Curd Initiation in the Cauliflower: I. JUVENILITY   总被引:1,自引:0,他引:1  
Hand, D. J. and Atherton, J. G. 1987. Curd initiation in thecauliflower. I. Juvenility.—J. exp. Bot. 38: 2050–2058. Four cauliflower (Brassica oleracea var. botrytis L.) cultivarswere screened for differences in juvenility, measured as thephase of insensitivity to vernalization. Juvenility persisteduntil the initiation of a critical number of leaves which sharplydefined transition to the sensitive, mature form at 13 to 15leaves in cv. Perfection and at 17 to 19 leaves in cv. WhiteFox. Preliminary investigation showed transition in cv. AlphaCliro to occur between 9 and 18 leaves initiated and in cv.Dole after 19 leaves. Leaf number was a stable marker of theend of juvenility in plants grown under different light conditions,whereas time, leaf area and leaf dry weight were not. Leaf numberwas linearly related to log whole shoot dry weight. The rateof leaf initiation in plants of cv. Perfection growing duringthe juvenile phase was approximately one third that of plantsin the mature phase, when measured on a thermal time base. Chilling mature, vegetative plants at 5 °C for 28 d advancedcurd initiation by up to 35 leaves in cv. Perfection; 27 leavesin cv. White Fox; 27 leaves in cv. Alpha Cliro and 21 leavesin cv. The beet western yellows virus (BWYV) was identified in sugar beet plants with leaf yellowing symptoms. When transmitted toSinapis alba L. the virus isolate caused severe symptoms of yellowing and violetting of the interveinal leaf tissue of this plant. By aphidsMyzus persicae (Sulz.) the virus isolate was transmitted toLactuca sativa L.,Raphanus sativus L. var.radicula Pers.,Baphanus sativus L. ssp.sativus L. ap., and toBrassica oleracea L. var.gemmifera DC. InLactuca sativa plants the virus induces a yellowing along with thickenning and brittleness of leaves and with mild dwarfing of the plants. InBaphanus sativus var.radicula andBaphanus sativus ssp.sativus plants it brings about a yellowing of the leaf margins with a change in consistency as was the case in lettuce, and inBrassica oleracea var.gemmifera it causes violet spots on the lower leaf sides. The transmission was proved in repeated experiments by a backtransmission to beet andSinapis alba and further transmission from beet toSinapis alba. The transmission of the virus isolate toVicia faba L.,Datura stramonium L., andPetunia hybrida hort. was unsuccessful. In the course of transmissions the isolate properties did not change. In its host range the virus resembles the Duffus’ strain 3 BWYV, isolated from beet in the U.S.A. This is the first characteristic of an Europian BWYV isolate, as obtained from naturally infected beet plants.  相似文献

Broccoli (Brassica oleracea var. italica cv Premium Crop) plantswere germinated in soil, transferred to vermiculite three weekslater and grown in the glasshouse, then either supplied continuouslywith boron levels ranging from 0.0 (deficient) to 12.5 (toxic)mg l–1 of nutrient solution or transferred from 2.5 to0.0 mg B l–1 at the initiation of inflorescence development.At commercial maturity the concentrations of various inorganicand organic solutes in phloem exudates and xylem saps, as wellas plant characteristics and elemental composition of the variousplant parts, were determined. Under deficient B levels leaf midrib and stem corkiness wereevident, together with signs of stem pith breakdown, symptomswhich resemble the initiation of the hollow stem disorder. Thexylem sap B concentration declined by about 50 % when B wasnot supplied or was removed after a period of adequate supply;the phloem concentration was unaffected. Also, the decreasingB concentration gradients from mature transpiring tissues toyoung developing sinks disappeared. Therefore, it is concludedthat when B is deficient, it is retranslocated from source leavesin the phloem stream supplying the developing leaves and inflorescence.The data also suggested that at toxic levels B undergoes extensivelateral transfer, probably from xylem to xylem, thereby enhancingthe B concentration of developing sinks. The B regime influenced dry-matter partitioning, retranslocationof some elements, and the synthesis and distribution of aminoacids and sugars, reflecting the general nature of B involvementin plant processes. Evidence for Repair Processes in the Invigoration of Seeds by Hydration   总被引:1,自引:0,他引:1  
