Summary A sample of South African Negroids (n=791) was scored for each individual's Bf and GLO phenotype. (The genes for the Bf and GLO polymorphisms are included in a known cluster of linked genes on chromosome 6.) Following a 2-test the respective two series of alleles were found to be distributed at random, i.e., there was no evidence for a linkage disequilibrium. Summary The newly described genetic polymorphism of glyoxalase I (GLO) is studied in seven ethnically defined Negroid samples from South Africa (total: n=843). The allele frequencies between the different Negroid samples studied vary only marginally. Summary A fetuin, fucosyl transferase has been identified in the smooth microsomal fraction from the rat exocrine pancreas. This enzyme is involved in the glycosylation of secretory proteins and is bound to membranes, predominantly of the Golgi complex. Optimal in vitro conditions for the assay of the enzyme activity were established: a pH of 5.5–6.0, a temperature of 21° C and concentrations of Mg+ + at 5.0 mM and ATP at 2.0 mM.Supported by a grant from the Deutsche Forschungsgemeinschaft, Bonn-Bad Godesberg (Ke 113/10). Synopsis The numbers of trout,Salmo trutta, in Granslev », Denmark, were estimated by the removal method on 18 dates from March 1974 to March 1976. Populations density varied from 0.39 to 0.74 trout m–2 in 1974–1975 and from 0.36 to 0.59 m–2 in 1975–1976 and at all times four or five year classes were present. The age structure of the population was unstable and the variable natural survival, immigration into and emigration from the study site could not be separated. An annual growth cycle with the most rapid growth for all year classes taking place from May to early August was found. Statistically significant differences between different years occurred in the growth of the 0,I and II age groups, but no evidence of density-dependent growth was found. The biomass ranged from 35.4 to 9.5 g m–2. The total mean annual biomass was 22.8 and 14.7 g m–2 in the two years and the II group made the greatest contribution, 44 and 48%, respectively. During 1975–1976 the mean annual biomass of each year class only was about two-thirds of that in 1974–1975. Annual production in the two years was 25.7 (range 24.7–28.5) and 12.6 g m–2 (range 11.7–15.0) and the II group accounted for about 46 and 38% of the production. In addition eel,Anguilla anguilla, produced about 0.5 g m–2 yr–2. The unstable age structure of the trout population was compared with trout populations from other streams. The importance of immigration as a recruitment process in middle and lower reaches of streams and of migrations as a mechanism to optimise utilization of the total stream habitat, as well as temperature as a factor controlling the growth rate are discussed.  相似文献

The production and chlorophyll concentration of epipelic and epilithic algae was measured during four years (1972–1975) in two shallow, Swedish subarctic lakes. One lake (Lake Hymenjaure) was fertilized with phosphorus or a combination of phosphorus and nitrogen while the other (Lake Stugsjön) served as a reference. The benthic algae in both lakes were dominated by Cyanophyceae of the same species during the whole investigation. The chlorophyll concentration of epipelic and epilithic algae was 100 and 20 mg·m–2 respectively and fairly constant during the season. In 1974–1975 there was a significant increase in chlorophyll concentration of the benthic algae in Lake Hymenjaure, probably as a response to the poorer light climate in the lake due to a large phytoplankton development. The annual benthic production was 3.4–7.2 gC·m–2 and it was not enhanced by the fertilization. Compared to the other primary producers (phytoplankton and macrophytes) the benthic algae constituted 70–83% of the total production in Lake Stugsjön. In Lake Hymenjaure, however, the importance of the benthic algae decreased from 50 to 22% of the total due to the great increase in phytoplankton production induced by the lake fertilization.  相似文献

Summary RNA labelled during oogenesis or early embryogenesis was isolated from eggs of the leaf hopperEuscelis plebejus. The polyadenylated RNA fraction deposited during early oogenesis accounted for approximately 2.7% of the total RNA content of the newly laid egg. This fraction differed significantly in molecular weight (15–32 S) from poly(A)-containing RNA synthesised between early cleavage and early germ anlage stages (4–20S). Locally injected3H-uridine spread through the egg within approximately 3 h. A considerable fraction (25–35%) of label injected as3H-uridine during early cleavage was recovered in DNA at subsequent stages (10–20 h later); labelled RNA was not found prior to the cellular blastoderm stage. When the yolk-endoplasm was separated from the blastoderm cells, only the latter contained demonstrable amounts of RNA synthesised by the embryo. Of the precursor incorporated into embryonic RNA, approximately 10% was found in the polyadenylated fraction at the early blastoderm stage, but only 3% at the early germ anlage stage. No differences in size distribution of polyadenylated RNA were evident between anterior and posterior halves of the early germ anlage stage.Supported by the Deutsche Forschungsgemeinschaft, SFB 46  相似文献

