A method has been described for the detection of maltase activity in starch gels. This method has been used to resolve the difficulties encountered when analysing animals with weak AmIB activity for Ami genotype. It was found that, with the herd of the Hannah Research Institute, the heterozygote excess observed could be explained largely on the basis of these weak AmIB bands.  相似文献

Summary A method is described for thein situ assay of acetylene reduction activity in the rhizosphere of rice involving the use of inexpensive, lightweight equipment. The assay is quick to set up and thus many replicates can be run simultaneously. Control of any acetylene-reducing blue-green algae in the assay chamber is achieved with a photosynthetically active herbicide which has no measurable effect on rhizosphere activity. Results using this method are compared with those from an excised root assay of the same plants. Only a very weak correlation between the two methods was found.  相似文献

A histochemical method for the determination of IAA-oxidase has been used in sections of various aerial parts of winter wheat plants. High IAA-oxidase activity was localized in the cell walls of sclerenchyma near the periphery of the stem, in the vascular bundle sheath of sclerenchyma and in xylem, both in the stem and in the leaf. The cell wall—bound IAA-oxidase activity therefore appeared in lignifying tissues. The staining was very weak or absent in the cell walls of parenchyma tissues and phloem. The positive reaction of the cytosol at the bulbous ends of guard cells and in the leaf primordia is presumed to be due to cytosolic IAA-oxidase. These results are discussed in relation to peroxidase localization and to our previousin vitro studies.  相似文献

Summary The pH decrease in a phosphate buffer due to fermentation of glucose to lactic acid by non-growing Lactobacillus plantarum cells has been studied. The method used offers a quick and reproducible way of measuring the glucose-fermenting activity of L. plantarum. The maximum observed velocity of pH decrease is linear with the biomass concentration and is defined as the activity of the cell suspension. With L. plantarum, recalculation of this arbitrary unit (pH·min–1 per gram dry weight) to a conceivable unit of lactic acid production rate (mol·min–1 per gram dry weight) is possible. This recalculation is based on the titration theory of a weak base with a weak acid. Para-tert-octylphenol (OP) is an important chemical intermediate mainly used for production of phenolic resins and lacquers. OP has weak estrogen-like activity in vitro, with induction of vitellogenin in fish hepatocytes and competitive estrogen receptor binding and estrogen-responsive gene expression in mammalian systems. Induction of vitellogenin in fish plasma and liver, in vivo, has also been measured and weak estrogen-like activity in rodent screening assays occurs but at much lower potency than predicted by the in vitro activity. Conventional aquatic wildlife toxicity testing shows effects on survival, growth and development, and reproduction occurring at concentrations lower than the estrogen-like responses predicted from screening assays. Studies in rodents have provided evidence of metabolic saturation at high doses of OP, and a multigeneration reproductive study confirms the lack of estrogen-like activity below this metabolic saturation as well as the lack of reproductive toxicity. This article reviews the weight of evidence for the aquatic and mammalian data and compares the results of the definitive assays to those of the screening tests. The results show that the estrogen-like activity in screening tests is not predictive of results from definitive testing, and that risk assessment decisions should be made using conventional toxicity endpoints.  相似文献

Alkaline phosphatases have been demonstrated enzymhistochemically in the testis of the common frog Rana temporaria, caught in the month of January. The follicle cells in the cyst walls and the lamina propria of the seminiferous tubules showed strong enzymatic activity; also a weak activity was observed in the peripheral region of some of the germinal cells lying within the cysts. The possible interaction of alkaline phosphatases in the transport processes across the cyst walls has been discussed.  相似文献

Cellulase and β-D-glucosidase activity have been identified in Venturia inaequalis, the causal agent of apple scab. Most of the activity of the enzymes was associated with the mycelium after homogenization. The activities were relatively weak in vitro compared to those of other plant pathogens. With a new screening technique, β-D-glucosidase hyperproducing mutants were obtained.  相似文献

Thirty-five ketone-isobenzofuranone hybrids ( 1 – 35 ) were designed, synthesized, and evaluated for their herbicidal activity against Chinese amaranth (Amaranthus tricolor) and barnyard grass (Echinochloa crus-galli). The structure-activity relationship (SAR) results revealed that the position and type of substituent were crucial for activity. The o-substituted derivatives outperformed the m- and p-substituted derivatives. Compounds with strong electron-donating groups (OH, OMe) had low activity, while those with heterocycles (N-methylpyrrole, furan, and thiophene) had a moderate herbicidal effect. Compounds with a weak electron-donating group (Me) and weak, moderate, and strong electron-withdrawing groups (F, Cl, Br, and NO2) showed promising herbicidal activity. Among these, the o-F substituted compound ( 20 ) was the most effective against Chinese amaranth, and the o-Cl substituted compound ( 23 ) was the most potent against barnyard grass. This is the first time the herbicidal potential of ketone-isobenzofuranone hybrids has been studied. The discovery of current chemical clues would be beneficial for the development of novel herbicides.  相似文献

