The paper demonstrates the applicability of fluorescent microparticles (FMP) for the determination of cladoceran feeding rates. For this purpose we examined the relation between filtering rate, ingestion rate, gut passage time and body length of Daphnia pulex at different food concentrations in laboratory experiments, and diurnal changes of filtering rates of Daphnia longispina in in situ experiments, using FMPs as tracer particles. Good agreement of our results with published data based on cell counts and radiotracer methods proves the suitability of the FMP-method. Potentials and limitations of the method are discussed.  相似文献

Abstract Instar-specific data on head length and body length of Hyalella azteca was obtained from live specimens reared individually at 23 ± 1 °C under a 14L:10D photoperiod and with food ad lib. Both head length and body length were strong predictors of instar number through instars 1 – 10. The relationship between instar number and head length was linear (head length [m] = 104.28 + 61.05[instar]; r 2=0.98, P<0.001), whereas that between instar and body length was not (body length [m] = 1339.28 + 55.82[instar]2; r 2 = 0.97 , P< 0.001). Head length measurements were easier to obtain and were more accurate than body measurements. Both measurements can be obtained without injury to the animal, permitting repeated observations on the same individual.  相似文献

To ascertain the feeding habits of benthic juvenile yellowfin goby Acanthogobius flavimanus, the gut contents of 599 specimens (15–41 mm in standard length, SL), collected on a tidal mudflat in the Tama River estuary throughout the diel cycle, were examined. The major prey items changed from harpacticoid copepods to errant and sedentary polychaetes at ca. 20 mm SL. Prey width increased with fish size. Fish of 26–28 mm SL fed mainly from sunset to morning, with highest feeding intensity during twilight hours and/or high tide. Based on the gut evacuation rate estimated from a forced feeding experiment in the laboratory and data for the diel change of mean gut-content volume in the field, the daily ration of juvenile yellowfin goby (26–28 mm SL) was calculated to be 13.8 mm3 fish−1 day−1. This volume is approximately equivalent to 3.9 individuals of the errant polychaete Ceratonereis erythraeensis (9.7 mm in body length, BL) or 8.1 individuals of the sedentary polychaete Prionospio japonica (14.8 mm BL), both species occurring abundantly on the mudflat during the study.  相似文献

The larva of Discoverichthys (Aulopiformes: Ipnopidae) is described for the first time based on a specimen 39.5 mm in standard length collected in surface waters near the Marianas in the western North Pacific. Despite its remote location from the previous record in eastern North Atlantic (type locality), this larva was identified as Discoverichthys praecox by general agreements of meristic counts and other morphological features. It is characterized by the following possible autapomorphic features: body moderately elongate, with uniformly distended abdomen, terminating in long, stout trailing gut; all fins, particularly the pectoral and pelvic, are extensively produced; body pigmentation is scanty, but all fins except the caudal are polka-dotted distally and covered by unusually thick skin; the skeleton is largely cartilaginous, with poorly differentiated axial components and uniquely expanded dorsal- and anal-fin pterygiophores. Its peculiar morphology is discussed with special reference to transformation events.  相似文献

We have studied some of the changes in the vacuolar shape and size during autolysis of Neurospora crassa mycelium. The fungus was grown in the Vogel's medium in a small fermenter. Microscopic examination of the periodically taken samples was carried out and the measurement of the size of vacuoles and of the hyphal and vacuolar areas were made on camera lucida drawings.Before autolysis the vacuolar radius increased 44%. In the non-circular vacuoles there is an increase in the mean length of the major vacuolar axis from 5.4±3.3 to 9.1±4.8 m (70% increase) before autolysis, and a little more than 50% during autolysis. The minor vacuolar axis undergoes a steady increase in length throughout the whole period of autolysis. About 39% of the total vacuolar area is formed before autolysis sets in, whereas the vacuolar area produced during autolysis amounts to 30%. Adopting as a criterion for autolysis the loss in mycelial dry weight, we can conclude that the process of vacuolation, measured as the increase in vacuolar area in mycelium of Neurospora crassa, takes place at a similar rate before and during autolysis.  相似文献

