This study focused on the acute physiological responses to saltwater exposure in juvenile shortnose sturgeon Acipenser brevirostrum. In two separate laboratory experiments, 2 year‐old A. brevirostrum were exposed to either full (32) or half‐strength (16) seawater for up to 24 h. First, oxygen consumption rates were used to estimate the metabolic costs over 24 h. Secondly, blood and muscle samples were analysed at 6, 12 and 24 h for water loss, various measures of osmoregulatory status (plasma osmolality and ions) and other standard haematological variables. Juveniles exposed to full‐strength seawater showed significant decreases in oxygen consumption rates during the 24 h exposure. Furthermore, seawater‐exposed fish had significantly increased plasma osmolality, ions (Na+ and Cl?) and a 17% decrease in total wet mass over the 24 h exposure period. To a lesser extent, increases in osmolality, ions and mass loss were observed in fish exposed to half‐strength seawater but no changes to oxygen consumption. Cortisol was also significantly increased in fish exposed to full‐strength seawater. While plasma protein was elevated following 24 h in full‐strength seawater, haemoglobin, haematocrit and plasma glucose levels did not change with increased salinity. These results imply an inability of juvenile A. brevirostrum to regulate water and ions in full‐strength seawater within 24 h. Nonetheless, no mortality occurred in any exposure, suggesting that juvenile A. brevirostrum can tolerate short periods in saline environments.  相似文献

The osmolality and ionic composition of the blood of juvenile Atlantic cod Gadus morhua and their response to conditions of reduced temperature and salinity in summer‐ and winter‐acclimated individuals was investigated. Haematocrit percentage was relatively stable throughout the experimental procedures. Summer‐acclimated juvenile Atlantic cod had higher plasma osmolality than winter‐acclimated fish in ambient conditions. Plasma Na+ levels were, however, higher in winter conditions, while Cl did not vary between seasons. Temperature reduction (12, 9 and 6° C in summer and to 6 and 4° C in winter) induced a significant response in plasma osmolality and Na+ levels in summer, but only in Na+ levels in winter‐acclimated fish. A pronounced effect was seen in the summer 6° C treatment. Salinity treatments (24, 16 and 8) had a significant effect on almost all the variables in both summer and winter and resulted generally in dilution of ionic and osmotic concentrations of the plasma. This effect was pronounced in the lowest temperature treatments, with the greatest reduction observed in the summer 6° C treatment. This could suggest that winter‐acclimated fish are physiologically adapted to cope with lower seawater temperatures as opposed to summer‐acclimated fish.  相似文献

Flounders Pleuronectes flesus with an implanted vascular catheter were exposed to a 50% dilution of the water soluble fraction (WSF) of Omani crude oil (c. 6ppm total aromatic hydrocarbons) and serial blood samples taken to determine their endocrine status (cortisol, catecholamines and thyroid hormones) and the resultant and/or causal physiological (haematological, ionoregulatory and respiratory) disturbances. This resulted in a progressive increase in plasma cortisol concentrations from 3 h onwards (rising from 18 to 51 ng ml−1 after 48-h exposure), and increased plasma glucose concentrations indicating a generalized stress response. Plasma T3 and T4 concentrations of both control and WSF-exposed groups declined progressively over the experimental period; exposure to the WSF of crude oil further depressed plasma T4 concentrations but not plasma T3 concentrations relative to those of control fish. Plasma osmolality and sodium and chloride concentrations were stable in WSF-exposed fish, however, plasma potassium concentrations were increased significantly at the 24-and 48-h sampling points. The most profound physiological disturbance in WSF-exposed fish was a dramatic decline in blood oxygen content (CvO2) (from 2–8 to 0–8 ml 100 ml−1 after 48-h exposure), which is likely to be the cause of the increased plasma noradrenaline concentrations from 3 h onwards. Increased noradrenaline is likely in turn to have been responsible for the significant increase in blood haematocrit and blood haemoglobin at the 3-h sampling point, although the dominant effect in the longer-term was a significant decline in both of these haematological parameters.  相似文献

