Effect of feeding defatted millet (Sorghum vulgarie) flour at 5, 10 and 14.5% protein levels respectively for six weeks has been studied on rat liver mitochondrial, microsomal and supernatant fractions total lipids, cholesterol, triglycerides, total phospholipids, phosphatidyl choline and phosphatidyl ethanolamine. The results have been compared with rats fed casein at 10% level for the same period. The metabolism of liver subcellular fractions lipids of millet diet and casein diet fed rats has been studied by the incorporation of acetate-1-14C and . A significant increase in mitochondrial triglycerides of rats fed millet diet at 5 and 10% protein level, in microsomes of rats fed millet diet at 5, 10 and 15% protein levels and in supernatant fractions of rats fed millet diet at 5 and 15% protein levels was observed. A significant increase in total cholesterol in mitochondria and microsomes and a significant decrease in supernatant fraction of rats fed millet diet at 10% protein level was observed. A significant increase in mitochondrial total phospholipids, phosphatidyl choline and phosphatidyl ethanolamine in rats fed millet diet at 10% protein level and a decrease in these in rats fed millet diet at 5 per cent protein level was observed. In microsomes total phospholipids were increased in rats millet diet at 10% protein level and phosphatidyl choline was increased in rats fed millet diet at 15% protein level. Total phospholipids, phosphatidyl choline and phosphatidyl ethanolamine were significantly reduced in the supernatant fraction of rats fed millet at 10% protein level.

Incorporation of acetate-1-14C into nonsaponifiable fraction of mitochondria, microsomes and supernatant fractions of rats fed millet diet at 5 and 15 % protein levels was significantly greater, and in saponifiable fractions of the above subcellular fractions was greater in rats fed millet diet at 5 per cent protein level. The specific activity (counts/min/mg) of free cholesterol in mitochondria, microsomes and supernatant fractions of millet diet fed rats was significantly greater, whereas the specific activity of triglycerides was not significantly different from the controls. The acetate-1-14C specific activity of phosphatidyl choline and phosphatidyl ethanolamine was significantly greater in all the above subcellular fractions of millet diet fed rats (except of phosphatidyl choline in rats fed millet diet at 5 % protein level). The specific activities of phosphatidyl choline were significantly greater in mitochondria of rats fed millet diet at 5 % protein level and of phosphatidyl choline and phosphatidyl ethanolamine in microsomes and supernatant fractions of rats fed millet diet at 5 and 15% protein levels. The specific activities of phosphatidyl choline were significantly decreased in mitochondria and microsomes of rats fed millet diet at 10% protein level. The total acetate-1-14C activities (counts/min/g equivalent wet liver) of free and esterified cholesterol triglycerides, phosphatidyl choline and phosphatidyl ethanolamine showed that their synthesis from acetate-1-14C was either enhanced in millet diet fed rats or was comparable to the controls. The total activity of (counts/min/g equivalent wet liver) into phosphatidyl choline and phosphatidyl ethanolamine showed that their synthesis was decreased in microsomes of rats fed millet diet at 10% protein level, increased in rats fed millet diet at 5 and 15% protein levels.  相似文献

Mitochondria isolated from tobacco leaves incorporated 14C-leucine into the protein and the rate was enhanced by tobacco mosaic virus (TMV) infection as compared with noninfected level. In vitro amino acid incorporation by mitochondria required adenosine triphosphate (ATP), Mg2+, and KC1 and the energy sources from oxidative phosphorylation as well as from ATP-generating system. This incorporation was inhibited by ribonuclease (RNase), deoxyribonuclease (DNase), actinomycin D, mitomycin C, puromycin, and chloramphenicol added in the reaction medium. The pretreatment of the mitochondria with DNase and actinomycin D reduced the rate of incorporation. The mitochondria incorporated 3H-guanosine triphosphate (GTP) and this activity was blocked by actinomycin D. The presence in this system of 15,000 g supernatant cell sap fraction or bacterial contamination was carefully checked obtaining a negative result. The reaction product into which l4C-amino acids incorporated was solubilized by trypsin. The nature of the amino acid incorporating activity of isolated mitochondria obtained from TMV-infected tobacco leaves is discussed.  相似文献