Seeds with low vigour as a result of ageing showed a large improvementin seed quality following a 2 h soak in water, reflected inan increased rate of germination, higher emergence in soil,and the retention of high germination after 24 h rapid ageingin a controlled deterioration vigour test. Similar, but smallerimprovements were seen after a priming treatment using polyethyleneglycol. Seeds dried to their initial moisture content aftertreatment retained these beneficial effects. The treatmentshad little effect on high vigour seed. These findings suggestthat seed invigoration by hydration treatments results fromthe repair of previously sustained deterioration. Ammonium Nutrition Enhances Chlorophyll and Glaucousness in Kohlrabi   总被引:2,自引:0,他引:2  
Kohlrabi (Brassica oleracea var.gongylodes) plants were grownin the greenhouse under autumn conditions and fertilized eitherwith pellets containing nitrogen as 40% ammonium sulphate and60% urea or with nutrient solution containing nitrogen predominantlyas nitrate. Plants given nitrogen as ammonium ions developedglaucous leaves compared to those supplied with nitrate whichformed glossy leaves. Ammonium-induced glaucousness was theresult of a two-fold increase in the amount of epicuticularwax and a markedly altered fine structure. Leaves from ammoniumfertilized kohlrabi plants also showed a 21% increase in chlorophyllcontent together with a reduction in the chlorophyll a:b ratioand decreased ground state fluorescence compared to plants suppliedwith nitrate. Photosynthesis and stomatal transpiration wereunaffected by the form of supplied nitrogen. Root segments taken from aseptically-grown seedlings of Brassicaoleracea var. italica cv. Green Comet were used in an investigationof factors affecting in vitro regeneration. Shoot regenerationwas found to increase with seedling age and to be highest inroot segments adjacent to the hypocotyl and lowest in segmentsadjacent to the root tip. In a comparison of a range of mediaand agar concentrations shoot formation was favoured by complexmedia containing reduced nitrogen and was higher on gelled mediathan in liquid medium. The effects of various cytokinins andauxins were investigated; KN was the best cytokinin and IAAand Picloram the best auxins for shoot induction. Root segmentsfrom six other Brassica cultivars were grown on the medium devisedfor Green Comet; shoots were regenerated from two B. oleraceacultivars and two B. napus cultivars, but not from the B. campestriscultivars tested. Water loss was studied in regenerated plantlets of Brassicaoleracea var. botrytis cv. Currawong derived through apicalmeristem culture. Hardening of plantlets was eliminated by asingle application of a polyvinyl resin (S600) sprayed immediatelyafter transplanting. Plantlets sprayed with S600 had highercuticular resistances than unsprayed plantlets; this treatmenthad no effect on stomatal resistance. Leaves formed during theculture period showed very little wax formation and using markedleaves it was found that only reduced levels of wax formed onthese leaves even after transplanting. New leaves formed aftertransplanting, showed typical wax formation compared to seedgrown plants. Abscisic acid (ABA) at 10–4 M applied as a leaf sprayto transplants did not cause a substantial increase in stomatalresistance in leaves which had been initiated during the cultureperiod. Leaves of seed-grown plants as well as leaves of plantletsformed after transplanting did respond to a leaf spray of ABAat 10–4 M by a large increase in stomatal resistance. Relative concentrations of K, Na, Ca, P, S and Mg in guard cellswere calculated for each leaf type by X-ray micro-probe analysis.K/Na values decreased in the order: seedling > leaves formedafter transplanting > leaves intiated during culture. A highpositive correlation was also found between K/Na and K/P forthe three leaf types. K:Mg and K:Ca ratios for leaves formedduring culture were low in comparison to the values obtainedfor leaves formed after transplanting and seedlings for whichthe values were similar. Isolated root cultures which could be maintained over severalmonths by serial subculture were established from Brassica oleraceavar. italica cv. Green Comet F1. A modified White's medium wasfound to be the best of several salt compositions tested. Theeffects on isolated root growth of the following were also examined;nutritional components, culture vessel type and closure, pHof the medium and auxin type and concentration. Using a mediumdevised for Green Comet, root cultures were established fromsix other B. oleracea, B. napus and B. campestris cultivars. It was possible to regenerate shoots from segments of culturedroots by incubation on agar-solidified