White sturgeon, Acipenser transmontanus (Richardson), are at risk of entrainment from dredging, with young-of-the-year fish at greatest risk. To evaluate this entrainment risk, swimming performance trials were conducted in a laboratory swim tunnel with hatchery-reared juvenile white sturgeon with varying experience levels including: naïve (only tested once), tested (re-tested after being kept in no flow) and trained (re-tested after kept in flow for nearly three weeks). Individuals of various sizes (80–100 mm TL) and all experience levels were strongly rheotactic (> 80%), but endurance was highly variable among fish. Small juveniles [< 82 mm total length (TL)] had lower escape speeds (< 40 cm s−1) than medium (82–92 mm TL) and large (> 93 mm TL) naïve fish (42–45 cm s−1), all of which had lower escape speeds than trained fish (72 cm s−1). Behavior was also highly variable among fish. Overall, benthic station-holding behaviors were least frequent in small fish, intermediate in medium and large fish, and most frequent in trained large fish. Probability of entrainment of juvenile white sturgeon can be reduced by maintaining dredge head flow fields at less than 45 cm s−1 for wild-spawned fish or by rearing hatchery fish to > 93 mm TL and exposing the fish to moderate flow velocities (10–12 cm s−1) prior to their release.  相似文献

The effect of 17beta-estradiol (E(2)) on osmoregulatory performance was examined in the euryhaline killifish, Fundulus heteroclitus. Fish were injected once with 1, 2 and 5 microg g(-1) E(2) and, 6 h after injection, transferred from 1 ppt seawater (SW) to full strength SW (40 ppt) or from SW to 1 ppt SW. In another set of experiments, fish were injected four times on alternate days with 2 microg g(-1) E(2) and then, 6 h after the last injection, transferred from 1 ppt SW to SW or from SW to 1 ppt SW. Fish were sampled 18 h after transfer (i.e., 24 h post-injection), and plasma osmolality, Na(+) and Cl(-) concentration and gill K(+)-pNPPase activity (a reflection of the sodium pump) were examined. Transfer from 1 ppt SW to SW resulted in significantly increased plasma osmolality, but did not affect gill K(+)-pNPPase activity. A single dose of E(2) (1, 2 and 5 microg g(-1)) prior to transfer from 1 ppt SW to SW increased plasma osmolality and decreased gill K(+)-pNPPase activity in a dose-dependent manner. Prolonged treatment with E(2) increased plasma osmolality and decreased gill K(+)-pNPPase activity in 1 ppt SW-adapted fish. Transfer of fish thus treated from 1 ppt SW to SW increased plasma osmolality and did not alter gill K(+)-pNPPase activity. Transfer from SW to 1 ppt SW had no significant effect on plasma osmolality or gill K(+)-pNPPase activity. Only the highest single dose of E(2) (5 microg g(-1)) prior to transfer from SW to 1 ppt SW decreased gill K(+)-pNPPase activity. Prolonged treatment with 2 microg g(-1) E(2) decreased gill K(+)-pNPPase activity only following transfer from SW to 1 ppt SW. The results substantiate an inhibitory action of E(2) on hypoosmoregulatory capacity in this euryhaline teleost.  相似文献

Twenty-month-old bighead carp, Aristichthys nobilis (Richardson), were fed prepared dry diets for 20 months in cages in Laguna de Bay, Philippines, to determine the effect on reproductive performance. The experimental diets were similar in composition except for the combinations of vitamins being tested. Diet 1 was supplemented with vitamins A, E and C; diets 2, 3 and 4 each lacked one of the supplementary vitamins; and diet 5 did not include any vitamin supplementation. Bighead carp that relied solely on natural food without a prepared diet served as a control. The total of six treatments each had two replicates. Results showed that the onset of gonad maturation was 2–3 months earlier in the fish that were fed the prepared diets regardless of vitamin supplementation, when compared with the fish that were fed natural food (control) . Moreover, the prepared diets enhanced egg hatchability which was significantly higher in fish that were fed diet 1 (+ vitamins A, E and C, 80.5 ± 18.1%) and diet 3 (– vitamin E, 78.5 ± 1.1%) than in those fish that were fed natural food (control) (36.5 ± 31.3%). Mean number of 3-day-old larvae was highest in fish fed on diet 1 (34 525 ± 1732), followed closely by fish that were fed diet 3 (32 420 ± 3909). A low number of 3-day-old larvae was obtained from fish fed the natural diet (14 490 ± 4331) as well as in fish that were fed diet 2 (– vitamin A, 14 347 ± 4863), diet 4 (– vitamin C, 21 407 ± 5840) and diet 5 (– vitamin A, E and C, 12 191 ± 1439). Other criteria for reproduction such as relative fecundity, fertilization rate, and hatching rate did not differ significantly (P > 0.05) among treatments. The addition of vitamins also had no significant effects on weight gain of adult fish.  相似文献

