This study determined the optimum number of tubules to be counted per testis cross section, and the number of animals per treatment group, when changes in stage frequencies in the cycle of the seminiferous epithelium are criteria for assessing effects of treatment on spermatogenesis. A data base of 9,672 observed and staged tubules was collected from testicular cross sections of 15 Sprague-Dawley rats. A significant variation between animals was found for the frequencies of Stages I, II, IV, VI, VIII, and XIII. Computer simulation was used to randomly select different combinations of animal and tubule numbers from the observed data. Stage frequency means from each simulation experiment were compared statistically to observed mean frequencies. A model that used data from all 14 stages was analyzed. The following conclusions were made: a) a minimum of 200 tubule cross sections/testis is recommended for estimating stage frequencies; b) for a fixed number of tubules scored, the number of animals sampled is more important than the number of tubules per animal in reducing variance; c) to detect a difference of 2 standard deviations from the mean with a 2% error rate and examining 200 tubules/testis, at least 12 animals must be used per group when assessing all 14 stages; d) when individual stages are examined using 10 animals per group, only Stage VII has 80% or greater power of test (alpha = 0.05) to detect a frequency difference; e) pooling stages into 3-4 groups is recommended to improve the power of detecting a treatment difference.  相似文献

Experiments were performed using X. laevis embryos during gastrulation and neurulation (stages 10, 11 1/2, 12 1/2, 13 1/2, 15 and 18). Part of presumptive epidermis and lateral plate mesoderm was removed, and embryos raised until stage 25. The size of axial structures (notochord, somite mesoderm, central nervous system) was determined using serial histological sections and compared with that of control embryos. In experimental embryos, the size of axial structures was decreased. Until a specific stage of development, close correlation was found between the volume of embryonic compartment corresponding to a particular, structure and the volume of presumptive epidermis and lateral plate mesoderm. This stage is individual for each axial organ: middle gastrula (stage 11 1/2) for notochord, late gastrula (stage 12 1/2) for somite mesoderm, and late neurula (stage 18) for central nervous system. This data suggest that differentiation pattern of ecto-mesodermal rudiment is subject to regulation during gastrulation-neurulation, and subdivision of ectoderm and mesoderm into axial and non-axial tissues is a self-organizing process.  相似文献

Iron is essential for the normal functioning of cells but since it is also capable of generating toxic reactive oxygen species, the metabolism of iron is tightly regulated. The present article advances the view that astrocytes are largely responsible for distributing iron in the brain. Capillary endothelial cells are separated from the neuropil by the endfeet of astrocytes, so astrocytes are ideally positioned to regulate the transport of iron to other brain cells and to protect them if iron breaches the blood-brain barrier. Astrocytes do not appear to have a high metabolic requirement for iron yet they possess transporters for transferrin, haemin and non-transferrin-bound iron. They store iron efficiently in ferritin and can export iron by a mechanism that involves ferroportin and ceruloplasmin. Since astrocytes are a common site of abnormal iron accumulation in ageing and neurodegenerative disorders, they may represent a new therapeutic target for the treatment of iron-mediated oxidative stress.  相似文献

The results of experimental trawl fishing in the littoral zone of the Amur Bay are summarized. Using a motor boat and a launch for trawling with experimental trawl during the research works made it possible to obtain new data on the species composition, distribution, and stock density of the fishes in the littoral zone of Peter the Great Bay. Light trawl is promising not only for research purposes but also for seasonal fishing.  相似文献

Estradiol-2-hydroxylase, the enzyme responsible for the conversion of estrogens to catechol estrogens was measured in the brain of female rats at specific stages of the estrus cycle. Radiometric measurements of the enzyme activity in microsomal, mitochondrial, and synaptosomal fractions of the brain revealed a sharp increased in activity at proestrus just prior to the preovulatory LH surge. The enzyme activity declined to lower levels at diestrus and metestrus. No comparable fluctuations were noted in the liver enzyme. These changes in brain enzyme activity in conjunction with demonstrated positive feedback of exogenous catechol estrogens on pituitary LH release, suggest that a rise in endogenous catechol estrogen formation in the brain may be responsible for the physiological induction of the preovulatory LH surge.  相似文献