Summary Four brook-dwelling populations of Ancylus fluviatilis were studied. Data on life-cycle, specimen number, biomass and egg capsule biomass are reported. By means of experimental data on ingestion, assimilation, growth and egg-production, energy balances are calculated for whole populations.Mean annual biomass turned out to be 26.58–64.88 mg C/m2. Mean annual biomass turn-over ratio (production/biomass) was 2.64–5.95. Cohort biomass turnover ratio was 4.11–6.09 for the whole population. Young specimens show considerably higher production efficiencies than adult ones. Production by the smallest size classes is a very considerable part of whole population production. Net production efficiency of the whole population was 11.2–11.8%, gross production efficiency 5.6–6.5%. An important factor for variation between different populations is temperature, but other factors, such as food supply, may be important, too. Experimental production estimates were compaerd with the Allen curve method and the instantaneous growth method.Differences due to different methods of production measurement are discussed. The range of values for different efficiencies lies within the range reported for other freshwater snails. Interpopulation variation in turnover ratios nearly equals variation reported for different species of univoltine snails.Supported by the Deutsche Forschungsgemeinschaft.  相似文献

Protoplasts of Cyamopsis tetragonoloba were isolated from leaves of in vitro grown plants. The yield of the protoplasts, their viability and subsequent divisions were greatly influenced by the pH of the media used for isolation and culture of protoplasts. Sustained divisions of the cultured protoplasts were best supported by a modified Kao and Michayluk (1975) nutrient medium containing glucose (0.4 M), NAA (4 mgl–1), 2,4-D (1 mgl–1) and KIN (2 mgl–1 ). The protoplast derived cells developed calli on transfer to Murashige and Skoog (1962) medium supplemented with 1 mgl–1 each of 2,4-D, NAA and KIN.  相似文献

Primary productivity and trophic status of a Kashmir Himalayan Lake   总被引:3,自引:3,他引:0  
The paper deals with general limnology, phytoplankton production and trophic status of a Himalayan lake of Kashmir. The primary production as measured by in situ 14C istope technique ranged from 36 to 906 mgC.m–2 d–1 with characteristic seasonal variation. The photosynthetic efficiency was from 0.02 to 0.48% during the same period. The yearly (1975–1976) production was estimated at 134 gC.m–2 m–2, which shows that the lake has not yet reached a high level of eutrophication. Phosphoenolpyruvate carboxylase (PEPCase; EC 4.1.1.31) activity was found to be modulated by light and darkness when measured in the presence of K+, which had been added to induce swelling of guard-cell protoplasts (GCPs) from Vicia faba L., whereas no modulation was detected in the absence of K+ (PEPcase activity remained constant at 1.5±0.15 pmol PEP metabolized · GCP–1 ·h–1; subsequently, pmol GCP–1 ·h–1 will be used). The activity of PEPCase increased by 100% (from 1.5 to 3 pmol·protoplast–1·h–1) in darkness and by 200% (from 1.7 to 5 pmol·protoplast–1· h–1) in light and oscillations in activity of these magnitudes were repeated at intervals of 2 min (dark) and 2.5 min (light) for a period of 10 min during K+-induced increase in the volume of GCPs. The oscillations were reflected in changes in malate-pool sizes determined in plastids, mitochondria and the supernatant fraction (consisting of the cytosol and the vacuole). Malate probably functioned as a mitochondrial substrate, thus supplying ATP for K+ uptake and the swelling of the protoplasts. On the basis of the present paper and previous results (H. Schnabl and B. Michalke 1988, Life Sci. Adv. Zusammenfassung Durch Vergleich licht- und elektronenmikroskopischer Befunde am Nebennierenmark wird als praktische Erfassungsgrenze für Catecholamine 10–11g Adrenalin pro Zelle ermittelt. Das entspricht einer Konzentration im Zytoplasma von 1,0%. Diese Konzentration wird auch als Verdünnungsgrenze in Modellversuchen festgestellt. Aufgrund theoretischer Überlegungen wird dargelegt, daß 10–11 bis 10–12g/Zelle allgemein die für eine beliebige histochemische Reaktion notwendige Konzentration darstellt.
Summary Comparing light and electron-microscopical data from adrenal medulla, the identification limit for catecholamines has been calculated to be in the order of 10–11g adrenaline/cell. This is equivalent to a concentration of 1,0% adrenaline/cell which has been found to be the dilution-limit in model experiments, too. On the basis of theoretical calculation it has been demonstrated that 10–11–10–12g/cell is the concentration needed for any histochemical reaction.

