Decontamination studies investigating the effectiveness of products and processes for the inactivation of Bacillus species spores have traditionally utilized metering viable spores in a liquid suspension onto test materials (coupons). The current study addresses the representativeness of studies using this type of inoculation method compared to when coupons are dosed with a metered amount of aerosolized spores. The understanding of this comparability is important in order to assess the representativeness of such laboratory-based testing when deciding upon decontamination options for use against Bacillus anthracis spores. Temporal inactivation of B. anthracis surrogate (B. subtilis) spores on representative materials using fumigation with chlorine dioxide, spraying of a pH-adjusted bleach solution, or immersion in the solution was investigated as a function of inoculation method (liquid suspension or aerosol dosing). Results indicated that effectiveness, measured as log reduction, was statistically significantly lower when liquid inoculation was used for some material and decontaminant combinations. Differences were mostly noted for the materials observed to be more difficult to decontaminate (i.e., wood and carpet). Significant differences in measured effectiveness were also noted to be a function of the pH-adjusted bleach application method used in the testing (spray or immersion). Based upon this work and the cited literature, it is clear that inoculation method, decontaminant application method, and handling of non-detects (i.e., or detection limits) can have an impact on the sporicidal efficacy measurements.  相似文献

Aims: To obtain data on the efficacy of various liquid and foam decontamination technologies to inactivate Bacillus anthracis Ames and Bacillus subtilis spores on building and outdoor materials. Methods and Results: Spores were inoculated onto test coupons and positive control coupons of nine different materials. Six different sporicidal liquids were spray‐applied to the test coupons and remained in contact for exposure times ranging from 10 to 70 min. Following decontamination, spores were recovered from the coupons and efficacy was quantified in terms of log reduction. Conclusions: The hydrogen peroxide/peracetic acid products were the most effective, followed by decontaminants utilizing hypochlorous acid chemistry. Decontamination efficacy varied by material type. Significance and Impact of the Study: The study results may be useful in the selection of technologies to decontaminate buildings and outdoor areas in the event of contamination with B. anthracis spores. These results may also facilitate selection of decontaminant liquids for the inactivation of other spore‐forming infectious disease agents.  相似文献

Aims: We evaluated whether lowering pH (with acetic acid) and raising free available chlorine (FAC) levels in bleach solutions would improve efficacy in inactivating Bacillus spores on different materials. We also determined how varying pH and FAC levels affected bleach stability. Methods and Results: Acidified bleach solutions with pH levels of 4·5, 6 and 7·5 and FAC levels between 5000 and 10 000 ppm were evaluated for decontamination efficacy against Bacillus subtilis spores inoculated onto test coupons made from wood, ceramic and galvanized steel. Lowering the pH or increasing the FAC level improved efficacy in some of the tests, but depended on the material, which significantly affected decontamination efficacy. The acidified bleach at pH of 7·5 was significantly less effective than bleach at a pH of 4·5 or 6. The FAC levels in the bleach were the most stable at pH 4·5, and stability at pH 4·5 was not significantly affected by the initial FAC level. Conclusions: It may be advisable to use bleach solutions with lower pH (rather than high FAC levels) in light of both the decontamination efficacy and bleach stability results. For wood materials, use of sporicides other than acidified bleach may be warranted. Significance and Impact of the Study: These results may be useful in preparing acidified bleach solutions for decontamination of materials contaminated with spores such as Bacillus anthracis.  相似文献

Aims: The purpose of this study was to evaluate the effects of environmental conditions and material type on persistence and inactivation of Brucella suis. Methods and Results: Brucella suis (approx. 1 × 108 CFU) was spiked onto surfaces (glass, aluminium and wood) by liquid inoculation. Persistence was evaluated over 56 days at 22 ± 2°C, 40 ± 15% r.h. and 5 ± 3°C, 30 ± 15% r.h. In addition, three readily available decontaminants (pH‐adjusted bleach, 70% ethanol and 1% citric acid) were evaluated for their effectiveness at inactivating Br. suis on these materials. Decontaminations were conducted following 0 and 28 days exposure to the two conditions. Results indicated that Br. suis can persist on environmental surfaces for at least 56 days. Persistence was highest at low temperature. Decontamination was most challenging on wood with all three decontaminants. Conclusions: Following a Br. suis contamination incident, passive decontamination (through attenuation) may not be feasible, as this organism can persist for months. In addition, the results suggest that some sporicidal decontaminants may be ineffective on materials such as wood, even for vegetative biological agents such as Br. suis. Significance and Impact of Study: This study aids incident commanders and remediation experts to make informed decisions regarding decontamination after a biological contamination incident.  相似文献