Analysis of the mouse chromosomes is becoming increasingly important in many fields of genetic research. It is generally considered that the mouse chromosomes are more difficult to analyse than, for example, human chromosomes which has often led to their misidentification. This article presents a guide to the correct identification of trypsin-Giemsa banded chromosomes from the mouse. The variability in the G-banded structure of each chromosome is presented pictorially together with some suggestions for their unequivocal identification. Since many of the mouse chromosomes have similar banding patterns, those chromosomes which are more frequently misidentified have been compared and contrasted. Finally a summary of the main features for the identification of each chromosome is presented.  相似文献

The identification of the whole set of protein interactions taking place in an organism is one of the main tasks in genomics, proteomics and systems biology. One of the computational techniques used by many investigators for studying and predicting protein interactions is the comparison of evolutionary histories (phylogenetic trees), under the hypothesis that interacting proteins would be subject to a similar evolutionary pressure resulting in a similar topology of the corresponding trees. Here, we present a new approach to predict protein interactions from phylogenetic trees, which incorporates information on the overall evolutionary histories of the species (i.e. the canonical "tree of life") in order to correct by the expected background similarity due to the underlying speciation events. We test the new approach in the largest set of annotated interacting proteins for Escherichia coli. This assessment of co-evolution in the context of the tree of life leads to a highly significant improvement (P(N) by sign test approximately 10E-6) in predicting interaction partners with respect to the previous technique, which does not incorporate information on the overall speciation tree. For half of the proteins we found a real interactor among the 6.4% top scores, compared with the 16.5% by the previous method. We applied the new method to the whole E.coli proteome and propose functions for some hypothetical proteins based on their predicted interactors. The new approach allows us also to detect non-canonical evolutionary events, in particular horizontal gene transfers. We also show that taking into account these non-canonical evolutionary events when assessing the similarity between evolutionary trees improves the performance of the method predicting interactions.  相似文献

In 1998, the government of India enforced a ban on performance/exhibition of 5 species of nonhuman animals: (a) lions, (b) tigers, (c) leopards, (d) bears, and (e) monkeys. The Ministry of Environment and Forests gave the responsibility to the Central Zoo Authority (CZA) for rehabilitation of these animals. Between 1999 and 2001, the CZA created rescue centers for rehabilitation of lions and tigers in the off-display areas of 5 zoos: (a) Bangalore, (b) Chennai, (c) Vishakhapatnam, (d) Tirupathi, and (e) Jaipur. Today, the CZA has rehabilitated 314 lions and tigers from circuses. The CZA has been meeting the expenses toward maintenance (feeding of and providing health care for) of the animals, outsourcing of staff, and maintaining enclosures. This article focuses on the mammoth work that the selected zoos had to carry out in rehabilitating the lions and tigers from the circuses.  相似文献

Circadian rhythms are demonstrated in the tongue of adult rats for both the mitotic index of the basal epithelium and the uptake rate of injected 3H-thymidine by the tongue tip. The animals were entrained to a light-dark cycle for four weeks prior to the experiments with the light phase extending from 0600 to 1800 hours (CST). The daily fluctuation is approximately 300% for the mitotic index and 185% for the uptake rate of 3H-thymi-dine. The highest mitotic index occurs at 1100, and the highest uptake of 3H-thymidine occurs four hours earlier at 0700. The least activity for both parameters occurs during the first part of the dark span of the light-dark cycle. Estimates of several other rhythmic parameters are determined by a computerized method.  相似文献

1. The effect exerted by a residue on the conformation of neighbouring residues was analysed by using data from nine globular proteins of known sequence and conformation. 2. An information measure was used which estimated the role of a residue in influencing neighbouring conformations and also its tendency to influence the lengths of runs of residues in that conformation. This measure was estimated for each residue in all conformations defined by domains on the varphi, psi diagram. 3. Plots of the information measure yielded an intercept, which was a measure of intra-residue information for a residue. The slope was a measure of the statistical co-operativity or tendency of the residue to influence the occurrence of its neighbours in runs of a particular conformation. Both parameters are a function of the residue type. Statistical co-operativity is found in the alpha(1)-helical (H(1)) and beta-pleated-sheet (P(2)) conformations and, to a lesser extent, in their distorted variants H(2) and P(1). 4. The directional nature of these influences for H(1) and P(2) conformations is illustrated by plots of the information measure against the distance m from the residue, for m=-10 to +10. 5. The results for statistical co-operativity are discussed in relation to theories of helix-coil and pleated-sheet-coil transitions. The value of the information-theory-derived parameters in obtaining s parameters for the Zimm & Bragg (1959) equations is illustrated. 6. Directional effects are discussed with particular relation to mechanisms of the termination of helices and the involvement of the alpha(II) conformation and also to discontinuities in pleated-sheet conformations.  相似文献

