We have studied the activity and composition of several geochemically significant physiological groups of bacteria in more than twenty alkaline salt lakes of the north-east Mongolia steppe with water salinity from 3 to 390 g l?1 and pH values ranging from 9.0 to 10.6. Active and diverse microbial communities have been found in most of the lakes. The methanotrophic bacteria were represented by the Type I members. Among the culturable forms of sulfur-oxidizing bacteria obligately chemolithoautotrophic and haloalkaliphilic representatives of the genera Thioalkalimicrobium and Thioalkalivibrio were detected in the sediments at high numbers (up to 106 cells ml?1). The largest population of anaerobic phototrophic bacteria was represented by purple sulfur bacteria of the Ectothiorhodospiraceae family. Salinity was the key factor in determining the activity and the composition of the microbial communities. The most diverse and active prokaryotic populations, including aerobic and anaerobic phototrophic, methanogenic, methanotrophic, sulfur-oxidizing, sulfate-reducing and nitrifying bacteria, were found in lakes with salinity less than 60 g l?1. In hypersaline lakes with a salinity >100 g l?1, the sulfur cycle remained active due to the activity of extremely halotolerant and alkaliphilic sulfur bacteria, while other important functional groups responsible for nitrification and methane oxidation processes were not detected. Overall, the prokaryotic communities of the Mongolian alkaline salt lakes represent an interesting new example of a diverse community of haloalkaliphilic bacteria well adopted to a broad salinity range.  相似文献

Two gram-positive anaerobic bacteria (strains E1H and MLS10) were isolated from the anoxic muds of Mono Lake, California, an alkaline, hypersaline, arsenic-rich water body. Both grew by dissimilatory reduction of As(V) to As(III) with the concomitant oxidation of lactate to acetate plus CO2. Bacillus arsenicoselenatis (strain E1H) is a spore-forming rod that also grew by dissimilatory reduction of Se(VI) to Se(IV). Bacillus selenitireducens (strain MLS10) is a short, non-spore-forming rod that grew by dissimilatory reduction of Se(IV) to Se(0). When the two isolates were cocultured, a complete reduction of Se(VI) to Se(0) was achieved. Both isolates are alkaliphiles and had optimal specific growth rates in the pH range of 8.5–10. Strain E1H had a salinity optimum at 60 g l–1 NaCl, while strain MLS10 had optimal growth at lower salinities (24–60 g l–1 NaCl). Both strains have limited abilities to grow with electron donors and acceptors other than those given above. Strain MLS10 demonstrated weak growth as a microaerophile and was also capable of fermentative growth on glucose, while strain E1H is a strict anaerobe. Comparative 16S rRNA gene sequence analysis placed the two isolates with other Bacillus spp. in the low G+C gram-positive group of bacteria. Received: 21 May 1998 / Accepted: 31 August 1998  相似文献