The hydraulic conductivity of the lateral walls of early metaxylem vessels (Lpx in m · s–1 · MPa–1) was measured in young, excised roots of maize using a root pressure probe. Values for this parameter were determined by comparing the root hydraulic conductivities before and after steam-ringing a short zone on each root. Killing of living tissue virtually canceled its hydraulic resistance. There were no suberin lamellae present in the endodermis of the roots used. The value of Lpx ranged between 3 · 10–7 and 35 · 10–7 m · s–1 · MPa–1 and was larger than the hydraulic conductivity of the untreated root (Lpr = 0.7 · 10–7 to 4.0 · 10–7 m · s–1 · MPa–1) by factor of 3 to 13. Assuming that all flow through the vessel walls was through the pit membranes, which occupied 14% of the total wall area, an upper limit of the hydraulic conductivity of this structure could be given(Lppm=21 · 10–7 to 250 · 10–7 m · s–1 · MPa–1). The specific hydraulic conductivity (Lpcw) of the wall material of the pit membranes (again an upper limit) ranged from 0.3 · 10–12 to 3.8 · 10–12 m2 · s–1 · MPa–1 and was lower than estimates given in the literature for plant cell walls. From the data, we conclude that the majority of the radial resistance to water movement in the root is contributed by living tissue. We have identified a whole-cell Cl current activated by hyposmotic stress in rat lacrimal acinar cells using the patch-clamp technique. Superfusion of isolated single cells with hyposmotic solution (80% of control osmolarity) caused a gradual increase of the current, which was reversed on return to the control solution. The current-voltage relationship showed outward rectification, and the current showed time and voltage dependence: slowly activated by depolarizing voltages and rapidly inactivated by hyperpolarizing voltages. The increase in current was not observed when intracellular Ca2+ was chelated with EGTA. It was also inhibited by the absence of extracellular Ca2+, or the presence of gadolinium ions (20 m Gd3+). We conclude that in rat lacrimal acinar cells hyposmotic stress activates Ca2+-dependent Cl channels as a result of Ca2+ influx through a Gd3+-sensitive pathway. The Cl channels involved appear to be indistinguishable from those activated by muscarinic stimulation. The inhibitory effect of Gd3+ suggests that stretch-activated nonselective cation channels may be responsible for the Ca2+ influx.The authors are grateful to Prof. R.M. Case, Dr. A.C. Summary The light-mediated, time-dependent rise in the pH value at the center of an alkaline band was analyzed using the methods of numerical analysis. From this analysis an expression of the time-dependent build-up of OH efflux was obtained for these bands. This information can now be employed to determine whether the light-activated transport of OH and HCO 3 influences the electrical properties of the plasmalemma. The dark-induced deactivation of OH transport was also characterized, revealing a transition from efflux to a transient influx phase during deactivation.Numerical analysis of the steady-state OH diffusion pattern, established along the surface of an alkaline band, revealed that the OH efflux width was wider than previously envisaged. It was also found that OH sink regions exist on either side of the efflux zone. These, and other characteristics revealed by the numerical analysis, enabled us to extend the OH transport model proposed by Lucas (J. Exp. Bot. 1975,26:347).  相似文献

Summary Phenotype and gene frequencies of the ceruloplasmin polymorphism are reported. In all populations considered here (Germans, Icelanders, Iranians, Pakistani, and Koreans) high frequencies of the allele CpB (0.978–0.996) could be observed, whilst the frequencies of the alleles CpA (0.003–0.013) and CpC (0.000–0.013) are very low.
Zusammenfassung Die Verfasser berichten über Phänotypen-und Gen-Frequenzen des Coeruloplasmin-Polymorphismus. In allen untersuchten Populationen (Deutsche, Isländer, Iraner, Pakistani und Koreaner) konnten sehr hohe Frequenzen des Allels CpB beobachtet werden (0,978–0,996), während die Frequenzen der Allele CpA (0,003–0,013) und CpC (0,000–0,013) deutlich geringer sind.