The effect of denture adhesives on Candida albicans growth in vitro Objective: Denture‐wearing favours the growth of Candida. In view of the fact that many denture wearers regularly use adhesives to enhance denture retention, stability and function, the aim of this work was to study the effect of denture adhesives on Candida albicans growth in vitro. Materials and methods: The denture adhesives tested were Corega® cream, Kukident® cream, Novafix® cream, Polident® cream, Protefix® cream, Steradent® cream, Aderyn® powder, Corega® ultra powder, Protefix® powder and Corega® strip. C. albicans growth curves were obtained in the presence or absence of a 1% solution of the denture adhesive diluted in Sabouraud broth. Macro‐ and microscopic morphological changes in C. albicans were analysed, as was microbial contamination of the denture adhesive. Results: Most of the denture adhesives studied induced morphological changes in C. albicans cells and colonies, but only two had any significant inhibitory effect on yeast growth. Kukident® cream markedly inhibited C. albicans growth in a concentration‐dependent way, reducing the growth rate by 95%, whereas Corega® cream also inhibited C. albicans growth but in a non‐concentration‐dependent way, reducing the growth rate by 37%. In addition, denture adhesives available as powders had detectable microbial contamination. Conclusion: Some commercially available denture adhesives showed microbial contamination and some had significant inhibitory effect on C. albicans growth.  相似文献

Abstract. Since many wood‐living forest species are influenced by the dynamics of coarse woody debris (CWD), information about the spatial pattern of CWD under natural conditions is essential to understand species distributions. In this study we examined the spatial pattern of downed logs and wood‐decaying fungi in an old‐growth boreal Picea abies forest in northwestern Sweden that is governed by gap‐phase dynamics. The spatial pattern of wood‐decaying fungi was studied to draw conclusions about species dispersal abilities. A total of 684 logs with a diameter > 10 cm were mapped and analysed with Ripley's K‐function. The distribution of all logs taken together displayed a significant aggregated pattern up to 45 m. The different decay stages also deviated from random expectations. Fairly fresh logs and logs in the middle decay stage were clumped up to about 25 and 35 m respectively, and late decayed logs aggregated up to 95 m. Logs with diameters from 10–29 cm were aggregated up to 25 m, whereas logs ≥30 cm diameter were randomly distributed. The result suggests that gap‐dynamics do have an impact on the spatial pattern of the CWD, creating fine‐scale clumping. The random distribution of large logs may result from the slightly regular spacing of large living trees. The spatial patterns of 16 species (n > 20) of wood‐decaying fungi were analysed with Ripley's K‐function. Three patterns were aggregated, for Gloeophyllum sepiarium, Coniophora olivacea and Vesiculomyces citrinus. These results indicate that the distribution of most species at the stand level is generally not influenced by dispersal limitations.  相似文献

A wide range of analytical methods are available for the detection and identification of biological warfare agents. These technologies are often hampered in their performance when the inactivated samples are analyzed. To work with pathogens outside of biosafety level 3 laboratories, a complete inactivation is mandatory when appropriate protection equipment is unavailable. When methods of inactivation are used, the detection of bacteria becomes more difficult. In contrast to measuring viable organisms, inactivation steps can have a massive impact on the intrinsic cellular information. This study examined the effects of autoclaving and chemical inactivation methods on Bacillus spores using biological warfare detection setups like real‐time PCR and MALDI‐TOF‐MS. Here, the inactivation of Bacillus atrophaeus spores with formaldehyde, which is a suggested model for biological warfare spore agents, was compared with other inactivation reagents like Wofasteril®E400, a commercially available decontaminant based on peroxyacetic acid. With Wofasteril®E400 the critical factor of inactivation time was reduced to about 15 min and a limit of detection of 8500 spores by PCR was still measurable using five‐times‐washed spores. It has also been shown that MALDI‐TOF‐MS peak information can be hampered by inactivation methods.  相似文献

