Heat shock protein (HSP) synthesis results from various types of injury, including heat shock (HS) and some oxidants. The intracellular signals leading to HSP synthesis are not yet fully elucidated. We have studied the influence ofNNN’N’-tetrakis(2-pyridylmethyl)ethylene diamine (TPEN), a metal chelator known to induce cellular zinc and copper deprivation, on resistance to heat and on hsp70 synthesis in HaCaT keratinocytes. TPEN was shown to sensitize HaCaT cells to heat shock. The effect of TPEN was neutralized by equimolar Zn2+. By the use of sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis and Western blotting characterization of hsp70, it was shown that cultured HaCaT cells constitutively express the inducible form of hsp70. The application of TPEN alone slightly increases the level of hsp70 but inhibits its induction by HS. This inhibitory effect is related to metal deprivation, because it is eliminated when Cu2+ or Zn2+ ions are supplied together with TPEN. These results suggest that these metals are involved in the expression by keratinocytes of a stress protein which has a protective action against environmental stress.  相似文献

The expression of metallothionein (MT) and heat shock protein gene families was investigated in normal and in HeLa-derived cadmium-resistant cells, named H454. In the absence of amplification of MT genes H454 cells accumulated elevated concentrations of cadmium ions and synthesized higher levels of MT proteins than unselected HeLa cells. Northern blot analyses revealed higher levels of MT mRNAs in the resistant cells than in wild-type cells after Cd2+and Zn2+exposure. Evaluation of the cytotoxic potential of the different metals confirmed the high resistance to cadmium of the H454 cells. Two proteins of the heat shock family, hsp70 and GRP78, were synthesized in Cd2+-exposed H454 cells at levels comparable to the ones present in Cd2+-treated normal cells. Northern blot analyses of the mRNA levels corresponding to these proteins revealed elevated expression of both hsp70 and GRP78 mRNAs in H454 cells upon exposure to cadmium ions and no response to zinc induction. These data suggest the existence in the H454 cells of a cadmium-specific pathway of regulation of MT and heat shock genes.  相似文献

The major heat shock protein (hsp) of Hydra vulgaris has recently been found to be a 60 kDa protein. Since in all organisms studied so far, the major heat shock protein is a 70 kDa protein, we have analyzed the relationship of hydra hsp60 to the highly conserved 70 kDa heat shock protein family. Genes and proteins related to the 70 kDa class of stress proteins are present in hydra. However, antibodies known to cross-react with hsp70 proteins in several different organisms do not cross-react with hydra hsp60 suggesting that hsp60 is not related to the conserved hsp70 proteins.  相似文献

Heat shock proteins are ubiquitous and highly conserved. Recently they have become implicated in the import of proteins into organelles. All the heat shock genes characterized to date, however, are known or assumed to be encoded in the nuclear genome even if the corresponding protein can be localised in the mitochondrion or chloroplast. In contrast, we identify here an hsp70 gene in the unicellular chromophytic alga Pavlova lutherii which is located on the chloroplast genome. Localisation of this gene to the chloroplast chromosome is confirmed by Southern blot analysis and pulse-field gel electrophoresis which also reveals that the length of the P. lutherii chloroplast chromosome is 115 kb. We compare the predicted protein of this hsp70 gene with that of maize and of the analogous proteins in the prokaryotic organisms Escherichia coli and Synechocystis PCC6803. The greatest identity is found with the cyanobacterium Synechocystis PCC6803.  相似文献

