Previous literature on racial self-identification among multiracials demonstrates that self-identification differs by context. Moreover, among multiracial adolescents, identity, usually measured in school, is correlated with achievement. In addition, a few studies have indicated that for half-white, half-minority adolescents, school achievement falls in between the achievements of their monoracial counterparts. Using the in-school and in-home components of the National Longitudinal Study of Adolescent Health (Add Health), we examine the relationship between racial self-identification and school belonging and achievement. We find that among black/white and Asian/white adolescents, adolescents who self-identify as white are particularly disadvantaged in school, reporting lower grade point averages (GPA) than their multiracial counterparts. Our conclusions suggest that multiple contextual measures of self-identification better capture the relationship between racial identification and academic achievement among multiracial adolescents.  相似文献

This article investigates whether cultural models of behaviour can explain the persistently higher unemployment rate of non-whites in the UK. Such models argue that the prevalence of unemployment among non-whites may induce a greater tolerance of joblessness and poorer attitudes towards working which act to further reduce their employment chances. However, estimates from a hazard model of unemployment durations indicate that such factors play no role in explaining longer ethnic minority spells. This casts some doubt on the power of cultural models to account for the UK experience.  相似文献

In the course of DNA profiling of grapevine cultivars using microsatellite loci we have occasionally observed more than two alleles at a locus in some individuals and have identified periclinal chimerism as the source of such anomalies. This phenomenon in long-lived clonally propagated crops, such as grapevine, which contains historically ancient cultivars, may have a role in clonal differences and affect cultivar identification and pedigree analysis. Here we show that when the two cell layers of a periclinal chimera, Pinot Meunier, are separated by passage through somatic embryogenesis the regenerated plants not only have distinct DNA profiles which are different from those of the parent plant but also have novel phenotypes. Recovery of these phenotypes indicates that additional genetic differences can exist between the two cell layers and that the Pinot Meunier phenotype is due to the interaction of genetically distinct cell layers. It appears that grapevine chimerism can not only modify phenotype but can also impact on grapevine improvement as both genetic transformation and conventional breeding strategies separate mutations in the L1 and L2 cell layers. High assimilation sacrum is fusion of the caudal‐most lumbar vertebra to the first sacral vertebra. Previous studies have shown that high assimilation is associated with clinical problems, including obstetrical difficulty. This study used adult American males (n = 1,048) and females (n = 1,038) of the Hamann–Todd and Terry skeletal collections to determine the prevalence of high assimilation and its effect on pelvic size, and to consider the obstetrical and evolutionary implications of high assimilation. The prevalence of high assimilation in this sample is 6.3%, with males and females not differing significantly from one another in their prevalence. This prevalence is near the median for that reported in 41 other samples. In both males and females, individuals with high assimilation have significantly longer anteroposterior and posterior sagittal diameters of the inlet, and shorter sacrum compared to those with a nonassimilated sacrum. Females with high assimilation have a significantly narrower sacral angulation (i.e., reduced inclination of ventral axis of sacrum), and shorter posterior sagittal diameter of the outlet compared to those with a nonassimilated sacrum. A short posterior sagittal diameter of the outlet is associated with childbirth difficulty. As high assimilation is partial homeotic transformation of a lumbar vertebra, this study supports previous research that homeotic transformation of vertebrae is selectively disadvantageous. Levels of carbonic anhydrase activity were determined on a total (60 EUmg?1 protein), external (7.36 EU), internal (50.14 EU) and protoplast (15.63 EU) basis for Ranunculus penicillatus (Dumort.) Bab ssp. pseudofluitans (Syme) S. Webster, a freshwater aquatic macrophyte, by conventional electrometric methods. The site of activity of ‘external’ carbonic anhydrase (CA) has been visualized using 5-Dimethylaminonapthalene-1- sulphonamide (DNSA)-CA fluorescent complex formation, and is postulated to be closely associated with the epidermal cell wall. The photosynthetic rate of R. penicillatus ssp. pseudofluitans at pH 9.0 is in excess of the uncatalysed rate of production of CO2 from HCO?3, and this plant is therefore using HCO?3 for photosynthesis. The possible contribution of CA activity to inorganic carbon assimilation, and specifically to transport of HCO?3, in submerged aquatic plants is discussed.  相似文献