Potato virus Y (PVY), the potato virus with the highest economic impact in Europe, is transmitted by aphids in a non‐persistent manner. A two‐year field experiment was conducted in Switzerland to evaluate the efficacy of three strategies for controlling aphid populations and the spread of PVY, consisting of treatment with one insecticide (Karate Zeon®), one elicitor (Bion®) and one oil (Telmion®), respectively. The elicitor strategy proved to be ineffective for controlling aphid populations and inadequate for controlling PVY spread. The insecticide strategy gave incomplete protection from aphid infestations, owing to the selection of aphid‐resistant clones. The insecticide gave too little protection against PVY spread for it to be considered a suitable candidate for the purpose. The oil strategy had no effect on aphid populations, but was the best option to reduce PVY spread.  相似文献

Aims

To develop test methods and evaluate the survival of Bacillus anthracis ?Sterne and Bacillus thuringiensis Al Hakam spores after exposure to hot, humid air.

Methods and Results

Spores (>7 logs) of both strains were dried on six different test materials. Response surface methodology was employed to identify the limits of spore survival at optimal test combinations of temperature (60, 68, 77°C), relative humidity (60, 75, 90%) and time (1, 4, 7 days). No spores survived the harshest test run (77°C, 90% r.h., 7 days), while > 6·5 logs of spores survived the mildest test run (60°C, 60% r.h., 1 day). Spores of both strains inoculated on nylon webbing and polypropylene had greater survival rates at 68°C, 75% r.h., 4 days than spores on other materials. Electron microscopy showed no obvious physical damage to spores using hot, humid air, which contrasted with pH‐adjusted bleach decontamination.

Conclusions

Test methods were developed to show that hot, humid air effectively inactivates B. anthracis ?Sterne and B. thuringiensis Al Hakam spores with similar kinetics.

Significance and Impact of the Study

Amylocystis lapponica (Romell) Singer is a widely distributed wood‐decaying polypore fungus found throughout the Northern Hemisphere. Despite its huge distribution range it occurs rather patchily and seems narrowly associated with old‐growth forest stands. Notably, it has been used as an ‘indicator species’, believed to reflect the long‐term presence of dead wood, naturalness of forest stands, and indirectly, species richness and possibly composition. In this study we focused on the last issue – whether or not there is a link between the occurrence of A. lapponica and the species richness and composition of other wood‐decaying fungi. Selecting log characteristics and microclimate as similar as possible, we compared 12 logs with and 12 logs without visible fruit bodies of A. lapponica to examine: 1) if visible fruit bodies corresponded with molecular identification of the mycelia, 2) if fungal species richness and composition of the substrate were related to A. lapponica occurrence, and 3) if A. lapponica was restricted to certain parts of the log. Fungal species were recorded by inspecting visible fruit bodies and by culture isolation and ITS sequencing from wood disc samples. Laboratory and field identification of A. lapponica had 71% correspondence, and mycelia were identified in two logs without visible fruit bodies. Twice as many fungal species were detected using ITS sequencing compared to fruit body identification. Total species richness was similar between the two log categories, but number of species per log was slightly higher in A. lapponica logs. Antrodia serialis (Fr.) Donk, and possibly also Fomitopsis pinicola (Sw.:Fr.) P. Karst. and Phellinus nigrolimitatus (Romell) Bourdot & Galzin, occurred more frequently in A. lapponica logs. Mycelia of A. lapponica were restricted to less decayed parts of the wood in the centre of the middle part of the logs.  相似文献

