Larvicidal activity of essential oil and isolated compounds from Clausena dentata leaves were tested against early fourth instar Aedes aegypti larvae. GC–MS analysis of essential oil revealed the presence of fourteen compounds of which the major compounds were sabinene (21.27%), biofloratriene (19.61%), borneol (18.34%) and β-bisabolol (17.68%). The essential oil of C. dentata exhibited significant larvicidal activity, with 24 h LC50 and LC90 values of 140.2 and 341.6 mg/l, respectively. Larvicidal activities of the four major compounds of essential oil were also tested. The LC50 values of sabinene, biofloratriene, borneol and β-bisabolol were 27.3, 47.4, 43.5 and 33.2 mg/l, respectively. Results of this study show that the leaf essential oil of C. dentata and its four major compounds may be a potent source of natural larvicides.  相似文献

Developing efficient controlled release system of insecticide can facilitate the better use of insecticide. We described here a first example of photo-controlled release of an insecticide by linking fipronil with photoresponsive coumarin covalently. The generated coumarin-fipronil (CF) precursor could undergo cleavage to release free fipronil in the presence of blue light (420 nm) or sunlight. Photophysical studies of CF showed that it exhibited strong fluorescence properties. The CF had no obvious activity against mosquito larvae under dark, but it can be activated by light inside the mosquito larvae. The released Fip from CF by blue light irradiation in vitro retained its activity to armyworm (Mythimna separate) with LC50 value of 24.64 μmol L?1. This photocaged molecule provided an alternative delivery method for fipronil.  相似文献

The mosquito Aedes aegypti is the vector agent responsible for the transmission of yellow fever and dengue fever viruses to over 80 million people in tropical and subtropical regions of the world. Exhaustive efforts have lead to a vaccine candidate with only 30% effectiveness against the dengue virus and failure to protect patients against the serotype 2. Hence, vector control remains the most viable route to dengue fever control programs. We have synthesized a class of 1,2,4-oxadiazole derivatives whose most biologically active compounds exhibit potent activity against Aedes aegypti larvae (ca. of 15 ppm) and low toxicity in mammals. Exposure to these larvicides results in larvae pigmentation in a manner correlated with the LC50 measurements. Structural comparisons of the 1,2,4-oxadiazole nucleus against known inhibitors of insect enzymes allowed the identification of 3-hydroxykynurenine transaminase as a potential target for these synthetic larvicides. Molecular docking calculations indicate that 1,2,4-oxadiazole compounds can bind to 3-hydroxykynurenine transaminase with similar conformation and binding energies as its crystallographic inhibitor 4-(2-aminophenyl)-4-oxobutanoic acid.  相似文献

A series of cinnamic acid derivatives and its heteroaromatic ring analogues were synthesized and evaluated for acaricidal activity in vitro against Psoroptes cuniculi, a mange mite. Among them, eight compounds showed the higher activity with median lethal concentrations (LC50) of 0.36–1.07 mM (60.4–192.1 µg/mL) and great potential for the development of novel acaricidal agent. Compound 40 showed both the lowest LC50 value of 0.36 mM (60.4 µg/mL) and the smallest median lethal time (LT50) of 2.6 h at 4.5 mM, comparable with ivermectin [LC50 = 0.28 mM (247.4 µg/mL), LT50 = 8.9 h], an acaricidal drug standard. SAR analysis showed that the carbonyl group is crucial for the activity. The type and chain length of the alkoxy in the ester moiety and the steric hindrance near the ester group significantly influence the activity. The esters were more active than the corresponding thiol esters, amides, ketones or acids. Replacement of the phenyl group of cinnamic esters with α-pyridyl or α-furanyl significantly increase the activity. Thus, a series of cinnamic esters and its heteroaromatic ring analogues with excellent acaricidal activity emerged.  相似文献