Plasma cortisol levels of fingerling rainbow trout were measured as an index of the stress resulting from various procedures used for transport of the fish for stocking. When transported under 'normal' conditions, which included water at the hatchery acclimation temperature (10–11°C), O2 saturation or supersaturation, and neutral pH, there was a marked increase in plasma cortisol levels within 0.5 h, which was maintained over the next 4 h of transport; there was a significant decrease in plasma cortisol by 8 h of transport. It was found that the plasma cortisol levels at 4 and 8 h were not appreciably altered by transport under partial O2 desaturation, O2 saturation, O2 supersaturation, or 0.5% NaCl, or by anaesthesia with tricaine methanesulfonate (MS 222) prior to capture and transport in MS 222-free water or 0.5% NaCl. A 15 min exposure to an immobilizing dose of buffered or unbuffered MS 222, or 2-phenoxyethanol, caused an increase in plasma cortisol of about 2 h duration, indicating that anaesthetics are themselves stressful. Exposure to chilled water (1° C) caused a large increase in plasma cortisol levels by 4 h after initiation of exposure; plasma cortisol had decreased at 1 day, and by 2 days a constant level was reached which was above the level in fingerling trout under 'normal' hatchery conditions. Trout acclimated to chilled water for 24 h and transported in chilled water had an increase in plasma cortisol during transport. Anaesthesia prior to transport or addition of salt did not reduce the stress of transport as judged by plasma cortisol levels. The results indicate that stress from capture and transport during stocking cannot be avoided using present methods.  相似文献

The diet of the dusky grouper Epinephelus marginatus , from the Balearic Islands, western Mediterranean, consisted primarily of crustaceans, molluscs and fishes, but diet composition varied with body size. The smaller dusky groupers (<300 mm L T) fed primarily on crustaceans, and particularly on brachyurans, which accounted for 46% of the prey identified. As dusky groupers grew, cephalopods became increasingly important and constituted 10 to 40% of the prey identified in subadult and adult specimens. The largest dusky groupers fed primarily on fishes that represented 40·9% of prey identified. These shifts in diet were accompanied by a positive selection of increasingly large prey and by an expansion of trophic niche. δ 15N values of dusky grouper white muscle ranged from 8·8 to 13·1% and 71% of the variation in δ 15N was explained by differences in dusky grouper size. δ 13C values ranged from – 17·9 to – 15·9%, and no significant body size effect on δ 13C was detected. Stomach content and 13C values indicated that from 1 year old, the diet of E. marginatus was based on the benthic food web. The enrichment in δ 15N registered from juvenile to large males was c . 3·8%. Overall, there was good agreement between gut content and stable isotope data indicating that the latter may be useful as a tool in trophic studies in marine protected areas where sampling for fishes is not allowed.  相似文献

This study investigated the effect of five sampling procedures on serum cortisol, glucose, total protein, osmolality, Na+, Cl, K+ and Ca++ concentrations in 2-year-old cultured sea bass ( Dicentrarchus labrax L).
Mild disturbance caused by rapid removal of fish and brief handling did not induce significant variation in any of the blood parameters investigated. Confinement and crowding elicited a high and significant increase in serum cortisol, glucose, osmolality, Na+, Cl, and Ca++ concentrations. Exposure to MS 222 (140 mg L–1) significantly increased osmolality, but not ionic concentration. Site of blood withdrawal (cardiac sinuses/caudal vein) had no effect on the concentration of analysed blood constituents, except for K+ levels. Scattered literature of sea bass blood chemistry is reviewed and compared with 'normal' ranges of blood constituents measured in this study. We conclude that it is necessary to select and rigorously execute an opportune blood sampling procedure whenever blood constituents are used as indicators of fish functional state.  相似文献