We have used the newly introduced method of De Lorenzo & Freedman (1978) for isolating synaptic vesicles to determine if such vesicles contain both serotonin (5-HT) and serotonin binding protein (SBP). Two fractions were obtained. A 55, 000 g fraction was morphologically heterogeneous and contained coated vesicles. A 135, 0000 vesicle (dia. 51.3 nm) fraction was homogeneous in ultra-structure and contained no coated vesicles. The specific activity of SBP in this fraction was much higher than that in the supernatant. Unlike SBP, very little lactic dehydrogenase activity appeared in the 135, 000 g fraction. Qualitative and quantitative differences were observed between the polypeptide profiles of soluble proteins extracted from the vesicles and supernatant proteins on SDS gels. Therefore, entrapment of cytosol in the vesicles of the 135, 000 g fraction was minimal. The 5-HT concentration of the 135, 000 g vesicles was 5.5 ng/mg protein and in the supernatant, 11.3 ng/mg protein. The ATP concentration in the 135, 000 g vesicle fraction was only 0.8 ng/mg Pr. Rabbit spinal cords were transected in order to determine if SBP is moved proximo-distally in axons by rapid axonal transport as would be predicted for a constituent of synaptic vesicles. SBP accumulated above the cut at a rate consistent with fast transport (78 mm/day). SBP activity fell caudal to the point of transection and there was no evidence, such as an accumulation below the lesion, that might indicate retrograde transport of SBP. These experiments indicate that SBP is probably synthesized in the cell bodies of serotonergic neurons and some is rapidly transported down axons to be stored in terminals in vesicles.  相似文献

In an attempt to study on metabolic changes in rats fed on an amino acid diet devoid of one branched chain amino acid and of niacin, rats were force-fed a leucine-free, isoleucine-free, valine-free or complete amino acid diet for 3 or 4 days and killed 3 hr after the feeding on day 4 or 5 to observe the body weight changes, the urinary nitrogen and N1-methylnicotinamide (MNA), and liver tryptophanpyrrolase (TPase) and tyrosine-α-keto-glutarate transaminase (TKase) activities.

The excretion of the urinary nitrogen and MNA, TPase and TKase activities, and fat content of livers of rats force-fed these amino acid deficient diets were higher than those fed the complete amino acid diet. It was further confirmed in the present study that changes in TPase activity of rats given diets devoid of one essential amino acid were in the same direction with changes in urinary MNA which was observed in the previous studies on rats given threonine-free, tryptophan-free, methionine-free, lysine-free and complete amino acid diets. However, such metabolic changes in rats fed the leucine-free diet were not so remarkable, compared with those of rats fed the other amino acid deficient diets.  相似文献

Urinary excretion of acid soluble peptide (ASP)-form amino acids was lower in rats deprived of protein than in rats fed on a 20% casein or 20% gluten diet. However, the amino acid pattern of urinary ASP was similar among each of the three dietary groups, suggesting that urinary ASP is mainly endogenous origin under these nutritional conditions.

College women who were given a meat-free protein diet for 3 days after 10 days’ protein deprivation excreted 1.4 times the amount of ASP-form amino acids during protein deprivation.

The rate of urinary excretion of ASP-form amino acids in the state of protein deprivation was proportional to the metabolic body size of organisms as far as rats and women were concerned.

Streptozotocin-induced diabetic rats excreted two times the amount of ASP-form amino acids compared with normal rats. This suggests that endogenous protein catabolism doubled in diabetic rats.

When labelled urinary ASP was injected into rats, approximately 40% of the label was recovered as urinary ASP within 24 hr. This excretion rate was far higher than that after the injection of free leucine.

The rate of urinary excretion of ASP-form amino acids correlated with that of Nτ-methylhistidine in rats.