Infrared (IR) thermography is used increasingly to estimate body temperature in small ectotherms such as insects. We used the thermal behaviour of an agricultural pest, the desert locust, Schistocerca gregaria, as a case study to demonstrate the application of this method to thermal biology. During microbial infection, the desert locust uses environmentally derived heat to elevate its body temperature. This ‘behavioural fever’ delays onset of disease caused by a fungal biopesticide. Understanding the thermal biology of S. gregaria is therefore a prerequisite for the development of a more effective mycoinsecticide. To accurately use IR thermography as a method of temperature measurement, IR data must first be calibrated with body temperature. Here, we identify two major factors which affect the IR data output and hence need to be incorporated into a camera calibration: (1) emissivity, predominantly determined by colour, and (2) the internal temperature of the camera. We demonstrate the limitations of thermocouple-based methods of temperature measurement in comparison to IR thermography. The detail provided by the large data sets revealed for the first time an early onset of fever in S. gregaria during infection with Metarhizium acridum viz. 20–25 h post-inoculation in comparison to the 48 h demonstrated previously.  相似文献

Two sulfate-reducing bacteria (SRB) were isolated from a mixed culture enriched with benzoate obtained from gut homogenate of the soil-feeding higher termite, Cubitermes speciosus. The organisms were vibrioid rods, staining Gram-negative, which performed incomplete substrate oxidation. They differed in several features. The smaller one, strain STp, was motile with a single polar flagellum. This strain differed from Desulfovibrio desulfuricans only by its inability to oxidize malate and pentanol. The bigger one, strain STg, differed from Desulfovibrio giganteus only by its nonmotility and a lower length. It is the first evidence of the presence of SRB in termite gut.  相似文献

随着塑料在人类社会中的普及,越来越多的废弃塑料及其前体物质被遗留在环境中,且其在自然环境中的降解速度十分缓慢,为此寻找有效的降解途径成为亟待解决的科学问题。【目的】探究利用黄粉虫(Tenebrio molitor)幼虫取食聚苯乙烯对其肠道微生物种群及其代谢路径的响应,以期通过食物诱导寻找一条生物降解和利用聚苯乙烯的有效途径。【方法】以聚苯乙烯为唯一食物来源喂饲黄粉虫幼虫,通过测量幼虫存活率和个体的体重来测定其生长发育情况;通过对其肠道内容物进行16S rRNA基因测序,分析其肠道菌群结构的变化;采用京都基因与基因组百科全书(Kyoto encyclopedia of genes and genomes,KEGG)分析法来预测相关功能基因。【结果】取食聚苯乙烯黄粉虫幼虫存活率和体重均下降,聚苯乙烯塑料明显减少;取食聚苯乙烯的黄粉虫幼虫肠道菌群丰度与多样性明显减少,在门水平,取食聚苯乙烯的黄粉虫幼虫肠道的优势菌为变形菌门(Proteobacteria)、软壁菌门(Tenericutes)和厚壁菌门(Firmicutes);在属水平,取食聚苯乙烯的黄粉虫幼虫肠道优势菌为螺旋体菌属(Spiroplasma)、肠杆菌(Enterobacillus)和大肠埃氏-志贺氏菌(Escherichia-Shigella);通过KEGG功能预测,找到与芳香类和烷烃降解功能相关的基因共18种,取食聚苯乙烯组黄粉虫幼虫肠道菌群降解聚苯乙烯相关通路丰度升高,相关基因表达增强。【结论】聚苯乙烯可以为黄粉虫幼虫生长发育提供一定的物质和能量,且能够使其完成一个世代过程;幼虫长时间取食单一食物后,其肠道菌群结构会发生目标性变化,利用KEGG预测能够找到与聚苯乙烯代谢相关的基因,为后续研究工作提供了有价值的依据。  相似文献

