days of treatment, Na2SO4 slowed growth of plants as indicated by root, stem and leaf dry weight, root length, shoot height and leaf area, and the effects were major of those induced by NaCl. Photosynthetic response was decreased more by chloride salinity than by sulphate. No effects in both treatments on leaf chlorophyll content, maximal efficiency of PSII photochemistry (F v/F m) and electron transport rate (ETR) were recorded. Therefore, an excess of energy following the limitation to CO2 photoassimilation and a down regulation of PSII photochemistry was monitored under NaCl, which displays mechanisms that play a role in avoiding PSII photodamage able to dissipate this excess energy. Ionic composition (Na+, K+, Ca2+, and Mg2+) was affected to the same extent under both types of salinity, thus together with an increase in leaves Cl, and roots SO4 2− in NaCl and Na2SO4-treated plants, respectively, may have resulted in the observed growth retardation (for Na2SO4 treatment) and photosynthesis activity inhibition (for NaCl treatment), suggesting that those effects seem to have been due to the anionic component of the salts.

The response of two tomato lines (Lycopersicon esculentum Mill. Ca2+ use efficient line 113 and Ca2+ use inefficient line 67) to a range of constant low Ca2+ concentrations was investigated in a sand culture system. Four Ca2+ concentrations were established and maintained throughout the experiment: 0.038, 0.75, 1.51 and 3.75 mM CaCl2 on a constant background of 1.1 mM NaCl. Response to Ca2+ was determined by analysis of growth parameters and of shoot Ca2+, Na+, K+ and Cl concentrations. Differences in Ca2+ and K+ use efficiencies were expressed as the calcium utilization efficiency ratio, or CaER, and potassium utilization efficiency ratio, or KER, (mg of dry weight produced·mg−1 of Ca2+ or K+ in plant). Dry weight production of line 113 was significantly higher than line 67, and was associated with a higher CaER and KER. The Ca2+ treatments differentially affected shoot Ca2+, Na+, Cl and K+ concentrations. As expected, shoot Ca2+ and Cl concentrations increased whereas Na+ concentration decreased with Ca2+ treatments. Line 113 had more than twice the amount of Na+ in shoot tissue than line 67. The K+ to Na+ ratio was twice as high in line 67 than in line 113. No evidence for higher soluble Ca2+ contributing to higher Ca2+ utilization was observed. The relationship between Ca2+ use efficiency and growth was not correlated with higher percentages of soluble Ca2+ in leaf tissue or with differences in root morphology. Differences in Ca2+ use efficiency alone could not explain the higher growth rate in line 113. This study demonstrated that the physiological factors involved in the genetic control of Ca2+ use efficiency should be assessed under a range of constant low Ca2+ concentrations in order to observe the physiological changes taking place. Thus, the use of Ca2+ deficient conditions are to be avoided as it may interfere with the expression of the physiological factors involved in Ca2+ use efficiency.

Cyclocarya paliurus seedlings were subjected to 85?mM NaCl and 0, 6, 12 or 18?mM Ca(NO3)2 treatments to study changes in plant growth, photosynthetic parameters and distribution and/or accumulation of organic and inorganic solutes. Na+ toxicity symptoms were observed in plants non-treated with Ca(NO3)2, while 12?mM Ca(NO3)2 supplementation produced a significant promotion of shoot growth; meanwhile chlorophyll content, photosynthetic rate and optimum quantum yield of photosystem II (PSII), represented by the Fv/Fm ratio and pigments content as well as proline and soluble sugars, significantly increased. Ca(NO3)2 supply increased K+ and Ca2+ concentration, whereas the Na+ transport to the shoot was inhibited. There was a strong increase in the K+/Na+ ratio in shoot of Ca(NO3)2-treated plants. X-Ray microanalysis of roots showed that K+, Ca2+ and Na+ accumulated mainly in the epidermal cells and cortical cells of roots with 12?mM Ca(NO3)2 supply, and low accumulation was observed in stelar parenchyma, indicating exogenous Ca2+ possibly induced or strengthened effects of Casparian bands on ion transport. These results suggest that Ca(NO3)2 supplement increased inorganic and organic solutes accumulation in shoot and leaf, and restricted Na+ transport to the shoot by reinforcing barrier effects for attenuating salt injuries in plants, which could be a strategy of Ca2+ alleviating Na+ toxicity in C. paliurus seedlings subjected to salt stress.