In this paper, a novel chemiluminescent (CL) method for the determination of benzhexol has been developed by combining the flow injection technique and its sensitizing effect on the weak CL reaction between sulfite and acidic cerium(IV). A mechanism for the CL reaction has been proposed on the basis of CL spectra. Under the optimized conditions, the proposed method allows the measurement of benzhexol hydrochloride over the range 0.1–10 μg/mL with a correlation coefficient of 0.9992 (n = 8), a detection limit of 0.02 μg/mL (3σ), and a relative standard deviation for 2.0 μg/mL benzhexol (n = 11) of 1.65%. Synopsis Serial transverse sections of porcine longissimus dorsi muscle (18 pigs, 50 to 178 kg live weight) were reacted for NAD tetrazolium reductase and ATPase at pH9.4, and for glycogen with the periodic acid-Schiff (PAS) reaction. Three histochemical types of muscle fibre were identified; (1) strong ATPase and weak NADH oxidative activity; (2) strong ATPase and intermediate NADH oxidative activity; and (3) weak ATPase and strong NADH oxidative activity. Immediatepost mortem samples from one side of each animal were compared with a laterpost mortem sample from the other side by measuring the absorbance of PAS-stained glycogen at 570 nm with a microscope photometer. Laterpost mortem absorbance was expressed as a percentage of immediatepost mortem absorbance in each category of muscle fibre in order to compensate for distributional error and different starting levels of glycogen. Muscle fibres with weak ATPase and strong NADH oxidative activity showed a progressive decrease in absorbance of PAS-stained glycogenpost mortem. In some animals, fibres with strong ATPase and intermediate or weak NADH oxidative activity showed an initialpost mortem increase in absorbance of PAS-stained glycogen which was then followed by a progressive decrease. The maximum rates of decrease in absorbance in the three fibre types did not differ to any great extent.  相似文献

Two new pairs of enantiomeric butenolides, (+)- and (−)-suberiteslide A, (+)- and (−)-subertieslide B had been obtained from the marine sponge Suberties sp. The structures with absolute configurations of these compounds were unequivocally determined by spectroscopic analyses and ECD (Electronic Circular Dichroism) method. It was the first separation of butenolides from the marine sponges of genus Suberites. Additionally, the anti-inflammatory, antibacterial and cytotoxic activities of these compounds were evaluated. The result indicated that only (−)-subertieslide B showed weak anti-inflammatory activity with the IC50 value of 40.8 μM.  相似文献

Quantitative assessment, using three Pseudomonas sp. strains, of the activity of the microbial biocide Soncid 8101 demonstrated that the values of effective sublethal concentrations (L50) differed by 500% (because of individual variations in the sensitivity of the test strains). The spread of parameters of biocidal activity could be narrowed by using a mixture of microorganisms with high, medium, and weak resistance. A method for quantitative assessment of the activity of microbial biocides was proposed based on the use of natural associations of soil bacteria.  相似文献

Two chlorophenyl-containing antibiotics have been isolated from a strain ofActinoplanes (ATCC 33002). Antibiotic A 15104 Y is a chlorinated phenylpyrrole compound whose structure has been confirmed by chemical synthesis. Antibiotic A 15104 Z is a chlorophenol derivative for which a structural formula is proposed on the basis of its physicochemical properties. A 15104 Y shows antimicrobial activity against Gram-positive, Gram-negative, and acid-fast bacteria, yeasts, fungi, and protozoa, while A 15104 Z possesses a low activity against Gram-positive bacteria andTrichophyton. A 15104 Y has a weak activity in curing experimentally infected mice, at a dose that is one-fifth the LD50 for the same species. This is the first example of a chloropyrrole derivative isolated from an actinomycete.  相似文献

A new acetylenic fatty acid, 1 , has been isolated from the title sponge. The structure of the molecule was elucidated to contain an enyne and a thiophene by spectroscopic methods. Compound 1 showed a weak cytotoxic effect against NBT‐T2 rat bladder epithelial cells (IC50>20 μg/ml), and antimicrobial activity with minimal‐inhibitory concentrations (MIC) of 64 and 128 μg/ml against Staphylococcus aureus and Escherichia coli, respectively.  相似文献

