A consistent supply of healthy tarnished plant bug, Lygus lineolaris (Palisot de Beauvois) (Hemiptera: Miridae), is necessary for the development of novel management strategies targeting this pest. After being in culture for several years, a substantial portion of a tarnished plant bug colony was found to be infected with a Nosema (Microsporidia) species. Studies were subsequently undertaken to evaluate the impact of Nosema infection on tarnished plant bug productivity and to test the efficacy of fumagillin to treat this infection. Using buffalo black stain, infections could not be reliably detected in adult tarnished plant bugs until adults were 6-8days post eclosion. Nosema infections reduced adult longevity and fecundity. Maximum fecundity was restored using a concentration of 16.8ppm fumagillin while maximum longevity for females was at a concentration of 33.6ppm fumagillin incorporated into the tarnished plant bug diet. Minimum infection scores were obtained at 67.2ppm, the highest concentration tested. A field survey of tarnished plant bugs in Mississippi found Nosema infections in 3% of wild tarnished plant bugs.  相似文献

The accurate measurement of insect mortality by parasites is critical in biological control research, both in baseline studies to determine the absence or inadequacy of native parasites and in subsequent efforts to measure the effectiveness of introduced endoparasitic species. Although rearing has been most frequently used to measure parasitism, dissection has been shown to be more accurate in several cases. Selection of the host instar, whether for rearing or dissection, was also found to be important in this study. In two species [Lygus lineolaris (Palisot) and L. hesperus Knight], parasitism by Peristenus digoneutis Loan and P. howardi Shaw, respectively, was highest in instars 3 and 4. Parasitism was underestimated in instars 1 and 2 (because of reduced exposure time) and in instar 5 (because of parasites killing the hosts in instar 4).  相似文献

Several species of Lygus (Heteroptera: Miridae: Mirini) are serious crop pests in North America where their parasitism rate by native nymphal parasitoids is generally lower than in Europe. Peristenus relictus (Ruthe) (formerly P. stygicus Loan) (Hymenoptera: Braconidae: Euphorinae) is the predominant nymphal parasitoid of several Lygus spp. in the warm Mediterranean region and has been a candidate for introduction against Lygus hesperus Knight and L. lineolaris (Palisot de Beauvois) in the southern US. We report a rapid, sensitive, and specific PCR-based assay for diagnosis of P. relictus immature stages within Lygus nymphs that entails three steps: DNA extraction, PCR of the partial mitochondrial COI gene, and agarose gel electrophoresis. The PCR-based methodology is species-specific because the target DNA of other sympatric, congeneric species was not amplified with use of the primers developed for P. relictus diagnosis. The sensitivity of the PCR method, assessed through spike tests, was established by the detection of a ratio of 1:10,000 P. relictus DNA to Lygus DNA. Molecular diagnosis of parasitism of field collected nymphs is achievable in one day, eliminating the need to rear nymphs to obtain adult parasitoids for morphological identification.  相似文献

Host dissection, host rearing and PCR analysis were used to estimate parasitism level and identify the parasitoid complex in Lygus spp. populations in alfalfa. Parasitism level estimates by PCR were higher than by other methods. The PCR results from all samples tested with available primers were consistent with Peristenus pallipes, although the parasitoid was identified as Peristenus near mellipes by morphological characters.  相似文献

Lygus hesperus Knight (Hemiptera: Miridae) is a key pest of fruit and vegetable crops, forages, and cotton (Gossypium spp.) in the western United States. Accurate models describing relationships between temperature and L. hesperus development are critical to the study of seasonal L. hesperus population dynamics. Development of L. hesperus nymphs was assessed at nine constant temperatures from 10 to 37.8 degrees C. The relationships between temperature and development for each L. hesperus instar, and for the entire nymphal stage, were best described by six-parameter biophysical models indicating both low- and high-temperature inhibition of development. Development rates asymptotically approached zero with decreasing temperature in the lower thermal range, and decreased with increasing temperatures above 32.2 degrees C. Nymphs did not survive from egg hatch to adulthood at either 10 or 37.8 degrees C, and nymph mortality was > 90% at both 12.8 and 35.0 degrees C. The fifth instar exhibited the longest stadium, whereas the shortest stadia were associated with the second and third instars. Development rates of males and females did not differ, and the ratio of males to females was not different from 1:1 at any temperature. Our temperature-dependent development rate models for L. hesperus nymphs will facilitate control of insect physiological age in controlled laboratory experiments, and should be useful in planning and interpreting field studies on L. hesperus population dynamics.  相似文献

