An important fishery for the West African hemi-estuarine clam, Egeria radiata exists in the lower reaches of the Cross River (CR) system. The stock is heavily exploited but its productivity and potential yield have not been studied. This paper discusses the distribution and ecology of, and presents data on growth, abundance, mortality, biological production and potential yield of the CR stock of E. radiata based principally on length structured relative age. Growth was exponential and conformed to the von Bertalanffy's growth model (VBGM), with the parameters L = 9.3 cm, K = 0.36 and T o = –0.10 year. The instantaneous total mortality coefficient (Z) by the catch curve method was 0.82 yr–1; natural mortality coefficient (M) was estimated at 0.32 yr–1. Total mean biomass computed from data obtained in two month's grab sample surveys in the area of occurrence of E. radiata was 1923 kg ha–1 for total (i.e. flesh + shell) and 628 kg ha–1 for flesh (i.e. shell-free) production units. The production-to-mean-biomass (or P/B ratio, R) were 0.31 and 0.52 for total and shell-free production units respectively. The maximum sustainable yield (Y max) was estimated at 398.4 kg ha–1 for total and 112.2 kg ha–1 for shell-free production units. With an exploitation ratio (E) of 0.61 and an estimated present catch rate of 1970 tonnes(t) yr–1 total weight (or about 82% of Y max) the Egeria stock of the CR is currently being fished above the level of its optimum yield. Much of the fishing pressure is on the very young clams, thus lowering the age at first capture (T c) to 1 year. This is an unhealthy situation for a species with limited area of distribution and low (< 1.0) P/B ratio; it can lead to rapid extinction of the stock if for some reason there is a substantial reduction in annual recruitment. It is recommended that in order to conserve this valuable resource and keep it at a level of sustained optimum yield, there should be a regulation prohibiting the landing of young clams (< 6.0 cm shell length). Furthermore the fishermen should learn to cultivate the animal by transplanting the young clams to new beds upstream to give them time to grow larger before harvesting. This type of management has been practised in Ghana for a long time and has helped to conserve the Egeria stock of the Volta River. The need for a continuous monitoring of the effluents from the Nigerian Newsprint Manufacturing Company's factory located at the bank of the CR in the centre of the clam industry is stressed; such monitoring would ensure that dangerous toxic chemicals are not discharged into the river to damage the Egeria industry.  相似文献

A family of 10 competing, unstructured models has been developed to model cell growth, substrate consumption, and product formation of the pyruvate producing strain Escherichia coli YYC202 ldhA::Kan strain used in fed-batch processes. The strain is completely blocked in its ability to convert pyruvate into acetyl-CoA or acetate (using glucose as the carbon source) resulting in an acetate auxotrophy during growth in glucose minimal medium. Parameter estimation was carried out using data from fed-batch fermentation performed at constant glucose feed rates of qVG=10 mL h–1. Acetate was fed according to the previously developed feeding strategy. While the model identification was realized by least-square fit, the model discrimination was based on the model selection criterion (MSC). The validation of model parameters was performed applying data from two different fed-batch experiments with glucose feed rate qVG=20 and 30 mL h–1, respectively. Consequently, the most suitable model was identified that reflected the pyruvate and biomass curves adequately by considering a pyruvate inhibited growth (Jerusalimsky approach) and pyruvate inhibited product formation (described by modified Luedeking–Piret/Levenspiel term).List of symbols cA acetate concentration (g L–1) - cA,0 acetate concentration in the feed (g L–1) - cG glucose concentration (g L–1) - cG,0 glucose concentration in the feed (g L–1) - cP pyruvate concentration (g L–1) - cP,max critical pyruvate concentration above which reaction cannot proceed (g L–1) - cX biomass concentration (g L–1) - KI inhibition constant for pyruvate production (g L–1) - KIA inhibition constant for biomass growth on acetate (g L–1) - KP saturation constant for pyruvate production (g L–1) - KP inhibition constant of Jerusalimsky (g L–1) - KSA Monod growth constant for acetate (g L–1) - KSG Monod growth constant for glucose (g L–1) - mA maintenance coefficient for growth on acetate (g g–1 h–1) - mG maintenance coefficient for growth on glucose (g g–1 h–1) - n constant of extended Monod kinetics (Levenspiel) (–) - qV volumetric flow rate (L h–1) - qVA volumetric flow rate of acetate (L h–1) - qVG volumetric flow rate of glucose (L h–1) - rA specific rate of acetate consumption (g g–1 h–1) - rG specific rate of glucose consumption (g g–1 h–1) - rP specific rate of pyruvate production (g g–1 h–1) - rP,max maximum specific rate of pyruvate production (g g–1 h–1) - t time (h) - V reaction (broth) volume (L) - YP/G yield coefficient pyruvate from glucose (g g–1) - YX/A yield coefficient biomass from acetate (g g–1) - YX/A,max maximum yield coefficient biomass from acetate (g g–1) - YX/G yield coefficient biomass from glucose (g g–1) - YX/G,max maximum yield coefficient biomass from glucose (g g–1) - growth associated product formation coefficient (g g–1) - non-growth associated product formation coefficient (g g–1 h–1) - specific growth rate (h–1) - max maximum specific growth rate (h–1)  相似文献