Anaerobic alkalithermophiles, a novel group of extremophiles   总被引:2,自引:0,他引:2  
Although some anaerobic and aerobic mesophiles have long been known to grow at alkaline pH (above 9.5), little was known until recently about thermophilic alkaliphiles, termed now alkalithermophiles. This minireview describes presently known and recently validly described anaerobic alkalithermophilic bacteria (pHopt 55C > 8.5; Topt > 55°C) and alkalitolerant thermophiles (pHopt 55C < 8.5 but pHmax 55C above 9.0). Some of these are widely distributed, but others have been isolated (thus far) only from one specific location. This novel group of anaerobic bacteria is comprised of physiologically different genera and species which, so far, all belong to the Gram-type positive Bacillus-Clostridium phylogenetic subbranch. An interesting feature of these anaerobic alkalithermophiles is that most of the isolates have short doubling times. The fastest growing among them are strains of Thermobrachium celere, with doubling times as short as 10 min while growing above pH 9.0 and above 55°C. A detailed survey was undertaken of the microbial communities of 16 saline lakes in the Vestfold Hills (Princess Elizabeth Land, eastern Antarctica), which ranged in salinity from slightly brackish (4–5‰) to hypersaline (maximum: 174‰). Temperatures at comparable sampling depths in the lakes ranged from −12.2°C to +10.5°C. Ranges in the abundances of bacteria, heterotrophic nanoflagellates (HNAN) and phototrophic nanoflagellates (PNAN) were 1.40 × 107 l−1–1.58 × 1010 l−1, 4.83 × 104 l−1–1.70 × 107 l−1 and 0–1.02 × 107 l−1, respectively. There was considerable variation across the salinity spectrum, though in the case of bacteria and PNAN significantly higher concentrations of cells were seen in two of the most saline lakes. The autotrophic ciliate Mesodinium rubrum occurred in all but five of the lakes and was found at salinity levels up to 108‰. Heterotrophic ciliates were generally scarce. Dinoflagellates, particularly Gonyaulax c.f. tamarensis, Gyrodinium lachryma and Gymnodinium sp., occurred in the majority of the lakes. On the basis of chlorophyll a concentrations, nutrient levels and microplankton concentrations the lakes spanned the spectrum from ultra-oligotrophic to oligo/mesotrophic. The most saline lakes had much reduced species diversity compared with the less saline environments. More than half of China's lakes are saline (viz. have salinities > 3 g L−1). Most salt lakes are in northwestern China (Tibet, Qinghai, Sinkiang, Inner Mongolia). Most Mongolian salt lakes are in the west of that country. Tectonic movements have been of the greatest importance in lake origins, but aeolian activity and deflation have also played a role. Many salt lakes in Qinghai-Tibet lie at altitudes > 4 000 m.a.s.l.; Aiding Hu (Sinkiang) lies at −154 m.a.s.l. Again, many lakes are large in area and deep. Small, shallow lakes are also common. Dimictic thermal patterns prevail in deep lakes, polymictic patterns in shallow ones. The highest salinity recorded is 555 g L−1. The salinity of Qinghai Lake, the largest Chinese salt lake, is 14 g L−1, but mean lake salinity on the northern Tibetan plateau is about an order of magnitude greater. Lop Nor has a salinity of ∼ 5 g L−1. Dominant ions are Na and Cl; Mg, Ca, SO4 and HCO3 + CO3 are important in certain lakes. Most major ions originate by weathering and leaching from rocks. pH values are generally high (often > 9.0). There are no bird or fish species confined to salt lakes, though many are associated with lakes of low or moderate salinity. Artemia occurs widely inland and in coastal salt pans, but is the only major macroinvertebrate of highly saline lakes. In lakes of only low to moderate salinity, invertebrate communities comprise widespread halotolerant freshwater forms and halophiles, some regionally endemic. Submerged and emergent macrophytes occur in lakes of low salinity, but phytoplankton species are more halotolerant. Ctenocladus circinatus, a green alga, is known from a Tibetan salt lake with a salinity of 200 g L−1. There is a dearth of basic limnological information on Chinese and Mongolian salt lakes. More work in particular is needed on a variety of geographically widespread lakes to (a) document seasonal physico-chemical events, and (b) compile comprehensive biological inventories of taxa present. Chinese salt lakes are significant sites for palaeoclimatic research, for conservation purposes, and for the resolution of several important biological questions (especially of an ecological and biogeographical sort). They also have important economic values. Unfortunately, the natural existence of many appears to be threatened by decreased inflows, largely the result of human impact on catchments.  相似文献

The spring benthos of 22 lakes ranging from 1–88 gl−1 salinity contained 58 species of macroinvertebrates, but only 23 species occurred in waters >3 gl−1. The amphipod Hyalella azteca and the chironomids Procladius freemani, Chironomus nr. muratensis and Cryptochironomus spp. were important at lower salinities (1–12 gl−1) whereas the chironomids Tanypus nubifer, Cricotopus ornatus and Chironomus nr. annularis dominated at moderate salinities (5–30 gl−1) and dolichopodid and ephyrid dipterans were the only species in hypersaline lakes (>50 gl−1). Diversity decreased significantly with increased salinity. Mean dry biomass ranged from 0–9.12 gm−2, showing little correlation with salinity, though hyposaline lakes often had elevated values and hypersaline lakes very low values. Shallow lakes (<5 m) had significantly lower standing crops. There were long term changes in biomass (over 45 years) in some lakes due to cultural eutrophication or secular changes in salinity. Chironomids were by far the dominant contributors to biomass at salinities to 50 gl−1, above which dolichopodid and ephyrid dipterans dominated. The lakes were classified into four groups—subsaline, hyposaline, shallow hypo-mesosaline and hypersaline, reflecting the importance of salinity and also relative depth as major controlling factors.  相似文献

