The potential distribution within the internodal axon of mammalian nerve fibers is derived by applying known node potential waveforms to the ends of an equivalent circuit model of the internode. The complete spatial/temporal profile of action potentials synthesized from the internodal profiles is used to compute the node current waveforn, and the extracellular action potential around fibers captured within a tubular electrode. For amphibia, the results agreed with empirical values. For mammals, the amplitude of the node currents plotted against conduction velocity was fitted by a straight line. The extracellular potential waveform depended on the location of the nodes within the tube. For tubes of length from 2 to 8 internodes, extracellular wave amplitude (mammals) was about one-third of the product of peak node current and tube resistance (center to ends). The extracellular potentials developed by longitudinal and radial currents in an anisotropic medium (fiber bundle) are compared.  相似文献

The action potential of the non-seismonastic plant, Luffa cylindrica,was studied and characterized. Single action potentials of 30to 60 mV were evoked by electrical or cooling stimuli appliedto the stem internode. Action potentials were transmitted withoutdiminution along stimulated internodes shorter than 40 mm inlength. No difference in velocity between acropetal and basipetaltransmission was observed. This behavior is very similar tothat of Mimosa and Biophytum. The velocity of transmission ofaction potentials along internodes of Luffa was dependent upondistance from the point of stimulation; 10 to 20 mm sec–1upto 40 mm, and 70 mm sec–1 at around 50 mm from the pointof stimulation. The action potential was not transmitted tothe neighboring internode. Action potentials were also observedin the petiole and leaf. Beyond the threshold stimulus, theamplitude of the transmitted action potential depended on thestrength of the stimulus. The absolute and relative refractoryperiods for the transmission of action potentials were 2 minand 2 to 5 min, respectively. Elongation growth of the stem, recorded using a differentialtransformer, decreased after the generation of a single actionpotential in the upper most internode. Exact cable theory is used to calculate voltage distributions along fully myelinated axons and those with various patterns of demyelination. The model employed uses an R-C circuit for the soma, an equivalent cable for the dendrites, a myelinated axon with n internodes and a cable representing telodendria. For the case of a voltage clamp at the soma, a system of 2n + 1 equations must be solved to obtain the potential distribution and this is done for arbitrary n. An explicit calculation is performed for one internode whereas computer-generated solutions are obtained for several internodes. The relative importance of the position of a single demyelinated internode is determined. An approximate expression is given for the critical internodal length necessary for action potential generation.  相似文献

A mathematical model of relation between the miorelaxant effect value of aminocyclitol antibiotics and their blood levels is proposed. The kinetics of the reduction of the neuromuscle transmission damaged under the action of the drugs was studied in detail in acute experiments with cats treated with gentamycin or streptomycin administered intravenously. The changes in the effect value in time were satisfactorily described by the logistic function. Parallel determination of the blood levels of the antibiotics in the animals provided construction of an adequate model of their pharmacokinetics. Tabulation of the corresponding biexponential equations and logistic functions for the same time intervals during reduction of the neuromuscle transmission provided necessary information for plotting the "effect: concentration" curves. Comparison of the drug levels at which the effect of inhibition of the neuromuscle transmission was equal to 50% of the maximum one, revealed a statistically reliable difference in the miorelaxant activity of gentamycin and streptomycin. A high miorelaxant activity of gentamycin in comparison to streptomycin was also shown on comparison of the average "effect: concentration" curves plotted on the basis of tabulation of the general equations presenting a combination of the logistic and biexponential equations.  相似文献