We studied the decomposition of Cyrilla racemiflora logs over a 13‐yr period in tropical dry and wet forests in Puerto Rico. The mean mass loss, ratio of soft to hard wood, nutrient concentrations, and the diversity of wood‐inhabiting organisms were greater in logs decomposing in the dry forest than in the wet forest. Termites were also more abundant in the logs collected from the tropical dry forest than the tropical wet forest. High moisture content and a low animal diversity on the logs in the wet forest seem to retard wood decay in this habitat. Wood decay rates in the tropical dry forest can be related to the high diversity of species and functional groups of wood‐inhabiting organisms.  相似文献

Initial microbial adhesion to surfaces is a complicated process that is affected by a number of factors. An important property of a solution that may influence adhesion is pH. The surface properties of the cedar wood were characterized by the sessile drop technique. Moreover, the interfacial free energy of surface adhesion to the cedar wood was determined under pH values (2, 3, 5, 7, 9 and 11). The results showed that cedar wood examined at different pH levels could be considered hydrophobic ranged from Giwi = ?13.1 mJ/m2 to Giwi = ?75 mJ/m2. We noted that the electron-donor character of cedar wood was important at both basic and limit acidic conditions (pH 11 and pH 3) and it decreased at intermediate pH (pH 5). The cedar wood substratum presents a weak electron acceptor under various pH’s. In addition, the adhesion of conidia from Penicilllium expansum to the cedar wood surfaces at different pH values (2, 3, 5, 7, 9 and 11) was investigated using Environmental Scanning Electron Microscopy and image analysis was assessed with the Mathlab® program. The data analysis showed that the conidia from P. expansum were strongly influenced by the pH. The maximum adhesion occurs in the pH 11 and pH 3 and decreased to 24% at pH 5.  相似文献

1 Spiders (Araneae) were collected on and near downed woody material (DWM) in a Populus‐dominated forest to determine if spiders utilize wood surfaces, and to ascertain the importance of DWM habitat and wood elevation for spider assemblages. 2 Over 10 000 spiders representing 100 species were collected. Although more spiders were collected on the forest floor, spider diversity was higher in traps located on wood surfaces than on the forest floor, and 11 species were collected more frequently on wood surfaces. 3 Spiders utilized DWM at different stages in their development. Female Pardosa mackenziana (Keyserling) (Lycosidae) carrying egg sacs were caught most often on the surface of DWM, possibly to sun their egg sacs. Additionally, the proportion of immature spiders was higher on the wood surface than on the forest floor. 4 Spiders collected on logs with and without bark were compared to assemblages collected on telephone poles to assess what features of DWM habitat may be important. Web‐building species were seldom collected on telephone poles, suggesting that they depend on the greater habitat complexity provided by DWM. In contrast, hunting spiders did not distinguish between telephone poles and logs. 5 Fewer spiders and a less diverse fauna utilized elevated compared to ground‐level wood. Additionally, Detrended Correspondence Analysis revealed that the spider community from elevated wood was distinguishable from the spider community from ground‐level wood, and from the forest floor spider community.  相似文献

Aims: The adhesion to an inert surface (the first step of biofilm formation) of the two main pathogenic Campylobacter species, Campylobacter jejuni and Campylobacter coli, isolated from diverse origins, was compared. Methods and Results: Adhesion assays were conducted in 96‐well, polystyrene microtiter plates using the BioFilm Ring Test® method. This new technique, based on magnetic bead entrapment, was shown to be suitable for analysing the adhesion of Campylobacter sp. strains by comparing the adhesion of four C. jejuni strains as revealed by the BioFilm Ring Test® and immunodetection. Among the 46 strains tested, C. jejuni and C. coli displayed different adhesion capabilities ranging from no adhesion to strong adhesion. However, no strain of C. coli was strongly adherent, and statistically, C. coli adhered less to an inert surface than C. jejuni. In addition, strains isolated from animals or carcasses were less adherent than those isolated from food‐processing and clinical cases. Conclusions: These observations suggest that the food environment and the human body could have selected strains with greater adhesion. Significance and Impact of the Study: The adhesion capability of strains could partly explain the cross‐contamination or re‐contamination of food products by Campylobacter. This property could provide a mode of survival for Campylobacter in the food chain.  相似文献