Presently, the food chains of the famous saline alkaline flamingo-lakes of East Africa are the focus of intense scientific discussion as the lakes host toxic cyanobacteria, which when consumed by Lesser Flamingos, weaken the birds and therefore make them susceptible to attacks by infective diseases. The distribution, genetic and toxicological aspects of Microcystis in Kenya has been studied extensively. Although there are reports on the occurrence of Microcystis in Kenya’s hypersaline alkaline lakes, they have not been confirmed. Our investigations carried out over a 10-year period in about 50 inland waters showed that Microcystis occurs exclusively in freshwaters, but never in the hypersaline alkaline lakes. Microscopic examinations of the phytoplankton of these lakes revealed the presence of Anabaenopsis abijatae (Nostococales) whose lumpy structure makes it roughly similar to Microcystis when viewed under an inverted microscope. We conclude that the possible occurrence of Microcystis in hypersaline alkaline lakes is doubtful and, as such, confirmatory studies including microphotographic documentation of findings should be carried out.  相似文献

Soda lakes are naturally occurring highly alkaline and saline environments. Although the sulfur cycle is one of the most active element cycles in these lakes, little is known about the sulfate-reducing bacteria (SRB). In this study we investigated the diversity, activity, and abundance of SRB in sediment samples and enrichment cultures from a range of (hyper)saline soda lakes of the Kulunda Steppe in southeastern Siberia in Russia. For this purpose, a polyphasic approach was used, including denaturing gradient gel electrophoresis of dsr gene fragments, sulfate reduction rate measurements, serial dilutions, and quantitative real-time PCR (qPCR). Comparative sequence analysis revealed the presence of several novel clusters of SRB, mostly affiliated with members of the order Desulfovibrionales and family Desulfobacteraceae. We detected sulfate reducers and observed substantial sulfate reducing rates (between 12 and 423 μmol/dm3 day−1) for most lakes, even at a salinity of 475 g/liter. Enrichments were obtained at salt saturating conditions (4 M Na+), using H2 or volatile fatty acids as electron donors, and an extremely halophilic SRB, strain ASO3-1, was isolated. Furthermore, a high dsr gene copy number of 108 cells per ml was detected in a hypersaline lake by qPCR. Our results indicate the presence of diverse and active SRB communities in these extreme ecosystems.  相似文献

Summary Sodium chloride-tolerant plantlets of Dendrocalamus strictus were regenerated successfully from NaCl-tolerant embryogenic callus via somatic embryogenesis. The selection of embryogenic callus tolerant to 100 mM NaCl was made by exposing the callus to increasing (0–200 mM) concentrations of NaCl in Murashige and Skoog medium having 3% (w/v) sucrose, 0.8% (w/v) agar, 3.0 mg l−1 (13.6 μM) 2,4-dichlorophenoxyacetic acid (2,4-D), and 0.5mg l−1 (2.3μM) kinetin (callus initiation medium). The tolerance of the selected embryogenic callus to 100 mM NaCl was stable through three successive transfers on NaCl-free callus initiation medium. The tolerant embryogenic callus had high levels of Na+, sugar, free amino acids, and proline but a slight decline was recorded in K+ level. The stable 100 mM NaCl-tolerant embryogenic callus differentiated somatic embryos on maintenance medium [MS medium +3% sucrose +0.8% agar +2.0 mg l−1 (9.0 μM) 2,4-D+0.5 mg l−1 (2.3 μM) kinetin] supplemented with different (0–200 mM) concentrations of NaCl. About 39% of mature somatic embryos tolerant to 100 mM NaCl germinated and converted into plantlets in germination medium [half-strength MS+2% sucrose+0.02 mg l−1 (0.1 μM) α-naphthaleneacetic acid +0.1 mg l−1 (0.49 μM) indole-3-butyric acid] containing 100 mM NaCl. Of these plantlets about 31% established well on transplantation into a garden soil and sand (1:1) mixture containing 0.2% (w/w) NaCl.  相似文献

