Rat skeletal muscle triacylglycerol utilization during exhaustive swimming   总被引:1,自引:0,他引:1  
The utilization of triacylglycerol in slow oxidative, fast oxidative-glycolytic, and fast glycolytic skeletal muscle fiber types was examined in rats subjected to a prolonged exhaustive swim. Significant reductions of intramuscular triacylglycerol occurred following 2 h and 40 min of swimming in all muscles containing a predominance of slow oxidative and fast oxidative-glycolytic fibers, which possess a high capacity for free fatty acid oxidation. Triacylglycerol content in the soleus decreased by 48%, and reductions of 41, 29, and 27% were measured in the red vastus lateralis, red gastrocnemius, and plantaris muscles, respectively. In the white vastus lateralis and white gastrocnemius muscles (fast glycolytic fibers) triacylglycerol concentrations were unaffected. In all muscles the variability of intramuscular triacylglycerol measurements between animals was 20-50% and the within animal variance (right vs. left hindlimb) was similar. Analytical repeatability was approximately 10% in all muscles and significantly less than the between- and within-animal variances. It was concluded that a real biological variation exists in the triacylglycerol content of all rat skeletal muscles and that intramuscular triacylglycerol is an important energy source during prolonged exercise of moderate intensity.  相似文献

An electron microscopical and histochemical investigation of bioptates obtained from the external broad femoral muscle of 74 sportsmen-skaters has been performed. Structural adaptation of the muscles to speed loadings and to endurance loadings has been analysed taking into consideration contents of muscle fibers (MF) of various type. Peculiarities in ultrastructural organization of the skeletal MF in the sprinter- and stayer-skaters, who undergo training according to a special program, are described; the character of the muscle changes is also followed in the sportsmen who undergo training according to the program that does not correspond to the contents of their MF. Lesions in the latter up to the necrotic ones are mostly found in the muscles of the stayer-skaters, as well as in the sprinters trained according to the stayer program. Presence of essential destructive alterations of the muscles in the skaters, especially in those whose contents of slow muscles do not correspond to their specialization, demonstrates, evidently, the fact that the given physical loading is not adequate to the functional or potential possibilities of the organism.  相似文献

The effects of classical strength training (CT) and low intensity strength training without relaxation (TwR) upon size, strength and fatigability of leg muscles in men were compared. A 8-10-week strength training led to an increase of size and maximal voluntary contraction of trained muscles. After the CT, the increment of strength was higher; on the other hand, strength increments related to total work performed increased after the TwR noticeably higher than after the CT. Two training programs influenced the size of total muscle and of muscle fibers (MF) differently: the volume of m. quadriceps femoris increased more after the CT than after the TwR. The CT induced a significant increase of cross sectional area (CSA) of fast MF, and the TwR induced an increase of CSA of slow MF. Resistance to fatigue after the TwR was higher than after the. The effects of TwR were more pronounced in single-joint movements training than in multi-joint movement.  相似文献

The purpose of this study was to determine the electromyographic (EMG) power spectral characteristics of seven trunk muscles bilaterally during two complex isometric activities extension-rotation and flexion-rotation, in both genders to describe the frequency-domain parameters. Eighteen normal young subjects volunteered for the study. The subjects performed steadily increasing isometric extension-rotation and flexion-rotation contractions in a standard trunk posture (40 degrees flexed and 40 degrees rotated to the right). A surface EMG was recorded from the external and internal oblique, rectus abdominis, pectoralis, latissimus dorsi, and erector spinae muscles at the 10th thoracic and the 3rd lumbar vertebral levels, at 1 kHz and 25%, 50%, 75% and 100% of maximal voluntary contraction (MVC). The median frequency (MF), mean power frequency (MPF), frequency spread and peak power were obtained from fast Fourier transform analysis. The MF and MPF for both extension-rotation and flexion-rotation increased with the grade of contraction for both males and females. The EMG spectra in flexion-rotation were different from those of extension-rotation (P < 0.001). The left external and right internal oblique muscles played the role of antagonists in trunk extension-rotation. There was an increase in the MF of the trunk muscles with increasing magnitude of contraction. Frequency-domain parameters for both the male and female subjects were significantly different (P < 0.001).  相似文献