The author proposes a new scheme for arranging the data about the assimilation of carbon sources in tables; this is illustrated with the Griseus species belonging to the Streptomyces genus and with the information about them presented by Bergey [2]. In the scheme, carbon sources in the tables are arranged in the order of their diminishing availability for streptomycetes, and the lists of species within the morphological groups are constructed according to the property of a carbon source not being assimilated. Such an arrangement of data in the tables is very clear and convenient to use; it allows one to make optimal schemes for determining the assimilation of carbon sources by actinomycetes, and makes it easy to divide any large number of cultures into groups according to a similar assimilation of carbon sources. The scheme has been used to analyse the data available in science and to establish correlations in the assimilation of carbon sources by species of the Griseus series belonging to the Streptomyces genus.  相似文献

The role of the methylation in the detoxication of arsenate in the rabbit   总被引:1,自引:0,他引:1  
The biotransformation, tissue retention, intracellular binding and biokinetics of arsenic were studied in rabbits exposed to [74As]arsenate (0.4 mg As/kg body wt., i.v.). Inhibition of the methyltransferase activity by injection of periodate-oxidized adenosine (PAD) caused a marked decrease of the formation of [74As]dimethylarsinic acid (DMA), which gave rise to 1.5-4 times increased tissue levels of 74As. This is almost the same as reported for rabbits given arsenite in combination with PAD and was due to a rapid reduction of the arsenate to arsenite which bound to the tissues. Only about 30% of the arsenate given was excreted unchanged in the urine, indicating that a large part was reduced to AsIII. Thus the methylation to DMA seems to be almost as important for the detoxication following exposure to arsenate as that following exposure to arsenite. In the rabbits with normal methylating capacity 50-70% of the produced AsIII was methylated to DMA. The liver was the only organ in which DMA was present 1 h after the administration, indicating that this is the main site of the methylation. The DMA was rapidly cleared from all tissues except the thyroid.  相似文献

A detailed study of the anterior pituitaries of pregnant and lactating mice revealed changes occurring in the ultrastructure of the somatotrophs. The rough endoplasmic reticulum cisternae become greatly dilated forming channels that are continuous with the perivascular space. This occurrence expands the membrane surface available for the release of hormone. Individual granules and cytoplasmic islands containing numerous granules appear in these channels. This suggests an alternative mechanism for granule release which would facilitate a rapid response to increased hormone requirement.  相似文献

The role of high-frequency ripple signals (HF signals) made by males of the water strider Aquarius remigis was studied in the contexts of competition for food and general spacing behaviour during the non-mating season. HF signals were played back through the ripple-producing legs of males during dyadic agonisdc encounters, using a signal-driven wire coil to oscillate a magnet glued to a leg. These signals were in addition to normal signals. The additional signals significantly increased the number of retreats by non-magneted males, showing that the signals increased the dominance of a non-territorial male. Hence, our results increase the number of contexts in which HF signals of A. remigis function. Males, but not females, avoided a site occupied by a magneted dead male through which HF signals were played. Thus, the communication system used by A. remigis males during competition for food seemed to be ignored by females, suggesting sex-specific signals even in a non-mating context. Evolutionary models of signalling often assume that contestants have evolved the same repertoire of signals in order to resolve conflicts peacefully. This water strider system thus poses an interesting challenge for future theoretical and empirical research on communication asymmetry.  相似文献

