CARMA1, BCL10 and MALT1 form a large molecular complex known as the CARMA1 signalosome during lymphocyte activation. Lymphocyte activation via the CARMA1 signalosome is critical to immune response and linked to many immune diseases. Despite the important role of the CARMA1 signalosome during lymphocyte activation and proliferation, limited structural information is available. Here, we report the dimeric structure of CARMA1 CARD at a resolution of 3.2 Å. Interestingly, although CARMA1 CARD has a canonical six helical-bundles structural fold similar to other CARDs, CARMA1 CARD shows the first homo-dimeric structure of CARD formed by a disulfide bond and reveals a possible biologically important homo-dimerization mechanism.  相似文献

A sensitive and selective LC-MS-MS method has been developed and validated for the determination of cryptotanshinone (CTS) and its active metabolite tanshinone II A (TS II A) in rat plasma using fenofibrate (FOFB) as internal standard. Liquid-liquid extraction was used for sample preparation. Chromatographic separation was achieved on a Waters symmetry ODS column using methanol and water (85:15) as mobile phase delivered at 1.0 mL/min. LC-MS-MS analysis was carried out on a Finnigan LC-TSQ Quantum mass spectrometer using atmospheric pressure chemical ionization (APCI) and positive multiple reaction monitoring. Ions monitored were m/z 297.0--> 251.0 for CTS, m/z 295.0--> 249.0 for TS II A, and m/z 361.1--> 233.0 for FOFB with argon at a pressure of 0.2 Pa and collision energy of 25 eV for collision-induced dissociation (CID). The assay was linear over the range 0.1-20 ng/mL for CTS and 0.2-15 ng/mL for TS II A. The average recoveries of CTS and TS II A from rat plasma were 93.7 and 94.7%, respectively. The established method has been applied in a pharmacokinetic study of CTS in rats.  相似文献

The CBM complex (CARMA1, BCL10 and MALT1) plays a crucial role in B and T lymphocyte activation. CARMA1 serves as a scaffold for BCL10, MALT1 and other effector proteins and regulates various signaling pathways related to the immune response. The assembly of CARMA1 and BCL10 is mediated through a CARD-CARD interaction. Here, we report the crystal structure of the CARD domain of CARMA1 at a resolution of 1.75 Å. The structure consists of six helices, as previously determined for CARD domains. Structural and computational analysis identified the binding interface between CARMA1-CARD and BCL10-CARD, which consists of a basic patch in CARMA1 and an acidic patch in BCL10. Site-directed mutagenesis, co-immunoprecipitation and an NF-κB activation assay confirmed that the interface is necessary for association and downstream signaling. Our studies provide molecular insight into the assembly of CARMA1 and BCL10.  相似文献

A multi-car elevator system is an elevator system that has more than one car installed in each elevator shaft. This system enables us to improve the transportation capability without increasing the occupied floor space. The primary purpose of this study is to consider the group control problem of operating cars efficiently without collision nor reversal in a multi-car elevator system, based on a detailed and realistic model where floor stoppage time of cars cannot be known in advance. In the context of elevator systems, reversal means that a car travels in the direction opposite to the desired direction of on-board passengers, which is prohibited because it makes passengers uncomfortable. We first propose an optimization-based collision and reversal avoidance method to operate cars in the same shaft. Next, we construct simple methods to allocate calls to individual cars under immediate and delayed guidance policies. Under the immediate guidance policy a call is allocated to a car immediately after it is registered, while under the delayed guidance policy the allocation may be changed until it is actually served. The effectiveness of the proposed group control method is examined by computer simulation.  相似文献

