Background

Chromoblastomycosis is a chronic, progressive fungal disease of the skin and subcutaneous tissue caused by a unique group of dematiaceous fungi. Fonsecaea monophora, a new species distinct from Fonsecaea pedrosoi strains, is the main pathogen responsible for chromoblastomycosis in south China. Macrophages can be polarized into two categories: classically activated and alternatively activated.

Objectives

Little is known about the relationship between F. monophora and macrophage polarization. This study aimed to study the effect of F. monophora on the polarization of THP-1 cells to macrophages.

Methods

We established coculture systems of F. monophora and THP-1-derived macrophages in different activation states.

Results

F. monophora enhanced the phagocytosis by macrophages in the initially activated state and weakened the phagocytosis by classically activated macrophages without affecting that by alternatively activated macrophages. Classically activated macrophages had the strongest killing effect on F. monophora, while the initially activated macrophages had the weakest. The pathogen could not be rapidly cleared by any type of macrophage. F. monophora promoted the expression of proinflammatory cytokines and inhibited that of anti-inflammatory cytokines.

Conclusions

F. monophora promoted the polarization of THP-1 cells to classically activated macrophages and inhibited that of THP-1 cells to alternatively activated macrophages.

Abstract

Context: Garcinia hombroniana Pierre, known as manggis hutan in Malaysia is a rich source of xanthones and benzophenones.

Objectives: This study was aimed to isolate and characterize potential cholinesterase inhibitors from the extracts of G. hombroniana bark and investigate their interactions with the enzymes.

Materials and methods: The dichloromethane extract afforded five triterpenoids which were characterized by NMR and mass spectral techniques. Cholinesterase inhibitory assay and molecular docking were performed to get insight of the inhibitory activity and molecular interactions of the compounds. The compounds were also tested for their antioxidant capacity.

Results: The isolated triterpenoids were identified as: 2β-hydroxy-3α-O-caffeoyltaraxar-14-en-28-oic acid (1), taraxerol (2), taraxerone (3), betulin (4) and betulinic acid (5). Compound 1 was the most active dual inhibitor of both AChE and BChE. Compound 1 also showed good antioxidant activities.

Conclusion: Compound 1 had dual and moderate inhibitory activity on AChE and BChE worthy for further investigations.  相似文献

Aims

Species within the Caulobacter genus have been termed ‘hub species’ in the plant microbiome. To understand these interactions, we assessed the interactions between several Caulobacter strains and a common host plant.

Methods

We identified a set of 11 Caulobacter strains that range in genetic diversity and tested them for their ability to increase the growth of Arabidopsis thaliana. In addition, biochemical assays were employed to determine if these Caulobacter strains produce common plant growth promoting (PGP) biosynthates. To identify potential PGP-related genes, genomic analyses were performed to compare the genomes of PGP Caulobacter strains to those of non-PGP Caulobacter strains.

Results

For the PGP Caulobacter strains, we observed that common PGP biosynthates did not contribute to the observed Caulobacter-mediated plant growth stimulation. Genomic analyses suggested that the genomes of PGP strains maintain similar metabolic pathways compared to those of non-PGP strains, and that common genes related to PGP factors do not explain the PGP mechanisms for the Caulobacter strains we analyzed.

Conclusions

Plant growth enhancement is not a conserved feature in the Caulobacter genus, and some Caulobacter strains even inhibit plant growth. Moreover, common PGP factors do not fully explain Caulobacter-mediated plant growth enhancement.

Aim

The secretion of allelochemicals from plant roots plays a key role in soil sickness and soil-borne disease. The goal of this study was to investigate the role of allelopathic chemicals in Ralstonia solanacearum-infected tobacco roots.

Methods

The organic acids investigated in the present study are major components of tobacco root exudates. Through a swarming assay, we assessed the chemotaxis and colonization of R. solanacearum in response to organic acids.

Results

Fumaric acid was detected, and the results showed that this acid could serve as a semiochemical for attracting R. solanacearum and inducing the formation of biofilms of this species. The results also revealed that cinnamic and myristic acids play significant roles on swarming motility and chemotaxis. In addition, cinnamic, myristic and fumaric acids could enhance the expression of chemotaxis- and motility-related genes in R. solanacearum cultured in minimal medium. Furthermore, these three acids promote R. solanacearum colonization and accelerate disease progression in tobacco.

Conclusion

Cinnamic, myristic and fumaric acids could serve as semiochemical attractants to induce the colonization and infection of R. solanacearum. The results of the present study enhance our understanding of the ecological effects of plant root exudates in plant-microbe interactions and help to reveal the relationship between tobacco bacterial wilt and the autotoxins and allelochemicals that accumulate from root exudates.