Thirty‐eight populations of woodlice (Oniscus asellus, Porcellio scaber) and millipedes (Julus scandinavius) from 28 differently metal‐polluted field sites were analysed for their 70‐kDa stress protein (hsp70) level. Although ANOVA revealed significant dependence of the hsp70 level on the concentrations of water‐soluble lead, cadmium and zinc and the soil pH, each of these parameters accounted for at most 18% of the intersite variability of the stress protein level only. A multivariate model based on multiple regression analysis explained more than 96% of hsp70 variance and revealed both the pollution history of a site (strong metal contamination for more than 70 years) and invertebrate species identity to act as the most important parameters. The model accounted for the observation that most of the populations from long‐term polluted sites exhibited comparatively low stress protein levels in response to their own (contaminated) habitats. In contrast, isopods (O. asellus) from a control site were not able to maintain a low hsp70 level when they were exposed to either an artificial metal cocktail or soil taken from one of the contaminated field sites. They did not acclimatize to the exposure conditions within 3 months. We propose that selection of insensitive phenotypes in long‐term polluted soils has taken place so as to minimize the stress protein level which, in turn, is indicative of high intracellular protein integrity. Long‐term selection for a high hsp70 level to compensate for adverse metal impact was not observed, which suggests that such a strategy may trade off against other fitness consequences. In this context, insensitivity to metal stress involved increased selectivity in food choice and reduced variability in stress response. Multiple regression models showed species‐specificity in those abiotic factors which determined (1) high hsp70 levels in sensitive populations as well as (2) low hsp70 levels in insensitive ones. Therefore, abiotic factors can be assigned to act as the main components of selection: lead and cadmium for J. scandinavius and O. asellus, zinc for P. scaber.  相似文献

The rate of survival and stress protein (hsc/hsp70) response were investigated in the freshwater amphipod, Gammarus fossarum Koch, 1835, during a 20-day stress and recovery experiment. Adult females and males, were separately exposed to 9 different cadmium concentrations for 5 days to simulate a short-term pulse of xenobiotics in an aquatic environment, followed by a recovery period of 15 days. In terms of mortality, females were much more sensitive to cadmium than males; 4.28±2.45 g Cd2+/l resulted in strong effects on the rate of survival of females but not males. In both sexes, mortality occurred predominantly within the first 5 days of the recovery period. At the cellular level, cadmium induced an hsc/hsp70 response. The lower Cd2+ concentrations we used led to an induction of stress proteins while higher Cd2+ concentrations resulted in a proportionately reduced hsc/hsp70 response, most likely due to pathological damage. Surviving individuals retained their capacity to induce stress protein production in the recovery period, even if the stress protein response system was overwhelmed by cadmium during the exposure period.  相似文献

The heat shock response has been studied extensively, yet the molecular signals that trigger the response remain elusive. The dogma of the heat shock response contends that denatured proteins initiate the response, but evidence is accumulating to point to a more complex system in which at least more than one signal is involved in this process. Thermal stress initiates changes in cellular phospholipid membrane physical state, which when acted upon by phospholipases may release lipid mediators that could serve as triggering signals during the heat shock response. We have examined the heat shock response in freshly isolated leukocytes from the pronephros of rainbow trout (Oncorhynchus mykiss). In this study, we show that leukocytes isolated from rainbow trout acclimated to 5 or 19°C express elevated levels of heat shock protein 70 (hsp70) mRNA when heat shocked at 5°C above their respective acclimation temperature and supplementation with exogenous docosahexaenoic acid or arachidonic acid followed by heat shock enhanced levels of hsp70 mRNA. The time course for docosahexaenoic acid induced enhancement of hsp70 mRNA was accelerated compared with heat shock alone, and staurosporine inhibited the docosahexaenoic acid induced increase of hsp70 mRNA. We also provide evidence that phospholipase A2 is involved in the heat shock response.  相似文献

The Mr 90000 heat shock protein (hsp 90) and one of the Mr 70000 heat shock cognate proteins (hsc 70) were localized by immunoelectron microscopy in salavary gland cells of normal and heat-shocked larvae of Chironomus thummi using polyclonal antibodies raised against Drosophila proteins. Immunoblotting after separation of proteins by gel electrophoresis shows that these antibodies cross-react with the corresponding proteins of Chironomus. Hsp 90 was localized both in the cytoplasm and in the nucleus, where it is associated with intrachromosomal and extrachromosomal ribonucleoprotein (RNP) fibrils, as well as with the peripheral region of compact chromatin. After heat shock the concentration of hsp 90 increases in the nucleus. This increase is prevented by actinomycin D administration during the heat shock. Hsp 90 is associated with the chromatin of puffs repressed by heat shock and with the RNP fibrils of actively transcribing heat shock puffs. Hsc 70 is mainly found in RNP fibrils and in the periphery of compact chromatin. During heat shock the concentration of hsc 70 decreases in the cytoplasm while it becomes more abundant in association with chromatin and intrachromosomal and extrachromosomal RNP fibrils. These results suggest a translocation of the existing protein from the cytoplasm toward the nucleus. Abstract