These results favor the hypothesis that urinary ASP reflects the catabolism of body proteins.  相似文献

The increase in the incorporation of amino acids into protein in vitro by preparations obtained from protein-fed rats as compared with preparations obtained from carbohydrate-fed rats has been described previously. After molecular sieving through Sephadex G-25 of cell-free preparations, the difference in incorporating activity between the two types of rats was diminished in systems containing ATP, phosphoenolpyruvate, pyruvate kinase, GTP, and a mixture of amino acids. When, after molecular sieving, a mitochondrial (15,000 g) supernatant was incubated for 4 min at 35°C the polysomal pattern of the preparations was unchanged. In the presence of ATP, phosphoenolpyruvate, and pyruvate kinase the polysomal incorporating activity was low and the polysomal pattern was only slightly changed. Addition of GTP increased the activity markedly, and a more pronounced activity was observed when a mixture of amino acids was added as well. As the amino acid incorporation ability increased, monosomes were formed from the polyribosomes. The activity of the polyribosomes was severalfold higher than that of non-Sephadex-treated preparations, indicating an activation of polysomal aggregates which under the usually applied conditions of incubation and prior to molecular sieving show little or insignificant activity. It was possible to activate polyribosomes from carbohydrate-fed and protein-fed rats to almost the same extent.  相似文献

SYNOPSIS. The presence of amino acid activating enzymes was demonstrated in the ciliated protozoan Tetrahymena pyriformis. By employing a sensitive hydroxamate assay procedure, the activation of L-valine was assayed in various subcellular fractions of the ciliate, and some characteristics of the enzyme activity in the most active fraction were determined. Most of the activity resided in pH 5 fractions isolated from high speed supernatants of ciliates disrupted by various physical and chemical methods. No activity could be demonstrated in isolated cilia, in pellicles with attached kinetosomes, in microsomes or in macronuclei, providing these organelles were thoroughly washed. A washed mitochondrial preparation isolated by the Mager and Lipmann procedure activated L-valine; mitochondria isolated by the procedure of Hogg and Kornberg did not. The pH 5 fraction isolated from the 102,000 X g supernatant of digitonin-lysed ciliates was stable for several weeks when stored in 0.1 M Tris buffer, pH 7.6 at – 25 C. The activity of this fraction with respect to L-valine activation was dependent on the presence of ATP1 and magnesium in the reaction mixture. The optimal concentrations of these components and of L-valine and hydroxylamine were determined, and the linearity of activity with time and enzyme concentration was demonstrated. Valine activation was not modified by dialysis of the pH 5 fraction, or treatment with RNase, or the addition of boiled pH 5 fraction.  相似文献

14C-Amino acid incorporating activity in the absence of exogenousmRNA was found in a cell-free system from embryos of light-germinatedPinus thunbergii seeds, but not in that from dark-imbibed seedembryos. Template activity in the cell-free system from thelight-germinated seed embryos was observed in the ribosome fraction,especially the polyribosome fraction, but not in the 100,000?gsupernatant fraction (s100). These facts suggest that the natureof the block in protein synthesis during the imbibition of seedsin the dark is due to the lack or inactivity of mRNA. Homogenates of kidney cortex obtained from control rats and rats treated with DDT have been separated into microsomes or ribosomes, and into postmicrosomal (S105) supernatant fraction or pH 5 supernatant fraction. The incorporation of [14C]phenylalanyl-tRNA into peptide was increased when microsomes derived from kidneys of DDT-treated rats were incubated with pH 5 supernatant fraction from control rats. Elongation factors (EF) 1 and 2, necessary for the binding of aminoacyltRNA to ribosomes and for translocation of peptidyl-tRNA from the A site to the P site of ribosomes, were present in the pH 5 supernatant fractions of kidney of control and DDT-treated rats and these fractions were incubated with KCl-washed ribosomes obtained from livers of control rats. The results provided evidence that the increased incorporation observed with the pH 5 supernatant fraction obtained from the DDT-treated animals could not be attributed to decreased ribonuclease activity or to increased elongation factor 2 activity but was due to an increase in elongation factor l activity.  相似文献

Tobacco callus fed L-tyrosine-UL?14C was sampled at 3-day intervals for 15 days, homogenized and studied with respect to distribution of incorporated radioactivity. The supernatant obtained by centrifuging of the homogenates at 270 g contained the bulk of the radioactivity although significant activity was also detected in the pellet. Sucrose density gradient centrifugation of the supernatant showed over 90% of the recovered label to be associated with a fraction designated as “less dense than mitochondria”, with the remainder being found in the fraction identified as “mitochondria”. During tissue culture, virtually all of the radioactivity in the fraction “less dense than mitochondria” was recovered in the supernatant obtained by centrifugation at 100,000 g. From 4 to 18% of the labeling in the 100,000 g supernatant fraction was attributable to tyrosine-containing protein, and the rest to free tyrosine and unidentified anionic constituents. The highest proportions of radio-activity in the 270 g pellet were associated with substances extractable with NaCl, pronase, 4.6 N NaOH, and acetolyzing reagent. Low but substantial labeling characterized the extracts obtained with Triton X-100 and 1 N NaOH. The final unextractable residue contained 20% of the 270 g pellet radioactivity.  相似文献