发育性髋关节脱位(Developmental dysplasia of the hip,DDH)是指一系列髋关节结构异常的疾病,病变严重程度涵盖轻度髋臼发育不良到不可逆的髋关节脱位。DDH的治疗手段进展迅速,但早期闭合复位使用最多的仍是Pavlik吊带。通过超声可监测髋关节复位情况从而评估Pavlik吊带的治疗效果。同时,随着超生髋关节检查技术在我国的推广,使得早期诊断DDH成为可能,这也促进了Pavlik吊带在临床的应用。未来研究的方向在于制定基于询证医学证据的Pavlik吊带使用规范和开发出效果更好、并发症更少的新型支具。  相似文献

When the proportions of glycerol and ethanol vary for a given water concentration, the stability of the amorphous state of the whole solution in the system water-glycerol-ethanol passes through a maximum. This ternary system may then be much more interesting for cryoprotection than the system water-glycerol-DMSO. The phase transitions on rewarming at several rates after rapid or slow cooling, or after annealing, were observed by calorimetry and the various states between the transitions including the amorphous state were observed by X-ray diffraction. However, this system is much more complicated than the system water-glycerol-DMSO, due to the existence of two ethanol hydrates. Since, beyond a certain ethanol concentration, a first melting occurs before the devitrification, the stability of the amorphous state could no longer be defined by the critical warming rate. It was then defined by the amount of crystals formed on cooling, which was surprisingly reproducible in the present experiments for each cooling rate. The maximum in the stability occurs for rather low ethanol concentrations, which is of interest since ethanol is more toxic that glycerol. The concentration corresponding to the maximum depends on the cooling rate. It occurs at about 20% (ww) ethanol/(glycerol + ethanol) for the 50 and 55% (ww) water solutions. It is shifted to 40% (ww) ethanol/(glycerol + ethanol) for the 60% (ww) water solutions.  相似文献

Experiments were conducted on male albino rats for 5 months; the activity of the liver aspartic and alaninaminotransferases underwent cyclic synchronized variations. Chronic 24-hour inhalation of vinylacetate in a concentration of 2.4 mg/m3 caused a transformation of the rhythms of these variations with the preservation of synchronization in the changes of the activity of the enzymes under study. With a greater concentration of vinylacetate (13.2 and 68 mg/m3) inducing chronic intoxication, desynchronization of transaminase variations was seen in the liver. A posssibility of correlation analysis use for determining the extent of synchronization in the activity of the enzymatic systems for the purpose of estimation of the toxic effect of the poisons is discussed.  相似文献

Summary Laboratory data for the loss of root material by barley and field data for the growth of barley plants in Syria and in England have been combined to predict the amount of material lost by barley roots during a season, and to predict the resulting microbial biomass in the rhizosphere. The predicted microbial biomass C in the rhizosphere ranged from 10–34% of the total plant biomass C depending mainly upon the value used for rate of loss of root material. Total loss of root material predicted during a season in England constituted 7.7–25.4 percent of C fixed by photosynthesis. The major assumptions made in these calculations are considered, and the predicted values discussed in relation to reported values for soil microbial biomass, CO2 fluxes from soil and associative nitrogen fixation.  相似文献

Endopeptidase-24.11, an ectoenzyme with a key role in metabolizing peptides at cell surfaces, is present in the adenohypophysis. A specific polyclonal antibody to the endopeptidase has been used to explore its localization in cryostat sections of pig pituitary glands by an immunoperoxidase method. Immunoreactivity was symmetrically but not uniformly distributed over the anterior lobe, with the highest intensity a zone just beneath the capsule along the anterior surface. In detail, the staining was observed to be in the cell membrane, but in some cells a small area of intense paranuclear staining was also observed. Serial 5 micron sections were immunostained alternately for endopeptidase-24.11 and for pituitary proteohormone. Luteinizing hormone (LH), follicular stimulating hormone (FSH), thyrotropin, adrenocorticotropin, prolactin and growth hormone were studied in this way. It was possible to identify groups of cells in adjacent sections and a good correlation was observed for endopeptidase-24.11-immunoreactivity with that for LH and FSH. The association of the endopeptidase with gonadotrophs was confirmed by double labelling. No evidence of colocalization was observed with the other proteohormone antibodies. We conclude that among the cells of the adenohypophysis only the gonadotrophs express endopeptidase-24.11 and discuss the possible significance of this observation in regard to the termination of peptide signals, such as that of luteinizing hormone-releasing hormone (LHRH) acting at this site.  相似文献