Several plant species possessing at least two leaves at eachnode, or well anatomically distinguished nodes, were examinedfor potential differences in node and internode construction,and unusual histological features were sought as markers ofthe nodes and internodes. In this capacity idioblastic cellsfilled with phenolic compounds can serve, in either node (Polygonum)or internode (Sambucus). When constructed in a special way intercellularspaces can also be markers, e.g. in Rumex, as can glands innodes and internodes (Angelica, Ugustrum, Astrantia). Such histologicaldifferences exist exclusively in internodes or nodes examined,permitting inferences to be made about the origins of theseplant parts. Multifactorial analysis of the combined action of rifampicin and a microbial peptidoglycan on the immune response to antigens of the vaccine EV fraction 1 was performed. The results were computer-processed and the second order equations describing delayed hypersensitivity (DH) and antibody titers were derived. Nomographs or equal level curves showing interrelations of the investigated factors were plotted. The character of the combined action on DH and antibody titers was heterogeneous. The peptidoglycan had a pronounced immunostimulant action on DH and, to a lesser extent, influenced the humoral response. Conditions for the peptidoglycan use aimed at immunostimulation were optimized with application of multifactorial analysis.  相似文献

Study of the lifespans of normal (non mutant) mice and growth delay mutants has shown that mortality rates for both kinds of animals exhibit reproducible fluctuations. In the case of the mutant mice, the positions of peaks on the differential mortality curves (mortality rate plotted against lifespan) coincided in different-sex groups of animals and in same-sex subgroups of animals. Differential mortality curves of the mutant mice also had a peak at 1 month of age that was absent from the differential mortality curves of the normal mice. In the case of normal animals, positions of most peaks were the same in the studied independent subgroups of males, and to a lesser extent – independent subgroups of females, which might be explained by a shift in mortality peak positions due to the reproductive activity of females. Similar positions of mortality rate peaks in the differential mortality curves for animals from independent groups and subgroups indicate the existence of increased risks of death at specific ages. The observed pattern could be due to the programming in the genome of both the periods of increased risk of death and the intermitting intervals of stable development.  相似文献

The formation of the lower nodes and internodes in maize (Zea mays L.) and the progression of their differentiation was investigated by generating clonal sectors from cells of the apical meristem. Marked clones were induced by irradiating dry seeds (kernels) and 2-, 8- and 13-day-old seedlings heterozygous for anthocyanin markers (b, pl) and a chlorophyll factor (wd). The extent and apparent number of cells generating the internodes 2–6, which normally remain condensed, were traced by promoting the elongation of these internodes with gibberellic acid. At the mature seed stage, internodes 2 and 3 are undergoing longitudinal expansion and each is represented by two or three circumferential populations of cells. Internodes 4 and 5 are in the process of radial expansion and each is represented by a single circumferential population of cells. At nodes 2–4, the cells for leaves and internodes have separated but such a separation has not occurred for nodes 5 and 6. The formation and expansion of basal six internodes progressed acropetally, i.e. from the base toward distal nodes. Analysis of sectors induced at the seedling stage shows that the formation of middle and top internodes also progress acropetally. The basal, middle and top internodes were found to develop at different apparent cell numbers in the apical meristem.  相似文献

The objective of this study was to determine whether the Hodgkin–Huxley model for unmyelinated nerve fibres could be modified to predict excitability behaviour at Ranvier nodes. Only the model parameters were modified to those of human, with the equations left unaltered. A model of a single Ranvier node has been developed as part of a larger model to describe excitation behaviour in a generalised human peripheral sensory nerve fibre. Parameter values describing the ionic and leakage conductances, corresponding equilibrium potentials, resting membrane potential and membrane capacitance of the original Hodgkin–Huxley model were modified to reflect the corresponding parameter values for human. Parameter temperature dependence was included. The fast activating potassium current kinetics were slowed down to represent those of a slow activating and deactivating potassium current, which do not inactivate. All calculations were performed in MATLABTM. Action potential shape and amplitude were satisfactorily predicted at 20, 25 and 37°C, and were not influenced by activation or deactivation of the slow potassium current. The calculated chronaxie time constant was 65.5 μs at 37°C. However, chronaxie times were overestimated at temperatures lower than body temperature.  相似文献

