Abstract The range of inositol phosphates and inositol phospholipids present in three filamentous fungi, Neurospora crassa, Fusarium graminearum and Phanerochaete chrysosporium has been investigated by HPLC analysis. The profiles obtained demonstrate that two isomers of phosphatidyl inositol monophosphate are present, and that an apparent complexity in the number of isomers of inositol bis- and trisphosphates is found in filamentous fungi that has not been observed in animal or plant cells.  相似文献

Omphalina basidiolichens are obligate mutualistic associations of a fungus of the genus Omphalina (the exhabitant) and a unicellular green alga of the genus Coccomyxa (the inhabitant). It has been suggested that symbiotic inhabitants have a lower rate of genetic change compared to exhabitants because the latter are more exposed to abiotic environmental variation and competition from other organisms. In order to test this hypothesis we compared substitution rates in the nuclear ribosomal internal transcribed spacer region (ITS1, 5.8S, ITS2) among fungal species with rates among their respective algal symbionts. To ensure valid comparisons, only taxon pairs (12) with a common evolutionary history were used. On average, substitution rates in the ITS1 portion of Omphalina pairs were 27.5 times higher than rates in the corresponding pairs of Coccomyxa since divergence from their respective ancestor at the base of the Omphalina/Coccomyxa lineage. Substitution rates in the 5.8S and the ITS2 portions were 2.4 and 18.0 times higher, respectively. The highest rate difference (43.0) was found in the ITS1 region. These are, to our knowledge, the highest differences of substitution rates reported for symbiotic organisms. We conclude that the Omphalina model system conforms to the proposed hypothesis of lower substitution rates in the inhabitant, but that the mode of transmission of the inhabitant (vertical versus horizontal) could be a prevailing factor in the regulation of unequal rates of nucleotide substitution between co-evolving symbionts. Our phylogenetic study of Coccomyxa revealed three main lineages within this genus, corresponding to free-living Coccomyxa, individuals isolated from basidiolichens Omphalina and Coccomyxa isolated from ascolichens belonging to the Peltigerales.  相似文献

Of 70 pure microbial cultures isolated from aquatic habitats, soil, and air according to the ability to lyse live blue-green algae, 62 were fungi representing the genera Acremonium, Emericellopsis, and Verticillium. Algal-lysing fungi were isolated from all habitat types sampled. The remaining isolates comprised four bacteria and four streptomycetes. All isolates lysed Anabaena flos-aquae and, in most cases, several other filamentous and unicellular blue-green algae. The fungi generally showed greater activity than most other isolates towards a wider range of susceptible algae, including green algae in some cases. Acremonium and Emericellopsis isolates, but not Verticillium, also inhibited the growth of blue-green algae and gram-positive bacteria, but did not lyse the latter. Lysis of blue green algae by Acremonium and Emericellopsis spp. was associated with the formation of diffusible heat-stable extracellular factors which, evidence suggests, could be cephalosporin antibiotic(s). Blue-green algae were also lysed by pure cephalosporin C. The frequent isolation of lytic fungi from algal habitats suggests a possible natural algal-destroying role for such fungi, which might be exploitable for algal bloom control.  相似文献

Local and chemical distribution of phlorotannins in brown algae   总被引:2,自引:2,他引:2  
The local and chemical distribution of phlorotannins among the Japanese Laminariaceae, Eisenia bicyclis, Ecklonia cava and Ecklonia kurome, was investigated. As a result of light microscopy observations with vanillin-HCl staining, phlorotannins were found to be accumulated within the vegetative cells of the outer cortical layer of the thalli, regardless of the species, stage of growth or organ. Crude phlorotannins comprised about 3.0% of the algal powder for each of the algae. High-performance liquid chromatography (HPLC) showed that the phlorotannins of E. bicyclis were composed of phloroglucinol (0.9%), phloroglucinol tetramer (4.4%), eckol (7.5%), phlorofucofuroeckol A (21.9%), dieckol (23.4%), and 8,8'-bieckol (24.6%), plus some other unknown phenolic compounds (17.3%). The composition of the phlorotannins differed little among the Laminariaceae, except for a significantly larger amount of the tetramer, MW 478, in E. bicyclis.  相似文献

Hitherto only gametangial and carposporangial phases of Thuretellopsis peggiana Kylin have been reported. A study of this rare member of the Dumontiaceae (Cryptonemiales) in culture has disclosed the existence of an encrusting tetrasporangial phase, previously unknown, whilst the life history has been shown to be of the ‘Bonnemaisonia’ type.  相似文献

The activity of choline acetyltransferase (ChAt; E.C. 2.3.1.6) measured as the bromoacetylcholine-sensitive portion of the maximal rate of acetylcholine synthesis has been determined in homogenates of nine regions of the heart of control rats and rats sacrificed 72-74 h after bilateral cervical vagotomy. When related to the content of protein, the activity of ChAT was lowered by 30% in the atria and unchanged in the ventricles of vagotomized rats. The highest absolute decline caused by vagotomy was observed in the sinoatrial region; it was somewhat less in the rest of the right atrium and in the interatrial septum and considerably less in the left atrium. It is concluded that preganglionic parasympathetic fibres supply mainly the sinoatrial region and the right atrium, and that they do not branch to the ventricles. Preganglionic ChAT nts about 30% of total ChAT activity in the atria. The sinoatrio-ventricular gradient in the distribution of ChAT in the heart is due to uneven distribution of both the pre- and postganglionic ChAT pools (i.e., of both the pre- and postganglionic cholinergic nerve fibres and nerve cell bodies).  相似文献