This paper reports on a 20-month study of chimpanzee nesting patterns in Issa, Ugalla, western Tanzania. Ugalla is one of the driest, most open, and seasonal habitats where chimpanzees are found. The methods used were ethoarchaeological, as the chimpanzees were not habituated and behavioural observations were rare. Systematic data on the spatial and temporal distribution of nests are presented. Places with no nests at the beginning of the study, despite being suitable for nesting, were used as controls. Similar to other chimpanzee study sites, nests were highly concentrated in some parts of the landscape. Issa chimpanzees preferred to nest on slopes. They extensively used the woodland vegetation type of their habitat for nesting throughout the annual cycle. Ninety percent of nest sites were used repeatedly throughout the study period, but none of the control places had nests during this period. The results indicate that chimpanzees ranged more widely during the dry season, when food abundance was lowest, food was available mainly in open vegetation types, and when drinking water was restricted to a few sources. Early hominins in similar habitats may have followed the ranging strategy of Issa chimpanzees. As with a previous study, the distribution of nests was spatially similar to archaeological distributions in early hominin sites. Hominin topography and vegetation type preferences may be misrepresented in the archaeological record. Nest sites may have been the antecedents of carcass processing sites.  相似文献

In an effort to understand the relationships between both the lipid content and 13C values of Collembola and their diet, isotopically labelled (C3 and C4) bakers yeasts were cultured and fed to two Collembolan species, Folsomia candida and Proisotoma minuta. The fatty acid composition of Collembola generally reflected that of the diet with the addition of the polyunsaturated components 18:2(n-6), 20:4(n-6) and 20:5(n-3), which appeared to be biosynthesised by the Collembola. Whilst ergosterol was the only sterol detected in the yeast diets, only cholesterol was detected in Collembola, and although the 13C values of diet and consumer sterols differed by >2, the 13C values indicated that cholesterol was derived entirely from dietary sterol. The bulk 13C values of Collembola were similar to those of the diets, but fatty acid 13C values did not necessarily reflect those of the dietary fatty acids, indicating significant de novo biosynthesis of fatty acids within Collembola. Switching the Collembola from C3 to C4 yeast enabled the determination of the rates of incorporation of dietary carbon into Collembolan lipids, and showed that half-lives of the incorporation of dietary carbon varied between 1.5 and 5.8 days at 20°C. Cholesterol exhibited the slowest rate of incorporation in both species, while bulk carbon in F. candida possessed an intermediate rate. These results demonstrate that an understanding of the sources of isotopic fractionation and the role of biochemistry in regulating the 13C values of individual compounds is important in the application of compound-specific isotopic analysis to the study of animal trophic activities.  相似文献

Natural and anthropogenic Iberian wetlands in southern Europe are well known for supporting large numbers of migratory Palaearctic waterbirds each winter. However, information on the geographical origin of dabbling ducks overwintering in these wetlands is scarce and mostly limited to data from ringing recoveries. Here, we used intrinsic isotopic markers to determine the geographical origin of male and female Northern Pintails Anas acuta and Eurasian Teal Anas crecca in Extremadura, inland Iberia, a key site for overwintering dabbling ducks. Additionally, we fitted six Northern Pintails with GPS‐GSM tags to complement the data derived from stable isotope analysis. Most (> 70%) first calendar‐year Northern Pintails were assigned to regions above 55°N, flying 2600–5600 km from their main natal regions to Extremadura. Mean values of δ2Hf varied significantly between male and female Northern Pintails, suggesting that the sexes had different geographical origins. Data from tagged adult Northern Pintails supported the isotopic data, one male flying more than 5000 km to the coast of the Pechora Sea (Russia). Most (> 70%) first calendar‐year Eurasian Teal were assigned to the region between 48° and 60°N, travelling 1500–4500 km to arrive in Extremadura. Male and female Eurasian Teal showed marginal differences in mean values of δ2Hf. In migratory dabbling ducks, pairing typically occurs on the wintering grounds, and ducks in their first winter can breed the following spring. For Northern Pintails, pair formation in Extremadura could occur between individuals with different geographical origins, which could contribute to the genetic variability of their offspring.  相似文献