media containing cytokininand auxin. The effects on regeneration of various auxins andcytokinins were investigated; the combination of Picloram withKN gave the highest frequency of shoot formation. It was demonstratedthat isolated roots retained their regenerative ability overa period of 5 months in culture. Electrical impedance spectra (100 Hz–800 kHz) were measuredin leaves of Peperomia obtusifolia L. (a succulent) and Brassicaoleracea L. (cabbage). By measuring impedances at three or moreinter-electrode distances in a single leaf, electrode impedanceand specific tissue impedance were separated. Analysis of impedance data from B. oleracea leaves in relationto an equivalent circuit model showed that leaf developmentwas accompanied by increases in extracellular resistance, cytoplasmicresistance and vacuole interior resistance, together with decreasesin plasma membrane capacitance and tonoplast capacitance. AfterB. oleracea leaves were subjected to a –6 °C freeze-thawstress, extracellular resistance, cytoplasmic resistance andvacuole interior resistance decreased, but plasma membrane capacitanceand tonoplast capacitance did not change. These results indicatethat useful measurements of leaf parameters can be obtainedby this technique. Examination of the electrode impedance spectrum showed thatelectrode insertion produced a damaged collar, 0·4–0·5mm wide, around the electrode. The Conductance model is a simple mechanistic model used topredict the growth of species in monoculture or mixtures fromparameter values derived from plants grown in isolation. Incontrast to many mechanistic models that require extensive parameterization,the Conductance model is able to capture the growth of a broadrange of species using a few simplified assumptions regardingplant growth and easily derived species-specific parameter values.We examine the assumptions within the Conductance model thattotal leaf area per plant is proportional to total plant weight,and that an isolated plant has a projected crown zone area thatis proportional to the 2/3 power of its weight. Power ratherthan linear relations were found between weight and leaf areafor Brassica oleracea, Daucus carota, Matricaria inodora, Solanumnigrum,Stellaria media , Trifolium repens and Veronica persica.For all seven species, the value of the power was less thanunity. All species also exhibited a power relation between crownzone area and weight, with the slope of this relation beingless than 2/3 for B. oleracea, D. carota and S. media. Althoughmorphology type accounted for some of the variation in the parametervalues relating to light interception, there were considerabledifferences between species within upright or prostrate foliagespecies groups. The Conductance model was used to predict yieldsof B. oleracea, S. nigrum and V. persica grown in both monocultureand binary weed-crop mixtures over a range of temporal and spatialscales. After calibrating the model to non-competing plants,the model was used to predict growth of the weed and crop speciesin contrasting densities and stand types. In some crop-weedcombinations, predicted crop and weed weights were within 17%of observed values, with no systematic deviations. Disease symptoms of speckled leaf blotch (Mycosphaerella graminicola)were first observed in early August, 86 d after sowing a winterwheat field trial in New Zealand. Disease hastened senescenceof all leaves, delayed the expansion of new leaves during springand reduced the maximum size of some later-formed leaves. Theabsolute green leaf area was reduced by disease through to fullsenescence of leaves, which occurred one week earlier than inhealthy plants. Disease indices calculated from the data onpercentage diseased area alone indicated a decline in diseaseduring rapid growth of the plants in spring as the temperatureincreased. However, this apparent decline was generated by themethod of calculation and was not evident when effects on leafarea were also considered. A water-soluble Chl a/b-protein (CP673) was isolated and purifiedfrom Brussels sprouts (Brassica oleracea L. var. gemmifera DC).The protein had a molecular mass of 78 kDa and an isoelectricpoint of 4.7, consisted of three or four subunits of 22 kDaand was extremely heat-stable. Although CP673 contained aboutone Chl a per protein, the blue and red absorption bands ofChl a that consisted of three or four Chl a forms with differentabsorption maxima suggested that there are several differentmodes or sites of binding for Chl a. Chl a/b ratio of largerthan 10 also indicated that