Several aspects of the basic biology of the western tarnished plant bug, Lygus hesperus Knight, are poorly known despite the economic importance of this species. Among these are the factors regulating the adult diapause. Reports of recent studies questioned the validity of earlier reports of diapause in L. hesperus, in part because of the demonstrated loss of diapause response in insects obtained from long-standing laboratory colonies. However, use of laboratory reared insects would facilitate additional diapause research, so long as those insects exhibit a diapause response similar to that of the field population. L. hesperus, originating as eggs from field-collected insects, were reared in the laboratory for four generations to examine corresponding changes in selected biological characteristics. Over the course of the four generations, incidence of diapause in both L. hesperus genders decreased whereas the frequency of oviposition by virgin females increased. Measurable changes were not observed in frequency of occurrence of a specific fat body type (glass bead fat) or nymphal development time. These results suggest L. hesperus used in diapause research should be as close to the field population as possible, but no further removed than three generations. Results further demonstrate variability among different biological characteristics in their responses to selection from laboratory rearing. Collectively, these findings demonstrate the importance of understanding the influences of rearing on specific biological characteristics under study, and the need to verify the similarity of laboratory-reared insects to their native counterparts in studies used to draw inferences regarding the field population.  相似文献

The tarnished plant bug, Lygus hesperus Knight, is a pest that causes considerable economic losses to vegetables, cotton, canola, and alfalfa. Detailed knowledge of its digestive physiology will provide new opportunities for a sustainable pest management approach to control this insect. Little is known about the different protease class contributions to the overall digestion of a specific protein. To this end, the proteolytic activities in female adult L. hesperus salivary gland and midgut homogenates were quantified over a range of pH's and time points, and the contribution of different classes of proteases to the degradation of FITC-casein was determined. In the salivary gland, serine proteases were the predominant class responsible for caseinolytic activity, with the rate of activity increasing with increasing pH. In contrast, both aspartic and serine proteases contributed to caseinolytic activity in the midgut. Aspartic protease activity predominated at pH 5.0 and occurred immediately after incubation, whereas serine protease activity predominated at pH 7.5 after a 9h delay and was resistant to aprotinin. The salivary serine proteases were distinctly different from midgut serine proteases, based on the tissue-specific differential susceptibility to aprotinin and differing pH optima. Collectively, the caseinolytic activities complement one another, expanding the location and pH range over which digestion can occur.  相似文献

The pathogenicity of 32 fungal isolates from the genera of Beauveria, Verticillium, Paecilomyces, Metarhizium, Mariannaea, and Hirsutella to second-instar tarnished plant bug, Lygus lineolaris (Palisot de Beauvois), was tested under laboratory conditions. These isolates originated from various insect hosts and substrates from France, Denmark, Austria, Italy, Turkey, Syria, and the United States. A single exposure concentration (1 x 10(7) conidia/ ml) assay for each isolate was first conducted by immersing the insects in 10 ml of a fungal suspension for 5s. These were followed by concentration-mortality assays on five of the most pathogenic isolates using four test concentrations ranging from 2 x 10(4) to 2 x 10(7) conidia/ml. B. bassiana 726 (Bemisia-passaged GHA strain) was used as a standard for comparison in all of the assays. Among the test isolates, three produced mortality not significantly different from the water control. Mortality ranged from 35 to 98% among the other 29 isolates. The LC50 values of the five most pathogenic isolates ranged from 0.8 to 5.0 x 10(5) conidia/ml. The LT50 values for these isolates ranged from 6.0 to 6.9, 3.1 to 5.1, and 2.5 to 4.0 d for concentrations of 2 x 10(5), 2 x 10(6), and 2 x 10(7) conidia/ml, respectively. Two strains of B. bassiana (ARSEF 1394,5665) and one M anisopliae (ARSEF 3540) were more pathogenic to the nymphs than the standard, having significantly lower LC50 and LT50, values. Our results demonstrated that several genera of entomopathogenic fungi have promise as microbial control agents against L. lineolaris.  相似文献