To study the mechanism of fatty infiltration in the liver due to added sulfur-containing amino acids to low casein diet, the effect of sulfur-containing amino acids and glutathione (GSH) on the incorporation of acetate-l-14C into lipid fractions were studied in liver slices from rats fed on 8% casein diet (Basal diet) with or without added methionine (Met).

The liver acetyl Co A carboxylase activities of rats on basal diet with or without added Met were similar.

Addition of Met, cystine or cysteine to the incubation medium had little effect on lipogenesis of slices. On addition of GSH to liver slices from rats fed on basal diet, lipid formation increased appreciably. On the other hand, addition of GSH to liver slices from rats fed on Met supplemented diet showed no accelerative effect on lipogenesis.

Addition of GSH to the incubation medium of liver slices from rats fed on basal diet tended to reduce the incorporation of acetate into the phospholipid fraction and to increase into the fatty acid fraction of liver slices.

The content of liver GSH was lower in rats on basal diet than in those on Met supplemented diet. The higher GSH level in rats on Met supplemented diet may be one factor causing fatty infiltration in the liver of these animals.  相似文献

The present study was conducted to investigate the effect of zinc deficiency on fatty acid desaturation in rats fed two different types of dietary fat, a mixture of coconut oil and safflower oil (7∶1, w/w, “coconut oil diet”) or linseed oil (“linseed oil diet”). In order to ensure an adequate food intake, all rats were force-fed by gastric tube. Zinc deficiency caused statistical significant reducion of Δ9-desaturase activity in liver microsomes of rats fed coconut oil diet and tendencial reduction (p<0.15) in rats fed linseed oil diet compared with control rats fed diets with the same type of fat. In agreement with this effect, zinc deficiency in the rats fed both types of dietary fat increased the ratio between total saturated and total monounsaturated fatty in liver phospholipids and liver microsomes. Zinc deficient rats on the coconut oil diet had unchanged Δ6-desaturase activity with linoleic acid as substrate and lowered activity with α-linolenic acid as substrate. In contrast, zinc deficient rats on the linseed oil diet had increased Δ6-desaturase activity with linoleic acid as substrate and unchanged activity with α-linolenic acid. Because linoleic acid is the main substrate for Δ6-desaturase in the rats fed coconut oil diet, and α-linolenic acid is the main substrate in the rats fed linseed oil diet, it is concluded that in vivo Δ6-desaturation was not changed by zinc deficiency in the rats fed both types of dietary fat. Activity of Δ5-desaturase was also not changed by zinc deficiency in the rats fed both dietary fats. Levels of fatty acids in liver phospholipids and microsomes derived by Δ4-, Δ5-, and Δ6-desaturation were not consistently changed by zinc deficiency in the rats fed both types of dietary fat. Thus, the enzyme studies and also fatty acid composition data of liver phospholipids and microsomes indicate that zinc deficiency does not considerably disturb desaturation of linoleic and α-linolenic acid. Therefore, it is suggested that similarities between deficiencies of zinc and essential fatty acids described in literature are not due to disturbed desaturation of linoleic acid in zinc deficiency. The present study also indicates that zinc deficiency enhances incorporation of eicosapentaenoic acid into phosphatidylcholine of rats fed diets with large amounts ofn-3 polyunsaturated fatty acids.  相似文献