Summary The growth of the yellow pigmented non-sporulating caldoactive bacterium Thermus aquaticus was investigated in different culture vessels and using differnt culture techniques. Each combination of these two variables led to very specific characteristic behaviour of the culture. A synthetic medium for a white cell type of T. aquaticus was optimized by means of pulse and medium-shift techniques. The main kinetic parameters of the organism have been determined to be =1.62h–1 and Y (glucose)=0.4 g g–1 at T=68 °C and pH=7.3. In complex medium only a mixed population of white and yellow cells could be cultivated. The cell yield was shown to be very low (Y=0.02 g g–1) due to incomplete substrate utilisation, but a very high maximal specific growth rate was determined ( max=3.5h–1) at 75 °C and pH=7.3. The maintenance coefficient for oxygen uptake was approximately Mo=16 mMol g–1 h–1. A discussion of the problems arising in the cultivation of thermophilic microorganisms with respect to their physiology and stability is given.  相似文献

Reproduction and growth of the vermilion snapper, Rhomboplites aurorubens, were studied in Trinidad and Tobago. The smallest individual caught measured 145mm total length (TL) and all fish appeared to be mature. It was not possible to precisely determine size at first maturity due to the use of macroscopic techniques. The smallest spent male and female measured 181 and 211mm TL respectively, suggesting a size at first maturity below these sizes. Spawning occurred throughout the year, with a period of peak spawning from about June to November in the rainy season when river runoff increased. Sagittal otolith sections were used for age determination and the opaque ring, which was counted as the annulus, was deposited from January to May in the dry season. A total of 11 age groups between the ages of 2–12 years (155–505mm total length) were found. The von Bertalanffy growth parameters were: L=532mm, K=0.13y–1, and t0=–0.17, where L is the asymptotic length, K is the growth coefficient and t0 is the theoretical age at zero length. The relationship between weight (WT) and length (TL) was WT=3.43×10–5 TL2.82. Vermilion snapper in this study area appears to grow slower and attain a smaller asymptotic length, but has a longer lifespan than found in populations in higher latitudes. This may be attributed to different levels of exploitation, which may be higher in the latter areas.  相似文献

Dissolved free amino acid (DFAA) concentration and composition and dissolved organic carbon (DOC) concentration were measured over 16 months at three depths in hypertrophic Hartbeespoort Dam, South Africa and in its two perenially inflowing rivers. The range of DFAA concentrations in the reservoir and both rivers were similar with dominant DFAA consisting of serine, glycine, alanine and ornithine in all three systems. The range of DOC concentrations in the rivers was 1.5–11.1 mg l–1, the major river (Crocodile) having about twice the DOC concentration of the Magalies River. The DFAA/DOC ratios ranged between 0.02–1.1% in the Crocodile River and 0.13–3.7% in the Magalies River. DFAA and DOC concentrations were positively correlated to the Magalies River flow, but for the Crocodile River, which received domestic and industrial effluents, DOC was inversely correlated to flow. The source of DFAA in both rivers was mainly terrestrial, in contrast to the main DOC source in the Crocodile River which was the effluents. The DFAA load of the Crocodile River ranged between 0.22 and 208 kg C d–1.DOC (5.0–24.8mg l–1) in Hartbeespoort Dam generally decreased with depth but DFAA (15–4800 nmol l–1) concentration showed no clear trend. The DFAA/DOC ratios varied between 0.02 and 2.9%. DFAA concentrations were correlated (r = 0.3, n = 30, p = 0.04) with bacterial numbers at 0 and 10 m only while no significant correlations were found with bacterial production, chlorophyll a concentration and phytoplankton primary and EDOC (extracellular DOC) production at any depth. The rate of bacterial utilization of DFAA was low compared with data from other lakes. Diurnal phytoplankton production of DFAA in the euphotic zone of the whole lake was calculated to vary between 268 and 30 780 t C d–1 indicating autochthonous DFAA sources were dominant to allochthonous DFAA sources. The autochthonous production of DFAA was > 2 × gross bacterial production of the euphotic zone indicating that although DFAA concentrations were frequently < 10 g C l–1, the rate of DFAA production exceeded bacterial requirements.  相似文献

