5α-Cholest-8(14)-en-3β-ol-15-one, at a level of 0.1% in a low cholesterol diet has been shown to have a profound hypocholesterolemic effect in rats. In one experiment the mean serum cholesterol level (mg per 100 ml ± S.E.M.) decreased from 71.2 ± 0.9 to 36.9 ± 3.3 after 7 days on the diet. In a second experiment the mean serum cholesterol value decreased from 86.4 ± 1.2 to 33.4 ± 3.9 after 8 days on the ketone-containing diet. The effects of the 15-ketosterol on serum cholesterol levels were significantly (p<0.001) different from those of eitheir ad libidum or pair-fed controls. The diet containing the ketone caused a significant decrease in food consumption and a slight decrease in body weight (10.2% and 3.1% in the two experiments). The ketone, at a level of 0.2% in the diet, caused a marked decrease in plasma cholesterol levels in mice. After 8 days of treatment the plasma cholesterol value (mg per 100 ml ± S.E.M.) of the treated mice was 35.8 ± 3.6 while that of pair-fed control animals was 114.4 ± 3.4.  相似文献

Understanding how pathogens and parasites diversify through time and space is fundamental to predicting emerging infectious diseases. Here, we use biogeographic, coevolutionary and phylogenetic analyses to describe the origin, diversity, and distribution of avian malaria parasites in the most diverse avifauna on Earth. We first performed phylogenetic analyses using the mitochondrial cytochrome b (cyt b) gene to determine relationships among parasite lineages. Then, we estimated divergence times and reconstructed ancestral areas to uncover how landscape evolution has shaped the diversification of Parahaemoproteus and Plasmodium in Amazonia. Finally, we assessed the coevolutionary patterns of diversification in this host–parasite system to determine how coevolution may have influenced the contemporary diversity of avian malaria parasites and their distribution among Amazonian birds. Biogeographic analysis of 324 haemosporidian parasite lineages recovered from 4178 individual birds provided strong evidence that these parasites readily disperse across major Amazonian rivers and this has occurred with increasing frequency over the last five million years. We also recovered many duplication events within areas of endemism in Amazonia. Cophylogenetic analyses of these blood parasites and their avian hosts support a diversification history dominated by host switching. The ability of avian malaria parasites to disperse geographically and shift among avian hosts has played a major role in their radiation and has shaped the current distribution and diversity of these parasites across Amazonia.  相似文献

Like many parasites, avian haematozoa are often found at lower infection intensities in older birds than young birds. One explanation, known as the “selection” hypothesis, is that infected young birds die before reaching adulthood, thus removing the highest infection intensities from the host population. We tested this hypothesis in the field by experimentally infecting nestling rock pigeons (Columba livia) with the malaria parasite Haemoproteus columbae. We compared the condition and fledging success of infected nestlings to that of uninfected controls. There was no significant difference in the body mass, fledging success, age at fledging, or post-fledging survival of experimental versus control birds. These results were unexpected, given that long-term studies of older pigeons have demonstrated chronic effects of H. columbae. We conclude that H. columbae has little impact on nestling pigeons, even when they are directly infected with the parasite. Our study provides no support for the selection hypothesis that older birds have lower parasite loads because parasites are removed from the population by infected nestlings dying. To our knowledge, this is the first study to test the impact of avian malaria using experimental inoculations under natural conditions.  相似文献