Survival, oxygen consumption (     ), total plasma cortisol and glucose levels and gill heat-shock protein 70 (hsp70) expression were measured in 10 and 50 g juvenile Atlantic cod Gadus morhua during an acute temperature increase (2° C h−1) to their critical thermal maximum. Ninety three per cent of the fish in both size classes survived to 24° C; however, mortality was 100% within 15 min of reaching this temperature. The     for both size classes increased significantly with temperature, reaching peak values at 22° C that were c. 2·8-fold those of control (10° C) fish. Resting plasma cortisol and glucose levels were lower in 10 g as compared to 50 g fish. Plasma glucose levels were highly variable in both size classes, and significant increases were only seen at >22° C for the 10 g fish. In contrast, plasma cortisol showed an exponential increase with temperature starting at 16° C in both size classes, and reached maximum levels at 22° C that were 19-fold (10 g fish) and 35-fold (50 g fish) higher than their respective control groups. Both the constitutive (73 kDa) and inducible (72 kDa) isoforms of hsp70 were detected in both size classes using the widely utilized mouse monoclonal antibody. Expression of these isoforms, however, did not change when Atlantic cod were exposed to elevated temperature, and the 72 kDa isoform was not detected using salmonid-specific antibodies. These results indicate that juvenile Atlantic cod are very sensitive to acute increases in water temperature. In addition, they (1) show that     and plasma cortisol, but not plasma glucose or gill hsp 70 levels, are sensitive indicators of thermal stress in Atlantic cod and (2) support previous reports that the upper critical temperature for this species is 16° C.  相似文献

An increased understanding of the juvenile life history stage of the lake sturgeon, Acipenser fulvescens , has been recognized as a key requirement for improving conservation efforts for this once abundant species. The objectives of the current study were to develop an effective methodology for capturing juvenile lake sturgeon in a large riverine environment; to describe the physical habitat characteristics (depth, water velocity and substrate) associated with the areas where juvenile lake sturgeon were captured; and to determine basic population parameters (length, body mass and condition factor) for juvenile lake sturgeon captured in those areas. Gillnets (mesh sizes 25–108 mm) were used to sample the Winnipeg River, a large river in the northern extent of the species range, from 12 June to 6 November 2006. A total of 2154 juvenile (<530 mm FL) lake sturgeon were captured, which represented over 74% of the total fish catch. Moderate (51–108 mm) and small (25 mm) mesh gill nets were found to be efficient for sampling juvenile lake sturgeon, with the former capturing a wider range of sizes and the latter capturing fewer fish but resulting in decreased mortality. Juvenile lake sturgeon were captured at high densities in discrete areas characterized by water depths >13.7 m, detectable water velocities >0.20 m s−1, and various substrate types.  相似文献

After conditioning Nile tilapia Oreochromis niloticus for 2 weeks, the photoperiod regime of 12 tanks of fish was changed to a 6L : 6D photoperiod while 12 further tanks were retained on the conditioning photoperiod regime (12L : 12D). Blood samples were collected 3 days (first sampling) and 3 months (second sampling) after changing the photoperiod regime. Blood was collected at 6 h intervals from both photoperiod regimes (1000, 1600, 2200 and 0400 hours). At the first sampling time, fish in the 6L : 6D had significantly higher cortisol both in the light and dark phases than levels in fish in the 12L : 12D photoperiod. At the second sampling time, the levels were significantly higher only in the light phase. The levels of cortisol, glucose and Cl in fish exposed to the 6L : 6D photoperiod, however, were far lower than those of acute stress‐induced levels observed in fish exposed to a stress experiment. Furthermore, there were no significant differences in overall values of all the variables between the photoperiod regimes at the second sampling time. This indicated that the fish exposed to the 6L : 6D photoperiod were not chronically stressed. Significantly higher blood lymphocyte counts were observed in fish exposed to the 6L : 6D compared to those of the 12L : 12D photoperiod during the light phase at the second sampling time. Other variables (glucose, Cl, haematocrit and neutrophil) did not show a significant difference between the treatments at either sampling time. These results demonstrated that the artificial photoperiod regime did not cause a significant acute or chronic stress response in Nile tilapia.  相似文献

