SYNOPSIS. Eimeria callospermophili was found in 6 species of ground squirrels and the white-tailed prairie dog. The hosts included Spermophilus armatus from Utah and Montana, S. richardsoni from Montana and Wyoming, S. beecheyi from California, S. lateralis and S. variegatus from Utah, and S. tridecemlineatus and Cynomys leucurus from Wyoming. Infections were generally transmissible from each species of ground squirrel to S. armatus and S. richardsoni. Oocysts from C. leucurus caused infections in S. armatus and S. richardsoni. No infections were found after inoculation of E. callospermophili oocysts into least chipmunks (Eutamius minimus), Mongolian gerbils (Meriones unguiculatus), or laboratory rats; however, excystation occurred in these animals. Resistance to infection did not develop in S. armatus, S. richardsoni, or S. variegatus, but did occur after 5 or more infections in S. lateralis. Eimeria callospermophili had little or no effect on the host in S. armatus, S. lateralis, or S. variegatus, but caused bloody diarrhea in severely infected individuals of S. richardsoni. The oocysts had an oocyst residuum consisting of several distinct bodies, which later coalesced to form a large homogeneous body. Each sporozoite had an unusually large refractile body. In experimentally infected specimens of S. armatus the prepatent period and patent period lasted for 5 and 9 days, respectively. Mature 1st-generation schizonts, first seen 2 days after inoculation, had 8–12 merozoites. Mature 2nd-generation schizonts, first seen 3 days after inoculation, had an average of 18 merozoites which were smaller than those of the 1st generation. SYNOPSIS. Cell lines or established cell lines of bovine, ovine or human origin and primary cells from whole embryos of groundsquirrels were used in a study of the in vitro development of Eimeria callospermophili and E. bilamellata from the Uinta ground squirrel, Spermophilus armatus. Monolayers in Leighton tube cultures were inoculated with sporozoites of either of these 2 species and examined with phase-contrast microscopy at various intervals. After such examination, coverslips were fixed in Schaudinn's or Zenker's fluid and variously stained. E. callospermophi sporozoites penetrated cells and underwent development to mature 1st generation schizonts in most cell types. At different times after inoculation, both species formed sporozoite-shaped schizonts, which later became spheroidal. Intracellular movements of sporo zoite-shaped schizonts of E. callospermophili were observed and such schizonts penetrated cells when freed by mechanical disintegration of the host cells. Merozoites were formed at the periphery of the schizont in both species. Mature 1st generation schizonts of E. callospermophili, with 6–14 merozoites, were first seen 15 hr after inoculation; the corresponding values for E. bilamellata were 12–27 merozoites and 4 days. Merozoites of both had anterior and posterior refractile bodies. Exposure to a trypsin-bile solution stimulated motility in merozoites of E. callospermophili. Second generation trophozoites and immature schizonts of E. callospermophili were seen in cultures of primary cells of whole ground-squirrel embryos 20–24 hr and 44–48 hr, respectively, after inoculation of sporozoites. 相似文献

This coccidium could not be experimentally transmitted to S. richardsoni; however, patent infections were established in S. armatus, S. lateralis, and S. variegatus. No infections were found after inoculation of least chipmunks (Eutamius minimus), Mongolian gerbils (Meriones unguiculatus), or laboratory rats even tho excystation occurred in these animals. Resistance to infection did not develop during repeated experimental infections of S. armatus, S. lateralis, or S. variegatus. No outward signs of coccidiosis were seen in any of the experimentally infected animals. In experimentally infected S. armatus, the prepatent period was 5 days, and the patent period lasted 3–7 (mean 6.5) days. The endogenous stages were located in the epithelial cells of the jejunum and ileum. Mature 1st-generation schizonts, 1st seen 2.5 days after inoculation, contained 16–32 merozoites. Mature 2nd generation schizonts were present 3.5 days after inoculation and contained 22–46 merozoites of a larger size than those of the 1st generation. Gametocytes were 1st seen 3.5 days after inoculation and developing oocysts were present 4 days after inoculation. Macrogametes contained eosinophilic granules which coalesced to form the oocyst wall. Formation of the macrogametes took place around cytoplasmic masses within the microgametocytes. 相似文献

Oocysts of Eimeria morainensis n. sp. are described from the golden-mantled ground squirrel, Spermophilus lateralis. in Northern Colorado. The oocysts of E. morainensis are double-walled and subspherical, 20.3 × 19.8 (18.7–26.2 × 17.5–21.2) μm; and the sporocysts are ellipsoid, 12.1 × 6.9 (8.7–13.7 × 6.2–8.7) μm. Oocyst residuum and micropyle are absent, but a polar granule is present. Sporocyst residuum and Stieda body are present. Differences in oocyst characteristics provide the basis for recognition of this new species of Eimeria. 相似文献