Like many other migratory birds, the black-throated blue warbler (Dendroica caerulescens) shows pronounced differences in migratory behaviour and other traits between populations: birds in the southern part of the breeding range have darker plumage and migrate to the eastern Caribbean during the winter, whereas those in the north have lighter plumage and migrate to the western Caribbean. We examined the phylogeography of this species, using samples collected from northern and southern populations, to determine whether differentiation between these populations dates to the Pleistocene or earlier, or whether differences in plumage and migratory behaviour have arisen more recently. We analysed variation at 369 bp of the mitochondrial control region domain I and also at seven nuclear microsatellites. Analyses revealed considerable genetic variation, but the vast majority of this variation was found within rather than between populations, and there was little differentiation between northern and southern populations. Phylogeographic analyses revealed a very shallow phylogenetic tree, a star-like haplotype network, and a unimodal mismatch distribution, all indicative of a recent range expansion from a single refugium. Coalescent modelling approaches also indicated a recent common ancestor for the entire group of birds analysed, no split between northern and southern populations, and high levels of gene flow. These results show that Pleistocene or earlier events have played little role in creating differences between northern and southern populations, suggesting that migratory and other differences between populations have arisen very recently. The implications of these results for the evolution of migration and defining taxonomic groups for conservation efforts are discussed.  相似文献

Incubations of allene oxide synthases of flax or maize with the E,E-isomers of the 13- and 9-hydroperoxides of linoleic acid (E,E-13- and E,E-9-HPOD, respectively) at pH 7.5 afforded substantial yields of trans-disubstituted cyclopentenones. Under the conditions used, (Z,E)-HPODs were converted mainly into -ketols and afforded only trace amount of cyclopentenones. These findings indicated that changing the double bond geometry from Z to E dramatically increased the rate of formation of the pericyclic pentadienyl cation intermediate necessary for electrocyclization of 18:2-allene oxides and thus the yield of cyclopentenones. The well-known cyclization of the homoallylic allene oxide (12,13-EOT) derived from -linolenic acid 13-hydroperoxide (E,Z-13-HPOT) into cis-12-oxo-10,15-phytodienoic acid was suppressed at pH below neutral and was not observable at pH 4.5. In contrast, cyclization of the allene oxide ((9E)-12,13-EOD) derived from (E,E)-13-HPOD was slightly favoured at low pH. The finding that the cyclizations of 12,13-EOT and (9E)-12,13-EOD were differently affected by changes in pH suggested that the mechanisms of cyclization of these allene oxides are distinct.  相似文献

This article analyses the Urban Poverty and Family Life Survey of Chicago and an urban subsample of the National Survey of Families and Households to examine race and ethnic variation in the occurrence and consequences of birth versus marriage pathways to family formation. Results based on multiple‐decrement life table analysis and multi‐variate life table regression analysis reveal strong race and poverty effects on pathways to family formation, but Hispanic origin does not systematically influence the odds that women will enter family life via marriage versus birth. We show stronger race effects on pathways to family formation for the Chicago sample owing to the much higher incidence of teen parenting among black inner‐city residents. Results also suggest lasting economic consequences of non‐marital fertility, irrespective of whether women eventually marry or divorce subsequent to a premarital birth.  相似文献