Fermentation is one of the most critical steps of the fuel ethanol production and it is directly influenced by the fermentation system, selected yeast, and bacterial contamination, especially from the genus Lactobacillus. To control the contamination, the industry applies antibiotics and biocides; however, these substances can result in an increased cost and environmental problems. The use of the acid treatment of cells (water‐diluted sulphuric acid, adjusted to pH 2·0–2·5) between the fermentation cycles is not always effective to combat the bacterial contamination. In this context, this study aimed to evaluate the effect of ethanol addition to the acid treatment to control the bacterial growth in a fed‐batch system with cell recycling, using the industrial yeast strain Saccharomyces cerevisiae PE–2. When only the acid treatment was used, the population of Lactobacillus fermentum had a 3‐log reduction at the end of the sixth fermentation cycle; however, when 5% of ethanol was added to the acid solution, the viability of the bacterium was completely lost even after the first round of cell treatment. The acid treatment +5% ethanol was able to kill L. fermentum cells without affecting the ethanol yield and with a low residual sugar concentration in the fermented must.

Significance and Impact of the Study

In Brazilian ethanol‐producing industry, water‐diluted sulphuric acid is used to treat the cell mass at low pH (2·0) between the fermentative cycles. This procedure reduces the number of Lactobacillus fermentum from 107 to 104 CFU per ml. However, the addition of 5% ethanol to the acid treatment causes the complete loss of bacterial cell viability in fed‐batch fermentation with six cell recycles. The ethanol yield and yeast cell viability are not affected. These data indicate the feasibility of adding ethanol to the acid solution replacing the antibiotic use, offering a low cost and a low amount of residue in the biomass.  相似文献

ABSTRACT The knockdown and lethal efficacies of five aerosol formulations including Combat Speed® (AIs: 0.1 % imiprothrin and 0.3% cyphenothrin), Raid Power® (AIs: 1.0% pyrethrin and 0.2% permethrin), Home Keeper®, (AIs: 0.2% tetramethrin and 0.3% permethrin), Super Killer® (AIs: 0.32% tetramethrin and 0.08% bioresmethrin), and Perma Kill‐K® (AIs: 0.3% dichlorvos and 0.1% tetramethrin) against five strains of the German cockroach, Blattella germanica (L.) were assessed. The results show that the mean value of KT50 (5.4 sec.) of Combat Speed® was 4.5 and 3.1‐folds lower than those of Perma Kill‐K® and Home Keeper®, respectively. The mean value of KT90 (9.0 sec; slope = 10.02) of Combat Speed® was 3.8 to 5.8‐folds lower than Perma Kill‐K®, Supper Killer® and Home Keeper®. As lethal effects, the mean value of LT50 (17.3 sec.) of Combat Speed® was over 26 folds lower than Supper Killer® and Perma Kill‐K®. The mean value of LT90 (32.9 sec.) of Combat Speed® was 37.4 and 15.1‐folds lower than those of Supper Killer® and Perma Kill‐K®, respectively. In general, Combat Speed® and Raid Power® were considered the insecticide aerosols with faster knockdown and higher lethal effects than Supper Killer®, Perma Kill‐K®, and Home Keeper® against five strains of German cockroaches in Korea. Also, the knockdown and lethal effects of Supper Killer®, Perma Kill‐K®, and Home Keeper® were highly variable depends on the strains.  相似文献

When does dead wood turn into a substrate for spruce replacement?   总被引:1,自引:0,他引:1  
Question: How many years must elapse for freshly fallen Picea abies stems to be transformed into a substrate for P. abies recruitment? Location: Natural sub‐alpine spruce forest, 1200–1300 m a.s.l., western Carpathians, Poland. Methods: Coarse woody debris (CWD) was measured on nine plots with a total area of 4.3 ha. All individuals of P. abies regeneration growing on dead wood were counted and their age was estimated. Decay rate of logs was determined using dendrochronological cross‐dating of samples from logs in different decay stages. Results: Although CWD covered only 4% of the forest floor, 43% of the saplings were growing on decaying logs and stumps. The highest abundance of P. abies recruitment occurs on logs 30–60 years after tree death, when wood is in decay stages no. 4–7 (on an 8 degree decay scale). However, much earlier colonization is possible. The first seedlings may germinate on a log during the second decade after tree death and survive for decades. Their slow growth is possibly due to the gradual progressive decomposition of wood. Conclusions: This study confirms the importance of decaying wood for P. abies recruitment. The decaying logs exhibit continuous and favourable conditions for the germination of P. abies seeds throughout their decay process. Logs, irrespective of their decay stage and age, are colonized by young seedlings. This recruitment bank is constantly renewed.  相似文献