SYNOPSIS. Stages in the endogenous cycle of Eimeria confusa from the grey squirrel, Sciurus carolinensis, are described from mixed infections with another species, Eimeria lancasterensis. All corresponding stages were markedly different in the 2 species. In E. confusa infections, the parasites were located below the host cell nuclei of the epithelial cells of the villi of the jejunum and ileum. Mature schizonts were ellipsoidal, averaged 20.9 × 18.6 μm and had 18–30 merozoites. The mature microgamonts measured 34.3 × 24.7 μm and had hundreds of microgametes. Fecal samples of 36 ground squirrels, Spermophilus beldingi, from Tioga Pass (elev. ca. 3315 m) in the Sierra Nevada, California, yielded oocysts of Eimeria beckeri in nine squirrels, E. citelli in four squirrels, E. beldingii n. sp. in two squirrels, and degenerated, unidentifiable oocysts in ten squirrels. Eimeria beldingii n. sp. oocysts are ellipsoidal, 30–34 × 24–30 (mean 32 × 26) μm with a two-layered, rough, striated wall, without a micropyle or residuum, with polar granules; they contain ellipsoidal or ovoid sporocysts 11–15 × 9–12 (mean 13 × 10) μm with a Stieda body and residuum. 相似文献

The range of the Uinta ground squirrel, Spermophilus armatus , is centred over one of the most tectonically active regions today, the Yellowstone hotspot. We document the role of Quaternary tectonic and climatic history on the genetic structure of this species by screening museum and extant individuals throughout its range. Phylogeographic, divergence time, and demographic analyses of partial mitochondrial cytochrome b and control region DNA sequences yield insight into the cadence of evolution across three spatiotemporal scales: (i) a relatively deep intraspecific divergence of S. armatus into three lineages coincident with the last major volcanic eruption in the region and maintained by the Snake River Plain; (ii) demographic expansion in two lineages corresponding to the time of last deglaciation of the region; and (iii) a recent (< 50 years) local extinction of the third lineage coincident with climatic change and conversion of habitat for agricultural purposes in eastern Idaho. Beyond these inferences, our study highlights the unique value of museum material to phylogeography, and shows that small mammal recolonization of previously glaciated montane 'islands' differs from northward postglacial expansion observed in areas previously covered by continental ice sheets. Montane 'islands' may harbour high genetic diversity because of admixture and recurrent expansion/extinction. 相似文献

Background
Echinococcus granulosus is usually transmitted between canid definitive hosts and ungulate intermediate hosts.Methodology/Principal Findings
Lesions found in the livers of ground squirrels, Spermophilus dauricus/alashanicus, trapped in Ningxia Hui Autonomous Region, an area in China co-endemic for both E. granulosus and E. multilocularis, were subjected to molecular genotyping for Echinococcus spp. DNA. One of the lesions was shown to be caused by E. granulosus and subsequently by histology to contain viable protoscoleces.Conclusions/Significance
This is the first report of a natural infection of the ground squirrel with E. granulosus. This does not provide definitive proof of a cycle involving ground squirrels and dogs or foxes, but it is clear that there is active E. granulosus transmission occurring in this area, despite a recent past decline in the dog population in southern Ningxia. 相似文献12.

Synopsis. Oocysts of Eimeria malabaricas sp. n. and Eimeria bandipurensis from the South Indian tree squirrel Funambulus tristriatus collected in Kerala, India, are described. Sporulated oocysts of E. malabaricas were ellipsoid to subspherical measuring 39.8 (35–45) × 32.1 (29–37) m?m, with a thick (2.5–3.0 m?m), 2-layered wall. The outer layer was yellow-brown, striated, and rough. There was no micropyle. but a polar granule was present in 34% of oocysts. The sporocysts were ovoid, 16.0 (14.0–18.0) × 11.2 (11.0–12.0) m?m, with a Stieda body and a granular residuum. Excysted sporozoites were 21.8 (19.0–23.0) × 3.4 (3.0–4.0) m?m, with a large refractile body. The sporulated oocysts of E. bandipurensis are redescribed. 相似文献