Our purpose was to characterize the relationship between EMG mean power frequency (MPF) or median frequency (MF) and rate of torque development in voluntary ballistic and electrically elicited isometric contractions. Twenty-three healthy adults participated in two sets of experiments performed on elbow flexor muscles. For Experiment 1, subjects were asked to generate voluntary ballistic contractions by reaching four different target torque levels (20, 40, 60 and 100% of the maximal voluntary contraction (MVC)) as fast as they could. For Experiment 2, electrical (M-waves) and mechanical (twitches) responses to electrical stimulation of the nerves supplying the biceps brachii and brachioradialis muscles were recorded with the subjects at rest and with a background isometric contraction of 15% MVC. MPF, MF and rate of torque development (% MVC/s) were calculated for both voluntary and elicited contractions. Significant positive correlations were observed between MPF and rate of torque development for the voluntary contractions, whereas significant negative correlations were observed between the two variables for elicited contractions. This suggests that factors other than muscle fiber composition influence the frequency content of EMG signals and/or the rate of torque development, and that the effect of these factors will vary between voluntary and elicited contractions.  相似文献

Studies have been made on Mg2(+)-ATPase, Ca2(+)-sensitivity, superprecipitation and fractional composition of natural and desensitized actomyosin from myocardium, slow and fast skeletal muscles after physical training (swimming, gravitational loading) and after monthly readaptation. Physical overloading makes physicochemical properties and protein composition of actomyosin from the myocardium and slow skeletal muscles similar to those in fast skeletal ones. Changes in actomyosin from the myocardium and slow skeletal muscles are more profound, whereas the recovery of the initial properties during readaptation reveals high plasticity of muscles of these phenotypes. Changes in Ca regulation depend mainly on muscle phenotype. Different plasticity of muscles of various phenotypes during readaptation results from differences in the synthesis of protein components of myofibrils.  相似文献

Studies have been made on glycogen content as well as on the activity of phosphorylase and glucose-6-phosphatase in fast and slow muscles from representatives of 6 classes of vertebrates (Lampetra fluviatilis, Cyprinus carpio, Rana temporaria, Rana ridibunda, Emys orbicularis, hen, rat). Glycogen level and glucose-6-phosphatase activity are either higher in slow muscles, or practically identical in both types of muscles (glucose-6-phosphatase is absent from the fast muscles of hens and rats). On the contrary, phosphorylase activity is higher in fast muscles, this finding being true only for higher vertebrates and lampreys.  相似文献

The effects of mechanical stimulation of the soles’ support zones in the modes of slow and fast walking (75 and 120 steps per minute) were studied using the model of supportlessness (legs suspension). 20 healthy subjects participated in the study. EMG activity of hip and shin muscles was recorded. Kinematics of leg movements was assessed with the use of videoanalysis system. In 80% of cases support stimulation was followed by leg movements, in 69% of which they had characteristics of locomotions being accompanied by the burst-like electromyographic activities. The order of involvement of leg muscles and organization of antagonistic muscles activities were analogous to those of voluntary walking. The latencies of electromyographic activity in hip and shin muscles composed 5.17 ± 1.08 and 14.01 ± 2.82 s, respectively, the frequencies of bursts differed significantly depending on stimulation frequency. In 31% of cases the electromyographical activity following the stimulation of the soles’ support zones had not burst-like but uninterrupted pattern. Its amplitude rose smoothly reaching a certain level that was subsequently maintained. Results of the study showed that soles’ support zones stimulation in the mode of locomotion could activate a locomotor generator provoking the appearance of locomotion-like activity and that effect evoked by this stimulation includes not only rhythmical but also non-rhythmical (probably postural) components of walking.  相似文献