Leptodora kindti (Crustacea: Cladocera) is a large species of zooplankton (2–18 mm length) that is exceptionally transparent. This transparency is believed to be a means by which it successfully coexists in lakes with planktivorous fishes. We investigated the gut remains of bluegill (Lepomis macrochirus) that had been feeding on L. kindti and Daphnia (D. galeata and D. retrocurva) in the wild (Lake Zurich, Illinois) and found that bluegill readily preyed on L. kindti as small as 3–5 mm length, and strongly selected L. kindti over Daphnia galeata and Daphnia retrocurva. The large compound eye of L. kindti is one half to one complete order of magnitude larger than Daphnia's eye, consistent with the hypothesis that eye area is an important visual cue for fishes. Moreover, the slope of the relationship between eye area and body length is an order of magnitude shallower in L. kindti than Daphnia, suggesting that eye area has been under stronger negative selection in L. kindti. Results suggest that L. kindti's large and dark eye compromises the transparent nature of its body.  相似文献

We studied the gut morphology of the ice rat Otomys sloggetti robertsi, a non-hibernating murid rodent endemic to the sub-alpine and alpine regions of the southern African Drakensberg and Maluti mountains. The gut structure of O. s. robertsi is well adapted for a high fibre, herbivorous diet, as is the case with other members of its subfamily Otomyinae. Despite the broad similarity in gross gut morphology with mesic- and arid-occurring otomyines, O. s. robertsi has a larger small intestine, caecum, stomach volume and parts of the colon, which we suggest are adaptations for increased energy uptake and/or poor diet quality in alpine environments. However, O. s. robertsi has a smaller larger intestine than other otomyines, perhaps because it occupies a mesic habitat. Seasonal sexual differences occurred, with females increasing dimensions of the stomach, small intestine length, caecum, and large intestine in summer. Sexual asymmetry in gut morphology may be related to increased energy requirements of females during pregnancy and lactation, indicating phenotypic plasticity in response to poor quality vegetation and a shorter growing season in alpine habitats.Communicated by G. 动物胃肠道是食物消化和营养吸收器官,对机体健康至关重要。果蝇与哺乳动物的肠道在细胞组成、遗传调控等方面高度相似,是研究肠道发育的良好模型。体外培养细胞中的研究发现,Nprl2通过作用于Rag GTPase,抑制雷帕霉素靶点复合物1(target of rapamycin complex 1,TORC1)的活性,参与细胞代谢的调节。前期报道nprl2突变果蝇具有前胃增大、消化能力降低等肠道衰老相关表型。但对于Nprl2是否通过Rag GTPase调控肠道发育等方面尚不清楚。为了探究Rag GTPase在Nprl2调控果蝇肠道发育中的作用,本研究利用遗传杂交结合免疫荧光等方法对RagA敲减和nprl2突变果蝇的肠道形态、肠道细胞组成等方面进行研究。发现单独敲减RagA可以引起肠变粗、前胃增大等表型,敲减RagA能挽救nprl2突变体中肠道变细、分泌型细胞减少的表型,但并不能挽救nprl2突变体中前胃增大的表型。以上结果表明,RagA在肠道发育中发挥重要作用,Nprl2通过作用于Rag GTPase调节肠道细胞分化和肠道形态,但Nprl2对前胃发育和肠道的消化功能的调节可能通过不依赖于Rag GTPase的机制实现。  相似文献

The quantification of nutritional status (e.g., total body fat) of animals is difficult, because the linear dimension (body length) required for the calculation of proxy parameters, such as the physique or body mass indices, cannot be measured without capture. One solution is photogrammetry of body length, provided the following two criteria are met: (1) the camera axes and subject are oriented vertically, and (2) anatomical landmarks are easily identified with low measurement error. By modifying Mori’s (Primates 20:371–397, 1979) approach, we devised an accurate photogrammetric method that uses a horizontal bar with an attached ruler for the monkey to traverse, and the anatomical landmarks of the eye and upper border of the ischial callosity to measure body length. We tested the applicability of this method on 11 adult female, habituated, free-ranging Japanese macaques (Macaca fuscata). Somatometric body length (crown–rump length and anterior trunk length) was statistically compared with the body length obtained using photogrammetry. The significant correlation of the photogrammetric body length with each somatometric measurement verified that the former could be employed to calculate various indices that are used to characterize fat mass (nutritional status) in Japanese macaques. The advantages and disadvantages of photogrammetry are also discussed.  相似文献