Effects of field dosages ofselected insecticides to Cotesiaplutellae (Kurdjumov) (Hymenoptera: Braconidae), a larval endoparasitoidof Plutella xylostella L. (Lepidoptera: Plutellidae), wereinvestigated under laboratory conditions.Emergence of adult C. plutellae frominsecticide-treated pupae was not significantlydifferent from the control treatment. Contacttoxicity to C. plutellae adults variedgreatly among the insecticides in a paperresidue contact bioassay. Threeazadirachtin-based insecticides, Agroneem(4.8 mg a.i.liter–1), Neemix (20 mga.i.liter–1) and Ecozin (20 mgai.liter–1) caused 11.1, 16.7 and 5.6%adult mortality, respectively. Of fourcommercial Bacillus thuringiensis (Bt)insecticides examined (all at 1.2 mga.i.liter–1), Crymax and Xentari had noeffect on adult parasitoids, whereas Mattchcaused 5.6% mortality, and Dipel caused 11.1%mortality. Indoxacarb (53 mg a.i.liter–1),-cyhalothrin (28 mg a.i.liter–1) andspinosad (53 mg a.i.liter–1) caused 100,88.5 and 50% adult mortalities, respectively.Low adult mortality (0–5.6%) was recorded fromingestion of azadirachtin-based, Btinsecticides and indoxacarb, compared with100% adult mortality in treatments of spinosador -cyhalothrin. Compared with the watercontrol, ingestion of azadirachtin-basedinsecticides significantly reduced parasitismby 50–57%, and Bt insecticides by 8–25%.However, ingestion of these insecticides didnot affect longevity of male and femaleparasitoid adults with one exception; femalelongevity was significantly reduced in theindoxacarb treatment. Insecticide residuescaused considerable mortality of C.plutellae adults, 39 and 44% mortality causedby 10 d old indoxacarb and -cyhalothrin,respectively, and 24 and 0% mortality causedby 7 and 10 d old residues of spinosad, respectively.  相似文献

During four cruises in continental shelf waters of the northern Gulf of Mexico in the winters of 1981–83, we performed quantitative studies on the grazing of the copepods Acartia tonsa, Centropages velificatus, and Eucalanus pileatus, on phytoplankton using natural particulate assemblages as food. Stations were in, or adjacent to the plume of the Mississippi River, thereby providing wide spectra of phytoplankton and suspended riverine particulate concentrations. Phytoplankton cell volume was converted to carbon, and this, coupled with carbon content measurements of these three copepod species, allowed comparisons of daily ingestion effort even though the copepods were of different sizes. Data were expressed in the same units (% of copepod body carbon ingested copepod –1 d–1) for each species. Over similar ranges of phytoplankton carbon concentrations (0.21–92.06 gCl–1), Acartia tonsa had higher carbon-specific ingestion rates (x = 22.31%, range = 0.08–152.37%) than C. velificatus (x = 2.8%, range = 0.00–31.09 %) or E. pileatus (x = 1.27%, range = 0.10–2.80%). Carbon-specific ingestion rates increased with increasing phytoplankton carbon concentration for A. tonsa (R2 = 0.81) and there was no evidence of saturated feeding on the carbon concentrations offered. A similar, but weaker trend was evident for E. pileatus (R2 = 0.71), but not C. velificatus (R2 = 0.49). Over a wide range of suspended particulate concentrations (10.6–95.2 mg l–1), there was no systematic effect of particulates on carbon-specific ingestion rate for any of the three copepod species. However, A. tonsa appeared more adept at grazing in highly turbid water than C. velificatus or E. pileatus.  相似文献