The baculovirus–insect expression system is a popular tool for recombinant protein production. The standard method for infecting insect larvae with recombinant baculovirus for protein production involves either feeding occlusion bodies or injecting budded virus into the cuticle. In this study, we showed that the recombinant Autographa californica multiple nucleopolyhedrovirus (AcMNPV) at titers >108 pfu/mL efficiently infected Trichoplusia ni (T. ni) larvae through aerosol inoculation of budded virus at a pressure of 5.5 × 104 Pa. The dipping T. ni larvae in virus‐containing solution efficiently infected them. These results indicate that surface contamination, either by aerosol or dipping, lead to infection via spiracles. The aerosol infection route for AcMNPV was restricted to T. ni and Plutella xylostella larvae, whereas Spodoptera litura and Helicoverpa armigera larvae were resistant to this inoculation process. The yields of the reporter proteins DsRed and EGFP from T. ni larvae following aerosol infection were nearly identical to those following oral feeding or injection. This alternative baculovirus infection strategy facilitates recombinant protein and virus production by insect larvae. © 2009 American Institute of Chemical Engineers Biotechnol. Poly(3‐hydroxybutyrate) (P(3HB)) and its copolymer poly(3‐hydroxybutyrate‐co‐3‐hydroxyvalerate) (P(3HB‐co‐3HV), are biodegradable thermoplastic polymers. They are members of the polyhydroxyalkanoate (PHA) family, synthesized and accumulated as a carbon and energy reserve by a variety of microorganisms. The aim of this study was to evaluate the use of the proteases Corolase® L10, Alcalase® 2.4L, Corolase® 7089 and Protemax® FC and glycosidases Celumax® BC, Rohament® CL and Rohalase® Barley for the recovery of P(3HB) and P(3HB‐co‐3HV) synthesized by Cupriavidus necator. The enzyme Celumax® BC provided better lysis of the bacterial cell membrane and the results for the optimization of the operating conditions showed that this enzyme is most stable in acetate buffer at pH 4.0, bath at 60°C, hydrolysis time of 1 h and concentration of 0.02% (w/w). The optimization of the operating conditions showed that the enzyme Celumax® BC provided better lysis of the bacterial cell in acetate buffer at pH 4.0, bath at 60°C, hydrolysis time of 1 h and concentration of 0.02% (w/w). These conditions resulted in lysis of the membrane of the bacteria with a recovery of 93.2% P(3HB‐co‐3HV) with 94% purity. The results showed that the use of enzymes for the polymer extraction is an efficient process that assists in the cell disruption of Cupriavidus necator. © 2012 American Institute of Chemical Engineers Biotechnol. Tree regeneration on rotten wood and on soil in old-growth stand   总被引:1,自引:0,他引:1  
Forest regeneration on soil and on decaying wood was studied in natural mixed stand of Facus sylvatica L., Abies alba Mill. and Picea abies Karst. in Babia Góra National Park, Western Carpathians.Downed wood, divided into five decay classes covered around 6% of the forest floor. Among seedlings, Fagus and Abies codominated, while Picea was less numerous. The average seedling density on the soil with herb layer (240 ind./100 m2) was higher than on the logs, even on the strongly decayed ones (177 ind./100 m2). However, the density of Abies and Picea seedlings was higher on the rotten wood than on soil. Seedling survival of all species was better on the logs, especially in conifers. Because of the total dominance of Fagus among saplings, the presence of Abies and Picea in the next generation of canopy trees can strongly depend upon their regeneration on decaying wood.  相似文献