A new genus, Papillonema gen.n., is erected to accomodate the two species P. danieli gen. et sp. n. and P. clavatum (Gerlach, 1957) comb.n. from intertidal sediments of a tropical mangrove. Papillonema gen.n. is characterized by prominent papilliform labial sensillae, an elongate muscular terminal bulb (up to 60% of pharyngeal length), and three precloacal supplements. Larvae of the tobacco budworm,Heliothis virescens, are resistant to high levels of ingested 20-hydroxyecdysone which could cause potential inhibition to the development of many other lepidopteran species. This resistance is attributed to the ability of the larvae to metabolize this molting hormone to its 22-acyl ester forms. When tobacco budworm larvae were fed large quantities of 20-hydroxyecdyone, the hormonal metabolites were found in gut and fat body tissues. When incubated with 20-hydroxyecdysone gut tissue converted 20-hydroxyecdysone into its 22-acyl ester metabolites. Lumen site of the midgut was found to be the major location of this bio-transformation. In contrast, fat body tissue failed to convert 20-hydroxyecdysone to 22-acyl ester metabolitesin vitro. After the oral injection of3H-ecdysone, the major metabolites formed were ecdysone 22-acyl esters whereas the majority of3H-ecdysone was transformed to polar metabolites after it was injected into the hemocoel of the larvae. Similar distributions of ecdysteroid 22-O-acyltransferase and alkaline phosphatase activity in subcellular fractions demonstrates the co-localization of these enzymes in plasma membrane of the gut epithelial cells. These results suggest that gut brush border membrane is the major site of ecdysteroid 22-acyl ester formation inH. virescens larvae.  相似文献

【背景】昆虫是世界上种类最多、肠道菌群资源最丰富且多样的动物类群之一。昆虫肠道微生物具有产生活性次级代谢产物的能力,是活性天然产物的重要来源。【目的】研究药用昆虫喙尾琵琶甲(Blaps rynchopetera)成虫肠道来源链霉菌(Streptomyces sp.) BPA71的次级代谢产物及其生物活性。【方法】利用正相硅胶柱色谱、葡聚糖凝胶Sephadex LH-20柱色谱等方法分离纯化该菌株的发酵粗提物,采用牛津杯法进行抗菌活性追踪,确定抗菌活性部位,通过ESI-MS、NMR等波谱数据分析对化合物结构进行鉴定,采用微量肉汤稀释法测定最低抑菌浓度(minimal inhibitory concentration, MIC),采用MTS法测定抗肿瘤活性。【结果】从Streptomyces sp. BPA71的固体发酵提取物中共分离得到4个已知化合物,通过对比核磁数据确定为糠酸甲酯(1)、吡咯甲酰胺A (2)、吡咯甲酰胺B (3)和吲哚-3-乙酸甲酯(4)。抗菌活性结果显示化合物2具有广谱抗菌活性。此外,化合物2对宫颈癌细胞HeLa、肺癌细胞A549、肝癌细胞SMMC-7721、乳腺癌细胞MDA-MB-231和结肠癌细胞SW480这5株肿瘤细胞均有明显的抑制活性。【结论】喙尾琵琶甲肠道来源Streptomyces sp. BPA71可产生丰富的生物活性物质,该研究结果为进一步挖掘喙尾琵琶甲肠道链霉菌的活性天然产物奠定了基础,同时丰富了人们对喙尾琵琶甲肠道微生物的认识。  相似文献

