用高效液相色谱法测定银杏叶提取物经酸水解后黄酮甙元含量.采用C18柱,甲醇:水:磷酸(55:44.5:0.5)为流动相,检测波长370nm,方法回收率95.1%~103.2%,变异系数(CV)3.17%,10批样品含测平均为25.4%.  相似文献

本文介绍一种用高效波相色谱(HPLC)同时测定血清中24-脱氢胆固醇、7-烯胆甾烷醇、菜油甾醇、胆甾烷醇和β-谷甾醇的方法。血清加内标(6-氯豆甾醇)后用氢氧化钾醇溶液皂化,用正己烷提取其中的各种甾醇,将提出的甾醇衍生为苯氨基甲酸酯,用HPLC分离测定。本法样品处理简单,色谱分析时间短,批内和批间变异系数分别为2.2~3.8%和3.3~6.7%。本文也首次报告我国成年人血清非胆固醇甾醇水平,5种甾醇血清浓度的总和平均为1.54mg/dl,是胆固醇的0.86%。  相似文献

用超临界萃取技术对产于青藏高原上的糖茶蔗种子中脂肪酸进行萃取,其萃取率为12%。用毛细管气相色谱对萃取的脂肪酸进行了分离和分析。结果表明不饱和脂肪酸的质量分数在90%以上。其中α-亚麻酸(α-linolenic acid)为27.4%,У-亚麻酸(У-linolenic acia)为2.70%。亚油酸(1inoleic acid)40.0%,油酸(oleic acid)21.0%。  相似文献

目的:建立可同时测定甲磺酸伊马替尼原料药中甲醇、乙醇和N''N - 二甲基甲酰胺(DMF)等3 种残留有机溶剂的毛细管气相色谱法。方法:采用WondaCAP WAX 毛细管柱为色谱柱,程序升温,N2 为载气,载气流速为4.0 mL ? min-1,氢火焰离子化检测器,检测器温度为250 ℃,进样口温度为200 ℃,进样量为1.0 μL。以正丁醇为内标,通过内标法计算样品中3 种有机溶剂残留量。结果:甲醇、乙醇和DMF 在所建立的色谱条件下均能完全分离,分别在2.94~44.11、4.91~73.68 和0.084~12.60 mg ? L-1 的质量浓度范围内线性关系良好(r =0.9940~0.9985),进样精密度(RSD)均小于5.0%(n =6),平均回收率为104.8%~105.8%(RSD<5.0%,n =9),定量限分别为0.20、0.33 和0.08 ng(信噪比为10 ∶ 1),检测限分别为0.05、0.11 和0.02 ng(信噪比为3 ∶ 1)。样品中未检出甲醇和DMF,乙醇含量低于有关限度规定。结论:该方法简便、准确、灵敏、重复性好,可用于甲磺酸伊马替尼原料药中残留有机溶剂的检测。  相似文献

采用气相色谱法测定抗毒素中的间甲酚含量。样品用苯甲醇作内标,经高氯酸沉淀蛋白、氯仿抽提后,通过HP-INNO-WAX柱,以氢火焰检测器测定间甲酚含量。内标峰与样品峰的分离度大于1.5,平均回收率及变异系数分别为100.067%和1.93%,测定线性范围0.0202~1.7588mg/ml,最小检测限为0.01272mg/ml,该方法准确快速,比化学方法好。  相似文献

采用胰脂酶分解的方法对几种乌桕[Sapium sebiferum(L.) Roxb.]种子桕脂甘油三酯组成进行了测定,桕脂的甘油三酯是由9-14种甘油三酯所组成,与可可脂相似之处是甘油三酯2-位上的脂肪酸,82-88%为油酸,二饱和酸甘油酯中81-88%,其中以POP为主(74-81%);单饱和酸甘油酯占1.8-6.0%;而三饱和酸甘油酯占9.6-15.6%,主要是PPP(8.5-14.0%)。桕脂经过适当的加工处理,可以做可可脂的代用品,将湖南产的乌桕与广西栽培的4个品种(“大粒鸡爪”,“蜈蚣”,“铜锤”和“小粒鸡爪”)桕脂中的甘油三酯组成比较,以“大粒鸡爪”桕脂中二饱和酸甘油酯的比较最高(88.0%),三饱和酸甘油酯的含量最低(9.6%)。  相似文献

摘要:本文建立了简单且准确的测定1,3-二羟基丙酮(DHA)的高效液相色谱(HPLC)方法。以Alltima C18(5μm,250×4.6mm)为分离柱,5%甲醇水溶液(0.05%H3PO4调pH至3.0),流速为1mL/min,用紫外检测器在200nm处检测DHA。结果测得DHA标准样品的保留时间为6.2min,并测得DHA的线性范围为0.1~10.0 g/L。用HPLC法测得以甘油为底物发酵产DHA发酵液中DHA含量为6.2 g/L。并证明高效液相色谱法可有效应用于产DHA发酵过程中产物的检测。  相似文献

应用高效液相层析技术,对重楼属十八个种植物的甾体皂甙进行了定性、定量分析。植物用甲醇提取,抽出物经DIAION柱,以90%甲醇洗出总甙。在HPLC上,用ODS柱先将总甙以用醇:水(9:1)洗脱分为三馏段,每馏再在ODS柱或Rp-8柱上以甲醇:水(8:2;7:3.5)洗脱,使各个皂甙成分完全分离。被分离的每个色谱峰与已知重楼皂甙的保留时间进行比较并配合HPLC的加入法,TLC分析及用HPLC制备少量样品做MS测定来加以定性鉴定。定量采用内标及校正曲线法。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