The population dynamics of trout (Salmo trutta L.) were studied in two high-altitude streams flowing westwards into the River Eden in northern England. The results from one (Knock Ore Gill), which is accessible to spawners from downstream, were compared with published data from an isolated population in a nearby stream which flows eastward into the River Tees. The Knock Ore Gill trout had a population density of 0.4–0.9 fish m–2, a biomass of 12.4 g m–2, annual production of 12–14 g m–2 year–1 and an instantaneous mortality rate of 0.97 year–1. Comparable values from the Tees tributary were 0.2, 3.3, 2.3 and 0.66 respectively. The Knock Ore Gill population also contrasted with that of the Tees tributary in having reliable recruitment from year to year. These differences were related to differences in the chemistry and spate characteristics of the two streams and to the fact that immigrants from downstream were responsible for over 30% of the annual oviposition in Knock Ore Gill.  相似文献

Sawfish are a group of endangered elasmobranchs that were common in tropical inshore, estuarine and freshwaters. The demography of two species of sawfish that occur in the western Atlantic – Pristis pectinata and P. perotteti – was investigated using age-structured life tables. Life history parameters for use in the life tables were obtained from published data. Five scenarios were tested for P. pectinata to incorporate uncertainties about life history data. Values of intrinsic rate of increase ranged from 0.08 to 0.13yr–1, and population doubling times from 5.4 to 8.5 yrs. Eight scenarios were tested for P. perotteti. The most likely range for the intrinsic rate of increase was 0.05–0.07yr–1, with population doubling times of 10.3–13.5yrs. Four scenarios investigating the sensitivity to methods of estimating natural mortality produced similar results. The demographic results were sensitive to changes in reproductive periodicity and natural mortality. The results indicate that if effective conservation plans can be implemented for sawfish and sawfish habitats, recovery to levels where there is little risk of extinction will take at least several decades.  相似文献

Nitrogen fixation was measured in four subarctic streams substantially modified by beaver (Castor canadensis) in Quebec. Acetylene-ethylene (C2H2 C2H4) reduction techniques were used during the 1982 ice-free period (May–October) to estimate nitrogen fixation by microorganisms colonizing wood and sediment. Mean seasonal fixation rates were low and patchy, ranging from zero to 2.3 × 10–3 µmol C2H4 · cm–2 · h–1 for wood, and from zero to 7.0 × 10–3 µmol C2H4 · g AFDM–1 · h–1 for sediment; 77% of all wood and 63% of all sediment measurements showed no C2H2 reduction. Nonparametric statistical tests were unable to show a significant difference (p > 0.05) in C2H2 reduction rates between or within sites for wood species or by sediment depth.Nitrogen contributed by microorganisms colonizing wood in riffles of beaver influenced watersheds was small (e.g., 0.207 g N · m–2 · y–1) but greater than that for wood in beaver ponds (e.g., 0.008 g N · m–2 · y–1) or for streams without beaver (e.g., 0.003 g N · m–2 · y–1). Although mass specific nitrogen fixation rates did not change significantly as beaver transform riffles into ponds, the nitrogen fixed by organisms colonizing sediment in pond areas (e.g., 5.1 g N · m–2 · y–1) was greater than that in riffles (e.g., 0.42 g N · m–2 · y–1). The annual nitrogen contribution is proportional to the amount of sediment available for microbial colonization. We estimate that total nitrogen accumulation in sediment, per unit area, is enhanced 9 to 44 fold by beaver damming a section of stream.  相似文献

Macrozoobenthos of the ultraoligotrophic Lake 95 (61°N, 46°W, 8 ha, zmax=18 m, ) is composed of about 14 taxa dominated by 12 Chironomidae species. Abundance, life cycle, biomass and production were estimated for the six dominant taxa. Abundance declined fromca. 4150 at 2.5 m depth toca. 1400 ind m–2 at 16 m depth and averagedca. 3200 ind m–2 on a lakewide basis. By numbers,Heterotrissocladius changi andH. oliveri dominated the average fauna.H. changi was common at the 2.5 m and 5 m depth stations, whereasH. oliveri dominated from 5 m depth downwards. Chironomids showed mainly a 1-yr life cycle, but apparently bothHeterotrissocladius species had two contemporary cohorts with emergence in midsummer and late autumn/early spring, respectively. Average annual ratio was 4.2 and 4.6 forH. oliveri andH. changi, respectively. Annual production varied from 0.3 g ash-free dry weight (AFDW) m–2 y–1 at 16 m depth to 1.6 g AFDW m–2 y–1 at 2.5 m depthH. changi contributed 45%, fiveMicropsectra spp. 17% andH. oliveri 15% to total average production, which on a lakewide basis wasca. 1.1 g AFDW or 25 kJ m–2 y–1. Lake 95 thus belongs at the very low end of measured lake zoobenthic productions, which range from 10 kJ m–2 y–1 in Arctic lakes toca. 1600 kJ m–2 y–1 in highly eutrophic shallow lakes.  相似文献