Aims A molecular genetic distance study has been used in an initial survey to identify subspecies and genotypes of the weed Acacia nilotica in Australia, information needed to find suitable biocontrol agents. We use patterns of DNA sequence variation (in two DNA fragments) from each of the nine described subspecies of Acacia nilotica (L.) Delile (Leguminosae: Mimosoideae) that is to determine their genetic similarity, to verify if the Australian populations are A. nilotica ssp. indica (Benth.) Brenan, and to establish if any other subspecies are present in Australia. Location Australia and southern Africa through the Arabian peninsular to the Indo‐Pakistan subcontinent. Methods Representative specimens from the global distribution of the nine A. nilotica subspecies were sourced primarily from herbaria sheet specimens where available, and secondarily from field collections. These specimens together with related outgroups from Mimosoideae were genetically analysed using the DNA fragments trnL and internal transcribed spacer one (ITS1). We calculated a similarity index as set out in paup * using upgma (Unweighted Pair‐Group Method Arithmetic average) methods to cluster taxa to produce a genetic distance phenogram. Results Sequence results from ITS1 and trnL DNA fragments identified seven of the described subspecies of A. nilotica. Acacia nilotica ssp. cupressiformis (J. Stewart) Ali & Faruqi and A. nilotica ssp. adstringens (Schumach. & Thonn.) Roberty were not found to be genotypically distinct from A. nilotica ssp. indica and A. nilotica ssp. nilotica, respectively, based on the two DNA fragments. Subspecific ITS1 genotypes were geographically distributed similarly to previous reports that were based on morphology, with the exception that the hemispherica ITS1 genotype also occurred in Somalia. We confirmed that the Australian A. nilotica populations are mostly comprised of subspecies indica, but in addition, some individuals were found to be genetically identical to an unidentified Pakistan genotype not previously reported as occurring in Australia. Main conclusions Australian A. nilotica populations originated from India and Pakistan and we recommend further analysis to determine the complete genetic diversity profile and origins of the Australian populations. We highlight the importance of determining any hybridization between Australian populations of A. nilotica and native subgenus Acacia species. This study demonstrates the importance of genotyping weed species targeted for biocontrol and/or listed host specificity test species that may be easily misidentified. Biocontrol practitioners can justify genetic studies by considering the costs should a project fail through misidentification.  相似文献

Fibroblast apoptosis plays a crucial role in normal and pathological scar formation and therefore we studied whether the putative apoptosis-inducing factor curcumin affects fibroblast apoptosis and may function as a novel therapeutic. We show that 25-μM curcumin causes fibroblast apoptosis and that this could be inhibited by co-administration of antioxidants N -acetyl- l -cysteine (NAC), biliverdin or bilirubin, suggesting that reactive oxygen species (ROS) are involved. This is supported by our observation that 25-μM curcumin caused the generation of ROS, which could be completely blocked by addition of NAC or bilirubin. Since biliverdin and bilirubin are downstream products of heme degradation by heme oxygenase (HO), it has been suggested that HO-activity protects against curcumin-induced apoptosis. Interestingly, exposure to curcumin maximally induced HO-1 protein and HO-activity at 10–15 μM, whereas, at a concentration of >20-μM curcumin HO-1-expression and HO-activity was negligible. NAC-mediated inhibition of 25-μM curcumin-induced apoptosis was demonstrated to act in part via restored HO-1-induction, since the rescuing effect of NAC could be reduced by inhibiting HO-activity. Moreover pre-induction of HO-1 using 5-μM curcumin protected fibroblasts against 25-μM curcumin-induced apoptosis. On a functional level, fibroblast-mediated collagen gel contraction, an in vitro wound contraction model, was completely prevented by 25-μM curcumin, while this could be reversed by co-incubation with NAC, an effect that was also partially HO-mediated. In conclusion, curcumin treatment in high doses (>25 μM) may provide a novel way to modulate pathological scar formation through the induction of fibroblast apoptosis, while antioxidants, HO-activity and its effector molecules act as a possible fine-tuning regulator.  相似文献