A culture-dependent enrichment technique was used to isolate endo-1,4-β-mannanase–producing fungi from a hypersaline environment. Galactomannan was used as carbon source and resulted in isolation of strains of Scopulariopsis brevicaulis, S. candida, and Verticillium dahliae. The Scopulariopsis isolates were found to be more dominant and could be isolated from consecutive evaporation ponds, whereas Verticillium was only isolated from one pond. The Scopulariopsis strains exhibited only endomannanase activity, whereas Verticillium displayed broad-activity spectrum by secreting endoxylanases and cellulases in addition to endomannanases. S. candida LMK004 and LMK008 produced 7420 and 14750 nkat g−1 biomass, respectively. Endomannanase production in these strains increased with an increase in NaCl concentration up to 10% (w/v), after which both growth and enzyme production was decreased. V. dahliae LMK006 grew and produced up to 5000 nkat g−1 biomass endomannanase in the absence of NaCl. Increased NaCl concentration had a negative effect on this strain. S. brevicaulis LMK002 showed poor endomannanase production but a similar growth trend as the other Scopulariopsis strains. In general, the Scopulariopsis strains exhibited better halotolerance than V. dahliae and could grow in the presence of 20% NaCl on solid medium.  相似文献

Response surface methodology employing central composite design (CCD) was used to optimize fermentation medium for the production of cellulase-free, alkaline xylanase from Streptomyces violaceoruber under submerged fermentation. The design was employed by selecting wheat bran, peptone, beef extract, incubation time and agitation as model factors. A second-order quadratic model and response surface method showed that the optimum conditions for xylanase production (wheat bran 3.5 % (w/v), peptone 0.8 % (w/v), beef extract 0.8 % (w/v), incubation time 36 h and agitation 250 rpm) results in 3.0-fold improvement in alkaline xylanase production (1500.0 IUml−1) as compared to initial level (500.0 IUml−1) after 36 h of fermentation, whereas its value predicted by the quadratic model was 1347 IUml−1. Analysis of variance (ANOVA) showed a high coefficient of determination (R2) value of 0.9718, ensuring a satisfactory adjustment of the quadratic model with the experimental data. The economical and cellulase-free nature of xylanase would enhance its applicability in pulp and paper industry.  相似文献

A survey of 11 sites covering three large (>10 km long, 6 sites) playa lakes and four pans (<1 km2, 5 sites) of the Yarra Yarra salt lake system in the Northern Agricultural Region of Western Australia commenced in 2001. These salt lakes are shallow and ephemeral, with inundation being more regular following winter rainfall, but summer inundation also occurred in 2001. Salinity was generally higher in playas (156–368 g l−1) than pans (30–284 g l−1), but salinity responded noticeably to heavy rainfall events, especially in pans. pH values in the playa lakes (6.68–7.82) were less variable than in the pans (6.81–8.08). The range of dissolved oxygen concentrations was greater in pans (3.7–14.4 mg l−1) than in playas (3.9–8.2 mg l−1). Cationic concentrations generally followed the pattern of sea water cation dominance. Benthic microbial communities comprised either cohesive to loosely mucilaginous mats, or thin films of diatoms. The hydrocarbon utilizing haloarchaea, Haloferax (two strains), Halobacterium and Halococcus from a hypersaline coastal area of the Arabian Gulf, had the potential for resistance and volatilization of Hg2+. Individual haloarchaea resisted up to between 100 and 200 ppm HgCl2 in hydrocarbon free media with salinities between 1 and 4 M NaCl, but only up to between 20 and 30 ppm in a mineral medium containing 3 M NaCl, with 0.5% (w/v) crude oil, as a sole source of carbon and energy. Halococcus and Halobacterium volatilized more mercury than Haloferax. The individual haloarchaea consumed more crude oil in the presence of 3 M NaCl than in the presence of 2 M NaCl. At both salinities, increasing the HgCl2 concentration in the medium from 0 to 20 ppm resulted in decreasing the oil consumption values by the individual haloarchaea. However, satisfactory oil consumption still occurred in the presence of 10 ppm HgCl2. It was concluded that haloarchaea with the combined potential for mercury resistance and volatilization and hydrocarbon consumption could be useful in removing toxic mercury forms effectively from oil free, mercury contaminated, hypersaline environments, and mercury and oil, albeit less effectively, from oily hypersaline environments.  相似文献