We examined seasonal prevalence in avian haemosporidians (Plasmodium and Haemoproteus) in migrant and resident birds in western Himalaya, India. We investigated how infection with haemosporidians in avian hosts is associated with temporal changes in temperature and mosquito abundance along with host abundance and life‐history traits (body mass). Using molecular methods for parasite detection and sequencing partial cytochrome b gene, 12 Plasmodium and 27 Haemoproteus lineages were isolated. Our 1‐year study from December 2008 to December 2009 in tropical Himalayan foothills revealed a lack of seasonal variation in Plasmodium spp. prevalence in birds despite a strong correlation between mosquito abundance and temperature. The probability of infection with Plasmodium decreased with increase in temperature. Total parasite prevalence and specifically Plasmodium prevalence showed an increase with average avian body mass. In addition, total prevalence exhibited a U‐shaped relationship with avian host abundance. There was no difference in prevalence of Plasmodium spp. or Haemoproteus spp. across altitudes; parasite prevalence in high‐altitude locations was mainly driven by the seasonal migrants. One Haemoproteus lineage showed cross‐species infections between migrant and resident birds. This is the first molecular study in the tropical Himalayan bird community that emphasizes the importance of studying seasonal variation in parasite prevalence. Our study provides a basis for further evolutionary study on the epidemiology of avian malaria and spread of disease across Himalayan bird communities, which may not have been exposed to vectors and parasites throughout the year, with consequential implications to the risk of infection to naïve resident birds in high altitude.  相似文献

A method for assaying cortisol and cortisone using chromatography on either paper or Sephadex LH-20 columns for isolation, followed by competitive protein binding, has been applied to umbilical cord and maternal plasma samples. In mixed cord plasma the mean cortisol concentration was 6.0 ± 0.8 μg/100 ml (n = 9) and the mean cortisone concentration was 13.5 ± 2.9 μg/100 ml (n = 9). In cord arterial plasma the mean cortisol concentration was 6.3 ± 2.9 μg/100 ml (n = 6) and the mean cortisone level was 10.1 ± 2.5 μg/100 ml (n = 6). For cord venous plasma, the mean level of cortisol was 5.6 ± 1.5 μg/100 ml (n = 6) and of cortisone was 13.5 ± 2.4 μg/100 ml (n = 6). Maternal plasma gave a mean value of cortisol of 42.3 ± 4.5 μg/100 ml (n = 6) and of cortisone of 6.2 ± 0.9 μg/100 ml. The results of this study suggest that the fetus at term-gestation produces cortisol. The significance of this production compared with placental transfer of maternal cortisol into the fetal circulation however is uncertain.  相似文献

Keratinophilic fungi were isolated from feathers of most common Indian birds,viz. domestic chicken (Gallus domesticus), domestic pigeon (Columba livia), house sparrow (Passer domesticus), house crow (Corvus splendens), duck (Anas sp.), rose-ringed parakeet (Psittacula krameri). Out of 87 birds, 58 yielded 4 keratinophilic fungal genera representing 13 fungal species and one sterile mycelium. The isolated fungi were cultured on Sabouraud's dextrose agar at 28±2°C.Chrysosporium species were isolated on most of the birds.Chrysosporium lucknowense andChrysosporium tropicum were the most common fungal species associated with these Indian birds. Maximum occurrence of fungi (47%) was recorded on domestic chickens and the least number of keratinophilic fungi was isolated from the domestic pigeon and duck. The average number of fungi per bird was found to be the 0.44.  相似文献

Malaria caused by Plasmodium parasites is one of the worst scourges of mankind and threatens wild animal populations. Therefore, identifying mechanisms that mediate the spread of the disease is crucial for both human health and conservation. Human‐induced climate change has been hypothesized to alter the geographic distribution of malaria pathogens. As the earth warms, arthropod vectors may display a general range expansion or may enjoy longer breeding season, both of which can enhance parasite transmission. Moreover, Plasmodium species may directly benefit for elevating temperatures, which provide stimulating conditions for parasite reproduction. To test for the link between climate change and malaria prevalence on a global scale for the first time, I used long‐term records on avian malaria, which is a key model for studying the dynamics of naturally occurring malarial infections. Following the variation in parasite prevalence in more than 3000 bird species over seven decades, I show that the infection rate by Plasmodium is strongly associated with temperature anomalies and has been augmented with accelerating tendency during the last 20 years. The impact of climate change on malaria prevalence varies across continents, with the strongest effects found for Europe and Africa. Migration habit did not predict susceptibility to the escalating parasite pressure by Plasmodium. Consequently, wild birds are at an increasing risk of malaria infection due to recent climate change, which can endanger both naïve bird populations and domesticated animals. The prevailing avian example may provide useful lessons for understanding the effect of climate change on malaria in humans.  相似文献