The objective of this study was to quantify spatial and temporal variability of anguillid glass eel ingress within and between adjacent watersheds in order to help illuminate the mechanisms moderating annual recruitment. Because single fixed locations are often used to assess annual recruitment, the intra-annual dynamics of ingress across multiple sites often remains unresolved. To address this question, plankton nets and eel collectors were deployed weekly to synoptically quantify early stage Anguilla rostrata abundance at 12 sites across two New Jersey estuaries over an ingress season. Numbers of early-stage glass eels collected at the inlet mouths were moderately variable within and between estuaries over time and showed evidence for weak lunar phase and water temperature correlations. The relative condition of glass eels, although highly variable, declined significantly over the ingress season and indicated a tendency for lower condition A. rostrata to colonize sites in the lower estuary. Accumulations of glass eels and early-stage elvers retrieved from collectors (one to >1500 A. rostrata per collector) at lower estuary sites were highly variable over time, producing only weak correlations between estuaries. By way of contrast, development into late-stage elvers, coupled with the large-scale colonization of up-river sites, was highly synchronized between and within estuaries and contingent on water temperatures reaching c. 10−12° C. Averaged over the ingress season, abundance estimates were remarkably consistent between paired sites across estuaries, indicating a low degree of interestuary variability. Within an estuary, however, abundance estimates varied considerably depending on location. These results and methodology have important implications for the planning and interpretation of early-stage anguillid eel surveys as well as the understanding of the dynamic nature of ingress and the spatial scales over which recruitment varies.  相似文献

Abstract:

The development of these updated guidelines was commissioned by the AACE, TOS, and ASMBS Board of Directors and adheres to the AACE 2010 protocol for standardized production of clinical practice guidelines (CPG). Each recommendation was re‐evaluated and updated based on the evidence and subjective factors per protocol. Examples of expanded topics in this update include: the roles of sleeve gastrectomy, bariatric surgery in patients with type‐2 diabetes, bariatric surgery for patients with mild obesity, copper deficiency, informed consent, and behavioral issues. There are 74 recommendations (of which 56 are revised and 2 are new) in this 2013 update, compared with 164 original recommendations in 2008. There are 403 citations, of which 33 (8.2%) are EL 1, 131 (32.5%) are EL 2, 170 (42.2%) are EL 3, and 69 (17.1%) are EL 4. There is a relatively high proportion (40.4%) of strong (EL 1 and 2) studies, compared with only 16.5% in the 2008 AACE‐TOS‐ASMBS CPG. These updated guidelines reflect recent additions to the evidence base. Bariatric surgery remains a safe and effective intervention for select patients with obesity. A team approach to perioperative care is mandatory with special attention to nutritional and metabolic issues.  相似文献

Sustained harvest of wild North Americanginseng (Panax quinquefolius L.) for overtwo centuries has led to heightenedconservation concern and a recent interest inthe population genetics of this species. Thisstudy examined allozyme variation from 32 wildand 12 cultivated populations of Americanginseng to: (1) document the amount anddistribution of genetic variation over a wideportion of the species' natural range, (2)examine genetic differences between wild andcultivated populations, and (3) provideindirect estimates of its breeding system. Strong genetic differences between wild andcultivated populations were found in the amountof variation within populations and thedistribution of variation among populations. Wild populations were significantly lower inall within-population diversity measures, butcontained significantly higher levels ofvariation partitioned among populations. Similarities between wild and cultivatedpopulations were also found. As a whole,cultivated and wild groups shared nearly allalleles, and populations of both groups showedstrong homozygote excess compared toexpectations under random mating. Thehomozygote excess is best explained by highlevels of selfing. In wild populations,significant correlations were found betweengenetic diversity and estimated populationsize, and between interpopulation geneticdistance and geographic distance. Overall, theresults for wild populations suggest that theyare influenced by high levels of genetic driftand low migration among populations. Conservation implications for American ginsengare discussed with particular emphases on: (1)the current debate surrounding the existence ofwild populations, (2) the ongoing practice ofintroducing cultivated seed into wildpopulations, and (3) the collection of geneticmaterial for the establishment of breedingprograms.  相似文献