This study describes the successful implementation of a modified cetyltrimethyl ammonium bromide (CTAB) protocol to isolate genomic DNA and amplify 14 microsatellite markers from fertilized eggs and larvae of an emerging South African farmed marine fish species, the dusky kob Argyrosomus japonicus. To test and validate the efficiency of this method, genetic data were utilized to resolve parentage and kinship of first‐generation (F1) offspring produced in mass‐spawning events of wild broodstock fish in a commercial hatchery.  相似文献

This study was carried out to determine the effects of gradual salinity increase on osmoregulatory ability of the Caspian roach Rutilus caspicus, under conditions which mimic stocking conditions of hatchery-raised fish. Initially, 30 juvenile fish (mean ± S.D. 3.20 ± 0.34 g) were transferred to 20 l circular tanks, in which salinities were changed in a stepwise fashion, from 0 to 5, 10 or 15 at 48 h intervals. The fish at salinity 15 were held for an additional 48 h at this salinity. Forty-eight hours after salinity transfer, survival rate, haematocrit, plasma Cl(-) , Na(+) and K(+) concentrations, osmolality and gill Na(+) /K(+) -ATPase (NKA) activity were measured. The only effect of exposure to 5 was a significant reduction in haematocrit compared to the freshwater control group. Exposure to salinity 10 raised haematocrit, Cl(-) and Na(+) concentrations and osmolality. At 48 h exposure to salinity 15, haematocrit, Cl(-) and Na(+) concentrations and osmolality were significantly higher than freshwater controls, and gill NKA activity was significantly lower, but the effect on NKA was no longer evident at 96 h exposure. There were no effects on survival. These results indicate that R. caspicus juveniles experience an initial non-lethal iono-osmotic perturbation following salinity increase but can adapt to brackish water at salinity 15.  相似文献

Several common situations in gilthead seabream (Sparus aurata L.) farming, such as air exposure, crowding and the use of anaesthetics, have been demonstrated to be stressful. In the present study, these conditions were simulated in the laboratory, after which head-kidney natural cytotoxic cell (NCC) activity was evaluated. For this, several specimens were air exposed for 2 min, returned to the aquarium and sampled from 0 to 4 days after exposure. NCC activity was significantly lower on the day following air-exposure compared with the control (rested fish) but not at any other time studied. Other fish were crowded (100 kg biomass m(-3), 2 h), returned to an aquarium with the same density as the control group (9 kg m(-3)) and sampled from 0 to 4 days after treatment. Head-kidney NCC activity was statistically increased compared with the control (resting) fish, 1 day after crowding. Anaesthesis for 1 h with 60 or 200 microl 2-phenoxyethanol l(-1)had no significant effect on NCC activity, while the use of 50 mg MS222 l(-1)for 1 h reduced such activity (by about 40%) compared with the control. In other experiments, fish were consecutively treated with crowding and anaesthetics. When treated with the lowest 2-phenoxyethanol concentration after crowding, the NCC activity inhibition was abolished compared with the activity in fish treated either with crowding or anaesthetic alone, while the use of the highest concentration increased such inhibition. The use of MS222 after crowding did not produce any differential effect compared with the fish treated with only one of the factors. In conclusion, NCC activity is affected differently according to the stress factor applied (hypoxia, crowding and/or anaesthetics). Differences in the effects provoked by these stressors on other seabream innate immune parameters are discussed.  相似文献