Abstract

The specific binding of lactoperoxidase-labelled 125I-labelled ovine prolactin was determined in a membrane particulate of the pigeon crop-sac mucosal epithelium. Binding was found to be dependent upon the particulate preparation used, its protein concentration and the length of the incubation at 5°C. Scatchard analysis of the binding to crop-sacs from saline or prolactin-injected (1.9 μg per pigeon) revealed that prolactin stimulated 7-fold its own receptors by increasing the number of binding sites per mg protein: saline - 392±75 fmol/mg protein and prolactin 2736±602 fmol/mg protein (p<0.01). This increase did not affect the affinity constant (Ka): saline - 5.28±0.75x108 l/mol and prolactin-3.28±0.40x108 l/mol (N.S.), in keeping with the stimulatory effect of prolactin in the rat liver and mammary gland. This study further demonstrates the physiological role of endogenous prolactin in maintaining its own binding-sites in the pigeon crop-sac, since the administration of 0.8 ml anti-serum to prolactin resulted in a 63% reduction in the specific binding of the labelled hormone in vitro. These results confirm the prolactin binding to the pigeon crop-sac mucosa, quantify the stimulation of this binding by prolactin itself, and demonstrate the role of the endogenous hormone in the maintenance of these receptors.  相似文献

The range of hosts a pathogen infects (host specificity) is a key element of disease risk that may be influenced by both shared phylogenetic history and shared ecological attributes of prospective hosts. Phylospecificity indices quantify host specificity in terms of host relatedness, but can fail to capture ecological attributes that increase susceptibility. For instance, similarity in habitat niche may expose phylogenetically unrelated host species to similar pathogen assemblages. Using a recently proposed method that integrates multiple distances, we assess the relative contributions of host phylogenetic and functional distances to pathogen host specificity (functional–phylogenetic host specificity). We apply this index to a data set of avian malaria parasite (Plasmodium and Haemoproteus spp.) infections from Melanesian birds to show that multihost parasites generally use hosts that are closely related, not hosts with similar habitat niches. We also show that host community phylogenetic ß‐diversity (Pßd) predicts parasite Pßd and that individual host species carry phylogenetically clustered Haemoproteus parasite assemblages. Our findings were robust to phylogenetic uncertainty, and suggest that phylogenetic ancestry of both hosts and parasites plays important roles in driving avian malaria host specificity and community assembly. However, restricting host specificity analyses to either recent or historical timescales identified notable exceptions, including a ‘habitat specialist’ parasite that infects a diversity of unrelated host species with similar habitat niches. This work highlights that integrating ecological and phylogenetic distances provides a powerful approach to better understand drivers of pathogen host specificity and community assembly.  相似文献

SYNOPSIS. Cytoplasmic polyribosomes were isolated from the avian malaria parasite Plasmodium lophurae by lysis with 0.15% Triton X-100 followed by high speed centrifugation through a discontinuous sucrose gradient. Polyribosomes were protected from nuclease degradation using 100 μg/ml heparin or 50 μg/ml dextran sulfate. Cell-free incorporation of radioisotope-labeled amino acids required a pH 5 fraction (duck reticulocyte), Mg2+, and an energy-generating system. The protein synthesizing system was stimulated by the addition of polyuridylic acid. Optimum conditions for protein synthesis by the plasmodial system are described. The effects of drugs on the cell-free protein synthesizing system using duck reticulocyte and plasmodial ribosomes are reported.  相似文献