A computer simulation was performed to investigate the size index as a motor unit identifier in electromyography. The size index calculated from the amplitude and area of the simulated motor unit action potential (MUP) was plotted against the distance between the needle electrode and current source to show how the index changes as a function of the distance. The index of the MUP also was plotted against the number of muscle fibers belonging to a single motor unit, the size of the motor unit territory, and the diameter of the muscle fibers in order to establish the major determinants of the index. The index was relatively constant for the distance less than 2 mm between the needle electrode and closest edge of the current source. It changed logarithmically with the number of muscle fibers and with the diameter of the fibers.  相似文献

With yeast subjected to the action of phenol or of phenol and alcohol the curves of survivors as measured by resistance to staining with methylene blue when plotted against time do not in general prove to be logarithmic. It is believed that such resemblances as have been found between such curves and monomolecular reaction or logarithmic curves are superficial and fortuitous. Any method therefore of evaluating disinfecting power based upon such a concept must prove misleading. Variations in resistance of individual cells and the distribution of such variations must be regarded as of fundamental importance in accounting for rates of death of microorganisms.  相似文献

This paper presents the construction, derivation, and test of a mesh model for the electrical properties of the transverse tubular system (T-system) in skeletal muscle. We model the irregular system of tubules as a random network of miniature transmission lines, using differential equations to describe the potential between the nodes and difference equations to describe the potential at the nodes. The solution to the equations can be accurately represented in several approximate forms with simple physical and graphical interpretations. All the parameters of the solution are specified by impedance and morphometric measurements. The effect of wide circumferential spacing between T-system openings is analyzed and the resulting restricted mesh model is shown to be approximated by a mesh with an access resistance. The continuous limit of the mesh model is shown to have the same form as the disk model of the T-system, but with a different expression for the tortuosity factor. The physical meaning of the tortuosity factor is examined, and a short derivation of the disk model is presented that gives results identical to the continuous limit of the mesh model. Both the mesh and restricted mesh models are compared with experimental data on the impedance of muscle fibers of the frog sartorius. The derived value for the resistivity of the lumen of the tubules is not too different from that of the bathing solution, the difference probably arising from the sensitivity of this value to errors in the morphometric measurements.  相似文献

The growth patterns of axillary buds of dayneutral tobacco (Nicotiana tabacum L. cv. Wisconsin 38) plants were assessed by using expiants of single buds attached to leafless stem cuttings and allowing the buds to grow to flowering without additional manipulation. Buds located 5, 10 and 15 nodes below the inflorescence were employed. For a given bud position, when a cutting had few internodes the growth pattern of a bud tended to fall into one of two groups: buds that produced few-noded shoots and buds that produced many-noded shoots. For example, in a group of 13 cuttings composed of bud 5 with 2 associated internodes, 11 buds produced 14.2 nodes (range, 11–17) and 2 buds produced 32.0 nodes (range, 30–34). As the number of internodes on the cutting increased, the number of buds producing few-noded shoots increased and the number of nodes produced decreased (e.g. in contrast to the data above, all 5th buds with 6 internodes produced 12.8 nodes; range 11–15). When cuttings from the 3 positions had the same number of internodes, the more apical cuttings had buds that produced fewer nodes (e.g. for cuttings with 6 internodes all 5th buds produced 12.8 nodes, all 10th buds produced 15.5 nodes and 85% of 15th buds produced few-noded shoots with 19.3 nodes). The number of nodes produced by a bud was a function of the original position of the stem piece and not the original position of the bud. That is, bud 5 associated with the 6 internodes below it produced 12.8 nodes and bud 10 associated with essentially the same 6 internodes (i.e. the 6 above it) produced 12.9 nodes while bud 10 associated with the 6 internodes below it produced 15.5 nodes. Thus, the number of nodes produced by a bud was dependent upon the original main-axis position of the cutting as well as the number of internodes on the cutting. Buds forced to grow out in situ on main axes devoid of leaves produced substantially more nodes than similar buds on cuttings. Buds isolated without associated internodes produced many-noded plants with a number of nodes similar to that of plants grown from seed. A depolarisation of the membrane potential difference (about-170 mV) of Chara corallina is observed in response to changes in cell turgor pressure using the pressure probe technique. The depolarisation occurs in phase with the pressure pulse (0.2 s duration) and is independent of the direction of the applied pressure gradient. This latter finding is in contradiction to results predicted on the basis of electro-kinetic phenomena. Pressure induced electrical leakages were ruled out by measuring the depolarisation in response to pressure in individual segments of the internode which were electrically isolated from one another. The changes in potential were recorded by external electrodes and an internal electrode which was positioned close to the micropipette of the pressure probe inserted through one of the electrically isolated nodes. The depolarisation in response to increasing positive or negative pressure gradients in the intact node region and in the intact middle segments was comparable to that monitored in the node region containing the pressure probe. Action potentials were initiated when the pressure gradients exceeded at least 2 bar. The action potentials were elicited at random in one of the two compartments adjacent to the node regions, but were never found to be initiated in the node regions themselves. The pressure-induced action potentials are explained in terms of an electro-mechanical compression (or expansion) of a local membrane area and discussed in their relevance to the propagation of pressure signals in response to water and salt stress in higher plants.Abbreviations PD potential difference  相似文献