Cadmium transport, resistance, and toxicity in bacteria, algae, and fungi   总被引:8,自引:0,他引:8  
Cadmium is an important environmental pollutant and a potent toxicant to bacteria, algae, and fungi. Mechanisms of Cd toxicity and resistance are variable, depending on the organism. It is very clear that the form of the metal and the environment it is studied in, play an important role in how Cd exerts its effect and how the organism(s) responds. A wide range of Cd concentrations have been used to designate resistance in organisms. To date, no concentration has been specified that is applicable to all species studied under standardized conditions. Cadmium exerts its toxic effect(s) over a wide range of concentrations. In most cases, algae and cyanobacteria are the most sensitive organisms, whereas bacteria and fungi appear to be more resistant. In some bacteria, plasmid-encoded resistance can lead to reduced Cd2+ uptake. However, some Gram-negative bacteria without plasmids are just as resistant to Cd as are bacteria containing plasmids encoding for Cd resistance. According to Silver and Misra (1984), there is no evidence for enzymatic or chemical transformations associated with Cd resistance. Insufficient information is available on the genetics of Cd uptake and resistance in cyanobacteria and algae. Mechanisms remain largely unknown at this point in time. Cadmium is toxic to these organisms, causing severe inhibition of such physiological processes as growth, photosynthesis, and nitrogen fixation at concentrations less than 2 ppm, and often in the ppb range (Tables 2 and 3). Cadmium also causes pronounced morphological aberrations in these organisms, which are probably related to deleterious effects on cell division. This may be direct or indirect, as a result of Cd effects on protein synthesis and cellular organelles such as mitochondria and chloroplasts. Cadmium is accumulated internally in algae (Table 4) as a result of a two-phase uptake process. The first phase involves a rapid physicochemical adsorption of Cd onto cell wall binding sites, which are probably proteins and (or) polysaccharides. This is followed by a lag period and then a slow, steady intracellular uptake. This latter phase is energy dependent and may involve transport systems used to accumulate other divalent cations, such as Mn2+ and Ca2+. Some data indicate that Cd resistance, and possibly uptake, in algae and cyanobacteria is controlled by a plasmid-encoded gene(s). Although considerable information is available on Cd toxicity to, and uptake in fungi, further work is clearly needed in several areas. There is little information about Cd uptake by filamentous fungi, and even in yeasts, information on the specificity, kinetics, and mechanisms of Cd uptake is limited.(ABSTRACT TRUNCATED AT 400 WORDS)  相似文献

Abstract Choline is an essential metabolite for the growth of filamentous fungi. It occurs most notably as a component of the major membrane phospholipid, phosphatidyl choline (lecithin), and fulfills a major role in sulphate metabolism in the form of choline- o -sulphate in many species. Choline is usually synthesised endogenously, but exogenous choline can also be taken up, either to compensate for metabolic deficiencies in choline-requiring mutants such as those of Aspergillus nidulans and Neurospora crassa , or as a normal function by species such as Fusarium graminearum which do not require added choline for growth. F. graminearum has a highly specific constitutive uptake system for this purpose. Recent studies have begun to indicate that choline also plays an important role in hyphal and mycelial morphology. Over a wide range of concentrations, choline influences mycelial morphology, apparently influences mycelial morphology, apparently by controlling branch initiation. At high concentrations of added choline, branching is inhibited but specific growth rate is unaffected, leading to the production of rapidly extending, sparsely branched mycelia. Reduction of choline concentration allows a progressive increase in branching. Additionally, in choline-requiring mutants which have a very reduced content of choline, multiple tip-formation and apical branching occurs. Just prior to cessation of growth in choline-starved cultures of A. nidulans choline-requiring mutants, hyphal morphology changes due to a brief phase of unpolarised growth to produce spherical swellings called ballons, at or near hyphal apices. The precise mechanism by which choline affects fungal morphology is not yet known, although in A. nidulans it appears to be at least partially due to the influence of membrane composition on the synthesis of the hyphal wall polymer chitin. Several hypotheses for the possible mode of action of choline in affecting fungal morphology are discussed here.  相似文献

以兰州五泉山为该地藻种资源库,对其中水生、陆生生境中藻类的种类多样性、群落结构、分布特点进行了研究。结果发现该地藻类植物65种(含4变种),包括蓝藻、绿藻、硅藻和红藻,其中硅藻种类最多(29种),其它依次为蓝藻(24种)、绿藻(11种)和红藻(1种)。水体中共42种,硅藻最多,有26种,其次蓝藻8种,绿藻7种,红藻1种,不同水体中优势种和亚优势种不同。土壤生境中发现20种,蓝藻13种,绿藻4种,硅藻3种,且非洲席藻和小球藻分为优势种和亚优势种。7个种类在水、陆两大生境都有分布,而且它们主要是丝状蓝藻。  相似文献