SYNOPSIS. The life cycle of a turkey strain of Eimeria dispersa Tyzzer was studied in Beltsville Small White turkeys. There were 4 asexual generations. Mature schizonts of the first generation were present 30 h postinoculation (PI); those of the 2nd, 3rd, and 4th generations were present 48, 72, and 96 h PI, respectively. Average size of schizonts and number and size of merozoites for each generation were as follows: first , 14.3 × 13.0 μm with 19.2 merozoites, each 4.5 × 1.2 μm; second , 8.0 × 7.2 μm with 13.5 merozoites, each 4.5 × 1.1 μm; third , 8.9 × 8.9 μm with 15.1 merozoites, each 5.6 × 2.1 μm; fourth , 11.6 × 10.5 μm with 6.7 merozoites, each 8.2 × 2.0 μm. Sporozoites and developmental stages of the first generation were in close association with an epithelial cell nucleus and located between the brush border and the "row" of epithelial cell nuclei; developmental stages of the other 3 generations were not associated with a nucleus and were located just under the brush border. Early macrogametes and microgametocytes were present 96 h PI. Development was confined to the epithelial cells of the villus and extended from the tip of the villus to ∼ 1/2 the distance down the sides in all areas of the intestine except the cecum. The prepatent period was between 114 and 120 h. Percentage of sporulation was 15, 57, and 90, at 24, 36, and 48 h, respectively. Sporulated oocysts averaged 24.5 × 20.2 μm. 相似文献

SYNOPSIS. A pure strain of Eimeria debliecki (University of Illinois strain A) established from a single oocyst was used to determine the endogenous cycle. Young parasite-free pigs 2 weeks to 3 months old were used throughout the study. The endogenous cycle was found to take place in the small intestine where the parasites were located in the distal portion of the striated simple columnar epithelial cells of the villi. The first generation schizonts were found in only the jejunum (15% of small intestine). The second generation schizonts and gametes occurred in the jejunum and ileum (70% of small intestine), a slight posterior progression occurring with each stage. The entire cycle required 6.5 days. The schizogonous cycle comprised 2 generations. The first generation schizonts required 2.5 days to reach maturity, measured 8-12 μ, contained 16 merozoites measuring 12-15 μ and had a polar residual mass. The second generation schizonts required 2 days to reach maturity, measured 13-16 μ, contained 32 rotund merozoites measuring 6–8 μ, and had only a few granules of residual material. Gametogony took place in 1.5 days. The macrogametes measured 12-16 μ, and the microgametocytes measured 9-14 μ with microgametes measuring 5–6 μ. 相似文献

Ten-day-old broiler chickens were inoculated with oocysts of a characterized strain of Eimeria mitis , and tissues were fixed at 4, 8, or 24-h intervals after inoculation for histopathological examination. Tissue collections were initiated at the time of inoculation and extended up to 168 h postinoculation. The preferred site of development of E. mitis was found to be the ileum although more limited development of the parasite also took in the jejunum, cecal pouches, cloaca, and bursa of Fabricius. No distinctive and consistent intestinal lesions were macroscopically evident even in heavily parasitized chickens. The prepatent period was approximately 92 h postinoculation. The histopathological features of the E. mitis infections were characterized using conventional bright-field microscopy as well as both scanning and transmission electron microscopy. SYNOPSIS. Forty grey squirrels Sciurus carolinensis were examined for coccidia during a 2-year period. Eimeria lancasterensis was found in all of them. The ellipsoidal oocysts of this species averaged 24.6 by 14.6 μ. They had no micropyle or oocyst residuum. A polar body was present. The sporocysts averaged 14.1 by 8.4 μ. The endogenous phases of the parasite were found in the epithelial cells of the villi thru the entire length of the small intestine. E. confusa was found in one of 40 squirrels. The oocysts of this species were subspherical, occasionally ellipsoidal or rarely spherical; they averaged 33.2 by 26.7 μ. Oocyst residuum and micropyle were absent. Polar granules ranged in number from 0–5. The sporocysts averaged 19.6 by 12.1 μ. The prepatent period for this species was 7–8 days and the patent period 6–13 days. An ultrastructural study of life cycle stages of Pneumocystis carinii in infected rat lungs in situ was undertaken utilizing 8 different modes of fixation. Three of the fixatives employed gave good fixation of cysts and intracystic bodies, but for the trophic forms fixation was only fair. Both the trophic forms and intracystic bodies have nuclear pores. The mitochondria of the organism have cristac that appear lamellar. One of the fixation modes revealed a thin, electron-dense layer on the outer surface of the cell wall, a "fuzzy coat" that had not been described previously. This material appears to mediate tight adhesion of trophic forms with other trophic forms, cysts, and with pneumocytcs of the lung alveolus. 相似文献