Isolated internodes of Chara corallina and Nitella flexilis have been used to determine the concentration of one passively permeating solute in the presence of non-permeating solutes. The technique was based on the fact that the shape of the peaks of the biphasic responses of cell turgor (as measured in a conventional way using the cell pressure probe) depended on the concentration and composition of the solution and on the permeability and reflection coefficients of the solutes. Peak sizes were proportional to the concentration of the permeating solute applied to the cell. Thus, using the selective properties of the cell membrane as the sensing element and changes of turgor pressure as the physical signal, plant cells have been used as a new type of biosensor based on osmotic principles. Upon applying osmotic solutions, the responses of cell turgor (P) exactly followed the P(t) curves predicted from the theory based on the linear force/flow relations of irreversible thermodynamics. The complete agreement between theory and experiment was demonstrated by comparing measured curves with those obtained by either numerically solving the differential equations for volume (water) and solute flow or by using an explicit solution of the equations. The explicit solution neglected the solvent drag which was shown to be negligible to a very good approximation. Different kinds of local beers (regular and de-alcoholized) were used as test solutions to apply the system for measuring concentrations of ethanol. The results showed a very good agreement between alcohol concentrations measured by the sensor technique and those obtained from conventional techniques (enzymatic determination using alcohol dehydrogenase or from measurement of the density and refraction index of beer). However, with beer as the test solution, the characean internodes did show irreversible changes of the transport properties of the membranes leading to a shift in the responses when cells were treated for longer than 1 h with diluted beer. Leymus chinensis (Trin.) Tzveh is a rhizomatous perennial herbage of Gramineae. Reproduction is mainly by vegetative reproduction. Tillering nodes and rhizomes of L. chinensis serve as organs for both vegetative reproduction and nutrient storage. Water-soluble carbohydrate (WSC) contents were measured in tillering nodes, nodes and Internodes of rhizomes of different age classes of L. chinensis populations at three development stages, namely the dough ripe stage, the vegetative growth stage after full ripeness, and the withering stage, in two habitats: sandy soil and saline-alkaline soil. The results showed that WSC content in tillering nodes of the three age classes of L. chinensis were all markedly decreased with increasing age in both sandy soil and saline-alkaline solh A similar trend of changes In WSC contents was observed in the nodes and internodes of rhizomes in different age classes In both habitats. The highest WSC contents were in 2-age-class nodes and internodes of rhizomes, followed by those In the 1 age class, with the lowest WSC contents found in 3-age-class nodes and internodes of rhizomes at the dough ripe and vegetative growth stages after full ripening. In turn, WSC contents decreased with Increasing age at the withering stage in both habitats. The WSC content in each age class of internode was higher than that in the node of rhizome at three development stages in both habitats.  相似文献