During investigations on the induction of the 70 kD stress protein family (hsp70, stress-70) in Julus scandinavius following exposure to different biohazards, the colour of the supernatant of the homogenate was closely correlated to the hsp70 level. Hsp70 has recently been shown to be a suitable biomarker for sublethal toxicity in soil animals. Control millipedes typically exhibited red or red-orange supernatants whilst the supernatant of starved or toxin-exposed diplopods was orange, orange-yellow, or even bright yellow. Based on these observations, a quantitative colour test was established which was found to be able to indicate the degree of stress situations caused by exposure to heavy metals (cadmium, zinc), organic pollutants (lindane, PCB 52), or by food deprivation in laboratory tests. It is suggested that this is caused by a breakdown of the red-orange bilirubins into orange-yellow urobilins.  相似文献

The effect of overproducing each of the three small heat shock proteins (Hsp; Hsp 18.5, Hsp 18.55, and Hsp 19.3) was investigated in Lactobacillus plantarum strain WCFS1. Overproduction of the three genes, hsp 18.5, hsp 18.55, and hsp 19.3, translationally fused to the start codon of the ldhL gene yielded a protein of approximately 19 kDa, as estimated from Tricine sodium dodecyl sulfate–polyacrylamide gel electrophoresis in agreement with the predicted molecular weight of small Hsps. Small Hsp overproduction alleviated the reduction in growth rate triggered by exposing exponentially growing cells to heat shock (37 or 40°C) and cold shock (12°C). Moreover, overproduction of Hsp 18.55 and Hsp 19.3 led to an enhanced survival in the presence of butanol (1% v/v) or ethanol (12% v/v) treatment suggesting a potential role of L. plantarum small Hsps in solvent tolerance.  相似文献

The highly conserved part of the nucleotide-binding domain of the hsp70 gene family was amplified from the soil diplopod Tachypodoiulus niger (Julidae, Diplopoda). Genomic DNA yielded 701, 549 and 540 bp sequences, whereas cDNA from heat shocked animals produced only one distinct fragment of 543 bp. The sequences could be classified as a 70 kDa heat shock protein (hsp70), the corresponding 70 kDa heat shock cognate (hsc70) and a glucose-related hsp70 homologue (grp78). Comparisons of genomic and cDNA sequences of hsc70 identified two introns within the consensus sequence. Generally, stress-70 expression levels were low, which hampered successful RT-PCR and subsequent subcloning. Following experimental heat shock, however, the spliced hsc70 was amplified predominantly, instead of its inducible homologue hsp70. This finding suggests that microevolution in this soil-dwelling arthropod is directed towards low constitutive stress-70 levels and that the capacity for stress-70 induction presumably is limited. hsc70, albeit having introns, apparently is inducible and contributes to the stress-70 response.  相似文献

Heat shock protein induction is often associated with a cellular response to a harmful environment or to adverse life conditions. The main aims of our study were (1) to evaluate the cytotoxic potential of cypermethrin; and (2) to investigate the suitability of stress-induced heat shock protein Hsp70 as a biomarker for environmental pollutants in transgenic Drosophila melanogaster (Hsp70-lacZ)Bg9. Different concentrations of cypermethrin (0.002, 0.2, 0.5 and 50.0 p.p.m.) were mixed with food. Third instar larvae of transgenic Drosophila melanogaster were allowed to feed on these mixtures for different time intervals (2, 4, 6, 12, 24 and 48h). Following feeding, hsp70 induction and tissue damage were evaluated. In the highest concentration treatment group (50 p.p.m.), 100% larval mortality was recorded after 12 h exposure. Hsp70 was found to be induced even at the lowest concentration (0.002 p.p.m.) of the insecticide, while tissue damage was observed in the larvae exposed for 48 h. While an insignificant decline in hsp70 expression was observed in the larvae exposed to cypermethrin at a dietary concentration of 0.002 p.p.m. after 48 h compared with those exposed for 24 h, in the next two higher concentrations of the toxicant, a similar but significant decline in hsp70 expression was evident in the exposed larvae after 48 h. The present study reveals the cytotoxic potential of cypermethrin and further proposes that hsp70 induction in transgenic Drosophila melanogaster could be used as a sensitive biomarker in risk assessment.  相似文献

