Eicosapentaenoic acid (EPA) is a member of the family of n-3 polyunsaturated fatty acids (PUFAs) that are clinically used to treat hypertriglyceridemia. The triglyceride (TG) lowering effect is likely due to an alteration in lipid metabolism in the liver, but details have not been fully elucidated. To assess the effects of EPA on hepatic TG metabolism, mice were fed a high-fat and high-sucrose diet (HFHSD) for 2 weeks and were given highly purified EPA ethyl ester (EPA-E) daily by gavage. The HFHSD diet increased the hepatic TG content and the composition of monounsaturated fatty acids (MUFAs). EPA significantly suppressed the hepatic TG content that was increased by the HFHSD diet. EPA also altered the composition of fatty acids by lowering the MUFAs C16:1 and C18:1 and increasing n-3 PUFAs, including EPA and docosahexaenoic acid (DHA). Linear regression analysis revealed that hepatic TG content was significantly correlated with the ratios of C16:1/C16:0, C18:1/C18:0, and MUFA/n-3 PUFA, but was not correlated with the n-6/n-3 PUFA ratio. EPA also decreased the hepatic mRNA expression and nuclear protein level of sterol regulatory element binding protein-1c (SREBP-1c). This was reflected in the levels of lipogenic genes, such as acetyl-CoA carboxylase α (ACCα), fatty acid synthase, stearoyl-CoA desaturase 1 (SCD1), and glycerol-3-phosphate acyltransferase (GPAT), which are regulated by SREBP-1c. In conclusion, oral administration of EPA-E ameliorates hepatic fat accumulation by suppressing TG synthesis enzymes regulated by SREBP-1 and decreases hepatic MUFAs accumulation by SCD1.  相似文献

During net nucleoside triphosphate synthesis by chloroplast ATP synthase the extent of water oxygen incorporation into each nucleoside triphosphate released increases with decrease in ADP, GDP or IDP concentration. Likewise, during net ATP hydrolysis by the Mg2+-activated chloroplast ATPase, the extent of water oxygen incorporation into each Pi released increases as the ATP, GTP, or ITP concentration is decreased. However, the concentration ranges in which substrate modulation occurs differs with each nucleotide. Modulation of oxygen exchange during synthesis and hydrolysis of adenine nucleotides, as measured by variation in the extent of water oxygen incorporation into products, occurs below 250 microM. In contrast, guanosine and inosine nucleotides alter the extent of exchange at higher and much wider concentration ranges. Activation of the chloroplast ATPase by either heat or trypsin results in similar catalytic behavior as monitored by ATP modulation of oxygen exchanges during hydrolysis in the presence of Mg2+. More exchange capacity is evident with octylglucoside-activated enzyme at all ATP concentrations. High levels of tentoxin were also found to alter the catalytic exchange parameters resulting in continued water oxygen exchange into Pi released during hydrolysis at high ATP concentrations. Little or no oxygen exchange accompanies ATP hydrolysis in the presence of Ca2+. The [18O]Pi species formed from highly gamma-18O-labeled ATP at lower ATP concentrations gives a distribution as expected if only one catalytic pathway is operative at a given ATP concentration. This and other results support the concept of catalytic cooperativity between alternating sites as explanation for the modulation of oxygen exchange by nucleotide concentration.  相似文献

An ATP pyrophosphohydrolase in a rat liver plasma-membrane subfraction was studied with respect to specific Ca2+ activation of the beta-phosphate bond hydrolysis. ATP and, in addition, adenosine 5'-[betagamma-imido]triphosphate and adenosine 5'-[betagamma-methlylene]triphosphate were substrates for Ca2+-stimulated enzymic hydrolysis of the beta-phosphate bond. A 15-fold activation was observed by raising the free Ca2+ concentration from 10(-7) to 10(-5) M. Mg2+ had little effect. Solubilization in 1% deoxycholate and partial purification on a sucrose density gradient resulted in a 5-fold increase in specific activity with unaltered Ca2+-stimulation pattern. The possible importance of the enzyme in Ca2+ transport is discussed.  相似文献

1. This paper reports studies on the metabolism of bone from normal chicks and from chicks with vitamin D-deficiency rickets. Both in vitro and in vivo there was an increased incorporation of [(14)C]proline into collagen hydroxyproline by rachitic bone. The proportion of the collagen that was soluble in cold salt solutions was greater with the rachitic bone. These results show that in rickets there is an increased synthesis of bone collagen, but they do not provide any evidence of a defect in the maturation of collagen. 2. Rachitic bone incubated aerobically in vitro consumed more glucose and released more lactate than normal bone. Bone from rachitic chicks treated with vitamin D 48hr. previously had rates of glycolysis that were nearly normal. Though we were unable to show any direct action of vitamin D in vitro, we consider that vitamin D probably has a direct action on bone, possibly related to matrix biosynthesis.  相似文献

The steady-state level of brain ATP was measured after the tissue had been treated with a focused microwave irradiation system. The brain ATP content (1.50 nmol/mg tissue) obtained by microwave fixation is similar to that observed by others using fast-freezing and microwave fixation techniques. The concentrations of ATP in different brain regions show a rather uniform distribution, ranging from 1.918±0.059 (brainstem) to 2.393±0.19 (caudate) nmol/mg tissue; however, insufficient microwave fixation time seems to produce a greater regional variation of ATP. Release of ATP was investigated by placing a cup on the sensory-motor cortex. The rate for basal release of ATP is 1.43±0.14 femtomole/min/mm2. A 30-fold increase in ATP release was obtained by direct stimulation of the cortex with 5 mA pulses of 0.2 msec duration at a rate of 20/sec over a period of 10 min. These results, in conjunction with others describing the potent pharmacological action of the nucleotide, seem to suggest that ATP could be a mediator in a purinergic system in the CNS.  相似文献

The effects of a number of hepatotoxic and other agents on the ATP content of rat liver are described. Changes in the distribution of ATP between the cell sap and the large-particle fraction were determined at intervals after rats had been dosed with various substances. Ethionine produced a rapid decrease in total liver ATP but no alteration in its intracellular distribution. Carbon tetrachloride, sodium salicylate, dimethylnitrosamine, 2,4-dinitrophenol, icterogenin, sodium succinate, sodium malonate and sodium taurocholate did not significantly alter the total ATP content of liver in the periods studied but changes in intracellular distribution were found. Carbon tetrachloride, malonate and taurocholate decreased, and salicylate treatment increased, the proportion of ATP in the cell sap. Treatment with sodium phenobarbitone increased the total liver ATP and the total amount of ATP in the cell sap. The changes in ATP concentration and in the intracellular distribution of ATP are correlated with changes previously reported in bile flow (Delaney & Slater, 1969). No general correlation was found between changes in total ATP and changes in bile flow rate, but there was a relationship between changes in bile flow and in ATP content in the case of ethionine. With the exception of taurocholate and icterogenin, which possibly act on a membrane site, an approximate correlation was found between changes in bile flow and changes in the amount of ATP in the cell sap. The findings are discussed in terms of possible mechanisms for biliary secretion.  相似文献

AMP, adenosine, inosine and hypoxanthine were present in perfusates collected from bovine adrenal glands during periods when catecholamine secretion was evoked by injections of carbamoylcholine. 2. The molar ratio of catecholamines to ATP-catabolites present in the perfusates was similar to that of catecholamines to ATP in chromaffin granules. 3.