The role of ovarian steroids in the preimplantation pig embryo was studied in vivo and in vitro. Twenty gilts were treated three times daily on days 1 to 4 after insemination with either 25, 100, 250, or 1000 mg progesterone in oil, and 17 gilts were injected with corresponding amounts of sesame oil (controls). All gilts were slaughtered 5 days after insemination and the embryos were recovered. Oviduct and plasma progesterone content were significantly (P<0.001) higher in gilts treated with 750 mg of exogenous progesterone per day. After 750 mg progesterone, oviduct progesterone content was twice as high as control levels, while after 3000 mg progesterone per day the levels in oviduct and uterus exceeded those of controls by five and seven times, respectively. In gilts treated with 750 mg progesterone per day, plasma progesterone levels were 177.4 ± 22.1 ng/ml (x ± SD) on day 3 and 186.4 ± 69.2 ng/ml on day 5 and resembled values found in superovulated pigs with more than 40 ovulations. Excessive plasma progesterone values of 1014.6 ± 840.4 ng/ml on day 3 and 473.2 ± 197.2 ng/ml on day 5 were found after treatment with 3000 mg of progesterone per day. Treatment with up to 750 mg of exogenous progesterone per day, did not affect embryonic development, but 3000 mg per day resulted in a significantly (P<0.001) higher percentage of retarded and degenerate embryos compared to controls (71.8% versus 3.2%).In addition, the amount and specificity of uptake of 3H-labelled progesterone and estradiol-17 beta by pig blastocysts recovered from superovulated gilts were investigated after 6 hrs in vitro culture. The uptake of 3H-progesterone was 131.9 ± 56.9 counts per million (cpm) per 10 blastocysts, corresponding to 1.1 fmoles progesterone. The uptake was non-specific for it was only slightly reduced in the presence of a 100-fold excess of unlabelled progesterone (20.1%) or estradiol-17 beta (27.0%). The uptake of 3H-estradiol-17 beta was 133.9 ± 74.12 cpm per 10 blastocysts, corresponding to 1.3 fmoles estradiol-17 beta. The uptake was significantly (P<0.01) reduced by 67.7% in the presence of a 100-fold excess of unlabelled estradiol-17 beta. Apparent specific binding was 0.87 fmoles estradiol-17 beta per 10 blastocysts or 72.5 fmoles estradiol-17 beta per mg protein. The uptake was only slightly reduced in the presence of a 100-fold excess of unlabelled progesterone (23.3%). This significant inhibition could be determined after 2 hrs in vitro culture. There was no competitive inhibition after 20 min. of culture.Uptake by unfertilized ova and degenerate embryos recovered on day 5 was significantly smaller (51.8 ± 10.3 cpm per 10 eggs; P<0.001) than by blastocysts recovered on the same day. No competitive inhibition could then be determined. In vivo, preimplantation pig embryos seem to be rather insensitive to high progesterone levels. Excessive amounts of progesterone probably can be metabolized to a great extent. Progesterone seems to be taken up rather non-specifically by the pig embryo. The uptake and binding of estradiol-17 beta seems to be more specific. Studies are in progress to investigate the physiological role of estradiol-17 beta uptake in early embryonic development.  相似文献

Capsule The reproductive rate of Rose-ringed Parakeets in the UK was higher in 2001-2003 than previously estimated.

Aim To measure reproductive rate and the factors affecting this in Rose-ringed Parakeets in England during a period of rapid expansion in population of this potential invasive species.

Methods During 2001–2003, 108 nests were located and monitored in southwest London, southeast London and the Isle of Thanet. Nest survival using the Mayfield method, fledging success and the characteristics of the nest and surrounding vegetation were measured.

Results The mean date of first egg was 26 March?±?1.3 days and the median clutch size was 4 eggs. Mayfield nest survival rate was 72%. Reproductive success was 1.4?±?0.3 young fledged per nest. Parakeets bred predominantly in ash (Fraxinus) and oak (Quercus). Nest trees had a diameter at breast height (DBH) of 73.7?±?4.7?cm (mean?±?se).