A preliminary study was conducted to delineate the ameliorating effect of dietary zinc nanoparticles (Zn-NPs) against thermal stress in Pangasius hypophthalmus reared under concurrent exposure to lead (Pb) and elevated temperature (34 °C). Three diets were formulated such as control (no Zn-NPs), Zn-NPs 10 and 20 mg/kg diet. Two hundred and thirty four fish were randomly distributed in to six treatments groups in triplicates; such as control group (no Zn-NPs in diet and unexposed to Pb and temperature, Ctr/Ctr), control diet with concurrent exposure to Pb and temperature (Pb-T/Ctr), Zn-NPs 10 and 20 mg/kg without stressors (Zn-NPs 10 mg/kg, Zn-NPs 20 mg/kg), Zn-NPs 10 and 20 mg/kg diet with concurrent exposure to Pb and temperature (Pb-T/Zn-NPs 10 mg/kg, Pb-T/Zn-NPs 20 mg/kg). The Pb in treated water was maintained at the level of 1/21th of LC50 (4 ppm) at 34 °C temperature in stressors groups. Post 60 days feeding trial, critical thermal minimum (CTmin), lethal thermal minimum (LTmin), and critical thermal maximum (CTmax), lethal thermal maximum (LTmax) and biochemical attributes on P. hypophthalmus were evaluated. The results indicated that, dietary supplementation of Zn-NPs increased the CTmin, LTmin and CTmax, LTmax in P. hypophthalmus. Positive correlations were observed between CTmin LTmin (Y = − 0.495 + 10.08x, R2, 0.896) and CTmax LTmax (Y = − 0.872 + 4.43x, R2, 0.940). At the end of the thermal tolerance study, oxidative stress and lipid peroxidation (LPO) were significantly reduced and neurotransmitter enzyme was significantly increased in the groups fed with Zn-NPs @ 10 mg and 20 mg/kg diet. Overall results indicated that dietary Zn-NPs can confer protection against thermal stress in P. hypophthalmus.  相似文献

The present study focused on extracting green larvicides from extracts of the combination of Foeniculum vulgare and Matricaria chamomilla using different solvents of increasing polarity in a Soxhlet extractor and evaluating their ovicidal, larvicidal, and cytotoxic activities. The most promising among all tested extracts was hexane extract. The ovicidal activity of the hexane PH2 extract resulted in a significant (p < 0.05) decrease in egg hatchability from 95.00 ± 6.16% to 15 ± 9.04% at doses ranging from 62.5 to 500 µg/mL. The larval mortality with the hexane extract ranged from 13.33 ± 3.3% to 93.33 ± 3.3% at doses ranging from 31.25 to 250 µg/mL, respectively. The LC50 and LC90 values of the larvicidal activity of the hexane extract were estimated to be 148.3 and 242.17 µg/mL, respectively, after 24 h of exposure. Similarly, the LC50 values after 48 and 72 h of exposure were 124.93 and 100.3 µg/mL, respectively, against the third instar of Cx. pipiens. PH2 treatment of larvae resulted in histopathological changes such as degenerated epithelial cells and destruction of microvilli on the epithelial cells. The PH2 extract achieved a dose-dependent decrease in the rate of cell survival. The IC50 value of PH2-treated HUVECs was 192.07 µg/mL after 24 h of incubation. The cells showed changes in cellular and nuclear morphology. In conclusion, the hexane extract of PH2 could be used in mosquito management programs.  相似文献

We evaluated in the present study the effect of deltamethrin resistance on the fitness cost of the filed populations of Anopheles labranchiae. A susceptible population was used as reference to do different comparisons. We selected the most resistant larvae population collected from northern Tunisia. Eggs were used for study of life history traits including developmental time, larvae mortality, fertility, hatchability and adult sex-ratio. Our results showed that deltamethrin resistance affected negatively (p < 0.05) the developmental time with the median range of 70 h, mortality with the rate of 7 folds in resistant population and hatchability which are lower than in susceptible population. Whereas, no significant differences were detected in adult sex-ratio and fertility of the two studied populations. Our results could help to determine the evolution of population dynamics of the resistant studied population in the areas where insecticide resistance is reported and resistance management is needed.  相似文献