Quantitative samples of Gammarus pulex L. taken from a small Danish stream during 1975 showed mean annual population densities varying from 500 m–2 in early May to 5 500 m–2 in late September. The mean annual biomass was 1.5 g dry weight m–2. No discrete cohorts could be distinguished from the size frequency distributions. Annual production, estimated by the size-frequency method, was 3.9 g dry weight M-2 and P/B ratio was 2.6. The contribution to trout energetics may have been as much as 17%.  相似文献

The aim of the study was to show which tissues and cell types of the cotyledon of Ricinus communis L. are responsible for uptake of sucrose by H+-sucrose symport. The cotyledons were incubated in labelled sucrose for up to 20 min and then the amount of radioactivity in each cell type of the cotyledon was assessed by microautoradiography. It was found that 50% of the label was present in the spongy mesophyll, and 10–15% was in the bundles, the epidermal layers and the palisade parenchyma. The sieve tubes contained only 2–3% of the label. The addition of sucrose to cotyledons depolarized the membrane of spongy-mesophyll cells by 33 mV. Therefore, it was concluded that the previously found H+-sucrose symport is at least partly located at the spongy mesophyll. No precursor-like behaviour of the label in mesophyll or bundle-sheath cells was observed in pulse-chase experiments, which indicates a direct uptake of sucrose by the sieve tube-companion cell complex from the apoplast.This work was funded by Deutsche Forschungsgemeinschaft. Annual egg production was determined for Artemia monica in Mono Lake, California, from 1983 to 1987. Annual oviparous (overwintering cyst) production was 3 and 7 million cysts m–2 yr–1 in 1986 and 1987, respectively, as measured by in situ sediment traps. Cyst production for the entire five year period was calculated using Artemia census data and inter-brood duration derived from mixolimnetic temperature. These estimates ranged from 2 to 5 million cysts m–2 yr–1. This method underestimated annual production by 30%, when compared to estimates using sediment traps. Cyst production was similar during 1983–1986 and showed a significant increase in 1987, which was due primarily to a larger reproductive population later in the year. Recruitment into the adult populations of the following spring ranged between 1.4 to 3.2%. Overall abundance of this generation reflected the patterns in annual cyst production. Compensatory effects must operate on the second generation of each year, since summer populations were similar in all years despite differences in cyst production.  相似文献

Summary Southern African Bantu-speaking negroid and San populations were examined with regard to the glucose-6-phosphate dehydrogenase (G6PD) PvuII restriction fragment length polymorphism (RFLP) showing alleles of 4kb and 1.6 kb, called Type 1 and Type 2, respectively. The standardized disequilibrium coefficient for the electrophoretic G6PD types and PvuII alleles in the Southern African population was 0.28. The molecular lesion causing the GdA mutation is the same in the San and Southern African negroid populations. GdA chromosomes are found in association with both the Type 1 and Type 2 alleles, whereas none of the 62 GdB chromosomes from the Southern African populations had the Type 2 allele. Five of the 44 GdB chromosomes studied in the American Black population had the Type 2 allele, indicating that the GdB allele in the two populations may have different origins. The presence of all 3 A deficiency mutations in the G6PD A gene, in a region where the ancestral population was thought to have predominantly G6PD B, may be explained by their origin in Africa after the divergence of the races.  相似文献

While the common red cell phenotype of lactate dehydrogenase in mammals includes A and B subunits, in some species, especially of the rodent suborder Myomorpha, LDH B subunits are absent in erythrocytes (phenotype B). A polymorphism involving LDH B+ and LDH B phenotypes was observed in Mus musculus, and it was found that LDH B is recessive in relation to LDH B+ (Shows and Ruddle, 1968). A similar polymorphism is described here in the field mouse, Apodemus sylvaticus. In family studies, the recessive mode of inheritance is confirmed. Induction of reticulocytosis reveals that the LDH B phenotype is not the consequence of degradation of the B subunits in aging red cells.