Macroscopic instantaneous and time-dependent currents have been measured in the vacuolar membrane of Beta vulgaris using a patch clamp configuration analogous to whole cell mode. At low cytosolic Ca2+ and in the absence of Mg2+, only an instantaneous current was observed. This current is carried predominantly by cations (PKPCl 71, pnapcl 41 and arginine is also conducted). The instantaneous current can be activated by ATP4– (e.g., ATP-activated mean K+ current density was –20 mA.m–2 at a membrane voltage of –20 mV) and by increasing cytosolic pH and Mg2+ (raising Mg2+ from 0 to 0.4 mm induced a mean current density increase of –7 mA.m–2 at –20 mV). Such current can be activated by simultaneous addition of putative in vivo concentrations of ATP4–/MgATP/Mg free 2+ (in the presence of bafilomycin to inhibit the vacuolar ATPase) and further modulated by cytosolic pH. With vacuolar K+ concentration greater than that of the cytosol, activation of the instantaneous current would mediate vacuolar K+ release over the range of physiological membrane voltage. It is argued that the ATP4–-activated current, in addition to acting as a K+ mobilization pathway, could provide a counter-ion (shunt) conductance, allowing the two electrogenic H+ pumps which reside in the vacuolar membrane to acidify the vacuolar lumen.A separate time-dependent current, which was not observed at low Ca2+ concentrations (less than 500 nm) could also be elicited by addition of Mg2+ at the cytoplasmic membrane face. This current was stimulated by increasing cytoplasmic pH.The authors are grateful to the BBSRC for financial support (Grant PG87/529) and to the Royal Society (University Research Fellowship to J.M.D.). Lactobacillus helveticus grown in milk with pH control at 6.2 had a slower growth rate (=0.27 h–1) and produced less exopolysaccharide (49 mg l–1) but increased lactic acid production (425 mM) compared to cultures without pH control (=0.5 h–1, 380 mg exopolysaccharide l–1, and 210 mM lactate), respectively. Both cultures displayed a mixed-acid fermentation with formation of acetate, which is linked not only to citrate metabolism, but also to alternative pathways from pyruvate.  相似文献

Field research was conducted on four Atlantic Coastal Plain soils in the United States to evaluate response of corn (Zea mays L.) plants to Mn application. The soils under study were classified as either Aeric or Typic Ochraquults. Manganese application increased corn grain yields by an average of 1195 kg ha–1 on the four soils. The average grain yields on the soils were 7955 kg ha–1 for the control and 9150 kg ha–1 for the +Mn treatment. A Mitscherlich plant growth model was used to establish relationships between percent maximum grain yield and Mn concentration in the ear leaf at early silk (r=0.87, =0.01) and in the mature grain (r=0.58, =0.01). Based on 90% of maximum yield as the definition of the critical deficiency level, the critical Mn deficiency levels calculated with parameters from the Mitscherlich model were 10.6 mg kg–1 in the ear leaf and 4.9 mg kg–1 in the grain.  相似文献