This paper summarizes recent data on the occurrence and properties of lithotrophic prokaryotes found in extremely alkaline, saline (soda) lakes. Among the chemolithotrophs found in these lakes the obligately autotrophic sulfur-oxidizing bacteria were the dominant, most diverse group, best adapted to haloalkaline conditions. The culturable forms are represented by three new genera, Thioalkalimicrobium, Thioalkalivibrio and Thioalkalispira in the Gammaproteobacteria. Among them, the genus Thioalkalivibrio was most metabolically diverse, including denitrifying, thiocyanate-oxidizing and facultatively alkaliphilic species. Culturable methane-oxidizing populations in the soda lakes belong to the type I methanotroph group in the Gammaproteobacteria, mostly in the genus Methylomicrobium. The nitrifying bacteria in hyposaline soda lakes were represented by a new species Nitrobacter alkalicus (Alphaproteobacteria), and by an alkaliphilic subspecies of Nitrosomonas halophila (Betaproteobacteria). Both belonged to the low salt-tolerant alkaliphiles. The facultatively autotrophic haloalkaliphilic isolates able to grow with hydrogen as electron donor were identified as representatives of the alpha-3 subclass of the Proteobacteria (aerobic) and of the Natronolimnicola - Alkalispirillum group in the gammaproteobacteria (nitrate-reducing). While all chemolithotrophic isolates from soda lakes belong to the alkaliphiles with a pH optimum for growth around 10, only the sulfur-oxidizing group included species able to grow under hypersaline conditions. This indicates that carbon and nitrogen cycles in the hypersaline alkaline lakes might not be closed.  相似文献

The objective of this study is to describe the biogeochemical characteristics in the waters of Mongolian lakes, particularly those related to parameters limiting phytoplankton growth and the trophic state. Investigations into the distribution of chemical and biological parameters were carried out in the following 18 lakes: Har Us, Har, Hovsgol, Achit, Dalai, Bayan, Tolbo, Holboo, Bust, Sangiyn Dalai, Tunamal, Dorgon, Uureg, Telmen, Hyargas, Uvs, Erkhel and Oygon, all of which showed a wide range of salinity between 0.16 and 24.9 g l−1. Lake water was classified into four types: six fresh (less than 0.5 g l−1 salinity), three subsaline (0.5–3 g l−1), seven hyposaline (3–20 g l−1) and two mesosaline (20–50 g l−1) lakes. Predominant cations and anions in the order of dominance were Ca, Mg > Na > K and HCO3 > SO4, Cl in freshwater lakes, Na > Mg > Ca, K and HCO3, Cl > SO4 in subsaline lakes, and Na > Mg > K, Ca and Cl, SO4 > HCO3 in hyposaline and mesosaline lakes. Nitrogenous and phosphorus nutrients in the waters were low, seemingly caused by the low loads from their watersheds, where the ground was free of vegetation with an extremely low level of human activity. The present investigations revealed some 234 taxa of phytoplankton and 38 of zooplankton. The PC:PN:PP stoichiometric ratio by weight was (22–202):(3–27):1. Phosphorus was assessed as the potential limiting parameter in eight lakes, nitrogen in six and both nutrients in four others. Twelve lakes showed an oligotrophic character, while six were mesotrophic type. The six oligotrophic lakes seemed to be subject to phosphorus limiting phytoplankton growth, four to nitrogen and two to both limitations. In the mesotrophic lakes, on the other hand, phosphorus limitation was verified in two lakes, nitrogen in two others and both in two lakes.  相似文献

Archaeal microorganisms that grow optimally at Na+ concentrations of 1.7 M, or the equivalent of 10% (w/v) NaCl, and greater are considered to be extreme halophiles. This review encompasses extremely halophilic archaea and their growth characteristics with respect to the correlation between the extent of alkaline pH and elevated temperature optima and the extent of salt tolerance. The focus is on poly-extremophiles, i.e., taxa growing optimally at a Na+ concentration at or above 1.7 M (approximately 10% w/v NaCl); alkaline pH, at or above 8.5; and elevated temperature optima, at or above 50°C. So far, only a very few extreme halophiles that are able to grow optimally under alkaline conditions as well as at elevated temperatures have been isolated. The distribution of extremely halophilic archaea growing optimally at 3.4 M Na+ (approximately 20% w/v NaCl) is bifurcated with respect to pH optima, either they are neutrophilic, with a pHopt of approximately 7, or strongly alkaliphilic, with pHopt at or above 8.5. Amongst these extreme halophiles which have elevated pH optima, only four taxa have an optimum temperature above 50°C: Haloarcula quadrata (52°C), Haloferax elongans (53°C), Haloferax mediterranei (51°C) and Natronolimnobius ‘aegyptiacus’ (55°C).  相似文献