The effects of magnesium (Mg) supplementation on the growth performance, oxidative damage, DNA damage, and photosynthetic pigment synthesis, as well as on the activity level of carbonic anhydrase (CA), ribulose-1,5-bisphosphate carboxylase (Rubisco), and antioxidant enzymes were studied in Vicia faba L. plants exposed to heat stress (HS) and non-heat-stress (non-HS) conditions. Seeds were grown in pots containing a 1:1 mixture of sand and peat, with Mg treatments. The treatments consisted of (i) 0 Mg and non-HS (ambient temperature; control); (ii) 50 mM Mg; (iii) HS (38 °C); and (iv) 50 mM Mg and HS (38 °C). HS was imposed by placing potted plants in an incubator at 38 °C for 48 h. Growth attributes, total chlorophyll (Total Chl), and CA, and Rubisco activity decreased in plants subjected to HS, whereas accumulation of organic solutes [proline (Pro) and glycine betaine (GB)]; superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activity; DNA damage; electrolyte leakage (EL); and malondialdehyde (MDA) and hydrogen peroxide (H2O2) content all increased. Application of Mg, however, significantly enhanced further proline (Pro), glycinebetaine (GB), SOD, POD, and CAT activity, and decreased DNA damage, EL, and MDA and H2O2 concentrations. These results suggest that adequate supply of Mg is not only essential for plant growth and development, but also improves plant tolerance to HS by suppressing cellular damage induced by reactive oxygen species through the enhancement of the accumulation of Pro and GB, and the actions of antioxidant enzymes.  相似文献

A new strain of Manila clam with orange shell color was produced after selection within a full-sib family for two generations. In the present study, the shell length, height, and width, and the live body weight of the orange strain were measured, and their correlation coefficients were calculated. The shell morphological traits were used as independent variables, and the live body weight was used as the dependent variable for calculating the path coefficients, correlation index, and determination coefficients. The results showed that the correlation coefficients between each shell morphological trait and the live body weight were all highly significant (P < 0.01). The correlation indices (R2) of morphological traits against the live body weight of clams were larger than 0.85, indicating that the morphology traits were the main factors affecting the body weight. Multiple regression equations were obtained to estimate shell length X1 (cm), shell height X2 (cm), and shell width X3 (cm) against live body weight Y (g): Y = ? 2.62 + 0.34 X1 + 0.145 X2, (X1 < 0.05, X2 < 0.05). The results suggest that the shell length could be used as the main trait for selective breeding and could indirectly make a large improvement in the weight trait.  相似文献

The bio-efficacy of Aloe vera leaf extract and bacterial insecticide, Bacillus sphaericus larvicidal activity was assessed against the first to fourth instars larvae of Aedes aegypti, under the laboratory conditions. The plant material was shade dried at room temperature and powdered coarsely. A. vera and B. sphaericus show varied degrees of larvicidal activity against various instars larvae of A. aegypti. The LC50 of A. vera against the first to fourth instars larvae were 162.74, 201.43, 253.30 and 300.05 ppm and the LC90 442.98, 518.86, 563.18 and 612.96 ppm, respectively. B. sphaericus against the first to fourth instars larvae the LC50 values were 68.21, 79.13, 93.48, and 107.05 ppm and the LC90 values 149.15, 164.67, 183.84, and 201.09 ppm, respectively. However, the combined treatment of A. vera + B. sphaericus (1:2) material shows highest larvicidal activity of the LC50 values 54.80, 63.11, 74.66 and 95.10 ppm; The LC90 values of 145.29, 160.14, 179.74 and 209.98 ppm, against A. aegypti in all the tested concentrations than the individuals and clearly established that there is a substantial amount of synergist act. The present investigation clearly exhibits that both A. vera and B. sphaericus materials could serve as a potential larvicidal agent. Since, A. aegypti is a container breeder vector mosquito this user and eco-friendly and low-cost vector control strategy could be a viable solution to the existing dengue disease burden. Therefore, this study provides first report on the mosquito larvicidal activity the combined effect of A. vera leaf extract and B. sphaericus against as target species of A. aegypti.  相似文献