Ulva rigida was cultivated in 7501 tanks at different densities with direct and continuous inflow (at 2, 4, 8 and 12 volumes d–1) of the effluents from a commercial marine fishpond (40 metric tonnes, Tm, of Sparus aurata, water exchange rate of 16 m3 Tm–1) in order to assess the maximum and optimum dissolved inorganic nitrogen (DIN) uptake rate and the annual stability of the Ulva tank biofiltering system. Maximum yields (40 g DW m–2 d–1) were obtained at a density of 2.5 g FW 1–1 and at a DIN inflow rate of 1.7 g DIN m–2 d–1. Maximum DIN uptake rates were obtained during summer (2.2 g DIN M–2 d–1), and minimum in winter (1.1 g DIN m–2 d–1) with a yearly average DIN uptake rate of 1.77 g DIN m–2 d–1 At yearly average DIN removal efficiency (2.0 g DIN m–2 d–1, if winter period is excluded), 153 m2 of Ulva tank surface would be needed to recover 100% of the DIN produced by 1 Tm of fish.Abbreviations DIN= dissolved inorganic nitrogen (NH inf4 sup+ + NO inf3 sup– + NO inf2 sup– ); - FW= fresh weight; - DW= dry weight; - PFD= photon flux density; - V= DIN uptake rate  相似文献

A total of 90, weekly exploited populations of the harpacticoid copepodTisbe holothuriae Humes were exposed to 148, 222, 333, 500, 750 or 1125µg Cd++ l–1, combined with exploitation rates of 10, 30, 50, 70 or 90% under conditions of surplus food supply at 22° C and 30 S. All populations exposed to concentrations down to 500µg Cd++ l–1 and 3 populations (out of 15) exposed to 333µg l–1 became extinct within the experimental period of 30 weeks. Survival time significantly depended on concentration. A recovery phase from an initially high mortality preceded eventual population extinction after adding 500µg Cd++ l–1. In the initial phase, higher nauplii mortality prevailed. During these experiments on acute intoxication, no relationship could be established between survival and exploitation rate. However, experiments on the effects of stepwise increases in Cd concentration (results not yet published) produced evidence of such a relationship. In spite of increased mortality, no significant numbers of dead copepods were detected in weekly samples because of their rapid decomposition and cannibalism, which depends on the amount of food available. Sampling regimes of 5 times per week yielded significant numbers of dead individuals.  相似文献

Summary Paired toad urinary bladders were prepared without or with an osmotic gradient (175 mosm) across them, stimulated for 2.5 (n=6), 5 (n=6), 30 (n=6) or 60 (n=6) min with ADH (20 mU/ml), and studied by freeze-fracture electron microscopy. Water permeability at these times was assessed in additional bladders (n=6 for each case) after tissue fixation according to the technique of Eggena. After both 60 and 30 min of ADH stimulation, the presence of a gradient compared with the absence of one was associated with fewer aggregates (242±35vs. 382±14 ×235 m–2 at 60 min,P<0.01; 279±36vs. 470±51 ×235 m–2 at 30 min,P<0.01) and lower water permeability (8.4±1.1vs. 18.8±1.8g×min–1×cm–1 ×mosm –1 at60min,P<0.005; 9.2±1.0vs. 22.0±2.1 g ×min–1×cm–2×mosm –1 at 30 min,P<0.001). In addition, with a gradient both maximum water permeability and maximum aggregate frequency were reached nearly together; a similar correspondence occurred without a gradient. We conclude that in the presence of an osmotic gradient both the ADH-associated aggregates and the water permeability response to ADH are prevented from reaching the higher levels observed in bladders not exposed to a gradient.  相似文献