A method of calculation of the circular dichroism (CD) of random-coil polypeptides has been developed by means of a Monte-Carlo approach to the treatment of statistical systems and exciton theory of optical activity of polymers. The contribution of π–π* and n–π* amide transitions to the CD has been taken into account. The π–π* transition gives rise to two CD bands: a negative short-wavelength band and a weak positive long-wavelength one. The n –π* transition gives rise to one negative CD band.  相似文献

Cytoadhesion of Plasmodium falciparum‐infected erythrocytes to endothelial protein C receptor (EPCR) is associated with severe malaria. It has been postulated that parasite binding could exacerbate microvascular coagulation and endothelial dysfunction in cerebral malaria by impairing the protein C–EPCR interaction, but the extent of binding inhibition has not been fully determined. Here we expressed the cysteine‐rich interdomain region (CIDRα1) domain from a variety of domain cassette (DC) 8 and DC13 P. falciparum erythrocyte membrane protein 1 proteins and show they interact in a distinct manner with EPCR resulting in weak, moderate and strong inhibition of the activated protein C (APC)–EPCR interaction. Overall, there was a positive correlation between CIDRα1–EPCR binding activity and APC blockade activity. In addition, our analysis from a combination of mutagenesis and blocking antibodies finds that an Arg81 (R81) in EPCR plays a pivotal role in CIDRα1 binding, but domains with weak and strong APC blockade activity were distinguished by their sensitivity to inhibition by anti‐EPCR mAb 1535, implying subtle differences in their binding footprints. These data reveal a previously unknown functional heterogeneity in the interaction between P. falciparum and EPCR and have major implications for understanding the distinct clinical pathologies of cerebral malaria and developing new treatment strategies.  相似文献

A sensitive and simple chemiluminescent (CL) method for the determination of diclofenac sodium has been developed by combining the flow injection technique and its sensitizing effect on the weak CL reaction between formaldehyde and acidic potassium permanganate. A calibration curve is constructed for diclofenac sodium under optimized experimental parameters over the range 0.040–5.0 µg/mL and the limit of detection is 0.020 µg/mL (3σ). The inter‐assay relative standard deviation for 0.040 µg/mL diclofenac sodium (n = 11) is 2.0%. This method is rapid, sensitive, simple, and shows good selectivity and reproducibility. The proposed method has been successfully applied to the determination of the studied diclofenac sodium in pharmaceutical preparations with satisfactory results. Furthermore, the possible mechanism for the CL reaction has been discussed in detail on the basis of UV and CL spectra. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

Summary The ability to convert cyclohexanecarboxylic acid to hippuric acid has been studied in liver from guinea pigs, rabbits, rats and mice using a gas chromatographic- mass spectrometric method employing selected ion monitoring. Guinea pig liver showed the highest activity, giving values double of those found in rabbit liver and five times those in rat liver. Only very weak activity was found in mouse liver. (Hydroxymethyl)cyclohexane, cyclohexanealdehyde and a-hydroxyethylcyclohexane, which are structurally related to cyclohexanecarboxylic acid but lack the carboxyl group, were not aromatized by guinea pig liver mitochondria. This finding indicates that the carboxyl group is essential for aromatization. Absence of aromatization was also found with the homologs cyclohexaneacetic acid and cyclohexanepropionic acid and with the di-acidstrans-1,2- andtrans-1,4-cyclohexanedicarboxylic acid. The effect of a methyl group in cyclohexanecarboxylic acid depended on its position. 2-Methyl-1-cyclohexanecarboxylic acid was not aromatized, however the 3- and 4-methyl derivatives underwent aromatization and subsequent conjugation with glycine. The rates of formation ofm-methyl- andp-methylhippuric acid were 16% and 9%, respectively, of that found for hippuric acid from cyclohexanecarboxylic acid (8.0 nmol/min/mg protein).  相似文献

The avian eggshell is one of the fastest calcifying processes known and represents a unique model for studying biomineralization. Eggshell strength is a crucial economic trait for table egg production, and ensures that a safe egg reaches the consumer kitchen. However, a common toolkit for eggshell mineralization has not yet been defined. In this issue, label‐free MS‐based protein quantification technology has been used by Sun et al. (Proteomics 2013, 13, 3523–3536) to detect differences in protein abundance between eggshell matrix from strong and weak eggs and between the corresponding uterine fluids bathing strong and weak eggs. Proteins associated with the formation of strong eggshells are identified, which are now candidates for further investigations to define the regulatory relationship between specific eggshell matrix proteins and calcite crystal texture.  相似文献