T lymphocyte (T cell) activation and proliferation is induced by the activation of multiple signal transduction pathways. Earlier studies indicate that CARMA1, a Caspase Recruitment Domain (CARD) and Membrane-associated GUanylate Kinase domain (MAGUK)-containing scaffold protein, plays an essential role in NF-kappaB activation induced by the costimulation of T cell receptor (TCR) and CD28 molecules. However, the molecular mechanism by which CARMA1 mediates TCR-CD28 costimulation-induced NF-kappaB activation is not fully understood. Here we show that CARMA1 is constitutively oligomerized. This oligomerization of CARMA1 is through its Coiled-coil domain. Disruption of the predicted structure of the Coiled-coil domain of CARMA1 impaired its oligomerization and, importantly, abrogated CARMA1-mediated NF-kappaB activation. Interestingly, disruption of the CC1 domain abrogates CARMA1 localization, whereas disruption of the CC2 domain seems to inhibit CARMA1 self-association. Together, our results demonstrate that the oligomerization of CARMA1 is required for TCR-induced NF-kappaB activation.  相似文献

Current nerve conduction studies (NCS) are influenced by the activity of the largest active fibers, making it difficult to assess the state of smaller nerve fibers. This study is aimed at alternative diagnostic techniques for assessing carpal tunnel syndrome (CTS). A conduction velocity distribution (CVD) estimator based on the collision technique that incorporates volume conductor modeling is proposed and discussed in this paper. Simulations were run to evaluate the accuracy of the CVD estimator and compare its performance with previous CVD estimators based on the collision technique. Results show the improved accuracy of the proposed approach, which is able to provide estimates with a percent mean square error (PMSE) lower than 1.1% for all CTS cases studied and lower than 2% in the presence of additive white Gaussian noise. Simulations also showed that conduction slowing in the carpal tunnel (CT) segment is detected by the proposed technique and displayed as an increase in the number of low velocity fibers. Results suggest that both CVD estimator and amplitude parameter proposed can help detect the severity of CTS in a patient more accurately than current NCS.  相似文献

Longitudinal profiles of water quality along a well-mixed tidal river are, ideally, based on simultaneous measurements at fixed stations distributed along the river. The resolution of the profiles is limited by the density of the stations. However, for a given number of stations the resolution is greatly increased if water quality date can be extrapolated upstream and downstream of the stations, making use of velocity data; the resolution is then determined by the density of the extrapolated data points, which may be an order of magnitude higher than the density of the stations. A 15-km length of river was investigated using 5 current meters equipped to measure depth, temperature, conductivity and dissolved oxygen. Data were recorded simultaneously every 10 minutes. When the average cross-sectional speed was 0.25 ms−1 (typical of tidal conditions), the extrapolated data points were 150 m apart, so the resolution of the resulting profiles (7 per kilometre) was much higher than that of the stations alone (0.3 per kilometre). The extrapolation process required a means of deducing the average cross-sectional speed from the speed measured at a given station. The key to this was provided by temperature data recorded during the onset of a spate, when tidal flow was suspended and the average cross-sectional speed was uniformly about 0.75 ms −1 at four of the stations. Profiles of temperature and dissolved oxygen were generated by this method; the resolution was about 2 data points km−1 during the onset of the spate, and 6 points km−1 during tidal flow.  相似文献

Collision avoidance models derived from the study of insect brains do not perform universally well in practical collision scenarios, although the insects themselves may perform well in similar situations. In this article, we present a detailed simulation analysis of two well-known collision avoidance models and illustrate their limitations. In doing so, we present a novel continuous-time implementation of a neuronally based collision avoidance model. We then show that visual tracking can improve performance of these models by allowing an relative computation of the distance between the obstacle and the observer. We compare the results of simulations of the two models with and without tracking to show how tracking improves the ability of the model to detect an imminent collision. We present an implementation of one of these models processing imagery from a camera to show how it performs in real-world scenarios. These results suggest that insects may track looming objects with their gaze.  相似文献