Conclusion Reproductive success in this parakeet was higher than previously estimated (0.8 young fledged per nest prior to 1999). This reproductive output is sufficient to explain the rapid increase in Rose-ringed Parakeet numbers (from an estimated 1500 birds in 1996 to nearly 10,000 birds by 2004) if Rose-ringed Parakeets have a low death rate similar to other parrots.  相似文献

The epidemiology of vector‐borne pathogens is largely determined by the host‐choice behaviour of their vectors. Here, we investigate whether a Plasmodium infection renders the host more attractive to host‐seeking mosquitoes. For this purpose, we work on a novel experimental system: the avian malaria parasite Plasmodium relictum, and its natural vector, the mosquito Culex pipiens. We provide uninfected mosquitoes with a choice between an uninfected bird and a bird undergoing either an acute or a chronic Plasmodium infection. Mosquito choice is assessed by microsatellite typing of the ingested blood. We show that chronically infected birds attract significantly more vectors than either uninfected or acutely infected birds. Our results suggest that malaria parasites manipulate the behaviour of uninfected vectors to increase their transmission. We discuss the underlying mechanisms driving this behavioural manipulation, as well as the broader implications of these effects for the epidemiology of malaria.  相似文献

Avian malaria parasites (Plasmodium) occur commonly in wild birds and are an increasingly popular model system for understanding host–parasite co‐evolution. However, whether these parasites have fitness consequences for hosts in endemic areas is much debated, particularly since wild‐caught individuals almost always harbour chronic infections of very low parasite density. We used the anti‐malarial drug MalaroneTM to test experimentally for fitness effects of chronic malaria infection in a wild population of breeding blue tits (Cyanistes caeruleus). Medication caused a pronounced reduction in Plasmodium infection intensity, usually resulting in complete clearance of these parasites from the blood, as revealed by quantitative PCR. Positive effects of medication on malaria‐infected birds were found at multiple stages during breeding, with medicated females showing higher hatching success, provisioning rates and fledging success compared to controls. Most strikingly, we found that treatment of maternal malaria infections strongly altered within‐family differences, with reduced inequality in hatching probability and fledging mass within broods reared by medicated females. These within‐brood effects appear to explain higher fledging success among medicated females and are consistent with a model of parental optimism in which smaller (marginal) offspring can be successfully raised to independence if additional resources become available during the breeding attempt. Overall, these results demonstrate that chronic avian malaria infections, far from being benign, can have significant effects on host fitness and may thus constitute an important selection pressure in wild bird populations.  相似文献

A technique using 51chromium‐labelled erythrocytes was used to measure blood volume in Scyliorhinus canicula following long‐term and acute salinity transfers. Basal whole‐blood volume was 5·6 ± 0·2 ml 100 g?1 (mean ±s .e .), this increased (6·3 ± 0·2 ml 100 g?1) following +14 day acclimation to 80% sea water (SW) and decreased (4·6 ± 0·2 ml 100 g?1) following acclimation to 120% SW. These changes were shown to be primarily due to changes in plasma volume, with no significant changes in extrapolated red‐cell volume being demonstrated. Blood volume was also measured in the same animals during 10 h acute transfer to 100% SW. Plasma volume in S. canicula during acclimation from 80% SW was significantly reduced (4·5 ± 0·3 ml 100 g?1) after 6 h of transfer to 100% SW. Blood volume in animals during acclimation from 120% SW was significantly increased (4·8 ± 0·2 ml 100 g?1) after 4 h of acute transfer. The osmoregulatory implications of these different timeframes during hyposaline and hypersaline transfer are discussed, along with the importance of this in vivo technique as context for in vitro studies with haemo‐dynamic stimuli.  相似文献

The role of introduced avian malaria Plasmodium relictum in the decline and extinction of native Hawaiian forest birds has become a classic example of the potential effect of invasive diseases on biological diversity of naïve populations. However, empirical evidence describing the impact of avian malaria on fitness of Hawai‵i's endemic forest birds is limited, making it difficult to determine the importance of disease among the suite of potential limiting factors affecting the distribution and abundance of this threatened avifauna. We combined epidemiological force‐of‐infection with multistate capture––recapture models to evaluate a 7‐year longitudinal study of avian malaria in ‵apapane, a relatively common native honeycreeper within mid‐elevation Hawaiian forests. We found that malaria transmission was seasonal in this mid‐elevation forest; transmission peaked during fall and during some years produced epizootic mortality events. Estimated annual mortality of hatch‐year birds typically exceeded 50% and mortality of adults exceeded 25% during epizootics. The substantial impact of avian malaria on this relatively common native species demonstrates the key role this disease has played in the decline and extinction of Hawaiian forest birds.  相似文献