Releases of Peristenus digoneutis against Lygus spp. in North America have been conducted for many years; however, no published procedures for mass production of the biological control agent were available. A laboratory rearing method was developed using Lygus lineolaris as the host to enhance establishment efforts and provide large numbers of wasps for inundative releases into high value fruit crops. Experiments were conducted to determine optimum host:parasitoid density and rearing temperature. The effects of nymph:wasp ratios and temperature on parasitism and wasp survival showed a 20:1 ratio at 20°C provided high parasitism (256 parasitized nymphs/wasp over lifetime) and excellent wasp survival of 27 days. Experiments on diapause-inducing conditions for P. digoneutis demonstrated that fluctuating temperatures of 23°C (day) and <16°C (night) and corresponding photo phases of 16 h light, for rearing parasitized nymphs, produced 100% diapausing parasitoids whereas non-diapausing parasitoids were only produced at more than 16 h light. Furthermore, parasitized Lygus nymphs need to be transferred to short day conditions no later than 10 days after parasitism to produce diapausing parasitoids. Critical life stages for exposure to conditions inducing diapause, the egg, first and second instar parasitoid larva, occurred from 0 to 10 days at 24°C constant temperature. Increased time in cold storage reduced the number of days to first emergence of parasitoids from diapausing cocoons when transferred to warm temperatures. The optimum storage time for diapausing P. digoneutis is between 25 and 44 weeks, depending upon the length of time that cocoons remain at warm conditions prior to chilling.  相似文献

【目的】筛选出适合牧草盲蝽Lygus pratensis在不同条件下稳定表达的内参基因。【方法】选取编码牧草盲蝽α-微管蛋白、β-微管蛋白、肌动蛋白、延伸因子、琥珀酸脱氢酶复合体A、泛素、转录起始因子TFIID亚基、谷胱甘肽S-转移酶、核糖体蛋白L32及TATA盒结合蛋白的10条基因为候选内参基因,采用qRT-PCR技术测定其在牧草盲蝽不同性别成虫、成虫不同组织、不同龄期、对功夫菊酯不同抗性成虫、不同温度及不同杀虫剂分别处理后的成虫共6类样品中的表达量;采用BestKeeper, geNorm, NormFinder及RefFinder 4种计算程序对各候选基因的表达稳定性进行评价。【结果】依据获得的有效qRT-PCR数据前提,利用不同计算方法综合分析得出:在牧草盲蝽成虫不同组织中和对功夫菊酯不同抗性成虫中最稳定表达的内参基因均为RPL32;不同温度处理、不同性别成虫中、不同杀虫剂处理和不同龄期牧草盲蝽中最稳定表达的内参基因分别为UBQ, SDHA, β-tubulin和TAF。通过geNorm软件判断配对变异数确定的内参基因最佳数目,结合RefFinder对基因表达稳定性综合排序的结果可得:牧草盲蝽不同成虫组织中、不同性别成虫中、不同龄期、对功夫菊酯不同抗性成虫中、不同温度及杀虫剂分别处理的成虫等各组的最优内参基因组合分别为RPL32+TAF, SDHA+GST, TAF+GST+UBQ, RPL32+GST, UBQ+ACT+β-tubulin和β-tubulin+TAF+RPL32。【结论】本研究获得的牧草盲蝽在不同条件下表达最稳定及最适内参基因组合,都可用于后续的基因定量表达研究。  相似文献

Parasitism of tarnished plant bug nymphs, Lygus lineolaris (Palisot de Beauvois) was investigated on different weeds and on lettuce, celery and alfalfa-clover, in a muck soil area of southwestern Quebec. The phenology of parasitism, defined as the time during which female parasitoids are active in the field, was evaluated. Two nymphal parasitoid species were found: Peristenus pallipes (Curtis) and P. pseudopallipes (Loan). Lygus lineolaris was most abundant on stinging-nettle with levels of parasitism of 17% (P. pallipes) and 27% (P. pseudopallipes) and on alfalfa-clover with levels of parasitism of 25% (P. pallipes) and 8% (P. pseudopallipes). On lettuce, parasitism by P. pseudopallipes was below 4% while no parasitism was observed on celery. The dissection and rearing methods were used to estimate parasitism, and dissection usually yielded parasitism levels higher than rearing. Degree-days requirements of both host and parasitoids were used to calculate the period of activity of parasitoid adults. The estimates of adult activity were 47 days from late May to early July for P. pallipes, and 42 days from mid-July to late August for P. pseudopallipes.  相似文献