Summary In the soleus muscle of the normal rat the number of cells containing fast troponin I decreased and those containing slow troponin I increased after birth until less than 10% stained for the fast form in the adult muscle. On denervation of soleus muscle this pattern of change was reversed with the result that the majority of cells stained for fast troponin I. The change was more rapid when denervation was carried out at 12 weeks rather than at 52 weeks of age. Denervation of extensor digitorum longus and tibialis anterior muscles produced little change in the distribution of fast and slow troponin I over a period of 12 weeks. After long periods (>24 weeks) of denervation of these fast muscles, fast troponin I was observed in cells in which originally only slow troponin I could be detected. Similar results to those obtained with troponin I in both fast and slow muscles were obtained using antibodies to the fast and slow forms of troponin C and troponin T.  相似文献

The mechanical properties of the slow soleus and the fast rectus femoris muscle under passive stretching were studied in endurance trained, untrained and lathyritic rats, aged 3 months. The soleus muscle with more abundant and cross-linked collagen had higher ultimate tensile strength and tangent modulus compared to the fast rectus femoris muscle which, on the other hand, had higher maximum strain. The inhibition of collagen cross-linking by lathyrism resulted in decreased tensile strength and stiffness, especially in the soleus muscle, whereas endurance training showed the opposite effects. It is supposed that the properties of collagen partly explain the capacity of slow muscles to maintain posture and to perform prolonged dynamic work. The effects of training on the tensile properties further indicate the close relationship between intramuscular collagen and the endurance capacity of muscles.  相似文献

Fibre bundles or whole muscles from Xenopus laevis, ranging in size from 0.5-60g, were studied. Maximum power output of predominantly fast (sartorius) and slow (adductor magnus) muscles was measured at cycle frequencies between 0.5 and 20Hz, using the work loop technique. Power output was highly dependent on cycle frequency, and in 50-60g adults was maximal at 6 Hz for fast fibres (65 Wkg-1), and 2 Hz for slow fibres (14 Wkg-1). The cycle frequency for maximum power output was dependent on body mass (Mb), and decreased as a function of Mb-0.07 in fast fibres, and Mb-0.23 in slow fibres. The functional significance of these differences is discussed.  相似文献

Summary Potassium (K-) contractures were recorded from slow-twitch (mouse soleus) and fast-twitch (mouse extensor digitorum longus (EDL) and rat sternomastoid) muscles. The mouse limb muscles responded to a maintained increase in external potassium concentration with a rapid increase in tension (fast contracture) which inactivated and was followed by a slow contracture. Rat sternomatoid muscles responded with fast contractures only. The threshold potassium concentration for contraction was higher in fast-twitch muscles than in soleus muscles, at 22 and at 37°C. After corrections had been made for the more rapid depolarization of soleus fibers, the threshold potential for soleus fiber contraction was 15 mV closer to the resting membrane potential than the threshold for fast-twitch fiber contraction. The K-contracture results were confirmed by two microelectrode voltage-clamp experiments. Activation of fast twitch fibers required depolarizing pulses that were 15 to 20 mV greater than the pulses required to activate soleus fibers. When the time courses of K-contractures were compared it was evident that inactivation with prolonged depolarization was much faster in the fast-twitch muscles than in the soleus muscles. The results suggest that the voltage dependence and kinetics of the process coupling T-tubule depolarization with calcium release from the sarcoplasmic reticulum may depend on fiber type in mammalian skeletal muscle.  相似文献

A comparative study of the isoenzyme patterns of esterase and peroxidase and overall peroxidase activity in stamens of male-sterile (MS) lines of Pearson ms-35 and P ms-35aa and of the respective male-fertile (MF) tomato plants have been conducted. The study has been made at two stages of stamens development — tetrad and pollen. Higher activities of the esterase isoenzymes in the MF stamens than that of MS in both ontogeny stages have been found. The slow moving esterase isoenzymes both of the MF and the MS stamens are the major isoenzymes in the early stage and are connected with tapetum development while the fast moving esterase isoenzymes are connected with pollen formation in the later ontogeny stage. Overall peroxidase levels in the MS stamens were higher than those of MF. The peroxidase patterns of the MS lines are also characterized by the greater number of isoenzymes and also the presence of specific isoenzymes, the contrast between the MF and the MS stamens being more strongly expressed at the later stage of development. A strong similitude between esterase and peroxidase patterns behaviour in both MS lines has been found.  相似文献

The problem of work-related neck and upper limb disorders among computer users has been reported extensively in the literature, and commonly cited risk factors include static posture, speed and force of keyboard operation. The present study examined changes in median frequency (MF) of the neck–shoulder muscles in symptomatic and asymptomatic office workers when they were exposed to these three physical stressors.