The effect of cadmium on the photosynthetic activity of Synechocystis PCC 6803 was monitored in this study. The oxygen evolving capacity of Synechocystis treated with 40 μM CdCl2 was depressed to 10% of the maximum in 15 min, indicating that Cd2+ penetrated rapidly into the cells and blocked the photosynthetic activity. However, neither photosystem II (PSII) nor photosystem I (PSI) activity showed a significant short-term decrease which would explain this fast decrease in the whole-chain electron transport. Thermoluminescence measurements have shown that the charge separation and stabilization in PSII remains essentially unchanged during the first few hours following the Cd2+ treatment. The electron flow through PSI was monitored by following the redox changes of the P700 reaction centers of PSI. Alterations in the oxidation kinetics of P700 in the Cd2+-treated cells indicated that Cd2+ treatment might affect the available electron acceptor pool of P700, including the CO2 reduction and accumulation in the cells. Perturbed angular correlation of γ-rays (PAC) using the radioactive 111mCd isotope was used to follow the Cd2+ uptake at a molecular level. The most plausible interpretation of the PAC data is that Cd2+ is taken up by one or more Zn proteins replacing Zn2+ in Synechocystis PCC 6803. Using the radioactive 109Cd isotope, a protein of approximately 30 kDa that binds Cd2+ could be observed in sodium dodecyl sulfate polyacrylamide gel electrophoresis. The results indicate that Cd2+ might inactivate different metal-containing enzymes, including carbonic anhydrase, by replacing the zinc ion, which would explain the rapid and almost full inhibition of the photosynthetic activity in cyanobacteria.  相似文献

Summary The component areas of chick blastula show differential expression of heat shock genes. The area opaca (ao), marginal zone (mz) and central area (ca) components of the epiblast display distinct quantitative and minor qualitative differences in the heat-induced and heat-repressible proteins, but are clearly different from the primary hypoblast (endoderm) in their expression of a given stress protein (hsp) as a response to heat shock. The major proteins synthesized in the component areas of epiblast in response to heat shock include hsp 18, 24, 70 and 89 Kd. Two-dimensional electrophoresis shows that each of these proteins consists of multiple charged species. The hypoblast expresses only hsp 70 Kd at non-significant levels and shows marked inhibition in the level of synthesis of heat-shock-repressible proteins. Heat shock during the blastula stage results in an increase in the size of the blastoderm and disrupts normal embryonic development. The heat shock genes provide an important molecular marker, which attests to regional specification in the chick blastula.  相似文献

G protein-coupled receptors (GPCRs) transduce extracellular signals to the interior of the cell by activating membrane-bound guanine nucleotide-binding regulatory proteins (G proteins). An increasing number of proteins have been reported to bind to and regulate GPCRs. We report a novel regulation of the alpha2A adrenergic receptor (α2A-R) by the ubiquitous stress-inducible 70 kDa heat shock protein, hsp70. Hsp70, but not hsp90, attenuated G protein-dependent high affinity agonist binding to the α2A-R in Sf9 membranes. Antagonist binding was unchanged, suggesting that hsp70 uncouples G proteins from the receptor. As hsp70 did not bind G proteins but complexed with the α2A-R in intact cells, a direct interaction with the receptor seems likely. In the presence of hsp70, α2A-R-catalyzed [35S]GTPγS binding was reduced by approximately 70%. In contrast, approximately 50-fold higher concentrations of hsp70 were required to reduce agonist binding to the stress-inducible 5-hydroxytryptamine1A receptor (5-HT1A-R). In heat-stressed CHO cells, the α2A-R was significantly uncoupled from G proteins, coincident with an increased localization of hsp70 at the membrane. The contrasting effect of hsp70 on the α2A-R compared to the 5-HT1A-R suggests that during stress, upregulation of hsp70 may attenuate signaling from specific GPCRs as part of the stress response to foster survival.  相似文献