Avoiding collisions is one of the most basic needs of any mobile agent, both biological and technical, when searching around or aiming toward a goal. We propose a model of collision avoidance inspired by behavioral experiments on insects and by properties of optic flow on a spherical eye experienced during translation, and test the interaction of this model with goal-driven behavior. Insects, such as flies and bees, actively separate the rotational and translational optic flow components via behavior, i.e. by employing a saccadic strategy of flight and gaze control. Optic flow experienced during translation, i.e. during intersaccadic phases, contains information on the depth-structure of the environment, but this information is entangled with that on self-motion. Here, we propose a simple model to extract the depth structure from translational optic flow by using local properties of a spherical eye. On this basis, a motion direction of the agent is computed that ensures collision avoidance. Flying insects are thought to measure optic flow by correlation-type elementary motion detectors. Their responses depend, in addition to velocity, on the texture and contrast of objects and, thus, do not measure the velocity of objects veridically. Therefore, we initially used geometrically determined optic flow as input to a collision avoidance algorithm to show that depth information inferred from optic flow is sufficient to account for collision avoidance under closed-loop conditions. Then, the collision avoidance algorithm was tested with bio-inspired correlation-type elementary motion detectors in its input. Even then, the algorithm led successfully to collision avoidance and, in addition, replicated the characteristics of collision avoidance behavior of insects. Finally, the collision avoidance algorithm was combined with a goal direction and tested in cluttered environments. The simulated agent then showed goal-directed behavior reminiscent of components of the navigation behavior of insects.  相似文献

The IEEE 802.11 network technology is the emerging standard for wireless LANs and mobile networking. The fundamental access mechanism in the IEEE 802.11 MAC protocol is the Distributed Coordination Function. In this paper, we present an analytical method of estimating the saturation throughput of 802.11 wireless LAN in the assumption of ideal channel conditions. The proposed method generalizes the existing 802.11 LAN models and advances them in order to take the Seizing Effect into consideration. This real-life effect consists in the following: the station that has just completed successfully its transmission has a better chance of winning in the competition and therefore of seizing the channel than other LAN stations. The saturation throughput of 802.11 wireless LANs is investigated by the developed method. The obtained numerical results are validated by simulation and lead to the change of the existing idea of the optimal access strategy in the saturation conditions.  相似文献

Flies are capable of extraordinary flight maneuvers at very high speeds largely due to their highly elaborate visual system. In this work we present a fly-inspired FPGA based sensor system able to visually sense rotations around different body axes, for use on board micro aerial vehicles (MAVs). Rotation sensing is performed analogously to the fly’s VS cell network using zero-crossing detection. An additional key feature of our system is the ease of adding new functionalities akin to the different tasks attributed to the fly’s lobula plate tangential cell network, such as object avoidance or collision detection. Our implementation consists of a modified eneo SC-MVC01 SmartCam module and a custom built circuit board, weighing less than 200 g and consuming less than 4 W while featuring 57,600 individual two-dimensional elementary motion detectors, a 185° field of view and a frame rate of 350 frames per second. This makes our sensor system compact in terms of size, weight and power requirements for easy incorporation into MAV platforms, while autonomously performing all sensing and processing on-board and in real time.  相似文献

Flight behaviour characteristics such as flight altitude and avoidance behaviour determine the species-specific collision risk of birds with wind turbines. However, traditional observational methods exhibit limited positional accuracy. High-resolution GPS telemetry represents a promising method to overcome this drawback. In this study, we used three-dimensional GPS tracking data including high-accuracy tracks recorded at 3-s intervals to investigate the collision risk of breeding male Montagu's Harriers Circus pygargus in the Dutch–German border region. Avoidance of wind turbines was quantified by a novel approach comparing observed flights to a null model of random flight behaviour. On average, Montagu's Harriers spent as much as 8.2 h per day in flight. Most flights were at low altitude, with only 7.1% within the average rotor height range (RHR; 45–125 m). Montagu's Harriers showed significant avoidance behaviour, approaching turbines less often than expected, particularly when flying within the RHR (avoidance rate of 93.5%). For the present state, with wind farms situated on the fringes of the regional nesting range, collision risk models based on our new insights on flight behaviour indicated 0.6–2.0 yearly collisions of adult males (as compared with a population size of c. 40 pairs). However, the erection of a new wind farm inside the core breeding area could markedly increase mortality (up to 9.7 yearly collisions). If repowering of the wind farms was carried out using low-reaching modern turbines (RHR 36–150 m), mortality would more than double, whereas it would stay approximately constant if higher turbines (RHR 86–200 m) were used. Our study demonstrates the great potential of high-resolution GPS tracking for collision risk assessments. The resulting information on collision-related flight behaviour allows for performing detailed scenario analyses on wind farm siting and turbine design, in contrast to current environmental assessment practices. With regard to Montagu's Harriers, we conclude that although the deployment of higher wind turbines represents an opportunity to reduce collision risk for this species, precluding wind energy developments in core breeding areas remains the most important mitigation measure.  相似文献