The state of aggregation and the activity of polyribosomes as well as the activity of the pH 5 enzyme fraction were studied at two stages of postnatal brain development, 9 and 50 days after birth. When the polyribosomes were prepared at 0°C in the presence of 5 mm -Mg2+, more than 85 per cent of the polyribosome material exhibited a sedimentation coefficient higher than 110 S. High Mg2+ concentrations are, therefore, unnecessary to obtain highly aggregated brain polyribosomes. The basal amino acid incorporating activity of both 9- and 50-day-old rat brain preparations is at least equal to that of rat liver. When prepared by the same procedure as above, 9-day-old rat brain polyribosomes seem to be more active (20 per cent) than those of adult brain. However, this difference in activity depends on the presence of a non-ribosomal inactive contaminant which is always present in higher amounts in adult brain preparations. When purified from this contaminant, the preparations do not differ in activity. High Mg2+ concentrations are also not necessary for optimal protein synthetic activity and, in fact, are inhibitory. When assayed with both types of highly aggregated polyribosomes, the pH 5 enzyme fraction from adult brain is clearly less active than that of 9-day-old rats. These results suggest that the loss of brain protein synthesis during development does not depend on the stability of the messenger RNA-ribosome complex but only on the soluble pH 5 enzyme fraction.  相似文献

Changes in the metabolism in vivo of amino acids with the lapse of time after feeding a diet were investigated by measuring the incorporation of 14C into some body components one hour after injection with 14C-amino acid mixture.

The incorporation of 14C into protein in the liver and carcass was rather constant, but that into blood sugar, liver glycogen, and lipids in the liver and carcass showed a change with the lapse of time after feeding a 25% casein diet or a protein-free diet. The incorporation of 14C into liver glycogen was stimulated shortly after feeding, but it was reduced at 7 hr, when a large amount of glycogen was still in the liver. On the contrary, the specific activity of blood sugar increased with the lapse of time after feeding. The conversion of 14C-amino acids into lipids in the liver and carcass was stimulated shortly after feeding.

The incorporation of 14C into protein was higher in the rats fed the protein-free diet than in those fed the 25% casein diet, and the higher incorporation was partly counterbalanced by the lower incorporation of 14C into lipids and glycogen in the rats fed the protein-free diet.  相似文献

Male rats of the Holtzman strain were fasted for 3 days and refed a diet high in carbohydrate (68.9%). The induction of liver glucose 6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase was monitored for up to 48 h after refeeding. Induction occurred by 24 h, and by 48 h, 4.2- and 1.5-fold increases were observed for glucose 6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase, respectively, compared with that of livers of pellet-fed rats. After refeeding, lysosomes increased in fragility as judged by an increased release of acid phosphatase activity during standard homogenization. Fragility was greatest 3 h after refeeding, but normal fragility was observed 24 h after refeeding. Nuclei were isolated from the liver samples before and after refeeding. Those isolated just before refeeding revealed small latent acid phosphatase activity (4–6%). However, after refeeding the carbohydrate-rich diet, a transient and significant (P < 0.01) increase in the latent activity occurred that was maximal (20%) at 1 h, returning to normal by 24 h. Cross-mixing the 800g nuclear pellet from livers of animals starved for 3 days with the 800g supernatant fraction from livers of animals refed the carbohydrate-containing diet did not alter the nuclear lysosomal-free (overt) or latent (detergent-released) enzyme activity. Similarly, mixing the 800g nuclear pellet from livers of animals refed for 1 h with the 800g supernatant fraction from livers of animals starved for 3 days, but not refed, did not change the nuclear lysosomalfree or latent enzyme activity. Purified nuclei, further washed in hypotonic buffer, lost acid phosphatase activity, but those isolated from livers of rats refed for 1 h retained 10% of the enzyme latency, whereas all latency was lost from those isolated from uninduced rats. A second lysosomal enzyme, β-galactosidase, became associated with the nuclei with the same temporal pattern as for acid phosphatase. However, no variation in nuclear content of cytosolic lactate dehydrogenase occurred as a result of feeding the high-carbohydrate diet to starved rats. When similarly starved rats were refed a diet high in lipid and carbohydrate-free, no induction of glucose 6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase was observed. Lysosomes were not temporarily fragile and purified nuclei did not exhibit increased latency of acid phosphatase activity. Though the evidence presented does not establish a direct correlation between lysosome migration to nuclei as a required function in enzyme induction, the temporal and specific nature of the phenomenon support the hypothesis that liver lysosomal enzymes participate in early signals in the induction of enzymes of lipogenesis.  相似文献