To determine the interactions between temperature and cadmium on zebrafish (Danio rerio) development, fertilized eggs were exposed to combinations of three temperature levels (21 °C, 26 °C, and 33 °C) and six cadmium concentrations (0, 0.25, 0.5, 2.0, 5.0, and 10.0 mg/L). Endpoints used included LC50 value (48 h), developmental rate, mortality, heart rate, hatching success, liver histopathology, embryo abnormalities, and heat shock protein (hsp) induction. Results showed a significant acceleration in the developmental rate with increasing temperature and irrespective of the presence of cadmium. Data on LC50 and ELS-test revealed that simultaneous exposure to both cadmium ions and cold stress (21 °C) was highly detrimental to growing embryos, causing a pronounced mortality and a significant reduction in average heart rate and embryo hatchability. In contrast, no similar reactions to cadmium were observed in pre-hatched embryos exposed to both control (26 °C) and high temperature (33 °C), and this can be explained by the significantly higher expression of hsp (hsp70) in embryos at these temperatures. Upon hatching, however, the larvae showed increased sensitivity to cadmium. The severity of malformations in the post-hatched larvae was in the order: hot cadmium stress>cold cadmium stress>cadmium stress alone>no stress at all. Liver histopathology as well as depletion in glycogen reserves exhibited greater severity with increasing cadmium concentration, irrespective of temperature. The present study confirms that temperature effectively confounds cadmium toxicity and needs to be considered for the accurate prediction and assessment of cadmium-induced toxicity in fish.  相似文献

In healthy pancreatic islets, glucose-stimulated changes in intracellular calcium ([Ca2+]i) provide a reasonable reflection of the patterns and relative amounts of insulin secretion. We report that [Ca2+]i in islets under stress, however, dissociates with insulin release in different ways for different stressors. Islets were exposed for 48 h to a variety of stressors: cytokines (low-grade inflammation), 28 mM glucose (28G, glucotoxicity), free fatty acids (FFAs, lipotoxicity), thapsigargin (ER stress), or rotenone (mitochondrial stress). We then measured [Ca2+]i and insulin release in parallel studies. Islets exposed to all stressors except rotenone displayed significantly elevated [Ca2+]i in low glucose, however, increased insulin secretion was only observed for 28G due to increased nifedipine-sensitive calcium-channel flux. Following 3–11 mM glucose stimulation, all stressors substantially reduced the peak glucose-stimulated [Ca2+]i response (first phase). Thapsigargin and cytokines also substantially impacted aspects of calcium influx and ER calcium handling. Stressors did not significantly impact insulin secretion in 11 mM glucose for any stressor, although FFAs showed a borderline reduction, which contributed to a significant decrease in the stimulation index (11:3 mM glucose) observed for FFAs and also for 28G. We also clamped [Ca2+]i using 30 mM KCl + 250 μM diazoxide to test the amplifying pathway. Only rotenone-treated islets showed a robust increase in 3–11 mM glucose-stimulated insulin secretion under clamped conditions, suggesting that low-level mitochondrial stress might activate the metabolic amplifying pathway. We conclude that different stressors dissociate [Ca2+]i from insulin secretion differently: ER stressors (thapsigargin, cytokines) primarily affect [Ca2+]i but not conventional insulin secretion and ‘metabolic’ stressors (FFAs, 28G, rotenone) impacted insulin secretion.  相似文献

Desert truffles are seasonal and important edible fungi that grow wild in many countries around the world. Truffles are natural food sources that have significant compositions. In this work, the antioxidant, chemical composition, anticancer, and antiangiogenesis properties of the Terfezia claveryi truffle were investigated. Solvent extractions of the T. claveryi were evaluated for antioxidant activities using (DPPH, FRAP and ABTS methods). The extracts cytotoxicity on the cancer cell lines (HT29, MCF-7, PC3 and U-87 MG) was determined by MTT assay, while the anti-angiogenic efficacy was tested using ex-vivo assay. All extracts showed moderate anticancer activities against all cancer cells (p < 0.05). The hexane extract inhibited the brain cell line (U-87 MG) with an IC50 of 50 μg/ml and significantly promoted cell apoptosis through the mitochondrial pathway and DNA fragmentation p < 0.001. The ethanol extract demonstrated potent antioxidants; DPPH, FRAP, and ABTS with an IC50 value of 52, 48.5 and 64.7 μg/ml, respectively. In addition, the hexane and ethyl acetate extract significantly (p < 0.001) inhibited the sprouting of microvessels by 100% and 81.2%, at 100 μg/ml, respectively. The GC analysis of the most active extract (hexane) showed the presence of several potent phytochemicals such as stigmasterol, beta-Sitosterol, squalene, lupeol, octadecadienoic acid, and oleic acid.  相似文献