The sensitivity of surface waters to acidic deposition is governed by the interaction of catchment geology, soils, topography, land use, climate and atmospheric deposition. Accordingly at the landscape scale, catchment attributes may be used to predict lake chemistry (for example, acid neutralising capacity (ANC), pH, calcium (Ca2+) and dissolved organic carbon (DOC)). Empirical (multiple linear regression) models based on average measured chemistry (2000–2006) for 204 lakes in Nova Scotia (NS) Canada, and their catchment attributes, were used to predict chemistry for all lakes in NS (n = 6104). Damage to aquatic biota, such as loss of species and/or reduced biodiversity has been widely evaluated using critical chemical thresholds commonly based on pH, ANC and Ca2+. The proportion of sensitive lakes in NS (that is, the stock at risk) was estimated as lakes with ANC less than 20 μeq l−1, pH below 6, and Ca2+ less than 75 μeq l−1 (13, 73 and 74%, respectively). Many lakes in NS are characterized by high DOC (>7 mg l−1); in these lakes organic acids contribute to total acidity, making anthropogenic influences difficult to discern. To account for the potential contribution of organic acidity, all lakes with pH below 6 (and DOC < 7 mg l−1) and lakes below a threshold for ANC adjusted for organic acids were quantified; 63% of the lakes fell below either of these thresholds. Despite substantial reductions in sulphur emissions in North America since the 1980s, many lakes in NS remain at risk to acidic deposition.  相似文献

The rates of photosynthesis and dark CO2 fixation were determined in 12 soda lakes of the Kulunda steppe. Characterization of the phototrophic communities was given, and the cell numbers of anoxygenic phototrophic bacteria (APB) were determined. The photosynthetic production in different lakes was substantially different, constituting from 0.01 to 1.32 g C m−2 day−1. The main part of carbon dioxide was assimilated in the process of oxygenic photosynthesis. Anoxygenic photosynthesis was recorded only in 5 of the 12 lakes studied. Its values varied between 0.06 and 0.42 g C m−2 day−1, constituting from 8 to 34% of the total photosynthetic activity. Anoxygenic photosynthesis was revealed in the lakes where the number of APB reached 107–109 CFU cm−3. Dark CO2 fixation constituted 0.01–0.15 g C m−2 day−1. Positive correlation was observed between the primary production value and water alkalinity. No relationship between productivity and water mineralization was revealed in the 30–200 g l−1 range, whereas an increase in salinity above 200 g l−1 suppressed the photosynthetic activity. The mechanisms of influence of the environmental factors on the rate of photosynthesis are discussed.  相似文献

An alkalophilic Streptomyces sp. RCK-SC, which produced a thermostable alkaline pectinase, was isolated from soil samples. Pectinase production at 45 °C in shaking conditions (200 rev min−1) was optimal (76,000 IU l−1) when a combination of glucose (0.25% w/v) and citrus pectin (0.25% w/v) was added along with urea (0.25% w/v) in the basal medium devoid of yeast extract and peptone. All the tested amino acids and vitamins greatly induced pectinase production and increased the specific productivity of pectinase up to 550%. In an immobilized cell system containing polyurethane foam (PUF), the pectinase production was enhanced by 32% (101,000 IU l−1) compared to shake flask cultures. In solid-state cultivation (SSC) conditions, using wheat bran as solid substrate, pectinase yield of 4857 IU g−1 dry substrate was obtained at substrate-to-moisture ratio of 1:5 after 72 h of incubation. The partially purified pectinase was optimally active at 60 °C and retained 80% of its activity at 50 °C after 2 h of incubation. The half life of pectinase was 3 h at 70 °C. Pectinase was stable at alkaline pH ranging from 6.0 to 9.0 for more than 8 h at room temperature retaining more than 50% of its activity.