Levels of norepinephrine (NE) in human plasma have been determined by a radioenzymatic technique sufficiently sensitive to measure 0.014 ng NE per ml plasma. Several procedures which raise plasma NE levels have been compared and a standard procedure developed to evaluate sympathetic neuronal function based on the increments in plasma NE produced by postural change and a standard amount of exertion. The mean basal level of NE in plasma of 74 resting, supine, normal subjects ranging in age from 10 to 70 (mean 32.7 years) was 0.292 ± 0.016 (± SEM) ng/ml and ranged from 0.112 to 0.738 ng/ml. There was a significant correlation between age and basal levels of NE (L.R. = 0.33, p < 0.01). In 44 subjects who stood for 5 minutes after the basal sample of blood was obtained, the mean plasma level of NE increased to 0.538 ± 0.044 ng/ml and further increased to 0.778 ± 0.080 ng/ml after a subsequent isometric hand grip for 5 minutes.  相似文献

Haemosporidians causing avian malaria are very common parasites among bird species. Their negative effects have been repeatedly reported in terms of deterioration in survival prospects or reproductive success. However, a positive association between blood parasites and avian fitness has also been reported. Here, we studied a relationship between presence of malaria parasites and reproductive performance of the host, a hole‐breeding passerine – the blue tit Cyanistes caeruleus. Since the malaria parasites might affect their hosts differently depending on environmental conditions, we performed brood size manipulation experiment to differentiate parental reproductive effort and study the potential interaction between infection status and brood rearing conditions on reproductive performance. We found individuals infected with malaria parasites to breed later in the season in comparison with uninfected birds, but no differences were detected in clutch size. Interestingly, infected parents produced heavier and larger offspring with stronger reaction to phytohemagglutinin. More importantly, we found a significant interaction between infection status and brood size manipulation in offspring tarsus length and reaction to phytohemagglutinin: presence of parasites had stronger positive effect among birds caring for experimentally enlarged broods. Our results might be interpreted either in the light of the parasite‐mediated selection or terminal investment hypothesis.  相似文献

The reduced form of ubiquinone-10 (coenzyme Q) has been shown to represent an important physiologic antioxidant principle in human blood. In order to establish a reference range for infants, we measured plasma levels of ubiquinone in 50 healthy European children aged 2 months to 15 years. A mean ±SD) value of 0.75±0.27 μg/ml plasma (0.87±0.31 μM) was determined; ubiquinone concentrations were not found to be sex-dependent (0.7±0.24μg/ml for girls, n=17, and 0.7±0.28μg/ml for boys, n=33) but correlated negatively with age (r = -0.37, P=0.0075). This negative correlation was mainly due to relatively high levels in infants approximately 1 year old.

The mean value determined does not significantly differ from the average ubiquinone plasma concentrations determined in healthy Nigerian children (0.85±0.40 μg/ml, n= 18) in a previous study (Becker K, Boetticher D, Leichsenring M. Internat J Vitam Nutr Res 1995, in press).  相似文献

Parasite range expansions are a direct consequence of globalization and are an increasing threat to biodiversity. Here, we report a recent range expansion of the SGS1 strain of a highly invasive parasite, Plasmodium relictum, to two non-migratory passerines in North America. Plasmodium relictum is considered one of the world''s most invasive parasites and causes the disease avian malaria: this is the first reported case of SGS1 in wild non-migratory birds on the continent. Using a long-term database where researchers report avian malaria parasite infections, we summarized our current understanding of the geographical range of SGS1 and its known hosts. We also identified the most likely geographical region of this introduction event using the MSP1 allele. We hypothesize that this introduction resulted from movements of captive birds and subsequent spillover to native bird populations, via the presence of competent vectors and ecological fitting. Further work should be conducted to determine the extent to which SGS1 has spread following its introduction in North America.  相似文献