As extinctions continue across the globe, conservation biologists are turning to species reintroduction programs as one optimistic tool for addressing the biodiversity crisis. For repatriation to become a viable strategy, fundamental prerequisites include determining the causes of declines and assessing whether the causes persist in the environment. Invasive species—especially pathogens—are an increasingly significant factor contributing to biodiversity loss. We hypothesized that Batrachochytrium dendrobatidis (Bd), the causative agent of the deadly amphibian disease chytridiomycosis, was important in the rapid (<10 years) localized extirpation of a North American frog (Rana boylii) and that Bd remains widespread among extant amphibians in the region of extirpation. We used an interdisciplinary approach, combining interviews with herpetological experts, analysis of archived field notes and museum specimen collections, and field sampling of the extant amphibian assemblage to examine (1) historical relative abundance of R. boylii; (2) potential causes of R. boylii declines; and (3) historical and contemporary prevalence of Bd. We found that R. boylii were relatively abundant prior to their rapid extirpation, and an increase in Bd prevalence coincided with R. boylii declines during a time of rapid change in the region, wherein backcountry recreation, urban development, and the amphibian pet trade were all on the rise. In addition, extreme flooding during the winter of 1969 coincided with localized extirpations in R. boylii populations observed by interview respondents. We conclude that Bd likely played an important role in the rapid extirpation of R. boylii from southern California and that multiple natural and anthropogenic factors may have worked in concert to make this possible in a relatively short period of time. This study emphasizes the importance of recognizing historical ecological contexts in making future management and reintroduction decisions.  相似文献

Angiogenesis and coronary artery collateral formation can improve blood flow and thereby prevent myocardial ischemia. The role of perivascular fibroblasts in neovascularization remains incompletely understood. Here we investigated the effects of epicardial and myocardial fibroblasts on angiogenesis in vitro by using a serum-free microcarrier-based fibrin gel angiogenesis system. To clearly distinguish between different cell types, we either stained endothelial cells or fibroblasts in the living with 1,1′-dioctadecyl-3,3,3′,3′-tetramethyl-indocarbocyanine-perchlorate (DiI). In cocultures, low numbers of heart fibroblasts stimulated endothelial sprouting, and capillary growth was also induced by fibroblast-conditioned media, indicating a paracrine mechanism. Capillary formation was decreased by increasing the density of fibroblasts in the cocultures, indicating contact-dependent inhibition. Using time-lapse studies, it turned out that close contacts between fibroblasts and endothelial cells resulted in rapid retraction of endothelial cells or, rarely, in cell death. Depending on the local ratio of fibroblasts to endothelial cell numbers, fibroblasts determined the location of capillary growth and the size of developing capillaries and thereby contributed to capillary network remodeling. In contrast to primary heart fibroblasts, NIH 3T3 fibroblasts did not display contact-dependent inhibition of endothelial sprouts. NIH fibroblasts were frequently seen in close association with endothelial capillaries, resembling pericytes. Contact-dependent inhibition of angiogenesis by epicardial fibroblasts could not be reversed by addition of neutralizing anti-TGF-β1 antibodies, by addition of serum, of medium conditioned by hypoxic tumor cells or myocardium, by various cytokines or by growing cocultures under hypoxic conditions. Our results implicate a pivotal role of periendothelial mesenchymal cells for the regulation of microvascular network remodeling and collateral formation. Current models to study Legionella pathogenesis include the use of primary macrophages and monocyte cell lines, various free-living protozoan species and murine models of pneumonia. However, there are very few studies of Legionella spp. pathogenesis aimed at associating the role of biofilm colonization and parasitization of biofilm microbiota and release of virulent bacterial cell/vacuoles in drinking water distribution systems. Moreover, the implications of these environmental niches for drinking water exposure to pathogenic legionellae are poorly understood. This review summarizes the known mechanisms of Legionella spp. proliferation within Acanthamoeba and mammalian cells and advocates the use of the amoeba model to study Legionella pathogenicity because of their close association with Legionella spp. in the aquatic environment. The putative role of biofilms and amoebae in the proliferation, development and dissemination of potentially pathogenic Legionella spp. is also discussed. Elucidating the mechanisms of Legionella pathogenicity development in our drinking water systems will aid in elimination strategies and procedural designs for drinking water systems and in controlling exposure to Legionella spp. and similar pathogens.  相似文献