A microsatellite‐enriched partial genomic DNA library of Lygus hesperus was generated and screened by sequencing. Ten polymorphic microsatellite marker loci were characterized by genotyping 92 insect samples. The observed number of alleles ranged from three to seven with an average of 4.5 (SE ± 0.45), while the effective number of alleles ranged from 1.21 to 3.02 with an average of 2.14 (SE ± 0.20). Significant deviations from Hardy–Weinberg expectations were detected at three loci. Significant linkage disequilibrium was also detected between the loci LhMS2‐54 and LhMS3‐32. Seven of the L. hesperus markers could be transferred to Lygus lineolaris.  相似文献

Helicoverpa spp. and mirids, Creontiades spp., have been difficult to control biologically in cotton due to their unpredictable temporal abundance combined with a cropping environment often made hostile by frequent usage of broad spectrum insecticides. To address this problem, a range of new generation insecticides registered for use in cotton were tested for compatibility with the assassin bug, Pristhesancus plagipennis (Walker), a potential biological control agent for Helicoverpa spp. and Creontiades spp. Indoxacarb, pyriproxifen, buprofezin, spinosad and fipronil were found to be of low to moderate toxicity on P. plagipennis whilst emamectin benzoate, abamectin, diafenthiuron, imidacloprid and omethaote were moderate to highly toxic. Inundative releases of P. plagipennis integrated with insecticides identified as being of low toxicity were then tested and compared with treatments of P. plagipennis and the compatible insecticides used alone, conventionally sprayed usage practice and an untreated control during two field experiments in cotton. The biological control provided by P. plagipennis nymphs when combined with compatible insecticides provided significant (P<0.001) reductions in Helicoverpa and Creontiades spp. on cotton and provided equivalent yields to conventionally sprayed cotton with half of the synthetic insecticide input. Despite this, the utilization of P. plagipennis in cotton as part of an integrated pest management programme remains unlikely due to high inundative release costs relative to other control technologies such as insecticides and transgenic (Bt) cotton varieties.  相似文献

Mobility and distribution of adult tarnished plant bug, Lygus lineolaris (Palisot de Beauvois), was studied on a day-neutral strawberry cultivar ('Selva') in 1991 and 1992 at L'Acadie, Quebec, Canada. On cage-covered plants, individuals were located mainly on reproductive parts. Study of the flight activity with sticky posts revealed that most captures were obtained < 1 m from ground level and that the time of the day at which the maximum counts occur varied among seasons. Three pest-sampling methods (white sticky trap, tapping of flower clusters, and D-Vac) were evaluated over continuous 24-h periods. Maximum captures of adults with white sticky traps were made at midday (1000-1400 hours), whereas the two other methods proved more effective at the beginning (0600-0800 hours) or the end of the day (2000-2200 hours) or during night time. For nymphs, maximum catches were obtained during the day with tapping and D-Vac; white sticky traps were ineffective. Because the D-Vac captures individuals present on all parts of the plant, these counts were used to monitor the effectiveness of the two other sampling techniques. The data suggest that tapping flower clusters throughout the day is a very effective sampling method to estimate nymphal tarnished plant bug populations in strawberries. However, sampling of populations with a high proportion of adults should take into account the bias caused by their flight activity, and sampling should be conducted early in the morning or at the end of the day.  相似文献

Lygus hesperus Knight (Hemiptera: Miridae) is a key pest of cotton (Gossypium spp.) in the western United States that injures floral buds (squares) and developing fruit (bolls). However, no clear relationship between Lygus population level and plant injury has been established. Age-dependent feeding activity by L. hesperus is a possible source of variation that has not been examined for its influence in studies of the impact of Lygus on cotton. Recent video-based laboratory studies indicated that feeding behaviors and trivial movement varied among L. hesperus adults of different gender and reproductive states (prereproductive; reproductive and unmated; and reproductive and mated). We compared within-plant distributions and accumulations of feeding injury to intact cotton plants corresponding to adult L. hesperus of different gender and reproductive states. Adult females, regardless of reproductive state, were observed on squares and axillary buds more often than were males. Additionally, prereproductive adults were observed on squares and axillary buds more often than were mated or unmated reproductive adults, regardless of gender. Plants that were exposed to prereproductive adults exhibited more abscised squares and more squares with injured anthers compared with plants exposed to reproductive adults. However, feeding injury did not differ by insect mating status or gender. These results are consistent with results of our previous video-based assays, and indicate adult reproductive state represents a source of variation that should be controlled in studies to evaluate Lygus-induced injury to cotton and other crop plants.  相似文献