Acartia spp. are the dominant copepod species in the Gironde estuary, seaward of the turbidity maximum area. Acartia bifilosa develop a large population in spring and early summer whereas Acartia tonsa appear in late summer. High values and high variability of chlorophyll a/suspended particulate matter ratio are found seaward of the turbidity maximum area. Feeding rates of A. bifilosa were measured by fluorometry. Phytoplankton ingestion was found to be highly variable, between 8 to 80% of copepod carbon body weight. Except for adult females, copepods were heavier in summer than in winter. PB ratios, estimated by the instantaneous growth rate method, varied from 0.03 d–1 to 0.14 d–1. The gut contents and P/B ratios of Acartia bifilosa were related to chl a/SPM ratio. From those data, and a few obtained for A. tonsa, it is concluded that only in summer months phytoplankton ingestion is enough to maintain secondary production.  相似文献

The present work investigated the effect of Morinda lucida (M. lucida) extract on isolated uterine smooth muscle of pregnant and non-pregnant mice. Pregnant and non-pregnant mice were pretreated with oral stilboesterol (0.1 mg/kg body weight) and killed by cervical dislocation. Thin strips of the uterus were cut and mounted in a 20ml organ bath containing De Jalon solution bubbled with 95%O2-5% CO2 gas mixture. The strips were connected to a force transducer coupled to a Grass 7D Polygraph for the recording of isometric tension. Effects of graded concentrations of oxytocin (OXY; 10-5-10-2 mol/L), acetylcholine (ACh; 10-9-10-5 mol/L) and M. lucida extract (0.015-1.5 mg/ml) were recorded. Fresh uterine strips were then incubated with M. lucida extract for 5mins and cumulative response to OXY was repeated. Another set of fresh strips was incubated in L-NAME for 15mins and the cumulative responses to M.lucida extract were repeated. OXY resulted in increased contractile responses in both pregnant and non-pregnant uterine muscles. M. lucida resulted in relaxation of the uterine smooth muscle in both pregnant and non-pregnant mice at all doses. However, at 1.5mg/ml, M. lucida completely blocked spontaneous uterine contractions. Following incubation with L-NAME, M. lucida extract led to a slightly greater relaxation of the uterine strips. In conclusion, M. lucida reduced contractility of uterine smooth muscle in both pregnant and non-pregnant mice as well as blocking contractile responses to OXY and Ach in uterine smooth muscle of pregnant and non-pregnant mice. There was no significant alteration of M. lucida activity by L-NAME suggesting that the action of the compound on uterine muscle is not associated with impaired nitric oxide synthase.  相似文献

The gut tissues and associated nervous system of the African migratory locust, Locusta migratoria, were found to contain FMRFamide-like immunoreactive (FLI) material throughout the five larval instars and 2 weeks into the adult stage in both males and females. FMRFamide-like immunoreactivity associated with the locust gut was described using camera lucida techniques. FMRFamide-like immunoreactivity is observed in the frontal connectives, recurrent nerve, and oesophageal nerves; projections from the ingluvial ganglion onto the anterior midgut, and from the proctodeal nerve onto the hindgut and posterior midgut; in the neuropils of the frontal ganglion, hypocerebral ganglion and ingluvial ganglia; 30 cell bodies in the frontal ganglion; multipolar sensory cells on the foregut; and endocrine-like cells in the gastric caecae and midgut. Radioimmunoassay (RIA) was used to determine the quantities of FLI material in foreguts, gastric caecae, anterior and posterior midguts, and hindgut of first-fifth instar larvae, 1-3- and 14-17-day male and female adult locusts. As expected, as the tissue size (assessed by total protein content) increases, so does the amount of FLI material in each tissue. Normalizing for tissue size reveals significant differences in FLI content among the stages for each tissue tested. Reversed phase-high pressure liquid chromatography (RP-HPLC) followed by RIA has identified four groups of FLI fractions present in the gut, and different members of these groups are present in the various gut tissues.  相似文献