A quasi-experimental Case–Control design was used to examine MF changes in two groups of female office workers when they were subjected to controlled doses of computer work involving prolonged static posture, increased typing speed and increased typing force. The MF of four major neck–shoulder muscles were examined bilaterally and compared between groups.

The MF changes over time-at-task did not clearly illustrate any muscle fatigue mechanism. However, Case Group consistently showed trends for higher MF than the Control Group, and this pattern was observed in response to all three physical stressors. The consistent group differences in MF suggest different muscle recruitment strategies between symptomatic and asymptomatic office workers. These results implied that symptomatic individuals had altered motor control, which may have important implications in understanding the etiology of work-related musculoskeletal disorders.  相似文献

The amount of the myelin fibers (MF) has been calculated in transversal serial sections of the nervous-vascular complexes of the splenic artery in newborns, in persons of mature (the 1st and 2d periods), elderly and old ages. In each age group 20 complexes have been studied. The total amount of the MF in persons of mature age (the 1st period) in 4.2-6 times greater (initial--terminal parts of the plexus) than in newborns, in the persons of old age it is 4-2.1 times less than in persons of mature age (in both cases P less than 0.001). Thin MF predominate, their relative contents in the aggregate of all classes of the MF in newborns are 92%, in mature persons--87%, in old persons--65%. Decreasing part of the thin MF together with increase of their absolute amount in the mature persons, comparing to the newborns, depends on higher rates in differentiation of middle and thick MF. Phenotyping peculiarities and stages of productive development, stabilization (the 1st-2d periods of the mature age), involutions of the nerve connections are defined in the splenic artery. The amount of the MF predominates in the nerves of the initial part of the plexus comparing to its terminal part. There is a direct correlative dependence between the amount of the MF in the plexus nerves and the size of the lumen in the splenic artery. Changes in the amount of the MF of different classes reflects qualitative differences of the splenic innervation relations at certain stages of the human postnatal development.  相似文献

This study was performed on rats with sustained (24 h) hyperadrenalinemia produced by sc implantation of retard adrenaline (A) tablets. Comparing with control, sham-operated animals, in A-treated rats duration of endurance exercise until exhaustion was shortened by approx. 40%. This was accompanied by: significant decreases of the pre-exercise muscle glycogen and creatine phosphate values in both "slow twitching" (soleus) and "fast twitching" (white portion of gastrocnemius) muscles, and of ATP content in soleus. Muscle lactate and pyruvate contents as well as blood glucose and FFA levels were elevated. After exercise muscle substrate and metabolite contents were similar in both groups in spite of the difference in exercise duration. It is concluded that prolonged hyperadrenalinemia diminishes the intramuscular energy substrate content, thereby reducing endurance capacity of rats.  相似文献

Summary 1. The formation of endplates outside the original endplate region of a muscle fibre was studied in slow and fast rat muscles. It was found that such new endplates are readily formed on the soleus muscle, whereas hardly at all in the fast extensor digitorum longus. Most new endplates appear to be morphologically normal within 2 months after nerve implantation.2. The time course of recovery of slow and fast cat muscles was followed after crushing the sciatic nerve. It was found that the slow soleus muscle recovers more rapidly than the fast flexor hallucis longus muscle.3. The endplates of reinnervated cat muscles are more complicated than those of the control muscles, but have nevertheless fewer nerve terminals per endplate. Reinnervated muscles are more sensitive to curare and it is suggested that this is due to a decrease in transmitter release, for it was found that less acetylcholine is released from reinnervated rat hemidiaphragms than from control ones.4. Motor and sensory reinnervation of spindles and tendon organs was studied. At the time when motor reinnervation is almost completed the sensory endings from spindles and tendon organs are highly abnormal. Thus sensory reinnervation proceeds much more slowly than motor.  相似文献