Chl b is present only in a smallfraction of CP673. The heterogeneity of CP673 in terms of compositionand binding of Chl suggests that Chl is not an intrinsic componentof the Chl-protein. Homology search showed that the N-terminalamino acid sequence of CP673 is highly homologous with thatof a 22 kDa protein that accumulates in water-stressed leavesof two Brassicaceae plants, rapeseed and radish, but not withthose of the light-harvesting Chl a/b-proteins of photosynthesis.A possible function of the water-soluble Chl-protein was discussed. The rate and final percentage of field emergence (seed quality)was measured in 55 seed-lots of carrots (Daucus carota L.),45 seed-lots of onion (Allium cepa L.) and 32 seed-lots of cabbage(Brassica oleracea var. capitata), together with the followingbiochemical indices: rates of ATP-synthesis; reduction of tetrazoliumby seed powder; AMP and malate content. Differences in field performance of seed-lots (seed quality)showed some correlation with particular biochemical indices.Seed quality was most significantly correlated with AMP contentand ATP synthesis in carrot, with malate content, reductionof tetrazolium and ATP synthesis in onion and with malate andAMP content and reduction of tetrazolium in cabbage. N-4-pyridyl-N′-phenyl urea (NC5392) showed similar activity to N6-benzyladenine (BA) in stimulating lateral bud development on Brussels sprout (Brassica oleracea var. gemmifera) seedlings particularly on de-rooted plants, but increased average bud weight far more than did BA. Both compounds increased flowering of young plants given an insufficient cold stimulus and seeds from treated plants showed a higher germination percentage than those from untreated plants.  相似文献

Nitrate Supply and the Biophysics of Leaf Growth in Salix viminalis   总被引:2,自引:0,他引:2  
The influence of nitrogen on leaf area development and the biophysicsof leaf growth was studied using clonal plants of the shrubwillow, Salix viminalis grown with either optimal (High N) orsub-optimal (Low N) supplies of nitrate. Leaf growth rate andfinal leaf size were reduced in the sub-optimal treatment andthe data suggest that in young rapidly growing leaves, thiswas primarily due to changes in cell wall properties, sincecell wall extensibility (% plasticity) was reduced in the LowN plants. The biophysical regulation of leaf cell expansion also differedwith nitrogen treatment as leaves aged. In the High N leaves,leaf cell turgor pressure (P) increased with age whilst in theLow N leaves P declined with age, again suggesting that foryoung leaves, cell wall plasticity limited expansion in theLow N plants. Measurements of cell wall properties showed thatcell wall elasticity (%E) was not influenced by nitrogen treatmentand remained constant regardless of leaf age. Plants of the C4 sedge Cyperus longus L. were grown at 10, 20and 30 °C. An asymptotic growth curve, the Richards function,was fitted to growth data for successive leaves. The mean rateof leaf appearance was a linear function of temperature with0.014 leaves appearing per day for every 1 °C increase intemperature. The instantaneous relative rate of leaf extensionshowed a marked ontogenetic drift which was most rapid at 30°C and slowest at 10 °C. The mean absolute extensionrate for foliage had a temperature coefficient of 0.16 cm d–1° C–1 in the range from 10 to 30 °C. The durationof leaf growth was independent of leaf number at 10 and 20 °Cbut increased linearly with leaf number at 30 °C. The smalldifferences in relative growth rate at the three temperaturesresulted in large differences in foliage area produced at theend of a 30 d growth period. The effects of root hypoxia on leaf growth of a Populus trichocarpa? deltoides hybrid have been assessed. Clonal plants were subjectedto hypoxic root conditions in pot culture by flooding and insolution culture by gassing with nitrogen. The rate of leafexpansion declined within 8 h and was suppressed for the durationof the treatment. Final leaf size was reduced by 35% to 60%compared to aerated plants. Final epidermal cell size and numberdepended both on the developmental stage of the leaf at theonset of stress and on the duration of the treatment. No differencesin bulk leaf water potential were measured between the hypoxicand aerated plants. Cell wall extensibility was lower, leafsolute potential was more negative and turgor potential washigher in leaves of hypoxia-treated plants than of aerated plants.These data suggest that leaf growth of hypoxia-stressed plantsis limited by cell wall extensibility.