Relations are derived that describe the combined effects of electrodiffusion, the Na/K pump, and Na/Ca transport by carrier on the resting membrane potential. Equations are derived that apply to both steady-state and non-steady-state conditions. Some example calculations from the equations are plotted at different permeability coefficient ratios, PK:PCa:PNa. The equations predict a depolarizing action of Na/Ca transport when more than two Na ions per Ca ion are transported by the carrier. For all permeability ratios examined, a steady state for Ca ions is achieved with at most a few millivolts of depolarization.  相似文献

1. Using the loss of turgidity of the cells as a criterion it is found that the toxicity curve of copper chloride with Nitella is sigmoid. An empirical equation can be constructed which will approximately fit the curve. 2. When the concentration of the copper chloride is varied the toxic effect varies as a constant, fractional, power of the concentration. This relation holds when the concentration is plotted against either (1) the time necessary to reach a given point on the ordinate of the survivor curve, (2) the maximum speed of toxic action as shown by the tangent to the survivor curve or (3) the first derivative of the equation which fits the survivor curve. 3. When the temperature is varied and the logarithm of the reciprocal of the time necessary to reach a given point on the survivor curves is plotted against the reciprocal of the absolute temperature the resulting figure consists of several intersecting curves. A hypothetical system is described which will give straight lines under normal conditions and curves when acted upon by a toxic agent.  相似文献

Membrane currents through potassium channels activated by nicorandil, which has a potent coronary vasodilating action, have been studied in ventricular cells of guinea pigs by using the single pipette whole-cell clamp technique. In the presence of 0.1 mM nicorandil, the duration of the action potential was shortened from 196 to 145 ms. Nicorandil markedly increased outward currents at potentials positive to the resting potential. When the difference in the currents before and after the application of nicorandil were plotted against the membrane potential, the current-voltage relation reversed close to the potassium equilibrium potential. The difference current during depolarizing pulses showed no time-dependent relaxation. These results indicate that the current evoked by nicorandil is carried by K+ ions and has voltage-independent kinetics. Power-density spectra obtained in the presence of nicorandil were fitted well by a single Lorentzian curve with a corner frequency of 4.4 Hz. The amplitude of the single-channel unit current was estimated from the relation between the variance and the mean current, and was 0.27 +/- 0.1 pA (n = 7) at -35 mV. The estimated slope conductance was 4.6 +/- 1.7 pS. Nicorandil did not affect Ca2+ currents. It is concluded that nicorandil activates a small-conductance K+ channel without affecting the Ca2+ channel.  相似文献

Fabric and growth mode of deep-water isidid gorgonian skeletons showing bright Mg-calcitic internodes and dark proteinageous nodes were investigated on modern, subrecent and fossil skeletons. The internodial microstructure is characterised by three-dimensionally interfingering calcitic fascicles accreting around a central axis. Macroscopic colour banding results from varying orientations of organic-rich fascicle bundles and intercalated bands of organic-poor granular crystals. This skeletal structure of isidid gorgonians strikingly differs from the density banding of scleractinians. Radiocarbon dating of a fossil skeleton gave an age of 3,985±35 to 3,680±35 years before present (BP) with a record of 305±35 years (±range). Linear extension rates of 0.4 mm year−1 average allow for an annual to sub-annual resolution on micrometer scale of colour bands or fascicles, respectively. The growth mode of branched skeletons is characterised by simultaneous secretion of vertically alternating nodes/internodes and lateral accretion of concentric increments enveloping the entire skeleton. Bifurcations at various growth stages imply that adjacent branches have different ages and show varying numbers of growth bands at any skeletal cross section. The scleroprotein gorgonin plays a crucial role in the formation of organic nodes and the secretion of calcitic internodes by providing a structural framework in the biomineralisation process.  相似文献