The sweep net is a standard sampling method for adults of the western tarnished plant bug, Lygus hesperus Knight (Hemiptera: Miridae), in cotton (Gossypium spp.). However, factors that influence the relationship between true population levels and population estimates obtained using the sweep net are poorly documented. Improved understanding of these factors is needed for the development and application of refined treatment thresholds. Recent reports of significant among-sampler differences in sweep net-based population estimates of the adult tarnished plant bug, Lygus lineolaris (Palisot de Beauvois), seem to preclude meaningful comparisons of population estimates collected by different samplers. We used a mark-release-recapture method and the standard sweep net to evaluate among-sampler differences in population estimates of L. hesperus adults. Adult lygus, marked with fingernail polish to facilitate identification and prevent flight, were released into 10-m sample rows on the evening before 10-sweep samples were collected the following morning. The experimental design was a randomized complete block with three replications of three treatments (sampler). Separate experiments were conducted in two plantings each of Pima (Gossypium barbadense L.) and Acala (Gossypium hirsutum L.) cotton. Collections of marked bugs from each study were evaluated for effects of sampler, sample date, and their interaction. Although differences in lygus collections were observed among sample dates in some tests, no differences were detected in the population estimates by different samplers. These results demonstrate that the sweep net technique can be sufficiently standardized to allow direct comparison of population estimates obtained by different samplers.  相似文献

Abstract Information on host plant preference of agriculturally important insect pests, such as Lygus hesperus (Knight), can be helpful in predicting its occurrence and future movement among crop and non-crop host plants. A field study was conducted during 2005 and 2006 to evaluate the host preference of Lygus to cotton and four other host plants in the Texas High Plains, including alfalfa (Medicago sativa L.), wild sunflower (Helianthus annuus L.), Russian thistle (Salsola iberica L.) and pigweed (Amaranthus palmeri L.). Sampling for both nymphs and adults during 2005 (July to November) and 2006 (June to November) showed that alfalfa and Russian thistle were the two most preferred hosts out of the five hosts evaluated. Abundance of nymphs (numbers per 50 sweeps per host plant) during the sampling period also indicated the superior reproductive suitability of alfalfa and Russian thistle. Cotton appeared to be the least attractive host plant for Lygus when Russian thistle and alfalfa were available in the host mosaic. Seasonal abundance of Lygus was found to be lower during 2006 compared to 2005, which may be explained by the difference in rainfall patterns during these two years. In terms of species dominance, L. hesperus was the most dominant species in the sampled population followed by an inconsiderable fraction of L. elisus and L. lineolaris.  相似文献

Trypsin-like enzymes from the salivary gland complex (SGC) of Lygus hesperus Knight were partially purified by preparative isoelectric focusing (IEF). Enzyme active against Nalpha-benzoyl-L-arginine-p-nitroanilide (BApNA) focused at approximately pH 10 during IEF. This alkaline fraction gave a single activity band when analyzed with casein zymograms. The serine proteinase inhibitors, phenylmethylsulfonyl fluoride (PMSF) and lima bean trypsin inhibitor, completely inhibited or suppressed the caseinolytic activity in the crude salivary gland extract as well as the IEF-purified sample. Chicken egg white trypsin inhibitor also inhibited the IEF-purified sample but was not effective against a major caseinolytic band in the crude salivary gland extract. These data indicated the presence of serine proteinases in the SGC of L. hesperus. Cloning and sequencing of a trypsin-like precursor cDNA provided additional direct evidence for serine proteinases in L. hesperus. The encoded trypsin-like protein included amino acid sequence motifs, which are conserved with five homologous serine proteinases from other insects. Typical features of the putative trypsin-like protein from L. hesperus included residues in the serine proteinase active site (His(89), Asp(139), Ser(229)), conserved cysteine residues for disulfide bridges, residues (Asp(223), Gly(252), Gly(262)) that determine trypsin specificity, and both zymogen signal and activation peptides.  相似文献