A fermentation medium based on millet (Pennisetum typhoides) flour hydrolysate and a four-phase feeding strategy for fed-batch production of baker's yeast,Saccharomyces cerevisiae, are presented. Millet flour was prepared by dry-milling and sieving of whole grain. A 25% (w/v) flour mash was liquefied with a thermostable 1,4--d-glucanohydrolase (EC 3.2.1.1) in the presence of 100 ppm Ca2+, at 80°C, pH 6.1–6.3, for 1 h. The liquefied mash was saccharified with 1,4--d-glucan glucohydrolase (EC 3.2.1.3) at 55°C, pH 5.5, for 2 h. An average of 75% of the flour was hydrolysed and about 82% of the hydrolysate was glucose. The feeding profile, which was based on a model with desired specific growth rate range of 0.18–0.23 h–1, biomass yield coefficient of 0.5 g g–1 and feed substrate concentration of 200 g L–1, was implemented manually using the millet flour hydrolysate in test experiments and glucose feed in control experiments. The fermentation off-gas was analyzed on-line by mass spectrometry for the calculation of carbon dioxide production rate, oxygen up-take rate and the respiratory quotient. Off-line determination of biomass, ethanol and glucose were done, respectively, by dry weight, gas chromatography and spectrophotometry. Cell mass concentrations of 49.9–51.9 g L–1 were achieved in all experiments within 27 h of which the last 15 h were in the fedbatch mode. The average biomass yields for the millet flour and glucose media were 0.48 and 0.49 g g–1, respectively. No significant differences were observed between the dough-leavening activities of the products of the test and the control media and a commercial preparation of instant active dry yeast. Millet flour hydrolysate was established to be a satisfactory low cost replacement for glucose in the production of baking quality yeast.Nomenclature C ox Dissolved oxygen concentration (mg L–1) - CPR Carbon dioxide production rate (mmol h–1) - C s0 Glucose concentration in the feed (g L–1) - C s Substrate concentration in the fermenter (g L–1) - C s.crit Critical substrate concentration (g L–1) - E Ethanol concentration (g L–1) - F s Substrate flow rate (g h–1) - i Sample number (–) - K e Constant in Equation 6 (g L–1) - K o Constant in Equation 7 (mg L–1) - K s Constant in Equation 5 (g L–1) - m Specific maintenance term (h–1) - OUR Oxygen up-take rate (mmol h–1) - q ox Specific oxygen up-take rate (h–1) - q ox.max Maximum specific oxygen up-take rate (h–1) - q p Specific product formation rate (h–1) - q s Specific substrate up-take rate (g g–1 h–1) - q s.max Maximum specific substrate up-take rate (g g–1 h–1) - RQ Respiratory quotient (–) - S Total substrate in the fermenter at timet (g) - S 0 Substrate mass fraction in the feed (g g–1) - t Fermentation time (h) - V Instantaneous volume of the broth in the fermenter (L) - V 0 Starting volume in the fermenter (L) - V si Volume of samplei (L) - x Biomass concentration in the fermenter (g L–1) - X 0 Total amount of initial biomass (g) - X t Total amount of biomass at timet (g) - Y p/s Product yield coefficient on substrate (–) - Y x/e Biomass yield coefficient on ethanol (–) - Y x/s Biomass yield coefficient on substrate (–) Greek letters Moles of carbon per mole of yeast (–) - Moles of hydrogen atom per mole of yeast (–) - Moles of oxygen atom per mole of yeast (–) - Moles of nitrogen atom per mole of yeast (–) - Specific growth rate (h–1) - crit Critical specific growth rate (h–1) - E Specific ethanol up-take rate (h–1) - max.E Maximum specific ethanol up-take rate (h–1)  相似文献

A field experiment in western Kenya assessed whether the agroforestry species Tithonia diversifolia (Hemsley) A. Gray, Tephrosia vogelii Hook f., Crotalaria grahamiana Wight & Arn. and Sesbania sesban (L) Merill. had access to forms of soil P unavailable to maize, and the consequences of this for sustainable management of biomass transfer. The species were grown in rows at high planting density to ensure the soil under rows was thoroughly permeated by roots. Soil samples taken from beneath rows were compared to controls, which included a bulk soil monolith enclosed by iron sheets within the tithonia plot, continuous maize, and bare fallow plots. Three separate plant biomass samples and soil samples were taken at 6-month intervals, over a period of 18 months. The agroforestry species produced mainly leaf biomass in the first 6 months but stem growth dominated thereafter. Consequently, litterfall was greatest early in the experiment (0–6 months) and declined with continued growth. Soil pH increased by up to 1 unit (from pH 4.85) and available P increased by up to 38% (1 g P g–1) in agroforestry plots where biomass was conserved on the field. In contrast, in plots where biomass was removed, P availability decreased by up to 15%. Coincident with the declines in litterfall, pH decreased by up to 0.26 pH units, plant available P decreased by between 0.27 and 0.72 g g–1 and Po concentration decreased by between 8 and 35 g g–1 in the agroforestry plots. Declines in Po were related to phosphatase activity (R2=0.65, P<0.05), which was greater under agroforestry species (0.40–0.50 nmol MUB s–1 g–1) than maize (0.28 nmol MUB s–1 g–1) or the bare fallow (0.25 nmol MUB s–1 g–1). Management of tithonia for biomass transfer, decreased available soil P by 0.70 g g–1 and Po by 22.82 g g–1. In this study, tithonia acquired Po that was unavailable to maize. However, it is apparent that continuous cutting and removal of biomass would lead to rapid depletion of P stored in organic forms.  相似文献