This study investigated the effect of high‐dose vitamin C supplementation on growth, tissue ascorbic acid concentrations and physiological response to transportation stress in juvenile silver pomfret, Pampus argenteus (initial average weight 6.2 ± 0.2 g). Three practical diets were formulated to contain 100 (control), 450 and 800 mg ascorbic acid/kg diet, respectively, supplied as l‐ascorbyl‐2‐polyphosphate. Each diet was fed to triplicate groups of fish in circular tanks (3 m ø, 1.5 m depth) (60 fish/tank) for 9 weeks. Growth did not change significantly with dietary vitamin C levels, although an improvement tendency with an increase in vitamin C supplementation was observed. Ascorbic acid concentrations in the liver and muscle of fish fed diets containing graded levels of vitamin C were positively correlated with dietary levels of this vitamin. Tissue ascorbic acid concentrations significantly increased with increasing vitamin C supplementation. After 9 weeks, the fish were subjected to transportation stress for 4 h to determine the influence of high vitamin C supplementation on the physiological response to this stressor. Serum cortisol, glucose and lactate levels significantly increased in stressed fish. Serum cortisol and glucose concentrations after stress were significantly higher in fish fed the control diet (7.91 μg L?1 and 0.80 mm , respectively) than in the other groups. However, there were no significant differences in serum cortisol and glucose levels after stress between the 450 and 800 mg kg?1 diets. No significant change could be found in serum lactate levels after stress among the different treatments. In conclusion, the dietary administration of high doses of vitamin C could reduce stress in silver pomfret and increase the survival of fish under stress conditions.  相似文献

Repeated blood withdrawal (5% of estimated blood volume at 0, 1, 4, 8, 24, 48 and 76 h) from tilapia acclimated to fresh water (FW) resulted in a marked increase in plasma levels of prolactin (PRL) during the first 8 h, reaching a peak above 300 ng/ml after 4 h. The increase in plasma PRL levels was significant except for the level after 72 h. A slight but significant decrease in plasma osmolality was observed at all time points after the blood withdrawal. Repeated blood withdrawal from fish acclimated to seawater (SW) resulted in a marked increase in plasma osmolality after 4 and 8 h. A significant increase was observed in plasma growth hormone (GH) in the fish in SW until the end of the experiment, but there was no change in plasma PRL. Plasma levels of cortisol were significantly higher in the fish in SW than in those in FW during the first 24 h. Blood withdrawal resulted in a significant reduction in hematocrit values in both FW- and SW-adapted fish, suggesting hemodilution. In a separate experiment, a single blood withdrawal (20% of total blood) stimulated drinking after 5 h, regardless of whether the fish were held in FW or SW. Plasma PRL level was also elevated following a single blood withdrawal in the fish acclimated to FW, but not in the fish in SW. Intraperitoneal injection of ANG II (1.0 microg/g) into the fish in FW significantly increased plasma PRL levels after 1 h. Activation of the renin-angiotensin system after blood withdrawal and the dipsogenic action of angiotensin II (ANG II) are well established in fish. The reduction in plasma osmolality after repeated blood withdrawal in FW and the increased osmolality in SW suggest that blood volume is restored, at least in part, by drinking environmental water. These results suggest that the marked increase in PRL concentration after blood withdrawal from the fish in FW is due, at least in part, to a facilitative effect between ANG II and reduced plasma osmolality.  相似文献

Four‐year‐old pollack Pollachius pollachius L., previously fed on dry pellets since their juvenile stage, were divided into three experimental groups two months prior their first spawning season. They were fed a commercial broodstock pellet enriched with 6% added oils, either: (1) capelin oil (control), (2) capelin oil plus arachidonic acid, or (3) DHA rich tuna oil. Spawning performance was determined in each group and egg lipids were analysed. During the vitellogenic period, the estradiol levels in plasma increased with oocyte enlargement, indicating that captivity and pelleted feed did not affect reproductive capacity. Females from each group spawned spontaneously between February and May. Egg production per kg of female was highest in the control group. Fertilization rate was highest (39%) in the group fed on diet enhanced in DHA. Lipid content in eggs reached 16% of dry mass, containing mainly phospholipids (75%). Egg fatty acid profiles showed few differences between dietary treatments. There was no significant difference in the concentration of Docosahexaenoic acid (22:6n − 3) between groups. Arachidonic acid (20:4n − 6) was lower in neutral and polar lipids of eggs from the control group than in the other groups. Tuna oil diet induced the highest DHA/EPA ratio in eggs and seemed to provide sufficient arachidonic acid for pollack broodstock. Egg fatty acid profiles are compared with 6 year‐old pollack broodstock fed on fish, and with other cultured and wild fish species.  相似文献