The article introduces and elaborates the hypothesis of carrier features, characteristics which in females are attractive to males as mate-choice cues. Female carrier features increase paternal resemblance and are ultimately attractive because they help pair-bonded males distinguish genetic offspring from those conceived by mates in extra-pair copulations. The proposed kin recognition mechanism, whether culturally evolved or innate, facilitates discriminatory paternal investment, and hence male fitness. Carrier features would be attractive to males in monogamous species where paternal investment is high, and cuckoldry represents a significant risk to male fitness. Logical space exists for male sender features, which tend to be expressed in offspring regardless of female characteristics. Conflict of genetic interests between the sexes should favor the evolution of carrier, rather than sender, features in both sexes. The argument centers on humans, for whom candidate carrier features are discussed with regard to physiognomy, behavior, recessive traits, and body odor. Criticisms are discussed, and testable predictions enumerated.  相似文献

This paper presents a mathematical programming model to help select equipment for a flexible manufacturing system, i.e., the selection of the types and numbers of CNC machines, washing stations, load/unload stations, transportation vehicles, and pallets. The objective is to minimize equipment costs and work-in-process inventory cost, while fulfilling production requirements for an average period. Queueing aspects and part flow interactions are considered with the help of a Jacksonian-type closed queueing network model in order to evaluate the system's performance. Since the related decision problem of our model can be shown to be NP-complete, the proposed solution procedure is based on implicit enumeration. Four bounds are provided, two lower and two upper bounds. A tight lower bound is obtained by linearizing the model through the application of asymptotic bound analysis. Furthermore, asymptotic bound analysis allows the calculation of a lower bound for the number of pallets in the system. The first upper bound is given by the best feasible solution and the second is based on the anti-starshaped form of the throughput function.  相似文献

High-resolution light-microscopy and high-throughput screening are two essential methodologies for characterizing cellular phenotypes. Optimally combining these methodologies in cell-based screening to test detailed molecular and cellular responses to multiple perturbations constitutes a major challenge. Here we describe the development and application of a screening microscope platform that automatically acquires and interprets sub-micron resolution images at fast rates. The analysis pipeline is based on the quantification of multiple subcellular features and statistical comparisons of their distributions in treated vs. control cells. Using this platform, we screened 2200 natural extracts for their effects on the fine structure and organization of focal adhesions. This screen identified 15 effective extracts whose fractionation and characterization were further analyzed using the same microscope system. The significance of combining resolution, throughput and multi-parametric analyses for biomedical research and drug discovery is discussed.  相似文献

Multi carrier code division multiple access (MC-CDMA) system is a promising multi carrier modulation (MCM) technique for high data rate wireless communication over frequency selective fading channels. MC-CDMA system is a combination of code division multiple access (CDMA) and orthogonal frequency division multiplexing (OFDM). The OFDM parts reduce multipath fading and inter symbol interference (ISI) and the CDMA part increases spectrum utilization. Advantages of this technique are its robustness in case of multipath propagation and improve security with the minimize ISI. Nevertheless, due to the loss of orthogonality at the receiver in a mobile environment, the multiple access interference (MAI) appears. The MAI is one of the factors that degrade the bit error rate (BER) performance of MC-CDMA system. The multiuser detection (MUD) and turbo coding are the two dominant techniques for enhancing the performance of the MC-CDMA systems in terms of BER as a solution of overcome to MAI effects. In this paper a low complexity iterative soft sensitive bits algorithm (SBA) aided logarithmic-Maximum a-Posteriori algorithm (Log MAP) based turbo MUD is proposed. Simulation results show that the proposed method provides better BER performance with low complexity decoding, by mitigating the detrimental effects of MAI.  相似文献