The dichloromethane/methanol (1:1) extract of the stem bark of Millettia usaramensis subspecies usaramensis was tested for its larvicidal activity against the 4th instar Aedes aegypti larvae and demonstrated activity with LC50 value of 50.8 ± 0.06 μg/mL at 48 h. Compounds isolated from the extract were also tested for their larvicidal activities, and the rotenoid usararotenoid-A (LC50 4.3 ± 0.8 μg/mL at 48 h) was identified as the most active principle. This compound appears to be the first rotenoid having a trans-B/C ring junction and methylenedioxy group at C-2/C-3 with high larvicidal activity. Related rotenoids with the same configuration at the B/C-ring junction did not show significant activity at 100 μg/mL.  相似文献

The objective of this study was to elucidate the impact of lactation stage on milk composition, hematological and biochemical parameters of dairy Baladi goats under Egyptian conditions. Forty-eight Baladi goats (32.8 ± 2.9 kg of BW) were enrolled in the current study. The lactation period has been divided into three stages; early (DIM less than 80 days), Mid (DIM 80–140 days), and Late (DIM over 140 days). Baladi goats had decreased daily-MY at a rate of 18.4% and 31.9% at mid and late stages of lactation, compared with early stage, respectively (p = 0.001). Furthermore, lactose% decreased significantly with progress of lactation (p = 0.017). Total solids%, however, decreased significantly at early stage of lactation in comparison with mid and late stages (p = 0.022). On the contrary, no significant differences were found in protein, fat and SNF percentages at different stages of lactation (p = 0.836, 0.625 and 0.281, respectively). Serum glucose and total protein were significantly reduced at late stage of lactation in comparison with early and mid stages (p = 0.001 and 0.001, respectively). On the contrary, no significant differences were found for erythrocytes count, hemoglobin, serum cholesterol, catalase and triiodothyronine at different stages of lactation. There were high and positive correlations between daily-MY and serum total protein (r = 0.87, P < 0.01) and triiodothyronine (r = 0.41, P < 0.01). However, negative estimates were reported between daily-MY and triglycerides (r = ?0.55, P < 0.01) and cholesterol (r = ?0.33, P < 0.05). Our results indicate that dairy Baladi goats produce milk with relatively stable protein, fat and solid not fat (SNF) contents at the different stages of lactation, encouraging the continuous utilization of their milk in processing. Also, dairy Baladi goats seem able to maintain the most vital biochemical parameters.  相似文献

Hydro-distilled essential oil from Kenyan Piper capense (Piperaceae) was analysed by gas chromatography mass spectrometry (GC–MS) and evaluated for larvicidal activity against the malaria vector, Anopheles gambiae. The oil consisted mainly of sesquiterpene hydrocarbons which accounted for 43.9% of the oil. The major sesquiterpenes were δ-cadinene (16.82%), β-bisabolene (5.65%), and bicyclogermacrene (3.30%). The oil also had appreciable amounts of monoterpene hydrocarbons (30.64%), including β-pinene (7.24%) and α-phellandrene (4.76%), and arylpropanoids (8.64%), including myristicin (4.26%). The oil showed larvicidal activity against third instar larvae of A. gambiae, with LC50 and LC90 values of 34.9 and 85.0 ppm, respectively. Most of the larvae died within the first few hours. The high larvicidal activity of this oil was indicated by the fact that over 80% mortality was observed at a concentration of 100 ppm after 24 h. These results compared favourably with the commercial larvicide pylarvex® which had LC50 and LC90 values of 3.7 and 7.8 ppm, respectively. Application of this oil or of products derived from it to larval habitats may lead to promising results in malaria and mosquito management programmes.  相似文献