Introduction: Oxidative stress induces atherosclerosis by triggering an inflammatory cascade within the vascular wall.

Objective: To investigate the role of pro-oxidant and antioxidant gene variations with CAD in Indian subjects.

Materials & methods: It’s a case-control study and genotyping for the variants MPO G-463A, CYBA G640A, SOD2 Val16Ala and CAT C-262T were performed by conventional PCR techniques.

Results: Only CYBA G640A variant allele was found to be significantly (p?=?0.0075) associated with CAD.

Conclusion: Although CYBA G640A variation was found to be significant, a larger study is needed to validate these results and establish its role as a biomarker.  相似文献

Background

The impact of host systemic conditions/diseases on the prosperity of oral Candida colonies remains unclear. The aim of the present study was to investigate whether a relationship exists between the quantity of oral Candida and the systemic condition/diseases of the host.

Patients and Methods

The cross-sectional relationship between Candida mannan concentrations and health check-up results was analyzed in consideration of local conditions that influence the prevalence of oral Candida.

Results

Candida mannan concentrations correlated with age, the number of untreated decayed teeth, number of prosthetic teeth, salivary pH, HbA1c, and the red blood cell count in a univariate analysis. In a multivariate analysis, Candida mannan concentrations correlated with age, the number of untreated decayed teeth, number of prosthetic teeth, salivary pH, and the red blood cell count. Candida mannan concentrations were higher in subjects older than 80 years, with a higher number of either untreated or prosthetic teeth, with a lower salivary pH, and with a decreased red blood cell count. Mannan concentrations were slightly higher in subjects with elevated HbA1c.

Conclusions

The present results suggest a close relationship between the quantity of oral Candida and the systemic condition/diseases of the host. Oral Candida may increase in immunocompromised hosts.

Objectives

To investigate mycothiol peroxidase (MPx) of Corynebacterium glutamicum that is a novel CysGPx family peroxidase using both the mycoredoxin and thioredoxin reducing systems as proton donors for peroxide detoxification and may be involved in the relief of acid stress.

Results

A Δmpx mutant exhibited significantly decreased resistance to acid stress and markedly increased accumulation of reactive oxygen species (ROS) and protein carbonylation levels in vivo. Over-expression of mpx increased the resistance of C. glutamicum to acid stress by reducing ROS accumulation. The stress-responsive extracytoplasmic function-sigma (ECF-σ) factor, SigH, mediated acid-induced expression of mpx in the wild-type under acid conditions, which in turn directly contributed to tolerance to acid stress.

Conclusion

MPx is essential for combating acid stress by reducing intracellular ROS levels induced by acid stress in C. glutamicum, which adds a new dimension to the general physiological functions of CysGPx.

Context: Early diagnosis of complications after severe trauma by specific biomarkers remains difficult.

Objective: Identify potential new biomarkers for early diagnosis of post-traumatic complications.

Material and methods: Mice underwent pressure-controlled hemorrhage or sham procedure. Four hours later, genome-wide expression of isolated Kupffer cells was compared with controls using Affymetrix-Genechip-Expression-Analysis and real-time-PCR.

Results: Expression analysis and real-time-PCR revealed a significant increase of gene expression of Cxcl10, Il4ra, Csf2rb2, Lcn2, and Gbp5.

Conclusion: Cxcl10, Il4ra, Csf2rb2, Lcn2, and Gbp5 might represent new biomarkers for early diagnosis of post-traumatic complications, if they are linked to the development of post-traumatic complications.  相似文献

Purpose

To analyze the complete genome sequence of the Brevibacterium frigoritolerans ZB201705, a Brevibacterium strain was isolated from the maize rhizosphere in drought- and salt-stressed soil, and the activity of the strain under simulated drought and high salt conditions was assessed.

Methods

We used a combination of the PacBio RS and Illumina sequencing platforms to obtain the complete genome sequence of B. frigoritolerans ZB201705.

Results

The genome consists of 5,475,560 bp in a linear chromosome with no gaps, 4,391 protein-coding sequences, 39 ribosomal RNAs, and 81 transfer RNAs. The genome analysis revealed many putative gene clusters involved in defense mechanisms. In addition, an activity analysis of the strain under high-salt and simulated drought conditions helped clarify its potential tolerance to these abiotic stresses.

Conclusions

Our data revealed the complete genome sequence of the new isolated strain, and showed that it produces many proteins involved in drought and salt stress responses, suggesting that B. frigoritolerans ZB201705 may be a potential factor to increase crop yield under abiotic stresses. The information provided here on the genome of B. frigoritolerans ZB201705 provides valuable insight into rhizobacteria-mediated plant salt and drought tolerance and rhizobacteria-based solutions for agriculture under abiotic stress.