The effects of thiamin (Thi) applied as seed soaking or foliar spray on some key physiological parameters were investigated in two differentially salt-responsive maize (Zea mays L.) cultivars, DK 5783 and Apex 836 F1, exposed to saline stress in two different experiments. An initial experiment (germination experiment) was designed to identify appropriate doses of Thi which could lessen the deleterious effects of salt on plants and screen all available maize cultivars for their differential tolerance to salt stress (100 mM NaCl). The seeds of nine maize cultivars were soaked for 24 h in solutions containing six levels of Thi (25, 50, 75, 100, 125 and 150 mg l?1). Based on the results obtained from the germination experiment, maize cultivar DK 5783 was found to be the most salt tolerant and Apex 836 as the most sensitive cultivar. Also, of six Thi levels used, two levels (100 and 125 mg l?1) were chosen for subsequent studies. In the second experiment (glasshouse experiment), two maize cultivars, DK 5783 (salt tolerant) and Apex 836 (salt sensitive) were subjected to saline regime (100 mM NaCl) and two levels of Thi (100 and 125 mg l?1) applied as foliar spray. Salt stress markedly suppressed shoot and root dry mass, total chlorophylls (“a” + “b”), leaf water potential and maximum fluorescence yield (Fv/Fm) in the plants of both maize cultivars, but it increased proline accumulation, leaf osmotic pressure, malondialdehyde (MDA) and hydrogen peroxide (H2O2) concentrations, electrolyte leakage (EL) as well as activities of some key antioxidant enzymes, superoxide dismutase (SOD; EC. 1.15.1.1), peroxidase (POD; EC. 1.11.1.7) and catalase (CAT; EC. 1.11.1.6). Salt-induced reduction in plant growth parameters was higher in the salt-sensitive cultivar, Apex 836, which was found to be associated with relatively increased EL, and MDA and H2O2 levels, and decreased activities of the key antioxidant enzymes. Application of Thi as seed soaking or foliar spray partly mitigated the deleterious effects of salinity on plants of both maize cultivars. The most promising effect of Thi on alleviation of adverse effects of salt stress on maize plants was found when it was applied as foliar spray at 100 mg l?1. Thiamin application considerably reduced tissue Na+ concentration, but improved those of N, P, Ca2+ and K+ in the salt-stressed maize plants. Exogenously applied thiamin-induced growth improvement in maize plants was found to be associated with reduced membrane permeability, MDA and H2O2 levels, and altered activities of some key antioxidant enzymes such as CAT, SOD and POD as well as increased photosynthetic pigment concentration under saline regime.

The influence of pre-sowing seed treatment with polyamines (2.5 mM putrescine, 5.0 mM spermidine and 2.5 mM spermine) on growth, photosynthetic capacity, and ion accumulation in two spring wheat (Triticum aestivum L.) cultivars MH-97 (intolerant) and Inqlab-91 (tolerant) was examined. The primed seeds of each treatment and non-primed seeds were sown in a field containing 15 dS m−1 NaCl. Although all three polyamines were effective in improving shoot growth and grain yield in both cultivars under saline conditions, the effect of spermine was very pronounced particularly in improving grain yield. Different priming agents did not affect the net CO2 assimilation rate and transpiration rate of either cultivar. However, pre-treatment with spermidine increased stomatal conductance (gs) in the tolerant cultivar, whereas with spermine stomatal conductance decreased in the intolerant cultivar under salt stress. Priming agents had different effects on the accumulation of different ions in wheat plant tissues. When spermidine and distilled water were used as priming agents, they were effective in reducing shoot [Na+] in the tolerant and intolerant cultivars, respectively under saline conditions. Although all priming agents caused an increase in shoot [K+], distilled water was more effective in improving shoot [K+] in both cultivars under salt stress. Pre-treatment with spermidine was very effective in reducing shoot [Cl] under saline conditions particularly in the tolerant cultivar. However, the pattern of accumulation of different ions in roots due to different seed priming treatments was not consistent in either cultivar except that root Na+ decreased due to priming with spermine and spermidine in the intolerant and tolerant cultivars under saline conditions. In conclusion, although all three priming agents, spermine, spermidine and putrescine, were effective in alleviating the adverse effect of salt stress on wheat plants, their effects on altering the concentration of different ions and growth were different in the two cultivars differing in salt tolerance.

Response of sugarcane to different types of salt stress
Summary Due to climatic conditions and prevailing water regime the yield and sucrose recovery in sugarcane are high in South Western India. However, excessive irrigation, poor drainage and luxuriant use of fertilizers have resulted in conversion of large fertile areas into saline lands. The salinity is due to the excess of Na+, Ca++, Mg++, SO4 and Cl ions. Individual salts of NaCl, Na2SO4, MgCl2 and MgSO4 were employed in culture experiments to study salt stress effect on sugarcane variety Co 740. It was observed that sulphate salinity was more toxic to sugarcane than the chloride one. Sulphate salts caused more inhibition of growth, chlorophyll synthesis, PEPCase activity, decreased the uptake of K+ and Ca++ ions but stimulated nitrate reductase. The stress did not result in proline accumulation in the sugarcane cultivar Co 740. The degree of toxicity of different ions in decreasing order in sugarcane cultivar Co 740 is SO4 >Na+>Cl>Mg++.

In pot experiments with Solanum tuberosum L. (cv Saturna) the application of KCl as compared to K2SO4 delayed tuber development. The solute composition of leaves of the KCl treated plants was significantly lower in K+ and NO3 -, but higher in Mg2+, Ca2+ and Cl-. Since the solute potential in the KCl treated plants was more negative and associated with a higher water content, a higher turgor pressure can be assumed. This could explain the enhanced shoot growth observed with KCl. Application of K2SO4, on the other hand, accelerated the development of tubers. This might result from a less competitive shoot sink in K2SO4 treated plants and a stimulated phloem loading and translocation of assimilates by higher concentrations of leaf-K.