Two strains (pedersoliT and girotti) of a new species of bacteria were isolated from the preen glands of wild Egyptian geese (Alopochen aegyptiacus) from the river Neckar in southern Germany in two subsequent years. The strains were lipophilic, fastidious, Gram-positive rods and belonged to the genus Corynebacterium. Phylogenetically, the isolates were most closely related to Corynebacterium falsenii DSM 44353T which has been found to be associated with birds before. 16S rRNA gene sequence similarity to all known Corynebacterium spp. was significantly <97%. Corresponding values of rpoB showed low levels of similarity <87% and ANIb was <73%. G + C content of the genomic DNA was 65.0 mol% for the type strain of the goose isolates, as opposed to 63.2 mol% in Corynebacterium falsenii. MALDI-TOF MS analysis of the whole-cell proteins revealed patterns clearly different from the related species, as did biochemical tests, and polar lipid profiles. We therefore conclude that the avian isolates constitute strains of a new species, for which the name Corynebacterium heidelbergense sp. nov. is proposed. The type strain is pedersoliT (=DSM 104638T = LMG 30044T).  相似文献

This study investigated the potential of Bacillus thuringiensis isolates obtained in the Cerrado region of the Brazilian state of Maranhão for the biological control of Aedes aegypti larvae. The isolates were obtained from soil samples and the identification of the B. thuringiensis colonies was based on morphological characteristics. Bioassays were run to assess the pathogenicity and toxicity of the different strains of the B. thuringiensis against third-instar larvae of A. aegypti. Protein profiles were obtained by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Polymerase chain reaction assays were used to detect the toxin genes found in the bacterial isolates. Overall, 12 (4.0%) of the 300 isolates obtained from 45 soil samples were found to present larvicidal activity, with the BtMA-104, BtMA-401 and BtMA-560 isolates causing 100% of mortality. The BtMA-401 isolate was the most virulent, with the lowest median lethal concentration (LC50) (0.004 × 107 spores/mL), followed by the Bacillus thuringiensis var. israelensis standard (0.32 × 107 spores/mL). The protein profiles of BtMA-25 and BtMA-401 isolates indicated the presence of molecular mass consistent with the presence of the proteins Cry4Aa, Cry11Aa and Cyt1, similar to the profile of Bacillus thuringiensis var. israelensis IPS-82. Surprisingly, however, none of the cry and cyt genes analyzed were amplified in the isolate BtMA-401. The results of the present study revealed the larvicidal potential of B. thuringiensis isolates found in the soils of the Cerrado region from Maranhão, although further research will be necessary to better elucidate and describe other genes associated with the production of insecticidal toxins in these isolates.  相似文献

Mosquitoes play a key role in the transmission of some important diseases. The need for controlling these insects is critical to reduce their risks to human and domesticated animals. Recently the trend to explore effective chemical compounds from local plants has begun as a safe means of control. The present study aimed to evaluate the anti-larval activity of Chrysanthemum extract and the prepared silver nanoparticle (AgNPs) against the Aedes aegypti mosquito, the dengue vector in Saudi Arabia. A series of different concentrations of ethanol extract and extract prepared AgNPs against the fourth-life larvae was tested. The effective concentrations of crude extract and AgNPs ranged from 50 to 250 and 10 to 30 ppm respectively, and the death percentages corresponding to these concentrations ranged from 18 to 92 and 36 to 96% respectively. According to the LC50 values of treated larvae, AgNPs (12.754 ppm) is more effective against A. aegypti mosquito larvae than the crude extract (228.345 ppm) at about 17.9 times. The mixing of the plant extract with the silver nitrate has led to potentiation. This is due to the synergy that occurs between the extract and the silver particles during the reduction process. The compounds in the extract are related to the surface of the particles, increasing the strength of their effects. It is recommend to separate the active elements in the Chrysanthemum plant and its preparation in the form of nanoparticles as a promising compound in mosquito control programs with least damage to human kind and the environment.  相似文献