The chemical composition of perinuclear crystals of Lychnis chalcedonica and intranuclear crystals of Dianthus barbatus in leaf tissue embedded in glycol methacrylate was tested by differential enzyme digestion of ultra-thin sections. The Lychnis crystals were digested readily by pepsin but with great difficulty and never to completion by trypsin. The Dianthus crystals were rapidly and completely digested by pronase; inconsistently and with difficulty by pepsin; and were unaffected by trypsin. Pre-incubation with water increased the effectiveness of pepsin digestion. Neither RNase nor DNase had any effect, either alone, or when preceded or followed by the proteinases. It was concluded that the Lychnis crystals are composed of protein that is probably high in aromatic amino acid content, and possibly low in the basic amino acids arginine and lysine. The Dianthus crystals also seem to be protein, but because of their amino acid composition or conformation, and possibly because of complex reactions with the fixative, these crystals are not readily hydrolyzed by pepsin and trypsin.  相似文献

Rapidly dividing Ehrlich ascites cells have a higher rate of protein synthesis than slowly-growing cells. This is true whether protein synthesis is measured in vivo or in a cell-free amino acid incorporating system. A lack of endogenous messenger RNA or a ribosomal defect in stationary cells is suggested as a likely cause of the decreased rate of protein synthesis. The soluble proteins synthesized by rapidly-dividing cells are qualitatively different from those synthesized by slowly-growing cells. The latter also have a larger intracellular pool of several amino acids and a slower protein turnover than rapidly-dividing cells.  相似文献

The microsomal fraction from the spleen (after perfusion) of immunized rabbits incubated for 20 min at 37° C under usual conditions in the presence of energy sources incorporates14C-labelled amino acids both into the solubilized (by adding deoxycholate), and into the nonsolubilized part (15%). The cell supernatant incorporates under these conditions the14C-labelled amino acids into total proteins in the absence of microsomes but in a lower degree. The cell supernatant contains gamma globulin detectable by immunoelectrophoresis. Gamma globulin obtained by specific precipitation of the solubilized microsomal fraction with antigamma-globulin serum had an measurable radioactivity. The precipitate of gamma globulin obtained from the supernatant of the incubation medium in the same manner (after removing the microsomes) had a specific activity twice as high. On separating the microsomal fraction extract and the incubation medium supernatant on DEAE cellulose most fractions show on extinction maximum at 260 nm in the first case and at 280 nm in the second case. The microsomal fraction isolated from the spleen and lymph nodes of immunized pigs-48 and 72 h after revaccination, when incubatedin vitro, incorporated14C-labelled amino acids into total protein. After ultrasonic disintegration in 0.14m NaCl and filtration through a Sephadex G 25 column it is specifically precipitated with the antigammaglobulin serum. Gamma globulin isolated after incubation of the microsomal fraction had a measurable radioactivity. AntiHSA antibodies determined by adsorption on immunosorbent did not possess significant radioactivity. Only the concentrated supernatant of the incubation medium showed minute radioactivity of 75–94 counts/min /ml. The problem of investigating the formation of nascent specific antibodies on a subcellular levelin vitro during the early period of secondary response to the antigen is discussed, in particular the problem of their detection. Male Sprague-Dawley rats, 4 weeks (young) or 8 months (adult) of age, were fed one of three purified diets free of or containing either 0.5% cholesterol or 0.5% oxidized cholesterol (92% oxidized cholesterol) for 3 weeks. Feeding of oxidized cholesterol caused a significant reduction of food intake, body weight gain, and relative liver weight in rats of both ages. The activity of the HMG-CoA reductase and cholesterol 7α-hydroxylase of liver microsomes, the key enzymes in cholesterol synthesis and catabolism, respectively, was lowered by oxidized cholesterol compared to the diet free of cholesterol in both ages, and the difference was significantly in the adult. On the other hand, the activity of Δ6-desaturase of liver microsomes, a key enzyme in linoleic acid metabolism to arachidonic acid, was significantly increased by oxidized cholesterol in adult rats, leading to the increase in linoleic acid desaturation index [(20:3n-6 + 20:4n-6)/18:2n-6] in liver phospholipids. Oxidized cholesterol reduced the concentration of cholesterol in serum and liver. Also, the fecal excretion of acidic steroids was lower in rats fed the oxidized cholesterol diet than in those fed the cholesterol-free diet. Thus, oxidized cholesterol significantly influenced cholesterol and fatty acid metabolism in particular in adult rats.  相似文献