A number of single fibres were isolated by dissection of four bovine masseter (ma) muscles, three rectus abdominis (ra) muscles and eight sternomandibularis (sm) muscles. By histochemical criteria these muscles contain respectively, solely slow fibres (often called type I), predominantly fast fibres (type II), and a mixture of fast and slow. The fibres were analysed by conventional sodium dodecyl sulphate/polyacrylamide-gel electrophoresis and the gels stained with Coomassie Blue. Irrespective of the muscle, every fibre could be classed into one of two broad groups based on the mobility of proteins in the range 135000-170000 daltons. When zones containing myosin heavy chain were cut from the single-fibre gel tracks and 'mapped' [Cleveland, Fischer, Kirschner & Laemmli (1977) J. Biol. Chem. 252, 1102-1106] with Staphylococcus proteinase, it was found that one group always contained fast myosin heavy chain, whereas the second group always contained the slow form. Moreover, a relatively fast-migrating alpha-tropomyosin was associated with the fast myosin group and a slow-migrating form with the slow myosin group. All fibres also contained beta-tropomyosin; the coexistence of alpha- and beta-tropomyosin is at variance with evidence that alpha-tropomyosin is restricted to fast fibres [Dhoot & Perry (1979) Nature (London) 278, 714-718]. Fast fibres containing the expected fast light chains and troponins I and C fast were identified in the three ra muscles, but in only four sm muscles. In three other sm muscles, all the fast fibres contained two troponins I and an additional myosin light chain that was more typical of myosin light chain 1 slow. The remaining sm muscle contained a fast fibre type that was similar to the first type, except that its myosin light chain 1 was more typical of the slow polymorph. Troponin T was bimorphic in all fast fibres from a ra muscles and in at least some fast fibres from one sm muscle. Peptide 'mapping' revealed two forms of fast myosin heavy chain distributed among fast fibres. Each form was associated with certain other proteins. Slow myosin heavy chain was unvarying in three slow fibre types identified. Troponin I polymorphs were the principal indicator of slow fibre types. The myofibrillar polymorphs identified presumably contribute to contraction properties, but beyond cud chewing involving ma muscle, nothing is known of the conditions that gave rise to the variable fibre composites in sm and ra muscles.  相似文献

The problem on presence of muscle fibers (MF) types is an actual one, since it is a prerequisite to forecasting certain changes in MF under different conditions. The aim of the investigation has been to find out whether independent groups of MF exist or whether they react as a whole population to gradual changes in pH of the preincubational medium, when dynamic changes in m-ATPhase activity are studied. In serial transversal cryostate slices 10 mcm thick m-ATPhase activity has been defined in MF at preincubation of the slices in media having various pH. The experiment has had three series. In the I--the ventral denticulate muscle with universal biomechanical properties has been investigated; preincubation has been carried out in media having pH from 3.0 up to 11.0 with the intervals 0.5 pH. In the II--besides the muscle mentioned the fast brachial muscle and the force triceps muscle have been studied; preincubation has been performed only at pH 9.5, since in the I series two dissociated groups of MF have been distinguished. In the III series, at studying the same three muscles m-ATPhase activity has been detailed with their preincubation at pH from 3.8 up to 4.5 with the interval 0.1. The m-ATPase activity begins to manifest in various MF at pH of the preincubational medium within the limits of 3.8-4.2. A gradual increase in amount of MF that are revealing, as pH is gradually increasing, evidences most likely against independence of separate groups.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The activities of five enzymes have been studied quantitatively in denervated extensor digitorum longus, gastrocnemius and soleus muscles of 24-month-old rats. The results have been compared with those obtained from normal muscles of a similar age group of rats. Three weeks after denervation, the activity of hexokinase was increased in gastrocnemius and extensor digitorum longus. Phosphofructokinase, lactate dehydrogenase, malate dehydrogenase and 3-hydroxyacyl-CoA-dehydrogenase showed decreased activities. These results suggest that enzyme which represents glucose uptake increased its activity in fast muscles and that enzymes for anaerobic glycolysis, lactate fermentation, citric acid cycle and beta-oxidation had a decreased activity in slow and fast muscles.  相似文献
